达乌尔黄鼠(Spermophilusdauricus)是国内研究冬眠生理学的主要模式动物,且日益为医学研究所关注。实验室条件下繁殖种群具有明确生活史特征,是相关研究的理想对象,但以往研究中,达乌尔黄鼠实验室条件下繁殖成功率及后代成活率均很低。本研究通过采取丰富饲料种类、雄性出眠后的暖温暂养、“双笼配对”等措施,对实验室条件下越冬达乌尔黄鼠进行配对,从21对动物中成功繁殖出18窝子代,繁殖成功率85.7 %。78只子代饲养至3月龄,仍有67只存活,存活率为85.9 %。通过本实验,得出改进达乌尔黄鼠实验室条件下繁殖的主要措施如下:(1)繁殖期母鼠及发育期幼鼠,除饲喂标准大鼠饲料外,适量添加幼犬粮、胡萝卜,以补充营养;(2)出眠的雄性达乌尔黄鼠放入18℃房间暂养2周,有助于其性腺发育,促进繁殖成功;(3)“双笼配对”能有效地降低外界干扰,减少动物的杀婴行为。    相似文献

冬眠哺乳动物的肠道微生物会发生季节性变化,同时在冬眠期间动物处于禁食状态,对肠道微生物的多样性和组成也产生影响。本研究通过16S rRNA基因高通量测序分析达乌尔黄鼠育肥阶段 (起始育肥期、快速育肥期、育肥完成期) 和冬眠阶段 (冬眠早期、冬眠晚期、出眠期) 共6个时期盲肠菌群的多样性、组成和功能,并通过冗余分析 (RDA) 探究其生理特征与菌群组成和功能之间的关系,揭示达乌尔黄鼠盲肠菌群的季节性变化。菌群组成的分析显示达乌尔黄鼠盲肠菌群主要由厚壁菌门 (Firmicutes)、拟杆菌门 (Bacteroidetes) 和疣微菌门 (Verrucomicrobia) 组成。与其他时期相比,冬眠早期厚壁菌门的相对丰度减少,拟杆菌门和疣微菌门的相对丰度增加。在Alpha多样性中,起始育肥期、快速育肥期和冬眠早期的Chao1和ACE指数显著低于出眠期,育肥完成期的Simpson指数显著低于快速育肥期 (P < 0.05) 。通过加权和非加权的UniFrac距离矩阵的主坐标分析发现盲肠菌群均显示出了明显的季节性聚类。PICRUSt分析中,丁酸代谢等代谢通路在育肥阶段富集,冬眠阶段集中在氮代谢等相关通路中。RDA分析显示达乌尔黄鼠不同时期的生理特征与其盲肠菌群的组成和功能显著相关。本研究表明,冬眠使达乌尔黄鼠盲肠菌群的多样性和相对丰度发生改变,盲肠菌群组成和功能的变化调节了达乌尔黄鼠的生理代谢,使达乌尔黄鼠适应季节性的环境变化。  相似文献

Summary R. meliloti primarily nodulates Medicago sativa but cannot nodulate Macroptilium atropurpureum. By introducing an 11.4 kb region into R. meliloti from the Symplasmid of Rhizobium strain MPIK3030, the host range of the R. meliloti transconjugants were shown to be extended to M. atropurpureum, one of the hosts of MPIK3030 but not normally nodulated by R. meliloti. The region responsible for host range extension was isolated by mass conjugating a clone bank from MPIK3030 into the R. meliloti wild type, and subsequent screening for nodulation on M. atropurpureum. Using deleted derivatives of a plasmid reisolated from endosymbiotic bacteria, the host range region was further narrowed down to three EcoRI fragments. Tn5 mutagenesis allowed the isolation of three discrete regions on an 11.4 kb section, which are involved in the extension of host range to M. atropurpureum. Finally, complementation experiments performed with R. meliloti common nod and hsn mutants indicated that none of the genes involved in the early steps of nodulation, including host-range functions, can be complemented by genes carried on the 11.4 kb fragment derived from MPIK3030.  相似文献

Regulation of thermogenic activity and uncoupling protein1 (UCP1) expression in brown adipose tissue (BAT) were studied in euthermic Daurian ground squirrel after acute and chronic cold exposure at 4°C. The UCP1 concentration was indirectly determined by titration with its specific ligand [3H]-labeled GTP, and Ucp1 mRNA was detected by using a [32P]-labeled antisense oligonucleotide probe. Both acute and chronic cold exposure stimulated up-regulation of Ucp1 mRNA. Although UCP1 concentration is not significantly increased after 24 h of cold exposure, it is markedly elevated by 75% in squirrels after 4-week cold adaptation compared with controls raised at 22°C. Changes in T4 5′-deiodinase activity were closely associated with variations of Ucp1 mRNA level. Ucp1 gene expression is significantly affected by cold exposure in BAT from euthermic Daurian ground squirrels. In addition, the activation of T4 5′-deiodinase may be an important regulatory factor in cold-induced Ucp1 expression.  相似文献

Summary Site-directed Tn5 mutagenesis by gene replacement method, via homologous recombination, was used to identify symbiotically essential regions in the genome of cowpea Rhizobium spp. IRc78. Transposon insertions with-in the nifK hybridizing region or in the regions spanning 10 kb downstream of the nifK have revealed the presence of functional genes required for nitrogen fixation. Six single Tn5 insertions resulted in nod+ fix phenotypes and one in nod+ but reduced fix+ phenotype. All seven Tn5 insertions were stable before, during and after plant passage. However, IRc78 transconjugants containing duplicated nif copies, (a normal and a Tn5 inserted copy separated by vector sequences) were unstable. In five IRc78::Tn5 strains, the mutant phenotypes were corrected by an extrachromosomally stable vector containing wild type nif alleles. Our experiments suggest that the correction to nod+ fix+ phenotype is by complementation although correction by recombination cannot be completely excluded.  相似文献

Summary A 70 kbp segment of the megaplasmid from a broad host range Rhizobium strain (MPIK3030) was mapped with the aid of cosmid clones made in the vector pJB8. A 7.9 kbp EcoRI fragment from this region, 55 kbp away from the nif gene cluster, was shown to hybridize to the common nod genes from R. meliloti. Using several R. meliloti nod probes it was possible to delimit an 830 bp region as being the center of greatest homology. Sequence data from two sections of this region gave a nucleotide homology of 73.7% to the nodC gene of R. meliloti. Using Tn5 mutagenesis a clone was isolated carrying Tn5 in the highly homologous region. When tested on Macroptilium atropurpureum, this MPIK3030 derivative was shown to have a Nod phenotype. When the wild-type allele was reintroduced into the Tn5 mutant, nodulation was restored. Interspecies complementation also showed that both R. meliloti and Rhizobium sp. 本文通过测定静止代谢率(RMR)、非颤抖性产热(NST)、线粒体呼吸酶、脂肪代谢酶活力、褐色脂肪 组织(BAT)线粒体GTP 结合能力、下丘脑促甲状腺激素释放激素(TRH)和促肾上腺激素释放激素(CRH)等指标,探讨短光照对达乌尔黄鼠产热的诱导和调节。结果表明,温暖(22℃ ) 短光照(8D∶ 6L)组,达乌尔黄鼠的RMR、NST、肝脏和BAT 线粒体细胞色素C 氧化酶活力以及BAT 线粒体GTP 结合能力均明显高于温暖长光照(16D∶ 8L)组中的动物,而体重、BAT 重量、肝细胞呼吸、BAT α - 磷酸甘油氧化酶活力则没有明显变化。短光照组黄鼠下丘脑TRH 水平显著高于长光照组,而血清中三碘甲腺原氨酸(T3 )及甲状腺素(T4 )浓度、T3 / T4 以及BAT 中T4 - 5’脱碘酶的活性没有明显变化。短光照组黄鼠下丘脑CRH 水平显著高于对照组,而肾上腺皮质酮含量无明显变化。结果表明短光照能够诱导达乌尔黄鼠产热增加,主要是通过激活细胞色素C 氧化酶活性和增加BAT 中解偶联蛋白浓度;短光照可能激活下丘脑TRH 和CRH,但它们没有直接诱导产热增加,推测其增加了黄鼠潜在的产热能力。  相似文献

大肠杆菌(Escherichia coli)是Ⅱ型脂肪酸合成系统的模式生物,3-羟基脂酰ACP脱水异构酶(FabA)是不饱和脂肪酸合成中的关键酶.生物信息学分析表明,乳酸乳球菌(Lactococcus lactis)的基因组中没有标注为3-羟基脂酰ACP脱水异构酶的基因,但有两个标注为3-羟基脂酰ACP脱水酶基因LlfabZ1和LlfabZ2,其编码的蛋白质与EcFabZ的相似性分别为41%和45.1%,且都具有3-羟基脂酰ACP脱水酶两个保守的α螺旋结构.用携带LlfabZ1和LlfabZ2的质粒载体遗传互补大肠杆菌fabA温度敏感突变株CY57,在42℃下不能恢复生长,但无细胞抽提物的结果显示LlFabZ1能够使反-2-癸烯酰ACP异构成顺-3-癸烯酰ACP,而LlFabZ2则不能.互补大肠杆菌fabZ突变株HW7显示,在诱导的条件下,含有LlfabZ2的转化子能够恢复生长,而LlfabZ1则不能.体外重建脂肪酸合成反应及蛋白质活性测定表明,LlFabZ1具有3-羟基脂酰ACP脱水异构酶功能,而LlFabZ2只具有3-羟基脂酰ACP脱水酶功能.另外,未得到LlfabZ1和LlfabZ2的突变株,表明LlFabZ1和LlFabZ2可能是乳酸乳球菌脂肪酸合成酶系中的必不可少的关键蛋白.上述结果证实了乳酸乳球菌fabZ1和fabZ2两个基因在脂肪酸合成中的功能.  相似文献

达乌尔黄鼠实验室饲养、繁殖及其冬眠阵   总被引:1,自引:0,他引:1  
为探索实验室条件下达乌尔黄鼠饲养与繁殖的方法及冬眠阵的发生规律,参照野生黄鼠冬眠洞穴的主要生态环境参数,建立人工冬眠屋,采用传统锯末技术记录冬眠阵。结果显示: (1) 处于春季繁殖期的黄鼠应以大鼠饲料为主,辅以少量黄瓜等,夏季活跃期交叉饲喂大鼠饲料与兔饲料,辅以多水的瓜果蔬菜,秋季育肥期以大鼠饲料为主,辅以高脂肪高蛋白的花生、豆类等。(2)雌鼠怀孕期为28 d 左右,哺乳期约一个月,雌鼠每窝产仔4 ~ 8 只,平均5.52 只;初生幼鼠两周内忌换垫料,并避免将异味带入鼠房。(3)黄鼠冬眠期从当年11月下旬至次年3 月上旬,平均93.95 d;冬眠阵睡眠时长平均7. 44 d,阵间激醒时长平均1.36 d,睡眠天数占整个冬眠期的89.9% ;整个冬眠期,黄鼠冬眠阵平均7. 55 个。(4)2009 年秋至2011 年春季,自野外共捕回黄鼠185 只, 存活146 只,存活率78. 9% 。在2006、2009 和2011 年的黄鼠繁殖期,共配对25 对,产仔138 只,成活92 只,成活率为66.7% 。结果表明,野生达乌尔黄鼠可在人工饲养条件下实现繁殖,并可在人工冬眠屋成功冬眠。  相似文献

The diversity of alleles at the gliadin loci Gli-U1 and Gli-M b 1 was studied in the tetraploid species Aegilops biuncialis (UUMbMb). The collection of 41 Ae. biuncialis accessions and F2 seeds obtained from five crosses served as the material used in this study. Gliadins were separated by acid polyacrylamide gel electrophoresis. To determine genomic affiliation (U or Mb) of components of Ae. biuncialis gliadin pattern, accessions of Ae. umbellulata and Ae. comosa were analyzed. In Ae. biuncialis accessions, 14 alleles were identified at the locus Gli-U1 and 12 alleles, at the locus Gli-M b 1. The results testify to a high degree of allele diversity at major gliadin-coding loci of homeologous group 1 chromosomes of Ae. biuncialis.  相似文献

The UCP concentration in mitochondria was indirectly determined by titration with its specific ligand [H3]-labeled GTP. Type II T4 5'-deiodinase of BAT was assayed concomitantly. We found two species of mRNA for UCP with lengths of about 1.9 and 1.5 kb, respectively, both occurring in almost the same concentration. UCP mRNA content was elevated significantly during hibernation, but the UCP concentration did not change compared with that of nonhibernating controls kept at room temperature. When hibernating squirrels were aroused, the UCP mRNA remained at the elevated level as during hibernation, but the UCP concentration increased in comparison with that of nonhibernating controls or during hibernating. Changes in T4 5'-deiodinase activity in BAT were similar to the variations of the UCP mRNA level. These results suggest that the activation of T4 5'-deiodinase in BAT may be an important factor for the up-regulation and maintenance of UCP mRNA content needed for the synthesis of sufficient UCP to acquire the thermogenic capacity for arousal from hibernation.  相似文献

The extent of feeding, fecundity and fertility of a new sugar beet pest, Aubeonymus mariaefranciscae Roudier (Coleoptera: Curculionidae), on a range of plants from different families was determined. We have found that the adult weevils were only able to feed and oviposit on two closely related cultivated plants, beet and sugar beet, both subspecies of Beta vulgaris L., suggesting a restricted host range for this curculionid. None of 16 secondary metabolites (widely distributed among plants, and representative of different chemical groups), nor any of 19 non-host extracts from 13 different families, deterred the adults of this curculionid from feeding on sugar beet. We found a phagostimulatory effect when beet and sugar beet homogenates from various plant tissues were supplied to adult weevils on weeds that they do not feed on. Conversely, no effect was observed when sucrose was provided. These findings suggest that host selection by A. mariaefranciscae adults is due to the presence of an undetermined phagostimulant compound(s) in beet and sugar beet, instead of the recognition of deterrent substances among the non-host plants.  相似文献

Summary A 6.7 kb HindIII fragment from the Sym-plasmid of strain NGR234 was found to code a nodD-like gene flanked by two loci which were required for siratro host range. Transfer of the 6.7 kb fragment from NGR234 to R. trifolii strain ANU843 conferred extended host range ability to this strain on siratro plants but not to other plants normally nodulated by strain NGR234. Tn5 mutagenesis of the 6.7 kb fragment showed that insertions located into loci flanking the nodD-like gene abolished the extended host range phenotype. A hybridization probe spanning one of the host specificity loci was shown to hybridize to three specific bands in the NGR234 genome. Complementation and DNA hybridization data showed that the nodD-like gene of strain NGR234 was functionally similar to that in R. trifolii. The introduction to R. trifolii of the 6.7 kb HindIII fragment containing Tn5 insertions located in the nodD-like gene did not abolish the ability to extend the host range of R. trifolii to siratro plants. However, transfer of the 6.7 kb HindIII to R. trifolii derivatives containing Tn5 insertions into either nodA, B or C or other R. trifolii nod genes failed to confer siratro nodulation to these recipients. Reconstruction experiments showed that the 6.7 kb fragment from strain NGR234 and the 14 kb nodulation region of R. trifolii could induce the nodulation of siratro plants when introduced together into Sym-plasmid-cured Rhizobium strains.  相似文献

该研究运用生物信息学方法鉴定甘蓝型油菜RPD3/HDA1基因家族,检测了其在‘黔油早2号’和‘中双11’中的表达水平及2个品种在低温(4℃)和ABA胁迫下该家族基因的表达特征,以探讨RPD3/HDA1基因在甘蓝型油菜中的潜在功能,为早熟油菜抗逆性遗传改良提供理论基础和候选基因。结果表明:(1)在甘蓝型油菜全基因组中共鉴定到28个RPD3/HDA1基因,将其命名为BnHDA1~BnHDA28,聚类为4个亚家族,同一亚家族成员的基因结构较为相似;在该基因家族中共检测到16对复制基因,均为片段重复。(2)顺式作用元件预测统计中共发现675个元件与植物激素、环境胁迫和光响应有关。(3)qRT-PCR分析显示,RPD3/HDA1基因在‘黔油早2号’中的表达量均高于‘中双11’;低温胁迫下,‘黔油早2号’和‘中双11’中RPD3/HDA1基因呈差异表达,与‘中双11’相比,RPD3/HDA1基因在‘黔油早2号’中的下调幅度较大;ABA处理后,RPD3/HDA1基因在2个品种中表达模式不一致,‘黔油早2号’中大部分RPD3/HDA1基因表达量较‘中双11’下调幅度小。研究认为,RPD3/HDA1基因可能在油菜开花中发挥调节作用,而且可能通过激素信号通路和防御信号通路参与油菜的生长发育和防御反应的调节。  相似文献

Gsp1p is a small nuclear-located GTP binding protein from the yeast Saccharomyces cerevisiae. It is highly conserved among eucaryotic cells and is involved in numerous cellular processes, including nucleocytoplasmic trafficking of macromolecules. To learn more about the GSP1 structure/function, we have characterized its Candida albicans homologue. CaGsp1p is 214 amino acids long and displays 91% identity to the ScGsp1p. There is functional complementation in S. cerevisiae, and its mRNA is constitutively expressed in the diploid C. albicans grown under various physiological conditions. Disruption of both alleles was not possible, suggesting that it could be an essential gene, but heterozygous mutants exhibited genomic instability.  相似文献
