In the light, transfer of lettuce seedlings precultured on liquid medium at pH 6.0 to fresh medium at pH 4.0 induces root hair formation. However, no root hairs form in the dark. Here, we investigated how light induces root hair formation. Randomization of the transverse cortical microtubule (CMT) arrays which occurs in root epidermal cells in the light prior to root hair initiation was not observed in the dark. However, addition of indole-3-acetic acid (IAA) or 1-aminocyclopropane-1-carboxylic acid (ACC) induced CMT randomization and root hair formation. In these cases, CMT randomization occurred in almost the same time-dependent manner as under light. However, root hair initiation was delayed for several hours in the dark. These results suggest that light promotes CMT randomization and root hair initiation via auxin and ethylene signaling but light additionally influences root hair initiation independently of these signaling mechanisms. Furthermore, addition of a microtubule-depolymerizing drug in the dark disrupted the transverse CMT arrays and initiated root hair formation; however, root hair elongation was still suppressed. Root hairs elongated when IAA or ACC was applied with the drug. These results suggest that light also promotes root hair elongation via auxin and ethylene signaling.  相似文献

The degree of relationships withinLactuca sativa and three wild relativesL. serriola, L. saligna, andL. virosa was studied by observing the performance, vigour and fertility of the F 1 hybrids obtained from crosses made in and between the four species. The crosses ofL. saligna ×L. virosa and the reciprocal crosses produced no hybrids.L. saligna andL. virosa are the least related of the four species.L. sativa ×L. serriola and the reciprocal crosses were successful and produced fertile hybrids These two species are genetically very closely related.L. saligna is known to produce, as a female parent, hybrids withL. sativa andL. serriola. Now the reciprocal cross was successful for the first time, so the unability to obtain hybrids in the past was based on the choice of accessions and not caused by unilateral incompatibility.L. virosa ×L. sativa and the reciprocal combination produced hybrids. The combinationL. serriola ×L. virosa produced hybrids with very limited fertility. In contrast to earlier reports (sterile hybrids) one combination of the reciprocal cross too produced hybrids with very limited fertility.—Some of theL. saligna ×L. sativa (and reciprocal) hybrids were found to look strikingly likeL. serriola. This adds evidence for the descent ofL. serriola andL. sativa:L. saligna also made part of the ancestral complex of the cultivated lettuce.  相似文献

Sesquiterpenoid lactones (SLs) from lettuce (Lactuca sativa L.) include constitutive components of latex such as lactucin and the induced phytoalexin, lettucenin A. A redundant primer strategy was used to recover two full length cDNA clones (LTC1 and LTC2) encoding sesquiterpene synthases from a cDNA library derived from seedlings with the red spot disorder, which accumulate phytoalexins. Recombinant enzymes produced from LTC1 and LTC2 in Escherichia coli catalysed the cyclisation of farnesyl diphosphate to germacrene A, potentially an early step in the biosynthesis of SLs. RT-PCR analysis showed LTC1 and LTC2 were expressed constitutively in roots, hypocotyls and true leaves but not in cotyledons. Expression in cotyledons was induced by challenge with the downy mildew pathogen Bremia lactucae in the disease resistant cultivar Diana. Southern hybridisation experiments showed that LTC1 and LTC2 were not part of a multigene family. The germacrene A synthases provide targets for modified expression to generate beneficial modifications to the SL profile in lettuce.  相似文献

Summary The influence of light quality on competence and determination for organogenesis was investigated using lettuce cotyledon explants. Lettuce seedlings from four genotypes were germinated in the dark or under white, red, or blue light. Cotyledon explants were excised and cultured on a shoot-inducing medium for 28 d under white light. Germination in the dark reduced shoot numbers, suggesting that light improves the competence of explants for organogenesis. When explants from seedlings germinated under white light were cultured under different light qualities, blue was found to inhibit shoot production while red light either promoted production or had no effect on shoot number compared to controls. Treatment with blue plus red light failed to overcome the inhibition by blue light. To ascertain the temporal responses of explants to light quality, they were cultured under red or blue light prior to transfer to the alternate treatment. Exposure to blue light within 7 d of excision permanently reduced explant competence for organogenesis. Exposure after this time had a minimal effect. These results suggest that both phytochrome and cryptochrome can regulate shoot production from lettuce cotyledon explants and blue light can only inhibit organogenesis, in lettuce, during a relatively small developmental window.  相似文献

Ferulic acid (FA) is released by living roots and by decaying plant material and is involved in chemical interactions between plants. Effects of FA on plant growth and root development of lettuce (Lactuca sativa L. cv. Grand Rapids) cultivated in axenic nutrient solution were studied in two factorial experiments. Root and shoot growth was impeded when 200 M trans-FA was added to the nutrient solution and the light intensity was in the range of 250–380 mol m-2 s-1. Root growth showed a stronger response to FA than did shoot growth. At 200 M, FA strongly inhibited root hair formation and reduced mean lengths of primary, secondary and tertiary roots, but stimulated primary and secondary root branching. Both isomerization to the cis isomer and the presence of the plant reduced the concentration of trans-FA in the nutrient solution during the two weeks exposure period. A third experiment was conducted to assess the influence of irradiance on the phytotoxicity of FA. At a light intensity of 489 mol m-2 s-1, or in the presence of microorganisms, the concentration of FA in the nutrient solution was lowered and the phytotoxic effects were reduced.  相似文献

采用水培技术,以油麦菜幼苗为材料,研究不同硝铵态氮配比(NO3-∶NH4+)对油麦菜苗期地上部和根系生长及生理特性的影响。结果表明:(1)油麦菜地上部和根系硝酸盐含量皆与营养液中NO3--N比例呈正相关关系,且各处理均达到无公害蔬菜的标准。(2)随着营养液中NH4+-N比例的增加,油麦菜地上部有机酸含量先降低后升高,且在硝铵态氮配比为5∶5时最低,可溶性糖含量呈上升趋势,而可溶性蛋白质含量先升高后降低,在硝铵态氮配比为5∶5时最高;油麦菜根系有机酸和可溶性糖含量先升高后降低,两者分别在硝铵态氮配比为5∶5和7.5∶2.5时最高,而可溶性蛋白质含量呈下降趋势,在全NO3--N时最高。(3)随着营养液中NH4+-N比例的增加,油麦菜地上部和根系中SOD活性先升后降,并分别在硝铵态氮配比为5∶5和7.5∶2.5时最高,而地上部和根系中MDA、脯氨酸含量和POD、CAT活性的变化趋势则与其相反。(4)随着营养液中NH4+-N比例的增加,油麦菜地上部和根系干重皆先升后降,根冠比则逐渐减小;在硝铵态氮配比为7.5∶2.5时干重最大,根冠比适宜且稳定。研究表明,水培油麦菜苗期地上部和根系生长及生理特性受到氮素形态配比的显著影响,且根系的生理响应更敏感;营养液中硝铵态氮配比为7.5∶2.5时,油麦菜受胁迫程度最低,地上部和根系生长较协调,油麦菜生长和生理状况最佳。  相似文献

以‘全年油麦菜’尖叶莴苣为试验材料,采用水培方式,研究3个浓度(0 mg·L-1、0.1 mg·L-1、1 mg·L-1)Ni2+在22.4 mg·L-1 N处理下对尖叶莴苣氮素吸收的生长及生理影响。结果显示:(1)尖叶莴苣根系和地上部生物量随处理时间的增加呈上升趋势。与对照T1(0 mg·L-1 Ni2+、112 mg·L-1 N)相比,T2处理(0 mg·L-1 Ni2+、22.4 mg·L-1 N)对尖叶莴苣根系及叶片生长具有一定抑制作用,植株鲜重、干重、根冠比、根系长度、平均直径、表面积、体积、根尖数、分根数、叶片表面积和体积在T3处理(0.1 mg·L-1 Ni2+、22.4 mg·L-1 N)下显著高于对照,T4处理(1 mg·L-1 Ni2+、22.4 mg·L-1 N)对尖叶莴苣根系及其叶片生长具有一定促进作用,但对其根尖数和分根数表现出一定抑制性。(2)随着Ni2+浓度的增加,尖叶莴苣叶片叶绿素a、叶绿素b和总叶绿素含量呈先升后降的变化规律,且均在T3处理下显著提高。(3)随着处理时间的增加,尖叶莴苣叶片的净光合速率(Pn)、气孔导度(Gs)和蒸腾速率(Tr)逐渐上升,胞间CO2浓度(Ci)逐渐下降,且T3处理叶片的Gs显著高于对照,其Ci最低,Pn最大。(4)施加Ni2+对尖叶莴苣有机酸、可溶性蛋白和可溶性糖含量以及SOD和POD活性有显著影响,在T3处理下有机酸含量降低,可溶性糖和可溶性蛋白含量显著增加,SOD和POD活性显著提高。(5)T3处理尖叶莴苣根系中N及叶片中B和Ca含量较高;根系中Ni含量高于叶片,T3处理叶片中的Ni含量较低,Mg含量较高;植株体内Cu含量随Ni2+浓度增加而下降。研究表明,外源Ni2+处理能影响低氮条件下(22.4 mg·L-1 N)尖叶莴苣幼苗生长及生理状况,适宜浓度(0.1 mg·L-1)Ni2+可有效提高尖叶莴苣根系对氮素的吸收利用效率,减少氮素施用量,促进尖叶莴苣根系和地上部叶片生长,增加光合色素含量,并提高净光合速率,进而改善植株的产量和营养品质。  相似文献

Light quality has previously been shown to influence morphogenesis in lettuce cotyledon explants, with white or red light promoting adventitious shoot production, and blue light inhibiting it. Endogenous polyamine (PA) concentrations were compared between explants cultured under different light qualities. Explants cultured under white or red light accumulated PAs during shoot primordia production, with a 5.6-fold increase compared to initial concentrations under white light, and 6.7-fold increase under red light. These results suggest polyamines are involved in the formation of shoot primordia. After 18 days in culture PA concentrations decreased under white light, and to a lesser extent under red light, signaling a shift in polyamine metabolism that correlates with shoot expansion, which occurs more readily under white light. Explants cultured under blue light accumulated polyamines for the first 7 days, to a level 1.3 times greater than initial values, followed by a gradual decline during the remainder of the culture period. Explants cultured under blue light also contained a greater proportion of PCA-insoluble conjugated PAs, compared to explants under white or red light, which contained greater proportions of free or PCA-soluble conjugated polyamines. The ratio of putrescine to spermidine was also different with a lower Put:Spd ratio being associated with shoot production under white or red light, and higher Put:Spd ratio being associated with culture under blue light.  相似文献

L. sativa L. cultivars were compared morphologically with accessions ofL. saligna L.,L. serriola L., andL. virosa L. in order to define the infra- and interspecific variation. Multivariate analysis (principal component analysis) distinguished four groups corresponding with the four species. Infraspecific variation (e.g., colour, leaf shape) withinL. sativa is the largest, because breeding has caused a large variation in vegetative features; infraspecific variation withinL. serriola is also large. Three plants of one accession were placed betweenL. sativa andL. serriola because they showed morphological characteristics of both species. They are still classified asL. sativa. The morphological differences betweenL. sativa andL. serriola are too large to consider these two as one species. The descent of the cultivar groups and the four species is described.  相似文献

采用室内土柱淋溶模拟试验,研究了不同施肥水平下无机复合肥中氮和磷在华南赤红壤中的淋失特征.结果表明: 铵态氮、硝态氮和总氮淋失量均随施肥量增加而提高;肥料中氮的淋失率在36.8%~49.2%之间,总氮淋失量(y)与施肥量(x)之间的回归方程为y=0.3667x+66.483(R2=0.992);铵态氮和总氮的淋失主要集中在前5次淋洗,硝态氮的淋失持续时间更长.施肥量对可溶性磷淋失量无显著影响,但颗粒磷和总磷淋失量均随施肥量增加呈上升趋势;肥料磷淋失率在0.002%~0.010%之间,总磷淋失量(y)与施肥量(x)之间的回归方程为y=7e-5x+0.0538(R2=0.931);磷的淋失动态与氮显著不同,为缓慢的持续累积过程.硝态氮与铵态氮淋失量比值和可溶性磷与颗粒磷淋失量比值均随施肥量增加呈下降趋势.  相似文献

The plant growth regulators, gibberellic acid (GA3), ethephon and chlormequat chloride (CCC) were sprayed on young lettuce, cauliflower and bean (Phaseolus vulgaris) plants, which had either been given or not been given a mechanically-induced stress (MIS) treatment. MIS was applied by brushing the plants with paper for 1.5 minutes each day. GA3 increased extension growth of bean and leaf length of lettuce in unbrushed plants as much as in brushed ones. CCC and ethephon were less effective at reducing the height of brushed bean plants compared to unbrushed ones. The effects of CCC on the growth of cauliflower and lettuce plants was not influenced by brushing, whereas unbrushed plants responded more readily to ethephon than did brushed ones. The effects of CCC on growth were generally similar to those of MIS whereas the effects of ethephon were in many ways different to MIS.The results are discussed in relation to the use of PGR and MIS treatments for modifying plant growth.  相似文献

We used a carrot (Daucus carota L. cv. Saint Valery) cell suspension culture as a simplified model system to study the effects of the allelochemical compound coumarin (1,2 benzopyrone) on cell growth and utilisation of exogenous nitrate, ammonium and carbohydrates. Exposure to micromolar levels of coumarin caused severe inhibition of cell growth starting from the second day of culture onwards. At the same time, the presence of 50 mumol/L coumarin caused accumulation of free amino acids and of ammonium in the cultured cells, and stimulated their glutamine synthetase, glutamate dehydrogenase, glucose-6-phosphate dehydrogenase and phosphoenolpyruvate carboxylase activities. Malate dehydrogenase, on the other hand, was inhibited under the same conditions. These effects were interpreted in terms of the stimulation of protein catabolism and/or interference with protein biosynthesis induced by coumarin. This could have led to a series of compensatory changes in the activities of enzymes linking nitrogen and carbon metabolism. Because coumarin seemed to abolish the exponential phase and to accelerate the onset of the stationary phase of cell growth, we hypothesise that such allelochemical compounds may act in nature as an inhibitor of the cell cycle and/or as a senescence-promoting substance.  相似文献

On the other hand, the inductive effect of 1,000 Jm−2 red light was partially reversed by subsequent far-red light irradiation at least twice. These results indicate the involvement of phytochrome in this response. The inductive effect of blue light was repeatedly reversed by subsequent far-red light irradiation, suggesting that the blue-light induction was mainly mediated by phytochrome. Potassium pyroantimonate was used to localize loosely-bound calcium in young ovules of lettuce (Lactuca sativa L.) during megasporogenesis to investigate the relationship between ionically available calcium and megaspore degeneration. At the megasporocyte (megaspore mother cell) stage, few calcium precipitates were located in the ovule. Following meiosis in the megasporocyte, a linear tetrad of four megaspores is formed, with three of the four megaspores degenerating from the micropylar end inward. Only the chalazal-most megaspore continues to develop, becoming the functional megaspore. A decrease in amount of calcium precipitates in the megaspore, particularly in the nucleus, precedes the breakdown of the micropylar megaspores, which subsequently undergo structural disintegration and loss of recognizable cellular features. A partial recovery of calcium precipitates occurs during later degeneration. The functional megaspore retains a consistently higher concentration of calcium precipitates during development, which is retained in the developing embryo sac. This, to our knowledge, is the first report related to calcium dynamics during megaspore degeneration, and may facilitate future research aimed at elucidating the mechanisms of megasporogenesis.  相似文献

Embryos of Avena sativa L. (cv. Moyencourt) show no high temperature dormancy. The dormancy is induced by the presence of endosperm-aleurone part of the seed. Germination of isolated embryos at 30°C can be prevented by ABA and the inhibition is reversed by GA. Inhibitors of GA synthesis also inhibit embryo germination. The embryos of dormant and non-dormant seeds vary greatly in their sensitivity to exogenous ABA. High temperature dormancy of the entire seeds can be relieved by low concentrations of ethanol. On the basis of these facts a hypothetic model is proposed showing how interaction between endogenous GA and ABA-like inhibitory substance, may regulate the high temperature dormancy of the seeds.  相似文献

Summary Symbiotic N2 fixation, NO 3 assimilation and protein accumulation in the shoots were measured simultaneously in alfalfa (Medicago sativa L.) grown in the field or in pots, in order to study how the balance between the two modes of nitrogen nutrition could be influenced by agronomic factors, such as harvest, mineral nitrogen supply and drought stress. During periods of rapid growth, fixation and assimilation may function simultaneously; they are antagonistic at the beginning and at the end of the growth cycle, when the nitrogen requirement of the plant is lower. When nitrogen nutrition does not limit growth, mineral nitrogen supply favours assimilation at the expense of fixation, but does not modify the amount of nitrogen accumulated, which is adjusted to the growth capacity of the plant. After cutting, nitrate assimilation compensated for the decrease in fixation and supplied the plant with the nitrogen required by the regrowth, the proliferation of which determined the fixation recovery. Drought stress decreased N2 fixation much more than NO 3 assimilation. The latter made growth recovery possible when water supply conditions became normal again. These results suggested the existence of an optimum level of nitrate assimilation, which differed depending on the age of the plants and allowed both maximum growth and fixing activity.  相似文献

Root hair formation is induced by low pH in lettuce (Lactuca sativa L. cv. Grand Rapids) seedlings cultured in mineral medium. The role of mineral concentrations in this phenomenon was investigated, especially for manganese. When lettuce seedlings were cultured in media that were deficient in calcium (Ca), manganese (Mn), boron (B) or molybdenum (Mo), morphological changes were induced in roots. Deficiency of other nutrients had little effect on root hair formation. Ca or B deficiency inhibited the growth of the main root and the formation of root hairs, regardless of pH. Mn or Mo deficiency increased root hair formation at pH 6 and suppressed main root growth slightly. In contrast, increasing the Mn concentration suppressed low-pH-induced root hair formation. The Mn content of roots grown at pH 4 was only about 15% of that at pH 6. In contrast, the Mo content of roots grown at low pH was about six times that of roots grown at neutral pH. These results suggest that root hair formation induced by low pH is at least partly mediated by decreased Mn uptake in root cells.  相似文献

Lactuca sativa L. cv. Grand Rapids) seedlings under acidic conditions by a phytochrome-mediated response. Microbeam irradiation of 1 mm root segments with the first (100 Jm−2) and second (1,000 Jm−2) maxima of the fluence response curve for red light induction of root hair initiation indicated that the ca. 5 mm apical portion of 12 mm long roots was the site of photoperception. The root hair-forming portion of the root was situated at a distance of 1.7 mm from the root tip at the time of red light irradiation and extended (at most 1.5 mm) towards the basal end at a later stage of development, irrespective of which portion of the root was irradiated with red light.
