The spatial scale at which samples are collected and analysed influences the inferences that can be drawn from landscape genetic studies. We examined genetic structure and its landscape correlates in the pitcher plant midge, Metriocnemus knabi, an inhabitant of the purple pitcher plant, Sarracenia purpurea, across several spatial scales that are naturally delimited by the midge’s habitat (leaf, plant, cluster of plants, bog and system of bogs). We analysed 11 microsatellite loci in 710 M. knabi larvae from two systems of bogs in Algonquin Provincial Park (Canada) and tested the hypotheses that variables related to habitat structure are associated with genetic differentiation in this midge. Up to 54% of variation in individual‐based genetic distances at several scales was explained by broadscale landscape variables of bog size, pitcher plant density within bogs and connectivity of pitcher plant clusters. Our results indicate that oviposition behaviour of females at fine scales, as inferred from the spatial locations of full‐sib larvae, and spatially limited gene flow at broad scales represent the important processes underlying observed genetic patterns in M. knabi. Broadscale landscape features (bog size and plant density) appear to influence oviposition behaviour of midges, which in turn influences the patterns of genetic differentiation observed at both fine and broad scales. Thus, we inferred linkages among genetic patterns, landscape patterns and ecological processes across spatial scales in M. knabi. Our results reinforce the value of exploring such links simultaneously across multiple spatial scales and landscapes when investigating genetic diversity within a species.  相似文献

Ten polymorphic microsatellite loci were cloned and characterized for the marine midge Clunio marinus (Chironomidae, Diptera). The number of alleles ranged from three to 31, the observed heterozygosity ranged from 0.06 to 0.83. During preliminary tests for polymorphisms, we identified subtly differing alleles due to the heteroduplexes they formed on nondenaturing gels. This allowed for the use of unlabelled primers in standard lower resolution PAGE and thus saved time and money in primer testing.  相似文献

The western cherry fruit fly (Rhagoletis indifferens) is a major pest of cherry crops in western North America, yet relatively little is known about its population structure or movement patterns due to the difficulty of studying these parameters directly in the field. We isolated and characterized a set of 16 polymorphic microsatellite loci specific to R. indifferens that display sufficient polymorphism to assess genetic structure and movement patterns. All 16 loci amplified in one or more other Rhagoletis species, indicating that they are useful tools for genetic analysis in other members of the genus, many of which are also agricultural pests.  相似文献

The house fly, Musca domestica L., is a cosmopolitan species with a capacity to transmit human pathogens. Here, we report on the development of polymorphic microsatellite loci for house flies and present preliminary results from four house fly subpopulations from Manhattan, Kansas. Twenty‐four microsatellite loci were isolated and characterized using DNA enriched for repeat sequences. Forty individuals from four locations in Kansas were assayed to identify for polymorphic loci. Eight loci were polymorphic with the number of alleles ranging from three to six.  相似文献

Abstract.  1. The effect of meso-scale (zone within bog and local plant density) and fine-scale (leaf length and resource availability) factors on the colonisation of pitcher plant leaves by arthropods was examined in an eastern Canadian bog.
2. In spring, the abundances of three arthropods, the mosquito Wyeomyia smithii , the midge Metriocnemus knabi , and the mite Sarraceniopus gibsoni , were determined for plots with low, moderate, and high densities of pitcher plants. All overwintering inhabitants were then removed from the plots. Newly opening leaves were colonised from outside the plots, and arthropod abundances were assessed again in autumn.
3. Pitcher plant fauna varied in their response to the meso-scale factors. In autumn (soon after colonisation), midges were more abundant in areas with high densities of pitcher plants. The relationship between mosquito abundance and plant density, and the variation in abundance among zones within the bog in the spring, were probably due to overwintering mortality.
4. All taxa responded to the fine-scale factors, leaf length, and capture rate, in the autumn, but the strength of the responses frequently depended on a meso-scale factor (plant density), in which responses were usually strongest where plants were sparse. Thus, the interaction between meso- and fine-scale processes needs to be considered when interpreting patterns of species abundance within arthropod assemblages in pitcher plant leaves.  相似文献

The population growth and biomass production of the pitcher-plant (Sarracenia purpurea L.) inquiline, Habrotocha rosa Donner (Rotifera: Bdelloidea), its consumption by other pitcher-plant inqulines, and its excretion of phosphorus (PO4–P) and nitrogen (NO3–N and NH4–N), were investigated in laboratory experiments. Observed population growth and production rate of H. rosa were higher at pH 4 (2.3 rotifers d-1) than at pH 3 (1.3 rotifers d-1), 5 (1.9 rotifers d-1), or 6 (0.8 rotifers d-1). Populations of H. rosa are an abundant and reliable food source for larvae of the dipteran inqulines Wyeomyia smithii (Coq.) and Blaesoxipha fletcheri (Aldrich) that co-occur with H. rosa in S. purpurea pitchers. Abundance of H. rosa within a pitcher is negatively associated with abundance of dipteran larvae, and these larvae consume rotifers in direct proportion to rotifer density (Type I functional response). Habrotrocha rosa may also account for the majority of the plant's supply of N and P. An average population of rotifers in the field (∼400 per pitcher) can excrete ∼5.2 μg NO3-N, ∼3.91 μg NH4-N, and ∼18.4 μg PO4–P per day into a single leaf, and excretion rate is independent of water pH. Over the six-month growing season of pitcher-plants in Massachusetts, U.S.A., we estimate that rotifers could supply 8.8–43 mg of N and 18.2–88 mg of P. These values far exceed the amount of N and P previously estimated to be supplied annually to the plants through insect capture or rainfall. Ten microsatellite loci were isolated from the Chinese alligator (Alligator sinensis), using a partial genomic DNA library and an enrichment method. Fifty microsatellite repeats were screened from the library of 888 positive clones, 10 of which were polymorphic. These microsatellite loci developed here are another set of novel molecular markers for A. sinensis, and they will be suitable for population genetic studies as well as for kinship analysis.  相似文献

The phytotelmata of the North American pitcher plant Sarracenia purpurea are colonised by a great variety of aquatic organisms and, thus, provide an ideal model to study trophic interactions in small freshwater ecosystems. Although algae are discussed as a potential food source for predators, little is known about the structure of algae coenoses in pitchers of S. purpurea. This study aims to elucidate temporal shifts in the algae community structure in pitchers of an allochthonous population of S. purpurea in Saxony, Germany. A total of 78 algae taxa was found in the pitchers. Mean algae abundances in new and old pitchers were similar (2.6 x 10(5) and 2.3 x 10(5) algae ml(-1), respectively). Taxa from the orders Chlamydomonadales, Chlorococcales, and Ochromonadales were the primary colonisers. With increasing age of the pitchers the filamentous green algae from the order Klebsormidiales became more abundant. In contrast, pennate diatoms dominated the algae coenoses in the fen. Algae community structure in vase-shaped 50 ml Greiner tubes was similar to those of natural pitchers. Differences in the temporal patterns of algae coenoses in individual pitchers suggested a colonisation of the pitchers by algae via trapped insects, air and rain water rather than via the surrounding fen. Biomass of algae approximated 0.3 mg C ml(-1), which corresponds to 82.8 % of the living biomass (bacteria, heterotrophic nanoflagellates, algae, protozoans and rotifers). Rotifers were abundant in new pitchers; nematodes and mites were seldom found in all pitchers. A similar qualitative and quantitative composition of the aquatic biocoenoses was observed in pitchers of another allochthonous S. purpurea population growing in Blekinge, Sweden. Biomass of algae represented nearly one quarter of the total organic matter content in the pitchers. Thus, nitrogen and phosphorus compounds present in the algae biomass might be used by the carnivorous S. purpurea plant as additional food source in allochthonous populations in Europe lacking top predators.  相似文献

Fennel (= Sago) pondweed (Potamogeton pectinatus L.) is a submersed macrophyte of nearly cosmopolitan distribution. The plant is of worldwide ecological importance as structuring component of shallow lakes, and as food for waterfowl. We developed nine polymorphic microsatellite primers for the population genetic analysis of P. pectinatus. The loci were identified using a GA/CT‐enriched genomic library using subtractive hybridization with magnetic particles. All nine loci were highly polymorphic with 6–9 alleles and heterozygosities ranging from 0.23 to 0.80 in a subset of N = 40 genotypes from five locations.  相似文献

Sarracenia purpurea L. plants were studiedin seven bogsalong the eastern seaboard of the United States fromNew Jersey to Georgia. Water sampled from 225 leaveswas found to contain several invertebrates. A singlebdelliod rotifer, a form of Habrotrocha cf. rosaDonner very similar to that described by Bateman(1987) in S. purpurea in western Newfoundland,was identified in 70% of the leaves. The invertebrate communities in Sarracenia purpurea were characterized by surveying the contents of 474 leaves from six pitcher plant populations in the eastern United States. More than 8200 invertebrate specimens, both living and dead, were recovered. Midge and mosquito larvae comprised 7907 (96%) of the specimens, with a mean of 16.9 dipteran larvae per leaf. Although midge larvae were present at all collection sites, mosquito larvae were absent from two of the sites. In the latter sites, the number of midge larvae per leaf was greater. This report documents for the first time that there is no statistical difference between the average number of Diptera larvae per leaf for sites that contain both midge and mosquito larvae compared to sites that contain only midge larvae. It appears that pitcher plants maintain an average number of larvae per leaf based on a number of interacting factors which we do not fully understand. This investigation documents that in the absence of mosquito larvae, more midge larvae are found per leaf per leaf than would be predicted based on data from sites that contain both larval taxa.  相似文献

We developed six microsatellite loci for song wrens (Cyphorhinus phaeocephalus). Polymorphism ranged from four to 12 alleles with heterozygosities ranging from 21 to 90%. Three of the six loci deviated significantly from the Hardy–Weinberg equilibrium suggesting the presence of one or more null alleles. Cross‐species amplification indicated that these loci will have limited utility in other wren species.  相似文献

We developed polymerase chain reaction primers for nine dinucleotide microsatellite loci in the marine deep‐sea fish, the tusk (Brosme brosme). All markers were obtained from a partial genomic DNA library, and characterized in 100 unrelated individuals from one putative population. The number of alleles ranged from two to 60 with an average of 15/locus. The observed heterozygosity ranged from 0.020 to 0.826 (average 0.438). Several of the markers amplified multiple alleles from the two other gadoid species tested.  相似文献

All markers were obtained from a partial genomic DNA library, and characterized in 90 unrelated individuals from one putative population sampled on the Mid-Atlantic Ridge. The number of alleles ranged from two to 61 with an average of 21 per locus. The observed heterozygosity levels ranged from 0.301 to 0.987 with an average of 0.672. Several of the markers amplified multiple alleles from either the Atlantic cod (Gadus morhua) or the deep-sea fish roughhead grenadier (Macrourus berglax).  相似文献

A microsatellite‐enriched partial genomic DNA library of Lygus hesperus was generated and screened by sequencing. Ten polymorphic microsatellite marker loci were characterized by genotyping 92 insect samples. The observed number of alleles ranged from three to seven with an average of 4.5 (SE ± 0.45), while the effective number of alleles ranged from 1.21 to 3.02 with an average of 2.14 (SE ± 0.20). Significant deviations from Hardy–Weinberg expectations were detected at three loci. Significant linkage disequilibrium was also detected between the loci LhMS2‐54 and LhMS3‐32. Seven of the L. hesperus markers could be transferred to Lygus lineolaris.  相似文献

Ten polymorphic microsatellite loci from Lasiopodomys brandtii have been isolated and characterized. Two to 11 alleles per locus were detected from 52 Brandt's voles samples collected from a single population. Expected heterozygosities ranged from 0.406 to 0.840. For the majority of loci observed heterozygosities were similar to or greater than the expected heterozygosity. One locus pair appeared to be in linkage disequilibrium. The microsatellite markers will enable the studies of genetic diversity, population structure and relatedness in this species, and perhaps in closely related species of vole.  相似文献

The stable fly, Stomoxys calcitrans (L.), is a significant pest of cattle. Twenty-three microsatellite markers were isolated from a repeat-enriched genomic library of S. calcitrans. We characterized variation at these markers and found that 17 loci were polymorphic in two fly populations from Florida. Two to nine alleles were observed among the variable microsatellite loci and expected heterozygosities ranged from 0.03704 to 0.85115. These markers will be useful for characterizing population genetic differentiation and for tracking the migration patterns of stable flies in the USA and worldwide.  相似文献

A total of nine polymorphic microsatellite loci were obtained from a genomic library of Pangasius pangasius (order Siluriformes, family Pangasiidae). Samples from rivers Bhagirathi (n = 22) and Mahanadi (n = 20) were genotyped for each of the nine microsatellite loci to determine genetic variation. The mean number of alleles per locus was 5.22 in Bhagirathi and 5.78 in Mahanadi; and expected heterozygosity ranged from 0.567 (Bhagirathi) to 0.578 (Bhagirathi). Significant deviation (P < 0.003) from Hardy–Weinberg expectations was evident at three loci, Ppa2 (Bhagirathi), Ppa14 (Mahanadi) and Ppa28 (Bhagirathi and Mahanadi). The identified microsatellite loci were found to be promising for population genetics studies of P. pangasius.  相似文献

In order to complement on‐going research on reproductive strategies and dispersal in the eastern phoebe (Sayornis phoebe), we developed five polymorphic microsatellite loci which would permit us to assign parentage to offspring and to evaluate population genetic structure. The number of alleles per locus ranged from 3 to 15; probability of exclusion, with a known maternal genotype, was 0.995. Preliminary screening revealed that primers were polymorphic in other Tyrannidae.  相似文献
