本文旨在构建沙门菌-斑马鱼感染模型,研究幼年斑马鱼体内的异源自噬。用不同浓度鼠伤寒沙门菌浸染受精后72h的幼年斑马鱼,绘制7d内斑马鱼的存活率曲线以确定适宜的细菌感染量。以上述细菌量感染斑马鱼后,荧光显微镜下观察沙门菌在体内的播散情况;在不同时间点计数细菌菌落,并用蛋白免疫印迹法检测自噬相关蛋白Lc3和p62的表达,同时在电子显微镜下观察自噬体结构。结果显示,以1×109CFU/ml细菌量感染斑马鱼后,2d内斑马鱼存活率为100%,第3天开始出现死亡。荧光显微镜下发现,感染后4h鱼体中有细菌侵入,3d后细菌扩散至全身。细菌计数结果显示,整条鱼感染的细菌量随时间延长不断增加,但感染后10h侵入鱼体细胞中的菌量显著低于8h。蛋白免疫印迹结果显示,感染后8hLc3-Ⅱ表达显著升高,p62表达显著降低。透射电子显微镜下,感染后8h幼鱼细胞中可观察到自噬体和自噬溶酶体结构。以上结果提示,用鼠伤寒沙门菌浸染幼年斑马鱼构建的模型可用于追踪病原菌感染引起的异源自噬的动态变化。  相似文献

为研究伤寒沙门菌质粒pRST98对人巨噬细胞THP-1自噬过程的影响,以携带伤寒沙门菌质粒pRST98的野生株ST6、消除pRST98的突变株ST6-ΔpRST98和将pRST98经接合转移的回补株ST6-c-pRST98为受试菌,与THP-1共培养建立感染模型,并加入自噬作用阻断剂氯喹(CQ)进行干预。首先测定CQ单独作用对细胞及细菌的影响,确定CQ最适浓度;在细菌和细胞共作用后的不同时间点收集细胞,通过蛋白免疫印迹法(WB)和mRFP-GFP-LC3 质粒转染法检测细胞LC3Ⅱ蛋白、p62蛋白、自噬体及自噬溶酶体的变化。结果显示,30 μmol/L CQ对细胞自噬的阻断效果明显,且细胞存活率超过50%,对细菌也无明显影响。WB结果显示,用该浓度CQ干预后,ST6-ΔpRST98感染组细胞的LC3Ⅱ及p62表达量上升程度显著高于野生株ST6及回补株ST6-c-pRST98;CQ干预组3株受试菌感染细胞LC3点状结构数量均高于未加CQ组,且ST6-ΔpRST98感染细胞的点状结构数量明显增加。以上结果提示,伤寒沙门菌质粒pRST98在自噬前期发挥作用,早于溶酶体降解的过程。  相似文献

探讨鼠伤寒沙门菌在感染鼠巨噬细胞早期与细胞器的相互作用。用pTassC-GFP质粒转染鼠巨噬细胞RAW264.7,结合多抗的溶酶体标志物溶酶体相关膜蛋白-1用键合了Alexa594的羊抗鼠二抗显色,以观察标记了绿色荧光蛋白的TassC与溶酶体的关系;用pTassC-GFP和pDsRed2-Perxi质粒共转染RAW264.7细胞,以观察TassC-GFP与过氧化物酶体的关系;用SYTO42标记鼠伤寒沙门菌,感染用pTassC-GFP和pDsRed2-Perxi质粒共转染的RAW264.7细胞,以观察细菌与TassC和过氧化物酶体的关系。免疫荧光显示TassC-GFP不与鼠巨噬细胞RAW264.7中的溶酶体结合,但与标记了红色荧光的过氧化物酶体共定位;感染1 h的RAW264.7胞内SYTO42标记的鼠伤寒沙门菌吞噬泡可招募TassC-GFP和过氧化物酶体。这些发现提示在鼠伤寒沙门菌感染早期过氧化物酶体携带杀菌成分通过TassC介导可参与发挥一定的杀菌作用。  相似文献

自噬(autophagy)是一种进化保守的溶酶体依赖性分解代谢途径,是细胞维持自稳态的重要机制之一,并参与多种疾病的发生. Beclin-1作为自噬体成核的关键分子之一,是1个调节自噬的关键靶点. Beclin-1有1个BH3结构域,Bcl-2、Bcl-XL等可以通过这个BH3结构域与Beclin-1结合而影响其活性. 抗凋亡Bcl-2家族蛋白和Beclin-1的表达水平、磷酸化、分子的亚细胞定位以及BH3-only蛋白等,均可调节Beclin-1蛋白和Bcl-2家族蛋白结合水平,进而调控自噬的发生,并可能对细胞最终走向自噬还是凋亡起着关键作用.  相似文献

用荧光定量PCR法检测鼠RAW264.7巨噬细胞感染与未感染鼠伤寒沙门菌后18种铁穗态相关基因的表达,评估宿主与病原体相互作用中铁稳态效应。研究显示,活的鼠伤寒沙门菌感染巨噬细胞1 h后可以诱导转铁蛋白受体表达,引起细胞内动态铁池相关基因的mRNA水平上长。基因表达分析显示,沙门菌通过诱导铁氧还原酶(Steap3)、铁膜转运蛋白(Dmt1)、铁调节因子Tfr2/Hfe以及铁调节蛋白(Irp1和Irp2)的表达主动吸收铁,而经铁转运蛋白(Fpn1)的铁外流并无明显改变。沙门菌在感染后1h积极地驱动了转铁蛋白介导的铁吸收程序。  相似文献

为探讨连香树精油的体外抗肿瘤活性。以人肝癌SMMC-7721细胞为受试细胞株,利用MTT法、流式细胞术、JC-1法和Western blot法评价连香树精油的体外抗肿瘤活性。结果表明:连香树精油处理24 h后可降低SMMC-7721细胞存活率;细胞中G0/G1期的比例下降,G2/M期的比例升高;细胞线粒体膜电位降低;自噬相关蛋白LC3-Ⅱ、Beclin-1表达上升,加入自噬抑制剂氯喹后LC3-Ⅱ蛋白与Bcl-2抗凋亡蛋白表达下降。综上说明连香树精油能抑制SMMC-7721细胞的增殖活性,通过阻滞G2/M期抑制细胞分裂,通过降低线粒体膜电位诱导SMMC-7721细胞凋亡,而自噬在SMMC-7721细胞凋亡过程中作为一种保护机制而存在。  相似文献

MG132(Z-Leu-leu-leu-CHO) 是一种蛋白酶体抑制剂,它可逆地抑制蛋白酶体活性,从而抑制泛素-蛋白酶体通路所介导的蛋白质降解,诱导细胞凋亡.实验研究MG132抑制肝癌细胞Bel-7404生长的机制.采用不同浓度梯度和时间梯度,通过荧光显微镜、透射电子显微镜、Hoechst33342染色、MTT检测、AnnexinⅤ/PI流式细胞术、Western blot分别检测MG132 对Bel-7404细胞的形态学变化、细胞内质网压力变化、自噬泡形成、凋亡小体形成、细胞存活率、细胞凋亡水平、凋亡及自噬信号途径中相关蛋白质表达的影响.结果显示,MG132能明显抑制Bel-7404细胞生长.通过增加内质网压力激活Caspase-12,也可通过线粒体途径调节Bcl-2/Bax水平,促进细胞色素c的释放,两者皆可激活下游效应Caspase-3,剪切PARP,诱导细胞凋亡.同时,MG132可诱导Beclin 1 和LC3B的上调,促使Bel-7404细胞发生自噬,可在透射电镜下观察到自噬泡形成.上述结果表明,MG132作用Bel-7404细胞涉及两类细胞程序性死亡途径:细胞凋亡和自噬.  相似文献

目的了解短链脂肪酸(SCFA)作用伤寒沙门菌诱导巨噬细胞凋亡机制。方法将SCFA作用伤寒沙门菌感染巨噬细胞8 h后,检测TNF-α、caspase3、caspase8、caspase9及NO的产生量,同时检测加入caspase3、caspase8、caspase9抑制剂和TNF-α抗体后的细胞凋亡率。结果作用8 h后caspase3、caspase8及NO、TNF-α的产生量均高于对照组(P0.01)。caspase3、caspase8抑制剂和TNF-α抗体均能不同程度抑制SCFA作用伤寒沙门菌诱导的巨噬细胞凋亡(P0.01)。结论 SCFA作用伤寒沙门菌诱导巨噬细胞凋亡可以通过NO及TNF-α介导,caspase3和caspase8参与的外源性凋亡途迳。  相似文献

目的:探讨白花蛇舌草多糖提取物(HDPE)对喉癌Hep-2细胞内质网自噬的影响。方法:实验分为对照组、HDPE 100、200、400 mg/L组和3-MA(自噬抑制剂)组,噻唑盐比色法(MTT)检测各组细胞培养24 h、48 h、72 h后增殖抑制率;原位末端转移酶标记法(TUNEL)法检测各组培养48 h细胞凋亡情况;单丹黄酰尸胺(MDC)染色观察各组培养48 h细胞自噬体及自噬溶酶体的变化;透射电镜观察培养48 h细胞内质网周围自噬囊泡的产生情况;蛋白印迹法(Western blot)检测各组培养48 h细胞Beclin-1蛋白(Beclin-1)、微管相关轻链蛋白3Ⅰ(LC3Ⅰ)、微管相关轻链蛋白3Ⅱ(LC3Ⅱ)、葡萄糖调节蛋白 78(GRP78)、活化转录因子6(ATF6)及CCAAT 增强子结合蛋白同源蛋白(CHOP)表达。结果:与对照组比较,HDPE 100、200、400 mg/L组和3-MA组细胞增殖抑制率、凋亡指数AI升高,MDC阳性细胞率量降低,内质网周围自噬囊泡减少,GRP78、ATF6及CHOP表达及LC3Ⅰ/LC3Ⅱ比值升高,Beclin-1表达降低(P<0.05);与3-MA组比较,HDPE 400 mg/L组细胞增殖抑制率、凋亡指数AI升高,MDC阳性细胞率、GRP78、ATF6及CHOP表达及LC3Ⅰ/LC3Ⅱ比值升高,Beclin-1表达降低(P<0.05)。结论:HDPE可能通过抑制喉癌Hep-2细胞内质网自噬,促进细胞内质网应激凋亡,进而抑制Hep-2细胞增殖能力。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

目的 针对医疗机构的合理用药水平进行评价研究。方法 根据医疗机构合理用药的具体要求,构建医疗机构合理用药评价指标体系,采用基于模糊群决策的方法和多指标评价分析法构建医疗机构合理用药评价模型。结果 构建了基于模糊群决策的医疗机构合理用药评价模型,并通过实例分析证明了评价模型的可行性。结论 建立的基于模糊群决策的医疗机构合理用药评价模型能够对医疗机构的合理用药水平进行科学评价,为提高医疗机构合理用药水平奠定基础。  相似文献
