The incorporation of 14C-galactose into primary AGMK-cells was studied in the presence and absence of Mn2+. The transport of galactose into the cells is not influenced by Mn2+. 1 mM MnCl2 inhibits the incorporation of galactose into acid-precipitable material up to 50% after 6 hours incubation. In the absence of Mn2+ a substantial amount of galactose is converted to glucose, which is mainly metabolized into aspartic acid and serine. The conversion of galactose into glucose is inhibited by the addition of Mn2+. However, Mn2+ does not influence the activity of the UDP-galactose-4'-epimerase in vitro. Using the SDS-polyacrylamide electrophoresis the labelling of protein bands is similar with 14C-galactose or a 14C-amino acid mixture, respectively. In the presence of Mn2+ the incorporation of both galactose or amino acids is inhibited: With amino acids the inhibition is observed in all protein bands, whereas with galactose some bands remain unaffected. It is concluded that these are galactoproteins.  相似文献

Role of excitatory amino acids in hypoxic preconditioning.   总被引:4,自引:0,他引:4  
We examined the effects of the extrinsic ionotropic NMDA receptor agonist (aspartate) and antagonist (ketamine) on the hypoxic preconditioning of mice and the concentration changes of intrinsic excitatory amino acids (EAAs), aspartate and glutamate, in the whole brain and different brain regions during preconditioning by an HPLC method. Our results showed that aspartate and ketamine significantly prolonged and shortened the standard tolerance time of mice during preconditioning and survival time in hypobaric chambers, respectively. After the 1st exposure, EAA concentrations in the whole brain and brain regions were increased. After run 4, they were decreased or maintained. It is suggested that the activation and suppression of ionotropic NMDA receptors is harmful and beneficial to hypoxic preconditioning, respectively. Degradation and/or inactivation of EAAs might be beneficial to the tolerance of mice to hypoxia.  相似文献

The effect of 20 essential and nonessential L-amino acids on the dynamics of development of spleen explants from 1- and 21-day-old rats on an organotypic tissue culture was studied. The hydrophilic amino acids with a higher molecular mass (asparagine, lysine, arginine, and glutamic acid) induced an inhibitory effect on the growth zone of explants of immature tissue from 1-day-old animals and an opposite, stimulating effect on the mature spleen tissue of 21-day-old rats. An immunohistochemical analysis revealed a reciprocal correlation between the expression of the proapoptotic protein p53 and the cell proliferation upon the action of lysine, asparagine, and glutamic acid. The role of polar amino acids in the modulation of cell proliferation and apoptosis in dependence on the period of ontogenesis was determined.
