Subunit interactions among the chloroplast ATP synthase subunits were studied using the yeast two-hybrid system. Various pairwise combinations of genes encoding a, p, y, 8 and e subunits of Spinach ATP synthase fused to the binding domain or activation domain of GAL4 DNA were introduced into yeast and then expression of a reporter gene encoding p-galactosidase was detected. Of all the combinations, that of y and e subunit genes showed the highest level of reporter gene expression, while those of a and p, a and e, p and e and p and 8 induced stable and significant reporter gene expression. The combination of 8 and e as well as that of 8 and y induced weak and unstable reporter gene expression. However, combinations of a and y, p and y and a and 8 did not induce reporter gene expression. These results suggested that specific and strong interactions between y and e, a and p, a and e, p and e and p and 8 subunits, and weak and transient interactions between 8 and e and 8 and y subunits occurred in the yeast  相似文献

内蒙古白绒山羊若干数量性状遗传参数的研究   总被引:12,自引:2,他引:10  
本研究应用女母回归法、公畜内女母回归法、半同胞相关法和单元内半同胞相关法对内蒙古阿尔巴斯白绒山羊的产绒量、绒厚、毛长、体重、绒伸直长度和细度等六个性状的遗传参数进行了估测。结果表明:(1)绒量遗传力介于0.26~0.45之间;绒厚遗传力介于0.33~0.56之间;毛长遗传力介于0.23~0.32之间;体重的遗传力介于0.16~0.36之间;绒伸直长度遗传力为0.24;绒细度的遗传力为0.14。(2)产绒量与绒厚、绒厚与毛长、长度与绒厚、长度与毛长的遗传相关分别介于0.33~0.79、0.51~0.69、0.38~0.60、0.74~0.90之间,存在较强的正向遗传相关;绒量与毛长、绒量与体重、绒厚与体重、细度与绒厚、细度与绒量、细度与体重的遗传相关分别为0.11~0.38、0.06~0.17、0.15~0.36、0.02~0.11、0.24~0.35、0.13~0.32之间,存在较弱的正向遗传相关;毛长与体重、长度与绒量、长度与体重、长度与细度、细度与毛长之间的遗传相关介于-0.14~-0.28、-0.09~-0.20、-0.18~-0.23、-0.27~-0.31、-0.17~-0.28之间,存在中等偏弱的负向遗传相关。(3)绒量、绒厚、毛长、体重的重复率分别为0.42、0.27、0.59、0.18。 Abstract:Daughter-dam regression, daughter-dam regression wit hin sire, half-sib correlation and intro-unit half?sib correlation were used in the study to estimate genetic parameters for several traits of Albas type of In ner Mongolia cashmere goats, Traits included in this paper were cashmere yield ( CY),cashmere thickness(CT),staple length(SL),body eight(BW),cashmere stretched length(CSL) and cashmere fineness(CF).The results showed: (1)Heritabilities for CY,CT,SL and BW ranged between 0.26 and 0.45,between 0.33 and 0.56, between 0.23 and 0.32 and between 0.16 and 0.36 respectively; Heritabilities for SL and CF were 0.24 and 0.14 respectively.(2)Genetic correlations between CY and CT,between CT and SL ,between SL and CT and between CST and SL ranged 0.33~0. 79, 0.51~0.69, 0.38~0.60, and 0.74~0.90,respectively,and they belonged to high positive one;Genetic correlations between CY and SL, between CY and betw een CY and BW,between CT and BW, between CF and CT, between CF and CY and betwe en CF and BW ranged 0.11~0.38, 0.06~0.17,0.15~0.36, 0.02~0.11, 0.24 ~0.35 and 0.13~0.32,respectively, and they belonged to low positive correla tion;Genetic correlations between SL and BW, between SL and CY,between SL ?and BW, between? SL and CF and between CF and SL ranged -0.14~-0.28, -0.09~-0 .20, -0.18~-0.23,-0.27~-0.31,and -0.17~-0.28,respectively,and they belonged to low negative correlations.(3)Repeatabilities for CY,CT,SL and BW were 0.42,0.27,0.59 and 0.18,respectively.  相似文献

绒毛用直接法制片,新生儿脐带血和成人外周血用半微量全血法制片。对绒毛、脐带血和外周血染色体的断裂和裂隙进行比较。结果表明,绒毛细胞的染色体断裂和裂隙比新生儿脐带血和成人外周血的显著增高,而新生儿脐带血和成人外周血之间则无明显差异。 Abstract:Chromosome breaks and gaps in chorionic villus cells and lymphocytes from newborn and abult were compared.The number of chromosome breaks and gaps in chorionic villus cells was higher than that in newborn and adult lymphocytes,This might be one of the reasons for higher chromosome aberration rate in chorionic villus.  相似文献

Brassinosteroid (BR) and gibberellin (GA) are two groups of plant growth regulators essential for normal plant growth and development. To gain insight into the molecular mechanism by which BR and GA regulate the growth and development of plants, especially the monocot plant rice, it is necessary to identify and analyze more genes and proteins that are regulated by them. With the availability of draft sequences of two major types, japonica and indica rice, it has become possible to analyze expression changes of genes and proteins at genome scale. In this review, we summarize rice functional genomic research by using microarray and proteomic approaches and our recent research results focusing on the comparison of cDNA microarray and proteomic analyses of BR- and GA-regulated gene and protein expression in rice. We believe our findings have important implications for understanding the mechanism by which BR and GA regulate the growth and development of rice.  相似文献

The achene wail anatomy and surface sculpturing of 14 species representing Lactuca L. and related genera were Investigated to evaluate inter-and Intrageneric relationships. The achene wall anatomy of the studied species can be divided Into two types: winged and ribbed. The winged type is present in Lactuca L., Pterocypsela Shlh, Clcerblta Wallr., Chaetoserls Shlh and Stenoseris Shlh, and is characterized by protruding wings and costae in transverse section. Winged type achene mesocarps are composed of parenchymatous cells and fiber ceils, and the distinct fibrous strands are confined to the costae or wings of the achenes. The ribbed type is present in Notoseris Shih and Paraprenanthes Chang ex Shih, and Is characterized by only having costae In transverse section. Fiber cells are continuous in the costae and intercostae, in surface sculpturing, the ornamentation and the shape of epidermal cells are different among these genera. The results Indicate that L. altalca and L. serrlola should be conspeclfic, and that L. dollchophylla is probably an Intermediate taxon between Lactuca and Chaetoserls. The results also support the separation of Pterocypseia, Paraprenanthes, Notoseris, Chaetoseris and Stenoserls from Lactuca and Clcerblta as independent entitles. Close affinities between Pterocypsela and Lactuca, Paraprenanthes and Notoserls, and Chaetoserls and Stenoserls are also proposed.  相似文献

单核苷酸多态性(single nucleotide polymorphism,SNP)是第三代遗传标记,在基因定位、遗传疾病和人类起源等理论研究中具有重大意义, 在抗药性或药物过敏反应中扮演着极其重要的角色,正逐步成为分子诊断、临床检验、新药研发的重要手段。随着人类基因组测序的完成,SNP分型和发现成为遗传和生命医学领域研究的热点之一。近年来,SNP的检测方法层出不穷,发展很快。文章综述和分析了几种新建立的SNP检测方法,包括基因芯片、分子探针、荧光偏振、荧光共振、质谱和磁性颗粒分析。在生物化学、工程学和分析软件等方面取得突破的基础上,有望建立灵敏准确、简便易行、高通量、低费用的SNP技术。Abstract: Single nucleotide polymorphism (SNP) is the third generation genetic marker. SNP detection now is becoming increasingly important means in molecular diagnostics, clinical assay and novel drug development. It plays an essential role in drug resistance and anaphylactic reaction and has the importamce in theoretical studies of gene location, hereditary diseases and human origin. With the accomplishment of human genome sequencing, the genotyping and discovering of SNP are becoming hot subjects in genetics and biomedicine researches. The methods for SNP detection were renewed rapidly and developed fast in past few years. In this review, several newly established detection methods including gene chip, molecular probe, fluorescence polarization and resonance, mass spectrometry, and bacterial magnetic particle are discussed. It could be expected that an accurate and sensitive, simple and easy-to-handle SNP technology with low cost and high throughput will be available on the basis of research breakthroughs of biochemistry, engineering and analytic software.  相似文献

Simultaneous silencing of multiple upregulated genes is an attractive and viable treatment strat-egy for many incurable diseases including cancer.Herein we used a dual gene-targeted siRNA conjugate composed of VEGF and Survivin siRNA sequences in the same backbone to inhibit proliferation and angiogenesis in two human osteosarcoma cell lines.We synthesized siRNA sequences targeting the VEGF and Survivin genes individually (VEGF siRNA and Survivin siRNA) or simultaneously (one-chain-double-target siRNA:dual siRNA).VEGF and Survivin mRNA and protein expression levels in human osteosarcoma MG-63 and Saos-2 cells were detected by qRT- PCR and western blot analysis.VEGF and Survivin protein location and expression were evaluated by immunohistochemistry and immunofluorescence staining.MG-63 and Saos-2 cell migration,proliferation,apoptosis,and angiogenesis were detected by scratch test,MTT assay,flow cytometry,and capillary tube assay respectively.The dual siRNA induced similar downregulation of VEGF and Survivin mRNA and protein levels,compared with VEGF siRNA or Survivin siRNA alone.The dual siRNA caused greater suppression of MG-63 and Saos-2 cell migration,proliferation and angiogenesis,and promoted more cell apoptosis than VEGF siRNA or Survivin siRNA alone,suggesting that the effects of the dual siRNA on inhibiting cell proliferation,migration,and angiogenesis and promoting apoptosis were superior to those of the single-target siRNAs.Simultaneous silencing of VEGF and Survivin using the dual siRNA may be an advantageous alternative for the development of therapeutic strategies against human osteosarcoma.  相似文献
