Ceramide and mono-, di-, tri-, and tetraglycosylceramide were isolated from the bran and endosperm of rice grains and chemically characterized. The detailed compositions of free ceramide were somewhat different between the bran and endosperm, but those of the ceramide moiety in glycosylceramides were substantially the same. There was a tendency in all the sphingolipid molecules in rice grains for hydroxy acids with C20 to be combined largely with the dihydroxy bases while hydroxy acids with C24< combined mainly with the trihydroxy bases. Representative molecular species of the sphingolipid classes were concluded to be as follows: for ceramide N-2′-hydroxylignoceroyl-4-hydroxysphinganine, for monoglycosylceramide l-O-β-glucosyl-N-2′-hydroxyarachidoyl-4,8-sphingadienine, for diglycosylceramide 1-O-[β-mannosyl(1→-4)-O-β-glucosyl]- and 1-O-[β-glucosyl(1→4)-O-β-glucosyl]-N-2′-hydroxylignoceroyl-4-hydroxy-8-sphingenine, for triglycosylceramide l-O-[β-mannosyl(1→4)-O-β-mannosyl(l→4)-O-β-glucosyl]- and l-O-[β-glucosyl(l→4)-O-β-mannosyl(1→4)-O-β-glucosyl]-N-2′-hydroxylignoceroyl-4-hydroxy-8-sphingenine, and for tetraglycosylceramide 1-0-[β-mannosyl(l→4)-O-β-mannosyl (1→4)-O-β-mannosyl(1→4)-O-β-glucosyl]- and l-O-[β-glucosyl(1→4)-O-β-mannosyl(l→4)-O-β-mannosyl(1β4)-O-β-glucosyl]-N-2′-hydroxylignoceroyl-4-hydroxy-8-sphingenine. 相似文献

Robinin has been proved by total synthesis and by methylation analysis using GC-MS to be kaempferol-3- O-(6-O-α-l-rhamnopyranosyl -β-d-galactopyranoside)-7-O-α-l-rhamnopyranoside and not, as claimed by Maksjutina et al., a mixture of 4 glycosides containing the 7-O-rhamnosyl moiety in the α- and β-furanoside as well as in the α- and β-pyranoside forms. The assumption that the 3-O-rhamnosido -galactose moiety contained furanoid rings was also disproved. 相似文献

The present paper deals with the study of the kinetics and thermodynamics of batch enzymic hydrolysis of multisubstrate media in the presence of more than one glucanase. A kinetic model, similar to the one proposed for competitive inhibition, is presented to describe the competition between two substrates, tri- and tetrasaccharides, for the same enzyme, glucoamylase. A literature research on the most recurrent values of the Michaelis-Menten constant also shows a linear relationship between this parameter and the molecular weight of the sugar substrate for a given glucanase.List of Symbols

岩黄连光合与蒸腾特性及其对光照强度和CO2浓度的响应 总被引:15,自引:2,他引:15
采用LI-6400便携式光合测定系统(Li-CorInc.,USA)对岩黄连叶片的气体交换进行了测定。结果表明(1)岩黄连叶片的光饱和点(LSP)为329.18μmol.m-2.s-1左右,光补偿点(LCP)为12.76μmol.m-2.s-1,最大净光合速率为2.96μmol.m-2.s-1,暗呼吸速率(Rd)为0.17μmol.m-2.s-1。光饱和点和光补偿点都比效低,表明岩黄连对光照的要求不高,属于阴生植物。(2)4月份,岩黄连Pn随CO2浓度升高而逐渐增大。当CO2浓度由50μmol.mol-1增加到600μmol.mol-1,Pn几乎呈直线上升,600~1000μmol.mol-1范围内逐渐缓和,到1000μmol.mol-1以后Pn变化平稳。由曲线估算CO2饱和点(CSP)大约在1000μmol.mol-1左右。CO2的补偿点为68.80μmol.mol-1。羧化效率为0.0308μmol.m-2.s-1。(3)岩黄连叶片水分利用率(WUE)随有效光辐射强度(PAR)的增强呈抛物线状变化,PAR在200μmol.m-2.s-1内呈直线上升,到200μmol.m-2.s-1时WUE达最大值,大于200μmol.m-2.s-1后WUE呈逐渐下降趋势。 相似文献

Treatment of 2-benzimidazolemethanol (4) with methanesulfonyl chloride and pyridine in chloroform afforded 2-(chloromethyl)-1-(methylsulfonyl)benzimidazole (6), which was also prepared by methanesulfonylation of 2-(chloromethyl)benzimidazole. Methanesulfonylation of α-(2-benzimidazolyl)benzyl alcohol (8) in chloroform yielded 2-(α-chlorobenzyl)-1-(methylsulfonyl)benzimidazole. 1-(Methylsulfonyl)-2-benzimidazolemethanol was obtained on methanesulfonylation of 4 pyridine at 0°, and α-[1-(methylsulfonyl)-2-benzimidazolyl]benzyl alcohol (12) was prepared from 8 by using the same reaction conditions. The reaction of 1-acetyl-2-(chloromethyl)-benzimidazole with silver methanesulfonate in benzene gave 1-acetyl-O-(methylsulfonyl)-2-benzimidazolemethanol. Compound 6 has some antitumor activity in the KB cell-culture system, and some antibacterial activity in the Staphylococcus aureus test-system; it is also active in preventing anaphylactic shock in a mouse test-system. 相似文献

In the presence of suitable acceptor molecules, dextransucrase makes a homologous series of oligosaccharides in which the isomers differ by a single glucosyl unit, whereas alternansucrase synthesizes one trisaccharide, two tetrasaccharides, etc. Previously, we showed that alternansucrase only forms certain isomers of DP > 4 from maltose in measurable amounts, and that these oligosaccharides belong to the oligoalternan series rather than the oligodextran series. We now demonstrate that the acceptor products from gentiobiose, also formed in good yields (nearly 90% in unoptimized reactions), follow a pattern similar to those formed from maltose. The initial product is a single trisaccharide, α-d-Glcp-(1→6)-β-d-Glcp-(1→6)-d-Glc. Two tetrasaccharides were formed in approximately equal quantities: α-d-Glcp-(1→3)-α-d-Glcp-(1→6)-β-d-Glcp-(1→6)-d-Glc and α-d-Glcp-(1→6)-α-d-Glcp-(1→6)-β-d-Glcp-(1→6)-d-Glc. Just one pentasaccharide was isolated from the reaction mixture, α-d-Glcp-(1→6)-α-d-Glcp-(1→3)-α-d-Glcp-(1→6)-β-d-Glcp-(1→6)-d-Glc. Our hypothesis that the enzyme is incapable of forming two consecutive α-(1→3) linkages, and does not form products with more than two consecutive α-(1→6) linkages, apparently applies to other acceptors as well as to maltose. Summary Mapping of monoaminergic systems in the brain of the newt Triturus alpestris was achieved with antisera against (1) thyrosine hydroxylase (TH), (2) formaldehyde-conjugated dopamine (DA), and (3) formaldehyde-conjugated serotonin (5-HT). In the telencephalon, the striatum was densely innervated by a large number of 5-HT-, DA-and TH-immunoreactive (IR) fibers; IR fibers were more scattered in the amygdala, the medial and lateral forebrain bundles, and the anterior commissure. In the anterior and medial diencephalon, TH-IR perikarya contacting the cerebrospinal fluid (CSF-C perikarya) were located in the preoptic recess organ (PRO), the organum vasculosum laminae terminalis and the suprachiasmatic nucleus. Numerous TH-IR perikarya, not contacting the CSF, were present in the posterior preoptic nucleus and the ventral thalamus. At this level, DA-IR CSF-C neurons were only located in the PRO. In the posterior diencephalon, large populations of 5-HT-IR and DA-IR CSF-C perikarya were found in the paraventricular organ (PVO) and the nucleus infundibularis dorsalis (NID); the dorsal part of the NID additionally presented TH-IR CSF-C perikarya. Most regions of the diencephalon showed an intense monoaminergic innervation. In addition, numerous TH-IR, DA-IR and 5-HT-IR fibers, orginating from the anterior and posterior hypothalamic nuclei, extended ventrally and reached the median eminence and the pars intermedia of the pituitary gland. In the midbrain, TH-IR perikarya were located dorsally in the pretectal area. Ventrally, a large group of TH-IR cell bodies and some weakly stained DA-IR and 5-HT-IR neurons were observed in the posterior tuberculum. No dopaminergic system equivalent to the substantia nigra was revealed. The possible significance of the differences in the distribution of TH-IR and DA-IR neurons is discussed, with special reference to the CSF-C neurons.Abbreviations

The occurrence of a new bacterial dehalogenase acting on both the optical isomers of 2-halogenated alkanoic acids was demonstrated. When the haloalkanoic acid-utilizing bacteria were screened in a medium containing dl-2-chloropropionate as a sole carbon source, two types of bacteria were isolated: (1) a few strains utilizing both d- and l-isomers of 2-chloropropionate and (2) strains utilizing only the l-isomer. A dehalogenating enzyme was obtained from the cells of Pseudomonas sp. which is able to utilize both isomers. The crude enzyme catalyzed the dehalogenation of d- and l-2-chloropropionates to yield l- and d-isomers of lactate, respectively. The enzyme showed the same pH optimum and heat inactivation rate for the d- and l-isomers. Apparent K
m values for d- and l-2-chloropropionates were 4.5 and 1.0 mM, respectively. The enzyme acted specifically on 2-haloalkanoic acids. Activity staining of disc-gels electrophoresed witg the crude enzyme preparation showed that the dehalogenation of d- and l-2-chloropropionates, monochloroacetate, dichloroacetate, 2,2-dichloropropionate, and dl-2-chlorobutyrate is due to a single protein.Abbreviations MCA

The objectives were to: 1) evaluate blood flow in the uterine (UA) and umbilical (Uma) arteries in the pregnant bitch, by measuring the resistive index (RI) and pulsatility index (PI); 2) to note the presence or absence of the early diastolic notch and diastolic flow in the UA and Uma flow waveforms, respectively; and 3) perform conceptus ecobiometry for fetal growth assessment during pregnancy. Six healthy bitches were examined on approximately Days -44, -42, -36, -31, -28, -25, -21, -18, -14, -8, -4, and -2 of pregnancy (whelping = Day 0). Triplex Doppler and B-mode ultrasonography were used to assess blood flow and conceptus ecobiometry. All pregnancies ended with a normal whelping and birth of live puppies. Prior to whelping, all conceptus dimensions increased significantly, whereas RI and PI of both the Uma and UA decreased significantly. For the UA, RI and PI were (mean ± SEM) 0.95 ± 0.02 and 2.75 ± 0.41, respectively, on Day -44, and were 0.60 ± 0.01 and 0.99 ± 0.03 on Day -4. For the Uma, RI and PI were 0.99 ± 0.01 and 2.42 ± 0.03 on Day -31, and were 0.62 ± 0.01 and 1.15 ± 0.02 on Day -4. The complete disappearance of the early diastolic notch in the UA, and the appearance of diastolic flow in the Uma occurred on Days -16 ± 5 and -21 ± 1. The authors concluded that UA and Uma perfusion were important end points to assess fetal vitality in bitches. Furthermore, the current reference values provided a baseline for monitoring normal and abnormal pregnancies in bitches. 相似文献

Vacuole represents a major storage organelle playing vital roles in pH homoeostasis and cellular detoxification. The chemical and functional properties of tonoplast in response to chilling temperature and their roles in chilling injury are largely unknown. In the current study, lipid composition of tonoplast and the activities of two vacuolar proton pumps, H+-ATPase (V-ATPase) and H+-pyrophosphatase (V-PPase), were investigated in accordance with the development of blackheart, a form of chilling injury in pineapple fruit (Ananas comosus). Chilling temperature at 10 °C for 1 week induced irreversible blackheart injury in concurrence with a substantial decrease in V-ATPase activity. By contrast, the activity was increased after 1 week at 25 °C. The activity of V-PPase was not changed under both temperatures. Level of total phospholipids of tonoplast decreased at 10 °C, but increased at 25 °C. There was no change at the level of total glycolipids under both temperatures. Thus, low temperature increased the ratio of total glycolipids vs. total phospholipids of tonoplast. Phosphatidylcholine and phosphatidylethanolamine were the predominant phospholipids of tonoplast. Low temperature increased the relative level of phosphatidic acid but decreased the percentage of both phosphatidylcholine and phosphatidylethanolamine. Unsaturated fatty acids accounted for over 60 % of the total tonoplast fatty acids, with C18:1 and C18:2 being predominant. Low temperature significantly decreased the percentage of C18:3. Modification of membrane lipid composition and its effect on the functional property of tonoplast at low temperature were discussed in correlation with their roles in the development of chilling injury in pineapple fruit. 相似文献

Eight sialyloligosaccharides have been isolated from the hemofiltrate of a patient with end stage renal disease using reverse osmosis, gel filtration, ion-exchange and high-performance liquid chromatography. The structures were predominantly elucidated by one- and two-dimensional 1H- and 13C-NMR spectroscopy: 1 NeuAc alpha 2-3Gal beta 1-4Glc; 2 NeuAc alpha 2-6Gal beta 1-4Glc; 3 NeuAc alpha 2-3Gal beta 1-4GlcNAc; 4 NeuAc-alpha 2-6Gal beta 1-4GlcNAc; 5 NeuAc alpha 2-3Gal beta 1-4-GlcNAc alpha 1-P; 6 NeuAc alpha 2-6Gal beta 1-4GlcNAc alpha 1-P; 7 NeuAc alpha 2-3Gal beta 1-3GalNAc alpha 1-P; 8 NeuAc alpha 2-8NeuAc. While compounds 1-7 are also components of normal human urine, di-N-acetyl-D-neuraminic acid (8) could be isolated for the first time from biological material. Chromone has emerged as one of the most important synthetic scaffolds for antitumor activity, which promotes the development of candidate drugs with better activity. In this study, a series of nitrogen mustard derivatives of chromone were designed and synthesised, in order to discover promising anti-breast tumour candidates. Almost all target derivatives showed antiproliferative activity against MCF-7 and MDA-MB-231 cell lines. In particular, methyl (S)-3-(4-(bis(2-chloroethyl)amino)phenyl)-2-(5-(((6-methoxy-4-oxo-4H-chromen-3-yl)methyl)amino)-5-oxopentanamido)propanoate showed the most potent antiproliferative activity with IC50 values of 1.83 and 1.90 μM, respectively, and it also exhibited certain selectivity between tumour cells and normal cells. Further mechanism exploration against MDA-MB-231 cells showed that it possibly induced G2/M phase arrest and apoptosis by generating intracellular ROS and activating DNA damage. In addition, it also inhibited MDA-MB-231 cells metastasis, invasion and adhesion. Overall, methyl (S)-3-(4-(bis(2-chloroethyl)amino)phenyl)-2-(5-(((6-methoxy-4-oxo-4H-chromen-3-yl)methyl)amino)-5-oxopentanamido)propanoate showed potent antitumor activities and relatively low side effects, and deserved further investigation. 相似文献

In order to examine the evolution of the phalangeal formula in a diverse clade of turtles, including hyperphalangy as a rare
condition in this group, we studied 210 specimens representing all extant genera of Trionychidae and their sister taxon, Carettochelyidae.
Both groups consist of highly aquatic species with elongated autopods that are either paddle-like (Trionychidae) or transformed
to flippers (Carettochelyidae). Phalangeal formulae were obtained mostly by radiographs of alcohol-preserved or dry specimens,
as well as by direct counts from skeletons. All trionychids and Carettochelys are pentadactylous, but their phalangeal formulae differ. Carettochelys exhibits the turtle-plesiomorphic state (manus and pes: 2-3-3-3-3), with no variation in adults. Trionychids exhibit intraspecific
variation, ranging from 2-3-3-3-2 to 2-3-3-6-5 for the manus, and from 2-3-3-3-2 to 2-3-3-5-3 for the pes. The extant Carettochelys as well as the Middle Eocene Allaeochelys crassesculpta are characterized by an elongation of phalanges, whereas trionychids consistently have shorter phalanges. All trionychid
genera exhibit some degree of hyperphalangy in digits IV and V, in both the manus and pes. Phalanges of the clawed digits
I–III are very robust compared to phalanges of the non-clawed digits IV and V. The latter contribute significantly to the
enlargement of the paddle by their additional phalanges. We hypothesize that this phalangeal pattern is coupled with prolongation
of growth processes in the non-clawed digits. The differences in autopod morphology between carettochelyids and trionychids
reflect different locomotor patterns related to different natural histories (elongated flippers for high-speed escape in the
mainly herbivorous Carettochelys; broad paddles for rapid turns during hunting in the mainly carnivorous trionychids). The autopod of Pelodiscus sinensis is proposed as an experimental model to examine the developmental basis of adult autopod variation. 相似文献

The analysis of photochemical activities of Photosystem I and Photosystem II in spheroplasts from normal and photobleached Anabaena cylindrica showed an increase in Photosystem II activity relative to Photosystem I in photobleached cells. We suggest that the reasons for this modification in photochemical activity are, (i) a rearrangement of pigments between the two photosystems, and (ii) improved functional condition of the photosynthetic units in Photosystem II.Abbreviations PSI

The seven isomeric optically inactive forms of methylcyclohexanol (i.e. 1-, and cis- and trans-2-, -3- and -4-) are excreted by rabbits mainly as glucuronides of the thermodynamically more stable forms of the alcohols. The eighth isomer, cyclohexylmethanol, however, undergoes aromatization in vivo, giving rise to benzoic acid and hippuric acid. The (±)-2-, (±-3- and 4-methylcyclohexanones are reduced in the rabbit and excreted mainly as the glucuronides of the thermodynamically more stable forms of the corresponding methylcyclohexanols. 2. Racemic cis- and trans-2-methylcyclohexanol and 2-methylcyclohexanone are all excreted as conjugated trans-2-methylcyclohexanol. However, when the (±)-cis-alcohol or the (±)-ketone is fed, (+)-trans-2-methylcyclohexanol is excreted, whereas when the (±)-trans-alcohol is fed it is excreted as the (±)-trans-alcohol. 3. Racemic cis- and trans-3-methylcyclohexanol and 3-methylcyclohexanone are all excreted as conjugated racemic cis-3-methylcyclohexanol. cis- and trans-4-Methylcyclohexanol and 4-methylcyclohexanone are all excreted as conjugated trans-4-methylcyclohexanol. 4. Cathepsins play a role in regulation of cell function through their presence in the cell nucleus. However, the role of Cathepsin K (Ctsk) as an epigenetic regulator in osteoclasts remains unknown. Our data demonstrated that Ctsk-/-Mmp9-/- mice have a striking phenotype with a 5-fold increase in bone volume compared with WT. RNA-seq analysis of Ctsk-/-, Mmp9-/- and Ctsk-/-/Mmp9-/- osteoclasts revealed their distinct functions in gene expression regulation, including reduced Cebpa expression, increased Nfatc1 expression, and in signaling pathways activity regulation. Western blots and qPCR data validated these changes. ATAC-seq profiling of Ctsk-/-, Mmp9-/-, and Ctsk-/-/Mmp9-/- osteoclasts indicated the changes resulted from reduced chromatin openness in the promoter region of Cebpa and increased chromatin openness in Nfatc1 promoter in Ctsk-/-/Mmp9-/- osteoclasts compared to that in osteoclasts of WT, Ctsk/- and Mmp9-/-. We found co-localization of Ctsk with c-Fos and cleavage of H3K27me3 in wild-type osteoclasts. Remarkably, cleavage of H3K27me3 was blocked in osteoclasts of Ctsk-/- and Ctsk-/-/Mmp9-/- mice, suggesting that Ctsk may epigenetically regulate distinctive groups of genes'' expression by regulating proteolysis of H3K27me3. Ctsk-/-/Mmp9-/- double knockout dramatically protects against ovariectomy induced bone loss. We found that Ctsk may function as an essential epigenetic regulator in modulating levels of H3K27me3 in osteoclast activation and maintaining bone homeostasis. Our study revealed complementary and unique functions of Ctsk as epigenetic regulators for maintaining osteoclast activation and bone homeostasis by orchestrating multiple signaling pathways and targeting both Ctsk and Mmp9 is a novel therapeutic approach for osteolytic diseases such as osteoporosis. 相似文献

We investigated the morphology of adult and juvenile stages of females and males of Bosmina longispina Leydig from a North Icelandic population. There are additional setae near the basis of the second antenna and between its branches in adult males, not described in Bosmina, but known from other Anomopoda. New features of female morphology are also found. The development of the first limb and postabdomen of the male is described in detail. In juvenile males of B. longispina (belonging to subgenus Eubosmina), characteristics of morphology, similar to those in adult males of other subgenera are found.Abbreviations
