Ten polymorphic microsatellite loci were isolated and characterized for the ferruginous pygmy owl (Glaucidium brasilianum). Observed and expected heterozygosity were 0.07–0.85 and 0.1–0.88, respectively. Allele numbers ranged from five to 16 per locus.  相似文献

Sequences from the mitochondrial cytochrome b gene were used to examine patterns of variation within and among populations of the ferruginous pygmy-owl (Glaucidium brasilianum) from both North America (including populations from Mexico) and South America. As currently conceived, G. brasilianum is paraphyletic, with North American and South American clades representing two distinct groups that should be recognized as the distinct species G. ridgwayi and G. brasilianum, respectively. Within the G. ridgwayi clade, populations from Arizona, Sonora, and Sinaloa are genetically distinct and share no mitochondrial haplotypes with populations occurring in Texas and other regions of Mexico. According to nested clade analysis this separation may be the consequence of past fragmentation that predates the origin of the Sonora desert. In addition, gene flow between the Arizona/Sonora/Sinaloa populations and the Texas/other Mexico populations is practically nonexistent, with estimates being approximately one individual every 10 generations. Collectively, these data suggest that the Arizona/Sonora/Sinaloa clade should be recognized as either a distinct subspecies or phylospecies from the group containing populations in Texas and the remainder of Mexico. These data should be used as guidelines for pygmy-owl recovery and conservation, as they meet the recommendations of task 3 of the pygmy-owl recovery plan that lists genetic data as essential information for pygmy-owl management.  相似文献

Eleven polymorphic microsatellite loci were used to assess genetic variation in the ferruginous pygmy owl (Glaucidium brasilianum) from North America. Analysis of genotypic variation suggests restricted gene flow between pygmy-owl populations in Arizona-Sonora and Sinaloa, and Texas and the remaining states in Mexico. The Arizona-Sonora population showed signs of a recent genetic bottleneck, an observation supported by low population estimates for Arizona (13–117 individuals). Heterozygosity in Arizona, however, was equal to levels recorded throughout Mexico and Texas. Congruent patterns revealed by nuclear (microsatellites) and mitochondrial DNA that indicate Arizona-Sonora and Texas populations are distinct from adjacent populations in Mexico, thus emphasizing need for the design and implementation of separate management plans for recovery and conservation efforts. Revealing evidence of distinct groups within the pygmy-owl populations in North America, results from this study may be used to make management decisions for the recovery and conservation of this species.  相似文献

The production of a battery of arabinoxylan-degrading enzymes by the fungus Penicillium brasilianum grown on brewer’s spent grain (BSG) under solid-state fermentation was investigated. Initial moisture content, initial pH, temperature, and nitrogen source content were optimized to achieve maximum production of feruloyl esterase, xylanase, and α-l-arabinofuranosidase. Under the optimum growth conditions (80% moisture, pH 6, 26.5°C, and 5 g/l nitrogen source), the maximum level of feruloyl esterase (1,542 mU/g BSG) was found after 196 h, whereas xylanase (709 U/g BSG) and ArabF activity (3,567 mU/g BSG) were maximal after 108 h and 96 h, respectively. Based on substrate utilization data, the feruloyl esterases produced by P. brasilianum was anticipated to subclass B. A crude enzyme (CE) preparation from P. brasilianum culture grown on BSG was tested for the release of hydroxycinnamic acids and pentoses from BSG. The P. brasilianum CE produced in this work contains a balance of cell wall-modifying enzymes capable of degrading arabinoxylan of BSG by more than 40%.  相似文献

Conspecific broadcasts are effective to increase detection of owls. To determine the most appropriate time of the year to survey owls, we played conspecific owl vocalisations monthly in a temperate rainforest of southern Chile. From 12 broadcast points surveyed we recorded detections of Glaucidium nana, Strix rufipes and Tyto alba. Glaucidium nana presented a bimodal detection curve throughout the year and we recorded two regular vocalisations in response to broadcasting: contact pair call and territorial call. Strix rufipes and T. alba both showed a peak of detection between February and May. Strix rufipes presented three vocalisations: territorial call, contact pair call and female contact pair call while T. alba uttered two vocalisations: territorial call and twittering call. We recommend surveys during the end of the breeding season (austral summer–autumn) when detection is higher in most owls. Surveys should also take into consideration the variability of the vocalisations and include covariates in monitoring to evaluate occupancy/detection models.  相似文献

This study investigated Plasmodium spp. infection in free-ranging neotropical primates from Brazilian Amazon regions under the impact of major anthropogenic actions. Blood samples from 19 new world primates were collected and analyzed with microscopic and molecular procedures. The prevalence of Plasmodium infection was 21.0% (4/19) and PCR positive samples were identified as P. brasilianum. Considering the social-economic changes that the Amazon is facing, the prevalence of P. brasilianum infection highlights the necessity to closely monitor the movement of both human and non-human primate populations, in order to mitigate pathogen exposure and the introduction of new agents into previously naïve areas.  相似文献

Plasmodium malariae mainly causes asymptomatic submicroscopic parasitemia in the endemic Amazon and non-endemic Atlantic Forest, where the number of cases and transmission of malaria through blood transfusion has increased. This study developed a P. malariae/P. brasilianum Real Time PCR (rtPCR) targeting the cytochrome b oxidase (cytb), a highly repetitive gene (20-150 copies/parasite) that should detect more cases than the 18S rRNA (4-8 copies/parasite) gene-based amplification systems. Cytb from human and non-human Plasmodium species (including P. brasilianum) aligned to the only 20 African P. malariae cytb sequences identified polymorphic regions within which we designed P. malariae species-specific primers. Non-human Plasmodium species, related parasites, anemia-causing microorganisms, normal human DNA and 47 blood bank donors samples that were truly negative to malaria accessed rtPCR specificity. Truly positive samples (n = 101) with species identification by semi-nested, nested or TaqMan PCR, and four samples from the Atlantic Forest that were suspected of malaria but three of them had negative genus TaqMan and 18S rRNA nested PCR. The cloned amplification product used in standard curves determined qPCR detection limit (0.5-1 parasite equivalent/μL). The 10 positive P. malariae samples among truly positives yielded positive rtPCR results and more importantly, rtPCR detected the four samples suspected of malaria from the Atlantic Forest. The rtPCR specificity was 100%, reproducibility 11.1% and repeatability 6.7%. In conclusion, the proposed rtPCR is fast, apparently more sensitive than all 18S rRNA amplification systems for detecting extremely low parasitemia. The rtPCR is also specific to P. malariae/P. brasilianum species. This new molecular tool could be applied to the detection of P. malariae/brasilianum infections with submicroscopic parasitemias in the context of epidemiological studies and blood bank safety programs.  相似文献

In the present paper, based on material from Brazil, we present a contribution to the taxonomy of the Neotropical two-winged Leptophlebiidae. The male imago of Perissophlebiodes Savage is described, in addition to a new species of Askola Peters from Amazonas. Askola emmerichi Domínguez, Molineri and Mariano, so far recorded from Venezuela, is reported from Brazil and compared to the new species herein described. Askola yanoman, sp. nov. can be separated from other species of the genus by the following combination of characters: (1) general colouration yellowish brown with areas washed with greyish brown; (2) dorsal portion of eyes brown, eyes meet on meson of head; (3) penis yellowish brown with darker margin, extending beyond the posterior margin of the styliger plate.  相似文献

In this study, the consumption of 4-bromobenzoic acid and 4-chlorobenzoic acid by the fungus Penicillium brasilianum, an endophyte from Melia azedarach is evaluated. This fungus metabolizes these halobenzoic acids to produce three new brominated compounds, which have been isolated and characterized, and three new chlorinated derivatives identified by HRMS. Among these products, (4-bromobenzoyl)proline has been also chemically synthesized and employed in biological assays, thus providing insights for the elucidation of the defense mechanism of P. brasilianum towards these halobenzoic acids.  相似文献

Zamia grijalvensis, a new cycad species from Chiapas, México, is described and illustrated. This species is similar to Z. lacandona with respect to leaves, but differs in habit, morphology and colour of the female and male cones. The female cone of Z. grijalvensis is reddish brown to orange brown, whereas that of Z. lacandona is dark brown with lighter beige tomentum. The male cone of Z. grijalvensis is erect, whereas that of Z. lacandona is decumbent. The new species may be of natural hybrid origin and is discussed in the light of other findings among cycads. Preliminary diploid chromosome counts for Z. grijalvensis 2n = 19, 20 and karyotypes appear variable, as previously found in its congeners with wide distribution, such as Z. paucijuga and Z. prasina, both of which are characterised by high diploid numbers with a high number of telocentric and few metacentric chromosomes. These changes in chromosome number are probably the result of stressful environmental factors.  相似文献

A new spectrophotometric procedure was developed for the quantitative determination of fucoidan and alginic acid and used for their analysis in extracts from biomass of 17 species of brown algae collected from coastal waters of the Kamchatka peninsula. In addition, neutral monosaccharides and mannitol were determined in the hydrolysis products of the alga biomass samples. The polysaccharide composition was shown to substantially depend on the algal species. The alginic acid content was maximal in the Alaria marginatablades; all the other representatives of the order Laminariales also are useful sources of this polysaccharide. At the same time, the fucoidan content is rather low in Laminariales. The highest content of fucoidan was found in Saundersella simplex, but Chordaria flagelliformisand Fucus evanescensare more practical fucoidan sources; the available supplies and the sugar composition make the latter alga the most suitable for the complex processing to prepare mannitol, fucoidan, and alginic acid.  相似文献

A new species ofPhaeosphaeria is described from mangroves in South Africa. InPhaeosphaeria capensis ascospores are 29–36×7–8 μm, cylindric-fusiform or somewhat clavate, widest at the middle, narrow at the base and rounded at the apex, (6-)7-septate, light brown or brown, minutely verruculose and surrounded by a spreading, mucilaginous sheath. Pseudomonas tolaasii, causing brown blotch disease on cultivated mushrooms, and yielding a white line precipitate towards P. “reactans”, has been shown to induce lysis of erythrocytes. Some Finnish strains isolated from diseased mushroom fruit bodies, although harboring the typical features of P. tolaasii, proved to be distinct, and have been allocated to a nov. sp. P. costantinii. We examined in these study whether all brown blotch causing agents were hemolytic. The induction of erythrocytes lysis seemed to be a rather common feature of mushroom associated-pseudomonads, especially for strains involved in the production of a white-line-in agar.  相似文献

Embryological features ofGlaucidium palmatum are as follows: the ovule is anatropous and bitegmic; the archesporium is hypodermal and multicelled, consisting of about 10 to 15 cells; all the archesporial, cells develop directly into megaspore mother cells, only three or four of which, however, generally complete meiotic divisions; before and during meiosis, dermal cells of the nucellar apical part undergo successive periclinal divisions forming a thick nucellar cap of as many as 20 cell-layers; embryo sac formation is of the Polygonum type; multiple embryo sacs occur frequently; antipodal cells are small in size and ephemeral or persistent; the inner integument is 3 to 5 cell-layers thick, and the outer integument 7 to 13 cell-layers thick; the outer integument is vascularized; a micropyle is formed by the inner integument alone; the endosperm is of the Nuclear type; embryogeny is of a type similar to the Onagrad type. In light of evidence from embryology and other sources it seems that there is ample reason for recognizing the family Glaucidiaceae which is distinct from the Ranunculaceae and its related families. Several common embryological features suggest an affinity between the Glaucidiaceae and the Paeoniaceae.  相似文献

Abstract

Red date palm weevil Rhynchophorus ferrugineus Oliv. is a widespread major pest of date palm in the Kingdom of Saudi Arabia. Three different forms (black and brown with and without spots on thoracic region) were investigated using PCR-based RAPD technique. Although weevils were collected from the same geographical region of Al-Hassa in Saudi Arabia, the banding profile acquired suggested that black and brown colored morphs are genetically closer compared to the brown with spots. Intra color variation remained minimum in black but brown and brown spotted morphs exhibited more genetic variation. This genetic variation may be either due to the generation of new mutants from the non-spotted or spotted weevil or they may belong to a different race.  相似文献

 A new species and a new variety of Cortinarius sect. Defibulati are described and illustrated from Niigata, Japan. Cortinarius rugosolilacinus sp.nov., found in deciduous forests, is most similar to Cortinarius livido-ochraceus (=C. elatior), but differs mainly in its longer basidiospores, slender basidiocarp, and pale violet lamellae in the button stage. Cortinarius pseudosalor var. niigatensis var. nov., found in deciduous forests, is distinguished from the type variety by its brown to dark brown pileus with a wrinkled surface even when young. Individuals ofPresbytis johnii with coats of brown hair, rather than of the normal black color, are reported from a population inhabiting forest fragments at a low elevation in the Western Ghats of South India. It is suggested that this coat color aberration is due to a relatively rare recessive allele, occurring homozygously as a result of inbreeding in small, isolated populations. Because similar color aberrations in other colobine monkeys seem most frequent in isolated populations, they too may be the result of founder effects rather than recent mutation.  相似文献

Spatial variation in resources is a fundamental driver of habitat quality but the realized value of resources at any point in space may depend on the effects of conspecifics and stochastic factors, such as weather, which vary through time. We evaluated the relative and combined effects of habitat resources, weather, and conspecifics on habitat quality for ferruginous pygmy-owls (Glaucidium brasilianum) in the Sonoran Desert of northwest Mexico by monitoring reproductive output and conspecific abundance over 10 years in and around 107 territory patches. Variation in reproductive output was much greater across space than time, and although habitat resources explained a much greater proportion of that variation (0.70) than weather (0.17) or conspecifics (0.13), evidence for interactions among each of these components of the environment was strong. Relative to habitat that was persistently low in quality, high-quality habitat buffered the negative effects of conspecifics and amplified the benefits of favorable weather, but did not buffer the disadvantages of harsh weather. Moreover, the positive effects of favorable weather at low conspecific densities were offset by intraspecific competition at high densities. Although realized habitat quality declined with increasing conspecific density suggesting interference mechanisms associated with an Ideal Free Distribution, broad spatial heterogeneity in habitat quality persisted. Factors linked to food resources had positive effects on reproductive output but only where nest cavities were sufficiently abundant to mitigate the negative effects of heterospecific enemies. Annual precipitation and brooding-season temperature had strong multiplicative effects on reproductive output, which declined at increasing rates as drought and temperature increased, reflecting conditions predicted to become more frequent with climate change. Because the collective environment influences habitat quality in complex ways, integrated approaches that consider habitat resources, stochastic factors, and conspecifics are necessary to accurately assess habitat quality.  相似文献
