Summary We developed efficient genetic transformation protocols for two species of duckweed, Lemna gibba (G3) and Lemna minor (8627 and 8744), using Agrobacterium-mediated gene transfer. Partially differentiated nodules were co-cultivated with Agrobacterium tumefaciens harboring a binary vector containing β-glucuronidase and nptII expression cassettes. Transformed cells were selected and allowed to grow into nodules in the presence of kanamycin. Transgenic duckweed fronds were regenerated from selected nodules. We demonstrated that transgenic duckweed could be regenerated within 3 mo. after Agrobacterium-mediated transformation of nodules. Furthermore, we developed a method for transforming L. minor 8627 in 6 wk. These transformation protocols will facilitate genetic engineering of duckweed, ideal plants for bioremediation and large-scale industrial production of biomass and recombinant proteins.  相似文献

In this study, laboratory tests were performed in order to examine growth characteristics of floating aquatic macrophytes (Lemna gibba and Lemna minor) in the presence of wastewater with circulation. The results showed that circulation of the waste water enhanced the kinetics of the process, as compared to the control systems. However, prolonged application of high circulation level had a different effect. In the presence of circulation with aquatic plants, there was additional 85.3-88.2% for BODs and 59.6-66.8% for COD decreases in the water quality indicators. In this study, the effectiveness of L. gibba and L. minor with circulation addition for the removal of four heavy metals (Pb, Ni, Mn, and Cu) from waste water was also investigated. Results from analysis confirmed the accumulation of different metals within the plant and a corresponding decrease of metals in the waste water. At the end of the study of circulation, L. gibba provided the metal removal for Cu, Pb, Ni, and Mn in the waste water as the ratio of 57%, 60%, 60%, and 62%, respectively. In this context, the best results were obtained when the action of L. gibba and L. minor plants, was combined with that of circulation. It is shown that in the presence of L. gibba and L. minor plants that are supplemented with circulation, the national standards of biochemical oxygen demand (BOD5) 27-33 mgL(-1) and chemical oxygen demand (COD) 62-78 mgL(-1) for L. minor and L. gibba, respectively, were reached after treatment. The new results can be used for design calculations regarding expected removal of pollutants by aquatic floating plants.  相似文献

Abstract

Aquatic plants have been identified as potentially useful for accumulating and bioconcentrating heavy metals. This study was developed to test the hypothesis that nutrient enrichment enhances the metal tolerance of floating macrophytes. Relative growth rates (RGR), photosynthetic pigments (chlorophyll a, b and carotenoid), malondialdehyde (MDA) content, and electrical conductivity (EC) were measured in Lemna gibba exposed to different cadmium and copper concentrations in laboratory conditions. Relative growth rates were negatively correlated with metal exposure, but nutrient addition suppressed this effect. Photosynthetic pigment levels were negatively correlated with metal exposures, and nutrient addition attenuated chlorophyll decrease in response to metal exposures. MDA content and EC also showed sharp increases at higher concentrations, indicating oxidative stress. This study indicates that nutrient enrichment increases the tolerance of Lemna gibba to metals, and that Lemna gibba is a suitable candidate for the phytoremediation of low-level copper and cadmium pollution.  相似文献

L-Pipecolic acid was found to be effective in inducing floweringof Lemna paucicostata 151, 381, 441 and 6746, and of Lemna gibbaG3. When the plants were grown on half-strength Hutner's medium,L-pipecolic acid caused profuse flowering of L. paucicostata151 maintained under 9 and 10 h of light daily. In L. paucicostata441 and 6746, L-pipecolic acid had a strong flower-promotingeffect under a near critical photoperiod. In L. paucicostata381, by contrast, L-pipecolic acid had only a very small effecton flowering. In L. gibba G3 substantial promotion of floweringwas observed under continuous light. When one-twentieth-strengthHutner's medium was used as the basic medium, L-pipecolic acidstimulated flowering in all strains of Lemna examined, evenunder continuous light. When L. paucicostata 151 was grown on one-tenth-strength M mediumor one-twentieth-strength Hutner's medium, the flower-inducingactivity of L-pipecolic acid was greatly enhanced by cytokininunder continuous light. However, when this strain was grownwith 9 h of illumination daily, this synergistic effect of cytokininwas only slight. A short-term (even 1-h) treatment with L-pipecolicacid resulted in flowering, suggesting that L-pipecolic acidis involved in the induction of flowering, rather than its evocation.D-Pipecolic acid also had flower-inducing activity, but itsactivity was 50 times lower than that of the L-isomer. Abstract

Gel filtration chromatography (with Sephadex G25, G50 and G100) was used to separate the different forms of thallium (Tl) and cadmium (Cd) in the cytosol fraction of Lemna minor and to examine the influence of the duration of metal exposure on the speciation of the two elements. A major proportion of Cd in the soluble phase was found to be bound to three groups of proteinaceous and polypeptide fractions; two of these, the high molecular weight protein fraction (Mr > 150,000) and the low polypeptide sized moieties of about Mr 1,500 or less were constitutive entities, whereas the intermediate sized fraction (Mr 7,000 - 8,000) could only be detected in plants previously exposed to Cd. After 12 days of exposure to Cd this fraction accounted for the greatest part of the bound Cd in Lemna tissues. Extending the period of exposure from 18 hours to 12 days resulted in a shift in the distribution of Cd between the low and intermediate fractions. Evidence for Tl-protein binding was limited and confined to the high molecular weight fraction, and most of the Tl in the soluble phase was present in a form closely allied to the free ion. The contrasting behaviour between the two elements has been interpreted in terms of the differences in their physicochemical properties.  相似文献

Brief exposure to light promotes frond multiplication in Lemnagibba G3 in darkness. Extent of promotion changes periodicallywith the time of the light-break. Response curves are interpretedin terms of a superposition of two modes of growth responseto light-break, which are, respectively, under the control ofdifferent physiological timing devices; circadian oscillationand the hourglass-type clock. Circadian oscillation, which consistsof a half-cycle of increasing photophily followed by anotherhalf-cycle of declining photophily, starts at a light-on signaland continues for a few days with rapid damping. The 24-hr periodof oscillation is the same at temperatures ranging from 16 to26°C. The hourglass is released by a light-off signal to‘accumulate sand’ or to increase photophily in asigmoidal way with time and is temperature-sensitive; the tempoof‘sand accumulation’ being quicker at 21°Cthan at 16 or 26°C. Oscillation is hastened to fade-outat 21°C, most likely due to the accelerated pace of thehourglass. Red and far-red reversibility is disclosed in bothmodes of growth response. Phytoremediation potential of Lemna gibba was evaluated for chromium (Cr) and cadmium (Cd) under laboratory conditions for variable metal load of 1?mg/l, 3?mg/l, 5mgl, 7?mg/l and 9?mg/l, respectively, for 7 and 15?days of treatment period. Effects of both metals on structural attributes of L. gibba were also analyzed by Scanning Electron Microscopic (SEM) study. The metal removal percentage by L. gibba for Cr metal was found in the range of 37.3% to 98.6% and for cadmium it was found within the range of 81.6% to 94.6%. Bio concentration factor (BCF) of L .gibba was observed within the range of 37 to 295 for Cr metal and for Cd metal it ranged from 237 to 1144, which shows that the plant is a hyper accumulator for Cd metal and moderate accumulator for Cr metal. Statistical analysis (Two-way ANOVA) was performed on experimental results to confirm the individual effect of metal concentration and treatment period as well as cumulative effect of both factors together on percentage metal removal and on BCF. Research studies indicated that with the progress of treatment period metal removal percentage increases but increasing metal load during experiment negatively co-relates the metal removal percentage and BCF.  相似文献

Flat forms of the fronds of Lemna gibba L., used widely in laboratoryexperiments, can be easily confused with Lemna minor Chemicalanalyses of the two species, grown under defined laboratoryconditions, showed clear distinctions between the two speciesin relation to total protein content and the content of individualmonosaccharides. Infra-red spectral analysis confirmed the chemicaldata for protein content and also revealed differences in thetypes of proteins present in the two species. Chemical and infra-redspectral analyses, particularly the amide I: carbohydrate absorbanceratio, provide a rapid and reliable means of verifying betweenthe two species, which are often difficult to identify withcertainty from morphological differences. A 10 to 20% stimulation of growth in Lemna gibba L. G3 occurred following the addition of 0.5 to 3 mM glycolate or glyoxylate, although concentrations of 5 mM or higher were inhibitory. Glyoxylate gave a higher stimulation than glycolate. The stimulating effect on growth was obtained in media with or without 2% added sucrose. A higher stimulation was obtained when the plants were cultivated in open flasks in comparison to cultivation in flasks plugged with cellulose stoppers, which presumably retarded gas exchange.  相似文献

In this study, the amounts of antioxidant vitamins (A, E and C), selenium (Se), reduced glutathione (GSH)/oxidized glutathione (GSSG) ratio and malondialdehyde (MDA) that is the indicator of lipid peroxidation were determined in Lemna gibba L. plants placed in the secondary clarifier and grown in natural water. The amounts of antioxidant vitamins (A, E and C), GSH, GSSG and MDA were determined with HPLC (high performance liquid chromatography) and the amounts of Se were determined fluorimetrically. While significant decrease in amounts of antioxidant vitamins (A, E and C) and Se takes place between the first and the second or third day depending on species and insignificant decrease takes place after that day, the amount of GSH/GSSG ratio decreases until the second day (p < 0.05) and insignificantly increases after that day. Amounts of vitamins (A, E and C), Se and GSH/GSSG ratio for the plants placed in the secondary clarifier are much less than that for control group while an opposite trend was observed in MDA level. The MDA for the plants placed in secondary clarifier has maximum value at the second day, which can be considered as the maximum stress occurring at that day. Consequently, the first two days of treatment time can be taken as acclimation time and after the acclimation time the plant lives in the hostile environmental condition with the certain amount of oxidative stress. As a result, it is determined that wastewater decreases the lifetime of plant by causing metabolic stress on it and affects antioxidant capacity. The lifetime of the plants in the secondary clarifier was determined to be five days since fading toward yellow color (necrosis) in the plants was observed at the fifth day.  相似文献

The influence of arsenic (As) on the growth and the antioxidant system of Lemna gibba L. exposed to five concentrations of As (0.0, 0.25, 0.5, 1.0, and 1.5 mg L?1) was studied. Although As exposure reduced relative growth rate, L. gibba continued to grow even after the high accumulation of this element after five days of exposure. The concentration of the superoxide anion was unaffected by As, whereas an increase of hydrogen peroxide concentrations was observed with the increasing of As concentration. Increasing concentrations of As also increased the enzyme activity of superoxide dismutase, peroxidase, and glutathione reductase and the total and nonprotein thiols, up to 0.5 mg L?1, whereas the anthocyanin content increased constantly with As concentration. Catalase and ascorbate peroxidase activities as well as the content of chloroplastic pigments were reduced in plants exposed to all As concentrations. These results support a major role of anthocyanins, nonprotein thiols, and antioxidant scavenging enzymes in L. gibba tolerance to toxic As concentrations.  相似文献

The occurrence of acetylcholine in a long-day duckweed, Lemnagibba G3 has been demonstrated. After a preliminary purificationof the formic acid-acetone extract by Sephadex G-15 column chromatography,acetylcholine was identified by paper chromatography, pharmacologicalactivity on frog muscle, and sensitivity to acetylcholinesterase. Acetylcholine contents relative to that at the start of theexperimental culture were 0.99?0.06, 1.61?0.27, and 1.17?0.16after 2 cycles of the [9(15)], [16(8)], and [24(0)] schedules,respectively. Duckweed(Lemna gibba) is a useful model system for elucidating plant development, but the techniques needed for regenerating fronds from calli are not yet well established. This study examined the effects of auxin, sucrose, and gelling agents on callus and frond formation inL. gibba G3. After three weeks of culturing on a solid medium, two types of calli were observed: watery, pale-green, and undifferentiated; or white, compact calli that were organized into nodules and which resembled somatic embryogenie calli. Homogeneous callus lines were produced through selective subculture. To induce nodular calli, auxin (2,4-D) was absolutely required, with an effective concentration of 5 to 20 μM; induction was found to be possible with up to a maximum concentration of 4.4%. The calli were then maintained on a medium with a reduced 2,4-D concentration (1 μM), and were transferred every three weeks. Optimal callus induction and growth were obtained by using 3% sucrose with a combination of 0.15% Gelrite and 0.4% agar. Fronds, however, could be regenerated only on distilled water solidified with a combination of 0.4% agar and 0.15% Gelrite. On this medium, 87% of the callus expiants regenerated into fronds after four weeks of culture. These new fronds were morphologically normal but small, approximately 15 to 20% of the size of stock fronds. Continued culture of these fronds in an SH medium produced normal duckweeds, and histological examination of the cultures revealed several distinct types of callus nodules. Nonetheless, because zygotic embryogenesis inL. gibba does not produce distinct bipolar structures, the developmental pathway of frond regeneration from these nodular cultures remains unknown.  相似文献

Uptake of uridine by a long-day duckweed, Lemna gibba G3 wasexamined. Km and Vmax for uptake were in the range of 1 to 2x10–5 M and of 5 to 10 x10–8 moles/g fresh weight/2hr, respectively. Uptake rate depended on temperature, and theoptimum pH was 5.0. Uridine uptake was competitively inhibitedby some compounds structurally analogous to uridine. However,the activity of uridine kinase was not affected by these compounds,except for cytidine. Uridine uptake was inhibited by metabolicinhibitors, in which uridine taken up was left unconverted toother forms, especially in the presence of DNP. These resultssuggest that uridine was taken up into the duckweed celb bya specific transport system and immediately phosphorylated byuridine kinase. Aquatic plants are used as a practical and effective method to remove toxic elements from secondary-treated municipal wastewater. In this study, Lemna gibba was investigated for its capacity to remove uranium, arsenic, and boron from secondary effluents. L. gibba was collected from a natural lake in Elaz??, Turkey, then acclimatized to the effluent in situ. The concentration of toxic elements in the plant material was monitored as a function of time for 7 days. L. gibba significantly accumulated the toxic elements, particularly in the first 2 days. Arsenic, uranium, and boron were accumulated in the highest concentrations (133%, 122%, and 40%, respectively). However, in the following days, accumulation levels showed both increases and decreases, most probably due to L. gibba reaching saturation levels.  相似文献

Plants growing in natural environments are exposed to radiofrequency electromagnetic radiation (EMR) emitted by various communication network base stations. The environmental concentration of this radiation is increasing rapidly with the congested deployment of base stations. Although numerous scientific studies have been conducted to investigate the effects of EMR on the physiology of humans and animals, there have been few attempts to investigate the effects of EMR on plants. In this study, we attempted to evaluate the effects of EMR on photosynthesis by investigating the chlorophyll fluorescence (ChF) parameters of duckweed fronds. During the experiment, the fronds were tested with 2, 2.5, 3.5, 5.5 and 8?GHz EMR frequencies, which are not widely studied even though there is a potentially large concentration of these frequencies in the environment. The duckweed fronds were exposed to EMR for 30?min, 1?h and 24?h durations with electric field strength of 45–50?V/m for each frequency. The results indicated that exposure to EMR causes a change in the non-photochemical quenching of the duckweeds. The changes varied with the frequency of the EMR and were time-varying within a particular frequency. The temperature remained unchanged in the duckweed fronds upon exposure to EMR, which confirms that the effect is non-thermal.  相似文献
