The principal forms of amino nitrogen transported in xylem were studied in nodulated and non-nodulated peanut (Arachis hypogaea L.). In symbiotic plants, asparagine and the nonprotein amino acid, 4-methyleneglutamine, were identified as the major components of xylem exudate collected from root systems decapitated below the lowest nodule or above the nodulated zone. Sap bleeding from detached nodules carried 80% of its nitrogen as asparagine and less than 1% as 4-methyleneglutamine. Pulse-feeding nodulated roots with 15N2 gas showed asparagine to be the principal nitrogen product exported from N2-fixing nodules. Maintaining root systems in an N2-deficient (argon:oxygen, 80:20, v/v) atmosphere for 3 days greatly depleted asparagine levels in nodules. 4-Methyleneglutamine represented 73% of the total amino nitrogen in the xylem sap of non-nodulated plants grown on nitrogen-free nutrients, but relative levels of this compound decreased and asparagine increased when nitrate was supplied. The presence of 4-methyleneglutamine in xylem exudate did not appear to be associated with either N2 fixation or nitrate assimilation, and an origin from cotyledon nitrogen was suggested from study of changes in amount of the compound in tissue amino acid pools and in root bleeding xylem sap following germination. Changes in xylem sap composition were studied in nodulated plants receiving a range of levels of 15N-nitrate, and a 15N dilution technique was used to determine the proportions of accumulated plant nitrogen derived from N2 or fed nitrate. The abundance of asparagine in xylem sap and the ratio of asparagine:nitrate fell, while the ratio of nitrate:total amino acid rose as plants derived less of their organic nitrogen from N2. Assays based on xylem sap composition are suggested as a means of determining the relative extents to which N2 and nitrate are being used in peanuts.  相似文献

The composition of xylem sap and exudate from stem incisionsof Nicotiana glauca Grah. was compared in detail. Exudationfrom stem incisions occurred over a 5 min period in certainplants, enabling collection of 5–30 µl of sap. Therate of exudation showed an exponential decline. Exudate hada high dry matter content (170–196 mg ml–1) andhigh sugar (sucrose) levels. Xylem sap had a low pH (5.8) andexudate a pH of 7.9. Glutamine dominated the amino compoundsin xylem sap and exudate, and K+ was the major cation. Totalamino compounds in stem exudate reached 10.8 mg ml–1 whereasxylem sap contained much lower levels (0.28 mg ml–1).All mineral elements and amino compounds with the exceptionof calcium were more concentrated in stem exudate than in xylemsap. Sucrose was labelled heavily in stem exudate following pulsingof an adjacent leaf with 14CO2. A concentration gradient ofsugar (2.1 bar m–1) was recorded for stems. Levels ofsucrose, amino compounds and K+ ions in stem exudate showeda diurnal periodicity. Each commodity reached maximum concentrationat or near noon and minimum concentration about dawn. The evidencesuggests that exudate from stem incisions of N. glauca is arepresentative sample of solutes translocated in the phloem. Growth and symbiotic performance of soybean (Glycine max (L.)Merrill) cv. Bragg and three of its induced nodulation mutants(nod49, non-nodulating; ntsl 116, intermediate supernodulator;nts1007, extreme supernodulator) were compared throughout developmentunder different nitrogen regimes (0, 2, 5 and 10 mol nitratem–3). Nitrogen fixation was assessed using 15N-isotopedilution and xylem sap analysis for ureide content. Both techniquesconfirmed a complete lack of N2 fixation activity in nod49.Plant reliance on nitrogen fixation by the other genotypes wasdependent on the nitrate regime and the developmental stage. The ntsl007 and ntsl 116 mutants fixed more nitrogen than theparent cultivar in the presence of 10 mol m–3 nitratein the nutrient solution, but higher input of symbioticallyderived nitrogen was still insufficient to offset the amountof nitrogen removed in the harvested seed. However, the mutantsutilized less nitrate for growth than Bragg. Comparison of estimatesof N2 fixation derived from the 15N-dilution technique withthose based on relative ureide content of xylem sap indicatedthat the latter offered a simple and reliable procedure forevaluating the symbiotic performance of supernodulating plants. The feed back control mechanism proposed to explain the inhibitionof N2 fixation by N was investigated using Vicia faba cv. Fiord.Plants were grown under controlled conditions without mineralN in coarse river sand. Asparagine was supplied to plants activelyfixing N2 by absorption through cut roots and via a wick ordirect injection into the stem just above the bottom leaf. Responsesin N2 fixation were measured by acetylene reduction (AR). Feedingplants with [14C]-labelled asparagine showed that the amidewas taken up when exogenously applied. Asparagine (10 mM) suppliedby the above procedures resulted in a 50-70% inhibition of ARby 48 h. Glutamine produced a similar effect. The cut root methodallowed higher levels of these amides to be supplied but theinhibition observed with 10 mM asparagine was only increasedslightly with higher levels of the amide. The antibiotic Securopenprevented bacterial contamination of root solutions of asparagineand glutamine and had no effect on nodule activity. It is concludedthat accumulation of asparagine of glutamine or the resultantincrease in the pool of soluble N in the plant cause a feedbackeffect on the activity of nitrogenase.Copyright 1993, 1999 AcademicPress Vicia faba, faba bean, asparagine, inhibition of N2 fixation  相似文献

The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. Seedlings (180-d-old) of Casuarina cunninghamianaM L., C. equisetifoliaMiq. and C. glauca Sieber inoculated with each of two differentsources of Frankia, were analysed for translocated nitrogenouscompounds in xylem sap. Analyses were also made on sap fromnodulated and non-nodulated plants of C. glauca grown with orwithout a range of levels of combined nitrogen. Xylem exudateswere collected from stems, roots, and individual nodules ofnodulated plants and from stems and roots of non-nodulated plants. While the proportional composition of solutes varied, the samerange of amino compounds was found in xylem sap from the threedifferent symbioses. In C. glauca asparagine was the major aminoacid in the root sap followed by proline, while in symbioticC. cunninghamiana arginine accounted for more than 25% of theamino compounds. Citrulline was the major translocated productfound in the stem exudate of symbiotic C. equisetifolia. Increasingconcentrations of ammonium nitrate in the nutrient solutionresulted in increasing levels of free ammonia and glutaminein xylem sap from stems of nodulated and non-nodulated C. glauca, but there was relatively little change in the prominent solutes,e.g. citrulline, proline, and arginine. The composition of nitrogenoussolutes in stem or root exudates of C. glauca was similar tothat of exudate collected from individual nodules and on thisbasis it was not possible to distinguish specific products ofcurrent N2 fixation in xylem. The main differences in N solutecomposition between the symbioses were apparently due to hostplant effects rather than nodulation or the levels of combinedN. Also, the data indicate that the use of the proportion ofN in sap as citrulline (or indeed any other organic N solute)could not be used as an index of nitrogen fixation.  相似文献

Nodulated 1-1.5-year-old plants of Acacia littorea grown inminus nitrogen culture were each partnered with a single seedlingof the root hemiparasite Olax phyllanthi. Partitioning of fixedN between plant organs of the host and parasite was studiedfor the period 4–8 months after introducing the parasite.N fluxes through nodules of Acacia and xylem-tapping haustoriaof Olax were compared using measured xylem flows of fixed Nand anatomical information for the two organs. N2 fixation duringthe study interval (635 µg N g FW nodules–1 d–1)corresponded to a xylem loading flux of 0.20 µg N mm–2d–1 across the secretory membranes of the pencycle parenchymaof the nodule vascular strands. A much higher flux of N (4891µg mm–2 d–1) exited through xylem at the junctionof nodule and root. The corresponding flux of N from host xylemacross absorptive membranes of the endophyte parenchyma of Olaxhaustorium was 1.15 µg N mm–1 d–1, six timesthe loading flux in nodules. The exit flux from haustorium toparasite rootlet was 20.0 pg N mm–1 d–1, 200-foldless than that passing through xylem elements of the nodule.Fluxes of individual amino compounds in xylem of nodule andhaustorium were assessed on a molar and N basis. N flux valuesare related to data for transpiration and partitioning of Cand N of the association recorded in a companion paper. Cycling of amino compounds in symbiotic lupin   总被引:2,自引:0,他引:2
The composition of amino acids was determined in the xylem andphloem sap of symbiotic lupins grown under a variety of treatmentsdesigned to alter the rate of nitrogen fixation. Asparaginewas the major amino acid in both xylem and phloem with glutamine, glutamate and aspartate also major components. GABA had a highconcentration in the xylem while valine was a major componentin the phloem. Exposure to combined nitrogen in the form ofeither ammonium or nitrate caused a reduction in specific nitrogenaseactivity and was associated with subsequent changes in bothof the translocated saps. Inhibiting nitrogen fixation by exposingnodules to oxygen produced a lower amide to amine ratio in thexylem sap (1.3:1) compared with control and nitrate ratios (2.6:1)and ammonium ratios (7.1:1). Similar ratios for amide aminewere also observed in the phloem sap. Labelling studies using15N2 to follow nitrogen fixation, ammonium assimilation andamino acid transport have shown rapid accumulation of labelinto glutamine with subsequent enrichment in glutamate, aspartate, alanine, and GABA. Asparagine was found in high concentrationsin nodules and became slowly enriched. Labelled nitrogen fixedand assimilated in nodules was detected 40 min later in stemxylem extracts, largely as the amides glutamine and asparagine. These experiments provide evidence that large amounts of nitrogenouscompounds are cycled through the root nodules of symbiotic plants(contributing approximately 50% of xylem N) and that differencesin the composition of the phloem sap may influence nodule growthand activity. The absorption and distribution of Cu in red clover (Trifoliumpratense L.) were measured in plants grown in flowing solutionculture with Cu maintained throughout at 0.5 µg 1–1and N supplied either as nitrate or through symbiotic fixation.Although there was a decrease in Cu absorption, both with time, and with a depleted nitrate supply, it increased to its formerrate when nitrate was adjusted to 10 mg N 1–1 after aperiod of depletion. Differences in absorption between plantsdependent upon fixation and those supplied with nitrate wererelated to the slower initial growth of the plants fixing N.Considerable proportions (> 30 per cent) of the absorbedCu were retained by the roots. At the final harvest, and withthe exception of plants grown with nitrate adjusted to 0.1 mgN 1–1 after a period of depletion, the proportion of theCu retained was related to the concentration of N in the roots. The different N treatments produced differences in Cu concentrationin the shoots, and the effects were greater in the youngestfully expanded leaves than in older leaves. An experimentally-based modelling technique was developed todescribe quantitatively the uptake, flow, storage and utilizationof NO3-N over a 9 d period in mid-vegetative growth of sandcultured castor bean (Ricinus communis L.) fed 12 mol m–3nitrate and exposed to a mean salinity stress of 128 mol m–3NaCl. Model construction used information on increments or lossesof NO3-N or total reduced N in plant parts over the study periodand concentration data for NO3-N and reduced (amino acid) Nin phloem sap and pressure-induced xylem exudates obtained fromstem, petiole and leaf lamina tissue at various levels up ashoot. The resulting models indicated that the bulk (87%) of incomingnitrate was reduced, 51% of this in the root, the remainderprincipally in the laminae of leaves. The shoot was 60% autotrophicfor N through its own nitrate assimilation, but was oversuppliedwith surplus reduced N generated by the root and fed to theshoot through the xylem. The equivalent of over half (53%) ofthis N returned to the root as phloem translocate and, mostly, then cycled back to the shoot via xylem. Nitrate comprised almosthalf of the N of most xylem samples, but less than 1% of phloemsap N. Laminae of leaves of different age varied greatly inN balance. The fully grown lower three leaves generated a surplusof reduced N by nitrate assimilation and this, accompanied byreduced N cycling by xylem to phloem exchange, was exportedfrom the leaf. Leaf 4 was gauged to be just self-sufficientin terms of nitrate reduction, while also cycling reduced N.The three upper leaves (5–7) met their N balance to varyingextents by xylem import, phloem import (leaves 6 and 7 only)and assimilation of nitrate. Petioles and stem tissue generallyshowed low reductase activities, but obtained most of theirN by abstraction from xylem and phloem streams. The models predictedthat nodal tissue of lower parts of the stem abstracted reducedN from the departing leaf traces and transferred this, but notnitrate, to xylem streams passing further up the shoot. As aresult, xylem sap was predicted to become more concentratedin N as it passed up the shoot, and to decrease the ratio ofNO3-N to reduced N from 0·45 to 0·21 from thebase to the top of the shoot. These changes were reflected inthe measured N values for pressure-induced xylem exudates fromdifferent sites on the shoot. Transfer cells, observed in thexylem of leaf traces exiting from nodal tissue, were suggestedto be involved in the abstraction process. Cotyledons of faba bean (Vicia faba L. cv. Fiord) were removedto determine whether an apparent delay in nodulation of thiscultivar could be attributed to an inhibitor from these organs.Cotyledons were left intact or excised from seedling plants14 and 18 d after sowing and plants grown with or without 2·5mm NO3. Seedling growth was depressed when cotyledons were removed onday 14 but not when removed on day 18. Removal of the cotyledonsat day 14 reduced nodule number and nodule weight in the absenceof NO3, but in the presence of NO2, nodule numberwas unaffected and only nodule weight was reduced. Cotyledonremoval at day 18 increased both nodule number and nodule weightwith +NO3 but not with –NO2. Acetylene reduction(AR) was markedly depressed by NO3. Cotyledon removalat day 14 decreased AR but removal at day 18 resulted in anincrease in AR. We suggest from these results that faba beancotyledons have an inhibitory effect on nodule activity andon nodulation and this interacts with NO3. This can beexplained through a ‘feed-back’ regulation of N2fixation by soluble N in the seedling. Transport of Nitrate and Calcium into Legume Root Nodules   总被引:5,自引:0,他引:5
Nitrate transport into nodulated plants of soybean (Glycinemax), cowpea (Vigna unguiculata) and faba bean (Vicia faba)was investigated. Nitrate entering the root system of soybeandid not pass out of the vascular system into nodular tissuesin detectable quantities. On the other hand, nitrate could passfrom soil through the outer surface of nodules but did not penetratethe infected tissue. Similarly, nitrate was restricted to corticaltissues of cowpea and faba bean. Thus, nitrate cannot inhibitnitrogen fixation as a result of reduction to nitrite by nitratereductase within the bacteroid zone. These results are, however, consistent with an effect of nitrate on an oxygen diffusionresistance located in the nodule cortex. Unlike nitrate, measurable quantities of 45calcium were transportedvia the xylem into infected and cortical tissues of soybeannodules: it also passed from the soil into the free space ofthe nodule cortex. Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen, total plant leaf area and specific leaf area (SLA) all increased. The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. Herridge, D. F., O'Connell, P. and Donnelly, K. 1988. The xylemureide assay of nitrogen fixation: sampling procedures and sourcesof error.—J. exp. BoL 39: 12–22. The relative abundance of ureides [(ureide-N/ureide-N + NO3–N+ amino-N)x 100] in vacuum-extracted xylem exudate of soyabean(Glycine max [L.] Merr.) can be used to quantify N2 fixationactivity. We report experiments designed to (1) develop appropriateprocedures for sampling and storage of xylem exudate extractedfrom plants in the field and (2) develop a single plant, non-destructivesampling procedure, useful for evaluation of segregating breedingmaterial. Soyabean cultivars Bragg, Essex, Davis, Hardee andDodds were grown at three sites at Brecza, New South Wales,on a deep, alkaline vertisol. All plots were inoculated witheffective Bradyrhizobium japonicum CB 1809. Vacuum strengthsof between 50•5 and 80•8 kN m–2 did not affectthe relative abundance of ureides of xylem exudate. Delays betweenplant sampling and extraction of exudate, however, resultedin time-dependent increases in the relative ureide contents.Extraction should immediately follow shoot detachment to allowvalid comparison of treatments and accurate interpretation ofdata. The relative ureide contents of exudate showed only minordiurnal fluctuations. Exudate, once collected, could be stabilizedby dilution (1:1) in absolute ethanoL Both diluted and undilutedsamples were stable for at least 7 d at 4°C. Differencesduring early flowering, but not subsequently, in the compositionsof N solutes of exudate extracted from either the whole shoot, the top half or the lower half indicated the possibility ofnon-destructive sampling where seed, as well as xylem exudate, could be harvested. Best results in terms of volume of exudateextracted, treatment separation and seed production were obtainedwhen single plants were sampled during late flowering by detachmentof shoots at internode 5. Quantification of N2 fixation by pigeonpea (Cajanus cajan (L.)Millsp.) in the field has proved difficult using techniquessuch as 15N isotope dilution, acetylene reduction and N difference. We report experiments to develop the ureide assay of N2 fixationbased on extraction and analysis of xylem exudate. Plants ofpigeonpea cv. Quantum, inoculated with effective Rhizobium spp.CB756, were grown in a temperature-controlled glasshouse inlarge pots filled with a sand: vermiculite mixture, in waterculture and in a slightly acidic, red-brown earth in replicatedfield plots. Xylem exudate was collected as bleeding sap fromboth nodulated and unnodulated roots, and from detached nodules.Exudate was extracted also from detached shoots and stems ofpigeonpea using a mild vacuum (60–70 kN m–2). Largedifferences in the composition of N solutes exported from rootsof N2-dependent and nitrate-dependent plants suggested thatshifts in plant dependence on N2 fixation may be reflected byconcomitant changes in N solutes. Thus, nodulated plants weresupplied throughout growth with either N-free nutrients or nutrientssupplemented with 1, 2, 5, 5, 10, or 20 mol m–3 15. Plants were harvested at regular intervals fordry matter and vacuum-extracted exudate. The relative abundanceof ureides ([ureide-N/ureide-N + nitrate-N + -amino-N] ? 100)in the exudate was highly correlated with the proportion ofplant N (calculated using a 15N isotope dilution technique)derived from N2 fixation. Two distinct phases of plant growthwere recognized and standard curves were prepared for each. The relationship between proportional dependence of plants onN2 and xylem relative ureides was unaffected by mineral-N source,i.e. nitrate or ammonium. This result is discussed in relationto interpretation of material from field-grown plants. The effectsof plant genotype, strain of rhizobia, section of stem extracted, removal of leaves, time delay between shoot detachment and extraction, and diurnal characteristics were examined in order to identifypotential sources of error and to optimize sampling procedures. Nodulation, nitrogen (N2) fixation and xylem sap composition were examined in sand cultured plants of Bambara groundnut (Vigna subterranea L.) and Kersting's bean (Macrotyloma geocarpum L.) inoculated with Bradyrhizobium strain CB756 and supplied via the roots for a 4 week period from the third week onwards with different levels of (15N)-nitrate (0–15 mM). The separate contributions of nitrate and N2 to plant nitrogen were measured by isotope dilution. Increasing levels of nitrate inhibited nodule growth (measured as dry matter or nodule N) of both species parallel with decreased dependence on symbiotically-fixed N. Specific nodule activity (N2 fixed g nodule dry−1 d−1 of nodules) was reduced progressively with time in V. subterranea at higher (5 or 15 mM) levels of NO3, but this was not so for M. geocarpum. Root xylem bleeding sap of both species showed ureides (allantoin and allantoic acid) as predominant (>90%) solutes of nitrogen when plants were relying solely on atmospheric N. Levels of ureide and glutamine decreased and those of asparagine and nitrate in xylem increased with increasing level of applied nitrate. Relative levels of xylem ureide-N were positively correlated (R2=0.842 for M. geocarpum and 0.556 for V. subterranea), and the ratio of asparagine to glutamine in xylem exudate negatively correlated (R2=0.955 for M. geocarpum and 0.736 for V. subterranea) with plant reliance on nitrogen fixation. The data indicate that xylem sap analyses might be useful for indirect field assays of nitrogen fixation by the species and that Kersting's bean might offer some potential as a symbiosis in which N2 fixation is relatively tolerant of soil N.  相似文献

Seedlings of Ricinus communis L. were cultivated in quartz sandand supplied with media which contained either different concentrationsof nitrate or ammonium nitrogen and were treated with a lowsalt stress. The concentration of ABA was determined in tissuesand in xylem and phloem saps. Between 41 and 51 day after sowing, abscisic acid (ABA) flows between roots and shoots were modelled.Long-distance transport of ABA was not stimulated under conditionsof nitrate deficiency (0.2 mol m–3). However, when ammoniumwas given as the only N source (1.0 mol m–3), ABA transportin both xylem and phloem was increased significantly. Mild saltstress (40 mol m–3 NaCl) increased ABA transport in nitrate-fedplants, but not in ammonium-fed plants. The leaf conductancewas lowered by salt treatment with both nitrogen sources, butit was always lower in ammonium-fed compared to nitrate-fedplants. A negative correlation of leaf conductance to ABA levelsin leaves or flow in xylem was found only in comparison of ammonium-fedto nitrate-fed plants. Carbon (C) fixation and nitrogen (N) assimilation rates havebeen estimated from 14C and 15N techniques for a 12 month periodin a Scottish sea loch. The maximum rate of nitrogen assimilated(29.92 mmol N m–2 day–1) was in April at the mostseaward station; similar high rates were experienced duringMay at the other stations. Carbon fixation rates were maximal(488–4047 mg C m–2day–1) at the time of highphytoplankton biomass (maximum 8.3 mg m–3 chlorophylla) during May, whilst nitrate concentrations remained >0.7µ.mol l–1. C:N assimilation ratios suggest nitrogenlimitation only during the peak of the spring bloom, althoughat times nitrogen (nitrate and ammonium) concentration fellto 0.2 µmol l–1 in the following months. The verticalstability of the water column, influenced by tidal and riverineflushing, varied along the axis of the loch, resulting in markeddifferences between sampling stations. Although ammonium waspreferentially assimilated by phytoplankton, >50% of productionwas supported by nitrate uptake and only during the summer monthswas the assimilation of ammonium quantitatively important.  相似文献

Low temperature (6 C) growth was examined in two cultivarsof Vicia faba L. supplied with 4 and 20 mol m–3 N as nitrateor urea. Both cultivars showed similar growth responses to increasedapplied-N concentration regardless of N-form. Total leaf areaincreased, as did root, stem and leaf dry weight, total carboncontent and total nitrogen content. In contrast to findingsat higher growth temperatures, 20 mol m–3 urea-N gavesubstantially greater growth (all parameters measured) than20 mol m–3 nitrate-N. The increased carbon content per plant associated with increasedapplied nitrate or urea concentration, or with urea in comparisonto nitrate, was due to a greater leaf area per plant for CO2uptake and not an increased CO2, uptake per unit area, carbon, chlorophyll or dry weight, all of which either remained constantor decreased. Nitrate reductase activity was substantial inplants given nitrate but negligible in plants given urea. Neitherfree nitrate nor free urea contributed greatly to nitrogen levelsin plant tissues. It is concluded that there is no evidence for a restrictionin nitrate reduction at 6 C, and it is likely that urea givesgreater growth than nitrate because of greater rates of uptake. Xylem transport of nitrogen and phosphorus was examined in maturemycorrhizal plants of 41 species in 15 genera of Epacridaceaein native habitat in south-west Australia. Glutamine was theprincipal nitrogenous solute of xylem of all but four species. In the latter species, arginine or asparagine predominated.Nitrate and ammonium comprised minor fractions of xylem (tracheal)sap N, except in two species in which nitrate contributed overhalf of the N. Ratios of total-N:phosphate-P in xylem sap variedwidely (mean 67±18, range 0.2–495) between speciesand habitats. Plants ofCroninia kingiana (syn.Leucopogon kingeanus)from the one habitat showed higher levels of N and P in xylemearly than late in the mycorrhizal season, but there was noconsistent evidence of higher N and P levels from upper thandeeper parts of their root systems. Study of juvenile populationsof four species of epacrids indicated that substantial fractionsof the yearly increment of N, P and dry matter was accumulatedduring the three summer months when infected mycorrhizal hairroots were absent. Glasshouse culture of mycorrhizal plantsof Epacridaceae in habitat soil enriched with decomposed andleached double (13C,15N)-labelled dry matter of wheat showedsubstantial labelling of shoots with15N but not with13C. Plantsfed similarly treated15N-labelled root residues of maize acquired15Nbut failed to generate 13C values different from those of controlplants. Possible avenues of mycorrhizal and non-mycorrhizalnutrition of Epacridaceae are discussed. amino acids; mycorrhizal nutrition; xylem transport; south-west Australia; Epacridaceae; nitrogen; phosphorus  相似文献
