Abstract: Long-Evans rat pups were exposed to either inorganic lead (400 mg Pb as lead acetate/kg body weight/day) or triethyltin sulfate (1.0 mg/kg body weight/day), by gastric intubation, from 2 days through 29 days of age. The rats were then weaned and placed on standard lab chow ad libitum. At 30 days of age, leadtreated rats exhibited statistically significant decreases in body and brain weights (22% and 17%, respectively), and the concentration of forebrain myelin was significantly reduced, by 21% relative to the 4.9 mg myelin protein/g brain in control rats. Although these animals recovered from the body weight deficits after several months, the deficits in brain weight and myelin concentration were still present at 120 days of age. This suggests that the lead-induced myelin deficits were permanent. Lead levels in brain, which were maximal at 30 days of age when the treatment was terminated, decreased more slowly than in other organs and were still 30% of maximal levels at 120 days of age. Triethyltin-treated animals also had significantly decreased body and brain weights (20% and 11%, respectively) at 30 days of age, and an even more severe reduction in forebrain myelin concentration (33%). These animals also regained a normal body weight by 120 days of age, but again the deficits in brain weight and myelin concentration persisted. Tin levels in brain and other organs had decreased to control levels by 60 days of age. Animals malnourished by maternal deprivation to match the body weights of the treated animals had myelin deficits that were less severe than those in the treated animals at 30 days of age (approximately 11% less than controls); however, these myelin deficits also persisted throughout the subsequent 90-day recovery period examined. The apparent lack of recovery from CNS myelin deficits produced by neonatal exposure to different heavy metals or to malnutrition reemphasizes the vulnerability of the developing nervous system to a wide range of metabolic insults.  相似文献

Effect of Triethyl Tin on Myelination in the Developing Rat   总被引:3,自引:2,他引:1  
Myelinogenesis in developing rats was studied following chronic dosing with triethyl tin (TET), at a level of 1.0 mg TET/kg body wt/day. Experiments included starved controls with body weights depressed by 17 to 40% to equal those of the TET-treated groups. Rats at ages of 16, 21, and 30 days showed decreases relative to well-nourished controls in body weight, forebrain weight, myelin yield, cerebroside level, and specific activity of brain 2',3'-cyclic nucleotide-3'-phosphohydrolase when dosed with TET. At 30 days, myelin and cerebroside yields were reduced by approximately 55%, while CNP activity was reduced by less than 20%. No differences in the forebrain myelin protein composition between control, starved, and TET animals were noted. The rate of myelin protein synthesis relative to brain total protein (assayed by incorporation of intracranially injected [3H]glycine into brain homogenate and myelin proteins) was decreased in the TET rats in proportion to the decreased yield of myelin, but no particular myelin protein was preferentially affected. Matching starved controls exhibited similar body weight decreases, less pronounced forebrain weight decreases, and little or no decrease in myelin concentration. There was a relative increase in the myelin protein synthesis rate in the starved rats, indicating preferential utilization of limited protein precursors for myelin protein synthesis. Spinal cord myelin was also decreased in the TET rats, but less severely than in the forebrain. At all ages optic, but not sciatic, nerves showed decreases in myelin concentration with TET treatment. We conclude that TET inhibits forebrain growth and CNS myelination more severely than can be accounted for by a general metabolic insult.  相似文献

Effects of inbreeding on the development and reproduction of the predatory pentatomid Podisus maculiventris were assessed throughout 30 generations following introduction in the laboratory. Developmental and reproductive fitness of two mildly inbred lines did not differ dramatically from that of a reference population. In most generations, egg weight, egg hatch and developmental rate of nymphs were greater in the reference strain than in both inbred strains, but fecundity and longevity did not differ among strains. In the 30th generation, fecundity of all strains had dropped to about half of that observed in the 15th generation, which was presumably related to non-genetic factors. There were consistent differences among strains for adult body weight and size, indicating heritable variation. A positive relationship between weight at day 14 and humeral width of P. maculiventris females was found, but neither of these parameters was associated with total fecundity. In the 30th generation, predators from both inbred lines had generally similar predation capacities to those from the reference population. No significant relationship could be detected between body weights and predation rates of either nymphs or adults. The use of inbreeding to minimize selective adaptation during rearing or to select for favourable traits in P. maculiventris is discussed.  相似文献

This is the first phenotypic analysis of 75 new recombinant inbred (RI) strains derived from ILS and ISS progenitors. We analyzed body weight in two independent cohorts of female mice at various ages and in males at 60 days. Body weight is a complex trait which has been mapped in numerous crosses in rodents. The LXS RI strains displayed a large range of weights, transgressing those of the inbred progenitors, supporting the utility of this large panel for mapping traits not selected in the progenitors. Numerous QTLs for body weight mapped in single- and multilocus scans. We assessed replication between these and previously reported QTLs based on overlapping confidence intervals of published QTLs for body weight at 60 days and used meta-analyses to determine combined p values for three QTL regions located on Chromosomes 4, 5, and 11. Strain distribution patterns of microsatellite marker genotypes, weight, and other phenotypes are available on WebQTL () and allow genetic mapping of any heritable quantitative phenotype measured in these strains. We report one such analysis, correlating brain and body weights. Large reference panels of RI strains, such as the LXS, are invaluable for identifying genetic correlations, GXE (Gene X Environment) interactions, and replicating previously identified QTLs. The relationship of brain and body weights can be expressed in log-log regression: log (brain weight) = log (A) + B log (body weight). To investigate further the weights' similarity, gestation length and brain and body weights were determined from the literature for 46 primate genera. The results of allometric and path regression analyses suggest that the relationship between brain and body weights may not be mainly pleiotropic in the order Primates. The correlation between brain and body weights appears to be due to epigenetic factors in hyperplastic growth related to time constraint by gestation length and to energy utilization limitations imposed by metabolic rate.  相似文献

The psychomotor maturation rate was studied in a heterogeneous stock of laboratory mice. Mean ages of several innate reflexes formation were compared for two groups of animals which represented extreme variants of brain weight phenotypes when adult. Mice with measured brain weights, by least 0.67 standard deviation higher (H) or lower (L) than the average for the whole population, reveal substantially different psychomotor development, the H-group appearing to be more advanced. This observation is at variance with those obtained using the lines of laboratory mice selectively bred for high and low brain weight. The apparent inconsistency of the two experiments may reflect the relative dominance of environmental (heterogeneous stock) or genetic (selected lines) factors, as well as occasional fixing of certain alleles in process of selection.  相似文献

目的建立野生来源TW(Tianjin Wild,TW)近交系小鼠的主要生理、血常规等指标。方法分别选用F28和F29代TW近交系成年雄性小鼠25只,雌性小鼠31只,检测动物的主要生理、血常规指标。结果TW小鼠体重雌雄差异无显著性,脏器指标中雌雄性比较仅肾脏重量、肾脏系数差异有显著性(P〈0.05);血常规检测指标中,雌雄小鼠在WBC、MCV、MCH、PLT、MONO等5项指标的均值比较差异具有显著性,其他各项均无差别。与ICR、KM、NIH及BALB/c等通用实验小鼠指标比较,TW小鼠的脾脏、脾脏系数和血小板均高于或多倍高于这些通用小鼠品系。结论TW小鼠在一些主要生理指标上与通用实验小鼠品系不同,呈现自身特点。  相似文献

The effects of ethanol injections on the F2 offspring of the cross between large-brain (LB) and small-brain (SB) mouse strains selected for high and low relative brain weights, respectively, have been studied. The parental strains have significantly differed in brain weight for many generations. The effects of ethanol (2.4 g/kg) have been compared in four subpopulations of mice that differ pairwise in brain weight. One pair of subpopulations has been isolated from the hybrid group and the other, from generation 22 of selection of the parental strains. The results of ANOVA have demonstrated that brain weight is related to the response to ethanol injections. The parameters of stereotyped behavior, which increased in after ethanol injections and reflected the decrease in exploratory activity) were different in mice with high and low relative brain weights. The pattern of behavioral changes after ethanol injections is the second (after increased learning ability) behavioral trait found to be correlated with brain weight.  相似文献

Fifty-three adult female and 51 adult male white-tailed rats (Mystromys albicaudatus) were killed with ether and weighed; the spleen, kidneys, liver, heart, lung, pancreas, brain and gonads were dissected free of adhering tissue and weighted. The mean absolute organ weight and organ:body weight ratios by sex and organ were calculated and compared. The male rats were significantly (p less than or equal to 0.05) heavier. The mean weight of the males was 110.0 +/- 23.8 g versus 82.9 +/- 16.1 g for the females. The absolute weights of the heart, liver and kidneys were significantly (p less than or equal to 0.05) greater for the males. The organ:body weight ratios, except for heart and brain (excluding ovary and testicle), were unaffected by sex. The heart to body weight ratio and the brain to body weight ratio were significantly (p less than or equal to 0.05) larger in female rats.  相似文献

The relationships among microencephaly, peak blood alcohol concentration (BAC), and dose of alcohol were examined in a rat model of third-trimester fetal alcohol effects. Ethyl alcohol was administered to neonatal rats from postnatal day 4 to day 10 during the brain growth spurt via an artificial rearing technique. Groups of rats received one of nine doses of alcohol (0.0, 2.5, 3.3, 4.0, 4.5, 5.3, 6.6, 7.5, or 8.5 g/kg body weight) administered in 8 hours each day. BACs were determined on postnatal days 6 and 7 at times corresponding to peak and trough BACs, respectively. On postnatal day 10, brains were removed, and total brain weights, cerebellar weights and brainstem weights were measured. Pups receiving 4.0 g/kg/day or less had mean peak BACs below 150 mg/dl and did not exhibit significant microencephaly when compared with controls. Higher dosages further increased the peak BAC and produced significant microencephaly. While a dose of 4.5 g/kg/day was sufficient to decrease significantly both total brain weight and cerebellar weight, a minimum dose of 6.6 g/kg/day was required for significant restriction of brainstem weight. The dose of 7.5 g/kg/day yielded a mean peak BAC of 420 mg/dl and reduced total brain weight, cerebellar weight, and brainstem weight by 33%, 52%, and 22%, respectively, relative to controls. Exposure to 8.5 g/kg/day was uniformly lethal. Peak BAC and total brain weight were highly correlated (r = -.916). As peak BAC increased, total brain weight decreased linearly. Comparisons with previous studies indicate that condensing the daily dose of alcohol effectively reduced the threshold doses for microencephaly and lethality.  相似文献

The outer membrane proteins of Vibrio vulnificus including isolates from humans, seawater and an asari clam were examined by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. A major outer membrane protein with an apparent molecular weight of 48,000 (48K protein) was common to all the strains grown in 3% NaCl-nutrient broth; however this 48K protein was not produced in any of the strains grown in chemically defined medium. Other major outer membrane proteins with molecular weights ranging from 33,000 to 40,000 varied in number, relative amount and molecular weight depending on the strain. One to three new outer membrane proteins with molecular weights ranging from 74,000 to 85,000 were produced in the cells grown in iron-deficient medium. The 48K protein and one or two major proteins with molecular weights ranging from 35,000 to 37,000 in the cells grown in 3% NaCl-nutrient broth were not solubilized by 2% SDS at 60 C for 30 min and were resistant to trypsin, indicating that they are porins. On the other hand, in cells grown in chemically defined medium, one or two major outer membrane proteins with molecular weights ranging from 33,000 to 40,000 might be porins.  相似文献

The internal variability of several varieties and strains of French bean is investigated, confining attention to the single character, seed weight. Out of eleven strains examined only two appeared to be homogeneous for seed weight, whilst two were obviously heterogeneous even by superficial examination of the seed.
A comparison of the distribution of seed weights in five strains of Canadian Wonder showed that each strain contains at least two or three lines out of the minimal estimate of four shared between them.
The natural out-crossing of fourteen varieties, including those analysed for seed weight, is compared. There are indications that varieties vary in their out-crossing propensities. No indication was found of the causes of the high frequencies of out-crossing occasionally encountered.
The possible sources of intra-varietal variation are briefly surveyed.  相似文献

The neuroprotective potential of mGluR1 and mGluR5 antagonists (group I), EMQMCM and MTEP, respectively was studied using the 3 min forebrain ischemia model in Mongolian gerbils and the hypoxia-ischemia model in 7-day-old rats. Hypoxia-ischemia was induced by unilateral carotid occlusion followed by 75 min exposure to hypoxia (7.3% O(2) in N(2)), forebrain ischemia in gerbils was evoked by bilateral common carotid artery occlusion. The postischemic rectal body temperature in rat pups or brain temperature of gerbils was measured. The drugs were administered i.p. three times every 2 h after the insult, each time in equal doses of 1.25, 2.5 or 5.0 mg/kg. After 2 weeks brain damage was evaluated as weight decrease of the ipsilateral hemisphere in the rat pups or damage to CA1 pyramids in the gerbil hippocampus. The results demonstrated a dose dependent neuroprotection in both ischemic models by EMQMCM, while MTEP was neuroprotective only in the gerbil model of forebrain ischemia. EMQMCM reduced postischemic hyperthermia in gerbils. Thus, the antagonists of mGluR1 and mGluR5 show differential neuroprotective ability in two models of brain ischemia. Postischemic hypothermia may be partially involved in the mechanism of neuroprotection following EMQMCM in gerbils.  相似文献

The single housing of laboratory rats may be recommended in some situations such as hypothesis-driven or test-specific studies, during electroencephalogram recording of phases of sleep and after surgical procedures. However, as single housing of laboratory rats has been shown to be stressful, modification of the housing environment is needed to improve the welfare of these animals. This experiment was carried out to investigate the long-term effects of environmental enrichment on some behavioural, physiological, pathological and psychological measures of welfare. With two batches of animals, 24 rats were housed singly in either enriched cages (EC) (n = 12 cages) or unenriched cages (UC) (n = 12 cages). Behaviour was sampled every week and so was body weight and weight gain over a six-week observation period. Behaviours of the rats in the elevated plus-maze were recorded on the seventh week, whereas organ weights were recorded postmortem. The results revealed that long-term single housing of rats in super-enriched cages increased levels of indicators of good welfare including sleep, exploration, movement and feeding behaviour, body weights, weight gains and the relative weights of the thymus gland and spleen, and decreased levels of indicators of poor welfare such as stationary behaviour and the relative weight of adrenal glands. Thus, enrichment of conventional cages of newly weaned singly-housed laboratory rats with multiple physical structures appeared to improve their ability to control the environment and to promote their species-specific behaviour; changes that can ultimately result in good welfare.  相似文献

Abstract We examined the penicillin-binding proteins (PBPs) of certain field strains of Streptococcus suis , as well as those from laboratory variants having different degrees of resistance to penicillin. Results indicated that (i) S. suis possesses three distinct groups of PBPs, arbitrarily named here PBP 1, PBP 2, and PBP 3, with approximate molecular weights of 97, 82, and 45 kDa respectively; (ii) PBP profiles of field strains of S. suis having different MICs (≤ 0.03 to 16.0 μg/ml) were not uniform (PBP 2 being difficult to detect in strains whose MICs exceeded 0.10 μg/ml, and PBP 3 which exhibited shifts in molecular weight of approximately 5 kDa); (iii) laboratory variant PBPs 1 and 2 showed decreased affinity for penicillin as compared to the parent strain in antibiotic competition experiments, even though the PBP profiles of both were similar. We suggest that PBP modifications (altered molecular weight and/or decreased affinity for penicillin) are involved in the mechanism of resistance to penicillin by S. suis .  相似文献

The relation between body weight and sexual maturation was examined in a hibernator, Belding's ground squirrel, by manipulating the availability of food to weaned juvenile males. Following body weight manipulation in the summer, testicular growth, serum testosterone, and spermatogenesis were monitored during the subsequent year, which included 7 mo when males were in the coldroom (ca. 8 degrees C), followed by 5 mo in the laboratory (ca. 20 degrees C). Juveniles (less than 1 yr old) maintained on a restricted diet entered the coldroom at normal body weights for their age class in nature and had immature gonads throughout the year, which is characteristic of this group in the field. In contrast, juveniles given abundant food during the summer entered the coldroom at body weights typical for free-living yearlings and exhibited mature gonads shortly after males were removed from the cold (high relative testis weights, high serum testosterone levels, and all stages of spermatogenesis). The high level of gonadal activity in overfed males was confined to a period of a few weeks in the spring, which coincided with the time when mating occurs in nature. The ability of male Belding's ground squirrels to accumulate body weight prior to hibernation seems important to sexual maturation in this seasonally breeding rodent.  相似文献

Data from 38 laboratories using 5 strains of Salmonella typhimurium (TA98, TA100, TA1535, TA1537, and TA1538) were analyzed to determine sources and magnitudes of test data variability. Each laboratory tested the mutagenicity of 4-nitroquinoline-N-oxide by the same protocol, using both its in-house cultures and a set of reference cultures provided to all laboratories. It was found that neither plate-to-plate nor day-to-day variability within a laboratory differed substantially between the in-house and reference cultures for any strain; this indicated no difference in the laboratories' handling of the two cultures. Not surprisingly, on average, plate-to-plate variability was substantially smaller than day-to-day variability within a laboratory, which, in turn, was substantially smaller than inter-laboratory variability. The solvent DMSO was found to have a small (6-7%) but statistically significant depressive effect on the spontaneous mutant frequency for the two plasmid-containing strains, TA98 and TA100, but not for the other three strains. When the mean value and variance of all laboratories for the in-house culture were compared with the corresponding reference culture values for each dose and strain, no major differences were seen. Any increase in mean or variance in the distribution of laboratory means in one of the two cultures could be ascribed largely to a small number of laboratories. Laboratories that reported 'high' or 'low' levels of spontaneous or induced revertants per plate tended to deviate in the same direction for most strains and for both in-house and reference cultures. If 'genetic drift' contributed to the inter-laboratory variability in this collaborative study, it was a minor component that went undetected in our analyses.  相似文献

This paper presents results from a number of studies where different BCG products and BCG strains were compared and evaluated by the immediate effect after vaccination. It was shown that BCG strains differ in terms of allergenic potency. However, two products prepared from the same strain in different laboratories may differ more than two products prepared from different strains in one laboratory. Thus for products prepared from different strains in different laboratories the allergenic potency may differ as a result of differences between strains, production methods and concentrations. Any difference in allergenic potency observed between products from different laboratories, therefore, cannot give conclusive information on a single characteristic.  相似文献
