横纹肌肌原纤维的第三肌丝──肌联蛋白   总被引:2,自引:0,他引:2  
实验研究证明,在动物横纹肌肌原纤维中,除包含有粗肌丝、细肌丝外,还有纤肌丝的存在,肌联蛋白(肌巨蛋白)是具有挠性的线状蛋白质,分子量为3000 000,长度约为0.9μm,跨越肌原纤维的M-线和Z-线,形成纤肌丝.其生理功能是在粗肌丝装配中具有分子模板作用,并将粗肌丝稳定于肌原纤维肌小节中央以及可参与肌球蛋白活性的调节.  相似文献

甲壳动物横纹肌肌原纤维的肌丝陈列,收缩蛋白质和收缩的Ca2+依赖性调节机制与脊椎动物横纹肌有不少差异.脊椎动物横纹肌、甲壳动物快肌与慢肌的粗丝与细丝的数量比依次为1:2,1:3和1:6,肌丝阵列各异.甲壳动物粗肌丝由肌球蛋白和副肌球蛋白组成,其分子装配与脊椎动物不同.细肌丝含有肌动蛋白、原肌球蛋白和肌钙蛋白,肌钙蛋白-T分子量较高,肌钙蛋白-C仅1个Ca2+结合位点.甲壳动物横纹肌兼有细肌丝调节与粗肌丝调节.  相似文献

利用电子显微镜观察了栉江珧平滑闭壳肌收缩装置的精细结构,它含有粗肌丝、细肌丝和致密体。分离的天然粗肌丝含肌球蛋白和副肌球蛋白,呈现带状和Bear-Selby网格状图象,其周期为14.5nm和7.2nm。以不含ATP的低离子强度溶液处理粗肌丝,则其近侧集聚大量细肌丝;而以微酸性、中等离子强度溶液处理粗肌丝,可以溶去肌球蛋白,但不破坏其周期性结构。以2M脲处理粗肌丝,它纵向分散成直径约有10nm的长带。  相似文献

用能溶解肌球蛋白但不溶解副肌球蛋白的溶液(300 mM KCI,pH6.0)处理分离的蜜蜂间接飞翔肌粗肌丝,经数分钟后可以看到粗肌丝端头散开成为多根微丝,微丝数最多为7根。延长处理时间,可以见到粗肌丝中央部分只剩下直径约为5 nm的徽丝。实验结果支持我们以前提出的蜜蜂间接飞翔肌粗肌丝的结构模式,并指示贯穿肌小节、两端都和Z线相连的内芯至少部分由副肌球蛋白组成。只存在于A带的,由6根微丝形成的外套是由肌球蛋白分子组成。  相似文献

自五十年代肌丝滑行模型建立以来,关于脊椎动物骨胳肌的蛋白质成分,肌丝排列以及肌肉收缩时结构变化的研究取得了很大的进展。骨胳肌肌原纤维由粗、细肌丝有规律地排列所组成。对于肌肉收缩蛋白的选择性抽提,专一性抗体标记以及重组肌丝的研究,证实肌球蛋白存在于粗肌丝;肌动蛋白、原肌球蛋白和原宁蛋白存在于细肌丝(Huxley,A.F.,1957;Huxley,H.E;,1972)。昆虫间接飞翔肌的结构和生理特性有许多不同于脊椎动物骨胳肌的特点。蜜蜂飞翔肌肌原纤维虽然也包含有粗、细两  相似文献

利用甘油梯度离心方法分离和纯化螯虾腹屈肌粗肌丝,电子显微镜照片显示粗肌丝上有数条纵行条纹,指示其可能由数根亚丝所组成。粗肌丝的 SDS-聚丙烯酰胺凝胶电泳表明其含有肌球蛋白和副肌球蛋白,肌球蛋白仅包含有二种轻链。副肌球蛋白类晶体呈针状,具有14.5nm 和72.5nm 的横纹周期。实验结果表明,螯虾腹屈肌粗肌丝是肌球蛋白-副肌球蛋白丝。  相似文献

给出了鸣鸣蝉发声肌肌原纤维的双阵列结构,其肌纤维中并存两种不同阵列的“快”和“慢”动肌原纤维(FSM和SSM).FSM和SSM虽然由粗肌丝构成相同的阵列骨架,但细肌丝对粗肌丝的比例(RTIF)不同,分别为3:1和5:1.明显区别于单音调鸣声的蝉类发声肌肌原纤维的RTIF为3:1的单阵列结构,即与鸣鸣蝉变音调声产生的原初机制相适应.  相似文献

The fine structure of the obliquely striated muscle cells of the longitudinal muscle of an annelid was investigated. The characteristics of myofilaments and cell components, such as sarcoplasmic reticular system (S.R.), T-systems and J-rods corresponded to those previously reported, but it was noted for the first time that the cells could be classified into two types with respect to the diameters of their thick myofilaments. In one type, the thick myofilaments were about 29 nm in diameter (A-type) and in the other they were about 41 nm in diameter (B-type). Most of the obliquely striated muscles described to date have been composed of a single type of cell, but we found two types of cell mixed together in the longitudinal muscle. The A-type cells with slender thick myofilaments were distributed mainly in the inner part of the muscle and the B-type cells with broader thick myofilaments were distributed in the outer part.  相似文献

The brachiopoda, Lingula unguis, has a pair of anterior adductors located in the center of the shell. Each muscle consists of an opaque and a translucent portion which is constructed of smooth and obliquely-striated muscle respectively. According to our ultrastructural observations, the opaque portion seems to have two types of cells. They differ only in the diameters of their thick myofilaments. The fine structure of their cell organelles resembles each other. We measured the diameters of the thick myofilaments in each type of cell to distinguish between the two cell types. About 500 measurements of myofilament diameters were made for each type of cell and statistically analyzed. For one type of cell, the distribution of diameters of the thick myofilaments fit a normal distribution curve with a peak at 37-60 nm. The distribution of diameters of the thick myofilaments for the other type fit a curve in which two normal distribution curves having peaks at 37-60 and 75-97 nm respectively partially overlapped. According to these results, we suggest that the opaque portion contains two types of cells, each having a different distribution of thick myofilament sizes.  相似文献

An investigation of developing skeletal muscle necessitatesthe study of three categories; the derivation of muscle cellsor fibers, myofilament synthesis and interactions, assemblyof myofilaments into functional sarcomeres of striated myofibrils.With few exceptions, skeletal muscle cells are of mesodermalorigin, and consist of rounded mononucleated cells which elongateand fuse with one another to become myotubes. Within the sarcoplasm,myofibrillar proteins are synthesized and grouped into interactingthick and thin filaments. Crude, non-striated myofibrils resultfrom linear arrangements of thick and thin filaments which arehorizontally aligned by the invaginating sarcotubular system.After Z-lines form, providing attachment sites for thin filaments,a typical banding pattern follows. The newly formed Z-linespull apart, followed by the attached thin filaments, and repeating"relaxed" sarcomeres are the resulting striated myofibrillarpattern.  相似文献

The ultrastructural features of cardiac muscle cells and their innervation were examined in the tarantula spider Eurypelma marxi Simon. The cells are transversely striated and have an A band length of about three microns. H zones are indistinct and M lines are absent. Thick and thin myofilament diameters are approximately 200 and 70 Å respectively. Eight to 12 thin filaments usually surround each thick one. Accumulations of thick and thin myofilaments occur perpendicular to the bulk of the myofilaments in some cells. The Z line is discontinuous and thick filaments may pass through the spaces in the Z line. Extensive systems of sarcoplasmic reticulum and transverse tubules are present; these form numerous dyadic junctions in both A and I band regions. Sarcolemmal invaginations form Z line tubules; lateral extensions of the plasma membrane portion of these invaginations form dyads. Nerve branches of the cardiac ganglion make multiple neuromuscular synapses with at least some of the cardiac muscle cells. Both large granular and small agranular vesicles are present in the presynaptic terminals. Intercalated discs similar to those present in other arthropod hearts occur between the ends of adjacent cardiac muscle cells.  相似文献

Summary The arrangement of myofilaments in the striated visceral muscle fibers of two arthropods (crayfish and fruitfly) and in the unstriated visceral fibers of one annelid (earthworm) was studied comparatively. Transverse sections through the A bands of arthropod visceral fibers indicate that each thick myofilament is surrounded by approximately 12 thin filaments. The myofilaments are less organized in the visceral fibers of the earthworm than in muscle fibers of the crayfish and fruitfly. The thick myofilaments of the earthworm are composed of subunits, 20–30 Å in diameter. The presence of two distinct sets of myofilaments in these slowly contracting striated and unstriated visceral muscle fibers suggests that contraction is accomplished via a sliding filament mechanism.In crayfish visceral fibers the sarcolemma invaginates at irregular intervals to form a long and unbranched tubular system at any level in the sarcomere. Dyads formed by the apposition of T and SR membranes are observed frequently. The distribution of the T and SR systems in the visceral fibers of the fruitfly and the earthworm is markedly reduced and dyads are infrequently observed. The reduced T and SR systems may be related to the slow contraction of these fibers. Transport of specific substances across the sarcolemma could initiate contraction or relaxation in these fibers.This study was supported by a training grant GM-00582-06 from the U.S. Public Health Service.  相似文献

Photoelectron count autocorrelation function of light scattered by Limulus thick myofilament suspensions was measured as a function of scattering angle in the relaxed, activated and rerelaxed states. By using the cumulants method of data analysis, the average linewidth over large ranges of KL (up to 120), has been calculated with K and L being, respectively, the magnitude of the momentum transfer vector and the length of the myofilament. We have observed a dramatic increase in the average linewidth denoting the presence of additional high frequency components for the myofilament suspension on the activated state. By confirming our results on the size of the myofilaments from electron micrographs, we are able to attribute the high frequency (kHz) components to the "correlated" cross-bridge motions, representing, to our knowledge, the first direct experimental evidence of such movements in isolated thick myofilament suspensions.  相似文献

The presence of striated subumbrellar musculature in hydromedusae can be related to the development in hydrozoans of a free-swimming life stage. The detailed ultrastructure of the striated subumbrellar musculature of the anthomedusan, Pennaria tiarella is presented. The striated musculature of Pennaria resembles vertebrate striated muscle in filament arrangement (L2 lattice pattern) and M line structure. The striation pattern, out-of-register myofilaments, filament structure as determined by rotational symmetry, Z line structure, types of intercellular junctions, and sarcoplasmic reticulum are more similar to structures found in other invertebrate striated muscle.  相似文献
