The entomopathogenic fungus Metarhizium anisopliae (Metschn.) Sorokin is widely used for biocontrol of pest insects, and many commercial products are on the market or under development. The aim of this review is to summarise all relevant safety data of this fungus, which are necessary for the commercialisation and registration process. The review contains the following sections: (1) identity, (2) biological properties (history, natural occurrence and geographical distribution, host range, mode of action, production of metabolites/toxins, effect of environmental factors), (3) methods to determine and quantify residues, (4) fate and behaviour in the environment (mobility and persistence in air, water and soil), (5) effects on non-target organisms (microorganisms, plants, soil organisms, aquatic organisms, predators, parasitoids, honey bees, earth worms, etc.), (6) effects on vertebrates (fish, amphibia, reptiles, and birds), and (7) effects on mammals and human health (allergy, pathogenicity/toxicity). On the basis of the presented knowledge, M. anisopliae is considered to be safe with minimal risks to vertebrates, humans and the environment. 相似文献

Transformation of the entomopathogenic fungus Metarhizium anisopliae to benomyl resistance 总被引:2,自引:0,他引:2
Abstract Protoplasts of the entomopathogenic fungus Metarhizium anisopliae were transformed to benomyl resistance using cosmid pSV50 which harbours a β-tubulin gene cloned from a Neurospora crassa benomyl-resistant mutant. Transformant colonies, which appeared at a frequency of 4 per 50 μg DNA, grew and sporulated on 10 μg/ml benomyl, whereas the wild type was inhibited by 3 μg/ml. Southern blot hybridization of DNA from transformants showed that, in each case, tandem repeats of the cosmid had integrated at several chromosomal loci. The transformants were mitotically stable when subcultured on non-selective agar and retained the ability to infect and kill larvae of Manduca sexta . Two transformants were less virulent than the wild type and one of them showed slower in vitro spore germination. The benomyl-resistant phenotype persisted in reisolates from insect cadavers. 相似文献

The entomopathogenic fungus Metarhizium anisopliae (Metsch.) Sorokin (Hypocreales: Clavicipitaceae) was applied in maize fields to control the Western Corn Rootworm Diabrotica virgifera virgifera Le Conte (Coleoptera: Chrysomelidae). Establishment and persistence of two strains of M. anisopliae were investigated after application as ‘fungal colonized barley kernels’ (FCBK) into the soil and as a spore suspension on maize leaves and on the soil surface in 2006 and 2007 at two locations in Hungary. The applied fungal strains were able to establish at both locations and a long‐term persistence of at least 15 months could be recorded in the soil. A positive correlation between density of colony forming units (CFU) in the soil and the soil inhabiting stages of the host insect D. v. virgifera could be found. M. anisopliae spores applied on maize leaves were able to survive for no longer than 3 days after application, whereas on the soil surface a noticeably increase of fungus densities were found after treatments. Investigations on the destruxin production of the entomopathogenic fungus Metarhizium anisopliae 总被引:3,自引:0,他引:3
The dynamics of cyclic peptide destruxins (dtxs) produced by Metarhizium anisopliae strains V245 and V275 were monitored both on solid and in liquid media. The results showed that both strains did not produce dtxs in large-scale fermenter cultures or solid Czapek Dox (CD) agar. Production of the major dtxs A and B could be determined in both strains when grown on rice for up to 10-30 days. The main dtxs A, B, E, and E diol were detected in CD liquid culture filtrate from both strains after three days post-inoculation on. Parallel decrease of dtx E and increase of E diol in the culture medium were found, indicating that the latter is the hydrolytic product from the former. Production of dtxs A and B was significantly positively correlated. A negative correlation was observed between the production of the metabolites and pH value of the medium. The influence of different nutrient sources on dtx production was evaluated by using media with different carbon and nitrogen ratios as well as with different insect homogenates. The findings showed that the amount of dtxs A, B, and E increased with the increasing content of peptone in the medium. When insect homogenate was used as single nutrient source or added to CD medium, no toxins were detected in the culture filtrate. The potential risk posed by the toxic metabolites during mass production is discussed. 相似文献

Fourteen polymorphic microsatellite markers were isolated from the entomopathogenic fungus, Metarhizium anisopliae, based on enriched genomic libraries. In order to assess allelic variability, the microsatellite loci were analysed in a collection of 34 isolates sampled from across Switzerland. The number of detected alleles in 14 loci ranged from two to eight and expected heterozygosity from 0.265 to 0.808. Because of the high expected heterozygosity, the 14 microsatellite loci are very useful for ecological studies and analysis of population diversity, and to identifying, monitoring, and tracking M. anisopliae strains applied as biological control agents. 相似文献

Mitochondria of Metarhizium anisopliae contain an alternative oxidase (AOX), which reduces oxygen to water by accepting electrons directly from ubiquinol. AOX activity is demonstrated in situ as a constitutive enzyme. Greatest activity of AOX appears at the beginning and at the end of the fungal developmental cycle, germination of aerial conidia and the formation of submerged conidia, respectively. Changes in nutritional conditions, e.g., the presence of host insect cuticle or nutrient starvation had no effect on the induction of AOX activity. Antimycin A, an electron transport chain inhibitor, induced AOX activity. Cloning of the AOX DNA and the alignment of the deduced amino acid sequence of a segment of the AOX gene from M. anisopliae shows structural similarities with other AOX sequences with differing levels of variation when compared with homologous sequences from plants, yeasts, and filamentous fungi. Alternative oxidase in entomopathogenic fungi may have a positive contribution to ecological fitness. 相似文献

Social insects employ many types of defense mechanisms against parasites and pathogens because they face high risks from infections
due to crowded living conditions with closely related nestmates. Grooming behavior, including self-grooming and allogrooming,
can remove fungal spores on the cuticles of social insects and may be a behavioral defense mechanism to improve survivorship.
Allogrooming between nestmates has been predicted to be especially important as a defense against ectoparasites. However,
little is known about the plasticity of grooming behavior in susceptible environments. Here, we show that the ant Lasius japonicus increases the frequency of allogrooming rather than self-grooming to improve survivorship against the entomopathogenic fungus
Metarhizium anisopliae. We found that increasing fungal dosage and ant group size influenced the plasticity of grooming behavior. Additionally,
the survival rate of the ants over 30 days improved with increased group size. Our results suggest that social insects opt
for altruistic behavior to prevent the spread of diseases. This study illustrates how ants maintain colonies through grooming

Purification and characterization of a novel chitinase from the entomopathogenic fungus, Metarhizium anisopliae 总被引:6,自引:0,他引:6
A novel chitinase was detected in extracellular culture fluids of the entomopathogenic fungus Metarhizium anisopliae (ATCC 20500) grown in liquid medium containing chitin as a sole carbon source. A chitinase was purified to near homogeneity from culture broth of M. anisopliae by DEAE-Sephacel, CM-Sepharose CL-6B ion-exchange chromatography, and gel filtration with Superose 12HR. The molecular mass of the enzyme determined by SDS-polyacrylamide gel electrophoresis was approximately 60 kDa and the optimum pH of the enzyme was 5.0. This molecular mass is different from values of 33, 43.5, and 45 kDa for endochitinases and 110 kDa for an exochitinase (N-acetylglucosaminidase) from M. anisopliae ME-1 published previously. In addition, N-terminal sequences of 60-kDa chitinase are different from those of 43.4- and 45-kDa endochitinases. The purified enzyme showed high chitinolytic activity against colloidal, crystalline chitin of crab shells as well as against p-nitrophenyl-beta-d-N-acetylglucosamide, p-nitrophenyl-beta-d-N, N'-diacetylchitobiose, and p-nitrophenyl-N, N'-N"-triacetylchitotriose, indicating that this enzyme has both endo- and exochitinase activity. 相似文献

Abstract Three different methods, (i) PEG, (ii) electroporation and (iii) biolistic, were employed to transform Metarhizium anisopliae using benomyl resistance as a selectable marker. Transformation frequencies and mitotic stability varied for each method, from 0.8 to 6.9 transformants μg−1 of DNA and 46%, respectively, by the PEG method; 1.3 to 1.8 transformants μg−1 of DNA and 67% by electroporation; and 32 to 201 transformants μg−1 of DNA and 90% by biolistic. Eight native isolates of the entomopathogenic fungus Metarhizium anisopliae (Metschnikoff) Sorokin were obtained by monitoring soils cultivated in a conventional manner. These isolates were compared in three areas: (a) conidial germination, (b) radial growth and sporulation and (c) ability of conidia to infect Tenebrio molitor larvae. All bioassays were carried out at constant temperatures of 10, 15, and 20 °C. Conidia of individual isolates demonstrated differences in germination after a 24-h long incubation at all evaluated temperatures. At 20 °C, the germination ranged from 67 to 100 % and at 15 °C from 5.33 to 46.67 %. At 10 °C, no germination was observed after 24 h; nevertheless, it was 8.67–44.67 % after 48 h. In terms of radial growth, the culture diameters and the associated production of spores of all isolates increased with increasing temperature. At 10 °C, sporulation was observed in three isolates while all remaining cultures appeared sterile. Three weeks post-inoculation, conidia of all assessed isolates caused 100 % cumulative mortality of treated larvae of T. molitor at 15 and 20 °C with the exception of isolate 110108 that induced 81.33 % mortality at 15 °C. At 10 °C, larval cumulative mortality ranged from 6.67 to 85.33 % depending on the isolate. Isolates 110108 and 110111 showed significantly slower outset and a much lower rate of infection at all temperatures compared to other tested isolates of M. anisopliae. The bioassays were carried out with the purpose to sort and select indigenous isolates of M. anisopliae useful as biocontrol agents in their original habitat. 相似文献

A general method for the assay of enzymes which produce ATP, or are susceptible to be coupled to ATP-producing enzymes, is described. We have applied it to the assay of glyceraldehyde-3-phosphate dehydrogenase and enolase. For these enzymes, the product of the reaction, 1,3-bisphosphoglycerate or phosphoenolpyruvate, were coupled to ADP and either phosphoglycerate kinase or pyruvate kinase, respectively. The ATP formed in both cases is used by hexokinase plus labeled glucose to produce labeled glucose 6-phosphate which is quantitatively separated in small Dowex 1 columns and measured by liquid scintillation spectrometry. The conditions described permitted the detection of 0.1 mU of glyceraldehyde-3-phosphate dehydrogenase or enolase. As a further example of the sensitivity of the radioassay, effluents of a CM-cellulose column charged with an extract prepared from one single frog oocyte were analyzed and shown to contain a single enolase and two glyceraldehyde-3-phosphate dehydrogenase fractions. 相似文献

However, a period of soaking in distilled water (10–44 h) accelerated the process of germination when a suitable nutrient source was provided. Conidial spores are often used as the infectious agent during insect biocontrol applications of entomopathogenic fungi. Here we show differential virulence of conidia derived from Metarhizium anisopliae strain EAMa 01/58-Su depending upon the solid substrata used for cultivation, where LC50 values differed by up to ~10-fold (5.3×106?4.5×105 conidia/ml) and LT50 values by ~40% (9.8?7.1 d). This fungal strain is also known to secrete proteins that are toxic towards adult Mediterranean fruit flies, Ceratitis capitata, and the Greater wax moth, Galleria mellonella, larvae. In vitro production and intrahemoceol injection using G. mellonella as the host was used to test fractions during purification of the protein toxins, demonstrating that they elicited defence-related responses including melanisation and tissue necrosis. Production of these proteins/peptides along with a number of potential cuticle degrading enzymes was confirmed both in vitro and during the infection process (in vivo). Two-dimensional gel electrophoresis, followed by gel elution and bioassay, was used to identify at least three proteins or peptides (molecular mass=11, 15 and 15 kDa) as mediating the observed insect toxicity. These data demonstrate that in vitro screening for insect toxins can mimic in vivo (i.e. during the infection process) secretion and applies the use of proteomics to invertebrate pathology. 相似文献

Entomopathogenic fungi, such as Metarhizium anisopliae (Metch.) Sorokin, are important agents for the biological control of insect pests. However, these fungi are not compatible with high temperatures. In this study, mutagenesis using ion beams or gamma rays was used to generate five potentially thermotolerant mutants from two wild-type isolates of M. anisopliae (two using ion beams and three using gamma rays). The mutant isolates had a higher upper thermal limit for vegetative growth compared to the wild types (by 2–3°C) and enhanced tolerance to wet–heat stress of 45°C for conidial germination. At 25°C and 30°C, most mutants were as virulent to maize weevil adults as the wild type, however, one mutant produced using ion beams almost lost virulence entirely. These results indicate that ion beams and gamma rays are useful tools for improving biological characteristics, such as thermotolerance, in entomopathogenic fungi, but that mutants must be carefully evaluated for unpredictable negative side effects. 相似文献
