A wheat-Dasypyrum breviaristatum partial amphiploid and its derivatives were analyzed by molecular cytological observation and tested for disease resistance in order to evaluate the potential use of the D. breviaristatum for wheat improvement. A fertility-improved partial amphiploid, TDH-2, was produced from the selfing population of Triticum aestivum cv. Chinese spring (CS)-D. breviaristatum amphiploid. Based on the results obtained from genomic in situ hybridization (GISH) and seed protein electrophoresis, we found the presence of fourteen D. breviaristatum chromosomes and the absence of D genome in TDH-2, indicating that the genomic composition of TDH-2 was AABBV(b)V(b). GISH analysis on BC(1)F(4) progenies of TDH-2xwheat demonstrated that alien D. breviaristatum chromosomes or segments were frequently transmitted. A survey of diseases resistance revealed that powdery mildew resistance from D. breviaristatum was totally expressed, however, the expression of stripe rust resistance from D. breviaristatum was dependent on the wheat background. The comparison of polymerase chain reaction (PCR), which was carried out using molecular marker SCAR(1400) linked to Pm21 D. villosum-derived powdery mildew resistance gene, suggested that D. breviaristatum possessed new resistance gene(s) different from that in D. villosum. The present study showed that the partial amphiploid TDH-2 and its derivatives could serve as novel sources for transfer of disease resistance genes to wheat. 相似文献

Stripe rust (caused by Puccinia striiformis) occurs annually in most wheat-growing areas of the world. Thinopyrum ponticum has provided novel rust resistance genes to protect wheat from this fungal disease. Wheat – Th. ponticum partial amphiploid line 7430 and a substitution line X005 developed from crosses between wheat and 7430 were resistant to
stripe rust isolates from China. Genomic in situ hybridization (GISH) analysis using Pseudoroegneria spicata genomic DNA as a probe demonstrated that the partial amphiploid line 7430 contained ten Js and six J genome chromosomes, and line X005 had a pair of Js-chromosomes. Giemsa-C banding further revealed that both lines 7430 and X005 were absent of wheat chromosomes 6B. The EST
based PCR confirmed that the introduced Js chromosomes belonging to linkage group 6, indicating that line X005 was a 6Js/6B substitution line. Both resistance observation and sequence characterized amplified region (SCAR) markers displayed that
the introduced chromosomes 6Js were responsible for the stripe rust resistances. Therefore, lines 7430 and X005 can be used as a donor in wheat breeding

A collection of 917 accessions of Spanish durum and bread wheat was screened for resistance to leaf rust (Puccinia triticina) under field conditions at three locations. Resistance levels ranged from very low to very high, high susceptibility being most frequent. Relative disease severity (referred to the most susceptible accession = 100%) was lower than 20% in about 6% of the accessions in each location. In the collection most of the lines (84%) displayed a susceptible infection type. A final selection of seven accessions (one of them durum) displaying low severity level in the field and high infection type in a growth chamber was chosen for further studies. High levels of partial resistant with longer latency period and high percentage of early aborted colonies without necrosis were found. They might be used in breeding programmes. 相似文献

Over 100 genes of resistance to rust fungi: Puccinia recondita f. sp. tritici, (47 Lr - leaf rust genes), P. striiformis (18 Yr - yellow rust genes) and P. graminis f. sp. tritici (41 Sr - stripe rust genes) have been identified in wheat (Triticum aestivum L.) and its wild relatives according to recent papers. Sixteen Lr resistance genes have been mapped using restriction fragments length polymorphism (RFLP) markers on wheat chromosomes. More than ten Lr genes can be identified in breeding materials by sequence tagged site (STS) specific markers. Gene Lrk 10, closely linked to gene Lr 10, has been cloned and its function recognized. Available markers are presented in this review. The STS, cleaved amplified polymorphic sequence (CAPS) and sequence characterized amplified regions (SCAR) markers found in the literature should be verified using Triticum spp. with different genetic background. Genetic resistance is the most effective approach to managing wheat leaf rust. The aim of this study was to characterize seedling and adult plant leaf rust resistance of a world wheat collection. Using controlled inoculation with ten races of Puccinia triticina, 14 seedling resistance genes were determined or postulated to be present in the collection. Lr1, Lr3, Lr10 and Lr20 were the most prevalent genes around the world while Lr9, Lr14b, Lr3ka and/or Lr30 and Lr26 were rare. To confirm some gene postulations, the collection was screened with gene-specific molecular markers for Lr1, Lr10, Lr21 and Lr34. Although possessing the Lr1 and/or Lr10 gene-specific marker, 51 accessions showed unexpected high infection types to P. triticina race BBBD. The collection was tested in the field, where rust resistance ranged from nearly immune or highly resistant with severity of 1 % and resistant host response to highly susceptible with severity of 84 % and susceptible host response. The majority of the accessions possessing the adult plant resistance (APR) gene Lr34 had a maximum rust severity of 0–35 %, similar to or better than accession RL6058, a Thatcher-Lr34 near-isogenic line. Many accessions displayed an immune response or a high level of resistance under field conditions, likely as a result of synergy between APR genes or between APR and seedling resistance genes. However, accessions with three or more seedling resistance genes had an overall lower field severity than those with two or fewer. Immune or highly resistant accessions are potential sources for improvement of leaf rust resistance. In addition, some lines were postulated to have known but unidentified genes/alleles or novel genes, also constituting potentially important sources of novel resistance. 相似文献

Components of resistance conferred by the Lr46 gene, reported as causing "slow rusting" resistance to leaf rust in wheat, were studied and compared with the effects of Lr34 and genes for quantitative resistance in cv. Akabozu. Lr34 is a gene that confers non-hypersensitive type of resistance. The effect of Lr46 resembles that of Lr34 and other wheats reported with partial resistance. At macroscopic level, Lr46 produced a longer latency period than observed on the susceptible recurrent parent Lalbahadur, and a reduction of the infection frequency not associated with hypersensitivity. Microscopically, Lr46 increased the percentage of early aborted infection units not associated with host cell necrosis and decreased the colony size. The effect of Lr46 is comparable to that of Lr34 in adult plant stage, but in seedling stage its effect is weaker than that of Lr34. 相似文献

Two partial amphiploid lines, B113 (32 plants) and B21 (13 plants), derived from a wheat-Psathyrostachys huashanica intergeneric cross were characterized by Giemsa C-banding and SDS-PAGE and evaluated for stripe rust resistance. All 15 partial
amphiploid plants were aneuploids with either 50 (8 plants), 51 (6 plants) or 54 (1 plant) chromosomes. Some showed regular
meiosis and all the P. huashanica chromosomes were included, although not in a single plant. Of 45 plants 34 showed specific bands on SDS-PAGE representing
high molecular weight glutenin subunit (HMW-GS) and 41 had bands representing P. huashanica low molecular weight glutenin subunit (LMW-GS), including two new subunits. All 45 plants were highly resistant (10) or immune
(35) to stripe rust mixed races CYR-30, CYR-31, Shuiyuan 7 and Shuiyuan 14. These amphiploid plants could be useful germplasm

molecular markers closely linked to Lr41, a set of near-isogenic lines (NILs) of the hard winter wheat cultivar Century were developed through backcrossing. A population
of 95 BC3F2:6 NILs were evaluated for leaf rust resistance at both seedling and adult plant stages and analyzed with simple sequence repeat
(SSR) markers using bulked segregant analysis. Four markers closely linked to Lr41 were identified on chromosome 2DS; the closest marker, Xbarc124, was about 1 cM from Lr41. Physical mapping using Chinese Spring nullitetrasomic and ditelosomic genetic stocks confirmed that markers linked to Lr41 were on chromosome arm 2DS. Marker analysis in a diverse set of wheat germplasm indicated that primers BARC124, GWM210, and
GDM35 amplified polymorphic bands between most resistant and susceptible accessions and can be used for marker-assisted selection

Chromosome 7E from Lophopyrum ponticum carries a valuable leaf rust resistant gene designated Lr19. This gene has not been widely used in common wheat breeding because of linkage with the yellow pigment gene Y. This gene tints flour yellow, reducing its appeal in bread making. However, a high level of yellow pigment is desirable in durum wheat breeding. We produced 97 recombinant chromosomes between L. ponticum transfer 7D.7E#1 and its wheat homoeologues, using the ph1b mutation that promotes homoeologous pairing. We characterized a subset of 37 of these lines with 11 molecular markers and evaluated their resistance to leaf rust and the abundance of yellow pigment. The Lr19 gene was mapped between loci Xwg420 and Xmwg2062, whereas Y was mapped distal to Xpsr687, the most distal marker on the long arm of chromosome 7. A short terminal 7EL segment translocated to 7A, including Lr19 and Y (line 1-23), has been transferred to durum wheat by backcrossing. The presence of this alien segment significantly increased the abundance of yellow pigment. The Lr19 also conferred resistance to a new durum leaf rust race from California and Mexico that is virulent on most durum wheat cultivars. The new durum lines with the recombinant 7E segment will be useful parents to increase yellow pigment and leaf rust resistance in durum wheat breeding programs. For the common wheat breeding programs, we selected the recombinant line 1-96, which has an interstitial 7E segment carrying Lr19 but not Y. This recombinant line can be used to improve leaf rust resistance without affecting flour color. The 7EL/7DL 1-96 recombinant chromosome did not show the meiotic self-elimination previously reported for a 7EL/7BL translocation. 相似文献

The genetic control of leaf rust resistance has been investigated by test crosses and the test-clone method in the rye samples Malysh 72-2, Chulpan 3, Immunnaya 4, Yaroslavna 3, Lovashpatonae 2, Gotor 2, Talovskaya 12-2, Braunrostresisten 2, Orlovskaya 9-2, 3098/18, and 4001/10. It has been found that this trait is controlled by a set of nonidentical oligogenes. Dominant gene Lr6 controls leaf rust resistance in Chulpan 3 and Immunnaya 4; Lr5, in Malysh 72-2; and Lr-c, in Yaroslavna 3. Test crosses have shown that genes Lr-a and Lr-b differ from the genes determining the resistance in Gotor 2 and Braunrostresisten 2. In German samples 3098/18 and 4001/10, the resistance to the Petersburg population of the pathogen is controlled by nonallelic genes Lr-a and Lr-b. It has been found that gene Lr-b is not identical to the resistance gene of Lovashpatonae 2, and Lr-a is not identical to Lr-c. In all samples, resistance to the leaf rust population is controlled by dominant genes from the stage of seedlings till maturity. Highly efficient Lr genes are present in the samples Malysh 72-2, Chulpan 3, Immunnaya 4, Yaroslavna 3, Lovashpatonae 2, Gotor 2, Talovskaya 12-2, 3098/18, and 4001/10. In the present study, F4-derived SSD population, developed from a cross between Australian cultivars ‘Schomburgk’ and ‘Yarralinka’, was used to identify
molecular markers linked to rust resistance genes Lr3a and Sr22. A total of 1,330 RAPD and 100 ISSR primers and 33 SSR primer pairs selected ob the basis of chromosomal locations of these
genes were used. The ISSR marker UBC 840540 was found to be linked with Lr3a in repulsion at a distance of 6.0 cM. Markers cfa2019 and cfa2123 flanked Sr22 at a distance of 5.9 cM (distal) and 6.0 cM (proximal), respectively. The use of these markers in combination would predict

The partial resistance to leaf rust in barley is a quantitative resistance that is not based on hypersensitivity. To map
the quantitative trait loci (QTLs) for partial resistance to leaf rust, we obtained 103 recombinant inbred lines (RILs) by
single-seed descent from a cross between the susceptible parent L94 and the partially resistant parent Vada. These RILs were
evaluated at the seedling and adult plant stages in the greenhouse for the latent period (LP) of the rust fungus, and in the
field for the level of infection, measured as area under the disease progress curve (AUDPC). A dense genetic map based on
561 AFLP markers had been generated previously for this set of RILs. QTLs for partial resistance to leaf rust were mapped
using the “Multiple Interval Mapping” method with the putative QTL markers as cofactors. Six QTLs for partial resistance were
identified in this population. Three QTLs, Rphq1, Rphq2 and Rphq3, were effective at the seedling stage and contributed approximately 55% to the phenotypic variance. Five QTLs, Rph2, Rphq3, Rphq4, Rphq5, and/or Rphq6 contributed approximtely. 60% of the phenotypic variance and were effective at the adult plant stage. Therefore, only the
QTLs Rphq2 and Rhpq3 were not plant-stage dependent. The identified QTLs showed mainly additive effects and only one significant interaction was
detected, i.e. between Rphq1 and Rphq2. The map positions of these QTLs did not coincide with those of the race-specific resistance genes, suggesting that genes
for partial resistance and genes for hypersensitive resistance represent entirely different gene families. Also, three QTLs
for days to heading, of which two were also involved in plant height, were identified in the present recombinant inbred population.
These QTLs had been mapped previously on the same positions in different populations. The perspectives of these results for
breeding for durable resistance to leaf rust are discussed.

Partial resistance is generally considered to be a durable form of resistance. In barley, Rphq2, Rphq3 and Rphq4 have been identified as consistent quantitative trait loci (QTLs) for partial resistance to the barley leaf rust pathogen
Puccinia hordei. These QTLs have been incorporated separately into the susceptible L94 and the partially resistant Vada barley genetic backgrounds
to obtain two sets of near isogenic lines (NILs). Previous studies have shown that these QTLs are not effective at conferring
disease resistance in all stages of plant development. In the present study, the two sets of QTL–NILs and the two recurrent
parents, L94 and Vada, were evaluated for resistance to P. hordei isolate 1.2.1 simultaneously under greenhouse conditions from the first leaf to the flag leaf stage. Effect of the QTLs on
resistance was measured by development rate of the pathogen, expressed as latency period (LP). The data show that Rphq2 prolongs LP at the seedling stage (the first and second leaf stages) but has almost no effect on disease resistance in adult
plants. Rphq4 showed no effect on LP until the third leaf stage, whereas Rphq3 is consistently effective at prolonging LP from the first leaf to the flag leaf. The changes in the effectiveness of Rphq2 and Rphq4 happen at the barley tillering stage (the third to fourth leaf stages). These results indicate that multiple disease evaluations
of a single plant by repeated inoculations of the fourth leaf to the flag leaf should be conducted to precisely estimate the
effect of Rphq4. The present study confirms and describes in detail the plant development-dependent effectiveness of partial resistance genes
and, consequently, will enable a more precise evaluation of partial resistance regulation during barley development. 相似文献

Growing resistant wheat varieties is a key method of controlling two important wheat diseases, leaf rust and stripe rust. We analyzed quantitative trait loci (QTL) to investigate adult plant resistance (APR) to these rusts, using 141 F5 RILs derived from the cross ‘Avocet-YrA/Francolin#1’. Phenotyping of leaf rust resistance was conducted during two seasons at Ciudad Obregon, Mexico, whereas stripe rust was evaluated for two seasons in Toluca, Mexico, and one season in Chengdu, China. The genetic map was constructed with 581 markers, including diversity arrays technology and simple sequence repeat. Significant loci for reducing leaf rust severity were designated QLr.cim-1BL, QLr.cim-3BS.1, QLr.cim-3DC, and QLr.cim-7DS. The six QTL that reduced stripe rust severity were designated QYr.cim-1BL, QYr.cim-2BS, QYr.cim-2DS, QYr.cim-3BS.2, QYr.cim-5AL, and QYr.cim-6AL. All loci were conferred by Francolin#1, with the exception of QYr.cim-2DS, QYr.cim-5AL, and QYr.cim-6AL, which were derived from Avocet-YrA. Closely linked markers indicated that the 1BL locus was the pleiotropic APR gene Lr46/Yr29. QYr.cim-2BS was a seedling resistance gene designated as YrF that conferred intermediate seedling reactions and moderate resistance at the adult plant stage in both Mexican and Chinese environments. Significant additive interactions were detected between the six QTL for stripe rust, but not between the four QTL for leaf rust. Stem rust resistance gene Sr43, transferred into common wheat (Triticum aestivum) from Thinopyrum ponticum, is an effective gene against stem rust Ug99 races. However, this gene has not been used in wheat breeding because it is located on a large Th. ponticum 7el2 chromosome segment, which also harbors genes for undesirable traits. The objective of this study was to eliminate excessive Th. ponticum chromatin surrounding Sr43 to make it usable in wheat breeding. The two original translocation lines KS10-2 and KS24-1 carrying Sr43 were first analyzed using simple sequence repeat (SSR) markers and florescent genomic in situ hybridization. Six SSR markers located on wheat chromosome arm 7DL were identified to be associated with the Th. ponticum chromatin in KS10-2 and KS24-1. The results confirmed that KS24-1 is a 7DS·7el2L Robertsonian translocation as previously reported. However, KS10-2, which was previously designated as a 7el2S·7el2L-7DL translocation, was identified as a 7DS-7el2S·7el2L translocation. To reduce the Th. ponticum chromatin carrying Sr43, a BC2F1 population (Chinese Spring//Chinese Spring ph1bph1b*2/KS10-2) containing ph1b-induced homoeologous recombinants was developed, tested with stem rust, and genotyped with the six SSR markers identified above. Two new wheat lines (RWG33 and RWG34) carrying Sr43 on shortened alien chromosome segments (about 17.5 and 13.7 % of the translocation chromosomes, respectively) were obtained, and two molecular markers linked to Sr43 in these lines were identified. Pearl millet [Pennisetum glaucum (L.) R.Br.] is a warm-season grass used for food, feed, fodder and forage, primarily in countries of Africa and India but
grown around the world. The two most-destructive diseases to pearl millet in the United States are rust (caused by Puccinia substriata var. indica) and pyricularia leaf spot (caused by Pyricularia grisea). Genes for disease resistance to both pathogens have been transferred into agronomically acceptable forage and grain cultivars.
A study was undertaken to identify molecular markers for three rust loci and one pyricularia resistance locus. Three segregating
populations were screened for RAPDs using random decamer primers and for RFLPs using a core set of probes detecting single-copy
markers on the pearl millet map. The rust resistance gene Rr

from the pearl millet subspecies P. glaucum ssp. monodii was linked 8.5 cM from the RAPD OP-G8350. The linkage of two RFLP markers, Xpsm108 (15.5 cM) and Xpsm174 (17.7 cM), placed the Rr
1
gene on linkage-group 3 of the pearl millet map. Rust resistance genes from both Tift 89D2 and ICMP 83506 were placed on linkage-group 4 by determining genetic linkage to the RFLP marker Xpsm716 (4.9 and 0.0 cM, respectively). Resistance in ICMP 83506 was also linked to the RFLP marker Xpsm306 (10.0 cM), while resistance in Tift 89D2 was linked to RAPD markers OP-K19350 (8.8 cM) and OP-O8350 (19.6 cM). Fragments from OP-K19 and OP-O8 in the ICMP 83506 population, and Xpsm306 in the Tift 89D2 population, were monomorphic. The Canadian common wheat (Triticum aestivum L.) cultivar 'Roblin' is resistant to both leaf rust (Puccinia recondita Rob. ex. Desm.) and stem rust (Puccinia graminis Pers. f. sp. tritici Eriks. and E. Henn.). To study the genetics of this resistance, 'Roblin' was crossed with 'Thatcher', a leaf rust susceptible cultivar, and RL6071, a stem rust susceptible line. A set of F6 random lines was developed from each cross. The random lines and the parents were grown in a field rust nursery artificially inoculated with a mixture of P. recondita and P. graminis isolates and scored for rust reaction. The same material was tested with specific races of leaf rust and stem rust. These data indicated that 'Roblin' has Lr1, Lr10, Lr13, and Lr34 for resistance to P. recondita and Sr5, Sr9b, Sr11, and possibly Sr7a and Sr12 for resistance to P. graminis. In a 'Thatcher' background, the presence of Lr34 contributes to improve stem rust resistance, which appears also to occur in 'Roblin'. 相似文献

Molecular markers for four leaf rust resistance genes introgressed into wheat from wild relatives. 总被引:12,自引:0,他引:12
Near-isolines carrying four different genes for resistance to leaf rust were used to find linked molecular markers for these genes. Clones used to detect polymorphism were selected on the basis of the reported chromosomal location of the resistance genes. Both Lophopyron-derived resistance genes, Lr19 and Lr24, cosegregated with eight molecular markers assigned to chromosomes 7DL and 3DL, respectively. One clone cosegregated with Lr9 and two closely linked RFLP markers were found for Lr32, mapping at 3.3 +/- 2.6 and 6.9 +/- 3.6 cM from the resistance gene. The Lophopyron-chromatin segment in isolines carrying chromosomes 7E (Lr19) and 3E (Lr24) replaced a large portion of chromosome 7D and the distal portion of chromosome 3D, respectively. Clones assigned to these chromosomes on the basis of aneuploid analysis hybridized to 7E and 3E segments, thus confirming cytological results that these introgressed segments represent homoeologous chromosomes. The linked RFLP markers could be used to identify the resistance genes and generate new combinations in breeding populations, especially in the absence of disease in the environment or when virulence is lacking. 相似文献
