The effect of sublethal exposure to copper on the diel activity of sea catfish, Arius felis, was examined after 72 h static exposure to copper (0.0, 0.05, 0.1, or 0.2 mg l–1 ). The locomotor behavior of 28 individual fish was electronically monitored in a 16-compartment rosette tank. Activity was quantified by the number of entries into compartments per hour. Controls and copper-exposed fish (0.05 mg l–1) behaved similarly after treatment and displayed no significant effects of handling or copper exposure on diel activity. However, fish exposed to 0.1 and 0.2 mg l–1 copper were hyperactive immediately following exposure; these fish had significantly greater numbers of entries into compartments per hour for entire 24-h monitoring periods (P < 0.01) and for the 12 h of photophase (P < 0.001) and of scotophase (P < 0.001) than fish in the other two experimental groups. The normal diel activity cycle was abolished. Additionally, exposure of fish to copper at 0.1 and 0.2 mg l–1 resulted in significantly less variability in activity (P < 0.05) compared to control fish and those exposed to 0.05 mg Cu l–1.  相似文献

The effect of exposure to different concentrations of food and suspended silt on filtration, respiration and condition were studied in the freshwater mussel Hyridella menziesi. Using a milk solids-based food and kaolin to simulate silt, mussels were maintained at different combinations of food and silt concentrations for 3 weeks. Between treatments mean filtration rates ranged from 0.97–1.66 l g–1 h–1, and respiration from 0.50–1.35 mg O2 g–1 h–1. Silt (non-volatile suspended solids up to 35 mg l–1) failed to have a significant effect on filtration rate or condition, but with increasing food levels (volatile suspended solids up to 35 mg l–1) filtration rate was reduced, and condition was reduced at the lowest food concentration (<5 mg l–1). Respiration showed a food × silt interaction between treatment blocks. When food was low respiration increased with increasing silt concentrations, and when silt was low (<5 mg l–1) respiration increased with increasing food concentrations. The observed effects of food and silt on filtration, respiration and condition are discussed in terms of their potential for affecting contaminant bioaccumulation. In low-food situations (i.e., <5 mg l–1), if mussels are pumping large volumes of water, contaminant uptake rates could be enhanced, whereas abundant food would result in lower pumping rates and lower uptake rates. Changes in metabolism with food concentration have implications for contaminant elimination, and changes in biochemical composition associated with changing condition could affect the tissue distribution and retention of contaminants.  相似文献

Intensively cultured fish stock when fed protein-rich feeds typically excrete high concentrations of total ammoniacal-nitrogen (TAN) into the water column which can have adverse effects on productivity, and upon the environment when aquaculture water is discharged. An immobilized culture of nitrifying bacteria isolated from prawn pond water and known to effectively remove TAN from saline water was tested for its ability to remove TAN from freshwater. The culture was readily adaptable to non-saline conditions and maintained TAN at less than 0.25 mg l–1, even with a daily addition of 3.2 to 4.2 mg TAN l–1 per d. The use of the immobilized culture of nitrifying bacteria represents an innovative and economical in situ treatment technology for removal of TAN in both saline and freshwater.  相似文献

Summary Simultaneous measurements have been made of the efflux rates of tritiated water and radiosodium from the lung-fishProtopterus annectens under conditions in which the fish can either use its gills, its lungs or both for respiration. Tritiated water efflux rate was similar (43–44% h–1) when the fish was in oxygenated water with or without access to air but decreased slightly in deoxygenated water. An efflux rate constant of 44% h–1 obtained in oxygenated water is similar to those of other inactive fish in fresh water. The efflux rate constant for sodium was 0.84% h–1 and total sodium flux was comparable to that of fresh-water fish. The decrease in both tritiated water and sodium efflux rates in deoxygenated water are explained in terms of diversion of blood flow away from the functional gills to the lungs. Oxygen consumption was highest (38.7 ml kg–1 h–1) in deoxygenated water and lowest when the fish was out of water. Kinetics of nitrate uptake by freshwater algae   总被引:2,自引:2,他引:0  
The kinetics of nitrate (NO3 ) uptake, the maximum uptake velocity (Vm) and the half-saturation constant (Ks), were determined for 18 species of batch-cultured freshwater algae grown without nitrogen limitation. Values of Ks ranged from 0.25 to 6.94 µM l–1 Chlorella pyrenoidosa Chick, and Navicula pelliculosa (Breb.) Hilse, respectively. Values of Vm ranged from 0.51 to 5.07 µM l–1 h–1 for Anabaena A7214 and Nitzschia W-32 O'Kelley, respectively. The mean positive values of Ks for Chlorophyta, Cyanophyta and Chrysophyta were 1.89, 3.67 and 4.07 µM l–1, respectively. The mean values of Vm for the same phyla were 1.61, 1.02 and 2.97 µM l–1 h–1 105 cells–1, respectively. The ranges of these kinetic parameters encompass values of kinetic parameters for marine and freshwater species in batch culture, for freshwater algae grown in N-limited chemostats and for natural populations of freshwater phytoplankton. Thus, in spite of variability between species, uptake parameters for both marine and freshwater algae are identical.  相似文献

Streptomyces sp. Y-110, isolated from soil, modified compactin to pravastatin, a therapeutic agent for hypercholesterolemia. In a batch culture, the highest production of pravastatin was 340 mg l–1 from 750 mg compactin l–1 in 24 h. By intermittent feeding of compactin into the culture medium, both the compactin concentration and its conversion increased to 2000 mg l–1 and 1000 mg pravastatin l–1, respectively, with the conversion rate of 10 mg l–1 h–1. Continuous feeding of compactin increased production of pravastatin to 15 mg l–1 h–1.  相似文献

A murine hybridoma cell line producing a monoclonal antibody against penicillin-G-amidase and a murine transfectoma cell line secreting a monovalent chimeric human/mouse Fab-antibody fragment were cultivated in three different media (serum-containing, low protein serum-free, and iron-rich protein-free) in flask cultures, stirred reactors and a fixed bed reactor. In static batch cultures in flasks both cell lines showed similar good growth in all three media.In suspension in a stirred reactor, the hybridoma cell line could be cultivated satisfactory only in serum-containing medium. In low protein serum-free medium, Pluronic F68 had to be added to protect the hybridoma cells against shear stress. But even with this supplement only batch, not chemostat mode was possible. In iron-rich protein-free medium the hybridoma cells grew also in continuous chemostat mode, but the stability of the culture was low. The transfectoma cell line did not grow in stirred reactors in any of the three media.Good results with both cell lines were obtained in fixed bed experiments, where the cells were immobilized in macroporous Siran®-carriers. The media, which were optimized in flask cultures, could be used without any further adaptation in the fixed bed reactor. Immobilization improved the stability and reliability of cultures of non-adherent animal cells in serum-free media tremendously compared to suspension cultures in stirred reactors. The volume-specific glucose uptake rate, an, indicator of the activity of the immobilized cells, was similar in all three media. Individuals of the fairy shrimp, Branchinecta longiantenna, were subjected to 5 concentrations (0.1 to 15 mg l–1) of Pb in diluted habitat water at 13 °C. Lead concentrations (mg kg–1 wet weight) in the animals were determined at 2-day intervals by digestion in nitric acid followed by atomic absorption analysis. The shrimp were also subjected to 0.1 mg l–1 media of Cd and Zn, separately.Uptake rates by the fairy shrimp for the three metal ions at 0.1 mg l–1 were: 0.111, 0.0885, and 0.0460 mg kg–1 day–1 for Zn, Pb, and Cd, respectively. After 2 days in 1.0 mg l–1 Cd or Zn, the animals expired; but they surviced for 8 days in a 10 mg l–1 Pb medium and for 2 days in 25 mg l–1 Pb. Lead uptake demonstrated a linear dependence on the Pb concentration in the media.Shrimp survived at much higher tissue accumulations of Pb compared to Zn and Cd. Estimated lethal doses were 20, 1.2–2.4, and 0.4–1.4 mg kg–1 wet weight for Pb, Zn, and Cd, respectively. Pb was found to be at much lower concentration than Cd or Zn in the natural pond water but between Cd and Zn levels in the sediment. S. mossambicus was exposed to toxic and sublethal concentrations of the fertilizer diammonium phosphate (0.2 to 1.0 g l–1). Mortality, food utilization and growth were studied. At a concentration of 0.6 g l–1 DAP, 100% mortality was observed within 96 h; no mortality occurred at 0.5 g l–1; LC50 was 0.55 g l–1. Rearing the fish in increasing sublethal concentrations of DAP, it was found that the feeding rate decreased from 25.4 mg g–1 fish–1 d–1 (fish reared in DAP-free water) to 10.1 mg g–1 d–1 at the highest sublethal concentration (0.5 g l–1). Growth rate was drastically reduced. At high sublethal concentrations of DAP, the fish lost reserve energy, in addition to the energy obtained from food intake for survival, as a result of increased swimming activity and opercular beats.  相似文献

Summary The kidneys of winter flounders transferred to hypotonic medium were investigated for glomerular and tubular handling of fluid and electrolytes and for the urinary excretion of proteins. Media were sea water (925 mosm·kg–1) and brackish water (70 mosm·kg–1).In sea water, the urine was hypertonic to the plasma in 7 fish of this study. Urine flow rate was correlated with the GFR. After adaptation to brackish water a delay of 1 to 3 days was observed until the kidneys switched from fluid retention to the excretion of large amounts of dilute urine. GFR and urine flow rate were increased from 0.61±0.08 to 1.58±0.29 ml·h–1·kg–1 and from 0.14±0.02 to 0.68±0.08 ml·h–1·kg–1, respectively . With increased filtered load the tubular reabsorption of fluid decreased from 74±2.4% to 45±11.2%. The excretion rates of sodium and potassium were increased due to decreased fractional sodium and potassium reabsorption. The urinary excretion of divalent cations, however, was reduced because the net tubular reabsorption of calcium was increased and the net secretion of magnesium was diminished.Both the urinary total protein concentration and the protein pattern showed no significant change, but the rate of protein excretion was increased from 0.21±0.04 to 0.60±0.05 mg·h–1·kg–1. The comparison of protein patterns obtained from urine and serum samples revealed that high molecular weight (HMW) proteins prevail in the serum whereas low molecular weight (LMW) proteins dominate in the urine. Co-metabolic degradation of trichloroethylene (TCE) by Pseudomonas putida F1 was investigated in a novel bioreactor with a fibrous bed. A pseudo-first-order rate constant for TCE degradation was 1.4 h–1 for 2.4 to 100 mg TCE l–1. Competitive inhibition of toluene on TCE removal could be prevented in this bioreactor. 90% TCE was removed over 4 h when 95 mg toluene l–1 was presented simultaneously.  相似文献

Summary Optimal growth conditions for Zymomonas mobilis have been established using continuous cultivation methods. Optimal substrate utilization efficiency occurs with 2.5 g l–1 yeast extract, 2.0 g l–1 ammonium sulfate and 6.0 g l–1 magnesium sulfate in the media. Catabolic activity is at its maximum with glucose uptake rates of 16–18 g l–1 h–1 and ethanol production rates of 8–9 g l–1 h–1, Qg values of 22–26 and Qp values between 11 and 13, which results in 40 g l–1 h–1 ethanol yields using a 100 g l–1 substrate feed. Any increase in these parameters goes on cost of substrate utilization efficiency. Summary The growth parameters ofPenicillium cyclopium have been evaluated in a continuous culture system for the production of fungal protein from whey. Dilution rates varied from 0.05 to 0.20 h–1 under constant conditions of temperature (28°C) and pH (3.5). The saturation coefficients in the Monod equation were 0.74 g l–1 for lactose and 0.14 mg l–1 for oxygen, respectively. For a wide range of dilution rates, the yield was 0.68 g g–1 biomass per lactose and the maintenance coefficient 0.005 g g–1 h–1 lactose per biomass, respectively. The maximum biomass productivity achieved was 2 g l–1 h–1 biomass at dilution rates of 0.16–0.17 h–1 with a lactose concentration of 20 g l–1 in the feed. This study examined the osmoregulatory status of the euryhaline elasmobranch Carcharhinus leucas acclimated to freshwater (FW) and seawater (SW). Juvenile C. leucas captured in FW (3 mOsm l–1 kg–1) were acclimated to SW (980–1,000 mOsm l–1 kg–1) over 16 days. A FW group was maintained in captivity over a similar time period. In FW, bull sharks were hyper-osmotic regulators, having a plasma osmolarity of 595 mOsm l–1 kg–1. In SW, bull sharks had significantly higher plasma osmolarities (940 mOsm l–1 kg–1) than FW-acclimated animals and were slightly hypo-osmotic to the environment. Plasma Na+, Cl, K+, Mg2+, Ca2+, urea and trimethylamine oxide (TMAO) concentrations were all significantly higher in bull sharks acclimated to SW, with urea and TMAO showing the greatest increase. Gill, rectal gland, kidney and intestinal tissue were taken from animals acclimated to FW and SW and analysed for maximal Na+/K+-ATPase activity. Na+/K+-ATPase activity in the gills and intestine was less than 1 mmol Pi mg–1 protein h–1 and there was no difference in activity between FW- and SW-acclimated animals. In contrast Na+/K+-ATPase activity in the rectal gland and kidney were significantly higher than gill and intestine and showed significant differences between the FW- and SW-acclimated groups. In FW and SW, rectal gland Na+/K+-ATPase activity was 5.6±0.8 and 9.2±0.6 mmol Pi mg–1 protein h–1, respectively. Na+/K+-ATPase activity in the kidney of FW and SW acclimated animals was 8.4±1.1 and 3.3±1.1 Pi mg–1 protein h–1, respectively. Thus juvenile bull sharks have the osmoregulatory plasticity to acclimate to SW; their preference for the upper reaches of rivers where salinity is low is therefore likely to be for predator avoidance and/or increased food abundance rather than because of a physiological constraint.  相似文献

We studied the relation between aquatic vegetation coverage, summer dissolved oxygen and density of sunfishes (Lepomis spp.) in 10 shallow bays of a eutrophic reservoir. The bays ranged 5.2–15.7 ha in area, 0.6–1.3 m mean depth and 6–91% vegetation area coverage. Over the 10 bays mean dissolved oxygen concentration ranged 5.8–9.0 mg l-1 in open water at least 20 m away from the vegetation, 0.8–8.1 mg l-1 at the vegetation-water edge and 0.5–7.7 mg l-1 in dense vegetation. Dissolved oxygen concentrations were inversely related to vegetation coverage. In open water, dissolved oxygen levels were consistently higher than 8 mg l-1 when vegetation coverage was <20% of the bay and decreased to about 6 mg l-1 at coverages near 80%. At the vegetation-water edge and within dense vegetation, dissolved oxygen levels dropped rapidly as vegetation coverage increased to 20% of the bay; when vegetation reached about 50% coverage, dissolved oxygen remained near 1.5 mg l-1at the vegetation-water edge, but oxygen dropped below 1 mg l-1in dense vegetation. Scarce vegetation harbored high Lepomis relative abundance (fish per m2 of vegetation) whereas extensive vegetation harbored low relative abundance, both contributing little to absolute abundance (total fish in all vegetation); however, intermediate coverage offered a combination of mid-level fish relative abundance that together with mid-level plant coverage translated into high absolute fish abundance. We suggest this response is related to hypoxia, and where aquatic vegetation is extensive, the effect of vegetation on hypoxia and water quality in general may influence fish populations in a way similar to that often attributed to reduced foraging efficiency and increased competitive interactions.  相似文献

Water reuse is a proposed strategy for utilizing or disposing of poor quality drainage water produced in the westside of central California. This 2-year field study evaluated the ability of two potential forage species to tolerate irrigation with water high in salinity, boron (B), and selenium (Se). The species used were: Sporobulus airoides var. salado (alkali sacaton) and Medicago sativa var. salado (alfalfa). After first year establishment with good quality water (<1 dS m–1), the two species were furrow-irrigated with drainage effluent that had an average composition of sulfate-dominated salinity ((electrical conductivity (EC) of 6.2 dS m–1)) B (5 mg l–1), and Se (0.245 mg l–1). Both crops were clipped monthly from June to October of each year. Total dry matter yields averaged between 11 and 12 mg ha–1 for both crops irrigated with effluent for two growing seasons. Plant concentrations of Se ranged from a low of 1.3 mg kg–1 in alkali sacaton to a high of 2.5 mg kg–1 in alfalfa, while B concentrations ranged from a low of 60 mg kg–1 in alkali sacaton to a high of 170 mg kg–1 in alfalfa. Chemical composition of the soil changed as follows from preplant to post-irrigation after two seasons with drainage effluent: EC from 2.78 to 6.5 dS m–1, extractable B from 1.9 to 5.6 mg l–1, and no change in extractable Se at 0.012 mg l–1 between 0 and 45 cm. Between 45 and 90 cm, EC values increased from 4.95 to 6.79 dS m–1, extractable B from 2.5 to 4.8 mg l–1, and no change in extractable Se at 0.016 mg l–1. Increased salinity and extractable B levels in the soil indicate that management of soil salinity and B will be necessary over time to sustain long term reuse with poor quality water.  相似文献

Three 5 l working volume fermenters were used to investigate the growth of the yeast Kluyveromyces fragilis in acid cheese whey under ambient temperature in order to assess the specific growth rate and yield, the lactose and oxygen uptake rates during the various phases of batch culture, the effect of increasing temperature on the various kinetic parameters, and the need for a cooling unit for single cell production batch systems. The initial dissolved oxygen in the medium was 5.5 mg l–1 and the pH was maintained at 4.5. The observed lag phase, specific growth rate and maximum cell number were 4 h, 0.2 h–1 and 8.4 × 108 cells ml–1, respectively. About 99% of the lactose in cheese whey was utilized within 20 h, 85% during the exponential growth phase. The specific lactose utilization rates by K. fragilis were 0.20 × 10–12, 1.457 × 10–12, 0.286 × 10–12 and 0.00 g lactose cell–1 h–1, for the lag, exponential, stationary and death phases, respectively. The dissolved oxygen concentration in the medium decreased as the cell number increased. The lowest oxygen concentration of 1.2 mg l–1 was observed during the stationary phase. The volumetric oxygen transfer coefficient was 0.41 h–1 and the specific oxygen uptake rates were 0.32 × 10–12, 2.14 × 10–12, 0.51 × 10–12 and 0.003 × 10–12 mg O2 cell–1 h–1, for the lag, exponential, stationary and death phases, respectively. The maximum temperature recorded for the medium was 33 °C, indicating that a cooling unit for batch production of single cell protein at ambient temperature is not needed for this type of bioreactor. The increase in medium temperature affected the cell growth and the lactose and oxygen uptake rates.  相似文献

The turnover times of glucose, averaged for 0–10 m in the upper waters of Lake Kinneret and measured by the addition of single or multiple concentrations of substrate, ranged from 23 to 188 hours and 1 to 87 hours respectively. Potential uptake rates (estimated as Vmax) ranged from 0.095 to 1.94 µg glucose l–1h–1, while measured uptake rates varied from 0.09 to 1.1 µg glucose l–1h–1. Concentrations of dissolved carbohydrates and glucose averaged 0.71 mg glucose equivalents l–1 and 39 µg glucose l–1 respectively. No evident relationships between glucose cycling and any fractions of dissolved organic matter, phytoplankton biomass or primary productivity were found. Turnover times were generally most rapid immediately after the decline of the spring Peridinium bloom. The respiration percentage of incorporated glucose ranged from 25% to 61% with highest values during the summer months. Respiration may be influenced by the nature of the indigenous bacterial population as well as by temperature. Glycerol at 10–20 g l–1 increased clavulanic acid production by Streptomyces clavuligerus in shake-flask culture. The biosynthesis of clavulanic acid continued for longer by feeding glycerol and production increased to 250 mg l–1 compared with 115 mg l–1 without feeding. In fermenter batch culture, degradation of clavulanic acid began after 72 h. With glycerol feeding in fed-batch culture, clavulanic acid production was not only increased further to about 280 mg l–1 but also remained stable up to 130 h.  相似文献

Summary Seepage caused salinity gradients in a 120 cm deep ditch adjacent to Lake Veere in the Dutch Delta area. The water was mixed by strong horizontal currents, forced by a pumping engine. The restoration of vertical chloride and oxygen gradients was studied.During summer when the level of Lake Veere was raised by 70 cm, a rapid seepage into the ditch was observed, ranging from 6 to 10 mm.h–1. This caused an anoxic saline layer. At the anaerobic-aerobic interface 1.4 to 2.4 mg O2.l–1.h–1 of the oxygen present in the upper water column was taken up. Sulfide oxidation required one quarter of this amount of oxygen.During winter the level of Lake Veere was lowered again. Normally seepage was absent then, and chloride entered the water column by diffusion; the mud chlorinity decreased from 15 to 3 Cl on its way to the ditch. Only after maximum discharge of polder water a moderate seepage of less than 4.4 mm.h–1 restored a chloride gradient; about 1.6 mg O2.l–1.h–1, produced by benthic diatoms, had to be supplied to the reducing materials in the seepage water in less than 12 hours.A saline seepage was simulated in the laboratory. These simulation tests showed that seepage below 1 mm.h–1, as well as benthic respiration only, had a small impact on the vertical oxygen profile in the water column.
