Hemoglobin alpha-chain variation in macaques:primary structures of the alpha 1 and alpha II chains from the adult hemoglobins of Malaysian Macaca nemestrina.

The complete primary structures of each of the two α chains commonly found in the adult hemoglobins of Malaysian Macaca nemestrina (pig-tailed macaques) were obtained from the intact chains and five fragments produced by two nonenzymatic and three enzymatic cleavage reactions. The two chains differ at a single site, the α^I chains having a glutaminyl residue and the α^II chains having a histidinyl residue in position 78. Both chains differ from their human counterpart at five positions, the extent of divergence being similar to that observed for most of the α chains from other species of Macaca that have been analyzed to date. Elucidation of the structural difference between the α^I and α^II chains demonstrates that the high degree of heterogeneity observed among the hemoglobin phenotypes of M. nemestrina is a consequence of underlying genetic variability and not a result of postsynthetic modification of genetically identical proteins. Comparisons with the hemoglobin phenotypes found in other species of Macaca support the contention that chromosomes bearing linked α^I and α^II genes, as well as those bearing single α^I and α^II alleles, combine in zygotes to produce the phenotypic variation observed in members of Malaysian populations of M. nemestrina.     

Synthesis of the mating factor of Saccharomyces cerevisiae and its truncated peptides : the structure-activity relationship.

The mating factor of Saccharomyces cerevisiae, Trp-His-Trp-Leu-Gln-Leu-Lys-Pro-Gly-Gln-Pro-Met-Tyr, has been synthesized to confirm the structure of the natural mating factor. The tridecapeptide has the same biological activity as the natural mating factor. From the studies on the biological activity of its truncated peptide synthesized the minimum sequence of the peptide require for the mating factor was deduced to be as His-Trp-Leu-Gln-Leu-Lys-Pro-Gly-Gln.     

Seasonal and geographic variations of the sterols from the starfish asterias vulgaris (verrill)

The levels of the major sterols of the starfish $\text{Asterias vulgaris}$ collected at one location in Nova Scotia varied considerably from month to month. After spawning in June the levels of the sterols in the starfish were very low, but a rapid assimilation of dietary sterols allowed the total sterol level to increase approximately two-hundred-fold to the annual maximum in July. The levels of a few minor sterols were unaffected by the spawning process, and during this period they emerged as the major components of the sterol mixture. The sterol mixtures from samples collected at different locations were compared.     

Bombesin-like peptides in small cell lung cancer: biochemical characterization and secretion from a cell line

High intracellular levels of BN-like peptides are present in tumors and cell lines of small cell carcinoma of the lung (SCCL) as well as the putative precursor cells of this tumor, the pulmonary endocrine cell. In cell line NCI-H209 the density of bombesin-like peptides was 8.9 +/- 1.1 pmol/mg total protein. Gel filtration chromatography of an extract of these cells revealed one major peak of immunoreactivity which coeluted with synthetic bombesin (1620 daltons). Also, high pressure liquid chromatography revealed one major peak of immunoreactivity was present which eluted before synthetic peptide. Therefore, SCCL bombesin-like peptides may be of similar size but are more hydrophilic than synthetic peptide. Cells maintained in culture continuously release bombesin-like peptides into the growth medium. Also, high concentrations of K+ stimulated the secretion of immunoreactive bombesin from cell lines in a Ca++-dependent manner. These SCCL bombesin-like peptides may function as important regulatory agents in the malignant lung.     

The antagonism of bombesin in the CNS by substance P analogues

The ability of substance P analogues to inhibit the action of bombesin in the CNS was investigated using receptor binding and biological assays. The putative substance P antagonists inhibited binding to central receptors for both substance P and bombesin-like peptides. Spantide, which was the most potent analogue tested, reversed the bombesin induced hypothermia and grooming. Therefore the putative substance P antagonists may also antagonize the actions of bombesin in the CNS     

Variations in the levels of asterone and asterogenol, two steroids from the saponins of the starfish, Asterias vulgaris (Verrill)

The levels of two steroids, asterone 1 and asterogenol 4, obtained by hydrolysis of the crude asterosaponin mixture from the starfish Asterias vulgaris, were highest in winter and spring, then the steroid levels fell to their annual minima in July, after the spawning period. Levels also varied geographically, but the ratio of these steroids remained approximately constant.     

Calmodulin stimulates human platelet phospholipase A2.

Calmodulin is a ubiquitous Ca²⁺-binding protein, mediating the effect of Ca²⁺ on many enzyme systems and cellular reactions. Phospholipase A₂ (phosphatide-2-acyl-hydrolase, EC 3.1.1.4) which governs the level of arachidonic acid in human platelets, requires Ca²⁺ for maximum activity. Results presented herein suggest that the stimulation of phospholipase A₂ by Ca²⁺ is also mediated through calmodulin. This finding adds to the growing list of enzymes whose activities are regulated by calmodulin.
