The relative requirement of N and P (the optimum N:P ratio)by Dunaliella tertiolecta, Phaeodactylum tricornutum, Prymnesiumparvum and Thalassiosira pseudonana was studied under variouslight intensities and spectra. The ratio was determined as theratio of the minimum cell N and P concentrations (q0N and q0pwhen either nutrient was limiting. The ratio varied widely amongspecies; under light-saturation for growth (116 µEin m–2s–1 it ranged from 11.8 in D. tertiolecta to 36.6 in P.tricornutum. The ratio appeared to be higher at a sub-saturatingintensity (24 µEin m–2 s–1 in all except P.tricornutum, mainly because of higher qoN with little changein qoP. In T. pseudonana QoP also increased, resulting in aninsignificant change in the ratio. The ratio varied little withinthe range of saturation intensity. Light quality affected qoNand qoP as well as the ratio, and the pattern of change variedfrom species to species. The optimum ratio of individual specieswas linearly correlated to their qoN except in P. tricornutum.qoN for all species showed a linear correlation with cell proteinconcentrations irrespective of light conditions. The changeof optimum N:P ratios in the three species thus appears to berelated to changes in cell protein contents. The ratio of carbohydratesto protein remained constant regardless of light intensity orquality and was higher in P-limited cultures. We conclude thatchanges in light regime can strongly influence algal nutrientrequirements and species interrelationships by altering theoptimum cellular N:P ratio.  相似文献

Cultures of Dunaliella tertiolecta were exposed to five concentrationsof cadmium in solution (0.1, 0.5, 1, 5 and 10ppm(mg 1-1)). Theaccumulation of cadmium by the algae was found to have two phases,an initial rapid uptake followed by a stabilisation of the cellularcadmium levels. D. tertiolecta concentrated cadmium from solution(cone, factor approx. 1350) at exposures up to 1 ppm Cd butexposure to the higher concentrations caused no further increasein the accumulated cadmium concentration of the algae whichreached a nmriimim at about 1.5 µg Cd mg1 Dunaliella.  相似文献

In a series of batch experiments in the dark the heterotrophicdinoflagellate Oxyrrhis marina grazed three phytoplankton prey(Phaeodactylun tricornutum, Isochrysis galbana and Dunaliellateriolecta) with equal efficiency. Growth rates of the dinoflagellateranged between 0.8 and 1.3 day–1 Maximum observed ingestionrates on a cell basis varied according to the size of the preyfrom about 50 cells flagellate–1 day–1 when D.tertiolectawas the prey to 250–350 cells fiagellate–1 day–1when the other species were eaten. However, when compared ona nitrogen basis, ingestion rates were independent of prey type.Both ingestion and growth ceased when prey cell concentrationsfell below a threshold concentration of about 105 cells ml–1.Maximum specific clearance rates were 0.8x1040ndash;5.7x104it day which is considerably lower than that found for heterotrophicdinoflagellates in oceanic waters and may explain why O.marinagenerally thrives only in productive waters. The timing of NHregeneration was linked to the C:N ratio of the prey at thestart of grazing. Regeneration efficiencies for NH4. never exceeded7%; during the exponential phase and were 45% well into thestationary phase. These results are comparable to those obtainedwith heterotrophic flagellates and demonstrate that the bioenergeticpatterns of grazing and nutrient cycling by different protozoaare very similar. Moreover, they support the notion that toachieve 90+% nutrient regeneration in the open ocean, as iscurrently believed, the microbial food loop must consist ofmultiple feeding steps. Cells of Dunaliella tertiolecta which had been grown in ordinaryair (low-CO2 cells) had high carbonic anhydrase (CA) activityon the cell surface and mainly utilized HCO3 for photosynthesis.When CA activity on the cell surface was inhibited by Diamoxor subtilisin, the cells utilized CO2. When bovine CA was added,the subtilisin-treated low-CO2 cells utilized mainly HCO3.When grown in air containing 2% CO2, the cells had low CA activityon the cell surface, and preferred CO2 to HCO3. Kineticanalysis of these results indicated that low-CO2 cells of D.tertiolecta absorb CO2 which was converted from HCO3via the CA located on the cell surface. Three marine phytoplankton species (Skeletonema costatum, Olisthodiscusluteus andGonyaulax tamarensis) were grown in batch culturesat 15°C and a 14:10 L:D cycle at irradiance levels rangingfrom 5 to 450 µEinst m–2 s–1. At each irradiance,during exponential growth, concurrent measurements were madeof cell division, carbon-specific growth rate, photosyntheticperformance (both O2 and POC production), dark respiration,and cellular composition in terms of C, N and chlorophyll a.The results indicate that the three species were similar withrespect to chemical composition, C:N (atomic) = 6.9 ±0.4, photo-synthetic quotient, 1.43 ± 0.09, and photosyntheticefficiency, 2.3 ±0.1 x 10–3 µmol O2 (µgChl a)–1 h–1 (µEinst m–2 s–1)–1.Differences in maximum growth rate varied as the –0.24power of cell carbon. Differences in growth efficiency, werebest explained by a power function of Chl a:C at µ = 0.Compensation intensities, ranged from 1.1 µEinst m–2s–1 for S. costatum to 35 forG. tamarensis and were foundto be a linear function of the maintenance respiration rate.The results indicate that interspecific differences in the µ–Irelationship can be adequately explained in terms of just threeparameters: cell carbon at maximum growth rate, the C:Chl aratio (at the limit as growth approaches zero) and the respirationrate at zero growth rate. Loliun perenne L. (cv.S. 23) was grown on vermiculite in winterin a heated greenhouse for 8 weeks under factorial combinationsof two potassium regimes (nominally 6 parts/106 and 156 parts/106in Hewitt's solution) and three densities of artificially supplementedvisible radiation flux (36.1, 7.3, and 2.2 W m–2). Growthand potassium uptake were studied through the calculation ofvarious growth functions from fitted curves. There was little effect of potassium treatment but the experimentalmaterial responded markedly to light. Leaf-area ratio in thethree treatments showed extreme plasticity in increasing from2–3 x 10–2 through 6 x 10–2 to 8–9 x10–2 m2 g–1 as light intensity decreased. Correspondingdecreases in unit leaf rate, however, caused over-all reductionsin relative growth rate. Specific absorption rates for potassium (AK, dry-weight basis)were strongly reduced at the lower light intensities but alsodisplayed complex ontogenetic drifts. Values of the allometricconstant, k (the ratio of root and shoot relative growth rates),decreased from c. 0.7 at 36.1 W m–2 through c. 0.3 at7.3 W m–2 to a value not significantly different fromzero (P < 0.05) at 2.2 W m–2. In material grown under the two higher light intensities a constantinverse relationship was found between the mass ratio of rootand shoot and the corresponding activity ratio. The resultsconform to this model: Mass ratio = –0.001+45.0 (1/activityratio) where activity ratio is expressed as specific absorptionrate for potassium (in µg g root–1 h–1)/unitshoot rate (rate of increase of whole-plant dry weight per unitshoot dry weight, in mg g shoot–1 h–1). In situ growth and development of Neocalanus flemingeri/plumchrusstage C1–C4 copepodites were estimated by both the artificial-cohortand the single-stage incubation methods in March, April andMay of 2001–2005 at 5–6°C. Results from thesetwo methods were comparable and consistent. In the field, C1–C4stage durations ranged from 7 to >100 days, dependent ontemperature and chlorophyll a (Chl a) concentration. Averagestage durations were 12.4–14.1 days, yielding an averageof 56 days to reach C5, but under optimal conditions stage durationswere closer to 10 days, shortening the time to reach C5 (fromC1) to 46 days. Generally, growth rates decreased with increasingstage, ranging from 0.28 day–1 to close to zero but weretypically between 0.20 and 0.05 day–1, averaging 0.110± 0.006 day–1 (mean ± SE) for single-stageand 0.107 ± 0.005 day–1 (mean ± SE) forartificial-cohort methods. Growth was well described by equationsof Michaelis–Menten form, with maximum growth rates (Gmax)of 0.17–0.18 day–1 and half saturation Chl a concentrations(Kchl) of 0.45–0.46 mg m–3 for combined C1–3,while Gmax dropped to 0.08–0.09 day–1 but Kchl remainedat 0.38–0.93 mg m–3 for C4. In this study, in situgrowth of N. flemingeri/plumchrus was frequently food limitedto some degree, particularly during March. A comparison withglobal models of copepod growth rates suggests that these modelsstill require considerable refinement. We suggest that the artificial-cohortmethod is the most practical approach to generating the multispeciesdata required to address these deficiencies.  相似文献

Grazing and ingestion rates of laboratory-born Thalia democraticaaggregates and Dolioletta gegenbauri gonozooids, phorozooidsand oozooids were determined while fed Isochrysis galbana (4–5µm diameter) alone or in combination with Peridinium trochoideum(16–18 µm diameter) at concentrations of 0.15–0.70mm3 x 1–1. Grazing rates (ml x zooid–1 x 24 h –1)ranged from 10 to 355, and at zooid weights greater than 5 µgcarbon were in order oozooid > gonozooid > aggregate.Grazing rates increased exponentially with increasing zooidweight. Weight-specific grazing rates (ml x µgC–1x 24 h–1) were independent of the four-fold initial foodconcentration. Mean weight-specific grazing rates increasedlinearly with increasing zooid weight for the aggregates andoozooids, but gonozooid mean rates were independent of zooidweight. Aggregate and gonozooid ingestion rates (106 µm3x zooid–1 x 24 h–1) ranged from 4 to 134 while oozooidrates ranged from 3 to 67. All ingestion rates were independentof the initial food concentration but increased linearly withincreasing zooid weight at similar rates. All mean weight-specificingestion rates (ml x µgC–1 x 24 h–1) wereindependent of zooid weight. The mean aggregate daily ration(µgC ingested x µg body C–1) was 59% and themean doliolid ration was 132%. Field studies indicate that normalconcentrations of D. gegenbauri in the Georgia Bight clear theirresident water volume (1 m3) in about 4 months, but that highlyconcentrated, swarm populations which occur along thermohalinefronts clear their resident water volume in less than 1 day. 1Current address: MacLaren Plansearch Ltd., P.O.Box 13250, sta.A.,St.John's, Nfld. Daphnia can suppress ciliates and rotifers through predationand interference competition, but it is not known whether thisproduces any direct benefit to Daphnia. We conducted survivorshipand cohort lifetable experiments to determine whether Daphniacan utilize ciliates and rotifers as food. Three species ofoligotrich ciliates (Halteria grandinella, Strobilidium gyransand Strobilidiumvelox) and one rotifer (Keratella cochlearis)were used. Lifetable experiments were conducted with a basallevel of algae (Cryptomonas sp.), plus either ciliates or rotifers,while survivorship experiments had only the rotifers or ciliates.Densities of 30 H.grandinella ml–1, 50 S.gyrans ml–1and 15 S.velox ml–1 enhanced Daphnia pulex's populationgrowth rate 35–50% over controls with only algae. TenS.gyrans ml–1 did not produce a significant change inDaphnia's growth rate. Densities of 100 and 300 K.cochlearis–1 increased Daphnia population growth rates by II and10%, respectively. Both 10 and 50 S.gyrans ml–1 enhancedDaphnia's survivorship compared to starved controls, but neither100 nor 300 K.cochlearis l–1 enhanced its survivorship.The amount of enhancement of Daphnia growth rates by rotifersand ciliates is roughly proportional to the death rates imposedby Daphnia. The death rate imposed by Daphnia on rotifers isa function of both algal density and Daphnia size. Two approaches to quantifying relationships between nutrientsupply and plant growth were compared with respect to growth,partitioning, uptake and assimilation of NO3 by non-nodulatedpea (Pisum sativum L. cv. Marma). Plants grown in flowing solutionculture were supplied with NO3 at relative addition rates(RAR) of 0·03, 0·06, 0·12, and 0·18d–1, or constant external concentrations ([NO3)of 3, 10, 20, and 100 mmol m–3 over 19 d. Following acclimation,relative growth rates (RGR)approached the corresponding RARbetween 0·03–0.12 d-1, although growth was notlimited by N supply at RAR =0.18 d-1. Growth rates showed littlechange with [NO3–] between 10–100 mmol m–3(RGR=0·15 –0·16 d-1). The absence of growthlimitation over this range was suggested by high unit absorptionrates of NO3, accumulation of NO3 in tissues andprogressive increases in shoot: root ratio. Rates of net uptakeof NO3 from 1 mol m–3 solutions were assessed relativeto the growth-related requirement for NO3, showing thatthe relative uptake capacity increased with RGR between 0·03–0·06d–1 , but decreased thereafter to a theoretical minimumvalue at RGR  相似文献

A comparison of photosynthesis-irradiance response curves (PEresponse curves) obtained through fast repetition rate (FRR)fluorometry and radiocarbon (14C) tracer method was made inthe chlorophyte, Dunaliella tertiolecta, grown under differentirradiance conditions. In FRR-based PE response curveexperiments, actinic light provided by white light-emittingdiodes (LEDs) was increased gradually from 0 to 1500 µmolquanta m–2 s–1 and the rate of photosyntheticelectron transport was determined at each light level. Short-termexperiments (20 min) of 14C-based PE response curvewere carried out with an improved photosynthetron, which containswhite LEDs as the light source. Irrespective of growth irradiance,the ratios of FRR to 14C-based initial slopes were almost uniform.The ratios of FRR- to 14C-based maximum rates were 25–36%higher than those of FRR- to 14C-based initial slopes. The relationshipbetween electron transport and carbon assimilation was non-linearwith increasing discrepancy towards high actinic light. Thisnon-linear relationship between FRR- and 14C-based estimatesis primarily due to the effect of physiological processes stimulatedat high levels of light, such as cyclic electron flow and theMehler reaction. The results of this study indicate that theFRR fluorometry can be used as a good indicator of photosyntheticrates from low to middle light levels, but becomes increasinglyquestionable as the maximum photosynthetic rate is approached.The degree to which this relationship is further affected bynutrient-status warrants investigation.  相似文献

Ingestion, respiration, and molting loss rates were measuredover the 3 – 29°C range in Neomysis intermedia. Weightspecific rates of these physiological processes ranged from2 to 140% body C day–1 for ingestion, from 2 to 15% bodyC day–1 for respiration, and from 0.1 to 5% body C day–1for molting loss. All weight-specific rates showed a logarithmicdecrease with a logarithmic increase in body weight, and a logarithmicincrease with a linear increase in temperature below 20 or 25°C.The effect of temperature, however, was different between thephysiological rates, with a large temperature dependency foringestion (Q10 = 2.6 –3.9) and molting loss (Q10 = 2.9– 3.6) and a moderate temperature dependency for respiration(Q10 = 1.9 – 2.1). Calculated assimilation efficiencychanged with body size, but was constant over the temperaturerange examined. Allocation of assimilated materials varied witha change in temperature, reflecting the different temperaturedependence between physiological processes. The goal of this research is to enhance our knowledge of thecontributions of doliolids to the planktonic community as consumersand secondary producers. The objectives are to quantify feedingand growth rates of Dolioletta gegenbauri gonozooids at fourfood concentrations and four temperatures in order to determinetheir impact as grazers throughout the water column. Althoughdoliolids are abundant in numerous regions of the coastal ocean,and are considered to be major planktonic grazers, data on ratesof feeding and growth are scarce. Laboratory experiments wereconducted at 16.5, 20, 23.5 and 26.5°C to quantify removalof a 50:50 volumetric concentration of Thalassiosira weissflogiiand Rhodomonas sp. at four different food concentrations of20, 60, 160 and 390 µg C l–1. Results from theseexperiments suggest that clearance rates are similar at concentrationsfrom 20 to 60 µg C l –1, and decrease as the foodconcentrations increase to 160 and 390 µg C l –1.The ingestion rates increase over a range of phytoplankton concentrationsfrom 20 to 160 µg C l –1, then decrease when abnormallyhigh concentrations of 390 µg C l –1 are offered.Clearance and ingestion rates increase as temperature increasesfrom 16.5 to 26.5°C. The exponential growth rates rangefrom k = 0.2–0.7, with the lowest rates occurring at thehighest food concentration. Growth rates increase with increasingtemperature from K = 0.1–0.3 at 16.5°C to 0.45–0.7at 26.5°C. In each case, the small- and medium-sized zooidshad higher growth rates than the larger gonozooids. These resultssuggest that doliolid feeding and growth rates are a functionof environmental food concentrations and temperatures, and implythat they can be important consumers in a changing neritic environment.  相似文献

The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. Using well plates of Phaeocystis pouchetii colonies isolatedfrom experimental mesocosms in western Norway, increases incolony size and division were documented. Median longest lineardimensions increased 0–7 µm h–1; literaturePhaeocystis globosa values are 0.9–4.7 µm h–1.Ten to twelve percent of colonies divided at rates of 0.21–0.28divisions day–1. Daughter colonies were 100 µm smallerthan mother colonies. Colonies delayed 3.5–4.9 days tofirst division, compared with literature values of 4–5days for P. globosa. This study provides the first experimentalevidence for colony division of wild P. pouchetii.  相似文献

Pseudocalanus species are important contributors to the secondaryproduction of the northern hemisphere mid- to high-latitudeoceans. In the coastal Gulf of Alaska, Pseudocalanus are presentyear round and are represented by three species. In 2001, Pseudocalanusmimus was the dominant Pseudocalanus species on the shelf duringspring and summer, comprising 30–100% of the total, whilePseudocalanus newmani dominated in Prince William Sound (10–90%).Pseudocalanus minutus were only abundant in Prince William Soundduring early spring. Egg production (by number and volume) wasa function of female prosome length and decreased from springto summer; however, significant variability was attributableto regional influences that were independent of size. For thesame sized female, P. newmani produced more eggs per clutchthan P. mimus. Pseudocalanus mimus, however, tended to havea larger mean egg size than P. newmani. Consequently, clutchvolumes of the two species were indistinguishable. Pseudocalanusegg production rates (EPRs) (eggs female–1 day–1)were lower in July and August (ca. 2–4) than April andMay (ca. 1–9), but total egg production by the population(eggs day–1) was nearly equivalent for the two time periodsdue to higher female concentrations in summer.  相似文献

Dunaliella tertiolecta, a green euryhaline flagellate, is unableto use glucose as a substitute for photosynthetically fixedCO2 to maintain growth. Glucose, acetate, pyruvate, succinate,sucrose, glycerol, alanine and -ketoglutarate do not stimulateendogenous respiration in this alga. By incubating whole cellswith these compounds labelled with 14C, it was shown that onlyacetate, pyruvate and glycerol penetrated the cell at rateswhich might affect growth. These rates were still only of theorder of 10 m/(moles/hr/mg protein. Only acetate and pyruvatewere metabolized to CO2 at appreciable rates, 20 and 80% ofthe total assimilated, respectively. Cell free preparations of D. tertiolecta metabolized glucoserapidly, up to 2 µmoles/hr/mg protein, with over 75% ofthe 14C-label being recovered as triose phosphate. Both thehexose monophosphate shunt and the Embden-Meyerhof pathway wereactive. When specifically labelled glucose was supplied, CO2from the C-6 carbon was released more rapidly than from theC-6 position, in both whole cells and in the cell free extract. It is concluded that the failure of D. tertiolecta to use glucoseis due to membrane impermeability, not lack of hexokinase. Selectivity-size spectra, clearance and ingestion rates andassimilation efficiencies of Acartia clausi (Copepoda), Peniliaavirostris (Cladocera) and Doliolum denticulatum (Doliolida)from Blanes Bay (Catalan Sea, NW Mediterranean) were evaluatedin grazing experiments over a wide range of food concentrations(0.02–8.8 mm3 L–1 plankton assemblages from BlanesBay, grown in mesocosms at different nutrient levels). Acartiaclausi reached the highest grazing coefficients for large algae>70 µm (longest linear extension), P. avirostris forintermediate food sizes between 15 and 70 µm, and D. denticulatumfor small sizes from 2.5 to 15 µm. Penilia avirostrisand D. denticulatum acted as passive filter-feeders. Acartiaclausi gave some evidence for a supplementary raptorial feedingmode. Effective food concentration (EFC) decreased linearlywith increasing nutrient enrichment for D. denticulatum andfollowed domed curves for A. clausi and for P. avirostris withmaximum values at intermediate and high enrichment levels, respectively.Clearance rates of crustacean species showed curvilinear responseswith narrow modal ranges to increasing food concentration. Clearancerates of D. denticulatum increased abruptly and levelled intoa plateau at low food concentrations. The calanoid copepod, Eudiaplomus graciloides, was reared fromegg to adult on uni-algal diets (0.1. 0.5 and 2.5 mg dry wt1–1) using the green alga, Chlamydomonas reinhardtii,as food, or on a mixed diet consisting of Lake Esrom water filteredthrough a plankton net with pore size 45 µm and supplementedwith C. reinhardtii (2.5 mg dry wt 1–1). On the mixeddiet at 21.0°C growth in body dry wt (W, µg dry wt)was exponential, and the growth constants were 0.21 day–1in the early to mid juvenile stage (N1 - C4) and 0.11 day–1in the late juvenile to early adult stage (C4-A). At 14.5°Cthe corresponding growth rate constants were 0.10 and 0.08 day–1.Similar growth rates were found at uni-algal concentrationsof 0.5 and 2.5 mg dry wt I–1, and it was argued that thethreshold concentration for growth in Eudiaptomus was closeto 0.1 mg dry wt I–1. The clearance (C, ml h–1)of copepodites was measured on the uni-algal diets. The constantsof the regression (C = aWb) were: a = 0.125, b = 0.858 (2000C. reinhardtii ml–1), a = 0.068, b = 0.849 (10 000), a= 0.028, b = 0.875 (50 000). Ingestion rates were calculatedfrom the clearances and the average algal concentrations. Atthe three food levels the average daily rations were 30, 67and 125% of body dry wt. The respiration rate (R, nl O2 h–1)was measured in individuals reared on the mixed diet. The constantsof the regression (R = aWb) were: a = 4.82, b = 1.07 (nauplii,14.5°C), a = 4.17, b = 0.904 (copepodites and adults, 14.5°C),a = 6.87, b = 0.757 (copepodites and adults, 21.0°C). Nosignificant difference in the respiration rate of copepoditesreared on uni-algal diets and the mixed diet could be demonstrated.Energy budgets were calculated. The assimilation efficiencyand the gross growth efficiency of copepodites decreased markedlywith increasing food concentration, the net growth efficiencyvaried from an average of 0.44 at the lowest algal concentrationto 0.60 on the mixed diet. The results are discussed in relationto previous findings with both freshwater and marine copepods.  相似文献

Biomass and phosphorus distribution and accumulation rates wereestimated for an undisturbed subtropical rainforest in northernNew South Wales. The accumulation rates were estimated overa 16-year period. It is estimated that the steady-state above-groundbiomass for this forest is 35.0 tonne ha–1. Most of theannual biomass production was replacing litterfall and mortality.The above-ground forest contained 52 kg P ha–1 with agross annual accumulation of about 0.4 kg P ha–1/yr–1.The forest understorey and forest floor contained 4.7 kg P ha–1and 7.9 kg P ha–1, respectively. The annual uptake wasapprox. 4 kg P ha–1 yr–1.
