Young seedlings ofGmelina arborea Roxb. were subjected to 2 weeks of drought. Despite the gradual reduction in stomatal conductance, leaf and root growth was not affected until the later part of the stress period. This was attributed to solute adjustment in the roots of the plants. As the severity of water stress increased, root growth was prolific in all the soil segments. As a result, water in the lowest soil segment was used to maintain plant turgor, which in turn sustains the leaf and root growth during the water-stress treatment. The influence of soil water content and soil water potential upon soil water uptake rate was also evaluated on soil profile basis. Rates of extraction began to decline in all soil segments as soon as soil water potential fell below -0.06 MPa, presumably as a result of vapour gaps between the root and soil (root: soil interface resistance). It is suggested that the growth of roots ofGmelina plants away from drying soil will minimize the resistance to water uptake.  相似文献

Summary Sugarcane (Saccharum spp. hybrid cv. CP 84-1198) embryogenic calluses were induced from young leaves cultured on modified Murashige and Skoog basal medium supplemented with 13.6 μM 2,4-dichlorophenoxyacetic acid. Five concentrations, 0.5, 1.0, 2.5, 5.0, and 10.0 μM, of five different growth regulators, 6-benzylaminopurine, kinetin, 6-γ,γ-(dimethylallylamino)purine, zeatin, and thidiazuron, were tested with or without 22.5 μM α-naphthaleneacetic acid to compare their ability to induce regeneration from embryogenic callus. After 4 wk on medium, the percentage of shoot meristem induction was evaluated, and after 10 wk the total number of shoots produced, as well as the percentage of shoots greater than 1 cm in length, was obtained. Although it had the lowest percentage of elongated shoots, medium containing thidiazuron alone performed better than all other growth regulators tested, with the highest percentage of shoot induction and the largest number of shoots, particularly at a concentration of 2.5 μM.  相似文献

Effect of various growth regulators on growth in vitro of cherry shoot tips   总被引:5,自引:0,他引:5  
An investigation was undertaken to examine the effects of nine different growth regulators on growth in vitro of shoot cultures of the semi-dwarfing cherry rootstock Colt (Prunus avium × P. pseudocerasus). The effects of each supplement on shoot extension and proliferation and also leaf and callus production were noted, and it was found that BAP has the ability to proliferate shoots, IBA nullifies this effect and that kinetin, ABA, GA3 and ethylene inhibit the growth of colt cultures. Conditions were established which resulted in a) optimum shoot growth prior to subsequent rooting or grafting; b) maximum shoot proliferation for rapid clonal multiplication and c) minimum shoot growth. This study will form the basis of an investigation into germplasm conservation of temperate fruit trees by both cryogenic storage and minimal growth techniques.  相似文献

The effect of growth regulators on induction of androgenesis in coconut was investigated using seven different growth regulators at various concentrations and combinations. Three auxins (1-naphthalene acetic acid—NAA, indoleacetic acid—IAA, picloram) and three cytokinins (2-isopentyl adenine-2-iP, kinetin, zeatin) were tested either alone or in combination with 2,4-dichlorophenoxyacetic acid (2,4-D), using modified Eeuwens Y3 liquid medium as the basal medium. Among the tested auxins, 100 μM NAA in combination with 100 μM 2,4-D enhanced the production of calli/embryos (123) whereas IAA and picloram showed negative and detrimental effects, respectively, for androgenesis induction over 100 μM 2,4-D alone. Kinetin and 2-iP enhanced the production of calli/embryos when 100 μM 2,4-D was present in the culture medium. Both cytokinins at 10 μM yielded the highest frequencies of embryos (113 and 93, respectively) whereas zeatin (1 or 2.5 μM) had no impact on microspore embryogenesis. When calli/embryos (produced from different treatments in different experiments) were sub-cultured in somatic embryo induction medium (Y3 medium containing 66 μM 2,4-D), followed by maturation medium (Y3 medium without growth regulators) and germination medium (Y3 medium containing 5 μM-6-benzyladenine—BA and 0.35 μM gibberellic acid—GA3), plantlets were regenerated at low frequencies (in most treatments ranging from 0% to 7%).  相似文献

The effects of some growth regulators on the dark-germinating seeds ofCucumis anguria have been investigated. GA3, 6-BA and ethrel strongly promoted the germination of seeds kept in white light. GA3 had no effect in counteracting the far red effect, but did counteract partially the blue light. Ethrel partially counteracted both the far red and blue light effect. No interaction could be found between red light and GA3 or 6-BA.  相似文献

Stem internodes with axillary buds were excised from 5-year old trees ofFicus benjamina cv. Exotica. The effect of 6-benzylaminopurine (BAP), gibberellic acid (GA3), indole-3-acetic acid (IAA), naphthaleneacetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D) on shoot growth and proliferationin vitro was investigated. Multiple shoots were developed after 3–4 weeks from stem internodes with axillary buds incubated in Murashige and Skoog (MS) medium supplemented with phloroglucinol (PG) and BAP. Optimum shoot proliferation took place in the presence of 1.0 mg l−1 BAP. Shoots obtained could be elongated in a medium with 0.5 mg l−1 GA3 prior to their rooting. The root initiation was successfully induced on MS medium either with IAA at 0.5–0.1 mg l−1 or in plant growth regulator-free medium. All rooted plantlets were subsequently transferred to a peat, humus and perlite mixture in a culture room with high humidity and covered with plastic bags. Three closely related growth regulators (G) that are present in some myrtaceous plants were examined for possible anti-transpirant effects. The bioassay material involved cuttings of mung bean and Eucalyptus rupicola. Stomatal resistance was determined by a diffusion porometer. Dormant seeds in permanent soil seed banks change their germination probability with age, but little is known about the causes of this behavior. Since current adaptive models do not consider the age structure of the seed bank, the sole explanation is that changes in germination probability with age are due to passive accidental factors or to decay of seeds. Here we present a theory, which shows that such changes might be adaptive. In particular, in density-regulated populations, the evolutionarily stable fraction of germinating seeds in each age class increases with age whenever age-dependent mortality of dormant seeds remains constant or increases with age.  相似文献

Echinochloa colona regeneration via organogenesis in callus cultures derived from leaf base and mesocotyl expiants andin vitro flowering were achived. Shoot bud regeneration was achieved on Murashige and Skoog’s (MS) basal medium supplemented with 6.66 μM 6-benzylaminopurine (BAP), 2.68 μM 1-naphthalene acetic acid (NAA) and 3 % (m/v) saccharose. Regenerated shoots were rooted on half strength basal MS medium with 2 % (m/v) saccharose devoid of growth regulators. About 90 -95 % of rooted plantlets survived in the greenhouse.In vitro flowering was induced in the regenerated shoots derived from callus on half strength MS medium supplemented with 4.4 μM BAP, 74.07 μM adeninesulphate, 0.72 μM gibberellic acid, and 3 % (m/v) saccharose. The frequency ofin vitro flowering was 80 – 90 % in three repeated experiments. Fertile seeds were recovered fromin vitro grown plantlets which were subsequently germinated into plants. Changes in soluble protein profile and chlorophyll (Ch1) content in detached soybean leaves incubated in darkness or light were delayed by application of benzyladenine or indole-3-acetic acid and enhanced by abscisic acid. The degradation in light differed significantly from the degradation in darkness. Chl and proteins were lost at a higher rate in darkness than in light.  相似文献

The effects of an amino acid mixture and of plant growth regulators added to the FHG barley anther culture medium were examined using three barley cultivars (Cadette, Léger, and Igri) grown in two environments (growth cabinet and glasshouse). ‘Léger’ and ‘Igri’ were known as responsive, and ‘Cadette’ as recalcitrant to androgenesis. Our first experiment showed that the amino acid-supplemented medium was best for embryogenesis and regeneration of ‘Cadette’ and ‘Igri’ in both environments, and if ‘Léger’ in the growth cabinet. The addition of ABA and TDZ did not improve embryogenesis and plant regeneration, and PAA decreased them in the growth cabinet. The addition of the amino acid mixture in the FHG medium also reduced the percentage of albino plants in the growth cabinet, but growth regulators did not improve the percentage of albino plants, and in some cases increased it. In the growth cabinet, disregarding media, ‘Léger’ produced more embryos than ‘Cadette’ and ‘Igri’, and Léger' and ‘Igri’ produced more green plants than ‘Cadette‘. Percentages of albino plants were higher or ‘Cadette’ than for ‘Igri’ or ‘Léger’. In a second experiment, we compared seven hybrids with their parents for androgenic responsiveness. Hybrids had a higher ability to generate green plants than expected based upon the weighted average reflecting the contribution of each parent. The intensification of lipid free radical oxidation processes and active oxygen spicies accumulation in roots and leaves of corn seedlings has been studied by chemiluminescence method. 10-days exposition on 0.1 M NaCl decreased their growth and influenced on peroxidation intensivity. The maximum of H2O2-initiated chemiluminescence in roots was noted at 9th hour whereas in leaves it was demonstrated at 48th hour of salt exposition. The effect of seeds treatment by synthetic growth regulators methiur and ivin on seedlings growth and peroxidation processes in seedlings has been studied. Under salt stress condition both substances intensified seedling growth and decreased initiated chemiluminescence level. Antioxidative effect of methiur has been more pronounced in roots whereas ivin one in leaves. Practical using of these growth regulators for salt-tolerance increasing of plants has been suggested.  相似文献

The use of the synthetic auxin 2,4-dichlorophenoxyacetic acid (2,4-d) has played an important role in the production and maintenance of totipotent cereal callus. However, 2,4-d has been implicated in the loss of totipotency from barley callus. To examine the effect of 2,4-d on barley callus, regenerability and karyotype were examined over time as influenced by cultivar differences and 2,4-d levels, during a period in which initially vigorous plant regeneration typically declines dramatically. Higher (20.4–27.1 M) versus lower (6.8–13.6 M) concentrations of 2,4-d were positively associated with the number of green plantlets recovered from calli maintained for 10 and 16 weeks before transfer to regeneration media, and with the longevity of regenerability. There was a positive relationship between 2,4-d concentration and normal karyotype. We also investigated the use of phenylacetic acid for the initiation of regenerable barley callus. The effects of plant growth regulators on somatic embryogenesis were studied in leaf cultures of Coffea canephora. The maximum number of somatic embryos were obtained on media that contained only cytokinin as a plant growth regulator. All of the auxins tested (NAA, IBA, IAA and 2, 4-D) inhibited the formation of embryos. The optimal concentration of each cytokinin (2-iP, BA and kinetin) for somatic embryogenesis was 5 M. Under optimal conditions, each explant formed more than 100 embryoids with little callus and few adventitious roots. Embryoids were formed only at the cut edges of the leaf discs. Effects of the growth regulators epibrassinolide-694 (EB), gibberellic acid (GA), and abscisic acid (ABA) on the ATP-dependent translocation of H+ through the membranes of plasma membrane vesicles of potato (Solanum tuberosum L.) tuber cells were studied. The ATP-dependent accumulation of H+ in the plasma membrane vesicles from dormant tubers was inhibited by EB and ABA and stimulated by GA. After the break of dormancy, the stimulatory effect of GA increased, the inhibitory effect of ABA decreased, and EB stimulated the accumulation of H+ in the vesicles. The data suggest that the plasma membrane H+ ATPase is a target of phytohormones that regulate the dormancy of potato tubers.  相似文献

Leaves from 14-d-old Capsicum annuum L. cv. Anaheim seedlings were cultured on Murashige and Skoog (MS) medium containing different combinations of indole-3-acetic acid (IAA) and 6-benzyladenine (BA). After 3 months, cultures were transferred to new medium where BA was replaced with 9 μM isopentenyladenine (2iP) to enhance the growth of shoot buds. Developing shoots were elongated and rooted on MS medium enriched with 9 μM indole-3-butyric acid (IBA). All cultures were maintained in 250 cm3 baby jars covered with a clear polypropylene lid with or without microporous polypropylene membrane. Vessel type and plant growth regulators significantly affected callus morphogenic appearance, organogenesis and in vitro plantlet growth. Ventilated vessels supported photomixotrophic culture and improved regeneration and growth of plantlets. Higher plantlet dry mass and content of photosynthetic pigments, and lower stomatal density of plantlets grown in ventilated than in non-ventilated vessels facilitated ex vitro acclimation and growth.  相似文献
