Nubbins of the coral Acropora aspera were artificially bleached and nitrogen fixation (acetylene reduction) rates were measured on the developing epilithic communities. Seasonal comparisons were made between corals that died in summer of heat stress and corals that died in winter from natural cold stress. Rates of acetylene reduction from artificially bleached corals peaked at 26.66 nmol cm−2 h−1 2 weeks after summer mortality, while rates from natural winter mortality peaked at 18.07 nmol cm−2 h−1 12 days after coral death. Comparative rates of acetylene reduction taken from live corals and coral rubble ranged between 0.56 and 1.16 nmol cm−2 h−1, and 0.15 and 12.77 nmol cm−2 h−1, respectively. N2-fixation rates from dead corals were up to 30 times greater than those measured on live corals. The observed increase in N2-fixation from dead corals may increase the availability of nitrogen for use in trophic processes within the reef for an extended period following the initial mortality event. If the spatial scale over which coral mortality has occurred in past thermal bleaching events is considered the ramifications of such an increase may be substantial.  相似文献

Nitrogen fixation was measured by the acetylene reduction method in a high Arctic ecosystem at Kongsfjorden, Spitsbergen (79°N, 12°E). The most important source of biologically fixed nitrogen was found in cyanobacteria either as free living colonies ofNostoc sp. in wet unvegetated or sparsely vegetated grounds or growing as epiphytes on bryophytes. Fixation associated with plant roots or in soil and peat samples had little or no significance for nitrogen input to the ecosystem. The ability to support an epiphytic flora of nitrogen-fixing cyanobacteria varied greatly between bryophyte species.Calliergon richardsonii andSanionia uncinata seemed especially well adapted for harbouring epiphytic cyanobacteria, but the extent of nitrogen fixation varied with the growing location. The rate of nitrogen fixation was greatly influenced by grazing by geese. In a geese-grazing area values were found with a maximum of 693.6±1.5 nmol C2H4 h−1 g (dry weight)−1 while the maximum value for ungrazed areas was 65.3±16.6 nmol C2H4 h−1 g (dry weight)−1. In the grazed area cyanobacteria were also found fixing nitrogen epiphytically on grass. The high plant productivity, supporting heavy grazing, clearly indicates an effective transfer of fixed nitrogen to the plant community. Under cliffs harbouring colonies of birds, the biological nitrogen fixation was inhibited by bird droppings.  相似文献

Increases of 23- (5.6 mmol acetylene reduced mg dry wt–1) and 16- (4 mmol acetylene reduced mg dry wt–1) fold in nitrogenase activity and 12- (671 l H2 mg dry wt–1 h–1) and 6- (349 l mg dry wt–1 h–1) fold in H2 photoproduction in Rhodopseudomonas palustris JA1 over 24 h were achieved with pyrazine 2-carboxylate (3 mM) and 3-picoline (3 mM), respectively, and were higher than earlier reports of enhancement (1.5 to 5- fold) in biological H2 production using various alternative methods.  相似文献

This study reports the effect of salinity and inoculation on growth, ion uptake and nitrogen fixation byVigna radiata. A soil ECe level of 7.5 dS m−1 was quite detrimental causing about 60% decline in dry matter and grain yield of mungbean plants whereas a soil ECe level of 10.0 dS m−1 was almost toxic. In contrast most of the studied strains of Rhizobium were salt tolerant. Nevertheless, nodulation, nitrogen fixation and total nitrogen concentration in the plant was drastically affected at high salt concentration. A noticeable decline in acetylene reduction activity occurred when salinity level increased to 7.5 dS m−1.  相似文献

Batch cultivation of Ralstonia eutropha NRRL B14690 attained 21 g biomass l−1 and 9.4 g poly(β-hydroxybutyrate) l−1 (0.45 g PHB g−1 dry wt−1) in 60 h. Repeated batch operation (empty-and-fill protocol) to remove 20% (v/v) of the culture broth and to supplement an equal volume of fresh media resulted in 49 g biomass l−1 and 25 g PHB l−1 (0.51 g PHB g−1 dry wt−1) with an overall productivity of 0.42 g PHB l−1 h−1 in 67 h. In the two cycles of repeated batch fermentation there was a 3-fold increase in productivity as compared to batch.  相似文献

The effects of changes in diurnal light patterns, salinity, and phosphorus on nitrogen fixation (as measured by acetylene reduction) by Nodularia spumigena Mertens were examined. As well, the effects of added inorganic nitrogen on growth, nitrogen fixation and heterocyt frequencies, and changes in nitrogen fixation and heterocyst frequencies during the growth cycle of Nodularia in cultures were determined.The diurnal pattern of nitrogenase activity in Nodularia was primarily light-induced, though dark activity did occur. Nitrogenase activity following a period of darkness exceeded the normal light rate (> 90 compared to 50 nmol · C2H2 reduced · ml–1 · h–1). Nitrogen fixation was reduced by high and very low salinities (5 to 10 was the optimum range), and added phosphorus stimulated nitrogenase in P-starved cells. Added nitrogen (ammonium or nitrate) had no effect on the growth of Nodularia, but in short term studies, ammonium completely inhibited nitrogenase activity. Heterocyst frequencies were greatest in the log phase of growth (to 40 per mm). During stationary phase, nitrogenase activity was negligable.  相似文献

Summary The nitrogen fixation rate in a 4-year-old stand of the woody legumeLeucaena leucocephala (Lam.) de Wit. was estimated in the field at a rather dry site in Tanzania by use of an acetylene reduction technique. The diurnal mean value during April–May was 35 nmol C2H4 mg–1 (dry weight) nodules h–1, with a variation between 22±8 and 48±12 nmol C2H4 mg–1 (dry weight) nodules h–1 in early morning and at midday, respectively. The nodule biomass was determined by auger sampling to be 51±16 kg (dry weight) ha–1. Most of the nodules were found at the 10–30 cm soil depth level. A rough calculation of the amount of nitrogen fixed annually arrived at 110±30 kg ha–1. The results give strong support for the use ofL. leucocephala for soil enrichment in less humid areas of tropical Africa.  相似文献

In this work, we characterized an ecto-ATPase activity in intact mycelial forms of Fonsecaea pedrosoi, the primary causative agent of chromoblastomycosis. In the presence of 1 mM EDTA, fungal cells hydrolyzed adenosine-5′-triphosphate (ATP) at a rate of 84.6 ± 11.3 nmol Pi h−1 mg−1 mycelial dry weight. The ecto-ATPase activity was increased at about five times (498.3 ± 27.6 nmol Pi h−1 mg−1) in the presence of 5 mM MgCl2, with values of V max and apparent K m for Mg-ATP2−corresponding to 541.9 ± 48.6 nmol Pi h−1 mg−1 cellular dry weight and 1.9 ± 0.2 mM, respectively. The Mg2+-stimulated ecto-ATPase activity was insensitive to inhibitors of intracellular ATPases such as vanadate (P-ATPases), bafilomycin A1 (V-ATPases), and oligomycin (F-ATPases). Inhibitors of acid phosphatases (molybdate, vanadate, and fluoride) or alkaline phosphatases (levamizole) had no effect on the ecto-ATPase activity. The surface of the Mg2+-stimulated ATPase in F. pedrosoi was confirmed by assays in which 4,4′-diisothiocyanostylbene-2,2′-disulfonic acid (DIDS), a membrane impermeant inhibitor, and suramin, an inhibitor of ecto-ATPase and antagonist of P2 purinoreceptors. Based on the differential expression of ecto-ATPases in the different morphological stages of F. pedrosoi, the putative role of this enzyme in fungal biology is discussed.  相似文献

Variations in nitrogen fixation (acetylene reduction) by Nodularia spumigena blooms in the Peel-Harvey estuarine system were examined with respect to spatial (sampling station location, and depth) and temporal (seasonal and diurnal) distribution. The annual contributions of nitrogen fixation by the blooms to the nitrogen budget of the estuary were estimated to range from 309 to 713t. Contributions by nitrogen fixation were similar to the riverine inputs in the Harvey Estuary, but lower in the Peel Inlet.The Harvey Estuary had higher biomass and total fixation rates (to 0.4 nmol C2H2 · ml–1 h–1), but the heterocyst nitrogen fixation rates were greater in the Peel Inlet (to 9 × 10–1 nmol C2H2 · heterocyst–1 · h–1). Nitrogen fixation decreased with depth in response to light, though other factors also appeared to be involved. The rates of fixation decreased concurrently with increasing bloom age, total soluble inorganic nitrogen and salinities. Maximum daily fixation rates occurred in the early morning.  相似文献

Nitrogen fixation by cyanobacteria in a moss community on East Ongul Island (69°00'S 39°35'E), Antarctica was investigated using the acetylene reduction method. The mean acetylene reduction rate at 10°C and 200 μE·m−2·s−1 photosynthetically active radiation was 7.12 nmol C2H4 per square centimeter of moss community per hour. The effects of temperature, radiation, desiccation and rehydration on the acetylene reduction rates were examined. A simple predictive model was constructed in order to estimate the amount of nitrogen fixed in the field. Using this model, the daily amount of nitrogen fixation was calculated from microclimatic data (temperature and radiation) measured in the experimental field at Syowa Station on East Ongul Island between 1983 and 1984. The cumulative amount of nitrogen fixation in the growing season during this period was estimated to be 329 mg N per square meter of moss community. It is suggested that nitrogen fixation by cyanobacteria in the moss community is important as a nitrogen source for the community growth on East Ongul Island.  相似文献

Summary Previous investigations indicated some forage grass roots in Texas are heavily colonized with N2-fixing bacteria. The most numerous N2-fixing bacteria were in the genera Klebsiella and Enterobacter. In the present investigation inoculation experiments were conducted using 18 isolates of these bacteria to determine if a N2-fixing association could be established between the bacteria and the grassesCynodon dactylon andPanicum coloratum. Plants were grown in soil for approximately 5 months in a greenhouse and were measured periodically for dry matter, nitrogen accumulation, and acetylene reduction activity. Results of the investigation indicated that 25% of the plant-soil systems were active in acetylene reduction and the activity was high enough to indicate agronomically significant quantities of N2 were being fixed (>8kg N ha−1). However, plant systems extrapolated to fix>8 kg N ha−1 contained less nitrogen and accumulated less dry matter than plants less active in acetylene reduction. Inocula could not be re-isolated from healthy grass roots indicating that the N2-fixing activity may have not have been closely assiciated with plant roots. Future research is needed to determine factors limiting colonization of grass roots.  相似文献

Nitrogenase activity (acetylene reduction activity) was found to occur universally in the Cyperus papyrus swamp in Lake Naivasha. Low rates of acetylene reduction activity (0.9–104.9 nmol C2H4 g d.wt. roots-1 h-1) were associated with excised roots of C. papyrus but higher rates of activity (89.0–280.4 nmol C2H4 g d.wt. roots-1 h-1) were associated with intact root systems of the plant. It was estimated that nitrogen fixation associated with young roots alone could supply about 26% of the nitrogen requirements of growing papyrus plants. Acetylene reduction activity in the lake bottom sediments was generally low and associated with adjacent papyrus stands. Plate counts of putative aerobic and facultatively anaerobic N2-fixing bacteria associated with papyrus roots showed the presence of high numbers of diazotrophs (5.4 × 106 CFU g d.wt. roots-1). Fewer numbers of N2-fixing bacteria were detected in the sediments (1.9 × 103-3.2 × 104 CFU g d.wt. sediment-1).  相似文献

Summary Tillage has been shown to affect the uptake of phosphorus (P) and yield of soybeans, [Glycine max (L.) Merr.], but there is little information concerning the effects of P fertilization on nitrogen (N2) fixation in soybeans under no-tillage. Two field experiments were conducted in 1980 and 1981 to determine the effects of soil P on N2 fixation under no-tillage and to study the interaction of P fertilization and tillage of N2 fixation, nutrient uptake, and yield of soybeans. In Exp. I, P was applied in 1977 at five rates up to 384 kg P ha−1 and the effects of residual soil P were evaluated in 1980 and 1981 under no-tillage management. Nitrogen fixation rates, as measured by acetylene reduction assay, were significantly affected by soil P in Exp. I, but the assay proved to be a poor technique for estimating total plant N in these tests. Acetylene reduction rates and plant P increased rapidly as soil P increased from 2 to 20 mg kg−1, with little additional increase above 20 mg P kg−1. In Exp. II, rates (0, 32, 64, and 128 kg P ha−1) and time (fall, spring and fall plus spring) of P application were compared under conventional tillage and no tillage. However, plant P increased with increasing levels of applied P. Applied P had no affect on acetylene reduction rates but rates were greater for no-tillage than conventional tillage at the V9 and R5 stages of growth in 1981. Plant uptake of P was more efficient under no-tillage than under conventional tillage in 1980 and 1981. Application of 64 kg P ha−1 under no-tillage resulted in equivalent plant P levels as the 128 kg P ha−1 applied under conventional tillage.  相似文献

The acetylene reduction method was used to measure nitrogen fixation in the phyllosphere of attached leaves of different phorophytes under natural conditions in a premontane rain forest in Costa Rica. Maximum rates of nitrogen fixation (26 ng N · cm−2 leaf area · h−1) – mainly due to the activity of two species of Scytonema (Cyanobacteria) – were measured in the rainy season in bright sunlight. Rates of nitrogen fixation were correlated with the leaf area covered by Scytonema. In periods without precipitation the fixation activity decreased to zero within 2–3 days. As long as the epiphylls were sufficiently supplied with water, other microclimatic factors like temperature and light intensity also influenced nitrogen fixation rates, but to a lesser extent. Relative humidity and species of phorophyte showed no direct influence. It was concluded that the most important factor for nitrogen fixation in the phyllosphere was the availability of liquid water. Linking these results to meteorological data, the input of nitrogen by biological nitrogen fixation in the phyllosphere in the investigation area was estimated to be as much as 1.6 ± 0.8 kg N · ha−1 · year−1 per unit of leaf area index (LAI). For an LAI of 2 for the understory the nitrogen input would vary between 2 and 5 kg N · ha−1 · year−1. This work also demonstrates the importance of detailed knowledge of variation in microclimate throughout the year as a basis for extrapolation of the annual nitrogen input. To maximize and sustain the productivity ofMonascus pigments, various environmental and nutritional parameters, such as the initial moisture content, pH, inoculum size, sample size, and nutrient supplement, that influence pigment production were evaluated in solid-state cultures as follows: initial moisture content, 50%; pH, 6.0; inoculum size 1 x 104 spore cells (grams of dry solid substrate)−1; sample size, 300 g. All supplementary nutrients (carbon, nitrogen, and mineral sources) added has inhibitory effects on the cell growth and red pigment production. In open tray culture the maximum biomass yield and specific productivity of red pigments were 223 mg DCW (grams of initial dry substrate)−1 and, 47.6 OD500 (DCW grams)−1 h−1, respectively.  相似文献

Summary The seasonal patterns of nodulation, acetylene reduction, nitrogen uptake and nitrogen fixation were studies for 11 pigeonpea cultivars belonging to different maturity groups grown on an Alfisol at ICRISAT Center, Patancheru, India. In all cultivars the nodule number and mass increased to a maximum around 60–80 days after sowing and then declined. The nodule number and mass of medium- and late-maturing cultivars was greater than that of early-maturing cultivars. The nitrogenase activity per plant increased to 60 days after sowing and declined thereafter, with little activity at 100 days when the crop was flowering. At later stages of plant growth nodules formed down to 90 cm below the soil surface but those at greater depth appeared less active than those near the surface. All the 11 cultivars continued to accumulate dry matter until 140 days, with most biomass production by the late-maturing cultivars (up to 11 t ha−1) and least by the early-maturing determinate cultivars (4 t ha−1). Total nitrogen uptake ranged from 69 to 134 kg ha−1. Nitrogen fixation by pigeonpea was estimated as the difference in total nitrogen uptake between pigeonpea and sorghum and could amount to 69 kg N ha−1 per season, or half the total nitrogen uptake. Fixation by pigeonpea increased with crop duration, but there were differences within each maturity group. The limitations of the methods used for estimating N2 fixation by pigeonpea are discussed. Abstract A total of 285 strains of Azospirillum were isolated from soils from seven geographic regions in New South Wales, Australia, using an immunomagnetic separation procedure which does not select strains according to their nitrogen-fixing ability. By combining amplification and restriction analysis of 16S rDNA (ARDRA) patterns with serological, morphological and biochemical results, we found that almost all isolates were A. brasilense and A. lipoferum. There was wide variation in the nitrogenase (acetylene reduction) activity of isolates grown in nitrogen-free, semisolid medium, with differences in average activities between regions. Isolates with zero or negligible nitrogenase activity were found in samples from only two regions, one of which had two out of 26 strains with no activity. Representative isolates, having the highest, the lowest, and intermediate nitrogen fixation rates for each site, were used to inoculate the roots of wheat plants in a model system. Most of the isolates, in association with wheat roots, reduced between 1 and 5 nmol C2H4· mg dry root−1· day−1, but certain strains gave considerably higher activities. The rank order of nitrogen fixation activity on wheat roots did not correlate well with that of nitrogen fixation in pure culture; some strains that fixed nitrogen vigorously in pure culture had low rates of fixation on roots, and vice versa. This inconsistency could not be explained by variations in the root colonizing ability of different strains. However, isolates of A. lipoferum had a higher average nitrogenase activity than A. brasilense, both in Nfb medium and in association with wheat roots. The majority of the most active nitrogen fixers were A. lipoferum. When wheat plants were inoculated with mixtures of two or four strains, nitrogen fixation rates were generally between the rates for the component strains when inoculated individually. Traditionally it has been thought that most boreal forest communities lack a significant input of biologically fixed nitrogen. Recent discoveries of nitrogen fixation by cyanobacteria associated with mosses have resulted in a re-evaluation of this view. While it is recognized that rates of nitrogen fixation in mosses can be highly variable, there is little understanding as to why this occurs. I monitored nitrogen fixation, using acetylene reduction, in wet lowland and dry upland boreal forest communities, in central Canada, over a growing season. At the peak of nitrogen fixation in mid summer, Sphagnum capillifolium had an 11 times higher rate of fixation than Pleurozium schreberi. Variation in canopy openness and precipitation had no effect on rates of fixation over the growing season. In P. schreberi fixation rates did not vary between sites. Temperature had a positive effect on fixation rates in both S. capillifolium and P. schreberi, but the effect was 4 times more pronounced in S. capillifolium. Seasonal rates of nitrogen fixation were estimated at 193 mg N m−2 for S. capillifolium and 23 mg N m−2 for P. schreberi. With moderate increases in climate warming, predicted increases in nitrogen fixation in S. capillifolium are sufficient to raise its decomposition rate. Increased temperatures may therefore act synergistically to change boreal systems from a sink to a source of carbon.  相似文献

The effect of agitation and aeration on the growth and antibiotic production by Xenorhabdus nematophila YL001 grown in batch cultures were investigated. Efficiency of aeration and agitation was evaluated through the oxygen mass transfer coefficient (K L a). With increase in K L a, the biomass and antibiotic activity increased. Activity units of antibiotic and dry cell weight were increased to 232 U ml−1 and 19.58 g l−1, respectively, productivity in cell and antibiotic was up more than 30% when K L a increased from 115.9 h−1 to 185.7 h−1. During the exponential growth phase, DO concentration was zero, the oxygen supply was not sufficient. So, based on process analysis, a three-stage oxygen supply control strategy was used to improved the DO concentration above 30% by controlling the agitation speed and aeration rate. The dry cell weight and activity units of antibiotic were further increased to 24.22 g l−1 and 249 U ml−1, and were improved by 24.0% and 7.0%, compared with fermentation at a constant agitation speed and a constant aeration rate (300 rev min−1, 2.5 l min−1).  相似文献

Rates of nitrogen fixation in seagrass beds (Zostera capricorni) were determined with15N and reduction of acetylene in intact cores of sediment and seagrass. There was good agreement in the results from the two techniques, with a molar ratio of 3∶1.9 ethylene: ammonia produced. Fixed nitrogen was rapidly utilized by the plants, with significant amounts of15N found in the roots and rhizomes and 50% of fixed15N apparently translocated to the leaves. Rates of fixation were high in summer (25 to 40 mg N m−2 day−1) and lower in winter (10 mg N m−2 day−1) and were estimated to supply between one-third and one-half of the nitrogen requirements of the seagrass. Rates of nitrogen fixation were greater in the light than in the dark, and in cores of intact seagrass than in defoliated cores, indicating that the bacteria were dependent on organic compounds secreted by the plants.  相似文献
