The content of histamine, serotonin, adrenaline and noradrenaline in the thoracic and lymphatic duct lymph and blood as well as absolute quantity of lymph transported biogenic amines and mediators into the general circulation were studied on 68 dogs during anaphylactic and endotoxin shock (ASh and ESh, respectively). Both ASh and ESh were accompanied by considerable changes in the hemodynamics, lymph circulation, content of biologically active substances in lymph and blood and in their lymph transport to the blood stream. The most profound and early changes in the content of biologically active substances during ASh and ESh were found in the lymph, which shows an important role of the lymphatic system in their resorption and transport from organs and tissues into the general circulation.  相似文献

To elucidate the source and physiological significance of plasma 3,4-dihydroxyphenylalanine, the immediate product of the rate-limiting step in catecholamine biosynthesis, plasma 3,4-dihydroxyphenylalanine was quantified in conscious rats after administration of reserpine, desipramine, clorgyline, or forskolin, treatments that affect tyrosine hydroxylase activity. Plasma 3,4-dihydroxyphenylalanine was also examined during infusions of norepinephrine with or without clorgyline, reserpine, or desipramine pretreatment. After reserpine, the plasma 3,4-dihydroxyphenylalanine level decreased by 22% and then increased by 40%, a result consistent with modulation of tyrosine hydroxylase activity first by an increased axoplasmic norepinephrine content and then by depletion of norepinephrine stores. After desipramine, the plasma 3,4-dihydroxyphenylalanine level decreased by 20%, reflecting the depressant effect of neuronal uptake blockade on norepinephrine turnover. Forskolin increased the plasma 3,4-dihydroxyphenylalanine level by 30%, consistent with activation of tyrosine hydroxylase by cyclic AMP-dependent phosphorylation. Acute administration of clorgyline was without effect on the plasma 3,4-dihydroxyphenylalanine level. Norepinephrine infusions decreased the plasma 3,4-dihydroxyphenylalanine concentration, as expected from end-product inhibition of tyrosine hydroxylase. Pretreatment with desipramine prevented the norepinephrine-induced decrease in plasma dihydroxyphenylalanine content, indicating that inhibition of tyrosine hydroxylase required neuronal uptake of norepinephrine. Both reserpine and clorgyline augmented the norepinephrine-induced decrease in plasma 3,4-dihydroxyphenylalanine level, suggesting that retention of norepinephrine in the axoplasm--due to inhibition of norepinephrine sequestration into storage vesicles or catabolism--caused further inhibition of tyrosine hydroxylase. Changes in plasma 3,4-dihydroxyphenylalanine concentration during norepinephrine infusions were negatively correlated with those in plasma 3,4-dihydroxyphenylglycol level, a finding consistent with modulation of tyrosine hydroxylase activity by axoplasmic norepinephrine. In reserpinized animals, clorgyline and norepinephrine infusion together decreased the plasma 3,4-dihydroxyphenylalanine content by 50%, a result demonstrating that hydroxylation of tyrosine was depressed by at least half. The results indicate that quantification of plasma 3,4-dihydroxyphenylalanine can provide a simple and direct approach for examination of the rate-limiting step in catecholamine biosynthesis.  相似文献

This paper concerns structural and functional organization of the human lymphatic system during the prenatal period. For the first time the data is presented on the development of thoracic duct based on the concept of lymphangion as a structural and functional unit of a lymphatic vessel. The formation of the duct is considered in connection with structural changes of its wall and variants of duct roots fusion. Experiments on dogs were made to simulate aphthous process on the oral mucosa by occlusion of the common bile duct. The content of adrenaline, noradrenaline, serotonin, histamine and ascorbic acid was measured during the development of pathological process in the areas of oral mucosa tissue characterized by most frequent appearance of aphthas and in the tissue of the duodenal, small and large intestine mucosa. The magnitude of the characteristics indicated was determined 2, 6 and 12 h and 1, 3, 7 and 10 days after operation. Aphthas appeared on the 3d day. Within the first hours of the experiment there was a considerable increase in the content of noradrenaline, adrenaline and serotonin in the oral mucosa accompanied by a reduction in the histamine and ascorbic acid content. In the intestinal mucosa, shifts in the correlations between the hormones and transmitters were less pronounced. During aphthas appearance, the histamine content rose whereas other parameters decreased. The period of the reverse development was characterized by the recovery of the correlations of the hormones under study and transmitters.  相似文献

In 196 human embryos, prefetuses, fetuses and newborns, by means of a complex of morphological methods, development of the jugular lymphatic sacs and the process of settling of the thoracic duct cervical part topography have been studied. The jugular lymphatic sac anlages take place on the 6th week of the development. From the lymphatic cleft, situating in the mesenchyme near the anterior cardinal veins, multichambered cavities laid with endotheliocytes are forming,--the jugular lymphatic sacs. Connection of the initially close lymphatic sacs with the venous system takes place secondarily by the end of the embryonic period of development. In the area of the sac ostia a valve is formed, that makes morphological premises for unidirected lymph flow into the venous system. The lymph nodes developing at the place of the reducing jugular lymphatic sacs, ensure formation: from the left jugular lymphatic sac--the cervical part of the thoracic duct, from the right jugular lymphatic sac--the right lymphatic duct and the jugular and the subclavicular lymphatic trunks. Variability in the form and topography of these structures are determined both by the form and construction of the jugular lymphatic sacs and by developmental peculiarities of the lymph nodes at their place. The process of settling of the thoracic duct cervical part topography depends on age changes of its size and form, as well as on development of structures situating nearby, and by the time of birth it is not completed.  相似文献

The lymphatic bed has been studied in 141 hepatic preparations of the man, dog and white rat. Development of obturation in the common bile duct in the man and at modelling a similar process in the animals results in essential morphological adaptive-compensatory and destructive changes in the hepatic lymphatic bed. The adaptive-compensatory reconstruction is especially well seen at initial stages of the disease. It is manifested as a total dilatation of the bed, certain reserve elements get into work, the bed capacity increases. Then certain new structural units of the bed, collateral pathways develop. When the obturational process lasts long, certain destructive changes of the hepatic lymphatic bed elements take place. The change in the relief of endotheliocytes is their most characteristic sign. With increasing age of the patients the degree of the destructive rearrangements of the hepatic lymphatic bed increases. In the experiment carried out on the animals the lymphatic outflow from the thoracic duct is determined, being an indirect index of the hepatic lymph-forming function. An essential increase of the lymph volume, several times greater than in the control, gets from the duct into the venous bed. A connection is revealed between the morphological transformations of the bed components and its drainage function. The lymphatic bed of the liver performs also an active work concerning resorption and transport of bilirubin, its content in the organ rather increases, when the common bile duct is obturated.  相似文献

Aphthous lesions of the oral mucosa were simulated in dogs by common bile duct ligation. In one of the experimental groups, the animals were administered the beta-adrenoblocker obsidan 30 min before operation. Two hours after operation the regions of the oral mucosa mostly affected by aphthous lesions were examined for the content of adrenaline and noradrenaline (NA). The data obtained indicate a significant increase in the content of NA and adrenaline in the oral mucosa 2 h following operation. In animals pretreated with obsidan, the content of catecholamines remained at the level seen in the control group. Therefore, blocking the transmission of nerve impulses to the sympathetic nervous system obsidan interferes with the reflex influence from the involved organs of the abdominal cavity, thereby protecting the oral mucosa tissues from the damage-inducing action of high concentrations of NA.  相似文献

Distribution of catecholamines and serotonin in the thalamus and hypothalamus of the brain was studied as affected by hypothermia. Both single and double hypothermia are found to induce considerable shifts in distribution of these amines. In the both regions of the brain the content of serotonin increases greatly, that of dopamine and adrenaline rises to a less extent. Background aims: Manufacturing of potent chimeric antigen receptor (CAR) T cells requires phenotypically naive and early memory T cells. We hypothesized lymphatic fluid collected from the thoracic duct of children would serve as a unique reservoir for early T cells, which could then be used for CAR T-cell therapy. Methods: We evaluated lymphatic fluid collected from 25 pediatric patients undergoing thoracic duct cannulation for other clinical indications. Results: Lymphatic fluid in the thoracic duct was rich in T cells, with higher percentage of naive and stem central memory T-cell subsets compared with paired blood samples. T cells from lymphatic fluid showed decreased negative checkpoint regulators on the surface and increased rapid expansion with bead activation. Creation of CD19-directed CAR T cells from blood and lymphatic T cells showed similar lentiviral transduction properties, but CAR T cells generated from lymphatic fluid produced superior cytotoxicity in a murine leukemia model because they were able to achieve equivalent tumor eradication at lower doses. Conclusions: These results are the first characterization of T cells from the thoracic duct of pediatric patients and suggest an alternative approach for manufacturing of cellular therapy that will improve both expansion and cytotoxic effect.  相似文献

In 80 Wistar rats by means of electroroentgenolymphography lymphatic pathways and thoracic duct have been studied, normal and after ligation of the latter in 1-3 days, 1 and 2-3 months. Contrasting of the thoracic duct in the control animals reveals its additional ducts, that run in parallel to the main collector and they get into it after a certain distance, as well as different time phases of filling the duct are noted. When the thoracic duct is ligated, in 1-3 days dilatation of the ductal cistern, contrasting of lymphatic vessels and nodes in other regions are observed. In 1 month of the experiment the thoracic duct cistern is poorly expressed in comparison with the early time. The contrast substance continues to get into the group of the lymph nodes in other regions. A network of anastomoses of lymphatic vessels is formed in the area of ligation. In 2-3 months, when the contrast substance continues to get retrogradely into other groups and regions of the lymph nodes, final restoration of the lymph-flow takes place at the expense of formation of collateral pathways.  相似文献

Lactating ewes received continuous intravenous infusions of adrenaline (0.05 micrograms/kg liveweight) for 4 days. Prior to, during and after adrenaline infusions, milk yield and composition were monitored. Plasma concentrations of metabolites and hormones were measured each day and glucose biokinetics were measured in non-steady state at the start and end of adrenaline infusions. During adrenaline infusion, milk yield and content of solids-not-fat decreased and milk fat content was reduced on the first day of infusion. Plasma glucose was raised throughout the period of adrenaline infusion, plasma lactate increased over the first 4 h from the start of infusion and plasma non-esterified fatty acids increased for 2 h at the start of infusion and tended to increase during the first 2-3 h after withdrawal of adrenaline. Plasma growth hormone remained relatively stable except for a marked increase at 30 min after withdrawal of adrenaline. At the start and immediately after withdrawal of adrenaline infusion plasma insulin was increased approximately twofold. Glucose production, but not utilization, increased at the start of infusions. Immediately after withdrawal of adrenaline glucose utilization increased 2.5-fold with a smaller response in glucose production. There was essentially no change in glucose clearance during adrenaline infusion but a marked increase occurred after withdrawal of adrenaline.  相似文献

Neurokinin A and B are putative inflammatory mediators. We assessed their ability to alter prenodal lymphatic resistance. Intralymphatic neurokinin A (3.0 x 10(-6), 3.0 x 10(-5) and 3.0 x 10(-4) mol l(-1)) significantly constricted lymphatics at the two highest doses. Preliminary experiments suggested that neurokinin B might dilate lymphatics. To test this, lymphatic pressure was increased by norepinephrine (3.1 x 10(-6) mol l(-1)). Neurokinin B (2.7 x 10(-4) mol l(-1)) was then infused intralymphatically during norepinephrine infusion. Norepinephrine increased perfusion pressure from 5.6 +/- 0.6 mmHg to 12.1 +/- 1.4 mmHg. Subsequent infusion of neurokinin B significantly decreased lymphatic perfusion pressure from 11.9 +/- 1.3 mmHg to 9.9 +/- 1.1 mmHg. These data indicate that neurokinin A and B can alter lymphatic resistance and are consistent with the hypothesis that lymph vessel function may be subject to modulation by neurokinins.  相似文献

The mechanisms regulating the water-salt metabolism have been investigated in the experiment on dogs with a disturbance of the lymph flow system. The function of a regulator of the water-salt metabolism is found to be peculiar to the lymphatic system which is effected by means of the humoral properties of the lymph and discreteness of its static pressure in the thoracic lymphatic duct. The central lymph exerts a stabilizing effect on both the osmo- and volumoregulatory functions of the kidneys, cardio- and hemodynamics. The state of acute lymphohypertension in the system of the thoracic lymphatic duct activates the mechanisms of the water-salt metabolism aimed at decreasing the volume of the extracellular fluid.  相似文献

Influence of voluntary exercise on hypothalamic norepinephrine   总被引:5,自引:0,他引:5  
We combined hypothalamic tissue and plasma determinations ofnorepinephrine, dihydroxyphenylalanine, and dihydroxyphenylglycol withmeasurements of abdominal fat in voluntary running rats to examine therelationship among exercise training, hypothalamic and sympatheticnervous function, and body fat stores. The hypothalamic concentrationsof norepinephrine, dihydroxyphenylalanine, and dihydroxyphenylglycolwere reduced after exercise training(P < 0.01), with the amount ofnorepinephrine being strongly associated with the plasma norepinephrine(r = 0.58, P < 0.05) and dihydroxyphenylglycol (r = 0.65, P = 0.01) concentrations. Exercisetraining resulted in a diminution in abdominal fat mass(P < 0.01). A strongrelationship existed between fat mass and hypothalamic norepinephrinecontent (r = 0.83, P < 0.001). The presence of apositive relationship between the arterial and hypothalamicnorepinephrine levels provides presumptive evidence of an associationbetween noradrenergic neuronal activity of the hypothalamus andsympathetic nervous function. The observation that abdominal fat massis linked with norepinephrine in the hypothalamus raises thepossibility that alterations in body fat stores provide an afferentsignal linking hypothalamic function and the activity of thesympathetic nervous system.

The content of catecholamines and dihydroxyphenylalanine in larvae of the nudibranch Phestilla sibogae was analyzed by high-performance liquid chromatography with electrochemical detection. Dihydroxyphenylalanine, norepinephrine and dopamine were identified in larvae of all ages examined (5 through 12 days post-fertilization). Dihydroxyphenylalanine could be accurately quantified only in larvae of ages 8 through 12 days, when its average concentration increased from 0.62 to 6.71 × 10−2 pmol μg protein−1. Between ages 5 and 12 days dopamine rose from 0.081 to 0.616 pmol μg protein−1, and norepinephrine from 0.45 to 2.17 × 10−2 pmol μg protein−1. Dihydroxyphenylalanine, dopamine and norepinephrine were also measured at different stages of metamorphic progress in 10- to 12-day larvae. Dihydroxyphenylalanine increased by a factor of 2.4 between the onset and completion of metamorphosis, but levels of dopamine and norepinephrine remained stable. One millimolar alpha-methyl-dl-m-tyrosine, an inhibitor of catecholamine synthesis, inhibited natural metamorphosis and depleted endogenous norepinephrine and especially dopamine, respectively, to 75% and 35% of control values. The existence of unexpectedly high levels of catecholamines in metamorphically competent larvae, and the association of catecholamine depletion with inhibition of metamorphosis, indicate that these compounds may participate in the control of gastropod development. The lymphatic system has become the subject of great interest in recent years because of its important role in normal and pathological processes. Progress in understanding the origins and early development of this system, however, has been hampered by difficulties in observing lymphatic cells in vivo and in performing defined genetic and experimental manipulation of the lymphatic system in currently available model organisms. Here, we show that the optically clear developing zebrafish provides a useful model for imaging and studying lymphatic development, with a lymphatic system that shares many of the morphological, molecular and functional characteristics of the lymphatic vessels found in other vertebrates. Using two-photon time-lapse imaging of transgenic zebrafish, we trace the migration and lineage of individual cells incorporating into the lymphatic endothelium. Our results show lymphatic endothelial cells of the thoracic duct arise from primitive veins through a novel and unexpected pathway.  相似文献

This paper deals with the effect of the fatty acid chain length of dietary triglyceride on the intestinal uptake and lymphatic transport of exogenous and endogenous cholesterol in the rat. This question seemed of interest as the chain length of the monoglyceride and fatty acids formed in the intestinal lumen from the triglyceride fed could be expected to affect the concentration of cholesterol in the micellar or isotropic phase of intestinal content. Feeding rats medium- or short-chain triglycerides (C(12) to C(2)) did not affect the lymphatic transport of endogenous cholesterol from the intestine compared to the fasting state. The extent of lymphatic transport of cholesterol added to these fats increased proportionally with chain length (C(6)-C(18)) of the component fatty acids. The uptake of exogenous cholesterol into the intestinal wall was similarly related to the chain length of the carrier triglyceride, with the exception of triacetin, which gave a much higher intestinal uptake than lymphatic transport. When cholesterol was fed in octadecane, negligible amounts only were transported to the thoracic duct lymph. This again indicates the importance of the polar split products of dietary fat for cholesterol absorption.  相似文献

An anatomical investigation of extraorganic lymphatic vessels and regional renal lymph nodes has been performed in 70 dogs. The regional lymph nodes in the right kidney are stated to be quantitatively constant, as well as cranial and caudal lateroaortal lymph nodes in the left kidney in regard to the middle left lateroaortal nodes, that get lymph from the left kidney parenchyma. One middle left lateroaortal lymph node is found in 47 animals examined, two lymph nodes--in 17 animals. In 6 cases a lymphatic vessel, that gets lymph from the renal parenchyma and independently runs into the cistern of the thoracic duct is found for the first time. The variant revealed is an exception from the rule known in lymphology: lymph in its way from periphery to the central collector runs, at least, through one lymph node.  相似文献

The morphofunctional state of lymphatic nodes, thyroid gland and testis have been studied with morphometric, cell photometric and biochemical methods in experiments on adult white rats under the influence of nonspecific adaptation stress and activation reactions caused by small doses of adrenaline (125 mg/kg and 5 mg/kg). The research showed that the changes in the structure and function of the lymphatic organs under stress and activation were markedly different. If stress results in depression of functional state of these organs, the activation reaction, on the contrary, stimulates it.  相似文献

The spontaneous contractile activity of isolated lymph nodes and the lymph flow from intestine lymphatic vessel in antiorthostatic posture of rats with an inclination angle of 30 degrees during 7-14 days, was decreased. Contractions of the rat lymph nodes in response to actions of adrenaline, acetylcholine and histamine were diminished. There are changes of biochemical components of lymph and blood plasma with simultaneous decrease of the blood plasma volume. It is concluded that the lymphatic system on antiorthostatic posture plays the compensatory role with the purpose of stabilization of homeostasis in the brain.  相似文献
