Two eudesmanolides, eight lactucin-like guaianolides and five phenolic compounds were isolated for the first time from roots of Cichorium pumilum, along with two previously reported eudesmane-type sesquiterpene lactones. Rearrangements of some lactucin-like guaianolides during isolation procedures were also discussed.  相似文献

A transformed root culture of Cichorium intybus L. (Asteraceae) was found to produce sesquiterpene lactones of guaiane and germacrane type. Lactucopicrin, 8-desoxylactucin and three sesquiterpene lactone glycosides: crepidiaside B, sonchuside A and ixerisoside D were isolated from the roots. The yield of 8-desoxylactucin reached 0.03 g l(-1) at the early stationary phase of the culture.  相似文献

A new neolignan glucoside was isolated from hairy roots of Cichorium intybus. Its structure was determined as (7S, 8R)-3′-demethyl-dehydrodiconiferyl alcohol-3′-O-β-glucopyranoside based on a combination of 1D and 2D NMR techniques and CD data. In addition, further water soluble constituents, including four known phenolic compounds and one known sesquiterpene lactone glucoside, were isolated from the same source.  相似文献

A re-investigation of V. arkansana afforded several new sesquiterpene lactones, three bourbonenolides, obviously closely related to those isolated from this plant previously, a glaucolide and a methoxy derivative most probably formed by fragmentation of the corresponding bourbonenolide. From V. profuga in addition to known lactones a new guaianolide was isolated. The biogenetic relationships of the Vernonia sesquiterpene lactones are discussed briefly.  相似文献

The aerial parts of Eupatoriastrum nelsonii afforded, in addition to known sesquiterpene lactones, four new lactones all closely related to the germacranolides isolated previously from other genera of the tribe Eupatorieae. In this work extracts from roots of the common vegetable Cichorium intybus L., highly appreciated for its bitter taste, were studied to investigate their possible biological activity on fungi from a variety of ecological environments: some are parasites on plants (phytopathogens) or of animals and humans (zoophilic and anthropophilic dermatophytes), others live on the soil and only seldom parasitize animals (geophilic dermatophytes). The extracts were ineffective on geophilic species and on tested phytopathogens, with the exception of Pythium ultimum, whereas they inhibited the growth of zoophilic and anthropophilic dermatophytes, in particular Trichophyton tonsurans var. sulfureum, whose treatment caused morphological anomalies, here observed by scanning electron microscopy. This behaviour is discussed on the basis of the presence in the chicory extract of the two main sesquiterpene lactones, 8-deoxylactucin and 11β,13-dihydrolactucin.  相似文献

Investigation of the cytotoxic active components of Michelia compressa afforded two new cytotoxic sesquiterpene lactones, michelenolide and micheliolide. Parthenolide, costunolide, santamarine, reynosin and liriodenine were also isolated and exhibited cytotoxic activity. The known sesquiterpene lactones, lanuginolide and dihydroparthenolide, were isolated but were not active. Two other new inactive sesquiterpene lactones, compressanolide and dihydroreynosin, were also obtained. The structures of michelenolide and micheliolide were confirmed by partial synthesis from parthenolide and the structure of compressanolide by partial synthesis from dihydroparthenolide.  相似文献

The characteristic pungency of the European liverwort, Chiloscyphus polyanthus, is due to a mixture of four sesquiterpene lactones, two new structures ent-5β-hydroxydiplophyllin and ent-3-oxodiplophyllin, and the previously known diplophyllin and diplophyllolide. A new dihydrodiplophyllin, together with the pungent diplophyllolide and diplophyllin, has been isolated from Diplophyllum albicans. Ent-dihydrodiplophyllolides derived from the above pungent diplophyllolides produce intense numbness of the tongue. All pungent sesquiterpene lactones showed inhibitory activity towards the germination and root elongation of rice husks.  相似文献

The aerial parts of Artemisia douglasiana afforded, in addition to some known sesquiterpene lactones, 22 new closely related guaianolides. A new thiophene acetylene was isolated from the aerial parts of A. schmidtiana together with some known sesquiterpene lactones. Chicory (Cichorium intybus) is known to contain guaianolides, eudesmanolides, and germacranolides. These sesquiterpene lactones are postulated to originate from a common germacranolide, namely (+)-costunolide. Whereas a pathway for the formation of germacra-1(10),4,11(13)-trien-12-oic acid from farnesyl diphosphate had previously been established, we now report the isolation of an enzyme activity from chicory roots that converts the germacrene acid into (+)-costunolide. This (+)-costunolide synthase catalyzes the last step in the formation of the lactone ring present in sesquiterpene lactones and is dependent on NADPH and molecular oxygen. Incubation of the germacrene acid in the presence of 18O2 resulted in the incorporation of one atom of 18O into (+)-costunolide. The label was situated at the ring oxygen atom. Hence, formation of the lactone ring most likely occurs via C6-hydroxylation of the germacrene acid and subsequent attack of this hydroxyl group at the C12-atom of the carboxyl group. Blue light-reversible CO inhibition and experiments with cytochrome P450 inhibitors demonstrated that the (+)-costunolide synthase is a cytochrome P450 enzyme. In addition, enzymatic conversion of (+)-costunolide into 11(S),13-dihydrocostunolide and leucodin, a guaianolide, was detected. The first-mentioned reaction involves an enoate reductase, whereas the formation of leucodin from (+)-costunolide probably involves more than one enzyme, including a cytochrome P450 enzyme.  相似文献

An investigation of seven Vernonia species, mainly from northern Brazil, afforded in addition to known compounds three new unusual sesquiterpene lactones with an allenic group. Intensive NMR studies fully established the structures. Furthermore, two lactones derived from bourbonene, two new glaucolides, a new furansesquiterpene, a nerolidol derivative and two other sesquiterpenes derived from eudesmane were isolated. The biogenetic relationships of the sesquiterpene lactones present in Veronia species as well as the chemotaxonomic aspects are discussed.  相似文献

Three new and one known furofuran lignans—syringaresinol derivatives—along with the known phenylpropanoids cichoriin and syringin were isolated from a callus tissue of Cichorium intybus. The investigation of Stokesia laevis afforded in addition to known compounds five new sesquiterpene lactones, four hirsutinolides and one guaianolide. The structures are very similar to those isolated from several Vernonia species. Since hirsutinolides have only been previously isolated from this latter genus, these results suggest that Stokesia may be closely related to Vernonia.  相似文献

The effects of methyl jasmonate (MJ) or salicylic acid (SA) on the sesquiterpene lactone content and biomass accumulation were investigated in a hairy root culture of Cichorium intybus. The guaianolides crepidiaside B (1), 8-deoxylactucin (2), and the germacranolide sonchuside A (3) were quantified by RP-HPLC. Neither MJ nor SA affected the growth of examined hairy root culture. Jasmonate up-regulated biosynthesis of the analysed sesqiterpene lactones in the culture (maximum after 72 h). SA caused a transient increase in sonchuside A accumulation in the roots (up to twofold increase compared with the control) and decrease of guaianolide content.  相似文献

Twenty‐nine compounds were isolated from the aerial parts of the Greek plant C. zuccariniana DC. The structures of the isolated compounds were established by means of NMR‐ (1H,1H‐COSY, 1H,13C‐HSQC, HMBC, NOESY, and ROESY) and mass‐spectral analyses. These compounds comprise 13 sesquiterpene lactones, 14 flavonoids, two lignans, and one simple lactone. Among the isolated sesquiterpene lactones, three are new, namely one heliangolide, (1E,4Z)‐15‐hydroxy‐8αO‐(4′‐acetoxy‐3′‐hydroxy‐2′‐methylidenebutanoyl)‐6βH,7αH‐germacra‐1,4,11(13)‐trien‐6,12‐olide; and two eudesmanolides, 8α‐(4′,5′‐diacetoxyangeloyl)sonchucarpolide and one unusual eudesmanolide with an oxygenated bridge linking C(1) and C(4), named zuccarinin. The main sesquiterpene lactones were malacitenolide, cnicin, and 4′‐O‐acetylcnicin. These results are in agreement with those obtained from the previously studied Greek Centaurea sp. belonging to the section Acrolophus (Cass .) DC.; this finding could be of chemotaxonomic significance for the genus Centaurea. The in vitro antimicrobial activities of the isolated new sesquiterpene lactones were against eight bacteria and eight fungal species. A 96‐well microbioassay procedure for fast and easy evaluation of antibacterial and antifungal activities was applied to compare these compounds with commercial antibiotic and fungicide standards, and with previously isolated analogous sesquiterpene lactones tested by the same bioassay. All of the compounds tested showed moderate antibacterial, but significant antifungal activities; the present results corroborate with previous data, indicating that these types of compounds exhibit low or moderate antibacterial, but potent antifungal activities. The unusual eudesmanolide zuccarinin proved to be the most potent among the present tested sesquiterpene lactones, as well as among all previously tested eudesmanolides isolated from Greek Centaurea sp.  相似文献

From the blue perianth segments of Cichorium intybus we isolated four anthocyanins. The pigments were identified as delphinidin 3,5-di-O-(6-O-malonyl-beta-D-glucoside) and delphinidin 3-O-(6-O-malonyl-beta-D-glucoside)-5-O-beta-D-glucoside and the known compounds were delphinidin 3-O-beta-D-glucoside-5-O-(6-O-malonyl-beta-D-glucoside) and delphinidin 3,5-di-O-beta-D-glucoside. The aerial parts of Artemisia iwayomogi afforded, in addition to already known terpenes, eleven new cudesmanolides whereas four new guaianolides were found in A. gmelinii. No sesquiterpene lactones were detected in the aerial parts of A. santolinifolia which afforded two new nerolidol derivatives besides several other already known compounds. From A. caerulescens var. cretacea six eudesmanolides were isolated, one of which has not been reported previously. The structures were elucidated by high field 1H NMR spectroscopy and chemical transformations. The chemotaxonomic significance of sesquiterpene lactones within the section Abrotanum and the subgenus Seriphidium is briefly discussed.  相似文献
