DNA分子标记技术为真菌系统进化研究提供了许多新的方法,真菌分子系统学已成为一门成熟的学科。简述了真菌分子系统学的发展简史和代表性的研究方法以及对真菌系统学的主要贡献,包括将广义的真菌划分为3个类群,粘菌和卵菌不再属于真菌界成员。真菌生命之树项目的研究结果对真菌界高阶分类系统作出重大调整,将先前的4个门(壶菌门、接合菌门、子囊菌门和担子菌门)变为7个门(微孢子虫门、壶菌门、新丽鞭毛菌门、芽枝霉门、球囊菌门、子囊菌门和担子菌门)和4个亚门,并对真菌各类群概念作出修订。此外,DNA分子标记技术对真菌种概念的认识、有性型-无性型关联及分子生态学等研究领域产生了重要影响。  相似文献

Root-associated fungi, including ectomycorrhizal and root-endophytic fungi, are among the most diverse and important belowground plant symbionts in dipterocarp rainforests. Our study aimed to reveal the biodiversity, host association, and community structure of ectomycorrhizal Basidiomycota and root-associated Ascomycota (including root-endophytic Ascomycota) in a lowland dipterocarp rainforest in Southeast Asia. The host plant chloroplast ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit (rbcL) region and fungal internal transcribed spacer 2 (ITS2) region were sequenced using tag-encoded, massively parallel 454 pyrosequencing to identify host plant and root-associated fungal taxa in root samples. In total, 1245 ascomycetous and 127 putative ectomycorrhizal basidiomycetous taxa were detected from 442 root samples. The putative ectomycorrhizal Basidiomycota were likely to be associated with closely related dipterocarp taxa to greater or lesser extents, whereas host association patterns of the root-associated Ascomycota were much less distinct. The community structure of the putative ectomycorrhizal Basidiomycota was possibly more influenced by host genetic distances than was that of the root-associated Ascomycota. This study also indicated that in dipterocarp rainforests, root-associated Ascomycota were characterized by high biodiversity and indistinct host association patterns, whereas ectomycorrhizal Basidiomycota showed less biodiversity and a strong host phylogenetic preference for dipterocarp trees. Our findings lead to the working hypothesis that root-associated Ascomycota, which might be mainly represented by root-endophytic fungi, have biodiversity hotspots in the tropics, whereas biodiversity of ectomycorrhizal Basidiomycota increases with host genetic diversity.  相似文献

Submerged cultures of some 1500 Ascomycota and Basidiomycota isolated from their fruit-bodies or as soil-borne, coprophilous or endophytic fungi were screened for activity against Candida albicans and a range of other pathogenic and saprotrophic fungi. Considerably more Ascomycota (11-16%) than Basidiomycota (3.5%) produced metabolites with activity against C. albicans. From five species of endophytes, six bioactive compounds were isolated and identified, viz. cerulenin (1), arundifungin (2), sphaeropsidin A (3), 5-(1,3-butadiene-1-yl)-3-(propene-1-yl)-2-(5H)-furanone (4), ascosteroside A (formerly called ascosteroside; 5) and a derivative of 5, ascosteroside B (6). 1, 3 and 5 were isolated from fungi belonging to different orders than previously described producers. Antifungal activities of 2 and 4-6 in the agar diffusion test were comparable with those of amphotericin B. Compound 6 exhibited a similar antifungal activity as 5 but its cytotoxicity towards Hep G2 cells was considerably lower. This study points to endophytic fungi related to hemibiotrophic or latent plant pathogens as an important source of bio- and chemodiversity.  相似文献

Benthic fungal diversities in Bohai Sea and North Yellow Sea were investigated using pyrosequencing of 18S rDNA. Overall, Chytridiomycota dominated, followed by Basidiomycota, Ascomycota and Cryptomycota, in terms of alpha diversities and relative abundance. The beta diversity of benthic fungi showed a significant seasonality but no regional differences, accounted for by contrasting relative abundances of Chytridiomycota and Basidiomycota. Significantly seasonal changes in Chytridiomycota and Basidiomycota assemblage structure were also observed, but not for Ascomycota and Cryptomycota. Environmental filtering was more important than water depth and geographic distance in shaping the distribution of benthic fungi in the neritic oceans. The overall fungal beta diversity co-varied with concentration of chlorophyll a, pH, and salinity, distance from land, and water depth. The assemblage structure of benthic Chytridiomycota, Basidiomycota, Ascomycota and Cryptomycota co-varied with different sets of environmental parameters, suggesting their niche differentiations in the coastal sediments.  相似文献

Fungi are ubiquitous with many capable of causing disease by direct infection, toxicoses, or allergy. Fungal spores are present in outdoor air throughout the year, yet airborne diversity is poorly characterised. Airborne fungal spores are routinely counted by microscopy, enabling identification to genera at best. We generated traditional microscopic counts over a year, then used environmental sequencing techniques to assess and compare 3 d selected from the main fungal spore season. The days selected corresponded to one with a high quantity of spores unidentifiable by microscopy, and two representing dry and wet summer periods. Over 86 % of genera detected by sequencing were not routinely identifiable by microscopy. A high degree of temporal variability was detected, with the percentage of clones attributed to Basidiomycota or Ascomycota, and composition of genera within each phylum varying greatly between days. Throughout the year Basidiomycota spores were found at higher levels than Ascomycota, but levels fluctuated daily with Ascomycota comprising 11-84 % of total spores and Basidiomycota 7-81 %. No significant difference was found between the proportion of clones attributed to each morphological group detected by sequencing to that counted by microscopy (P = 0.477, 0.985, and 0.561). The majority of abundant genera detected by DNA analysis are not routinely identified by microscopy (>75 %). Of those, several are known human and plant pathogens, and may represent unrecognised aeroallergens.  相似文献

Cryopreservation of filamentous micromycetes and yeasts using perlite   总被引:2,自引:2,他引:0  
The viability, growth and morphology of 48 strains of Ascomycota (including 17 yeasts) and 20 strains of Zygomycota were determined after a 2-d and then after 1-year storage in liquid nitrogen using a new cryopreservation method with perlite as a particulate solid carrier. In case of Ascomycota, 45 strains (94 %) out of 48 survived both 2-d and 1-year storage in liquid nitrogen, respectively. In case of Zygomycota, all 20 strains survived both storage. In addition, 3 strains of Basidiomycota counted among yeasts were tested and all survived the 1 year storage. In all surviving cultures no negative effects of cryopreservation by this method have been observed after 1-year of storage in liquid nitrogen. The results indicate that the perlite protocol can be successfully used for cryopreservation of taxonomically different groups of fungi and also for fungi which failed to survive other routinely used preservation procedures.  相似文献

One hundred and eleven strains of Basidiomycota, 39 strains of Ascomycota and 2 strains of Mucoromycotina belonging to wood decomposers that cause white-rot (WR) or brown-rot (BR), other wood associated saprotrophs (WA), litter decomposing cord-forming Basidiomycota (LDF), and saprotrophic microfungi (SA), were screened for the production of hydrolytic enzymes and laccase. The presence of enzyme-encoding genes was also analysed in the published genomes of saprotrophic fungi. Several genes, including those for acidic phosphatase, β-glucosidase and N-acetylglucosaminidase, were common in the genomes with enzyme activity widely displayed by fungi, while other enzymes, such as certain hemicellulases or laccase, were produced less frequently. Enzyme production by saprotrophic fungi was shaped by the combination of their ecophysiology and taxonomy. Basidiomycota exhibited higher activities of all enzymes, except alkaline phosphatase, α-glucosidase, N-acetylglucosaminidase, α-mannosidase and α-fucosidase, than Ascomycota. The SA and BR fungi showed distinct enzyme production patterns, while the enzyme production by WR, LDF and WA was similar. Differences among species were typically reflected in the level of enzyme activity rather than in the absence of enzymes. Enzyme screening results showed that in several cases, fungi exhibited enzyme activity without the presence of the corresponding gene and vice versa. This indicates that the use of genome-derived information for the prediction of potential enzyme production has substantial limitations and cannot replace functional screening of fungal cultures.  相似文献

Horizontally transmitted fungal endophytes are an ecological group of fungi, mostly belonging to the Ascomycota, that reside in the aerial tissues and roots of plants without inducing any visual symptoms of their presence. These fungi appear to have a capacity to produce an array of secondary metabolites exhibiting a variety of biological activity. Although the ability of fungi to produce unique bioactive metabolites is well known, endophytes have not been exploited, perhaps because we are only beginning to understand their distribution and biology. This review emphasizes the need to routinely include endophytic fungi in the screening of organisms for bioactive metabolites and novel drugs; it also underscores the need to use information obtained concerning fungal secondary metabolite production from other groups of fungi for a targeted screening approach.  相似文献

Histidine kinases (HK) sense and transduce via phosphorylation events many intra‐ and extracellular signals in bacteria, archaea, slime moulds and plants. HK are also widespread in the fungal kingdom, but their precise roles in the regulation of physiological processes remain largely obscure. Expanding genomic resources have recently given the opportunity to identify uncharacterised HK family members in yeasts and moulds and now allow proposing a complex classification of Basidiomycota, Ascomycota and lower fungi HK. A growing number of genetic approaches have progressively provided new insight into the role of several groups of HK in prominent fungal pathogens. In particular, a series of studies have revealed that members of group III HK, which occur in the highest number of fungal species and contain a unique N‐terminus region consisting of multiple HAMP domain repeats, regulate morphogenesis and virulence in various human, plant and insect pathogenic fungi. This research field is further supported by recent shape‐function studies providing clear correlation between structural properties and signalling states in group III HK. Since HK are absent in mammals, these represent interesting fungal target for the discovery of new antifungal drugs.  相似文献

Fungal spores are widespread and common in the atmosphere. In this study, we use a metagenomic approach to study the fungal diversity in six total air samples collected from April to May 2012 in Seoul, Korea. This springtime period is important in Korea because of the peak in fungal spore concentration and Asian dust storms, although the year of this study (2012) was unique in that were no major Asian dust events. Clustering sequences for operational taxonomic unit (OTU) identification recovered 1,266 unique OTUs in the combined dataset, with between 223?96 OTUs present in individual samples. OTUs from three fungal phyla were identified. For Ascomycota, Davidiella (anamorph: Cladosporium) was the most common genus in all samples, often accounting for more than 50% of all sequences in a sample. Other common Ascomycota genera identified were Alternaria, Didymella, Khuskia, Geosmitha, Penicillium, and Aspergillus. While several Basidiomycota genera were observed, Chytridiomycota OTUs were only present in one sample. Consistency was observed within sampling days, but there was a large shift in species composition from Ascomycota dominant to Basidiomycota dominant in the middle of the sampling period. This marked change may have been caused by meteorological events. A potential set of 40 allergyinducing genera were identified, accounting for a large proportion of the diversity present (22.5?7.2%). Our study identifies high fungal diversity and potentially high levels of fungal allergens in springtime air of Korea, and provides a good baseline for future comparisons with Asian dust storms.  相似文献

The All-Russia Collection of Microorganisms comprises about 2500 yeast strains, which represent over 500 species of 100 genera belonging to 6 classes of Ascomycota and Basidiomycota. Type strains are available for almost all species. The isolates collected represent all the continents; however, the majority of isolates have been recovered in Europe. Differences in rates of nucleotide or amino acid substitutions among major groups of organisms are repeatedly found and well documented. A growing body of evidence suggests a link between the rate of neutral molecular change within populations and the evolution of species diversity. More than 98% of terrestrial fungi belong to the phyla Ascomycota or Basidiomycota. The former is considerably richer in number of species than the latter. We obtained DNA sequences of 21 protein-coding genes from the lichenized fungus Rhizoplaca chrysoleuca and used them together with sequences from GenBank for subsequent analyses. Three datasets were used to test rate discrepancies between Ascomycota and Basidiomycota and that within Ascomycota: (i) 13 taxa including 105 protein-coding genes, (ii) nine taxa including 21 protein-coding genes, and (iii) nuclear LSU rDNA of 299 fungal species. Based on analyses of the 105 protein-coding genes and nuclear LSU rDNA datasets, we found that the evolutionary rate was higher in Ascomycota than in Basidiomycota. The differences in substitution rates between Ascomycota and Basidiomycota were significant. Within Ascomycota, the species-rich Sordariomycetes has the fastest evolutionary rate, while Leotiomycetes has the slowest. Our results indicate that the main contribution to the higher substitution rates in Ascomycota does not come from mutualism, ecological conditions, sterility, metabolic rate or shorter generation time, but is possibly caused by the founder effect. This is another example of the correlation between species number and evolutionary rates, which is consistent with the hypothesis that the founder effect is responsible for accelerated substitution rates in diverse clades.  相似文献

为了解我国食药用菌资源的开发利用情况,需要对我国食药用菌栽培驯化情况进行统计分析。通过公共数据库关键词搜索,统计栽培驯化食药用菌种类,经过拉丁名以及中文名称的核实,本名录收录了192种驯化栽培的食药用菌,隶属于2门15目48科89属,包括子囊菌门(Ascomycota) 26个种,担子菌门(Basidiomycota)166个种,其中在科水平上,多孔菌科(Polyporaceae)、蘑菇科(Agaricaceae)、侧耳科(Pleurotaceae)以及泡头菌科(Physalacriaceae)种类最多;在属水平,侧耳属(Pleurotus)、蘑菇属(Agaricus)、木耳属(Auricularia)、灵芝属(Ganoderma)等栽培驯化种类较多。通过本名录发现,我国已经完成栽培驯化的食用菌种类繁多,还有大量食药用菌有开发利用前景。  相似文献

A plethora of bioactive plant metabolites has been explored for pharmaceutical, food chemistry and agricultural applications. The chemical synthesis of these structures is often difficult, so plants are favorably used as producers. While whole plants can serve as a source for secondary metabolites and can be also improved by metabolic engineering, more often cell or organ cultures of relevant plant species are of interest. It should be noted that only in few cases the production for commercial application in such cultures has been achieved. Their genetic manipulation is sometimes faster and the production of a specific metabolite is more reliable, because of less environmental influences. In addition, upscaling in bioreactors is nowadays possible for many of these cultures, so some are already used in industry. There are approaches to alter the profile of metabolites not only by using plant genes, but also by using bacterial genes encoding modifying enzymes. Also, strategies to cope with unwanted or even toxic compounds are available. The need for metabolic engineering of plant secondary metabolite pathways is increasing with the rising demand for (novel) compounds with new bioactive properties. Here, we give some examples of recent developments for the metabolic engineering of plants and organ cultures, which can be used in the production of metabolites with interesting properties.  相似文献

黄泥田土壤真菌群落结构和功能类群组成对施肥的响应   总被引:4,自引:0,他引:4  
研究长期不同施肥黄泥田土壤真菌群落组成和功能特性,深入认识真菌对不同施肥的响应机理,可以为合理施肥和保证农田土壤健康发展提供科学依据.设置不施肥(CK)、单施无机肥(NPK)、无机肥配施农家肥(NPKM)、无机肥加秸秆还田(NPKS)4个处理,采用Illumina高通量测序和FUNGuild对不同施肥处理下黄泥田土壤真菌群落结构和功能群进行分析.结果表明:从门水平上看,土壤真菌群落主要由子囊菌、担子菌和接合菌构成,且以子囊菌为主(47%~74%).NPKM和NPKS处理中子囊菌的相对丰度分别为49%和47%,显著低于CK(71%)和NPK(74%)处理,从目水平上看,减少的主要为肉座菌、格孢腔菌和散囊菌.NPKM和NPKS处理担子菌相对丰度分别为18%和28%,高于CK(14%)和NPK(10%)处理,从目水平上看,增加的主要为银耳菌、糙孢伏革菌和伞菌.与CK相比,单施无机肥降低了担子菌含量.不同施肥处理均在一定程度上增加了接合菌门的丰度,从目水平看,以内囊霉菌和粪蛙霉菌为主.NPK处理真菌α多样性指数显著低于其他处理,NPKM和NPKS处理的丰富度指数(Chao1和ACE)高于CK和NPK处理.不同处理之间真菌的营养类型以腐生营养型为主(48%~57%),NPKM和NPKS处理的共生营养型真菌相对丰度为17%,显著高于CK和NPK处理,增加的以丛枝菌根真菌、外生菌根真菌为主.NPK处理动物病原菌含量(10%)显著高于其他处理.冗余分析(RDA)结果表明,土壤含水量、孔隙度和盐度是影响真菌群落结构和功能类群组成的主要因子,其次为有机质和总氮.长期单施无机肥降低真菌种群多样性,增加致病菌含量,不利于土壤健康,而有机无机肥配施可在一定程度上提高真菌种群丰富度指数和共生真菌比例,有利于保持黄泥田稻田土壤健康的生态环境和真菌群落的多样性.  相似文献

More than 70 species of halotolerant and halophilic actinomycetes belonging to at least 24 genera have been validly described. Halophilic actinomycetes are a less explored source of actinomycetes for discovery of novel bioactive secondary metabolites. Degradation of aliphatic and aromatic organic compounds, detoxification of pollutants, production of new enzymes and other metabolites such as antibiotics, compatible solutes and polymers are other potential industrial applications of halophilic and halotolerant actinomycetes. Especially new bioactive secondary metabolites that are derived from only a small fraction of the investigated halophilic actinomycetes, mainly from marine habitats, have revealed the huge capacity of this physiological group in production of new bioactive chemical entities. Combined high metabolic capacities of actinomycetes and unique features related to extremophilic nature of the halophilic actinomycetes have conferred on them an influential role for future biotechnological applications.  相似文献

Traditional techniques for studying the fungal community composition in streams favour the detection and identification of aquatic hyphomycetes. Our objective was to use molecular techniques to determine the presence and contributions of other fungal groups. We designed primers specific for the ITS regions in Ascomycota, Basidiomycota, Chytridiomycota, Zygomycota and Oomycota. The primers were used to amplify DNA from linden, maple, and beech leaves, and birch wood submerged in a stream for 4 weeks in summer, autumn, winter and spring. The amplification products were separated by denaturing gradient gel electrophoresis. Ascomycota were present in large phylotype numbers (up to 21) on all substrates and all dates and represented ≥ 75 % of the fungal biomass. Basidiomycota were the second most abundant group in summer and autumn (up to 13 % on wood) and were absent only on linden and maple in spring. There were consistently large numbers of phylotypes from Chytridiomycota and their relative contribution to the microbial community peaked in winter on all substrates. Oomycota were present in summer and abundant only on wood. Zygomycota were present in low numbers and their estimated contribution to fungal biomass was ≤ 1%. Using primers to target individual groups facilitates a more balanced approach to studying fungal diversity in freshwater ecosystems.  相似文献

The cytoplasmic 5S ribosomal RNA sequence from Pneumocystis carinii was determined and compared with those of 382 eukaryotes and an evolutionary tree was constructed to establish the phylogenetic position of Pneumocystis. The data suggest that Pneumocystis is associated with the Rhizopoda/Myxomycota/Zygomycota group but not with common fungi, such as Ascomycota or Basidiomycota, nor with other protozoa.  相似文献

菌生真菌是指以其他真菌为宿主的真菌,是重要的自然生物资源.它们不是系统学上的分类群,而是特殊生境真菌,包括子囊菌、担子菌和接合菌的种类.其中菌生子囊菌是动植物以及其他真菌的内生菌、寄生菌或腐生菌,少数种己用于植物病害的生物防治,对其进行研究具有重要的理论意义和应用潜力.作者汇总了我国丰富的非地衣型菌生子囊菌资源,目前己报道该类真菌132种,其中以担子菌为宿主的约63种,以子囊菌为宿主的38种,其余或对基物的选择性不强或宿主真菌的分类地位不详,部分种类表现出对基物真菌或者宿主真菌的选择性.同时,对菌生真菌与宿主真菌相互作用方式、菌生子囊菌在植物病害防控中的应用以及少数种类对食用菌栽培的为害进行了简要概述.  相似文献
