A bacterial strain of Acinetobacter sp., which was capable of enzymatic production of pyruvate from lactate, was cultured in a 5-l reactor with a basal salt medium. After 14 h of fed-batch fermentation, 9.56 g l–1 cell concentration in the broth was obtained with 20 g l–1 (178 mM) sodium lactate and 4 g l–1 NH4Cl in the medium; and the biotransformation ability was 2.51 units ml–1. The cells were harvested from one reactor and then used for pyruvate production from lactate in the same reactor. l-lactate at a concentration about 527 mM was almost stoichiometrically converted to pyruvate in 28 h. After a total 42 h of cell culture and biotransformation, the transformative yield was about 0.72 g g–1 pyruvate from lactate and the rate of pyruvate production was calculated as 1.33 g l–1 h–1 during the process. The results suggested this simple enzymatic production of pyruvate from lactate should be a promising process and may bring a yield higher than that by microbial fermentation. By this process, the recovery of pyruvate from such a simple reaction liquid is relatively easy and inexpensive to perform.  相似文献

Cell-free extract (crude extract) of Agrobacterium tumefaciens grown on d-glucuronate or d-glucarate converts d-glucarate and galactarate to a mixture of 2-keto-3-deoxy- and 4-deoxy-5-keto-d-glucarate. These compounds are then converted by partially purified crude extract to an intermediate tentatively identified as 2,5-diketoadipate. The same enzyme preparation further decarboxylates this intermediate to alpha-ketoglutarate semialdehyde, which is subsequently oxidized in a nicotinamide adenine dinucleotide-dependent reaction to alpha-ketoglutaric acid. Since A. tumefaciens converts d-glucuronic acid to d-glucarate, a pathway from d-glucuronate to alpha-ketoglutarate in A. tumefaciens was determined.  相似文献

The oxidation of D-(+)-glucose to D-glucaric acid using the TEMPO-like nitroxide oxidation catalyst, 4-acetamido-2,2,6,6-tetramethyl-1-piperidinyloxy (4-acetamido-TEMPO) was carried out using several oxidizing agents and co-catalyst. A new spectrophotometric method for determining low hydrogen peroxide concentrations by using horseradish peroxidase in the presence of NADH at pH 7.5 has been described. Both total NADH consumption and initial reaction rate may be used for the determination. Using the NADH consumption, a linear response with respect to hydrogen peroxide was observed in the concentration range 7 x 10(-8)-2.5 x 10(-6) M. Due to the presence of superoxide dismutase, hydrogen peroxide is partly regenerated and an amplification of the signal results, which explains the sensitivity.  相似文献

Treatment of partially hepatectomized male rats with urethan 6 hr after operation resulted in 50–55% inhibition of the incorporation of orotic acid-5-3H into nuclear ribosomal RNA and heterogeneous RNA 18 hr later. Neither partially hepatectomized female rats similarly treated with urethan nor operated male animals treated with an equitoxic dose of butyl carbamate presented evidence of an impairment of nuclear RNA synthesis.  相似文献
