The objectives of this research were to prepare and characterize inclusion complexes of clonazepam with β-cyclodextrin and hydroxypropyl-β-cyclodextrin and to study the effect of complexation on the dissolution rate of clonazepam, a water-insoluble lipid-lowering drug. The phase-solubility profiles with both cyclodextrins were classified as AP-type, indicating the formation of 2:1 stoichiometric inclusion complexes. Gibbs free energy ( DGtro ) \left( {\Delta {G_{tr}}^o} \right) values were all negative, indicating the spontaneous nature of clonazepam solubilization, and they decreased with increase in the cyclodextrins concentration, demonstrating that the reaction conditions became more favorable as the concentration of cyclodextrins increased. Complexes of clonazepam were prepared with cyclodextrins by various methods such as kneading, coevaporation, and physical mixing. The complexes were characterized by Fourier transform infrared spectroscopy and differential scanning calorimetry studies. These studies indicated that complex prepared kneading and coevaporation methods showed successful inclusion of the clonazepam molecule into the cyclodextrins cavity. The complexation resulted in a marked improvement in the solubility and wettability of clonazepam. Among all the samples, complex prepared with hydroxypropyl-β-cyclodextrin by kneading method showed highest improvement in in vitro dissolution rate of clonazepam. Mean dissolution time of clonazepam decreased significantly after preparation of complexes and physical mixture of clonazepam with cyclodextrins. Similarity factor indicated significant difference between the release profiles of clonazepam from complexes and physical mixture and from plain clonazepam. Tablets containing complexes prepared with cyclodextrins showed significant improvement in the release profile of clonazepam as compared to tablet containing clonazepam without cyclodextrins.  相似文献

Rate of net CO2 assimilation by soil-grown soybean plants were studied over a range of relative leaf water contents at each of four levels of irradiance. There was a large interaction between light level and leaf water deficit on the rate of CO2 assimilation. The effect of leaf water deficit on assimilation became larger as irradiance increased. Both stomatal resistance to CO2 transport and mesophyll resistance to CO2 assimilation increased as leaf-water deficit increased. The increase in both resistance with changing leaf-water content was largest at high irradiance and became smaller as irradiance decreased. Relief of soil-moisture stress by watering induced large oscillations of CO2 assimilation, stomatal resistance, and mesophyll resistance. The oscillation of the mesophyll resistance occurred in the absence of changes in relative water content and appeared to be related to oscillations in leaf temperature. The observed increase in mesophyll resistance with decreasing leaf-water content under nonoscillative conditions may be caused by changes in leaf temperature rather than leaf water content.  相似文献

Summary At constant cladode temperature the stomatal resistance of O. inermis increased when the cladode-air vapor pressure difference was increased and stomatal resistance decreased when the cladode-air vapor pressure difference was lowered. Net CO2 fixation in the dark was very responsive to these humidity dependent changes in stomatal resistance. Net CO2 fixation and stomatal resistance in the light did not respond to changes in cladode-air vapor pressure differences in the light under the conditions tested. When temperature response functions for dark CO2 fixation were examined at constant ambient humidity, the reduction in dark CO2 fixation at higher temperatures was largely due to stomatal closure in response to the increased vapor pressure difference. The water requirement for net CO2 fixation in the dark at typical nocturnal vapor pressure differences was about 10 times lower than that of net CO2 fixation in the light at vapor pressure differences typical of the late afternoon. The role of the stomatal responses to humidity in determining the patterns and rates of net CO2 exchange in the light or dark, and its possible ecological significance is discussed.  相似文献

Summary The structural characteristics, water relations, and photosynthesis of Ferocactus acanthodes (Lemaire) Britton and Rose, a barrel cactus exhibiting Crassulacean acid metabolism (CAM), were examined in its native habitat in the western Colorado desert. Water storage in its succulent stem permitted nighttime stomatal opening ot continue for about 40 days after the soil water potential became less than that of the stem, a period whe the plant would be unable to extract water from the soil. After 7 months of drought and consequent unreplenished water loss from a plant, diurnal stomatal activity was not observed and the stem osmotic pressure was 6.4 bars, more than double the value measured during wet periods with nighttime stomatal opening. F. acanthodes had a shallow root system (mean depth of 8 cm) which responded within 24 h to rainfall.When the nocturnal stem surface temperature was raised from 8.0° C to 35.0° C, the stomatal resistance increased 4-fold, indicating that cool nighttime temperatures are advantageous for gas exchange by F. acanthodes. Moreover, the optimal temperature for CO2 uptake in the dark was only 12.6° C. CO2 uptake at night became maximal for 3.0 mEinsteins cm-2 of photosynthetically active radiation incident during the preceding day, and the minimum number of incident quanta absorbed per CO2 fixed was 68. The transpiration ratio (mass of water transpired/mass of CO2 fixed) had the relatively low value of 70 for an entire year, consistent with values obtained for other CAM plants. The total amount of water annually diverted to the floral structures was about 6% of the stem wet weight. The annual growth increment estimated from the net CO2 assimilation corresponded to about 10% of the stem mass for barrel cacti 34 cm tall, in agreement with measured dimension changes, and indicated that such plants were about 26 years old.  相似文献

Seedlings of Alnus incana (L.) Moench were studied in controlled environmental conditions. Net photosynthetic capacity of four plants simultaneously as well as light, temperature, and atmospheric water pressure deficit were continuously recorded during approximately two months. The potted plants were continuously given known quantities of water. Two different effects of water stress were found in the experimental plants. When sufficient amounts of water had been available to them, photosynthetic CO2 fixation rates largely followed the variation in temperature and light. On the other hand, after prolonged water stress, higher temperatures caused a large decrease in net CO2 uptake even if the plant apparently had sufficient water during the actual measurements. Possible mechanisms for this effect are discussed. Effects of water stress on photosynthesis were studied at a constant temperature as well as in conditions where temperature, light, soil water content, and atmospheric water vapor pressure were allowed to vary in a complex pattern. Mathematical models for expressing net CO2 uptake as a function of environmental variables were constructed for both of these experimental situations. However, only the latter approach clearly demonstrated the fundamental role of temperature in controlling the photosynthesis of plants under water stress.  相似文献

The purpose of this study was to characterize leaf photosynthetic and stomatal responses of wheat (Triticum aestivum L.) plants grown under two N-nutritional regimes. High- and low-N regimes were imposed on growth-chamber-grown plants by fertilizing with nutrient solutions containing 12 or 1 millimolar nitrogen, respectively. Gas-exchange measurements indicated not only greater photosynthetic capacity of high-N plants under well-watered conditions, but also a greater sensitivity of CO2 exchange rate and leaf conductance to CO2 and leaf water potential compared to low-N plants. Increased sensitivity of high-N plants was associated with greater tissue elasticity, lower values of leaf osmotic pressure and greater aboveground biomass. These N-nutritional-related changes resulted in greater desiccation (lowered relative water content) of high-N plants as leaf water potential fell, and were implicated as being important in causing greater sensitivity of high-N leaf gas exchange to reductions in water potential. Water use efficiency of leaves, calculated as CO2 exchange rate/transpiration, increased from 9.1 to 13 millimoles per mole and 7.9 to 9.1 millimoles per mole for high- and low-N plants as water became limiting. Stomatal oscillations were commonly observed in the low-N treatment at low leaf water potentials and ambient CO2 concentrations, but disappeared as CO2 was lowered and stomata opened.  相似文献

Exposing plants to long-term CO2 enrichment generally leads to increases in plant biomass, total leaf area and alterations on leaf net photosynthetic rates, stomatal conductance and water use efficiency. However, the magnitude of such effects is dependent on the availability of other potentially limiting resources. The aim of our study was to elucidate the effects of elevated CO2, applied at different temperature and water availability regimes, on nodulated alfalfa plants. Regardless of water supply, elevated CO2 enhanced plant growth, especially when combined with increased temperature although no differences were detected until 30 days of treatment. Absence of differences in leaf relative growth rate, and gas exchange measurements, suggested that plants grown in a low water regime adjusted their growth to the amount of available water. Elevated CO2 enhanced water use efficiency because of reduced water consumption and a greater dry mass production. Increased dry matter production of plants grown under elevated CO2 and temperature was the result of stimulated photosynthetic rates, greater leaf area and water use efficiency. Lack of CO2 effect on photosynthesis of plants grown at ambient temperature might be consequence of down-regulation phenomena. Plants grown at 700 μmol mol−1 CO2 maintained control nitrogen levels, discarding enhanced nitrogen availability as the main factor explaining enhanced dry matter.  相似文献

Catalytic steam reforming of glycerol for H2 production has been evaluated experimentally in a continuous flow fixed-bed reactor. The experiments were carried out under atmospheric pressure within a temperature range of 400–700 °C. A commercial Ni-based catalyst and a dolomite sorbent were used for the steam reforming reactions and in situ CO2 removal. The product gases were measured by on-line gas analysers. The results show that H2 productivity is greatly increased with increasing temperature and the formation of methane by-product becomes negligible above 500 °C. The results suggest an optimal temperature of ∼500 °C for the glycerol steam reforming with in situ CO2 removal using calcined dolomite as the sorbent, at which the CO2 breakthrough time is longest and the H2 purity is highest. The shrinking core model and the 1D-diffusion model describe well the CO2 removal under the conditions of this work.  相似文献

It has been reported that elevated temperature accelerates the time‐to‐mortality in plants exposed to prolonged drought, while elevated [CO2] acts as a mitigating factor because it can reduce stomatal conductance and thereby reduce water loss. We examined the interactive effects of elevated [CO2] and temperature on the inter‐dependent carbon and hydraulic characteristics associated with drought‐induced mortality in Eucalyptus radiata seedlings grown in two [CO2] (400 and 640 μL L?1) and two temperature (ambient and ambient +4 °C) treatments. Seedlings were exposed to two controlled drying and rewatering cycles, and then water was withheld until plants died. The extent of xylem cavitation was assessed as loss of stem hydraulic conductivity. Elevated temperature triggered more rapid mortality than ambient temperature through hydraulic failure, and was associated with larger water use, increased drought sensitivities of gas exchange traits and earlier occurrence of xylem cavitation. Elevated [CO2] had a negligible effect on seedling response to drought, and did not ameliorate the negative effects of elevated temperature on drought. Our findings suggest that elevated temperature and consequent higher vapour pressure deficit, but not elevated [CO2], may be the primary contributors to drought‐induced seedling mortality under future climates.  相似文献

Plants from clonal cuttings of Salix sp. were subjected to a drying cycle of 10 d in a controlled environment. Gas exchange and fluorescence emission were measured on attached leaves. The light-saturated photosynthetic CO2 uptake became progressively inhibited with decreased leaf water potential both at high, and especially, at low intercellular CO2 pressure. The maximal quantum yield of CO2 uptake was more resistant. The inhibition of light-saturated CO2 uptake at leaf water potentials around-10 bar, measured at a natural ambient CO2 concentration, was equally attributable to stomatal and non-stomatal factors, but the further inhibition below this water-stress level was caused solely by non-stomatal factors. The kinetics of fluorescence emission was changed at severe water stress; the slow secondary oscillations of the induction curve were attenuated, and this probably indicates perturbations in the carbon reduction cycle. The influence of light level during the drought period was also studied. Provided the leaves were properly light-acclimated, drought at high and low light levels produced essentially the same effects on photosynthesis. However, low-light-acclimated leaves became more susceptible to photoinhibitory treatment under severe water stress, as compared with well-watered conditions.  相似文献

Terrestrial responses to increasing atmospheric CO2 are important to the global carbon budget. Increased plant production under elevated CO2 is expected to increase soil C which may induce N limitations. The objectives of this study were to determine the effects of increased CO2 on 1) the amount of carbon and nitrogen stored in soil organic matter and microbial biomass and 2) soil microbial activity. A tallgrass prairie ecosystem was exposed to ambient and twice-ambient CO2 concentrations in open-top chambers in the field from 1989 to 1992 and compared to unchambered ambient CO2 during the entire growing season. During 1990 and 1991, N fertilizer was included as a treatment. The soil microbial response to CO2 was measured during 1991 and 1992. Soil organic C and N were not significantly affected by enriched atmospheric CO2. The response of microbial biomass to CO2 enrichment was dependent upon soil water conditions. In 1991, a dry year, CO2 enrichment significantly increased microbial biomass C and N. In 1992, a wet year, microbial biomass C and N were unaffected by the CO2 treatments. Added N increased microbial C and N under CO2 enrichment. Microbial activity was consistently greater under CO2 enrichment because of better soil water conditions. Added N stimulated microbial activity under CO2 enrichment. Increased microbial N with CO2 enrichment may indicate plant production could be limited by N availability. The soil system also could compensate for the limited N by increasing the labile pool to support increased plant production with elevated atmospheric CO2. Longer-term studies are needed to determine how tallgrass prairie will respond to increased C input.  相似文献

Leaf photosynthesis rate of the C4 species Paspalum plicatulum Michx was virtually CO2-saturated at normal atmospheric CO2 concentration but transpiration decreased as CO2 was increased above normal concentrations thereby increasing transpiration efficiency. To test whether this leaf response led growth to be CO2-sensitive when water supply was restricted, plants were grown in sealed pots of soil as miniature swards. Water was supplied either daily to maintain a constant water table, or at three growth restricting levels on a 5-day drying cycle. Plants were either in a cabinet with normal air (340 mol (CO2) mol-1 (air)) or with 250 mol mol-1 enrichment. Harvesting was by several cycles of defoliation.With abundant water supply high CO2 concentration did not cause increased growth, but it did not cause an increase in growth over a wide range of growth-limiting water supplies either. Only when water supply was less than 30–50% of the amount used by the stand with a water-table was there evidence that dry weight growth was enhanced by high CO2. In addition, with successive regrowth, the enhancing effect under a regime of minimal water allocations, became attenuated. Examination of leaf gas exchange, growth and water use data showed that in the long term stomatal conductance responses were of little significance in matching plant water use to low water allocation; regulation of leaf area was the mechanism through which consumption matched supply. Since high CO2 effects operate principally via stomatal conductance in C4 species, we postulate that for this species higher CO2 concentrations expected globally in future will not have much effect on long term growth.  相似文献

A system for measurement of leaf gas exchange while regulating leaf to air vapour pressure difference has been developed; it comprises an assimilation chamber, leaf temperature controller, mass flow controller, dew point controller and personal computer. A relative humidity sensor and air and leaf temperature sensors, which are all used for regulating the vapour pressure difference, are mounted into the chamber. During the experiments, the computer continuously monitored the photosynthetic parameters and measurement conditions, so that accurate and intenstive measurements could be made.When measuring the light-response curve of CO2 assimilation for single leaves, in order to regulate the vapour pressure difference, the leaf temperature and relative humidity in the chamber were separately and simultaneously controlled by changing the air temperature around the leaf and varying the air flow rate through the chamber, respectively. When the vapour pressure difference was regulated, net CO2 assimilation, transpiration and leaf conductance for leaves of rice plant increased at high quantum flux density as compared with those values obtained when it was not regulated.When measuring the temperature-response curve of CO2 assimilation, the regulation of vapour pressure difference was manipulated by the feed-forward control of the dew point temperature in the inlet air stream. As the vapour pressure difference was regulated at 12 mbar, the maximum rate of and the optimum temperature for CO2 assimilation in rice leaves increased 5 molCO2 m–2 s–1 and 5°C, respectively, as compared with those values obtained when the vapour pressure difference took its own course. This was reasoned to be due to the increase in leaf conductance and the decrease in transpiration rate. The water relations and photosynthesis of Agave deserti Engelm., a plant exhibiting Crassulacean acid metabolism, were measured in the Colorado desert. Although no natural stomatal opening of A. deserti occurred in the summer of 1975, it could be induced by watering. The resistance for water vapor diffusion from a leaf (RWV) became less than 20 sec cm−1 when the soil water potential at 10 cm became greater than −3 bars, as would occur after a 7-mm rainfall. As a consequence of its shallow root system (mean depth of 8 cm), A. deserti responded rapidly to the infrequent rains, and the succulent nature of its leaves allowed stomatal opening to continue for up to 8 days after the soil became drier than the plant. When the leaf temperature at night was increased from 5 to 20 C, RWV increased 5-fold, emphasizing the importance of cool nighttime temperatures for gas exchange by this plant. Although most CO2 uptake occurred at night, a secondary light-dependent rise in CO2 influx generally occurred after dawn. The transpiration ratio (mass of water transpired/mass of CO2 fixed) had extremely low values of 18 for a winter day, and approximately 25 for an entire year.  相似文献

叶面积指数是作物生长状况的一个重要表征参数,也是研究陆地生态系统的一个重要的参数.当今世界温室气体排放逐年上升,气候变暖趋势明显,对气候变化敏感的农业将受到影响.在全球变化的背景下,采用农业技术转移决策支持系统(DSSAT)系统,通过在黄淮海平原典型站点模拟3种CO2浓度条件下冬小麦在水分充足和水分亏缺2种情境下的生长过程,分析不同CO2浓度下水分亏缺对冬小麦叶面积指数的影响差异.研究发现,CO2浓度升高对叶面积指数增长有促进作用,且在干旱情况下对叶面积指数的正效应比湿润情况下更为明显,在CO2浓度倍增条件下,发生水分亏缺的作物叶面积指数数倍增长.研究结论有助于分析CO2浓度变化对农作物生长过程的影响,为农田水分管理提供依据,又为估算叶面积指数提出了一种模型的方法.  相似文献

The effects of elevated CO2 and temperature on principal carbon constituents (PCC) and C and N allocation between needle, woody (stem and branches) and root tissue of Pseudotsuga menziesii Mirb. Franco seedlings were determined. The seedlings were grown in sun‐lit controlled‐environment chambers that contained a native soil. Chambers were controlled to reproduce ambient or ambient +180 ppm CO2 and either ambient temperature or ambient +3.5 °C for 4 years. There were no significant CO2 × temperature interactions; consequently the data are presented for the CO2 and temperature effects. At the final harvest, elevated CO2 decreased the nonpolar fraction of the PCC and increased the polar fraction and amount of sugars in the needles. In contrast, elevated temperature increased the nonpolar fraction of the PCC and decreased sugars in needles. There were no CO2 or temperature effects on the PCC fractions in the woody tissue or root tissue. Elevated CO2 and temperature had no significant effects on the C content of any of the plant tissues or fractions. In contrast, the foliar N content declined under elevated CO2 and increased under elevated temperature; there were no significant effects in other tissues. The changes in the foliar N concentrations were in the cellulose and lignin fractions, the fractions, which contain protein, and are the consequences of changes in N allocation under the treatments. These results indicate reallocation of N among plant organs to optimize C assimilation, which is mediated via changes in the selectivity of Rubisco and carbohydrate modulation of gene expression.  相似文献

Seedling structure influences tree structure and function, ultimately determining the potential productivity of trees and their competitiveness for resources. We investigated changes in shoot structure for seedlings of Pseudotsuga menziesii (Douglas-fir) grown under climate change scenarios of ambient or elevated CO2 (+180 mol mol–1) plus ambient or elevated temperature (+3.5°C), for 4 years in outdoor, sunlit chambers. Mass allocation and allometry were measured for buds, leaves, branches, and stems, and anatomy was evaluated for leaves and stems. Seedlings became more xeromorphic with elevated temperature: allocation of total mass to branches over stems and leaves increased, sapwood area to height ratio increased, number of growing points relative to seedling size increased, and stem and branch length and mass decreased for sections initiated during the three full CO2 and temperature seasons. Neither stem nor leaf anatomy was affected by elevated temperature. Elevated CO2 increased specific mass of leaves, but had few other effects on mass allocation, allometry, or anatomy for any shoot organ. There were no CO2 × temperature interactions for any important parameter. Thus, under realistic simulated field environmental conditions representative for in at least some P. menziesii forests (i.e., OR, USA, forests with limited soil nitrogen and summer soil moisture), elevated temperature, but not elevated CO2, may affect seedling shoot structure and, hence, function.  相似文献

Predicted climate changes in the US Central Plains include altered precipitation regimes with increased occurrence of growing season droughts and higher frequencies of extreme rainfall events. Changes in the amounts and timing of rainfall events will likely affect ecosystem processes, including those that control C cycling and storage. Soil carbon dioxide (CO2) flux is an important component of C cycling in terrestrial ecosystems, and is strongly influenced by climate. While many studies have assessed the influence of soil water content on soil CO2 flux, few have included experimental manipulation of rainfall amounts in intact ecosystems, and we know of no studies that have explicitly addressed the influence of the timing of rainfall events. In order to determine the responses of soil CO2 flux to altered rainfall timing and amounts, we manipulated rainfall inputs to plots of native tallgrass prairie (Konza Prairie, Kansas, USA) over four growing seasons (1998–2001). Specifically, we altered the amounts and/or timing of growing season rainfall in a factorial combination that included two levels of rainfall amount (100% or 70% of naturally occurring rainfall quantity) and two temporal patterns of rain events (ambient timing or a 50% increase in length of dry intervals between events). The size of individual rain events in the altered timing treatment was adjusted so that the quantity of total growing season rainfall in the ambient and altered timing treatments was the same (i.e. fewer, but larger rainfall events characterized the altered timing treatment). Seasonal mean soil CO2 flux decreased by 8% under reduced rainfall amounts, by 13% under altered rainfall timing, and by 20% when both were combined (P<0.01). These changes in soil CO2 flux were consistent with observed changes in plant productivity, which was also reduced by both reduced rainfall quantity and altered rainfall timing. Soil CO2 flux was related to both soil temperature and soil water content in regression analyses; together they explained as much as 64% of the variability in CO2 flux across dates under ambient rainfall timing, but only 38–48% of the variability under altered rainfall timing, suggesting that other factors (e.g. substrate availability, plant or microbial stress) may limit CO2 flux under a climate regime that includes fewer, larger rainfall events. An analysis of the temperature sensitivity of soil CO2 flux indicated that temperature had a reduced effect (lower correlation and lower Q10 values) under the reduced quantity and altered timing treatments. Recognition that changes in the timing of rainfall events may be as, or more, important than changes in rainfall amount in affecting soil CO2 flux and other components of the carbon cycle highlights the complex nature of ecosystem responses to climate change in North American grasslands.  相似文献
