Four anthocyanins, cyanidin 3-O-(2″-(5?-(E-p-coumaroyl)-β-apiofuranosyl)-β-xylopyranoside)-5-O-β-glucopyranoside, cyanidin 3-O-(2″-(5?-(E-p-coumaroyl)-β-apiofuranosyl)-β-xylopyranoside), cyanidin 3-O-(2″-(5?-(E-caffeoyl)-β-apiofuranosyl)-β-xylopyranoside) and cyanidin 3-O-(2″-(5?-(E-feroyl)-β-apiofuranosyl)-β-xylopyranoside) were isolated from leaves of African milk bush, (Synadeniumgrantii Hook, Euphorbiaceae) together with the known cyanidin 3-O-β-xylopyranoside-5-O-β-glucopyranoside and cyanidin 3-O-β-xyloside. The four former pigments are the first reported anthocyanins containing the monosaccharide apiose, and the three 5?-cinnamoyl derivative-2″-(β-apiosyl)-β-xyloside subunits have previously not been reported for any compound.  相似文献

A novel acylated cyanidin 3-sambubioside-5-glucoside was isolated from the purple-violet flowers of Matthiola longipetala subsp. bicornis (Sm) P. W. Ball. (family: Brassicaceae), and determined to be cyanidin 3-O-[2-O-(2-O-(trans-feruloyl)-β-xylopyranosyl)-6-O-(trans-feruloyl)-β-glucopyranoside]-5-O-[6-O-(malonyl)-β-glucopyranoside] by chemical and spectroscopic methods. In addition, two known acylated cyanidin 3-sambubioside-5-glucosides, cyanidin 3-O-[2-O-(2-O-(trans-sinapoyl)-β-xylopyranosyl)-6-O-(trans-feruloyl)-β-glucopyranoside]-5-O-[6-O-(malonyl)-β-glucopyranoside] and cyanidin 3-O-[2-O-(β-xylopyranosyl)-6-O-(trans-feruloyl)-β-glucopyranoside]-5-O-[6-O-(malonyl)-β-glucopyranoside] were identified in the flowers.  相似文献

The structures of the peracetylated derivatives of the following alditols obtained from oligosaccharides of human milk have been established by two-dimensional, J-resolved and J-correlated, 1H-n.m.r. spectroscopy at 360 MHz: β- d-Galp-(1→3)-β- d-GlcpNAc-(1→3)-β- d-Galp-(1→4)- d-Glc-ol, α- l-Fucp-(1→2)-β- d-Galp-(1→3)-β- d-GlcpNAc-(1→3)-β- d-Galp-(1→4)- d-Glc-ol, and β- d-Galp-(1→3)-β- d-GlcpNAc-(1→3)-[β- d-Galp-(1→4)-β- d-GlcpNAc-(1→6)]-β- d-Galp-(1→4)- d-Glc-ol.  相似文献

A group of 1-(4-methane(amino)sulfonylphenyl)-5-(4-substituted-aminomethylphenyl)-3-trifluoromethyl-1H-pyrazoles (12af) was synthesized and evaluated as anti-inflammatory agents. While all the compounds (20 mg/kg) showed significant anti-inflammatory activity after 3 h of inflammation induction (69–89%) as compared to celecoxib (80%), 1-(4-methanesulfonylphenyl)-5-(4-methylaminomethylphenyl)-3-trifluoromethyl-1H-pyrazole (12a) was found to be the most effective one (89%). The synthesis of model hybrid nitric oxide donor N-diazen-1-ium-1,2-diolate derivatives of 1-(4-methanesulfonylphenyl)-5-(4-substituted-aminomethylphenyl)-3-trifluoromethyl-1H-pyrazoles (10af) requires further investigation since the reaction of N-(4-(1-(4-(methylsulfonyl)phenyl)-3-(trifluoromethyl)-1H-pyrazol-5-yl)benzyl)ethanamine (12b) or 1-(4-(1-(4-(methylsulfonyl)phenyl)-3-(trifluoromethyl)-1H-pyrazol-5-yl)benzyl)piperazine (12c) with nitric oxide furnished N-nitroso derivatives (13 and 14), respectively, rather than the desired N-diazen-1-ium-1,2-diolate derivatives (10b and 10c).  相似文献

Abstract: To search for new and bioactive compounds from traditional Chinese medicines, a new glycoside, 3-O-[α- L -rhamnopyranosyl-(1→3)-( n -butyl-β- D -glucopyranosiduronate)]-28-O-β- D -glucopyranosyloleanolic acid ( 1 ), was isolated from the roots of Cyathula officinalis Kuan, along with 3-O-(methyl-β- D -glucopyranosiduronate)-28-O-β- D -glucopyranosyl oleanolic acid ( 2 ), 3-O-β- D -glucopyranosyl oleanolic acid ( 3 ), 3-O-β- D -glucuronopyranosyl oleanolic acid ( 4 ), 3-O-[β- L -rhamnopyranosyl-(1→3)-(β- D -glucuronopyranosyl)] oleanolic acid ( 5 ), 3-O-(β- D -glucuronopyranosyl)-28-O-β- D -glucopyranosyl oleanolic acid ( 6 ), 28-O-β- D -glucuronopyranosyl-(1→4)-β- D -glucopyranosyl hederagenin ( 7 ), 3-O-[β- L -rhamnopyranosyl-(1→3)-β- D -glucuronopyranosyl]-28-O-β- D -glucopyranosyl oleanolic acid ( 8 ), and 3-O-[β- D -glucopyranosyl-(1→2)-α- L -rhamnopyranosyl-(1→3)-β- D -glucuronopyranosyl]-28-O-β- D -glucopyranosyl oleanolic acid ( 9 ). The structures of these compounds were determined based on spectral and chemical evidence. The 50 per cent growth-inhibiting (GI50) of compounds 1 and 5 against MDA-MB-231 (a human breast cancer cell line) was 3.44 × 10-4 and 4.66 × 10-4 mol/L, respectively.

Three spirostanol and two furostanol glycosides were isolated from a methanol extract of the roots of Asparagus curillus and characterized as 3-O-[α-l-arabinopyranosyl (1→4)- β-d-glucopyranosyl]-(25S)-5β-spirostan-3β-ol, 3-O-[{α-l-rhamnopyranosyl (1→2)} {α-l-arabinopyranosyl (1→4)}-β-d-glucopyranosyl]-(25S)-5β-spirostan- 3β-ol, 3-O-[{β-d-glucopyranosyl (1→2)} {α-l-arabinopyranosyl (1→4)}-β- d-glucopyranosyl]-(25S)-5β-spirostan-3β-ol, 3-O-[{β-d-glucopyranosyl (1→2)} {α-l-arabinopyranosyl (1→4)}-β-d-glucopyranosyl]-26-O-[β-d-glucopyranosyl]- 22α-methoxy-(25S)-5β-furostan-3β, 26-diol and 3-O-[{β-d-glucopyranosyl (1→2)} {α-l-arabinopyranosyl (1→4)}-β-d-glucopyranosyl]-26-O-[β-d-glucopyranosyl]- (25S)-5β-furostan-3β, 22α, 26-triol respectively.  相似文献

Three new diarylheptanoids, designated 1-(3,4-dihydroxyphenyl)-7-(4-hydroxyphenyl)-(6E)-6-hepten-3-ol (1), 1-(3-hydroxyphenyl)-7-(3,4-dihydroxyphenyl)-3-methoxy-(6E)-6-heptene (2), (3R, 5R)-1-(3,4-dihydroxyphenyl)-7-phenyl-heptane-3,5-diol (3) and three known compounds, were isolated from rhizomes of Curcuma comosa. Three glycopeptides, obtained in quantity from ovalbumin by exhaustive digestion with Pronase and purified by ion-exchange chromatography and gel filtration, had mannose-2-acetamido-2-deoxyglucose-aspartic acid ratios of 5:4:1, 6:2:1, and 5:2:1. The structures of the glycopeptides have been investigated by sequential digestion with purified exo-glycosidases, Smith degradation, and selective acetolysis, and by methylation analysis of the glycopeptides and their degradation products. The resulting data indicated the structures to be α-d-Manp-(1→6)-[α-d- Manp-(1→3)]-α-d-Manp-(1→6)-[β-d-GlcNAcp-(1→4)]-[β-d-GlcNAcp-(1→2)-α-d- Manp-(1→3)]-β-d-Manp-(1→4)-β-d-GlcNAcp-(1→4)-β-d-GlcNAcp→Asn, α-d- Manp-(1→6)-[α-d-Manp-(1→3)]-α-d-Manp-(1→6)-[α-d-Manp-(1→2)-α-d-Manp- (1→3)]-β-d-Manp-(1→4)-β-d-GlcNAcp-(1→4)-β-d-GlcNAcp→Asn, and α-d-Manp- (1→6)-[α-d-Manp-(1→3)]-α-d-Manp-(1→6)-[α-d-Manp-(1→3)]-β-d-Manp-(1→4)- β-d-GlcNAcp-(1→4)-β-d-GlcNAcp→Asn. The glycopeptides had a common-core structure consisting of five mannose and two hexosamine residues, but the two larger glycopeptides were not homologous.  相似文献

Five anthocyanins, cyanidin 3-(2′′-(6′′′-caffeoyl-β-glucopyranosyl)-6′′-(E-p-coumaroyl)-β-glucopyranoside)-5-β-glucopyranoside, cyanidin 3-(2′′-(6′′′-E-sinapoyl-β-glucopyranosyl)-6′′-(E-p-coumaroyl)-β-glucopyranoside)-5-β-glucopyranoside, cyanidin 3-(2′′-(6′′′-feroyl-β-glucopyranosyl)-6′′-(E-p-coumaroyl)-β-glucopyranoside)-5-β-glucopyranoside, pelargonidin 3-(2′′-(6′′′-E-sinapoyl-β-glucopyranosyl)-6′′-(E-p-coumaroyl)-β-glucopyranoside)-5-β-glucopyranoside, and pelargonidin 3-(2′′-(6′′′-E-p-coumaroyl-β-glucopyranosyl)-6′′-(E-p-coumaroyl)-β-glucopyranoside)-5-β-glucopyranoside, together with five known anthocyanins have been identified in flowers of Cleome hassleriana Queen line. One monoacylated and four diacylated cyanidin 3-sophoroside-5-glucosides were identified as the main anthocyanins in flowers with mauve colouration, while a homologous glycosidic pattern based on pelargonidin was found in the five main anthocyanins from flowers with pink colouration. The anthocyanins identified in C. hassleriana share the same glycosidic pattern as anthocyanins isolated from the genera Raphanus, Brassica and Iberis in the sister family Brassicaceae.  相似文献

The N-terminal domain of glucose-dependent insulinotropic polypeptide (GIP) plays an important role in regulating biological activity. This study examined biological properties of several N-terminal truncated forms of GIP and two novel forms with substitutions at Phe position-6 with Arg or Val. GIP(6-42), GIP(R6-42), GIP(V6-42), GIP(7-42) and GIP(9-42) stimulated cAMP production in BRIN-BD11 cells similar to native GIP, whereas responses to GIP(3-42), GIP(4-42), GIP(5-42) and GIP(8-42) were reduced (P < 0.01 to P < 0.001). GIP-induced cyclic AMP production was significantly inhibited by GIP(3-42), GIP(4-42), GIP(5-42), GIP(6-42), GIP(R6-42), GIP(7-42) and GIP(8-42) (P < 0.001). Compared with native GIP, in vitro insulinotropic activity of GIP(3-42), GIP(4-42), GIP(5-42), GIP(7-42) and GIP(8-42) was reduced (P < 0.05 to P < 0.001), with GIP(4-42), GIP(5-42), GIP(7-42) and GIP(8-42) also potently inhibiting GIP-stimulated insulin secretion (P < 0.001). In ob/ob mice, GIP(4-42) and GIP(8-42) increased (P < 0.05 to P < 0.01) plasma glucose concentrations compared to the glucose-lowering action of native GIP. When GIP(8-42) was co-administered with native GIP it countered the ability of the native peptide to lower plasma glucose and increase circulating insulin concentrations. These data confirm the importance of the N-terminal region of GIP in regulating bioactivity and reveal that sequential truncation of the peptide yields novel GIP receptor antagonists which may have functional significance.  相似文献

A series of 2-(arylidene)-1-(4-chlorophenyl)-4,4,4-trifluorobutane-1,3-diones (24), 4-(arylidene)-3-(4-chlorophenyl)-5-(trifluoromethyl)-4H-pyrazoles (57), 1-(4-chlorophenyl)-4,4,4-trifluoro-2-(2-(aryl)hydrazono)butane-1,3-diones (8, 9), 3-(4-chlorophenyl)-4-(2-(aryl)hydrazono)-5-(trifluoromethyl)-4H-pyrazoles (10, 11), 2-((3-(4-chlorophenyl)-1-phenyl-5-(trifluoromethyl)-1H-pyrazol-4-yl)methylene)malononitrile (13), 2-((5-(4-chlorophenyl)-1-phenyl-3-(trifluoromethyl)-1H-pyrazol-4-yl)methylene)cycloalkan-1-ones (14, 15) and 1-(aryl)-3-(5-(4-chlorophenyl)-1-phenyl-3-(trifluoromethyl)-1H-pyrazol-4-yl)prop-2-en-1-ones (16, 17) were designed, synthesized and evaluated for their in vitro antitumor activity. 1-(4-Chlorophenyl)-4,4,4-trifluoro-2-(2-(4-methoxyphenyl)hydrazono)butane-1,3-dione (8) showed potential and broad spectrum antitumor activity compared to the known drug 5-FU with GI50, (6.61 and 22.60 µM), TGI (42.66 and <100?µM) and LC50 (93.33 and <100?µM) values, respectively. On the other hand, compound 8 yielded selective activities toward melanoma, colon, non-small lung and breast cancer cell lines compared with erlotinib and gefitinib. Molecular docking methodology was performed for compound 8 into binding site of B-RAFV600E and EGFR kinases which showed similar binding mode to vemurafenib (PLX4032) and erlotinib, respectively.  相似文献

A new model is presented to predict the plant uptake of nitrate supplied by diffusion and mass flow to its roots. Plant growth, root-shoot ratio and the plant's nitrate uptake capacity are all set dependent on the plant's N nutrition state. The androgen receptor (AR) is an attractive target for the treatment and molecular imaging of prostate cancer. New carbon-11-labeled propanamide derivatives were first designed and synthesized as selective androgen receptor modulator (SARM) radioligands for prostate cancer imaging using the biomedical imaging technique positron emission tomography (PET). The target tracers, (S)-N-(4-cyano-3-(trifluoromethyl)phenyl)-2-hydroxy-3-(2-[11C]methoxyphenoxy)-2-methylpropanamide ([11C]8a), (S)-2-hydroxy-3-(2-[11C]methoxyphenoxy)-2-methyl-N-(4-nitro-3-(trifluoromethyl)phenyl)propanamide ([11C]8e), (S)-N-(4-cyano-3-(trifluoromethyl)phenyl)-2-hydroxy-3-(4-[11C]methoxyphenoxy)-2-methylpropanamide ([11C]8c) and (S)-2-hydroxy-3-(4-[11C]methoxyphenoxy)-2-methyl-N-(4-nitro-3-(trifluoromethyl)phenyl)propanamide ([11C]8g), were prepared by O-[11C]methylation of their corresponding precursors, (S)-N-(4-cyano-3-(trifluoromethyl)phenyl)-2-hydroxy-3-(2-hydroxyphenoxy)-2-methylpropanamide (9a), (S)-2-hydroxy-3-(2-hydroxyphenoxy)-2-methyl-N-(4-nitro-3-(trifluoromethyl)phenyl)propanamide (9b), (S)-N-(4-cyano-3-(trifluoromethyl)phenyl)-2-hydroxy-3-(4-hydroxyphenoxy)-2-methylpropanamide (9c) and (S)-2-hydroxy-3-(4-hydroxyphenoxy)-2-methyl-N-(4-nitro-3-(trifluoromethyl)phenyl)propanamide (9d), with [11C]CH3OTf under basic conditions and isolated by a simplified C-18 solid-phase extraction (SPE) method in 55 ± 5% (n = 5) radiochemical yields based on [11C]CO2 and decay corrected to end of bombardment (EOB). The overall synthesis time from EOB was 23 min, the radiochemical purity was >99%, and the specific activity at end of synthesis (EOS) was 277.5 ± 92.5 GBq/μmol (n = 5).  相似文献

Two new saponins, agavasaponin E and agavasaponin H have been isolated from the methanolic extract of Agave americana leaves and their structures elucidated. A series of (E)-1-(substituted benzylidene)-4-(3-isopropylphenyl)thiosemicarbazone derivatives were synthesized and characterized by FT-IR spectrum, elemental analysis, NMR spectrum, HR-MS spectrum, and X-ray single crystal diffraction technology. The crystal structures and packing of (E)-1-(4-fluorobenzylidene)-4-(3-isopropylphenyl)thiosemicarbazone and (E)-1-(3-fluorobenzylidene)-4-(3-isopropylphenyl)thiosemicarbazone were maintained through the intramolecular hydrogen bond (N3-H6⋅⋅⋅N1) and intermolecular hydrogen bonds (N2-H4⋅⋅⋅S1, C14-H14⋅⋅⋅F1 and C7-H7⋅⋅⋅S1). The results obtained by employing the DPPH free radicals scavenging assay indicated that (E)-1-(4-methoxylbenzylidene)-4-(3-isopropylphenyl)thiosemicarbazone had a more significant antioxidant activity compared with other compounds. The results measured by adopting the disc diffusion method elucidated that (E)-1-(4-trifluoromethylbenzylidene)-4-(3-isopropylphenyl)thiosemicarbazone possessed a more prominent antifungal activity than other compounds. Molecular docking showed that (E)-1-(4-chlorobenzylidene)-4-(3-isopropylphenyl)thiosemicarbazone had the highest affinity with receptor protein (1NMT). Moreover, the drug-likeness characteristic, physicochemical properties, pharmacokinetic profiles, and bioactivity scores of all the compounds were predicted through in silico studies. The results illustrated that (E)-1-(4-fluorobenzylidene)-4-(3-isopropylphenyl)thiosemicarbazone had the drug-likeness characteristic and all the compounds were considered as moderately biological active molecules.  相似文献

Treatment of 2-benzimidazolemethanol (4) with methanesulfonyl chloride and pyridine in chloroform afforded 2-(chloromethyl)-1-(methylsulfonyl)benzimidazole (6), which was also prepared by methanesulfonylation of 2-(chloromethyl)benzimidazole. Methanesulfonylation of α-(2-benzimidazolyl)benzyl alcohol (8) in chloroform yielded 2-(α-chlorobenzyl)-1-(methylsulfonyl)benzimidazole. 1-(Methylsulfonyl)-2-benzimidazolemethanol was obtained on methanesulfonylation of 4 pyridine at 0°, and α-[1-(methylsulfonyl)-2-benzimidazolyl]benzyl alcohol (12) was prepared from 8 by using the same reaction conditions. The reaction of 1-acetyl-2-(chloromethyl)-benzimidazole with silver methanesulfonate in benzene gave 1-acetyl-O-(methylsulfonyl)-2-benzimidazolemethanol. Compound 6 has some antitumor activity in the KB cell-culture system, and some antibacterial activity in the Staphylococcus aureus test-system; it is also active in preventing anaphylactic shock in a mouse test-system.  相似文献

Chemical investigation of the ethanol extract of the branch and leaves of Illicium majus resulted in the isolation of four new phenylpropanoid glycosides ( 1 – 4 ) and one new phenolic glycoside ( 9 ), along with 13 known ones. Spectroscopic techniques were used to elucidate the structures of the new isolates such as 3-[(2R,3S)-7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-3-(hydroxymethyl)-2,3-dihydro-1-benzofuran-5-yl]propyl β-D-glucopyranoside ( 1 ), [(2R,3S)-7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-5-(3-hydroxypropyl)-2,3-dihydro-1-benzofuran-3-yl]methyl 2-O-α-L-rhamnopyranosyl-β-D-glucopyranoside ( 2 ), [(2R,3S)-7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-5-(3-hydroxypropyl)-2,3-dihydro-1-benzofuran-3-yl]methyl 2-O-α-L-rhamnopyranosyl-β-D-xylopyranoside ( 3 ), 3-[(2R,3S)-3-({[2-O-(4-O-acetyl-α-L-rhamnopyranosyl)-β-D-xylopyranosyl]oxy}methyl)-7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-2,3-dihydro-1-benzofuran-5-yl]propyl acetate ( 4 ), and 4-(2-hydroxyethyl)phenyl 3-O-β-D-glucopyranosyl-β-D-glucopyranoside ( 9 ). Free radical scavenging activities of the isolates were elucidated through the DPPH assay method. The most active compounds, 1-O-caffeoyl-β-D-glucopyranose ( 17 ) and soulieana acid 1 ( 18 ), exhibited moderate radical scavenging activities (IC50=37.7±4.4 μM and IC50=97.2±3.4 μM, respectively). The antibacterial activities of the isolates against Staphylococcus aureus and Escherichia coli were also assessed, and no activity was shown at the measured concentration (<32 μg/mL).  相似文献
