The aquatic leech, Theromyzon tessulatum, secretes a proteinaceous cocoon with extraordinary physical properties (e.g., proteolytic, thermal resiliency). The deduced amino acid sequence of a major protein (Tcp—Theromyzon cocoon protein) from the T. tessulatum cocoon wall has been used to model the endogenous structure of the Tcp protein. The Tcp protein sequence comprises six internal repeats, each containing 12 ordered Cys residues. Amino acid alignments suggest that the region Cys1→6 is homologous to antistasin, a leech anticoagulant, and Cys7→12 is homologous to an epidermal growth factor-like domain found in notch-class proteins, which play critical roles in development, signaling, and adhesion throughout the Animalia. Modeling of individual domains (i.e., antistasin and notch) positions multiple hydrophobic and charged residues on the surface. When the antistasin and notch domains were fused, hydrophobic pockets appeared that may facilitate a polymerization mechanism. Collectively, the predicted features of our Tcp model are consistent with the physical properties of the leech cocoon wall.   相似文献

One distinguishing feature of clitellate annelids is the presence of specialized segments comprising the clitellum, whose primary function is to secrete a cocoon. Using histological analyses, we have documented cell types (I-V) and cellular processes associated with cocoon secretion in the aquatic leech, Theromyzon tessulatum. Our data indicate that the bulk of the cocoon's biomass arises from precursor cells of a single type that hypertrophy and proliferate ∼1 week prior to egg laying, and then differentiate into either of two cell types (i.e., Type II or Type III) depending on their position within the clitellum. Type II cells are concentrated along the lateral edges and venter of the clitellum and secrete alcian blue-staining granules that form opercula (i.e., glue-like material that seals both cocoon ends), while Type III cells populate the dorsal midline and secrete azocarmine-staining granules that build the cocoon wall. Both cell types occupy spaces between deep muscle layers and extend long-neck tubules to the surface epithelium as they fill with granules a few days prior to egg laying. Other cell types appear to make minor contributions to the cocoon (e.g., Type I, Type IV) or have supporting or signaling roles (e.g., Type V). Our observations suggest that post-translational modification (i.e., glycosylation) of the same core protein(s) distinguishes the granules of Type II/III cells, and that the default state of the Type II/III precursor may be evolutionarily linked to secretory cells in basal polychaetes.  相似文献

Theromyzon tessulatum were reared individually from hatching until they attained sexual maturity. They were fed at different seasons and cultured at one of two temperature regimes. A series of weight measurements enabled growth curves to be constructed. Observations were also made on the digestive processes. In keeping with at least one other sanguinuous glossiphoniid, a restricted and constant number of blood meals precede reproduction and the hungry and sated stages show contrasting and distinctive behaviour. A notable feature is the great increase in weight after feeding resulting from the absorption of water and its incorporation into the body tissues. It probably enters through the body surface. The role of microorganisms within epithelial cells of certain crop caecae, was also considered.  相似文献

1. Nerve terminals associated with longitudinal muscle in the leech show FMRFamide-like immunoreactivity. 2. Structure-activity studies using FMRFamide analogs show that the C-terminal RFamide portion of the molecule is crucial for biological activity on leech longitudinal muscle. 3. The putative protease inhibitor FA (Phe-Ala) increases the peak tension produced by longitudinal muscle in response to superfused FMRFamide and the majority of its analogs, suggesting the presence of peripheral proteases capable of degrading RFamide peptides. 4. FMRFamide decreases the relaxation rate of neurally evoked contractions of longitudinal muscle. FA also decreases the relaxation rate of neurally evoked contractions. 5. Intact and isolated muscle cells respond to superfused FMRFamide with a conductance increase, that leads to depolarization and often with a delayed conductance decrease as the membrane potential is restored to resting levels. 6. The depolarizing response of isolated muscle cells to FMRFamide is dependent on external calcium.  相似文献

Summary Microvillar (receptive) and external (non-receptive) portions of the plasmalemma of photoreceptor cells of Hirudo were compared electron microscopically in thin sections and freeze-fracture replicas. A morphometric approximation showed that the surface area of the microvillar membrane is about 19 times larger than that of the external membrane. The microvillar membrane most probably undergoes extensive membrane turnover. In both segments of the membrane the particles associated with the P- and the E-fracture faces are randomly distributed except at some specific sites. The particles adhere predominantly to the P-faces. The particle densities on the fracture faces of the microvillar membrane differ from those of the external membrane. The P-face particles of the external membrane appear to be larger than those of the microvillar membrane. It is suggested that the P-face particles of the microvillar membrane represent sites where the photopigment is incorporated into the membrane. The distinguishing structural features correspond to the functional differences postulated for both portions of the plasma membrane.  相似文献

Summary After the staining of nervous tissue with phosphotungstic acid in absolute ethanol (E-PTA), a selective opacification occurs at certain specific sites, while other structures, especially the plasma and intra-cellular membranes, remain electron-lucent. Among those selectively stained sites, our studies have been focussed on: (1) The dense synaptic material consisting of several presynaptic clumps, termed projections, an intrasynaptic dense line and a subsynaptic web from which fine fibrillar wisps extend into the surrounding ground substance; (2) Neurofilaments and neurotubules, the surface of which is bristled by numerous side-arms; (3) A microfilamentous network intertwines the neurotubules, the neurofilaments and the mitochondria in the dendrites and axon, and is also connected to the E-PTA dense undercoating delineating the inner aspect of the plasma membrane and to the fine wisps emanating from the subsynaptic web. A three-dimensional microfilamentous latticework is thus formed in the nerve cell processes; (4) Dense cytoplasmic inclusions, termed nematosomes, which are usually located in the ground substance of the perikaryon among or in the vicinity of clusters of ribosomes. Tiny microfilaments emanate from the peripheral strands of these bodies. The presence of basic residues in the chains of structural proteins of which consist the subsynaptic web and the nematosome is plausible, since the specificity of the E-PTA staining procedure for the detection of basic residues has previously been put forth. The occurrence of a three-dimensional microfilamentous network in the nerve cell processes led us to hypothesize that it plays a role in translocation of materials. It may provide the motive force for the axoplasmic transport, for instance, with the neurotubules, as well as, plausibly, with the neurofilaments, serving as attachment sites and guideways.Supported by grant MA-3448 from The Medical Research Council of Canada.  相似文献

The present study focuses on the ultrastructural characteristics of both spermiogenesis and the spermatozoon in the order Trypanorhyncha. New ultrastructural data are presented for two species of the unexplored superfamily Tentacularioidea, Nybelinia queenslandensis, and Kotorella pronosoma. The present study also provides supplementary data on the superfamily Eutetrarhynchoidea, with the analysis of spermiogenesis and spermatozoon of two progrillotiids, Progrillotia dasyatidis and Pro. pastinacae, and new ultrastructural data concerning spermiogenesis in the eutetrarhynchids Dollfusiella spinulifera and Parachristianella trygonis. Spermiogenesis in trypanorhynchs follows the Bâ and Marchand's type I and the ultrastructural organisation of the mature spermatozoon corresponds to the Levron et al.’s type I. The most remarkable characters concerns the number of electron-dense plates constituting the intercentriolar body during spermiogenesis and in the variability of the arc-like row of thick cortical microtubules present in the anterior areas of the spermatozoon because of its variability according to the species.  相似文献

Summary The ultimobranchial glands of the chicken were examined by electron microscopy and immunocytochemistry using a calcitonin antiserum. Electron microscopy confirmed the presence of C-cells, containing numerous secretory granules storing calcitonin, in the luminal lining of cyst-like structures found in these glands. These cells were furnished with prominent microvillar projections at their luminal surface, and the cytoplasm of the apical region was filled with fibril material. Furthermore, the cells contained prominent junctional complexes and desmosomes at their apico-lateral surfaces. In these C-cells, secretory granules were concentrated near the lumen and some were attached to the apical cell membrane. The luminal content of the cysts had a colloid-like and flocculent appearance, and was frequently seen attached to the cytoplasmic projections or apical cell membrane of the C-cells. Since the cysts progressively increase in volume and number with age, it is suggested that they may partly play a role in the storage of excess or unneeded hormonal products.  相似文献

Summary An ultrastructural investigation of the gametes and their interaction during the early events of fertilization in molluscs has been performed. A gamete binding event involving large numbers of sperm has been identified and examined in detail. The surface of the oocyte is projected into numerous microvilli which extend through the vitelline envelope. Tufts of fibrillar material radiate from the tips of these microvilli, forming a layer external to the vitelline envelope. The acrosomal vesicle of the mature spermatozoon contains two major components, which function differently during fertilization. The vesicle is indented at its adnuclear surface, constituting a preformed acrosomal tubule. This tubule does not elongate during the acrosome reaction. Completion of the reaction results in the formation of an extracellular coat, derived from one component of the acrosomal vesicle, on the anterior surface of the sperm. Sperm-egg binding is accomplished by an association of the extracellular coat on the reacted sperm and the fibrous tufts on the tips of the microvilli of the oocyte. Evidence that gamete membrane fusion occurs by fusion of the acrosomal tubule and a microvillus is presented. The present investigations describe the fine structure of the excretory complex, rectal cells and the anal vesicle of the microfilaria of Breinlia sergenti. The structure of the excretory cell and rectal cells is found to be similar to nerve cells. Axonal (digitiform) processes in the excretory and anal vesicles are described and a possible sensory function is ascribed to these structures.  相似文献

Chemically induced carcinogenesis models in the rat are widely used for studying the biology of cancer and for developing and evaluating cancer prevention strategies. The azoxymethane(AOM)-induced rat colon tumor is a valuable tool for studying the interaction between tumor development and exogenous factors. Malignant conditions are characterized by enhanced levels of lipid peroxidative products, protein bound carbohydrates indicative of membrane damage and an ineffective antioxidant scavenging system. In the present study, AOM-induced rat showed lipid peroxidative damage, alterations in the membrane glycoprotein component and antioxidant defense system, along with ultrastructural changes like disruption in goblet cell and its membrane, swollen mitochondria with cristae dispersed, elongated endoplasmic reticulum and other features of neoplastic invasion. Berberine significantly attenuated the increases in lipid peroxidation, protein bound carbohydrates and enhanced the antioxidative status. The present study suggests that berberine prevents the appearance of malignant morphology and ultrastructural changes of AOM induced cancer by producing apoptosis like changes. Thus berberine inhibits neoplastic transformation by the induction of antioxidant defence system and ability to induce apoptotic like changes—thus elucidating its anti cancer role.  相似文献

Summary The ultrastructure of Cyclidium, including the cilia, kinetosomes, pellicle, microtubules and kinetodesmal fibers is similar to that recorded for other ciliates. Of special interest is the attachment of rod-shaped bacteria within the longitudinally directed shallow surface folds of the protozoan. Both the bacteria and the surface of Cyclidium seem to possess an outer coating of a sticky substance which upon contact holds the bacteria to the protozoan. The bacteria appear to be attached by only a relatively small area of their surfaces. A dense substance appears within the alveolus of the pellicle at the regions of the attachment of the bacteria. The association of the organisms is probably a temporary one, and it is unknown whether either organism is benefited or harmed by the association. The position of the mitochondria in Cyclidium is unusual in that they all lie flattened against the inner membrane of the pellicle, usually in a position directly opposite to that of the attachment of the bacteria to the surface, thus being separated from the bacteria by only the outer cell membrane and the pellicle. Summary Electron microscopic studies on developing follicles of Locusta migratoria show the vitelline membrane to be composed of two ultrastructurally distinguishable components: The vitelline membrane bodies (VMBs) and, in addition, fine granular material, cementing the VMBs together. VMBs form first in the oocyte-near zone within the oocyte-follicle cell space. Subsequently, the second vitelline membrane substance is secreted between the VMBs through apical protrusions of the follicle cells. The possible origin of the VMBs is discussed.Yolk uptake in Locusta seems to occur predominantly by pinocytosis. During oocyte development the oocyte membrane is enlarged by numerous microvilli and folds. In addition pinocytotic vesicles are pinched off. 在蛋白质组学研究中 ,双向聚丙烯酰胺凝胶电泳是现行蛋白质分离的最重要的方法之一。实验发展了一种提高固定pH梯度 (IPG)凝胶双向电泳的重复性、分辨率和通量的方法 :在一块SDS 聚丙烯酰胺凝胶上同时进行多块固定pH梯度(IPG)凝胶 (Multi stripsononeSDSgel,MSOG)电泳。用此方法比较了人肝癌细胞、不同生长状态的人肝癌细胞、3T3细胞的蛋白质以及同一个样品在不同大小的第二向凝胶系统 (大型和中型凝胶 )的双向电泳图谱。结果表明 ,同一样品在 13cmIPGStrip双向电泳可分离 2 0 0 0以上蛋白质点且图谱蛋白质点的匹配率可超过 95 %以上。同时又可以最大程度地降低凝胶背景对蛋白质点比较分析的干扰 ,从而提高了双向电泳分离蛋白质的分辨率和通量。这些优点都有助于差异蛋白质组学特别是细胞器差异蛋白质组学研究的自动化。  相似文献

Summary The ultrastructural changes taking place in the median eminence of Bufo bufo 3 hours to 4 months after transection of the brain at different levels, are described.5 types of neurons in the zona externa of the median eminence of normal toads are described. All types of neurons degenerate, and profound changes of the ultrastructure of the capillaries are observed after transection just behind, or immediately in front of the optic chiasma. A few neurons containing dense granules with a diameter of about 1,000–1,300 Å remain intact, however. The degeneration following denervation in front of the optic chiasma was considerably delayed compared to degeneration after denervation behind the optic chiasma.After transection more rostral to the optic chiasma, no significant degeneration of the median eminence was observed.The results are discussed with regard to degenerative dynamics and origin of the different nerve types. It is concluded, that all types of neurons terminating in the median eminence, originate at a level between the caudal and rostral parts of the preoptic nucleus, some fibres, however, containing dense, 1,000–1,300 Å granules, originate caudally to the optic chiasma, in the posterior hypothalamus.Part of this study was presented at the Vth International Symposium on Neurosecretion, Kiel, Germany, August 1969.  相似文献

Aims: Validation of stereology and three‐dimensional reconstruction for monitoring the probiotic effect of Aeromonas hydrophila on the gut development of germ‐free Artemia franciscana nauplii. Methods and Results: Germ‐free Artemia nauplii were cultured using Baker’s yeast and dead Aer. hydrophila. Live Aer. hydrophila were added on the first day to the treatment group. The gut length and volume were monitored on days two and four using stereology and three‐dimensional reconstruction. Both methods showed comparable results. Stereology was least labour intensive to estimate volumes, while three‐dimensional reconstructions rendered architectural and topographical data of the gut. Moreover, a positive effect of probiotic bacterium, Aer. hydrophila is likely. Conclusion: Slight increment in the growth of the digestive tract of A. franciscana nauplii exerted by probiotic bacteria could be detected using stereology and three‐dimensional reconstruction. Significance and Impact of the Study: The gnotobiotic Artemia rearing system is unique to investigate the effects of micro‐organisms on the development of nauplii. However, in the base of this model system, only survival counts and length measurements exist as monitoring tools. Therefore, additional tools such as stereology and three‐dimensional reconstruction are prerequisite to obtain more powerful analysis.  相似文献

In this study, we present a comparative assessment of the effects of two polyether ionophorous antibiotics (monensin and salinomycin) on the concentrations of lead (Pb), cooper (Cu), zinc (Zn) and iron (Fe) in the kidneys, spleen, liver and brain of Pb-intoxicated animals. Our data demonstrated that the intoxication of ICR male mice with Pb salt resulted in a significant accumulation of Pb in all studied organs of the mice compared to the untreated control animals. The biodistribution of the toxic metal was in the order kidneys > spleen > liver > brain. The treatment of the Pb-intoxicated animals with tetraethylammonium salts of monensic and salinomycinic acids significantly decreased the concentration of the toxic metal ion compared to the toxic control. The effect varied in the interval 38% (for kidneys) to 52% (for brain) compared to the toxic control group (Pb). The tetraethylammonium salt of salinomycinic acid was more effective in reducing the Pb concentration in the brain of the Pb-treated mice compared to monensin. Pb-intoxication did not affect significantly the Zn endogenous concentration compared to the normal values. The treatment of ICR male mice with Pb-salt decreased the Cu concentration in the spleen and increased the Cu concentration in the liver compared to the untreated control animals. The detoxification of the Pb-intoxicated mice with tetraethylammonium salts of salinomycinic and monensic acids restored the Cu concentration in the spleen, but did not affect the Cu levels in the liver. The Pb-intoxication of the ICR mice resulted in a significant decrease of the Fe-concentration in the spleen and liver compared to the untreated control animals. The administration of the tetraethylammonium salts of salinomycinic and monensic acids to the Pb-treated animals restored the levels of Fe in both organs.  相似文献

A peroxisomal C-tail-anchored type-II membrane protein, Pex26p, recruits AAA ATPase Pex1p-Pex6p complexes to peroxisomes. We herein attempted to gain mechanistic insight into Pex26p function. Pex26pΔ33-40 truncated in amino-acid residues at 33-40 abolishes the recruiting of Pex1p-Pex6p complex to peroxisomes and fails to complement the impaired phenotype of pex26 CHO cell mutant ZP167, thereby suggesting that peroxisomal localization of Pex1p and Pex6p is indispensable for the transport of matrix proteins. In in vitro transport assay using semipermeabilized CHO cells, Pex1p is targeted to peroxisomes in a manner dependent on ATP hydrolysis, while Pex6p targeting requires ATP but not its hydrolysis. This finding is confirmed by the assay using Walker-motif mutants. Transport of Pex1p and Pex6p is temperature-dependent. In vitro binding assays with glutathione-S-transferase-fused Pex26p, Pex1p and Pex6p bind to Pex26p in a manner dependent on ATP binding but not ATP hydrolysis. These results suggest that ATP hydrolysis is required for stable localization of Pex1p to peroxisomes, but not for binding to Pex26p. Moreover, Pex1p and Pex6p are altered to a more compact conformation upon binding to ATP, as verified by limited proteolysis. Taken together, Pex1p and Pex6p are most likely regulated in their peroxisomal localization onto Pex26p via conformational changes by the ATPase cycle.  相似文献
