The light response of miracidia was studied by means of several original methodics. The heterogeneity in the character of light response between various larvae of the same age population was determined in the course of experiments. It was shown that a part of miracidia possesses the strict positive phototaxis. Moreover, in their movement to the light the larvae seem to orientate themselves on the light intensity gradient. Not all the miracidia possess the positive phototaxis ("+" miracidia). Some of them ("-" miracidia) leave for the dark side of camera under the influence of direct ray of light. There are also larvae indifferent to the light conditions in thepopulation. They can move in different directions in spite of the light ray direction ("0" miracidia). The number of "+", "-" and "0" miracidia in the population is inconstant. The number of the negative phototactic larvae grows with age and respectively the number of "+" miracidia decreases. It is obvious the "+" miracidia can transform into "0" or "-" forms, while there is no opposite transformation. The reasons of differences in the movement character of "+" and "-" miracidia are under discussion.

Schistosoma japonicum miracidia swim directed along a chemical gradient toward the snails Oncomelania hupensis and Biomphalaria glabrata, and they turn back when the concentration of attractive chemicals decreases. The host signal for this chemotactic response has a molecular weight of more than 30,000. When swimming miracidia encounter the surface of O. hupensis or agar containing O. hupensis snail-conditioned water (SCW) they perform the host-specific responses "contact with return," "repeated investigation," and "attachment," but they do not exhibit such behavior when encountering B. glabrata surface or agar containing B. glabrata SCW. Thus S. japonicum miracidia respond to different host signals when they approach snails than when they attach to snails.

A striking difference was found in the responses of Schistosoma mansoni and S. haematobium miracidia to the intensity and direction of light and to gravity. S. mansoni miracidia were found to be positively phototactic and sensitive to small changes in light intensities. They were indifferent to gravity. S. haematobium miracidia however were negatively phototactic and unable to distinguish low light intensities from darkness. They showed a strong positive geotaxis. Both S. mansoni and S. haematobium miracidia could successfully find and infect snails to a depth of 2 metres of water: the significance of these findings in relation to transmission of schistosomiasis in the field is discussed.

The effects of light intensity, water temperature and river spate conditions on the rate of migration of hatchery-reared Atlantic salmon smolts down a release ladder were examined. Low light intensity and high day time water temperatures raised smolt migration rates: water temperature had littleor no effect at night. Thediel patternofdown-ladder movement wasdetermined by these two environmental stimuli. Smolts demonstrated a threshold response to both light and water temperature: a fall in light intensity, or increase in water temperature, below or above their respective thresholds elicited no further response. As fish progressed through the ladder their movement became increasingly nocturnal, and most entry into the estuary occurred at night. Heavy rainhll caused rapid migration of smolts through the ladder. As ladder discharge remained constant, smolts must have been responding to some other change in the water conditions. Smolts responded to spate conditions irrespective of water temperature and light intensity, and they responded to light intensity irrespective of water temperature. A hierarchy of environmental cues, responsible for triggering migratory behaviour on a day to day basis during the smolt run, of spate> light intensity > water temperature is therefore suggested.

The effects of salinity, pH, and temperature on the longevity and hatchability of miracidia of Philophthalmus megalurus and P. gralli were determined. Miracidia of both species are able to hatch and survive at saline concentrations much above physiological levels, although these processes are reduced in 2.0--2.4% saline and completely inhibited at 2.6%. The greatest hatching rates for both species were found near neutrality (pH 6--8) but some miracidia hatched at the extreme pH levels of 3 and 12. Philophthalmus megalurus miracidia exhibited longer half-lives under acid conditions (pH 2--6) than P. gralli miracidia; conversely, P. gralli miracidia showed longer half-lives in alkaline conditions (pH 8--11). Hatching and longevity were much greater below room temperature (5--20 C) than above (30--50 C) for miracidia of both species. Temperatures above 50 C proved lethal for eyefluke eggs. Except in acid pH, P. gralli miracidia showed longer half-lives than miracidia of P. megalurus. Comparison to studies on schistosomes revealed that the inhibitory effects of physiological saline and host body temperature on the hatching process of schistosome eggs does not occur in these 2 species of eyeflukes.

Chemical communication among freshwater organisms is an adaptation to improve their coexistence. Here,we focus on the chemical cues secreted by the freshwater gastropod Lymnaea stagnalis, which are known to stimulate behavioural responses of Trichobilharzia ocellata (Plathelminthes, Digenea, Trematoda) miracidia. Such responses are commonly claimed to influence transmission positively, but in response to chemical cues miracidia randomly change their swimming direction. This kind of response does not necessarily increase transmission, because miracidia may be trapped at the periphery of very large snail odour-clouds, which may prevent them from approaching the snail. On the other hand, the odour clouds may be too small to improve host-localisation. To shed light on these scenarios, the spreading of molecules released around L. stagnalis (active space) was visualised by recording host-finding responses of T. ocellata miracidia when they approached snails. Behavioural responses of miracidia indicated the spreading of compounds forming an attractive active space only around the host-snail L. stagnalis, but not around sympatric non-host-snail species. The active space increased approximately linearly with the time the snail rested at the same spot and within 5 min it reached a volume of more than 30 times that of the snail. We also demonstrated in a large-scale experiment, that the active space of L. stagnalis significantly increases the transmission success of T. ocellata miracidia. Additionally, the microhabitat selection of T. ocellata miracidia was studied, demonstrating that peripheral locations near the water surface were preferred, which are also preferred sites of L. stagnalis. Improved chemoperception and microhabitat selection may have been a consequence of coevolution with snails and benefited miracidia, which became efficient transmissive stages. During a short period of the year, Daphnia may perform a phenotypically induced diel vertical migration. For this to happen, light-induced swimming reactions must be enhanced both at dawn and at dusk. Enhanced swimming in response to light intensity increase can be elicited by fish-associated kairomone in the laboratory, if food is sufficiently available. However, during the light change at dusk the Daphnia are still in the hypolimnion, where no fish kairomone is present and both temperature and food availability is low. Still, what goes down must come up. This raises questions about how Daphnia tunes its light-induced swimming behaviour to prevailing conditions such that a normal diel vertical migration can be performed. We investigated the symmetry in behavioural mechanism underlying these diel vertical migrations in the hybrid Daphnia galeata × hyalina (Cladocera; Crustacea), with special interest for the environmental cues that are known to affect swimming in response to light increase. That is, we tested whether fish- associated kairomone, food availability, and temperature affected both swimming in response to light intensity increase and decrease similarly. We quantified swimming behaviour during a sequentially increased rate of light change. Vertical displacement velocity was measured and proved to be linearly related to the rate of the light change. The slope (PC) of the function depends on the value of the factors kairomone concentration, food availability, and temperature. The changes of the PC with kairomone concentration and with temperature were similar both at light intensity increases and decreases. The PC also increased with food concentration, although during light increases in a different way from during light intensity decreases. Low food availability inhibited swimming in response to light intensity increase, but enhanced swimming in response to light intensity decrease. Hence, ascent from the deep water layers with low food concentration at dusk is facilitated. These causal relations are part of a proximate decision-making mechanism which may help the individual Daphnia to tune migration to predation intensity and food availability.

In a four-tube vertical system, Echinostoma caproni miracidia exhibited a strong negative geotaxis which was dominated by a positive phototaxis. In horizontal chambers a positive phototactic response was also demonstrated. These miracidia showed a positive chemoresponse, as determined in phi-chambers, to glutamic and aspartic acids but not leucine. Positive responses were also elicited to snail-conditioned water and sulfuric and acetic acids. Ammonia, Mg2+, and HCl produced no significant reactions. Responses of E. caproni and Schistosoma mansoni miracidia, both of which develop in Biomphalaria glabrata snails, were similar providing further evidence that miracidia mimic the behavioral patterns of compatible snail species.

The influence of temperature and ultra-violet radiation on the degree of activity, survival and infectivity of schistosome miracidia is profound. Miracidia of Schistosoma mansoni and S. haematobium were affect eaually. Only miracidia classified as "active" or "slow" were capable of penetration, a capacity they retained for about 17 hours at 19 degrees C. Miracidia that were "lethargic" as a result of low temperature, old age or ultra-violet radiation lost their infective capacity. The conclusion, however, is that neither the temperatures encountered in the field nor the solar ultra-violet radiation penetrating turbid waters are likely to be harmful to miracidia and thus have no effect on the level of transmission.

Carrot fly displays a preference for moist air but the female does not differentiate humidities above 65% RH. In a temperature gradient aggregation takes place at 18–24°.An apparatus is described in which the effects of temperature, humidity and light intensity on activity were studied. Activity was always on a higher level below 65% RH and it is suggested that this orthokinetic response is the mechanism by which flies aggregate in moist air. Light intensity influenced the kinetic response to humidity; low light intensity tending to produce similar activity at all levels. These reactions are discussed in relation to field behaviour.

Biomphalaria glabrata and Schistosoma mansoni relationship was studied by light microscopy (LM) and freeze-fracture replica technique (FFR). We observed very thin cytoplasmic extensions of hemocytes in the LM, which then surround immobilize the miracidia. FFR images showed that the contact site between hemocytes cytoplasmic extensions and the external tegumentary coat involved only superficial layers of miracidia. Numerous vacuoles and filopodia were observed in the hemocyte cytoplasm, the latter binding with those from neighboring cells. In spite of the close interfilopodia contact, no cellular junctions were seen at these sites nor between filopodia-miracidia contact areas. The observed migration of hemocytes and their disposition in layers surrounding the miracidia in vitro correspond to previous studies.

Suspensions of miracidia and cercariae of Schistosoma mansoni were subjected to repeated freeze-thaw cycles and then injected into resistant Salvador strain Biomphalaria glabrata snails. A pronounced increase in the number of mitotic figures, relative to uninjected, sham-injected, or diluent (water)-injected controls, was observed in the amebocyte-producing organ (APO) at 3 days postinjection (PI). After centrifugation of miracidia freeze-thaw extract (FTE), the resulting supernatant (FTS) and pellet possessed equal stimulatory activity that was approximately half that seen with FTE. Ultracentrifugation of miracidia FTS resulted in a supernatant that retained full activity, indicating a soluble molecule. Heat treatment of miracidia FTE reduced but did not eliminate activity, suggesting a nonprotein active component. Concentration or dilution of FTS by a factor of 10 gave a nonlinear dose-response relationship. Susceptible NIH albino snails injected with miracidia FTE had increased mitotic activity in the APO, which was much less than that seen in Salvador snails, whereas injection of miracidia FTE into Helisoma duryi had no discernable effect. Measurement of mitotic activity as a function of time PI showed no increase in numbers of mitotic figures in the APO at 18 hr but a large increase at 24 hr PI. Mitotic activity returned to preinjection levels by 96 hr PI, although a subsequent increase occurred at 120 hr PI.

SYNOPSIS. The effect of temperature on photoaccumulation and photophobic response of Volvox aureus were studied. The algae exhibited positive photoaccumulation at room temperature and negative at low temperature. When stimulated with light of intermediate intensiy (~ 5 × 103 lux), the phobic response of the algae consisted of a decrease in the frequency or the cessation of flagellar movement in the anterior cells. At room temperature, an increase in light intensity elicited the phobic response, whereas at low temperature a decrease in light intensity was the effective stimulus. The phobic response lasted only a few seconds. The positive and negative photoaccumulations of the algae could be explained by the brief cessation of flagellar movement in the anterior cells, elicited by an increase of stimulus light at room temperature or a decrease of stimulus at low temperature.

Summary The germination response of seeds of Ludwigia octovalvis (Jacq.) (=Jussiaea suffruticosa L.; Raven, 1963), to continuous light and to various types of intermittent irradiations is determined at 25, 30, 35 and 40° at different light intensities. At 25 and 40° intermittent irradiations are an effective substitute for continuous ones, while at 30 and 35° no promotion of germination is observed in the intermittent irradiations tested, except if the dark interval between light pulses is reduced to 1 min. Previous results obtained in the photocontrol of germination in this species are confirmed and extended to a variety of light cycles and intensities, indicating that the response to temperature is bimodal unless light is supplied continuously with high intensity, and that germination depends on light intensity more markedly in the temperature region of minimum germination (30–35°). As germination in this species is controlled by the red far-red system, hypothesis to account for the temperature dependence are based on the interaction between temperature and the reactions in which phytochrome is involved.Abbreviations L light - D darkness  相似文献

The effects of light intensity and temperature on Arthrospira platensis growth and production of extracellular polymeric substances (EPS) in batch culture were evaluated using a three-level, full-factorial design and response surface methodology. Three levels were tested for each parameter (temperature: 30, 35, 40°C; light intensity: 50, 115, 180 μmol photons m−2 s−1). Both growth and EPS production are influenced mainly by the temperature factor but the interaction term temperature*light intensity also had a significant effect. In addition, conditions optimising EPS production are different from those optimising growth. The highest growth rate (0.414 ± 0.003 day−1) was found at the lowest temperature (30°C) and highest light intensity (180 μmol photons m−2 s−1) tested, no optima were detectable within the given test range. Obviously, optima for growth must be at a temperature lower than 30°C and a light intensity higher than 180 μmol photons m−2 s−1. For EPS production, light intensity had a positive linear effect (optimum obviously higher than 180 μmol photons m−2 s−1), but for the temperature parameter a maximum effect was detectable at 35°C.

Schistosoma mansoni occurs in tropical regions where levels of ultraviolet B (UVB; 290-320 nm) light are elevated. However, the effects of UVB on parasite transmission are unknown. This study examines effects of UVB on the miracidia and sporocysts of S. mansoni, focusing specifically on intramolluscan development, infectivity, and the ability to photoreactivate (repair DNA damage using visible light). Histology revealed that miracidia irradiated with 861 J x m(-2) underwent abnormal development after penetrating Biomphalaria glabrata snails. Total number of sporocysts in snail tissues decreased as a function of time postinfection (PI), among both nonirradiated and irradiated parasites; however, this decrease was greater in the latter. Moreover, whereas the proportion alive of nonirradiated sporocysts increased PI, that of irradiated sporocysts, i.e., derived from irradiated miracidia, decreased. Irradiation of miracidia with UVB resulted in decreased prevalence of patent infection (defined by presence of daughter sporocysts) in a dose-dependent manner, and no infections occurred at a dose of 861 J x m(-2). Like many aquatic organisms, including the snail host, parasites were able to photoreactivate if exposed to visible light following UVB irradiation, even subsequent to penetrating snails. These photoreactivation results suggest cyclobutane-pyrimidine dimers in DNA as the primary mechanism of UVB damage, and implicate photoreactivation, rather than nucleotide excision, as the main repair process in S. mansoni.

This study investigated the effects of the photophase light intensity on the scotophase melatonin response. Twelve, 8-month-old crossbred gilts were allocated to three groups of four and housed in temperature- and lighting-controlled climate rooms. The rooms had a light intensity of 40, 200 or 10,000 lx and a light-dark cycle of 12 L:12 D. The gilts were allowed to acclimatize to a new lighting regimen for 1 week before being sampled at 2h intervals for 24h. Following the sampling, pigs were transferred under a different light intensity, allowed to adjust for 1 week and sampled again. The procedure was repeated three times so that all the groups went through all three lighting regimens (light intensities). All the gilts exhibited a clear circadian serum melatonin rhythm under each lighting regimen with high melatonin concentrations occurring during the scotophase. There was no difference in the scotophase melatonin response in terms of mean concentrations or duration of increased melatonin levels within or between the groups under different lighting regimens. There was considerable inter-individual variation in the dark phase melatonin response but the individual profiles were consistent under the different lighting regimens. It is concluded that when a certain threshold light intensity (<40lx) is exceeded, the photophase light intensity has no effect on the scotophase melatonin response. These results imply that extremely high light intensities during the photophase would provide no additional benefits compared with normal comfortable light intensity, if artificial lighting programs were introduced to commercial piggeries in order to reduce seasonal effects on reproduction.

Movement patterns of Schistosoma mansoni miracidia were examined in several concentrations and gradients of snail-conditioned water (SCW). Miracidia surrounded by uniform concentrations of SCW swam at the same speed and exhibited the same rate of turning (angular velocity) as did control miracidia swimming in spring water. However, miracidia in gradients of SCW exhibited a 3-fold increase in their angular velocity without altering their swimming speed. Miracidia ascending gradients of SCW did not increase their angular velocity and failed to orient to the gradient of the stimulant. In contrast, miracidia which encountered sufficiently abrupt decreases in SCW concentration, while descending the gradient, sharply increased their angular velocity. This behavior caused miracidia to remain in regions of high concentration of stimulant. The magnitude of decrease in SCW concentration needed to evoke this response depended on the absolute concentration of SCW. Thus, the miracidial response is a "boundary reaction", a form of chemoklinokinesis, and not a chemotaxis.

The effects of light and temperature on the germination and growth of Luffa aegyptiaca were investigated both in the laboratory and in the field. The seeds germinated in both darkness and light but germination was better in the light. At constant temperatures germination was best at 21°C, while alternating temperatures of 21 and 31°C and 15 and 41°C caused higher germination than the most favourable constant temperature. Constant temperatures of 15 and 31°C and alternating temperatures of 21 and 41°C resulted in very low germination, whereas no germination occurred at 41°C and at alternating temperatures of 31 and 41°C. Soil depth caused only a delay in seed germination, as it did not affect the total germination. High temperature and high light intensity resulted in good seedling growth in terms of dry weight, leaf area and relative growth rate. High temperature and low light intensity caused increased plant height and high shoot weight ratio, both of which manifested in seedling etiolation. They also caused high leaf area ratio. Under low temperatures, irrespective of light intensity, growth was generally poor, but it was significantly poorer under low light intensity, which also caused high root weight ratio. High light intensity was principally responsible for high leaf weight ratio. The results help to explain the abundance of the species in newly cleared areas in Lagos and its environs.
