Background

Buruli ulcer (BU) is a skin disease caused by Mycobacterium ulcerans, with endemicity predominantly in sub-Saharan Africa and south-eastern Australia. The mode of transmission and the environmental reservoir(s) of the bacterium and remain elusive. Real-time PCR investigations have detected M. ulcerans DNA in a variety of Australian environmental samples, including the faeces of native possums with and without clinical evidence of infection. This report seeks to expand on previously published findings by the authors'' investigative group with regards to clinical and subclinical disease in selected wild possum species in BU-endemic areas of Victoria, Australia.

Methodology/Principal Findings

Twenty-seven clinical cases of M. ulcerans infection in free-ranging possums from southeastern Australia were identified retrospectively and prospectively between 1998–2011. Common ringtail possums (Pseudocheirus peregrinus), a common brushtail possum (Trichosurus vulpecula) and a mountain brushtail possum (Trichosurus cunninghami) were included in the clinically affected cohort. Most clinically apparent cases were adults with solitary or multiple ulcerative cutaneous lesions, generally confined to the face, limbs and/or tail. The disease was minor and self-limiting in the case of both Trichosurus spp. possums. In contrast, many of the common ringtail possums had cutaneous disease involving disparate anatomical sites, and in four cases there was evidence of systemic disease at post mortem examination. Where tested using real-time PCR targeted at IS2404, animals typically had significant levels of M. ulcerans DNA throughout the gut and/or faeces. A further 12 possums without cutaneous lesions were found to have PCR-positive gut contents and/or faeces (subclinical cases), and in one of these the organism was cultured from liver tissue. Comparisons were made between clinically and subclinically affected possums, and 61 PCR-negative, non-affected individuals, with regards to disease category and the categorical variables of species (common ringtail possums v others) and sex. Animals with clinical lesions were significantly more likely to be male common ringtail possums.

Conclusions/Significance

There is significant disease burden in common ringtail possums (especially males) in some areas of Victoria endemic for M. ulcerans disease. The natural history of the disease generally remains unknown, however it appears that some mildly affected common brushtail and mountain brushtail possums can spontaneously overcome the infection, whereas some severely affected animals, especially common ringtail possums, may become systemically, and potentially fatally affected. Subclinical gut carriage of M. ulcerans DNA in possums is quite common and in some common brushtail and mountain brushtail possums this is transient. Further work is required to determine whether M. ulcerans infection poses a potential threat to possum populations, and whether these animals are acting as environmental reservoirs in certain geographical areas.  相似文献

Demographic and genetic replenishment of populations through the exchange of individuals is essential for their persistence. Habitat loss and fragmentation can reduce the permeability of landscapes, hinder dispersal and compromise the genetic integrity of populations over time. We examined ecological connectivity in an arboreal marsupial, the common ringtail possum (Pseudocheirus peregrinus) in fragmented forests of southeastern Australia. This species is potentially robust to fragmentation based on its presence in degraded landscapes and known use of plantations for foraging and nesting. Using 312 individuals screened at 15 microsatellites, we measured dispersal and gene flow across seven native Eucalyptus forest remnants surrounded by exotic Pinus radiata plantations and three sites within a large continuous forest. The permeability of the pine matrix to dispersing possums was significantly lower than that of the native forest. Small, isolated patches exhibited signatures of genetic drift, having lower heterozygosity and allelic richness than possums in large patches. Most (87%) possums were born in their sampled patch or dispersed only short distances into neighbouring remnants. The continuous forest was identified as an important source of immigrants only for proximate patches (within 2.5 km), thus emphasizing for the common ringtail possum and more specialized arboreal mammals the need to conserve large, proximate forest remnants. Our findings highlight the importance of using genetic tools to understand the long-term biological consequences of fragmentation for effective management.  相似文献

We characterized 15 polymorphic tetranucleotide microsatellite markers for the ringtail, Bassariscus astutus. We tested these loci in 21 individuals captured in Arizona and Texas and found six to 19 alleles per locus. Observed and expected heterozygosities ranged from 0.381 to 1.000 and from 0.381 to 0.941, respectively. All loci were in Hardy-Weinberg equilibrium, and none were in linkage disequilibrium. These markers may be used to investigate population genetics and mating patterns in this species.  相似文献

This study examines the annual energetics of a small folivorous marsupial, Pseudocheirus peregrinus. Particular attention was given to the energy and time allocated to reproduction by the females. Daily energy expenditure was measured directly using the doubly labelled water technique. Energy transferred to the young via the milk was estimated from information on milk composition and production. There was no significant seasonal variation in the energy expenditure or water influx of males or females. The mean daily energy expenditure of a 1-kg non-lactating adult ringtail possum was 615 kJ day–1 or 2.2 times standard metabolic rate. Females showed significant changes in daily energy expenditure according to their reproductive status. Without the burden of lactation the total annual energy expenditure of an adult female was estimated as 212.4 MJ kg–1 year–1. The total annual energy expenditure of a female rearing two young was 247.5 MJ kg–1 year–1, with the late stage of lactation constituting the most energetically expensive period accounting for 30% of the total yearly energy expenditure during 24% of the time. Total metabolisable energy allocation during reproduction (22 MJ kg) was similar to estimates available for other herbivores, although, the peak metabolisable energy allocation during lactation (759 kJ day–1) was lower than values available for other herbivores. The total energy requirement for reproduction (metabolisable energy plus potential energy exported to young via milk) suggests that the ringtail possum also has a relatively low overall energy investment in reproduction. It is suggested that the lactational strategy of the ringtail possum has been selected in order to spread the energy demands of reproduction over time due to constraints on the rate of energy intake imposed by a leaf diet and/or to prolong the mother-young bond. The strategies a female ringtail possum may employ to achieve energy balance when faced with the energy demands of reproduction are discussed.  相似文献

1. Glucuronic acid was excreted in the urine of the brushtail possum, Trichosurus vulpecula, in response to dietary terpenes (essential oils found in Eucalyptus foliage). 2. The relationship between urinary glucuronic acid and the estimated terpene content of the diet was not equimolar, but varied from 1-17%, with levels of 5-39% found in animals maintained on terpenes or eucalypt leaves for several weeks. 3. Blood levels of ascorbate and the turnover rate of ascorbate were increased in the brushtail possum in response to dietary terpenes. 4. The presence of an active glucuronic acid pathway and associated glucose-ascorbate-glucose cycle was postulated for the brushtail possum and the other arboreal marsupials, Pseudocheirus peregrinus (common ringtail possum) and Petauroides volans (greater glider). 5. By means of these pathways these animals may utilise the high ascorbate content of Eucalyptus leaves to conserve glucose required for synthesis of glucuronic acid used for conjugation of dietary terpenes.  相似文献

Although critically endangered western ringtail possums (Pseudocheirus occidentalis) have been recorded from a variety of vegetation types in southwestern Australia, the extent to which many of these vegetation types are occupied by the species remains unknown. We conducted spotlight surveys for western ringtail possums between March and April 2018 in Albany, southwestern Australia, to determine the species’ occupancy in 2 vegetation types. Using occupancy models, we demonstrated that sheoak (Allocasuarina fraseriana) woodlands, previously unrecognized as providing western ringtail possum habitat, support the species, although the median probability of their occupancy was lower than in marri (Corymbia calophylla) and eucalypt (Eucalyptus marginata and E. staeri) woodlands. Use of trees and other habitat components by western ringtail possums varied within and between vegetation types. Sheoak woodlands are likely critical for western ringtail possums and given the apparent flexibility in vegetation types used by the species, investigation of the potential value of other vegetation types for this species has conservation merit. © 2019 The Wildlife Society.  相似文献

Habitat loss and invasive predators increasingly threaten global biodiversity. Here we use a landscape‐scale experimental approach to explore the individual and synergistic effects of logging and an invasive predator, the red fox Vulpes vulpes on two common native arboreal vertebrates (a predator and prey species) in south‐eastern Australia. We used site occupancy methods to evaluate different models evaluating the effects of site specific forest logging disturbance, lethal fox baiting and forest structural elements for explaining variation in site occupancy of a large monitor lizard Varanus varius, and a marsupial prey, the common ringtail possum Pseudocheirus peregrinus across a complex forest landscape. Site occupancy of ringtail possum was influenced by habitat resources and the structural complexity of forest, which indirectly mediated predation risk. Presence of fox baiting had no direct effect on the ringtail site occupancy. In contrast, access to prey resources and fox baiting appeared to best explain site occupancy variation in monitor lizards across the landscape. While these species are affected primarily by separate disturbances, synergistic interactions between the processes may intensify their effects. Our results demonstrate that species susceptibility to disturbance processes are highly idiosyncratic. This approach makes efficient use of integrated modelling to aid conservation management at both local and landscape levels.  相似文献

Abstract There is increasing realization that human perceptions of a ‘patchy’ environment do not necessarily relate to that of a particular organism and ecological characteristics of land adjacent to patches of natural vegetation (‘remnants’) may influence the ecological processes within. We investigated how the distribution and abundance of resources within and surrounding 39 small remnants located within the city of Melbourne, Australia, influenced the abundance of common brushtail possums Trichosurus vulpecula (Phalangeridae) and common ringtail possums Pseudocheirus peregrinus (Pseudocheiridae), two primarily folivorous marsupials. Bayesian modelling techniques were used to relate the abundance of both possum species to estimates of den and food availability both within the remnant and within a 100‐m buffer strip extending out into adjacent residential development. We found that both brushtail and ringtail possum abundance within remnants increased with the density of potential den sites within the remnant and food availability within the surrounding landscape. Ringtail possum density within the remnant further increased with den availability within the surrounding landscape. Our results lend support to the hypothesis that, in highly modified landscapes, processes originating outside a remnant vegetation patch may influence the abundance of species within.  相似文献

A comprehensive and contemporary understanding of habitat and resource requirements has been critical to the conservation of multiple taxa and ecosystems globally. Until recently, much of the ecological knowledge that contributes to conservation priorities and strategies for the Critically Endangered western ringtail possum (Pseudocheirus occidentalis) was largely derived from decades‐old observations in peppermint (Agonis flexuosa) and marri‐jarrah (Corymbia calophylla and Eucalyptus marginanta) woodlands in the northern parts of the species range. These observations do not account for more recent evidence of their flexible use of habitat resources in other regions of its range. This may represent a significant conservation opportunity for the species through the identification of additional habitats that warrant protection. In a region where knowledge of their ecology is scarce, we used scat analysis and quantitative spotlighting to determine the diet and density of western ringtail possums in three vegetation types: peppermint, sheoak (Allocasuarina fraseriana) and marri‐eucalypt (C. calophylla, E. marginanta and Eucalyptus staerii) woodlands. Given the species’ reported dependence on peppermint woodlands and dominant canopy species for food sources, we hypothesised that western ringtail possums would be most abundant in this habitat type and that their diet would comprise the foliage of few (≤2 species) canopy species. We found western ringtail possums consumed a higher diversity of plant species than expected (8–14), exhibited dietary preference for non‐dominant canopy species and were present in all sampled vegetation types at substantially higher densities than previously recorded for the region (as high as 17 possums ha?1). Our results confirm (i) the western ringtail possum is flexible in its use of habitat resources and (ii) the significant conservation value of sheoak and marri‐eucalypt woodlands in the southernmost portion of its distribution.  相似文献

The last 20 years has seen a significant series of outbreaks of Buruli/Bairnsdale Ulcer (BU), caused by Mycobacterium ulcerans, in temperate south-eastern Australia (state of Victoria). Here, the prevailing view of M. ulcerans as an aquatic pathogen has been questioned by recent research identifying native wildlife as potential terrestrial reservoirs of infection; specifically, tree-dwelling common ringtail and brushtail possums. In that previous work, sampling of environmental possum faeces detected a high prevalence of M. ulcerans DNA in established endemic areas for human BU on the Bellarine Peninsula, compared with non-endemic areas. Here, we report research from an emergent BU focus recently identified on the Mornington Peninsula, confirming associations between human BU and the presence of the aetiological agent in possum faeces, detected by real-time PCR targeting M. ulcerans IS2404, IS2606 and KR. Mycobacterium ulcerans DNA was detected in 20/216 (9.3%) ground collected ringtail possum faecal samples and 4/6 (66.6%) brushtail possum faecal samples. The distribution of the PCR positive possum faecal samples and human BU cases was highly focal: there was a significant non-random cluster of 16 M. ulcerans positive possum faecal sample points detected by spatial scan statistics (P<0.0001) within a circle of radius 0.42 km, within which were located the addresses of 6/12 human cases reported from the area to date; moreover, the highest sample PCR signal strength (equivalent to ≥106 organisms per gram of faeces) was found in a sample point located within this cluster radius. Corresponding faecal samples collected from closely adjacent BU-free areas were predominantly negative. Possums may be useful sentinels to predict endemic spread of human BU in Victoria, for public health planning. Further research is needed to establish whether spatial associations represent evidence of direct or indirect transmission between possums and humans, and the mechanism by which this may occur.  相似文献

The effect of two plant secondary metabolites, tannins and formylated phloroglucinol compounds (FPCs), on the intake of Eucalyptus foliage by common ringtail (Pseudocheirus peregrinus) and common brushtail possums (Trichosurus vulpecula) was studied. We manipulated the amount of tannin that was free to bind with protein by coating foliage with polyethylene glycol 4000 (PEG) and relied on natural intraspecific variation in FPC concentrations. In contrast to ringtail possums, brushtail possums showed a greater tolerance to FPCs and ate more foliage when it was coated with PEG, suggesting that tannins limited their food intake. Brushtails detected the effects of tannins through immediate oral sensations rather than through systemic effects. Ringtail possums appeared highly tolerant of foliar tannins yet susceptible to low concentrations of FPCs. We could not detect any interaction between tannins and FPCs that affected the intake of Eucalyptus foliage by either species of possum. Although ringtail and brushtail possums are widely regarded as specialist and generalist folivores, respectively, their differential susceptibility to co-occurring secondary metabolites suggests greater complexity. Each possum species appears to be a specialist in its own right, which leads to a partitioning of available foliage. Brushtails avoid tannins and ringtails avoid FPCs.  相似文献

The marsupials that eat Eucalyptus in south-eastern Australia provide an example of animals with similar niche requirements occurring sympatrically. They certainly differ in size, ranging from about 1 kg in the greater glider (Petauroides volans) and the closely related common ringtail possum (Pseudocheirus peregrinus), to 4 kg (common brushtail possum, Trichosurus vulpecula) and up to 15 kg in the koala (Phascolarctos cinereus). All species, however, may eat considerable amounts of eucalypt foliage, often favouring the same species, and thus appear to compete for food. In order to better understand the degree of competition for food, we measured feeding by the greater glider in response to increasing concentrations of a specific group of eucalypt plant secondary metabolites (PSM), the sideroxylonals, and then compared it to results published for the other species. The greater glider was more resilient than the other species to increasing concentrations of sideroxylonals. We suggest this allows gliders to feed on leaves from the eucalypt subgenus, Symphyomyrtus, while its small size and gliding ability allow it to feed where koalas cannot, on the young leaves on top of the canopy. In contrast, the common ringtail possum is well adapted to feeding from species of the subgenus Eucalyptus, which do not produce sideroxylonals but contain less available nitrogen (AvailN) than do the symphyomyrtles. These ‘nutritional niches’ segregate the forest and along with other factors, such as generalist and specialist feeding strategies and differences in body size and requirements for shelter, presumably minimise competition between the marsupial species.  相似文献

Abstract This study investigated the feeding ecology of the green ringtail possum, Pseudochirops archeri (Pseudocheiridae) in a tropical rainforest with 94 plant species in the canopy. Over 50% of tree use was from only four tree species, Aleurites rockinghamensis, Ficus fraseri, Arytera divaricata and Ficus copiosa. These species were used significantly more frequently than would be expected if tree species were selected randomly in proportion to their relative abundance in the forest. Conversely, 88 other tree species present were used less frequently than expected. Possums also favoured particular individual trees within some of the preferred tree species. In 91% of feeding observations, possums consumed mature leaves only. The availability of young leaves, flowers and fruit varied throughout the year, with a peak in availability of these resources during the early wet season. By primarily selecting mature leaves, green ringtail possums reduce their dependence on seasonally variable resources. We suggest that green ringtail possums should be considered as specialist folivores, focusing on only a few of the tree species available, possibly due to advantages associated with limiting the number of plant secondary metabolites in their diet. Furthermore, they favour certain individual trees within species, perhaps due to intraspecific variation in plant secondary metabolites or nutrient content, behaviour that has been well established in eucalypt folivores. We conclude that green ringtail possums are highly specialized in their feeding ecology, limiting their diet to a small number of continuously available food items.  相似文献

The Australasian marsupial order Diprotodontia includes ten extant families that are grouped into the suborders Vombatiformes (koalas and wombats), Macropodiformes (kangaroos and allies), and Phalangeriformes (possums and gliders). We investigated interfamilial relationships using mitochondrial 12S rRNA, valine tRNA, and 16S rRNA gene sequences. Our results support the monophyly of both Vombatiformes and Macropodiformes, but not Phalangeriformes. Among possums and gliders, there was strong support for a petauroid clade that includes Pseudocheiridae (ringtail possums), Petauridae (sugar glider, striped possums), Acrobatidae (feathertail possums), and the monotypic family Tarsipedidae, which is represented by the highly specialized and autapomorphic honey possum (Tarsipes rostratus). Other prior hypotheses for the phylogenetic placement of the honey possum were rejected by statistical tests. The inclusion of the honey possum within Petauroidea suggests that derived ultrastructural features of Tarsipes' spermatozoa evolved independently in Tarsipes versus polyprotodont Australasian marsupials.  相似文献

Myosin heavy chains (MyHCs) and fibre types in the masseter muscle of seven species of Australian marsupials (brushtail and ringtail possums, bettong, bandicoot, dunnart, two species of antechinuses) spanning three orders were studied by native myosin electrophoresis, SDS-PAGE, immunoblotting and immunohistochemistry. We found only two fibre types in the masseter muscles of these animals: (1) masticatory fibres expressing masticatory MyHC, and (2) hybrid α/β fibres that co-express α-cardiac and β-cardiac MyHCs. Masticatory fibres predominate in most species, being appropriate for predation or for chewing tough vegetable matter. The relative abundance of α/β fibres decreased from 60% to 0 in the order: ringtail possum > brushtail possum > bettong > bandicoot > dunnart/antechinus. These variations in masseter fibre type are correlated with decreasing amounts of vegetable matter in the diets of these animals. The results are in contrast to earlier work on masseter fibres of macropodids that expressed α-cardiac MyHC almost homogeneously. The fact that the bettong (Family: Potoroidae), which belong to the same marsupial superfamily (Macropodoidea) as kangaroos and wallabies (Family: Macropodidae), has not specialized in the exclusive expression of α-cardiac MyHC as members of the latter family suggests that this specialization was of recent phylogenetic origin (30 million years before present).  相似文献

Twenty-one polymorphic microsatellite markers were developed for the Virginia opossum (Didelphis virginiana). The number of alleles ranged from two to 13 and observed heterozygosities ranged from 0.464 to 0.964. Significant heterozygote deficiencies were observed at three loci and null alleles were detected at five loci. Evidence for gametic disequilibrium was observed between three sets of paired loci after a sequential Bonferroni correction was applied. These markers will enable us to investigate the mating tactics, movement behaviour and social structure of Virginia opossum populations inhabiting fragmented agricultural landscapes.  相似文献

Nine highly polymorphic microsatellite markers were developed for Gehyra variegata from an enriched library screened for tetranucleotide repeats. An average of 19.44 alleles per locus and an average observed heterozygosity of 0.86 were found in 81 samples from fragmented populations in the Western Australian wheatbelt. Different gecko species exhibit varying rates of extinction in fragmented landscapes and the genetic markers were developed to test whether these difference are due to different rates of dispersal. The loci were tested for their ability to cross amplify in 12 species of Australian Gekkonidae and Pygopodidae, in an attempt to identify species which might be amenable to population genetic analysis using these microsatellite markers.  相似文献

1. Mean retention times (MRTs) of fluid (marked with Co-EDTA), fine particles (mordanted with Yb) and large particles (mordanted with Cr) were measured in brushtail possums (Trichosurus vulpecula), ringtail possums (Pseudocheirus peregrinus) and laboratory rabbits fed semipurified diets. 2. In brushtail possums there were no significant differences in MRT among the three digesta markers. 3. In ringtail possums MRTs of the fluid and fine particle markers were approximately twice that of the large particle marker, indicative of selective retention of both fluid and fine particles in the caecum. 4. In the rabbit MRT of fine particles was also greater than that of large particles, again indicative of selective retention of fine particles in the caecum. 5. Fibre digestibility was greater in the rabbits than in the ringtail possums, and greater for neutral-detergent fibre (including agar) but less for acid-detergent fibre in the rabbits than in the brushtails. Differences in fibre digestibility between brushtails and rabbits were explained by differences in patterns of digesta flow. However, the higher digestibilities of fibre in the rabbits than in the ringtail possums could not be explained on a similar basis.  相似文献

The diversity of class II major histocompatibility complex (MHC) loci was investigated in the brushtail possum, an important marsupial pest species in New Zealand. Immunocontraception, a form of fertility control that generates an autoimmune response, is being developed as a population control method for the possum. Because the immune response is partly under genetic control, an understanding of immunogenetics in possum will be crucial to the development of immunocontraceptive vaccines. MHC molecules are critical in the vertebrate immune response. Class II MHC molecules bind and present exogenously derived peptides to T lymphocytes and may be important in the presentation of immunocontraceptives. We used polymerase chain reaction primers designed to amplify the peptide binding region of possum class II MHC genes to isolate sequences from 49 animals. We have previously described 19 novel alleles from the DAB locus in the possum (Holland et al., Immunogenetics 60:449–460, 2008). Here, we report on another 11 novel alleles isolated from possum DAB, making this the most diverse marsupial locus described so far. This high level of diversity indicates that DAB is an important MHC locus in the possum and will need to be taken into account in the design of immunocontraceptive vaccines.  相似文献

1. Lysozyme and alpha-lactalbumin from the milk of the common ringtail possum have been purified and partially sequenced. 2. The lysozyme had similar enzymic activity to the c-type lysozyme of the domestic hen and 43% homology over the N-terminal 49 residues. 3. alpha-Lactalbumin was present in the milk in two biologically active forms; the more acidic form had 66% sequence homology with the N-terminal 35 residues of red-necked wallaby, 54% with human and 43% with bovine alpha-lactalbumin. 4. SDS polyacrylamide-gel electrophoresis of milk samples showed that alpha-lactalbumin was present in the milk throughout lactation but that lysozyme first appeared only in mid-lactation.
