The concentrations of the Fusarium toxins deoxynivalenol (DON) and zearalenone (ZON) of a heavily contaminated wheat grain batch were followed over a period of 1 year by taking samples 15 times every 28 days. The air temperature and relative humidity at the top of the wheat batch ranged between 7 and 22°C and 44 and 55%, respectively, and corresponded to a variation in the moisture content of the wheat grain between 11.5 and 12.3%. None of these fluctuations were related to ZON and DON concentrations, which varied between 0.46 and 0.66 and 15.0 and 19.5 mg/kg DM. Therefore, the data were used to analyse the error sources for the analytical results. It was found that the variance proportions due to sampling and sample preparation plus analysis were not similar for DON and ZON. The variance proportion due to sampling was found to be 0.62 for ZON, which corresponded to a variance proportion of 0.38 due to sample preparation plus analysis. In contrast, the latter variance proportion for DON was estimated to be 1.0 and consequently completely superimposed the sampling error. It is concluded that long-term storage of contaminated wheat grain does not affect the concentrations of DON and ZON considering the measured fluctuations in ambient temperature, relative humidity and moisture content of the grain. Therefore, no degradation of DON and ZON occurred during the storage of wheat for a period of one year under ambient conditions. The effects of sampling and sample preparation plus analysis on the final analytical results are different for DON and ZON and require further consideration.  相似文献

Thefusarium mycotoxins deoxynivalenol (DON) and zearalenone (ZON) were determined in conventionally and organically grown grain harvested 1998 in Thuringia/Germany. A total of 196 wheat samples and 69 rye samples was analysed.In this year with heavy rainfalls during the summer months, high concentrations offusarium mycotoxins were typical in grain grown in Germany, as the DON concentrations found here. DON concentrations in conventionally grown wheat were found to be significantly higher than in organically grown wheat. 69% of the conventionally grown wheat were tested positive, containing a mean concentration of 1540 µg/kg DM. In 54% of the organically grown wheat samples DON was detected with a mean value of 760 µg/kg DM. DON concentration in rye and ZON concentration in wheat showed similar tendencies.The different cultivars of conventionally grown wheat showed large differences in DON contamination.  相似文献

Physiological samples of 100 piglets fed diets containing 0.01, 0.06, 0.15, 0.22 and 0.42 mg ZON and 0.2, 0.8, 1.0, 1.9 and 3.9 mg DON per kg over a period of 35±1.5 days were investigated for concentrations of deoxynivalenol (DON) and zearalenone (ZON) and their metabolites. DON was detected in serum, bile and urine in increasing concentrations corresponding to the diet contamination. The metabolite de-epoxy-DON was detected only in urine. The DON contamination of the diet was closely reflected by the serum concentrations of the piglets. ZON and its metabolite α-zearalenol were detected in bile fluid, liver and urine, while β-zearalenol was only detected in bile fluid. In serum neither ZON nor its metabolites were found. The total concentration of ZON plus its metabolites in the bile fluid corresponded well with the dietary contamination. For all analyses it has to be noted that toxin residues were detectable even in individual samples of piglets fed the control diet containing 0.01 mg ZON/kg and 0.2 mg DON/kg. The aim of the study was to examine the effects of dry matter intake level and the feeding ofFusarium-contaminated wheat on the toxin-turnover and ruminal fermentation of dairy cows. Fourteen dairy cows equipped with ruminal and duodenal cannulae were used. All animals were fed the same diet, only the daily feed amounts were adjusted to the current performance stage of the cow. On a dry matter basis, the diet consisted of 60% concentrate including 55% wheat (Fusarium-contaminated wheat [Mycotoxin period] or control wheat [Control period]). Each cow was fed with both the contaminated and the control wheat. TheFusarium-contamination of the wheat significantly decreased the flow of undegraded protein at the duodenum with increased intakes of organic matter. The duodenal flow of microbial protein and the activities of aspartate aminotransferase (ASAT), glutamate dehydrogenase (GLDH) and gamma glutamyl transferase (γ-GT) in the serum were not affected by dietary treatment, but increased with feed intake. The duodenal flow of deoxynivalenol (DON) and de-epoxy DON related to DON intake ranged between 12 and 77% when theFusarium-contaminated wheat was fed. DON was almost completely metabolized to de-epoxy DON independent of the feed intake level. The zearalenone (ZON) flow at the duodenum increased moderately with increasing ZON/feed intake.  相似文献

A dose response study was carried out with piglets to examine the effects of increasing amounts of Fusarium toxins in the diet on performance, clinical serum characteristics, organ weights and residues of zearalenone (ZON) and deoxynivalenol (DON) and their metabolites in body fluids and tissues. For this purpose, Fusarium toxin contaminated maize (1.2 mg ZON and 8.6 mg DON per kg maize) was incorporated into a maize based diet for piglets at 0, 6, 12.5, 25 and 50% at the expense of control maize. The experimental diets were tested on 100 female piglets allotted to 20 boxes (five animals per box) covering a body weight range of 12.4 ± 2.2 kg to 32.5 ± 5.6 kg. Voluntary feed intake and, consequently, body weight gain of the animals receiving the highest proportion of Fusarium toxin contaminated maize were significantly decreased while the feed conversion ratio was not affected by the treatment. The mean weight of the uterus related to the body weight of the animals of the same group was increased by almost 100% as compared to the control. For this group, significantly decreased values of total serum protein were determined, while the serum activity of the liver enzyme glutamate dehydrogenase and the serum concentration of the follicle stimulating hormone were decreased for all treatment groups receiving 6% contaminated maize or more in the diet. Serum concentrations of immune-globulins were not consistently altered by the treatment. Corresponding to the dietary exposure, increasing concentrations of ZON and α-zearalenol were detected in the bile fluid, liver and in pooled urine samples. The metabolite β-zearalenol was detected only in bile fluid. The total concentration of ZON plus its metabolites in bile fluid correlated well with the diet contamination (r = 0.844). DON was found in serum, bile fluid and pooled urine samples while de-epoxy-DON was detected only in urine. The serum concentration of DON correlated well with the respective toxin intake 3 - 4 h prior to slaughtering (r = 0.957). For all mentioned analyses of residues it has to be noted that toxin residues were detectable even if negligible concentrations were present in the diet.  相似文献

From 1998 to 2001 a total of about 1172 conventionally and organically produced samples of wheat, rye, barley and triticale were examined for the presence of deoxynivalenol (DON) and zearalenone (ZON). Furthermore, feedstuffs for pigs were included in the monitoring of Fusarium toxins. DON and ZON analyses were performed using ELISA or HPLC. The incidences and levels of toxins varied from year to year. Overall contamination levels were highest in wheat and triticale, followed by rye and barley. The highest DON contaminations were found in 1998. The probes of the years 1999-2001 showed lower incidences of Fusarium toxins. The second examined mycotoxin ZON was detected at lower levels in cereals. Similar results were observed in the monitoring of feedstuffs.  相似文献

Deoxynivalenol (DON) is a hazardous and globally prevalent mycotoxin in cereals. It commonly accumulates in the grain of wheat, barley and other small grain cereals affected by Fusarium head blight (caused by several Fusarium species). The concept of reducing DON in naturally contaminated grain of wheat or barley using a DON-degrading bacterium is promising but has not been accomplished. In this study, we isolated a novel DON-utilising actinomycete, Marmoricola sp. strain MIM116, from wheat heads through a novel isolation procedure including an in situ plant enrichment step. Strain MIM116 had background degradation activity, and the activity was enhanced twofold by the consumption of DON. Among Tween 20, Triton X-100 and Tween 80, we selected Tween 80 as a spreading agent of strain MIM116 because it promoted DON degradation and the growth of strain MIM116 in the presence of DON. The inoculation of MIM116 cell suspension plus 0.01% Tween 80 into 1,000 harvested kernels of wheat and barley resulted in a DON decrease from approximately 3 mg kg?1 to less than 1 mg kg?1 of dry kernels, even when cells had only basal levels of DON-degrading activity. To the best of our knowledge, this is the first report that describes (1) the isolation of a DON-degrading bacterium from wheat heads, (2) the effects of surfactants on the biodegradation of DON and (3) the decrease of DON levels in naturally contaminated wheat and barley grain using a DON-degrading bacterium.  相似文献

The objective of this study was to follow the mycotoxin formation and changes in nutrient composition of wheat (cv. Ritmo) artificially inoculated with Fusarium culmorum. From anthesis until harvest, samples were taken once a week from the inoculated and control plots. The investigations were focused on monitoring the progression of the contamination of the wheat kernels with deoxynivalenol (DON) and zearalenone (ZON). Both the uncontaminated control kernels and the contaminated kernels were examined also for the presence of zearalenone-4-beta-D-glucopyranoside and several trichothecenes at harvest. Furthermore, the impact of the Fusarium inoculation on some nutrients as starch, crude protein, amino acid composition, crude ash, non starch polysaccharides (NSP) as well as viscosity and thousand seed weight (TSW) was examined. Also proteolytic and amylolytic activity as well as the NSP-degrading enzyme activities of inoculated and control samples were analysed at the time of harvest. DON was detected in higher concentrations and in earlier stages, while ZON was found later and in smaller amounts. On average 7.79 mg/kg DM of DON and 100 microg/kg DM of ZON were found in the inoculated kernels at the time of harvest. Neither in the contaminated nor in the control samples glucose conjugates of ZON (Zearalenone-4-beta-D-glucopyranoside) were detected. Moreover, the infection with Fusarium culmorum had pronounced effects on some quality parameters. The crude protein content of the inoculated kernels showed significantly higher values over the whole period compared to the control kernels. The protein content of the inoculated kernels amounted 13.9% DM at harvest, while only a concentration of 12.5% DM was detected in the control samples. Similarly, in almost all stages of development the crude ash content of inoculated samples was higher than in control samples. These distinct differences in kernel composition resulted possibly from the changes of the thousand seed weight. In the present work the grain harvested from the control plots showed a significantly higher TSW (24.2 g) as compared to their inoculated counterparts (15.5 g). Despite lower extract viscosity of inoculated samples at time of harvest, the content of soluble NSP of inoculated plots was higher than in control samples at the same time. Moreover, inoculation resulted in markedly increased activities of protease, amylase and several NSP-degrading enzyme activities. This would suggest that the cell wall penetrating properties of the fungus itself and/or that the fungus induced alterations of the metabolic activity of the embryo or other constituents of the wheat kernel could be responsible.  相似文献

Forty-five samples from 1988–1995 of naturally contaminated grain, barley, wheat and oats, three samples of mixed feed, and 16 samples of grain artificially inoculated with Fusarium culmorum during the flowering stage were analysed for deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-acetyl-DON), culmorin and hydroxy-culmorins. These compounds are secondary metabolites produced by the fungal species F. culmorum and F. graminearum. Acetonitrile-water extract of the samples was purified on a MycosepTM#225 column, derivetized using pentafluoropropionic anhydride (PFPA) and analysed by gas chromatography-mass spectrometry (GC-MS). The amount of each of culmorin, 5-, 12-, 14 and 15-hydroxy-culmorin and one unknown hydroxy-culmorin were determined relative to the amount of DON plus 3-acetyl DON for each sample. The ratio between the total amount of culmorin compounds and the DON compounds ranged from 0.14 to 1.07 in the samples. This study shows that there is a strong correlation between the amount of DON present in the grain and the amount of culmorin and hydroxy-culmorins present. The ratio of each of the culmorin compounds relative to the amount of DON compounds were in the same range in the grain artificially inoculated by F. culmorum as found in an earlier study for F. culmorum strains cultivated on rice, while the hydroxy-culmorin profile in the naturally contaminated grain was more similar to what was found for the F. graminearum cultures in the same study [1]. These results indicate that F. graminearum may be a relatively important source for DON in grain also in relatively cold areas. Unground wheat kernels contaminated with 2.09 mg deoxynivalenol (DON) per kg dry matter were stored for up to 56 days at moisture contents of 15, 17.5 and 20% to study the alterations of DON concentration when the wheat was stored either unsupplemented or supplemented with 5 g sodium metabisulfite (Na2S2O5, SBS), 10 g propionic acid (PA) or 5 g SBS plus 10 g PA per kg. SBS addition alone or in combination with PA reduced the DON contamination to 1.2–4.3% of the initial DON concentration while DON concentration of unsupplemented and wheat batches supplemented only with PA varied inconsistently or remained unchanged. The SBS-related DON reduction was paralleled by a concomitant increase in the concentration of the non-toxic reaction product DON sulfonate. In contrast to the unsupplemented wet-stored controls, SBS addition prevented the growth of moulds and yeasts when added alone or in combination with PA. In conclusion, for the conditions examined, the wet preservation of DON-contaminated wheat with SBS seems to be promising as an on-farm detoxification measure.  相似文献

A study was conducted to determine the distribution of deoxynivalenol (DON) and ochratoxin A (OTA) in a lot of 261 of wheat kernels. Within this study, two different sampling and sample preparation strategies were carried out. On the one hand, following the official commission regulation 401/2006/EC, an aggregate sample out of 100 incremental samples was build, homogenized and prepared for laboratory analysis. On the other hand each individual subsample was investigated for its deoxynivalenol and ochratoxin A content. The determined concentration of DON in the individual samples was in a range from 830 up to 2655 (μg/kg, for OTA results ranged from < 0.2 up to 8.6 μg/kg. Thus, a coefficient of variance of 25% for DON and 200% for OTA was achieved. From this, a spot formation for OTA was observed and the average value of the 100 incremental samples did not correspond to the achieved value for the aggregate sample. While the DON contamination at this concentration range seems to be more even, consequently the result of the aggregate sample was in accordance with the average value. In addition a sample communition study was performed to answer the question whether the time consuming process of grinding of the whole aggregate sample is necessary or not. The results of this study show that contamination of whole wheat kernels with DON is at the same level within a 1 kg sample (CV 16%), while OTA contamination shows high variability (CV 94%). At least for OTA this study indicated that an extensive and complete sample communition of the high volume aggregate sample is necessary.  相似文献

Experiments were carried out to examine the effects of a Fusarium contaminated wheat (10mg deoxynivalenol and 0.76mg zearalenone, ZON, perkg dry matter) and of a detoxifying agent (Mycofix ®Plus, Biomin GmbH, Herzogenburg, Austria) on the growing performance of bulls, carry-over of ZON and its metabolites into body fluids and tissues, and on nutrient digestibility in wethers. The experiments were designed according to a complete two by two factorial approach which meant that both the uncontaminated control wheat and the Fusarium toxin contaminated wheat were tested both in the absence and presence of Mycofix ®Plus. The growing experiment with bulls (n = 14 per treatment) covered the live weight range between 244kg and 460kg. The respective wheat batches were included in the concentrate portion at 65%. Concentrates were fed according to plan whereas maize silage was offered for ad libitum consumption. Daily dry matter intake and live weight gain [kg per animal and day] were 7.40, 7.52, 7.51 and 7.49 and 1.367, 1.296, 1.380 and 1.307 for bulls fed the unsupplemented control wheat, the supplemented control wheat, the unsupplemented and Fusarium toxin contaminated wheat and the supplemented Fusarium toxin contaminated wheat, respectively. ZON and its metabolites were not detected in edible tissues. The most striking effects of feeding the Fusarium toxin contaminated wheat on carcass characteristics were a reduced dressing percentage, an increased weight of the emptied gastro-intestinal tract and a reduced weight of the testicles. No effect of the detoxifying agent was seen for these parameters whereas heart weight increased independently of Fusarium toxin contamination of the concentrates. Nutrient digestibility of the two wheat batches, unsupplemented or supplemented with Mycofix ®Plus was evaluated according to the difference method using wethers. Presence of Fusarium toxins in wheat did not influence its feeding value. The effects of the addition of the detoxifying agent were mycotoxin unspecific and resulted in an increase in apparent digestibility of crude protein and a decrease in crude fiber digestibility. It is concluded that feeding of Fusarium toxin contaminated wheat did not adversely affect performance of growing bulls (approximately 2.2mg DON and 0.1mg ZON perkg complete ration at a reference dry matter content of 88%) or nutrient digestibility in wethers. The effects of the detoxifying agent Mycofix ®Plus on growing performance and on nutrient digestibility were rather Fusarium toxin unspecific. The slightly negative effects on growing performance needs to be examined further.  相似文献

The amount of the Fusarium toxins deoxynivalenol (DON). 3-acety-DON. ivalenol and fusarenon × have been determined in about 3000 samples of barley, oats and wheat grown in Norway. The samples were collected in the period 1988–1994. and grouped according to grain species, year of production, district and sample category. The DON content was found to be significantly higher in oats than both barley and wheat in grain produced by ordinary grain producers in Norway. Of samples collected from grain silos. 68.7% of the barley. 97.1% of the oats and 66.3% of the wheat samples were found to contain 30 mg/kg of DON or more. The proportion containing 1000 μg kg or more was 2.1.13.4and0.0% in burley, oats and wheat, respectively. However, these variations in contamination level were not reflected in the results from two field trials, where the grain species were grown side-by-side in the same fields. The differences in contamination level between the grain species seem to be due partly to edaphic and agrotechnical factors and partly to variation in the susceptibility to different Fusarium species in interaction with climatic factors. Nivalenol(≥ 50 μg/kg) was detected in 8.3% of the barley samples. 8.2% of the oats and 0.4% of the wheat samples. Samples collected at the grain silos were then not included.  相似文献

The Fusarium head blight (FHB)-susceptible winter wheat cv. Ritmo was inoculated with spores of Fusarium culmorum at the beginning of full blossom. Samples of whole wheat plants were taken once weekly from anthesis until harvest and subsequently fractionated into straw, glumes and spindles, which were examined for deoxynivalenol (DON) and zearalenone (ZON). Additionally, the content of crude protein (CP) and non-starch polysaccharides (NSP) was scrutinized. Synthesis of the Fusarium toxins DON and ZON generally differed in terms of date of formation and concentration. Final mean DON concentrations of 37.5, 28.1 and 5.0 mg/kg DM were measured in glumes, spindles and straw, respectively, at the time of harvest. At this time, maximal mean ZON concentrations of 587, 396 and 275 microg/kg DM in spindles, glumes and straw, respectively, were determined. Moreover, Fusarium infected wheat residues contained higher CP but lower NSP contents at the last three sampling dates. In addition, collective samples of wheat straw and chaff were taken to investigate the effect of the Fusarium contamination on their in sacco DM degradation in dairy cows. Samples were analysed for mycotoxins and selected quality parameters. The dried and milled collective samples of straw and chaff were weighed into nylon bags and subjected to ruminal incubation for 4, 8, 16, 24, 48, 72, 96 and 120 h in two dairy cows equipped with a permanent rumen cannula. Marked differences in level of mycotoxin contamination as well as in ingredient composition between the variants of straw and chaff were detected. Moreover, after 120 h rumen incubation the in sacco DM degradation of inoculated straw and chaff were lower compared to the accordant controls. The soluble fraction was increased in inoculated samples, whereas a diminishment in the potentially degradable but insoluble fraction was more pronounced. Thereby, a decrease in the potential degradability was obtained for inoculated straw and even if less pronounced for chaff compared to the non inoculated corresponding controls. In conclusion, infection with F. culmorum of wheat involves an increased risk of mycotoxin contamination in straw. Also, a Fusarium infection may have an impact on chemical composition and may result in Fusarium growth-related modifications of host cell wall components.  相似文献

The impact of moisture on the development of Fusarium head blight (FHB) and accumulation of deoxynivalenol (DON) in Fusarium-infected wheat was examined. The field experiments were designed as split-split-plot with five replicates. Main plots were durations of mist-irrigation [14, 21, 28 and 35 days after inoculation (DAI)]; sub-plots were wheat cultivar; and sub-sub-plots were F. graminearum isolates differing in aggressiveness and DON production capacity. The wheat cultivars ‘Alsen’ (moderately resistant), ‘2375’ (moderately susceptible) and ‘Wheaton’ (susceptible) were inoculated at anthesis. Severity of FHB was assessed 21 days after inoculation. Visually scabby kernels (VSK) and mycotxin content (DON, 15-AcDON, 3-AcDON and nivalenol) were determined on harvested grain. The damage to grain, as measured by VSK, was significantly lower in the treatments receiving the least amount of mist-irrigation (14 DAI) suggesting that extended moisture promotes disease development. DON was, however, significantly lower in the 35-DAI misting treatment than in treatments receiving less post-inoculation moisture. The reduction of DON observed in treatments receiving extended mist-irrigation was greatest in ‘Wheaton’ which recorded the highest FHB severity, VSK and DON of the cultivars examined. Our results suggest that DON and other trichothecenes may be reduced by late-season moisture despite increased grain colonization. We suggest that leaching may explain much of the reduction of mycotoxins, and that differences in tissue morphology and metabolism may determine the rate of leaching from specific tissues.  相似文献

An experiment was carried out to examine the effects of aFusarium-contaminated wheat grain as a component of the concentrate portion (10 mg deoxynivalenol and 0.76 mg zearalenone, ZON, per kg dry matter) on performance of growing bulls, and on carry over of ZON into tissues and body fluids. In a second study, rumen physiological parameters were investigated in wethers equipped with rumen fistulae. Moreover, the influences of a detoxifying agent (Mycofix®, MP, Biomin GmbH, Herzogenburg, Austria) were considered as an additional experimental factor beside the contamination of the wheat (uncontaminated control wheat,Fusarium-toxincontaminated wheat).The fattening experiment with bulls (n=14 per treatment) covered the live weight range between 244 kg and 460 kg. Daily dry matter intake and live weight gain (kg per animal and day) were 7.40, 7.52, 7.51 and 7.49 and 1.367, 1.296, 1.380 and 1.307 for bulls fed the unsupplemented control wheat, the supplemented control wheat, the unsupplemented andFusarium toxin contaminated wheat and the supplementedFusarium toxin contaminated wheat, respectively. Concentration of ZON and its metabolites in edible tissues were lower than the detection limits of the applied HPLC-method.The results of the rumen physiological investigations revealed that the molar ratios of short chained volatile fatty acids and ammonia concentration in rumen fluid remained unchanged in response to dietary treatments whereas the addition of MP to the diets buffered the postprandial decrease in rumen pH.  相似文献

Nine gilts weighing 80 kg at the beginning of the trial were fed a mycotoxin contaminated diet containing 2 mg deoxynivalenol (DON) and 0.4 mg zearalenone (ZON) per kg (Diet M). Their daily weight gain until 103 kg BW was reduced in comparison to the nine control animals fed an uncontaminated diet (Diet C) (763 vs. 912 g; p = 0.02). There was no treatment effect on the age at first observed oestrus. Seven and eight gilts receiving Diet M and C, respectively, became pregnant after being mated once or being again mated three weeks later. The examination of the uteri of gilts slaughtered 35–61 days after mating showed that the exposure to DON and ZON had no effect on the number of foetuses per gilt (p = 0.54), but increased their growth rate (p = 0.003). Thus, low dietary DON and ZON levels had no negative effects on the reproductive parameters examined. The hypothesis that the bulbourethral gland weight of barrows can be used for the bioassay of low dietary ZON levels was rejected since feeding Diet M from 80–103 kg BW did not increase the weight of that accessory sex gland (p = 0.51).  相似文献

Surveys of corn (infected plants and commercial kernels) forFusarium species and their mycotoxins were carried out on samples collected all over Italy and from some European and mediterranean countries.Investigations on samples of corn stalk and ear rot standing in the field, mainly collected in southern Italy, proved to be contaminated with zearalenone (ZON), zearalenols (ZOL), and deoxynivalenol (DON). TheFusarium species most frequently isolated, and their recorded toxigenic capability (in parentheses), were:F. moniliforme;F. culmorum (ZON, ZOL, DON, 3AcDON);F. equiseti (ZON, ZOL); andF. proliferatum (MF). Thirty-seven antagonists, from a collection of 113 pre-screened microorganisms, were tested in field experiments on winter wheat for their ability to control ear blight and production of mycotoxins, particularly deoxynivalenol (DON), in grain by Fusarium culmorum and F. graminearum. Ears were spray-inoculated during anthesis with spore suspensions, first of test fungi and then of the pathogen. Isolates of F. equiseti were the most effective treatments. A strain of F. equiseti (G9) decreased DON consistently on wheat inoculated with F. culmorum, compared with the control, often by more than 70%. It performed well under severe disease pressure (F. culmorum at 105 conidia mL-1) and similarly to a standard fungicide, tebuconazole. Percentages of diseased grains and amounts of DON corresponded well. Small quantities of nivalenol (NIV) occurred in grain samples after treatment with some F. equiseti strains. On wheat inoculated with F. graminearum, F. equisti G2 decreased the percentage diseased grains by 92% and DON by 94%. One strain of F. equiseti (G2) was found to produce small amounts of NIV in pure culture.  相似文献
