Of the dewatering characteristics of activated sludges in our previous paper, the dewatering rate of sludge decreased in proportion to increasing amounts of extracellular polymer. As extracellular polymer in activated sludge was one of the important factors in the dewatering process, the change of the dewatering characteristics of thermochemically treated sludge (containing extracellular polymer) were compared with that of extracellular polymer extracted from sludge. The ultrafiltration rate of extracellular polymer flocculated by thermochemical treatment was much faster than that without treatment, indicating improved dewatering characteristics. Under the same treatment conditions, the dewatering characteristics of sludge were also much improved. The addition of the extracellular polymer treated thermochemically below pH 3 had no effect on the dewatering characteristics of the sludge. The addition of a flocculant to the thermochemically treated sludge was found to further improve the dewatering characteristics. The thermochemical treatment under low pH condition facilitated the flaking of the cake from the filter.  相似文献

Applications of nuclear magnetic resonance (NMR) to study a variety of physiological and biochemical aspects of bacteria with a role in the sulfur cycle are reviewed. Then, a case-study of high resolution13 C-NMR spectroscopy on sludges from bioreactors used for treating sulfate and sulfide rich wastewaters is presented.13 C-NMR was used to study the effect of sulfate and butyrate on propionate conversion by mesophilic anaerobic (methanogenic and sulfate reducing) granular sludge and microaerobic (sulfide oxidizing) flocculant sludge. In the presence of sulfate, propionate was degraded via the randomising pathway in all sludge types investigated. This was evidenced by scrambling of [3-13C]propionate into [2-13C]propionate and the formation of acetate equally labeled in the C1 and C2 position. In the absence of sulfate, [3-13C]propionate scrambled to a lesser extend without being degraded further. Anaerobic sludges converted [2,3-13C]propionate partly into the higher fatty acid 2-methyl[2,3-13C]butyrate during the simultaneous degradation of [2,3-13C]propionate and butyrate. [4,5-13C]valerate was also formed in the methanogenic sludges. Up to 10% of the propionate present was converted via these alternative degradation routes. Labeled butyrate was not detected in the incubations, suggesting that reductive carboxylation of propionate does not occur in the sludges.  相似文献

Five different bacterial extracellular polymer extraction methods and a combination of two of these methods were compared on cultures of activated sludge, synthetic activated sludge, and Klebsiella aerogenes. High-speed centrifugation was the most effective extraction method for the K. aerogenes culture, based on the comparatively small amount of cell disruption and the relatively high extracellular polymer yield. Steaming treatment was the most effective extraction method for the activated sludges, since it released a significant quantity of extracellular polymers from the flocs and caused less cellular disruption than ethylenediaminetetraacetic acid and sodium hydroxide treatments. Sodium hydroxide treatment caused extensive disruption in all cultures. Ultrasonication released low concentrations of extracellular polymers from all cultures. However, it caused no significant cell disruption and therefore may be useful as a preliminary treatment in conjunction with another extraction method.  相似文献

The sludge digestate stabilized by mesophilic anaerobic digestion was further degraded through thermophilic anaerobic digestion using 0–10 % (v/v) of thermophilic, proteolytic Coprothermobacter proteolyticus, and/or methanogenic granular sludge. The results demonstrated that the temperature shift to thermophilic condition promoted abiotic solubilization of proteins and reactivated the fermentative bacteria and methanogens indigenous in the sludge digestate, resulting in a final methane yield of 6.25 mmol-CH4/g-volatile suspended solid (VSS) digestate. The addition of C. proteolyticus accelerated the hydrolysis and fermentation of proteins and polysaccharides in the digestate during the early stage of thermophilic anaerobic digestion and stimulated methane production by syntrophic cooperation with methanogenic granular sludge. In the treatment with granular sludge and inoculated with 10 % (v/v) of C. proteolyticus, a final methane yield of 7 mmol-CH4/g-VSS digestate was obtained, and 48.4 % proteins and 27.0 % polysaccharides were degraded. The dissolved proteins were contributed by abiotic factor, C. proteolyticus, and indigenous digestate bacteria, respectively, by around 16, 28, and 56 %.  相似文献

The characteristics of the extracellular polymeric substances (EPS) extracted with nine different extraction protocols from four different types of anaerobic granular sludge were studied. The efficiency of four physical (sonication, heating, cationic exchange resin (CER), and CER associated with sonication) and four chemical (ethylenediaminetetraacetic acid, ethanol, formaldehyde combined with heating, or NaOH) EPS extraction methods was compared to a control extraction protocols (i.e., centrifugation). The nucleic acid content and the protein/polysaccharide ratio of the EPS extracted show that the extraction does not induce abnormal cellular lysis. Chemical extraction protocols give the highest EPS extraction yields (calculated by the mass ratio between sludges and EPS dry weight (DW)). Infrared analyses as well as an extraction yield over 100% or organic carbon content over 1 g g−1 of DW revealed, nevertheless, a carry-over of the chemical extractants into the EPS extracts. The EPS of the anaerobic granular sludges investigated are predominantly composed of humic-like substances, proteins, and polysaccharides. The EPS content in each biochemical compound varies depending on the sludge type and extraction technique used. Some extraction techniques lead to a slightly preferential extraction of some EPS compounds, e.g., CER gives a higher protein yield.  相似文献

The input impedance of muscle fibers of the crab was determined with microelectrodes over the frequency range 1 cps to 10 kc/sec. Care was taken to analyze, reduce, and correct for capacitive artifact. One dimensional cable theory was used to determine the properties of the equivalent circuit of the membrane admittance, and the errors introduced by the neglect of the three dimensional spread of current are discussed. In seven fibers the equivalent circuit of an element of the membrane admittance must contain a DC path and two capacitances, each in series with a resistance. In two fibers, the element of membrane admittance could be described by one capacitance in parallel with a resistance. In several fibers there was evidence for a third very large capacitance. The values of the elements of the equivalent circuit depend on which of several equivalent circuits is chosen. The circuit (with a minimum number of elements) that was considered most reasonably consistent with the anatomy of the fiber has two branches in parallel: one branch having a resistance Re in series with a capacitance Ce; the other branch having a resistance Rb in series with a parallel combination of a resistance Rm and a capacitance Cm. The average circuit values (seven fibers) for this model, treating the fiber as a cylinder of sarcolemma without infoldings or tubular invaginations, are Re = 21 ohm cm2; Ce = 47 µf/cm2; Rb = 10.2 ohm cm2; Rm = 173 ohm cm2; Cm = 9.0 µf/cm2. The relation of this equivalent circuit and another with a nonminimum number of circuit elements to the fine structure of crab muscle is discussed. In the above equivalent circuit Rm and Cm are attributed to the sarcolemma; Re and Ce, to the sarcotubular system; and Rb, to the amorphous material found around crab fibers. Estimates of actual surface area of the sarcolemma and sarcotubular system permit the average circuit values to be expressed in terms of unit membrane area. The values so expressed are consistent with the dielectric properties of predominantly lipid membranes.  相似文献

The effects of the cationic surfactant (cationic cetyl trimethyl ammonium bromide, CTAB) on the improvement of the sludge dewaterability using the cationic flocculant (cationic polyacrylamide, CPAM) were analyzed. Residual turbidity of supernatant, dry solid (DS) content, extracellular polymeric substances (EPS), specific resistance to filtration (SRF), zeta potential, floc size, and settling rate were investigated, respectively. The result showed that the CTAB positively affected the sludge conditioning and dewatering. Compared to not using surfactant, the DS and the settling rate increased by 8%–21.2% and 9.2%–15.1%, respectively, at 40 mg·L−1 CPAM, 10×10−3 mg·L−1 CTAB, and pH 3. The residual turbidities of the supernatant and SRF were reduced by 14.6%–31.1% and 6.9%–7.8% compared with turbidities and SRF without surfactant. Furthermore, the release of sludge EPS, the increases in size of the sludge flocs, and the sludge settling rate were found to be the main reasons for the CTAB improvement of sludge dewatering performance.  相似文献

Stem cutin from P. radiata seedlings grown under winter and summer environmental conditions comprised n-alkanoic, (C10–C26), α, ω-alkanedioic (C14–C22), ω-hydroxyalkanoic (C12–C24), hydroxy-α, ω-alkanedioic and polyhydroxyalkanoic acids. 9-Hydroxyheptadecane-1, 17-dioic, 9-hydroxyoctadecene-1, 18-dioic, 9-hydroxynonadecane-1, 19-dioic, and 10, 17-dihydroxyheptadecanoic acids are newly-identified constituents of gymnosperm cutin. Cutin grown under winter temperatures and photoperiod contained twice the amount of 9, 16-dihydroxyhexadecanoic acid than that in summer-grown cutin, suggesting that the winter-grown cutin was formed from a highly cross-linked polymer, and that summer-grown cutin contained more linear polyester portions in the polymer.  相似文献

Several benthic cyanobacteria were found to produce significant amounts of extracellular flocculants. The macromolecular flocculants produced by Phormidium sp. strain J-1 and Anabaenopsis circularis PCC 6720 were characterized. The Phormidium flocculant is a sulfated heteropolysaccharide to which fatty acids and protein are bound. The polysaccharide backbone is composed of uronic acids, rhamnose, mannose, and galactose. The A. circularis flocculant is also an acidic polysaccharide containing keto acid residues and neutral sugars, but to which no fatty acids, proteins, or sulfates are linked. Both flocculants could be recovered from growth medium by precipitation with cetyltrimethylammonium bromide and were found to bind the cationic dye Alcian-blue in a linear proportion to their concentration in solution. The latter property was used to quantify flocculant concentrations in culture supernatants and natural water samples and to compute their anion densities.  相似文献

Biosynthetically radiolabelled heparan sulphate proteoglycans have been isolated from the growth medium and the cell lysate of a human neuroblastoma cell line (CHP100). Chromatography on Sepharose CL-4B identified two heparan sulphate proteoglycans in the medium (Kav 0.220 and 0.3890, whereas in the cell lysate the major proteoglycan species were more heterogenous and of a smaller overall molecular size (Kav 0.407) than the medium-derived counterparts. Chromatography on Sepharose CL-6B of free heparan sulphate glycosaminoglycan chains showed that the majority of cell-layer-derived material heparan sulphate 2, Kav=0.509) was smaller than medium heparan sulphates (heparan sulphate 1 and heparan sulphate 2, Kav 0.230 and 0.317). Analysis of the patterns of polymer sulphation by nitrous acid treatment, gel chromatography and high-voltage electrophoresis established that in each heparan sulphate fraction there was on average 1.1 sulphate residues per disaccharide with an N:O sulphate ratio of 1.1 Heparan sulphate in the medium had a high proportion of di-O-sulphated disaccharides in regions of the chain with repeat disaccharide sequences of structure GlcA-GlcNSO3, whereas cell-associated material was enriched in di-O-sulphated tetrasaccharides of alternating sequences GlcA-GlcNAc-GlcA-GlcNSO3. The identification of several populations of heparan sulphate proteoglycans differing in molecular size and glycosaminoglycan fine structure may reflect the functional diversity of this family of macromolecules in the nervous system.  相似文献

The distribution and phylogenetic affiliations of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV)-degrading denitrifying bacteria in activated sludge were studied by a polyphasic approach including culture-independent biomarker and molecular analyses as well as cultivation methods. A total of 23 strains of PHBV-degrading denitrifiers were isolated from activated sludges from different sewage treatment plants. 16S ribosomal DNA (rDNA) sequence comparisons showed that 20 of the isolates were identified as members of the family Comamonadaceae, a major group of β-Proteobacteria. When the sludges from different plants were acclimated with PHBV under denitrifying conditions in laboratory scale reactors, the nitrate removal rate increased linearly during the first 4 weeks and reached 20 mg NO3-N h−1 g of dry sludge−1 at the steady state. The bacterial-community change in the laboratory scale sludges during the acclimation was monitored by rRNA-targeted fluorescence in situ hybridization and quinone profiling. Both approaches showed that the population of β-Proteobacteria in the laboratory sludges increased sharply during acclimation regardless of their origins. 16S rDNA clone libraries were constructed from two different acclimated sludges, and a total of 37 clones from the libraries were phylogenetically analyzed. Most of the 16S rDNA clones were grouped with members of the family Comamonadaceae. The results of our polyphasic approach indicate that β-Proteobacteria, especially members of the family Comamonadaceae, are primary PHBV-degrading denitrifiers in activated sludge. Our data provide useful information for the development of a new nitrogen removal system with solid biopolymer as an electron donor.  相似文献

This study followed the uptake, distribution, and elimination of sodium arsenate administered in a single dose and in multiple doses, respectively, to Iranian dairy sheep and goats. In the single dosing study, the blood concentration data fit an open two-compartment model of the form:C b (t)=?(A+B)e ?kat +Ae ?αt +Be ?βt Absorption distribution and elimination rate constants were statistically significantly different for the two animal species. In the multiple dosing study, arsenic accumulated in the blood of both animal species, as expressed by a one compartment model of the form:C t =C ss (1-e ?kt ) Arsenic was eliminated rapidly at the termination of dosing, with the blood washout half-life being shorter in sheep than in goats. This study extracted the soluble microbial products and loosely bound and tightly bound extracellular polymeric substances (EPS) from suspended sludge from a membrane bioreactor, original and aerobically/anaerobically digested, and compared their fouling potentials on a microfiltration membrane. The resistance of cake layer accounts for 95–98% of the total filtration resistances when filtering the whole sludges, with anaerobically digested sludge presenting the highest resistance among the three tested sludges. The tightly bound EPS has the highest potential to foul the membrane; however, the loosely bound EPS contribute most of the filtration resistances of the whole sludges. The foulants corresponding to the irreversible fouling have chemical fingerprints similar to those from loosely bound EPS, which have a greater predilection to proteins and humic substances than to polysaccharides.  相似文献

Bioethanol and enzymes were produced from fiber sludges through sequential microbial cultivations. After a first simultaneous saccharification and fermentation (SSF) with yeast, the bioethanol concentrations of sulfate and sulfite fiber sludges were 45.6 and 64.7 g/L, respectively. The second SSF, which included fresh fiber sludges and recycled yeast and enzymes from the first SSF, resulted in ethanol concentrations of 38.3 g/L for sulfate fiber sludge and 24.4 g/L for sulfite fiber sludge. Aspergillus niger carrying the endoglucanase-encoding Cel7B gene of Trichoderma reesei was grown in the spent fiber sludge hydrolysates. The cellulase activities obtained with spent hydrolysates of sulfate and sulfite fiber sludges were 2,700 and 2,900 nkat/mL, respectively. The high cellulase activities produced by using stillage and the significant ethanol concentrations produced in the second SSF suggest that onsite enzyme production and recycling of enzyme are realistic concepts that warrant further attention.  相似文献

Eighteen hair samples from Karakul newborn lambs with various colors were estimated for eumelanin and pheomelanin contents (Ce and Cp, respectively) by electron spin resonance (ESR) spectrometry and by high-performance liquid chromatography (HPLC). Correlation coefficients between the values estimated by the ESR and HPLC methods were 0.96, 0.93, and 0.99 for Ce, Cp, and Ce/Cp, respectively. The high correlation coefficients show that both methods fit well for estimation of relative values of these parameters. The absolute values of Ce and Ce/Cp coincide rather well when Ce is high, but considerable discrepancies appear when Ce is low. The reasons for these discrepancies are discussed. The HPLC method appears to be more sensitive for detection of low concentrations of pheomelanin, while the ESR method fits well for mass selection purposes.  相似文献

Trifunctional dialkyl [1,2-bis(diethylcarbamoyl)- ethyl] phosphonates, (RO)2P(O)CH[C(O)N(C2H5)2]- [CH2C(O)N(C2H5)2] R  CH3, C2H5, i-C3H7, n-C6H13 were prepared from the respective sodium salts, Na[(RO)2P(O)CHC(O)N(C2H5)2] and N,N- diethylchloroacetamide, and they were characterized by elemental analysis, mass, infrared and NMR spectroscopy. The molecular structure of (i-C3H7O)2- P(O)CH[C(O)N(C2H5)2][CH2C(O)N(C2H5)2] was determined by single crystal X-ray diffraction analysis and found to crystallize in the monoclinic space group P21/c with a=15.589(6), b=9.783(4), c= 16.283(7) Å, β = 110.90(3)°, Z = 4 and V= 2320(2) Å3. The structure was solved by direct methods and blocked least-squares refinement converged with Rf = 5.7% and RwF= 4.4% on 2266 unique data with F>4σ(F). Important bond distances include PO 1.459(3) Å, CHCO 1.228(3) Å and CHCH2CO 1.223(3) Å. The coordination chemistry of the ligand with several lanthanides was examined, and the structure of the complex Gd(NO3)3{[(i-C3H7O)2P(O)CH[C(O)N(C2H5)2][CH2C(O)N(C2H5)2]}2·H2O was determined. The complex crystallized in the monoclinic space group P21/n with a = 13.524(5), b = 22.033(4), c = 19.604(4) Å β = 106.22(2)°, Z = 4 and V= 5609(3) Å3. The structure was solved by heavy atom techniques and blocked least-squares refinement converged with RF = 5.9% and RwF = 4.1% on 5275 reflections with F > 4σ(F). Both trifunctional ligands were found to bond to Gd(III) through only the phosphoryl oxygen atoms. The remainder of the Gd coordination sphere was composed of three bidentate nitrate oxygen atoms and an oxygen bonded water molecule. Several important bond distances include GdO(phosphoryl)av = 2.343(5) Å, GdO(nitrate)av = 2.475(7) Å, GdO(water) = 2.354(5) Å, PO(phosphoryl)av = 1.467(6) Å, CHCOav = 1.242(10) Å and CHCH2COav = 1.209(11) Å.  相似文献

Nucleic acid duplexes featuring a single alpha-anomeric thymidine inserted into each DNA strand via 3′-3′ and 5′-5′ phosphodiester linkages exhibit local conformational dynamics that are not adequately depicted by conventional restrained molecular dynamics (rMD) methods. We have used molecular dynamics with time-averaged NMR restraints (MDtar) to explore its applicability to describing the conformational dynamics of two α-containing duplexes – d(GCGAAT-3′-3′-αT-5′-5′-CGC)2 and d(ATGG-3′-3′-αT-5′-5′-GCTC)?r(gagcaccau). In contrast to rMD, enforcing NOE-based distance restraints over a period of time in MDtar rather than instantaneously results in better agreement with the experimental NOE and J-data. This conclusion is based on the dramatic decreases in average distance and coupling constant violations (Δd av, J rms, and ΔJ av) and improvements in sixth-root R-factors (R x). In both duplexes, the deoxyribose ring puckering behavior predicted independently by pseudorotation analysis is portrayed remarkably well using this approach compared to rMD. This indicates that the local dynamic behavior is encoded within the NOE data, although this is not obvious from the local R x values. In both systems, the backbone torsion angles comprising the 3′-3′ linkage as well as the (high S-) sugars of the α-nucleotide and preceding residue (α?1) are relatively static, while the conformations of the 5′-5′ linkage and the sugar in the neighboring β-nucleotide (α+1) show enhanced flexibility. To reduce the large ensembles generated by MDtar to more manageable clusters we utilized the PDQPRO program. The resulting PDQPRO clusters (in both cases, 13 structures and associated probabilities extracted from a pool of 300 structures) adequately represent the structural and dynamic characteristics predicted by the experimental data.  相似文献

Various alkyltrimethylammonium bromides ATABs, R = C1 to C6 were added to a mixed liquor of sewage activated sludge and the effects on settling properties were investigated. The 30-min settled sludge volume decreased from 28% to 24, 20, 17, and 15% on addition of 1 mM C10, C12, C14, and C16 compounds to mixed liquor (sludge concentration, 1.28 g/l), respectively. With increasing alkyl chain length, the amounts of ATABs adsorbed by the sludge, and metal ions released from the sludge, increased. However, a quantitative correlation was not observed between the two. In addition, the adsorption of ATABs resulted in a small decrease in sludge wet weight. However, such a change was not observed on addition of ATABs bearing short alkyl chains. A good correlation was observed between the amounts of ATABs adsorbed and their organic solvent/water partition coefficients. These results indicated that the effectiveness of ATABs as sludge conditioners depends on their hydrophobicity.  相似文献

Chemical changes of polysaccharides and lignin in Prunus armeniaca decayed by the ascomycete fungus Hypocrea sulphurea were investigated. Solid-state 13C NMR spectra showed that polysaccharides were the main components of fresh and decayed wood. Decomposition of cellulose and hemicellulose by the fungus was minimal although a slight preference for crystalline as compared with amorphous cellulose was recognised. Comparison of the signal intensity of the resonance at 145 ppm in fresh and decomposed wood suggested that the fungus had decomposed tannin constituents. Thermochemolysis with tetramethylammonium hydroxide (TMAH) and product identification by gas chromatography–mass spectrometry revealed that the ratio of syringyl-type (S) units to guaiacyl-type (G) units decreased from 1.8 to 1.1 following fungal attack. Increases in both guaiacyl and syringyl acid-aldehyde ratios (Ad/Al)G, (Ad/Al)S together with an increase from 0.82 to 3.54 in the ratio of methyl 3,4-dimethoxybenzoate to the sum of 1-(3,4-dimethoxyphenyl)-1,2,3-trimethoxypropane (threo and erythro-isomers) ΓG from the decayed wood confirmed oxidative Cα–Cβ cleavage for the mechanism of lignin decay by this ascomycete.  相似文献

Understanding the influences of climatic changes on water use efficiency (WUE) of Tibetan alpine meadows is important for predicting their long-term net primary productivity (NPP) because they are considered very sensitive to climate change. Here, we collected wool materials produced from 1962 to 2010 and investigated the long-term WUE of an alpine meadow in Tibet on basis of the carbon isotope values of vegetation (δ 13Cveg). The values of δ 13Cveg decreased by 1.34‰ during 1962–2010, similar to changes in δ 13C values of atmospheric CO2. Carbon isotope discrimination was highly variable and no trend was apparent in the past half century. Intrinsic water use efficiency (W i) increased by 18 μmol·mol–1 (approximately 23.5%) during 1962–2010 because the increase in the intercellular CO2 concentration (46 μmol·mol–1) was less than that in the atmospheric CO2 concentration (C a, 73 μmol·mol–1). In addition, W i increased significantly with increasing growing season temperature and C a. However, effective water use efficiency (W e) remained relatively stable, because of increasing vapor pressure deficit. C a, precipitation, and growing season temperature collectively explained 45% of the variation of W e. Our findings indicate that the W e of alpine meadows in the Tibetan Plateau remained relatively stable by physiological adjustment to elevated C a and growing season temperature. These findings improve our understanding and the capacity to predict NPP of these ecosystems under global change scenarios.  相似文献
