A new type of apparatus for sectioning samples of hard, undecalcified bone is described. Slices of fresh or archeological human bone 4-5 mm thick are dehydrated and then embedded in epoxy resin. The apparatus used to prepare sections from the resulting blocks consists of a low-speed rim-type diamond cut-off wheel and a slowly advancing table carrying the specimen held in a rotating mount. Sections may be cut at a thickness of 80 micron +/- 1%. After cleaning in an ultrasonic bath, these can be mounted on slides for quantitative microscopic examination with transmitted light. Grinding and polishing are not necessary. The results obtained are illustrated.  相似文献

A new type of apparatus for cutting 100-200µ sections of bone is described. This apparatus consists of an especially designed bone vise and a mounted circular saw with three directional movement controls. The saw is driven by one D.C. motor and its vertical movement assembly is driven by another. Their speeds can be regulated individually. The bone sections may be made serially at intervals of 0.3 to 0.5 mm. and bones as large as 2 cm. in diameter or longitudinal slabs of larger bones may be sectioned.  相似文献

The cortical bone distributions in the femoral necks of apes and humans differ as a result of different loading environments caused by the realignment of the hip abductor apparatus. Femoral neck cortical bone in extant humans is very thin superiorly and thicker inferiorly, while the cortical bone in apes tends to be more uniformly thick. The unique internal anatomy of extant humans allows inferences to be made about primary locomotor function from incomplete femora. Here the differences in cortical bone distributions are quantified using moment coefficient of skewness. Skewness coefficients at two locations along the neck of the 6 million years old African femoral specimen BAR 1002’00 were compared to samples of 9 extant adult humans and 10 adult chimpanzees. The skewness coefficients of cortical bone in the femoral neck of BAR 1002’00 are more similar to those of chimpanzees than to humans, although the contrast is less pronounced in the region closer to the neck-shaft junction than more proximally toward the femoral head; this pattern indicates that in at least one respect this specimen attributed to Orrorin tugenensis manifests structural features suggesting influences of a hip abductor apparatus that had not yet evolved to the same extent as in extant humans.  相似文献

The periodontal ligament (PDL), a soft tissue connecting the tooth and the bone, is essential for tooth movement, bone remodeling and force dissipation. A collagenous network that connects the tooth root surface to the alveolar jaw bone is one of the major components of the PDL. The organization of the collagenous component and how it changes under load is still poorly understood. Here using a state-of-the-art custom-made loading apparatus and a humidified environment inside a microCT, we visualize the PDL collagenous network of a fresh rat molar in 3D at 1 μm voxel size without any fixation or contrasting agents. We demonstrate that the PDL collagen network is organized in sheets. The spaces between sheets vary thus creating dense and sparse networks. Upon vertical loading, the sheets in both networks are stretched into well aligned arrays. The sparse network is located mainly in areas which undergo compressive loading as the tooth moves towards the bone, whereas the dense network functions mostly in tension as the tooth moves further from the bone. This new visualization method can be used to study other non-mineralized or partially mineralized tissues, and in particular those that are subjected to mechanical loads. The method will also be valuable for characterizing diseased tissues, as well as better understanding the phenotypic expressions of genetic mutants.  相似文献

An apparatus has been constructed to cool biological material at a controlled rate. The material to be frozen is placed in glass ampuls which are immersed in an aluminum bath containing ethyl alcohol and the bath is placed inside a freezer cabinet. Liquid nitrogen is pumped intermittently into the cabinet by means of a single-speed electric pump. The rate of cooling is controlled by a device that varies the interval between successive pumping cycles. The temperature fall is monitored by thermocouples placed inside selected glass ampuls and recorded as a plot on moving graph paper.This simple instrument is capable of cooling at an accurately controlled rate over the range of 0 to 7 °C/min. We chose for our studies a cooling rate of 1 °C/min which we could maintain with an accuracy of ±0.1 °C. Temperature fluctuations were, however, observed at the freezing plateau and varied considerably in magnitude and temperature at onset even for the same material cooled under the same conditions. Mouse bone marrow cells frozen by our technique and stored for various periods of time may, on reconstitution, form colonies in vivo and in vitro identical in morphology and number to those from unfrozen control cells. Our results suggest that expensive and intricate devices may not be necessary to obtain optimal recovery of viable cells after storage in liquid nitrogen. The apparatus is now in regular use for the storage of human bone marrow cells intended for use in treatment of patients with leukemia refractory to conventional measures.  相似文献

Human adenovirus 1 was studied for its effect on the chromosomal apparatus both in bone marrow cells and male sex cells of mice. Chromosome aberrations were most early detected in spermatocytes of the 1st order mice infected with human adenovirus 1. In bone marrow cells of mice the highest level of chromosome aberrations was observed 30, 60, 90 days after the inoculation, which corresponds to a more frequent detection of the adenoviral antigen. The UV-irradiated-virus caused chromosome aberrations in the later periods after the inoculation which might be induced by the virus reactivation in a cell.  相似文献

The ossicular apparatus of golden moles in the genus Chlorotalpa has received comparatively little attention in the literature, although the malleus is known to be intermediate in size between the "unmodified" malleus of Amblysomus and the hypertrophied mallei found in some other golden moles. In the present study, the middle ear structures of three Chlorotalpa species (C. duthieae, C. sclateri, and C. arendsi) are described. Measurements of middle ear structures were applied into three existing models of middle ear function. The predictions from the models suggest that the airborne hearing of Chlorotalpa species is limited to relatively low frequencies, but the impedance transformation by the middle ear apparatus is expected to be reasonably efficient. The sensitivity of the middle ear apparatus to inertial bone conduction is intermediate between that predicted for Amblysomus and that predicted for species with hypertrophied mallei. Hearing in fossorial mammals may be limited by factors other than the middle ear apparatus: the predictions for Chlorotalpa must therefore be treated with caution. However, a consideration of the "intermediate" middle ear morphology of Chlorotalpa species sheds some light on the origin of ossicular hypertrophy in golden moles. The limited enlargement of the malleus seen in Chlorotalpa is expected to have improved seismic sensitivity by bone conduction significantly at low frequencies, while airborne hearing might not have been adversely affected.  相似文献

Mammalian NEFA and nucleobindin are calcium-binding proteins containing a signal peptide, two EF-hand motifs, acidic and basic regions and a leucine-zipper motif. Although they have been discussed to be involved in autoimmunity, apoptosis and calcium homeostasis in the Golgi apparatus and bone matrix, their exact role remains unknown. Here we report the cloning of their Drosophila homolog, nucb1, as well as the analysis of its expression pattern during embryogenesis and the subcellular localization of the NUCB1 protein. The nucb1 mRNA and the NUCB1 protein were found to be expressed maternally and zygotically, and they accumulate ubiquitously at low levels during all embryonic stages due to a maternal component. From stage 11 onward, high levels of zygotic expression can be detected specifically in the salivary glands and their placodes. In contrast to the known mammalian family members, the NUCB1 protein localizes in a subpattern of cytoplasmic substructures, probably the Golgi apparatus.  相似文献

The anatomy of the hyoid apparatus and positional changes of the hyoid bone during mastication and deglutition are described in the New Zealand White rabbit (Oryctolagus cuniculus). A testable model is constructed to predict the range of movement during function of the hyoid, a bone entirely suspended by soft tissue. Frame-by-frame analysis of a videofluorographic tape confirms the accuracy of the prediction through observation of hyoid bone excursion during oral behavior. During chewing, translation of the hyoid bone is diminutive and irregular, lacking a clearly discernible path of excursion. However, some movements of the hyoid occur with regularity. During fast opening, anterodorsal movement of the hyoid is interrupted with an abrupt posteroventral depression when the bolus is moved posteriorly toward the cheek teeth by the tongue. This clockwise rotation (when viewed from the right side) of the hyoid accompanies jaw opening and is reversed (posteroventral movement) for the jaw closing sequence. Lateral movements of the hyoid may be slightly coupled to mandibular rotation in the horizontal plane. The findings suggest that the hyoid bone maintains a relatively static position during the dynamics of chewing. The primary function would be to provide a stable base for the movements of the tongue. Another possible function would be to control the position of the larynx within the pharyngeal cavity. Some characteristic features of the rabbit hyoid apparatus may be consequential to relatively erect posture and a saltatory mode of locomotion.  相似文献

The osteology and myology of the illicial apparatus of Cryptopsaras couesii were examined in an effort to elucidate function. The apparatus consists of two bones, a tiny illicial bone, completely enveloped by tissue of the esca, and an extremely long supporting pterygiophore that lies within a deep groove on the dorsal surface of the head. The anterior end of the pterygiophore emerges on the tip of the snout from between the frontal bones, while the posterior end, encased in a dermal sheath when the illicial apparatus is retracted, emerges on the back just anterior to the dorsal caruncles. Five pairs of muscles control movement of the illicial apparatus: small, short erectors and depressors control movement of the illicial bone, while extremely long inclinators, protractors, and retractors control movement of the pterygiophore. The protractors and retractors of C. couesii are more robust and much longer than those of other lophiiforms, indicating that the pterygiophore of this species has an exceptionally wide range of movement in the anterior and posterior plane. Moreover, these muscles wind around the pterygiophore in opposite directions, a unique anatomy that suggests that C. couesii extends and retracts the pterygiophore by rotation.  相似文献

 Size effects of large magnitude are observed in the torsional shear modulus and damping of bovine plexiform bone. Damping increases and stiffness decreases with specimen size over all sizes studied. Measurements were conducted in torsion using a laser-based micromechanics apparatus capable of viscoelastic studies over a range of frequencies up to 100 kHz, upon samples of various size, with no parasitic friction or other errors that could mimic any size effect. Torsional tan δ at 1 Hz varies by about a factor of five over the size range 2.8–6.2 mm thick, and is more dependent on specimen thickness at 1 Hz than it is at higher frequency. The size effects are attributed to compliance and viscoelasticity of the interfaces between laminae. These laminae must be substantially stiffer than whole bone. Observed size effects are likely to play a role in understanding scaling laws of bones in living organisms. Received: 12 February 2002 / Accepted: 22 November 2002 R.S. Juveniles, females, and males of Ophidion rochei share similar external morphology, probably because they are mainly active in the dark, which reduces the role of visual cues. Their internal sonic apparatuses, however, are complex: three pairs of sonic muscles, and highly modified vertebrae and ribs are involved in sound production. The sonic apparatus of males differs from juveniles and females in having larger swimbladder plates (modified ribs associate with the swimbladder wall) and sonic muscles, a modified swimbladder shape and a mineralized structure called the “rocker bone” in front of the swimbladder. All of these male traits appear at the onset of sexual maturation. This article investigates the relationship between morphology and sounds in male O. rochei of different sizes. Despite their small size range total length (133–170 mm TL), the five specimens showed pronounced differences in sound‐production apparatus morphology, especially in terms of swimbladder shape and rocker bone development. This observation was reinforced by the positive allometry measured for the rocker bone and the internal tube of the swimbladder. The differences in morphology were related to marked differences in sound characteristics (especially frequency and pulse duration). These results suggest that male calls carry information about the degree of maturity. Deprived of most visual cues, ophidiids probably have invested in other mechanisms to recognize and distinguish among individual conspecifics and between ophidiid species. As a result, their phenotypes are externally similar but internally very different. In these taxa, the great variability of the sound production apparatus means this complex system is a main target of environmental constraints. J. Morphological, histochemical and ultrastructural investigations on epiphyseal apparatus of Rana Esculenta were made. The most important findings were the following: 1) metaphyseal cartilage is localized inside proximal diaphyseal compact bone as a plug; 2) metaphyseal cartilage do not reduce in thickness during ageing; 3) metaphyseal cartilage do not show vascular invasion and do not mineralize in degenerative zone; 4) trabecular bone was not at all evident in this animal; 5) external periosteum is well vascularized and proliferates in correspondence to marginal epiphyseal end of the diaphyseal. From these results the hypothesis that the ranid frog bone growth is not due to metaphyseal metabolism (as in avian and mammals) but to bone periosteal marginal mineralization is reached.  相似文献

With age, there is a progressive loss of body balance function. Yet, the potential influence of osteoporosis on body balance is largely unknown. Dentin matrix protein 1 (DMP1) is highly expressed in bone and required for phosphate homeostasis and mineralization. Dmp1 null mice display striking defects in bone structure. In this study we reported circling behavior and hyper reaction to touching in Dmp1 null mice. Our histology, tartrate resistant acid phosphatase (TRAP) staining and µCT data showed dramatic changes, such as an expansion of poorly mineralized matrices, in the Dmp1 null porous bony structure in the vestibular apparatus. The targeted re-expression of DMP1 in the Dmp1 null bone fully rescued not only the bone phenotype, but also circling behavior and hyper reaction. Furthermore, X-gal stain and DMP1 immunohistochemistry assay showed that DMP1 was not expressed in neuron cells or balance related cells in the inner ear, suggesting that a defect in the bony labyrinth of the internal ear is indirectly responsible for the circling behavior and/or hyper reaction to touching. Finally, discovery of DMP1 lacZ signal in pericyte-like cells may suggest a new function of DMP1 in angiogenesis.  相似文献

The equipment used for preparative polyacrylamide gel electrophoresis has been either difficult to construct or costly if purchased commercially. An inexpensive preparative acrylamide gel apparatus and peristaltic pump are described in this paper which are easy to use and may be constructed from readily available materials. The construction of the preparative gel apparatus requires no special machining or glass blowing.This report describes the use of the disc gel apparatus in the final purification step of three binding proteins which appear to be involved in the transport of the branched-chain amino acids in Escherichia coli. Two of these proteins have been described previously (1–4). The apparatus has also been successfully used in a number of other laboratories for the purification of a variety of other proteins (5–9).  相似文献

Endothelin 1 (Edn1), a secreted peptide expressed ventrally in the primordia of the zebrafish pharyngeal arches, is required for correct patterning of pharyngeal cartilage development. We have studied mutants and morpholino-injected larvae to examine the role of the Edn1 signal in patterning anterior pharyngeal arch bone development during the first week after fertilization. We observe a remarkable variety of phenotypic changes in dermal bones of the anterior arches after Edn1 reduction, including loss, size reduction and expansion, fusion and shape change. Notably, the changes that occur appear to relate to the level of residual Edn1. Mandibular arch dermal bone fusions occur with severe Edn1 loss. In the dorsal hyoid arch, the dermal opercle bone is usually absent when Edn1 is severely reduced and is usually enlarged when Edn1 is only mildly reduced, suggesting that the same signal can act both positively and negatively in controlling development of a single bone. Position also appears to influence the changes: a branchiostegal ray, a dermal hyoid bone normally ventral to the opercle, can be missing in the same arch where the opercle is enlarged. We propose that Edn1 acts as a morphogen; different levels pattern specific positions, shapes and sizes of bones along the dorso-ventral axis. Changes involving Edn1 may have occurred during actinopterygian evolution to produce the efficient gill-pumping opercular apparatus of teleosts.  相似文献

Long-term bone calcium metabolism was measured using a whole body counter apparatus and the radioisotopes calcium-47 and strontium-85. Strontium was given with calcium in a single intravenous injection in order to estimate long-term retention of calcium. Calcium-47, with a short radio-active half-life, was measured directly for 20 days, and then indirectly from strontium retention for periods up to 200 days.The standard parameters of calcium transport were measured and, as well, a new parameter, calcium accretion to the fixed bone pool. These studies were carried out on 13 subjects, six of whom were selected “normals” and seven were patients with osteoporosis. Calcium accretion to the fixed bone pool, the new index of bone formation, varied from 100 to 210 mg. of calcium daily in normal subjects and from 70 to 340 mg./day in patients with osteoporosis. These values are one-third of those for the accretion to the total bone pool, the previously reported bone formation rate.  相似文献

An external fixation apparatus immobilized to the skull has been used with satisfactory results in 15 cases of significant retrusion of the middle third of the face in which intermaxillary wiring and facial suspension had proved inadequate. The retrusion was a consequence of trauma or secondary to congenital malformation.This apparatus, developed by Delbet and Tessier, would be useful in difficult cases, such as for edentulous patients, and is an alternative to bone grafting.  相似文献

Summary Lyophyle desiccation is an extremely valuable method of preserving micro-organisms. The apparatus described above proves satisfactory. The material should be frozen as rapidly as possible at a temperature of −35°C. or lower, and the apparatus should be constructed in such a way that the pressure remains approximatively constant. During the sealing of the ampoules care should be taken that no dry material passes from the latter to the apparatus. A time limit for the desiccation process is hard to fix. When water has condensed on the outside surface of the ampoules, the apparatus is left untouched for about an hour. This brings the total duration of the desiccation as a rule up to 1 1/2 to 2 hours. A volume of 0.25 ml skimmed milk suffices for the preservation of a strain. It is advisable to start from a fresh culture that has been grown on a solid medium and to suspend the required amount in skimmed milk at a pH of 7.4–7.8. The suspension should directly be frozen in a thin layer. Bofore the frozen ampoules are submitted to the desiccation process, they may be kept for some time at a temperature of −35°C.
