The development of sustainable integrated aquaculture systems requires combining fed aquaculture (finfish) with extractive inorganic aquaculture (seaweed) and extractive organic aquaculture (shellfish). With the support of AquaNet, the Network of Centers of Excellence in Aquaculture in Canada, we are developing such a system at an industrial pilot scale by co‐cultivating salmon (Salmo salar), kelp (Laminaria saccharina) and blue mussel (Mytilus edulis) at aquaculture sites in the Bay of Fundy, Canada. This presentation will focus on the development of the extractive inorganic component. The entire cycle of rearing Laminaria saccharina has been completed and improved, both in the laboratory and at the integrated sites: release in the laboratory of spores from mature macroscopic sporophytes, seeding of ropes, germination of microscopic gametophytes, sexual maturation of male and female gametophytes, development of zygotes into juvenile sporophytes, which are then transplanted to the sites for rapid grow‐out. Another aspect of the project, food safety monitoring of chemical therapeutants and phycotoxins in mussel and kelp cultured in proximity to salmon, will also be described. The productivity, nutrient absorption capacity, and role of the seaweed component are being analyzed so that its appropriate scale to the other components can be defined in order to develop responsible aquaculture practices in which metabolic/physiological processes of the different co‐cultured organisms counter‐balance each other within acceptable operational limits. Adopting polytrophic strategies will be key to the aquaculture industry to develop its environmentally and economically‐balanced diversification and increase its social acceptability within a broader coastal management framework.  相似文献

The kelp Ecklonia radiata has become a target for controlled cultivation. However, to date there are no standardised protocols for the hatchery stage of this species that result in high rates of germination, gametophyte development and transition to sporophytes. Therefore, the objective of this study was to quantify the effect of photoperiod, light intensity, temperature, nutrient media and use of GeO2 on the key hatchery processes of germination, gametophyte development and transition to sporophytes in controlled laboratory experiments. Germination of E. radiata was high (≥?85%) throughout the study, regardless of treatments. Temperature had a major effect on the length of gametophytes, which increased with increasing temperature. The formation of sporophytes was favoured when individuals were maintained under 17 °C continuously, while reduced by approximately 30% when using F/2 compared to PES nutrient media. Overall, the recommended conditions for the hatchery stage of E. radiata are to maintain cultures under a 12 h L:12 h D photoperiod at 17 °C as this resulted in higher germination rates, good gametophyte development and higher transition to sporophytes compared to other treatments. Moreover, the use of GeO2 has to be limited to no more than 2 days as extended use has detrimental effects on the development of sporophytes. Finally, storage of sorus-bearing fronds of sporophytes up to 4 days after the collection from the field generally increased the number of released zoospores and is a simple mechanism to increase the fertility of brood stock.

The kelp Laminaria saccharina dominates soft bottoms in 4–10 m depth in Kiel Bay. Experimental sporophytes transplanted to 2 and 5 m depth showed the typical annual growth pattern of Laminaria species. Surprisingly, 2 m plants died after the first resting phase, whereas 5 m plants survived and showed outgrowth of a new blade generation. Thalli at both depths were infected with the brown algal endophyte Streblonema aecidioides, with host deformations being significantly stronger in 2 m plants. Growth rates of infected sporophytes were reduced. Exclusion of UV light in 2 m depth resulted in less infected thalli. Discs excised from L. saccharina and cultivated in different photon fluence rates from 10–600 µmol m–2 s–1 did not differ in growth rate, photosynthesis or dark respiration. Hence, an exclusion of L. saccharina from shallow depths caused by high light cannot be concluded. We suggest the biological interaction with the endophyte S. aecidioides, amplified by UV light, to be most important for the exclusion of L. saccharina from shallow depths in the western Baltic.  相似文献

Various stages of the life cycle of the marine brown alga Laminaria saccharina (L.) Lamour. (Laminariales, Phaeophyta) including male and female gametophytes, female gametes, zygotes and young sporophytes of different age were investigated for their potentials of carbon dioxide (14CO2) fixation. Rates of photosynthesis attain the same order of magnitude in all stages. Photosynthetic 14CO2-fixation is accompanied by a substantial light independent carbon assimilation. The seaweed Laminaria japonica (Phaeophyceae) has a two-generation life cycle consisting of haploid gametophytes and diploid sporophytes. Female and/or male gametophytes were transformed using particle bombardment and the histological LacZ assay was performed on sporophytes generated by either parthenogenesis or inbreeding. Female gametophyte-targeted transformation resulted in similar lower efficiencies in both parthenogenetic and zygotic sporophytes, and only a chimeric expression pattern was observed. Male gametophyte-targeted transformation led to a higher efficiency, with 3.5% of the zygotic sporophytes stained completely blue (all-blue), implying the integration of lacZ at the one-cell stage. Polymerase chain reaction analysis using primers specific for a lacZ-vector juncture fragment and subsequent blotting indicated the presence of the introduced gene in the sporophytes. Lessonia trabeculata is one of the major kelps found along the northern coast of Chile. In addition to its ecological and economic importance, L. trabeculata may be severely affected by environmental disturbances such as El Níño, which during 1982–1983 cleared wide areas along the coast of Peru and Chile. The main goal of this work was to mass culture L. trabeculata and to observe the growth of sporophytes obtained in the laboratory and cultured in the sea. Juvenile sporophytes obtained in the laboratory were attached between 1 and 6 m in depth. The linear growth rate, as blade elongation, was recorded weekly for seven months. No significant differences (p < 0.05) were found in sporophyte blade linear growth at different depths. The best elongation growth rate was 7.5 ± 1.6 mm d–1 at 3 m during March. This preliminary work suggests that L. trabeculata follows an annual growth cycle similar to that of other Laminariales with a high rate of blade elongation during the summer and decreasing towards autumn. This species can be considered a potential candidate for aquaculture to increase the availability of raw material and aid in repopulation of overexploited areas.  相似文献

The present is the first study on epi-endophytic algae on thalli of Undaria pinnatifida growing along Argentinian coasts. The main goal is to describe the nature and the morphology of this symbiosis. Individuals of Laminariocolax aecidioides were detected in both June and December 2004, growing on U. pinnatifida sporophytes. In nature, the epi-endophyte were macroscopically observed as dark zones that partially covered the hosts’ fronds. L. aecidioides vegetative thalli were irregularly branched uniseriate filaments. The life cycle is described from laboratory cultures started from Patagonian populations. Caryology revealed that the sporophytic diploid phase presented 16 chromosomes whereas the gametophytic haploid phase presented 8 chromosomes. Isolates made from thalli growing in the interior of infected hosts developed into filamentous, branched sporophytes that reproduced by both unispores and plurispores that were produced in unilocular and plurilocular sporangia, respectively. The results of this paper also allowed us to conclude that L. aecidioides uses the thalli of U. pinnatifida as a proper substrate. The penetration of endophitic filaments among the host′s cortical cells produced a lateral compression and, finally, their thalli development generated perforations in the host tissues. The effects of the epi-endophytic infection of L. aecidioides on U. pinnatifida are neither severe nor deleterious.  相似文献

The brown alga Laminaria saccharina is the dominant subtidal macroalga in Kiel Bay, western Baltic. It is infected by the microscopic brown alga, Streblonema aecidioides. Infected thalli may show symptoms of Streblonema disease, i.e. alterations of blade and stipe, ranging from dark spots to heavy deformations and completely crippled thalli. Samples taken from a single locality all year round show that (i) the host population is infected at a high rate of 87±13% (SD), but that (ii) a considerable proportion of thalli containing Streblonema does not show disease symptoms, and that (iii) juvenile hosts, which mainly appear in autumn, are infected at almost the same rate. Thus the infection seems to occur early in the host's life. Juveniles in nature show fewer symptoms of the disease than adults. Two months after infection, oxygen production and growth in laboratory-raised experimentally infected juvenile hosts was not different from uninfected controls. Experimental thalli showed more severe morphological alterations than uninfected controls only four months after infection. Both field and laboratory observations indicate that a lag phase exists between infection and outbreak of the disease.  相似文献

This paper describes a method to study the diversity of young kelp sporophytes that are recruited from the bank of microscopic stages. Small samples of rocky substratum (0.5 cm2) were collected from the low intertidal zone, which was dominated by the kelp Laminaria digitata. Samples were cultivated in the laboratory under conditions permitting gametogenesis. Sporophyte recruits in the cultures were isolated and identified at the species level using the barcoding mitochondrial marker rpl31–rns. Sixty per cent of the collected samples had at least one to a maximum of 30 kelp recruits, belonging to five different species (L. digitata, L. hyperborea, L. ochroleuca, Saccharina latissima and Sacchorhiza polyschides). As the examination of freshly collected rocky samples under a stereo microscope did not reveal any kelp sporophytes, the recruitment in these samples after culture probably occurred from the bank of microscopic forms present on the substratum. Despite the dominance of L. digitata in the field, the young sporophytes obtained after culturing were mainly S. polyschides. This study illustrates the suitability of culturing in combination with molecular identification of young sporophytes to address several key aspects of kelp ecology related to the existence of a bank of microscopic stages in the field.  相似文献

Growth of young sporophytes of the brown algae Laminaria digitata, L. saccharina and L. solidungula from Spitsbergen were measured in the laboratory after being exposed for 21 days to either photosynthetically active radiation (PAR=P) or to full light spectrum (PAR + UV-A + UV-B=PAB) using of cutoff glass filters. The plants were grown at 8±2°C and 16 h light : 8 h dark cycles with 6 h additional ultraviolet radiation (UVR) exposure in the middle of the light period. Growth was measured every 10 min using growth chambers with online video measuring technique. Tissue morphology and absorption spectra were measured in untreated young sporophytes while chlorophyll (Chl) a content and DNA damage were measured in treated thalli at the end of the experiment. In all species, growth rates were significantly higher in sporophytes exposed to P alone compared to sporophytes exposed to PAB. Tissue DNA damage is dependent on thallus thickness and absorption spectra characteristics of pigments and UV-absorbing compounds. In sporophytes exposed to UVR, energy demands for repair of DNA damage and synthesis of UV-absorbing compounds for protection effectively diverts photosynthate at the expense of growth. Photosynthetic pigment was not significantly different between treatments suggesting a capacity for acclimation to moderate UVR fluence. The general growth pattern in sporophytes exposed to P alone showed an increasing growth rate from the onset of light (0500–0900 hours) to a peak at the middle of the light phase (0900–1500 hours), a decline towards the end of the light phase (1500–2100 hours) and a minimum “low” growth in the dark (2100–0500 hours) relative to growth during the entire light phase. Under PAB, different growth patterns were observed such as growth compensation at night in L. digitata, delayed growth recovery in L. saccharina and minimal but continuous growth in L. solidungula. Growth as an integrative parameter of all physiological processes showed that the effect of UVR is correlated to the depth distribution of these species.  相似文献

The capacity to cope with high light stress was investigated in different life-history and developmental stages of Laminaria saccharina Lamour. sporophytes and gametophytes. Changes in photosynthetic efficiency and in the level of photoinhibition were measured by in vivo fluorescence changes of photosystem II. Pigment content was studied using high performance liquid chromatography. Additionally, the morphology of the various developmental stages during the life cycle was studied by light microscopy in relation to the photosynthetic parameters. High light stress (2 h, 500 μmol.m-2.s?1) induced photoinhibition of photosynthesis with fast kinetics in older sporophytes and gametophytes. In contrast, the absolute degree of photoinhibition after light stress was higher in younger than in older sporophytes. Photosynthesis recovered faster in older sporophytes and gametophytes compared to young sporophytes. In very young sporophytes, photosynthesis did not recover fully even after 12 h exposure to low light, indicating severe photodamage. Kinetics of recovery in old sporophytes and in gametophytes showed a fast and a slow phase, whereas younger sporophytes recovered only with a slow phase, The fast phase is indicative of a decline of the photoprotective process, whereas the slow phase indicates a recovery from photodamage. The capacity to cope with high light stress in Laminaria sporophytes increased with increasing age of the thalli. The gametophytes are less sensitive to high light stress and may be selected to endure unfavorable white light conditions. Investigation of the xanthophylls showed that the higher resistance to high light is not caused solely by a higher content of xanthophyll cycle pigments. Additionally, changes in the thallus structure during the development of the sporophytes seemed to cause a higher resistance to high light. The observed changes in the ability to cope with high light in the different life-history and developmental stages of Laminaria saccharina may influence the distribution of the species on the shore.  相似文献

The life cycle and mating behavior of Helicotylenchus multicinctus (Nematoda: Hoplolaimidae) were observed in vitro on excised roots of Musa cavendishii in gnotobiotic culture. Eggs hatched into juveniles whose appearance and structure were similar to those of the adults. Juveniles grew in size and each juvenile stage was terminated by a molt. H. multicinctus had four juvenile stages. The first molt occurred outside the egg shortly after hatching. After the final molt the juveniles differentiated into adult males and females. Mating was required for reproduction. After mating, fertilized females began to lay eggs. The life cycle from second stage juvenile to second stage juvenile was completed in 39 days.  相似文献

This paper describes the formation of gametophytes and the early stages of the development of sporophytes in the kelp seaweed Alaria angusta from Kamchatka. To establish laboratory cultures we used zoospores obtained from A. angusta sporophytes collected on October 29, 2014. The gametophytes were grown under different conditions: at 6–7°С and natural light and at 10°С and illumination with cool white fluorescent bulbs, 30 μmol photon m–2 s–1, 12: 12 h L: D cycle. In the first case (natural light, 6–7°С), the vegetative growth of male and female gametophytes lasted for more than 4 months; maturation of sexual products occurred 144 days after germination of the embryospores. In the second case (artificial light, 10°С), rapid development of the gametophytes occurred; the first juvenile sporophytes appeared on the 10th day after the zoospores settled onto the substrate. Our data contribute to understanding of the regulatory effect of temperature and light on the peculiarities of species vegetation in the natural environment and formation of the age structure of the species population, one particular feature of this process is the continuous appearance of juvenile sporophytes in the warm period of the year.  相似文献

Lessonia variegata J. Agardh (Laminariales, Phaeophyta) is endemic to New Zealand, where it occurs in subtidal kelp forests on wave exposed coasts in the North, South and Stewart Islands. This is the first account of the growth in culture and life history of L. variegata. Microscopic gametophytes alternate with macroscopic sporophytes, characteristic of members of the order Laminariales. The life history was completed in culture within 14 days under growth conditions of 12 °C, 12:12 (L:D) and 15 °C, 15:9 (L:D). Maximum growth of sporophytes occurred at 15 °C, 15:9, and slowest growth at 10 °C, 9:15. Under low light conditions (8–9 μmol photon m−2 s−1) filamentous growth of gametophytes predominated, and both the proportion of gametogenesis and the growth of sporophytes after 30 days was much reduced from equivalent cultures grown under conditions of higher light (16–17.5 μmol photon m−2 s−1). Interest in this species relates both to its potential for commercial utilisation as well as in the role it plays in coastal rocky reef ecosystems.  相似文献

In previous studies, Laminaria saccharina L. (Lamour.) sporophytes were found to exhibit two major peaks of sporogenesis and an annual life cycle in Long Island Sound, New York. Young sporophytes were observed shortly after the sporogenesis peaks in early autumn and spring, but most of the mature sporophytes decayed during summer. A new study was conducted to determine if the spring sporogenesis activity contributed to the recruitment observed in autumn through oversummering of gametophytic and juvenile sporophytic stages, as previously suggested. Reproduction and growth in gametophytes and growth in juvenile sporophytes were studied under crossed gradients of light and temperature. Periodic outplantings of substrata seeded with gametophytic and sporophytic stages to the field were conducted to assess actual survival. The optimum temperature and light conditions for gametophyte development, growth and reproduction varied with the time of year meiospores were obtained. Most of this variation was attributable to temperature. A seasonal adaptation to temperature in most developmental stages was observed. Higher temperatures resulted in greater numbers of male gametophytes. Gametophytic stages could develop at all times, suggesting that oversummering in this stage was possible. Juvenile sporophytes had a narrower optimum temperature range and again photon fluence rate contributed little to observed variances. Out planting of sporophytic stages at various times during the year indicated only sporophytes prepared from autumn and winter could survive summer conditions. The thalli of these plants grew rapidly in spring and eroded back to the meristematic region in summer. Most of these plants then quickly became reproductive, resulting in another autumn sporogenesis peak. Gametophytic and sporophytic outplantings prepared from spring meiospores did not survive the summer. Thus, the recruitment observed in autumn can only be the result of the previous autumn's sporogenesis activity. The sporogenous activities of spring and early summer appear to be unimportant, despite the fact that all reproductive indices are superior at those times.  相似文献

Herbivory can be an important factor structuring coastal algal communities. Herbivores may preferentially graze particular algal species or tissue types. Mesograzers, despite their small size, can critically weaken kelp thalli and impact entire kelp beds. We propose that when kelp beds are composed of several kelp cohorts, mesograzers will selectively choose to inhabit younger plants and grazing activities will have a greater impact on younger plants. This study investigated the effects of grazing by the littorinid gastropod, Lacuna vincta, on different age classes of the bull kelp, Nereocystis luetkeana by (1) testing food preference of L. vincta on juvenile, first-year adult, and second-year adult Nereocystis blades in the laboratory, (2) determining substrate (blades of different ages) preference of L. vincta in the laboratory, and by (3) estimating in-situ herbivore abundances and densities on juvenile and adult Nereocystis. Results demonstrated that grazing by L. vincta produced greater damage on juvenile than older Nereocystis tissues. Although L. vincta did not select juvenile versus older kelps as substrate in the laboratory, in situ surveys showed that differences existed between age classes with higher L. vincta densities on juvenile than adult kelp. We conclude that at a local scale, L. vincta can be an important structuring factor in Nereocystis populations due to its high density and grazing ability. Handling editor: K. Martens  相似文献

It has recently been reported that the Eisenia bicyclis beds on the coast of Tomarihama in the Oshika Peninsula in Miyagi Prefecture (northern part of Japan) have been progressively receding from the offshore side of the beds every year. In this study, to determine the factors causing the change in the distribution of E. bicyclis beds as well as the extent of the change, an ecological survey of E. bicyclis and grazing animals was conducted. A 150 m-long fixed line was set up in the rocky coastal area on the Oshika Peninsula in Miyagi Prefecture, and the number of E. bicyclis individuals inhabiting a 1-m width was then measured. In addition, the numbers of individuals of sea urchins (Strongylocentrotus nudus) and abalone (Haliotis discus hannai) were measured. Although the E. bicyclis bed extended to the 64-m point on the fixed line at the time of the survey in August 1999, by July 2001, its lower limit was at the 53-m point; thus, a retreat of the outer edge of the bed of 11 m was observed in 2 years. The density of adult E. bicyclis sporophytes in the bed was reduced from 1.08 to 1.80 individuals (ind.) m−2, respectively. Juvenile sporophytes were also observed, but most of those that had settled near the lower limit of the bed failed to survive to become adult sporophytes. There were few sea urchins, the main grazing animal on the site, inside the E. bicyclis bed (3.5 ind. m−2 in October 2001) while there were many near the bed lower limit and on the deep-site side (12.9 ind. m−2 in October 2001). These results indicate that the high grazing pressure of sea urchins reduced the survival of juvenile sporophytes near the bed lower limit, thus impeding the recruitment of juvenile sporophytes to replace adult sporophytes that died due to senescence, storm action and/or grazing.  相似文献

Sporophytes of some epiphytic species in the fern genus Pyrrosia exhibit Crassulacean acid metabolism (CAM), generally considered to be a derived physiological response to xeric habitats. Because these species alternate between independent sporophytic and gametophytic generations yet only the sporophyte has been characterized physiologically, experiments were conducted to determine the photosynthetic pathways present in mature sporophytes, immature sporophytes, and gametophytes of Pyrrosia longifolia. Diurnal CO2 exchange and malic acid fluctuations demonstrated that although the mature sporophytes exhibited CAM, only C3 photosynthesis occurred in the gametophytes and young sporophytes. Consideration of the above results and those from previous studies, as well as the life cycle of ferns, indicates that the induction of CAM probably occurs at a certain developmental stage of the sporophyte and/or following exposure to stress. Elucidation of the precise mechanisms underlying this C3-CAM transition awaits further research.  相似文献

A high-yield technique is described for the elaboration of single cell detritus (SCD) from Laminaria saccharina, based on the sequential action of C1H, enzymes (endoglucanases and cellulases) and 2 bacteria showing a high degree of cellobiotic, proteolytic, and alginolytic activity (CECT 5255 and CECT 5256). Over 85% of dried particles of L. saccharina were transformed into a suspension of free cell and bacterial and detrital particles after 24 hours of bacterial activity with this technique. These particles were less than 20 m in diameter, constituting a suitable diet for bivalve mollusks. After 72 hours 99% of the total particulate volume consisted of particles less than 20 m in diameter. Tests of hatchery diets for the seed of clam Ruditapes decussatus revealed increases of 54% and 68% for live weight and length, respectively, when SCD from L. saccharina was used as the sole dietary component compared with a live phytoplankton diet. However, SCD from L. saccharina is not a suitable food for the larvae of R. decussatus.  相似文献

Laminaria abyssalis occurs in deep water in tropical latitudes of the Brazilian coast (19° 23 S, 38° 28 W to 22° 54 S, 42° 13 09 W). Its life cycle has been completed in the laboratory in seven months using different conditions of light and temperature. The gametophytic stage required for growth the low photon flux density of 1.2 ± 0.3 µmol m–2 s–1 and 18 °C, while the juvenile and adult sporophytes needed 15 µmol m–2 s–1 and 18 °C. The sporophytes became fertile at 23 °C. Our results showed that light and temperature are the main factors regulating the growth and life history of this species under the culture conditions tested.  相似文献
