Cistus genus is widespread in the Mediterranean regions with several species and is traditionally known as a natural remedy, but few previous phytochemical researches have been reported on Cistus species growing in Sardinia. The aim of this work was to study the interpopulation variability of the volatiles of C. creticus subsp. eriocephalus to find out the natural chemotypes of this taxon for chemotaxonomic purposes. Plant material was collected from seven wild populations in Sardinia. The oils from aerial part were obtained by hydrodistillation and analyzed by GC/MS enabling to identify 185 compounds. The seven samples showed appreciable variations in their chemical composition. MA was found almost exclusively linear hydrocarbons (85%) and MN presented fatty acids (36.97%) as major fraction. CP and MN had high quantity of non‐terpenic carbonylic compounds (63% and 69%, respectively) and in CP was found also sclareol, a well‐known antimicrobial compound. PCA analysis showed the existence of a high interpopulation variability within the essential oils of C. creticus subsp. eriocephalus growing in Sardinia. For example, MN and CP are very close to each other as CG is close to PM. BN is isolated from the others Cistus populations due to lacking essential oil. These data suggest that the basis of variation in the volatile composition of seven C. creticus subsp. eriocephalus populations depends on hybridization and that the sample without essential oil is the only one no‐hybridized.  相似文献

  • The potential resilience of shrub species to environmental change deserves attention in those areas threatened by climate change, such as the Mediterranean Basin. We asked if leaves produced under different climate conditions through the winter season to spring can highlight the leaf traits involved in determining potential resilience of three Cistus spp. to changing environmental conditions and to what extent intraspecific differences affect such a response.
  • We analysed carbon assimilation, maximum quantum efficiency of PSII photochemistry (Fv/Fm) and leaf morphological control of the photosynthetic process in leaves formed through the winter season into spring in C. creticus subsp. eriocephalus (CE), C. salvifolius (CS) and C. monspeliensis (CM) grown from seed of different provenances under common garden conditions.
  • Intraspecific differences were found in Fv/Fm for CE and CS. Carbon assimilation‐related parameters were not affected by provenance. Moreover, our analysis highlighted that the functional relationships investigated can follow seasonal changes and revealed patterns originating from species‐specific differences in LMA arising during the favourable period.
  • Cistus spp. have great ability to modify the structure and function of their leaves in the mid‐term in order to cope with changing environmental conditions. The Fv/Fm response to chilling reveals that susceptibility to photoinhibition is a trait under selection in Cistus species. Concerning carbon assimilation, differing ability to control stomatal opening was highlighted between species. Moreover, seasonal changes of the functional relationships investigated can have predictable consequences on species leaf turnover strategies.

Cistus creticus L., an aromatic species from the Mediterranean area, contains various diterpenes bearing the labdane skeleton. The production of essential oil from this species has potential economic value, but so far, it has not been optimized. In order to contribute to a better knowledge of this species and to its differentiation, the morphological characters, volatile chemical composition and genetic data of two subspecies (C. creticus subsp. eriocephalus and C. creticus subsp. corsicus) were investigated. The leaf trichomes were studied using scanning electron microscopy. The chemical composition of Corsican essential oil (C. creticus subsp. corsicus) has been reported using GC, GC/MS and 13C NMR; the main constituents were oxygenated labdane diterpenes (33.9%) such as 13-epi-manoyl oxide (18.5%). Using plant material (54 samples) collected from 18 geographically distinct areas of the islands of Corsica and Sardinia, the basis of variation in the headspace solid-phase microextraction volatile fraction and an inter-simple sequence repeat genetic analysis were also examined. It was shown that the two subspecies of C. creticus differed in morphology, essential oil production, volatile fraction composition and genetic data.  相似文献

Short‐term physiological plasticity allows plants to thrive in highly variable environments such as the Mediterranean ecosystems. In such context, plants that maximize physiological performance under favorable conditions, such as Cistus spp., are generally reported to have a great cost in terms of plasticity (i.e., a high short‐term physiological plasticity) due to the severe reduction of physiological performance when stress factors occur. However, Cistus spp. also show a noticeable resilience ability in response to stress factors. We hypothesized that in Cistus species the short‐term physiological response to stress and that to subsequent recovery can show a positive trade‐off to offset the costs of the photosynthetic decline under drought. Gas exchange, chlorophyll fluorescence, and water relations were measured in C. salvifolius, C. monspeliensis, and C. creticus subsp. eriocephalus during an imposed experimental drought and subsequent recovery. Plants were grown outdoor in common garden conditions from seeds of different provenances. The short‐term physiological response to stress and that to recovery were quantified via phenotypic plasticity index (PIstress and PIrecovery, respectively). A linear regression analysis was used to identify the hypothesized trade‐off PIstress–PIrecovery. Accordingly, we found a positive trade‐off between PIstress and PIrecovery, which was consistent across species and provenances. This result contributes in explaining the profit, more than the cost, of a higher physiological plasticity in response to short‐term stress imposition for Cistus spp because the costs of a higher PIstress are payed back by an as much higher PIrecovery. The absence of leaf shedding during short‐term drought supports this view. The trade‐off well described the relative variations of gas exchange and water relation parameters. Moreover, the results were in accordance with the ecology of this species and provide the first evidence of a consistent trade‐off between the short‐term physiological responses to drought and recovery phases in Mediterranean species.  相似文献

Phenolic constituents and essential oil from the aerial parts of Cistus creticus subsp. eriocephalus (Viv.) Greuter et Burdet growing in central Italy were analysed by HPLC-MSn and GC–MS, respectively. Furthermore, six constituents were isolated by semipreparative HPLC from the methanol extract and their structures were determined on the basis of 1D and 2D NMR measurements as well as MS spectra. Isolated compounds were one new natural product, i.e. the shikimic acid ester 3,5-diihydroxy-4-(O-β-d-glucopyranosyl)-cyclohex-1-en-1-(O-β-d-glucopyranosyl)-ester (27), and six flavonoid glycosides, namely quercetin-3-O-β-D glucopyranoside (16), quercetin-3-O-rhamnoside (17), tricetin-4′-O-β-D glucopyranoside (24), tricetin-4′-O-β-D rutinoside (21), 3′-methoxy-quercetin-3-O-(3-β-Dglucopyranosyl-2-rhamnopyranosil-4-glucopyranosyl-2-rhamnopyranosil)-glucoside (25) and 3′,4′dimethoxyquercetin-3-O-rhamnopyranoside (26). GC–MS analysis of the essential oil highlighted the occurrence of aliphatic compounds, mainly fatty acids, whereas labdane-type compounds were very scant. Our results showed that C. creticus subsp. eriocephalus has a different chemical profile with respect to the other subspecies due to the lack of labdane derivatives. On the other hand, this subspecies contains several phenolic constituents like ellagitannins, gallotannins and flavonoids, some of which can be of chemotaxonomic value.  相似文献

Variations in physiology and metabolic products of Cistus creticus subsp. eriocephalus along an altitudinal gradient (350–750 m.a.s.l.) within the Monti Lucretili Regional Natural Park (central Italy) were studied. The results showed that the phenol production was in relationship with the net photosynthetic rates and the chlorophyll content. In particular, the increasing caffeic acid (CA) content with altitude suggested its role in providing an additional photo‐protection mechanism, by its ability to consume photochemical reducing power and acting as an alternative C‐atom sink under high light conditions. The metabolic production was tested by high performance thin layer chromatography (HPTLC) fingerprint analysis, highlighting the potential of this technique in biologic studies.  相似文献

  • Helichrysum microphyllum subsp. tyrrhenicum (Asteraceae) is an endemic taxon of Sardinia and Corsica, where it grows at different altitudes. The objective of this study was to investigate the seed traits and germination behaviour of four Sardinian populations of this taxon located at different altitudes.
  • Seed traits were evaluated, and germination tests were carried out by incubating seeds at a range of constant (5–30 °C) and alternating (25/10 °C) temperatures. The dry after‐ripening (DAR) pre‐treatment was also applied by storing seed in dry conditions for 3 months at 25 °C. Seed traits and germination behaviour data were statistically analysed to identify if there was a correlation with altitude.
  • Differences in seed size, area and mass among populations were recorded, however, no relationship was found with altitude. High germination percentages were obtained in all populations, both in untreated and DAR seeds, and were positively affected by alternating temperatures. The final germination percentage and time required to reach 50% final germination (T50) showed no relationship with altitude.
  • The differences in seed traits and germination detected among the studied populations of H. microphyllum subsp. tyrrhenicum were not correlated with altitude. This study provides new and important knowledge for this taxon. H. microphyllum subsp. tyrrhenicum is characterised by high germination percentages and low T50 values and does not seem to require any dormancy‐breaking treatment. This species represents a high‐potential native plant species that should be considered within environmental management plans.

  • This work aims to study seeds of the endemic species Astragalus aquilanus from four different populations of central Italy. We investigated seed morpho‐colorimetric features (shape and size) and chemical differences (through infrared spectroscopy) among populations and between dark and light seeds.
  • Seed morpho‐colorimetric quantitative variables, describing shape, size and colour traits, were measured using image analysis techniques. Fourier transform infrared (FT‐IR) spectroscopy was used to attempt seed chemical characterisation. The measured data were analysed by step‐wise linear discriminant analysis (LDA). Moreover, we analysed the correlation between the four most important traits and six climatic variables extracted from WorldClim 2.0.
  • The LDA on seeds traits shows clear differentiation of the four populations, which can be attributed to different chemical composition, as confirmed by Wilk's lambda test (< 0.001). A strong correlation between morphometric traits and temperature (annual mean temperature, mean temperature of the warmest and coolest quarter), colorimetric traits and precipitation (annual precipitation, precipitation of wettest and driest quarter) was observed.
  • The characterisation of A. aquilanus seeds shows large intraspecific plasticity both in morpho‐colorimetric and chemical composition. These results confirm the strong relationship between the type of seed produced and the climatic variables.

  • Morphological and functional seed traits have important roles in characterising the species regeneration niche and help to understand the reproductive biology of rare and threatened plants, which can thus support appropriate plant conservation measures.
  • Seed morphometric and dispersal kinetics of the critically endangered Dioscorea strydomiana were measured and compared with those of four other Dioscorea species, and seed germination response under constant temperatures (5–35 °C) was compared with that of the congeneric and widespread D. sylvatica.
  • Seed mass of D. strydomiana (ca. 14 mg) was twice that of D. sylvatica, but similar to or smaller than the other species examined. Seeds of D. strydomiana have the lowest speed of descent and lowest variability in most of the morphological traits considered, suggesting lower phenotypic plasticity but higher variance in the wing‐loading value. Seeds of D. strydomiana reached maximum germination at 15 °C (ca. 47%), which decreased slightly to ca. 37% at 25 °C and was completely inhibited at 35 °C. D. sylvatica seeds started to germinate at 10 °C (ca. 3%), reached 75–80% germination at 15–20 °C and maximum (ca. 90%) at 25–30 °C. Base temperatures for germination (Tb) were 9.3 and 5.7 °C, for D. strydomiana and D. sylvatica, respectively. Due to the higher germination percentages of D. sylvatica, ceiling and optimum temperatures could also be modelled for this species, suggesting higher sensitivity to high temperature for seeds of D. strydomiana.
  • The detected poor seed lot quality of D. strydomiana suggests difficulties in reproduction from seed, highlighting the need for further investigation and conservation actions for this threatened yam species.

  • Citrus canker is a worldwide‐distributed disease caused by Xanthomonas citri subsp. citri. One of the most used strategies to control the disease is centred on copper‐based compounds that cause environmental problems. Therefore, it is of interest to develop new strategies to manage the disease. Previously, we reported the ability of the siderophore pyochelin, produced by the opportunistic human pathogen Pseudomonas aeruginosa, to inhibit in vitro several bacterial species, including X. citri subsp. citri. The action mechanism, addressed with the model bacterium Escherichia coli, was connected to the generation of reactive oxygen species (ROS). This work aimed to find a non‐pathogenic strain from the lemon phyllosphere that would produce pyochelin and therefore serve in canker biocontrol.
  • An isolate that retained its capacity to colonise the lemon phyllosphere and inhibit X. citri subsp. citri was selected and characterised as Pseudomonas protegens CS1. From a liquid culture of this strain, the active compound was purified and identified as the pyochelin enantiomer, enantio‐pyochelin.
  • Using the producing strain and the pure compound, both in vitro and in vivo, we determined that the action mechanism of X. citri subsp. citri inhibition also involved the generation of ROS. Finally, the potential application of P. protegens CS1 was evaluated by spraying the bacterium in a model that mimics the natural X. citri subsp. citri infection.
  • The ability of P. protegens CS1 to reduce canker formation makes this strain an interesting candidate as a biocontrol agent.

In this study phytochemical compounds and antioxidant capacity, cytotoxic, antimicrobial and anti-biofilm activities of hydroethanolic extracts of five Cistus species (C. creticus L., C. laurifolius L., C. monspeliensis L., C. parviflorus Lam. and C. salviifolius L.) distributed in Turkey were investigated. (+)-catechin, epigallocatechin gallate, quercetin-3-O-rutinoside, quercetin-3-O-glucoside, kaempferol-3-O-glucoside, luteolin were detected in different amounts. Strongest antioxidant capacities were observed with C. creticus, and C. parvifolius (0.476 and 0.452, respectively). Minimum inhibitory concentrations (MIC) of the extracts were determined between 32 and 128 μg/mL against different bacteria and Candida strains. C. monspeliensis and C. laurifolius extracts were inhibited the biofilm production levels of three Gram-negative bacteria (E. coli, S. enterica, P. aeruginosa), two Gram-positive bacteria (S. aureus, B. subtilis) and three Candida strains (C. albicans, C. parapsilosis, C. krusei). C. creticus extract showed strongest cytotoxic activity against human breast adenocarcinoma (MCF-7) and prostate cell lines (PC-3) (IC50: 14.04±2.78 μg/mL and 34.04±2.74 μg/mL, respectively) among all plants tested.  相似文献

  • The relationship between the phytohormones, gibberellin (GA) and abscisic acid (ABA) and light and temperature on seed germination is still not well understood. We aimed to investigate the role of the ABA and GA on seed germination of Vellozia caruncularis, V. intermedia and V. alutacea in response to light/dark conditions on different temperature.
  • Seeds were incubated in GA (GA3 or GA4) or ABA and their respective biosynthesis inhibitors (paclobutrazol – PAC, and fluridone – FLU) solutions at two contrasting temperatures (25 and 40 °C). Furthermore, endogenous concentrations of active GAs and those of ABA were measured in seeds of V. intermedia and V. alutacea during imbibition/germination.
  • Exogenous ABA inhibited the germination of Vellozia species under all conditions tested. GA, FLU and FLU + GA3 stimulated germination in the dark at 25 °C (GA4 being more effective than GA3). PAC reduced seed germination in V. caruncularis and V. alutacea, but did not affect germination of V. intermedia at 40 °C either under light or dark conditions. During imbibition in the dark, levels of active GAs decreased in the seeds of V. intermedia, but were not altered in those of V. alutacea. Incubation at 40 °C decreased ABA levels during imbibition in both V. caruncularis and V. alutacea.
  • We conclude that the seeds of Vellozia species studied here require light or high temperature to germinate and ABA has a major role in the regulation of Vellozia seed germination in response to light and temperature.

This study aims to explore the effect of environmental factors (temperature, light, storage time) on germination response and dormancy patterns in eight Mediterranean native wildplants, belonging to the Euphorbia L. genus. In detail, we considered E. amygdaloides subsp. arbuscula, E. bivonae subsp. bivonae, E. ceratocarpa, E. characias, E. dendroides, E. melapetala, E. myrsinites, and E. rigida. We collected seeds from natural plant populations and performed germination assays in climatic chambers at seven constant temperatures (from 5 to 35°C, with 5°C increments), and four fluctuating temperature regimes (8/15, 8/20, 8/25, and 8/30°C, with a 12/12 hr thermoperiod). Germination assays were set up both in dark (D) and in light/dark conditions (L/D, 12/12 hr photoperiod), after short and long seed storage (SS around 30 days and LS around 150 days). For all these species, except E. amygdaloides subsp. arbuscula, results show that the final germinated proportions were improved by a long storage period (>150 days), which supports the existence of nondeep physiological dormancy. Optimal temperature levels ranged from 14.3 to 21.3°C and base temperatures ranged from 5.6 to 12.1°C, while ceiling temperatures from 25.6 to 34.7°C. For none of these species, germinations were favored by an alternating daily temperature regime, while in several instances, germinations were quicker and more complete in darkness, than in an alternating light/dark regime. In some instances, extreme temperature levels (5 and 30°C) induced dormancy and germinations did not resume when seeds were exposed at optimal temperature levels. Results are discussed in terms of the dynamics of emergences and how this might be affected by climate changes.  相似文献

  • Successful germination and seedling emergence in new environments are crucial first steps in the life history of global plant invaders and thus play a key role in processes of range expansion.
  • We examined the germination and seedling emergence success of three global plant invaders – Lupinus polyphyllus, Senecio inaequidens and Verbascum thapsus – in greenhouses and climate chambers under climate regimes corresponding to seven eco‐regions. Seed materials were collected from one non‐native population for L. polyphyllus and S. inaequidens, and from 12 populations for V. thapsus (six natives and six non‐natives).
  • Experimental climates had significant effects on species responses. No species germinated in the dry (humidity ≤ 50%) and cool (≤ 5 °C) experimental climates. But all species germinated and emerged in two moderately cool (12–19 °C) and in three warm (24–27 °C) experimental climates. In general, V. thapsus showed higher fitness than S. inaequidens and L. polyphyllus. The climate of the seed source region influenced responses of native and non‐native populations of V. thapsus. Non‐native populations of V. thapsus, originating from the warmer seed source, showed higher performance in warm experimental climates and lower performance in moderately cool experimental climates compared to native populations. Responses of V. thapsus populations were also related to precipitation of the seed source region in moderately dry experimental climates.
  • The warm, semi‐arid and humid experimental climates are suitable for the crucial first steps of invasion success for L. polyphyllus, S. inaequidens and V. thapsus. The species adaptation to its source region modified the responses of our studied plants under different experimental climates representing major eco‐regions of the world.

  • Dormancy cycles are an important mechanism for avoiding seed germination under unfavourable periods for seedling establishment. This mechanism has been scarcely studied in tropical species. Here, we studied three tropical and perennial species of Xyris, X. asperula, X. subsetigera and X. trachyphylla, to investigate in situ longevity and the existence of seasonal seed dormancy cycles.
  • Seeds of three species of Xyris were buried in their natural habitat, with samples exhumed bimonthly for 18 months. Germination of exhumed seeds was assessed under a 12‐h photoperiod over a broad range of temperatures. Seeds of X. trachyphylla were also subjected to treatments to overcome secondary dormancy.
  • Seeds of all species are able to form a persistent seed bank and exhibit seasonal changes in germinability. Secondary dormancy was acquired during the rainy summer and was overcome during the subsequent dry season (autumn/winter). Desiccation partially overcomes secondary dormancy in X. trachyphylla seeds.
  • Soil seed bank persistence and synchronisation of seed germination under favourable conditions for seedling establishment contribute to the persistence and regeneration of X. asperula, X. subsetigera and X. trachyphylla in their natural environment.

  • Climate change will alter the biotic and abiotic environment and dissipate ecological barriers, reorganising maps of current distribution of parasites and their hosts. In this study, we analyse the population dynamics of the parasitic plant Viscum album subsp. austriacum and explore key biotic (host availability and seed dispersal) as well as abiotic (temperature) factors influencing elevational distribution.
  • The study was conducted along an elevational gradient of a Mediterranean mountain, covering the distribution belts of three potential pine hosts: Pinus halepensis (1300–1500 m), P. nigra (1300–1900 m) and P. sylvestris var. nevadensis (1600–2000 m). Along this gradient, we measured multiple variables of mistletoe population (prevalence, abundance and demographic profile) and different factors that might define the current mistletoe distribution (host suitability and availability, temperature and seed dispersal services).
  • We found a decline in mistletoe prevalence and abundance with increasing elevation, detecting larger values of both variables at lower elevations of the most suitable host (Pinus nigra). Pinus sylvestris var. nevadensis was a suboptimal but suitable host for the parasite at high elevations. Mistletoe found suitable temperatures and seed dispersal services all along the gradient, being able to recruit at any site.
  • With warming temperatures, the presence of suitable vectors for parasite dispersion, and the presence of a sub‐optimal host (P. sylvestris var. nevadensis) at the mountain top, mistletoe currently has a window of opportunity to expand its present geographic distribution to the summits.

  • Seed germination of Citrullus colocynthis, as in many other species of Cucurbitaceae, is inhibited by light, particularly at low temperatures. Germination response to light and temperature has been attributed to day length and temperature during seed maturation. This study assessed the effects of these factors on the germination response of C. colocynthis to temperature and light quality.
  • Ripe fruits were collected from natural habitats during December and February and germinated at three temperatures (15/25, 20/30 and 25/35 °C) in five light treatments (dark, white light and Red:Far Red (R:FR) ratios of 0.30, 0.87 and 1.19). Additionally, unripe fruits were also collected from natural habitats and completed their maturation in growth chambers under different day lengths (6, 16 and 24 h of darkness) at 10/20 °C, and in darkness at both 10/20 °C and 25/35 °C. Mature seeds of the different treatments were germinated in the same five light treatments at 15/25 °C.
  • Germination was significantly higher in the dark than that in any light treatment. Seeds matured at higher temperatures (i.e. seeds from the December collection and those matured at 25/35 °C) had significantly higher germination than those matured at lower temperatures (i.e. seeds from the February collection and those matured at 10/20 °C). Dark germination was significantly higher for the December collection than for the February collection. Seeds of the two collections germinated in the dark only at 15/25 °C. However, seeds matured in a growth chamber at 10/20 °C in darkness germinated at 15/25 °C in all light treatments, except for the R:FR ratio 0.30. Seeds of the different treatments failed to germinate in FR‐rich light.
  • This study demonstrates that both temperature and day length during seed maturation play significant roles in the germination response of C. colocynthis. Additionally, the dark requirement for germination is likely beneficial for species with the larger seeds, such as C. colocynthis, which produce bigger seedlings that are able to emerge from deep soils and are competitively superior under dense vegetation and resource‐limited conditions.

  • Anogeissus leiocarpa (DC.) Guill. & Perr. (Combretaceae) has important economic and cultural value in West Africa as source of wood, dye and medicine. Although this tree is in high demand by local communities, its planting remains limited due to its very low propagation via seed.
  • In this study, X‐rays were used to select filled fruits in order to characterise their morphology and seed germination responses to treatment with sulphuric acid and different incubation temperatures.
  • Morphological observations highlighted a straight orthotropous seed structure. The increase in mass detected for both intact and scarified fruits through imbibition tests, as well as morphological observations of fruits soaked in methylene blue solution, confirmed that they are water‐permeable, although acid‐scarified fruits reached significantly higher mass increment values than intact ones. Acid scarification (10 min soaking in 98% H2SO4) positively affected seed germination rate but not final germination proportions. When intact fruits where incubated at a range of temperatures, no seeds germinated at 10 °C, while maximum seed germination (ca. 80%) was reached at 20 °C. T50 values ranged from a minimum of ca. 12 days at 25 °C to a maximum of ca. 34 days at 15 and 35 °C. A theoretical base temperature for germination (Tb) of ca. 10 °C and a thermal requirement for 50% germination (S) of ca. 195 °Cd were also identified for intact fruits.
  • The results of this study revealed the seed germination characteristics driven by fruit and seed morphology of this species, which will help in its wider propagation in plantations.
