Low efficiency of embryo maturation, germination and conversion to plantlets is a major problem in many species including Persian walnut. We studied the effects of abscisic acid (ABA) and sucrose, on the maturation and germination of Persian walnut (Juglans regia) somatic embryos. Individual globular somatic embryos were grown on a maturation medium supplemented with different combinations of ABA and sucrose for ca. 1 month, until shoot meristems and radicles had developed. White and opaque embryos in late cotyledonary stage were subjected to desiccation after the culture period on maturation media. The number of germinated somatic embryos was influenced by the concentrations of ABA in the maturation medium. The best treatment for germination, in which both shoot and root were developed contained 2 mg l−1 ABA and resulted in 41% conversion of embryos into plantlets. Regeneration was reduced at higher levels of ABA. While ABA always reduced the rate of secondary embryogenesis, treatments containing 4.0% sucrose significantly increased the number of secondary embryos. On the other hand, sucrose had little influence on maturation. Normal and abnormal embryos were verified anatomically.  相似文献

Experiments were performed to determine the influence of maturation medium carbohydrate content on the rates of germination and plantlet conversion (root and shoot growth) of somatic embryos from four embryogenic lines derived from leaf or internode explants of Quercus robur L. seedlings. The conversion rate was favoured by high carbohydrate content as long as the maturation medium contained at least 2% sucrose, which was necessary for healthy embryo development. Given this, sorbitol and mannitol favoured the conversion rate more efficiently than sucrose, the highest rate, 32%, being achieved by medium with 6% sorbitol and 3% sucrose. Maturation treatment did not affect the root or shoot lengths of converted embryos. In supplementary experiments, 2 weeks of gibberellic acid treatment between maturation and germination treatments did not improve germination rates, but did reduce root length and the number of leaves per regenerated plantlet. In the four embryogenic lines tested, plant recovery rate was enhanced by inclusion of benzyladenine into the germination medium following culture of the embryos on maturation medium with 6% sorbitol and 2-3% sucrose. In embryogenic systems it is important to assess the uniformity of the regenerants. Random amplified polymorphic DNA (RAPD) analysis using 32 arbitrary oligonucleotide primers was performed to study variability in DNA sequences within and between four embryogenic lines. No intraclonal nor interclonal polymorphism was detected between embryogenic lines originating from different types of explant from the same seedling, but every one of the primers detected enough polymorphism among clones originating from different plants to allow these three origins to be distinguished. No differences in DNA sequences between regenerated plantlets and their somatic embryos of origin were detected, but a nodular callus line that had lost its embryogenic capacity was found to be mutant with respect to three other clones originating from the same plantlet. This study shows that high carbohydrate levels in the maturation medium significantly increase plant conversion of oak somatic embryos, which exhibit no variation in DNA sequences when proliferated by secondary embryogenesis.  相似文献

Commercial deployment of clonal trees via somatic embryogenesis (SE) could increase forest productivity over conventional tree breeding techniques. However, some technical advances need to be made to use SE in clonal forestry with Pinus radiata. For example, the conversion of embryonal mass (EM) into plants is at present a major bottleneck. For this reason, maturation experiments were carried out to determine the effect of the initial amount of EM, activated charcoal (AC) and the best combination of abscisic acid (ABA), sucrose and amino acid concentration in the maturation medium. Germination was evaluated on different media formulations with and without AC. When 100 mg of EM were suspended in liquid medium without AC, cotyledonary somatic embryos were obtained in all the maturation media tested. Maturation medium supplemented with 60 μM ABA, 6% sucrose, and embryo development medium amino acid mixture produced the highest number of cotyledonary somatic embryos, between 10 and 1,550 embryos per gram of EM fresh weight. Approximately half of the tested 25 lines produced more than 600 embryos per gFW. Embryo development was the best when somatic embryos were germinated in half strength modified Quoirin and Lepoivre medium supplemented with 2 g L−1 AC. This protocol simplified and improved SE maturation and germination due to the elimination of subcultures, the large number of somatic embryos obtained from a very low amount of EM, and the elimination of pre-germination treatments, resulting in a significant saving of cost and labor.  相似文献

The effect of gibberellic acid (GA3) was tested on germination of coconut zygotic embryos, their conversion into plantlets and ex vitro survival. There were four treatments consisting of 5 wk of culture in semi-solid medium or liquid medium, with or without GA3. Embryos were then transferred to GA3 free-liquid medium for the rest of a 32-wk culture. Germination and conversion percentages were higher in semi-solid medium than in liquid medium, and with both media percentages increased with GA3 treatment (with the exception of the highest GA3 concentration). Embryos of two varieties (MGD and MYD) were used. The following are the results with MGD embryos. Optimum GA3 concentration in liquid medium was 0.46 μM, with 80% germination (62% in the control without GA3) and 4.6 μM in semi-solid medium with 98% germination (71% in the control). With GA3 treatment, germination was also faster. Conversion in semi-solid medium with GA3 was 87% (60% in the control), and 45% in liquid medium with GA3 (25% in the control). Once the plantlets had at least three bifid leaves and three primary roots at the time of transfer to ex vitro, they survived independently of the treatment. When MYD embryos were used, germination and conversion percentages were higher in semi-solid medium than in liquid medium, and they increased when GA3 was used, although percentages were lower than those obtained with MGD embryos. The results showed that the use of GA3 benefited coconut embryos in culture because it favored germination and conversion to plants on semi-solid medium, and hence improved previous protocols.  相似文献

The effects of sucrose on maturation and of plant growth regulators on germination of soybean somatic embryos were investigated for the purpose of developing an efficient culture method for plant recovery. Somatic embryos produced on medium with a low sucrose concentration (5 gl-1), less than 1 mm in length, 0.6 mg in fresh weight, and green in color, were grown for 2 weeks on MS medium containing 5 gl-1 or 30 gl-1 sucrose and then for another 5 weeks on MS medium containing 5–90 gl-1 sucrose. The highest increase in fresh weight of somatic embryos was obtained in the treatment of transferring from 30 gl-1 sucrose (2 weeks) to 60 gl-1 (5 weeks). With the increase in fresh weight, the somatic embryos gradually changed color from green to yellow, and finally to white, when they stopped growth. Soybean seed storage proteins (-conglycinin and glycinin) were accumulated in somatic embryos under tissue specific and stage specific control analogous to that in zygotic embryos. Exogenous gibberellic acid was effective in promoting precocious germination of premature soybean somatic embryos, but was not necessary for the germination of mature somatic embryos. For the mass production of chestnut trees with selected, hybrid, or genetically engineered genotypes, one potentially desirable propagation strategy is based on somatic embryogenesis. Although methods exist for the initiation of embryogenic cultures of Castanea sativa from immature zygotic embryos or leaf explants, the embryos produced have had low rates of conversion into plantlets. This study explored the possible benefits for somatic embryos that have already undergone maturation and cold treatments, of (a) partial slow or fast desiccation, and (b) of the addition of plant growth regulators or glutamine to the germination medium. Germination response was evaluated in terms of both conversions to plantlets and through embryos developing only shoots (shoot germination) that could be rooted following the micropropagation protocols developed for chestnut. Two or 3 wk slow desiccation in sealed empty Petri dishes resulted in a slight reduction in water content that nevertheless increased total potential plant recovery, shoot length, and the number of leaves per plantlet. However, best results were achieved by 2 h fast drying in a laminar flow hood, which reduced embryo moisture content to 57–58% and enhanced the potential plant recovery and quality of regenerated plantlets. Plant yield was also promoted by addition of 0.44 μM benzyladenine and 200–438 mg/l of glutamine to the germination medium, and plantlet quality (as evidenced by root, shoot, and leaf growth) by the further addition of 0.49 μM indole-3-butyric acid.  相似文献

Summary This study reports a protocol for germination of Givotia rottleriformis (var. Tel. Thella Poniki) using zygotic embryo culture. A 100% germination was obtained by culturing the embryos on Murashige and Skoog medium containing 30 gl−1 sucrose. A sucrose concentration lower or higher than 30 gl−1 resulted in lower germination or promoted callus formation. The seedling growth was promoted by the addition of 100 mgl−1 tyrosine in the medium. Seedlings germinated in the presence of 0.2–0.4 mgl−1 α-naphthaleneacetic acid and 0.3–0.5 mgl−1 indole-3-butyric acid were abnormal, showing a slender stem with slender roots or forming callus with stout roots. Germination also affected embryo orientation in culture; placing embryos upright on the medium was most beneficial for germination. The in vitro-germinated seedlings were acclimatized in soil under shady conditions with a survival rate of 60–70%. These plants were phenotypically normal, healthy, and similar to donor plants. This protocol will be useful for overcoming seed dormancy and for rapid multiplication and conservation of G. rottleriformis using zygotic embryo culture.  相似文献

该研究以蔗糖、麦芽糖、山梨醇及PEG(6000)为渗透剂,探讨了不同渗透剂对白刺花体细胞胚发育、胚成熟及萌发的影响。结果表明:白刺花下胚轴形成的胚性愈伤组织接种至MS+2,4-D 0.2 mg·L~(-1)+NAA 1.0 mg·L~(-1)+6-BA 2.0 mg·L~(-1)+TDZ 1.0 mg·L~(-1)+蔗糖40 g·L~(-1)+谷氨酰胺100 mg·L~(-1)+植物凝胶3g·L~(-1)的培养基上,体细胞胚发生率高达66. 21%,总胚数为79个; 7%蔗糖可使体细胞胚成熟率高达64.36%,同时也可提高多子叶畸形胚形成; 2%麦芽糖+2%山梨醇+4%蔗糖组合使体细胞胚成熟率最高达88.89%,畸形胚比例最低; 30 g·L~(-1)PEG培养时,体细胞成熟率最高,为82.35%;鱼雷期的体细胞胚最合适转接,可使体胚萌发率达90.58%,复合糖上培养得到的成熟体细胞胚生根率最高,为87.47%。这为实现白刺花体细胞胚育苗奠定了理论基础,并提供了可行的方案。  相似文献

The effects of amino acids, abscisic acid (ABA), polyethylene glycol (PEG), and elevated sucrose were tested on the maturation and germination of American chestnut (Castanea dentata) somatic embryos. Somatic embryos from three lines were matured over an eight week period through a two-stage process. After maturation, somatic embryos were randomly divided into three groups to measure dry weight/ fresh weight ratios, starch levels, and germination rates. Prior to transfer to germination medium, somatic embryos received a four week cold treatment. While some treatments with amino acids, elevated sucrose, PEG or ABA increased either dry weight/fresh weight ratios, starch content or both, only addition of 25mM L-asparagine significantly increased germination rate and taproot length, and this response was only obtained with one of the three lines tested. Six plants survived the transfer to potting mix, acclimatization to greenhouse conditions and field planting.  相似文献

Embryogenic masses (EMSes) of pistachio (Pistacia vera L.) were proliferated in liquid Murashige and Skoog (MS) medium without growth regulators. To determine the effects of benzylaminopurine (BAP), racemic (±) abscisic acid (ABA) and sucrose treatments during maturation on the subsequent germination and plantlet regeneration, clusters of mature somatic embryos were transferred from maturation medium onto the surface of 0.7% agar-solidified Murashige and Skoog medium. Neither germination nor plantlet development medium contained BAP or ABA. Germination studies were carried out using 80 somatic embryos at every combination of four sucrose concentrations, three maturation periods and either five concentrations of BAP or four of ABA, and the numbers germinating were recorded after four durations of culture. A similar experimental plan was used to study plantlet regeneration. The number of germinated somatic embryos increased markedly with duration of the culture on germination medium, and was influenced by the concentrations of BAP or ABA in the maturation medium; the concentration of sucrose in this medium had little influence. Plantlet regeneration also increased with culture duration and was reduced at the highest levels of BAP or ABA; with ABA, the probability of plantlet regeneration was lower for longer maturation periods. ABA and BAP have similar effects on somatic embryo germination (except at the highest levels used), but BAP is superior to ABA for promoting subsequent plantlet regeneration. Linear logistic models were used to investigate the significance of the treatments, and to estimate the optimum conditions for germination and plantlet regeneration. 为探求促进广金钱草萌发及生长的有效途径,该文使用不同浓度的费氏中华根瘤菌、放射型根瘤菌以及田菁茎瘤固氮根瘤菌菌液分别浸染广金钱草种子,以蒸馏水浸泡种子为空白处理,研究其对广金钱草种子萌发指标(发芽率、发芽势、发芽指数、活力指数)的影响,并在温室培养40、60、80 d后分别测定广金钱草幼苗的生长指标(叶片数、分枝数、株高)及部分生理指标(叶绿素含量、含氮量)的变化。结果表明:(1)在1×107 CFU·mL-1浓度条件下,三种菌液浸种均获得最大萌发指标,其中田菁茎瘤固氮根瘤菌实验组效果最显著,发芽率、发芽势、发芽指数和活力指数高于对照组16.00%、9.33%、9.51和41.34%。(2)除田菁茎瘤固氮根瘤菌实验组外,其余实验组的广金钱草幼苗叶片数、分枝数及株高均低于对照组。(3)放射型根瘤菌实验组和田菁茎瘤固氮根瘤菌实验组幼苗叶绿素含量相比对照组分别增加1.47%和7.47%,含氮量分别增加0.57%和5.17%。综上所述,三种根瘤菌菌液浸种均可在不同程度上提高广金钱草种子的发芽能力和植株生长期的叶绿素和含氮量,其中田菁茎瘤固氮根瘤菌具有最大正向影响,可有效提高种子萌发能力并促进植株生长。该研究可为不同地区广金钱草的种植和栽培提供技术指导,为开发田菁茎瘤固氮根瘤菌作为植物促生长菌剂提供理论支持。  相似文献

The present study evaluated the effects of chilling, partial desiccation, cotyledon excision and successive subculture of microspore-derived embryos on plant development in oilseed rape (Brassica napus L.). The results showed that out of the five media, all the genotypes showed the best response when the embryos were cultured on the half-strength Murashige and Skoog medium with 2.0 mg dm−3 benzylaminopurine. A cold treatment for 3 or 5 d further increased frequencies of embryo germination (90.0 %) and plantlet development (58.46 %). Desiccation for one day also increased the embryo germination and plantlet development in all genotypes tested. Cutting the cotyledons of the embryos at late cotyledonary stage significantly increased the frequency of plantlet development. The highest rate of plantlet development was obtained from cultures of embryos sampled with size of less than 4.0 mm. The successive subculture further improved the germination and development of plantlets from embryos. In the genotype ZJU452, the rate of plantlet development reached 99.78 % after the second subculture of embryos.  相似文献

山玉兰(Magnolia delavayi)是木兰科木兰属的常绿乔木或大型灌木,是重要的园林造景、庭院绿化素材,也是重要的育种资源。山玉兰花粉的研究对其杂交育种的成败具有重要影响,但目前尚未见其花粉活力的相关报道。该研究以新鲜的山玉兰花粉为对象,采用悬滴培养法分析了温度、光照以及培养液的蔗糖和硼酸浓度对山玉兰花粉萌发的影响。结果表明:(1)山玉兰花粉萌发时,最适宜的温度为27℃。(2)光暗条件下,山玉兰花粉以浓度为5%的蔗糖培养效果最佳,其萌发率在16%以上;而硼酸浓度则以0.001%的培养效果最佳。(3)蔗糖与硼酸共同作用可有效促进花粉萌发和花粉管生长。其中,在光照条件下,以5.0%蔗糖+0.001%硼酸为最适宜的培养液,花粉萌发率达41.27%,花粉管长达281.49μm;而在黑暗条件下,则以5.0%蔗糖+0.01%硼酸为最适宜的培养液,花粉萌发率达45.71%,花粉管长达254.00μm。该研究结果为进一步开展人工辅助授粉、发掘山玉兰的种质资源工作奠定了基础。  相似文献

恶劣环境下,人工海防林因面临养分胁迫而经营困难。为探讨盐、磷胁迫对主要海防林树种木麻黄和台湾相思种子萌发及生长的影响,该研究分别用不同浓度的NaCl(盐)和KH2PO4(磷)溶液处理种子和浇灌幼苗,测定种子萌发和幼苗生长指标。结果表明:(1)高盐胁迫显著抑制种子萌发,对幼苗生长有一定影响,但两种植物影响程度不同;台湾相思种子萌发耐盐性高于木麻黄,前者相对盐害率最大值为23.03%,后者为89.15%;随着盐浓度增加,木麻黄和台湾相思种子的发芽率、发芽势、发芽指数和活力指数均降低,对应最大值分别为38.70%、34.67%、18.70、0.055和76.67%、62.22%、48.46、6.11。(2)两种植物的株高和根长随盐浓度增加而降低,木麻黄和台湾相思株高分别为12.29~6.01 mm和48.27~17.33 mm,根长分别为8.57~1.45 mm和33.41~5.88 mm;台湾相思根、茎、叶生物量及根冠比均随盐浓度的增加逐渐减小,木麻黄各处理差异较小。(3)台湾相思的种子和幼苗较木麻黄更耐低磷环境,二者最适磷浓度存在差异;木麻黄种...  相似文献

The effect of various abiotic stresses on germination rate, growth and soluble sugar content in Sorghum bicolor (L.) Moench cv. CSH 6 seed embryos and endosperm during early germination was investigated. Under stress conditions germination, water potential and tissue water content decreased markedly. Subsequently, this reduction resulted in marked decreases in fresh weight both in embryos and endosperm. Conversely, a substantial increase in dry weight was observed. Furthermore, a considerable increase in the sugar contents in both embryo and endosperm was detected. The fructose level was always higher than glucose and sucrose in response to various stresses. However, as compared to the control the level of glucose and sucrose was higher in embryos and endosperm after stress treatments. Based upon these results a possible physiological role of sugars in the germination of sorghum seeds is discussed.  相似文献

火干扰不仅影响森林生态系统的结构与功能,而且影响群落更新与演替。为了研究草木灰对4种松属种子萌芽和幼苗生长的影响,在湖南省株洲市黄丰桥林场采集马尾松、云南松、湿地松和火炬松种子,以马尾松次生林火烧迹地的草木灰为基质,设置对照和3个草木灰处理,进行播种试验及生长观测。结果表明:(1)草木灰处理下4个物种的种子发芽率比对照试验低,并呈现随草木灰含量的增加而降低的趋势,低草木灰含量处理马尾松种子发芽率不及对照一半,中、高草木灰含量处理发芽率更低;云南松、湿地松和火炬松种子发芽率规律类似于马尾松,而高草木灰含量处理发芽率为0。(2)培养皿播种平均发芽时间比花盆播种长,无论草木灰处理还是培养条件(培养皿或花盆),马尾松和云南松种子平均发芽时间比湿地松和火炬松短。(3)4个物种种子播种14周后,对照试验中马尾松和云南松幼苗死亡率最低,而火炬松和湿地松幼苗死亡率相对增加。(4)播种14周后不同试验处理4个物种幼苗生长差异不明显。火炬松、湿地松、云南松和马尾松幼苗的单株平均干重分别达0.038、0.031、0.027、0.024 g。各物种叶干重按降序排列依次是火炬松(0.026 g)湿地松(0.019 g)云南松(0.017 g)马尾松(0.016 g),根和茎的干重比重相当,约占对应物种幼苗叶干重的三分之一。研究表明以草木灰为培养基质,马尾松和云南松种子的发芽率较高,平均发芽时间较短,更适合作为火烧迹地植被恢复先锋树种,为火干扰后选择人工促进天然更新树种提供参考。  相似文献

Explants of germinating zygotic embryos of Eleutherococcus sessiliflorus,an important medicinal plant, produced somatic embryos directly on Murashige and Skoog (MS) medium with 4.5 M 2,4-D. In addition, embryogenic callus formed at a low frequency (less than 7%) from hypocotyl segments after prolonged culture. High frequency somatic embryogenesis was obtained through cell suspension culture after the cells were transferred to medium lacking 2,4-D. Maturation and germination of embryos was influenced by the sucrose concentration of the medium. At a low concentration of sucrose (1%), maturation and germination of embryos occurred readily. At over 6% sucrose, somatic embryos did not germinate although this could be overcome by GA3 treatment. Cold treatment during acclimatization after transfer to soil enhanced survival. Surviving plantlets produced new sprouts after overwintering in the field.  相似文献

Somatic embryos of three Japanese pines, Pinus thunberghii, P. densiflora, and P. armandii var. amamiana, were subjected to different post-maturation treatments to improve both germination and plant conversion frequencies. Slow desiccation of somatic embryos at high relative humidity resulted not only in a marked increment in germination frequencies but also subsequently improved plant conversion rates in all genotypes of the three species tested. Overall three species, germination and plant conversion frequency of somatic embryos was improved by more than fourfold (19?C81%) and more than fivefold (15?C77%), respectively, compared to those of untreated control. In addition, this treatment resulted in a considerable improvement of synchronization of the germination period. The time required for root emergence was synchronized over a half period (28?C14?days) compared with the control. Somatic plants were acclimatized and their growth was monitored in the field.  相似文献

麻疯树因其种子含油率较高,种子油提炼的生物柴油可部分替代汽油,而成为一种极具潜能的能源作物,但由于产量低,麻疯树在热带、亚热带的发展受到极大限制。杂交育种是提高产量的重要手段,杂交亲本花粉生活力的高低直接影响到育种的成效。因此,寻求麻疯树离体花粉萌发的最适培养基配方,探明花粉萌发培养基中各主要培养基成分间的交互作用对生产上麻疯树杂交结实率和种子产量的提高具有重要意义。该研究以麻疯树开花初期雄花上花药刚散粉时的成熟花粉粒为材料,采用Box-Behnken设计(Box-Behnken design,BBD)的响应面法,对麻疯树花粉离体萌发培养基中各主要培养基成分的浓度配比及各主要培养基成分的交互作用进行了研究。以花粉萌发率为响应指标,建立了4种营养成分(蔗糖、硼酸、硝酸钙、硝酸钾)与花粉萌发率的响应面模型,并对各主要培养基成分的浓度配比进行了优化。通过R软件进行响应面分析的结果表明:4因素对花粉萌发率的影响顺序为蔗糖硼酸硝酸钙硝酸钾;蔗糖与硼酸、蔗糖与硝酸钙、蔗糖与硝酸钾之间的交互作用显著。响应面建模优化后的最佳培养基为13.77%蔗糖+32.14 mg·L~(-1)硼酸+22.21 mg·L~(-1)硝酸钙+19.95 mg·L~(-1)硝酸钾+200 mg·L~(-1)硫酸镁,在此条件下的理论萌发率为99.73%。采用此培养基成分配比得到麻疯树花粉离体试验萌发率为98.97%,与理论响应值相吻合,同时也表明利用BBD设计的响应面模型进行麻疯树花粉离体萌发培养条件优化方法的有效性。  相似文献

Experiments were performed to determine the influence of proliferation medium on the maintenance of embryogenic competence and on repetitive embryogenesis in Castanea sativa Mill. somatic embryos derived from leaf explants. Somatic embryo proliferation was carried out by both direct secondary embryogenesis and by the culture of nodular callus tissue originated from cotyledons of somatic embryos. Both systems led to the production of cotyledonary somatic embryos on Murashige and Skoog proliferation medium supplemented with 0.1 mg l-1 benzyladenine and 0.1 mg l-1 naphthaleneacetic acid. Carbon source and concentration had a marked influence on maturation and subsequent germination ability of chestnut somatic embryos. Plantlet conversion was achieved in embryos matured on media with 6 % sucrose, and on 3 or 6 % maltose, whereas mean shoot length, root length and leaf number of produced plants were not significantly affected by these maturation media. Overall, the best results were obtained with 3 % maltose-matured somatic embryos, giving rise to 6 % plant recovery in addition to 33 % of embryos exhibiting only shoot development. The application of a 2-month cold treatment at 4 degrees C to somatic embryos matured on medium with 3 % maltose was necessary for achieving plant conversion, while partial desiccation did not appear to influence this response. A total of 39 % of embryos eventually produced plants either through conversion to plantlets or indirectly through rooting of shoots. Shoots formed by somatic embryos could be excised, multiplied and rooted following the micropropagation procedures previously developed for chestnut.  相似文献
