Fine details of the internal and external morphology of the in vitro mycelial phase (MP) to yeastlike phase (YP) transition of the dimorphic fungal pathogen Sporothrix schenckii are shown in electron micrographs of ultrathin sections. Morphological transformation at the ultrastructural level was observed to occur by direct formation of budlike structures at the tips and along the hyphae and by oidial cell formation. Direct budding of yeast from conidiospores was not observed. Early transitional forms arising by direct blastic action from the MP possessed conspicuous electron-dense microfibrillar material at the outer limits of the cell wall. The electron density of this microfibrillar material was enhanced by staining with acidified dialyzed iron. It is believed that this extracellular material may be composed in part of an acid mucosubstance. No acid phosphatase activity was associated with this microfibrillar material. This substance was found to be a characteristic of the outer limits of the cell wall of the YP of S. schenckii. Oidial YP cell formation occurred later during the transition. The cell wall of the developing oidial YP transitional form arose from an inner layer of the converting hyphae. No consupicuous alterations of the cytoplasmic content of the parent MP cell was observed during MP-to-YP transition. It is suggested that the MP-to-YP transition of S. schenckii may be regulated by at least two mechanisms involving alterations of the biochemical and/or biophysical nature of the cell wall of the MP cell in response to the conversional stimuli.  相似文献

Summary Walls from cysts, hyphae and sporangia of Phytophthora palmivora consist chiefly (ca. 90% dry wt) of -glucans with 1,3-, 1,4- and 1,6-links. The glucans are predominatly -1,3-linked but there are significant differences in the relative proportion of 1,3-, 1,6- and 1,4-linked glucosyl residues among the three wall types. There are also differences in protein content, susceptibility to degradation by various -glucanases, and surface texture. The isolated cyst wall consists solely of a thin fabric of long, tightly interwoven, randomly oriented microfibrils. Both inner and outer surfaces of the cyst wall are distinctly microfibrillar. The hyphal wall has two different textures; the internal surface is distinctly microfibrillar while the external surface is non-fibrillar. In a germinated cyst, there is a zone of demarcation where the microfibrils of the cyst wall disappear into the smooth outer texture of the germ tube wall. An exo--1,3-glucanase preferentially removed the amorphous material of the outer surface of the germ tube leaving exposed a continuous microfibrillar fabric from cyst to hyphal tube. Conceivably, the textural and structural differentiation of the cell wall may play a decisive role in cellular morphogenesis.  相似文献

A series of environmental and clinical isolates of Sporothrix schenckii being studied in our laboratories under standard conditions of temperature, humidity and media, displayed, in some of the isolates, large dematiaceous hyphae. The large hyphae could be seen to produce the fine hyphae associated with S. schenckii on which typical microconidia developed. Typical microconidia also developed occasionally on the large hyphae, and strands of the unusually large hyphae also were seen to form arthroconidia. Some strains formed large, thin-walled cells on thin and/or thick hyphae resembling the balloon cells seen in mature colonies of Trichophyton tonsurans.All strains which have demonstrated these various and unusual structures histopathologically produced typical sporotrichosis in laboratory animals. When reisolated from these animals, the cultures again exhibited the unusual structures, as well as those typical of S. schenckii.  相似文献

To date, only one case of post-traumatic endophthalmitis caused by Scedosporium dehoogii has been reported, but its contamination or colonization might not be precluded due to the absence of pathogenic isolation and/or pathological examination. We report the first case to our knowledge of S. dehoogii-induced subcutaneous scedosporiosis in a psoriatic patient. A 58-year-old man with 5-year history of psoriasis vulgaris and immunosuppressant therapy developed pyrexia and multiple subcutaneous abscesses on both knees. Direct microscopy of the yellowish pus showed masses of bright green short spores. Skin biopsy revealed some branched septate hyphae within the granuloma. Two aspirated pus specimens collected at a 1-week interval produced white cottony colonies on Sabouraud dextrose agar. Bacterial cultures of one blood and two purulent samples were negative, and fungal culture of blood sample was not performed. The isolate was identified as S. dehoogii using β-tubulin phylogeny and species-specific PCR with primer MSDE1/MSA2. Without addition of antifungal treatment, subcutaneous lesions disappeared spontaneously after immunosuppressant withdrawal and no relapse occurred during 64-month follow-up. The spontaneous recovery may result from immune reconstitution following immunosuppressant discontinuation.  相似文献

The internal mycelium of Pseudoperonospora cubensis has been observed in transmission and scanning electron microscopic preparations. The internal mycelium may be inter- or intracellular. Haustoria of short swollen bundles of hyphae have been observed. Actively growing hyphae contain numerous mitochondria, nuclei, active dictyosomes, low amounts of storage materials (lipid) and microbody-like structures with a laminate inclusion. Thick walled hyphae with a diameter which is smaller than the actively growing hyphae have been observed. These thick wallcd hyphae contain large amounts of reserve material (lipid) and it is suggested that they may function as resting propagules.  相似文献

Summary Immature embryos, inflorescences, and anthers of eight commercial cultivars of Triticum aestivum (wheat) formed embryogenic callus on a variety of media. Immature embryos (1.0–1.5 mm long) were found to be most suitable for embryogenic callus formation while anthers responded poorly; inflorescences gave intermediate values. Immature embryos of various cultivars showed significant differences in callus formation in response to 11 of the 12 media tested. No significant differences were observed when the embryos were cultred under similar conditions on MS medium with twice the concentration of inorganic salts, supplemented with 2,4-D, casein hydrolysate and glutamine. Furthermore, with inflorescences also no significant differences were observed. Explants on callus formation media formed two types of embryogenic calli: an off-white, compact, and nodular callus and a white compact callus. Upon successive subcultures (approximately 5 months), the nodular embryogenic callus became more prominent and was identified as aged callus. The aged callus upon further subculture, formed an off-white, soft, and friable embryogenic callus. Both the aged and friable calli maintained their embryogenic capacity over many subculture passages (to date up to 19 months). All embryogenic calli (1 month old) from the different callus-forming media, irrespective of expiant source, formed only green shoots on regeneration media that developed to maturity in the greenhouse. There were no significant differences in the response of calli derived from embryos and inflorescences cultured on the different initiation media. Also, the shoot-forming capacity of the cultivars was not significantly different. Anther-derived calli formed the least shoots. Aged and friable calli on regeneration media also formed green shoots but at lower frequencies. Summary The oomyceteSaprolegnia ferax, unlike most walled organisms, does not regulate turgor. When hyphae were subjected to water stress by the addition of sucrose or other solutes to the growth medium, turgor pressure diminished progressively; yet the hyphae continued to extend with deposition of a more plastic apical wall. Even when turgor was no longer measurable with a micropipet-based pressure probe (0.02 MPa or less, compared with 0.4 MPa in unsupplemented medium) they produced regular hyphal tubes and tips. Such turgorless hyphae extended as rapidly, or more rapidly, than normal ones, but they were wider and their tips blunter. Despite the loss of turgor, hyphae put forth branches and cysts germinated. The organization of actin microfilaments was essentially normal, and the response to cytochalasin A was similar in turgorless and standard hyphae. However, as turgor diminished the hyphae's capacity to penetrate solid media was progressively impaired; aerial hyphae were no longer produced, and zoospore formation was inhibited. The results contradict the common belief that turgor supplies the driving force for hyphal extension, tip morphogenesis, and branching. Evidently, these functions do not intrinsically require hydrostatic pressure. Shoots of Miscanthus X ogiformis Honda Giganteus transferred from medium with benzyladenine (BA) to thidiazuron (TDZ) formed significantly more axillary shoots than shoots grown continuously on either medium, or transferred from TDZ to BA. Shoot formation on axillary shoots transferred from BA-containing media to media with kinetin or isopentenyladenine (2iP) or transferred from media with TDZ, kinetin or 2iP to media with BA, corresponded to the number of shoots formed in the previous subculture. Shoot formation on shoots transferred from medium containing BA to media containing combinations of BA and/or TDZ increased with increasing concentrations of TDZ in the first subculture, whereas shoot formation in the second and third subculture depended on the BA concentration. When shoots were transferred from media with BA to media with TDZ, the time for shoot formation, as well as shoot size, indicate that the combined effect of BA and TDZ is expressed only during the early phase of the subculture. Summary The ultrastructure of the surface membranes of the spores and aerial hyphae of S. glaucescens and S. acrimycini was studied by means of electron microscopy of air-dried whole mounts, thin sections, negative stainings and freezeetchings. The delicate sheath which envelopes the spore walls produces long protruding appendages which have previously been designated as hairs, a name which is based on silhouette views of untreated whole mounts, where the processes appear long and curled. Freeze-etching and negative staining revealed that the processes are composed of interlacing straight tubular units of varying length. These tubules exhibited a distinct fine structure. It is suggested that they are made of rolled up sheets with a regular striation pattern. The surface membrane of the non-sporulating aerial hyphae has no processes, but a pattern of irregularly arranged and ill-defined rodlets.  相似文献

Summary The microfibrillar component of the walls of zoosporangia and resistant sporangia of the phycomycete Allomyces arbusculus has been studied in the electron microscope, after chemical removal of the amorphous wall materials. In the zoosporangium wall the microfibrils are randomly arranged, as in the outer layer of the hyphal walls, and the sporangial wall is of even thickness. In the resistant sporangia the microfibrillar layer of the wall is perforated by numerous pores 0.25 in diameter. The microfibrils are randomly arranged over much of the wall but tend to be concentrically arranged in the vicinity of the pores. On the inside of the wall the microfibrils form a thickened rim around the pore.  相似文献

In vitro growth of Quercus robur L. buds taken from 1–7 month old seedlings or adult plant material was studied. The following were investigated for their effects on the establishment of explants and subsequent subcultures: original position and lignification of the primary explant, conditioning and ageing of source plants, incorporation of N6 benzyladenine (BA) and activated charcoal in the medium.For bud break the best results were obtained with explants from herbaceous twigs in all tested media. For shoot growth the results depended on the medium used. Medium containing activated charcoal produced episodic growth, leaf organogenesis was reduced, spontaneous rooting occurred, but subculturing from adult plant material failed. On medium containing more than 8.8 M of BA, all the buds developed abnormally and elongation did not occur. At lower concentrations of BA (4.4 M) shoots elongated, leaf organogenesis was increased and episodic growth tended to disappear on subcultured seedling explants. No spontaneous rooting was observed, but subculture from adult plant material was managed successfully.  相似文献

Summary Most of the hyphae forming the medulla of the stroma of the brown rot fungi are 4–7 in diameter and contain food reserves in large vacuoles and lipid bodies. Some stromatal hyphae have very thick walls and perform a protective function. Smaller hyphae (1–2 in diameter) form a network through the medulla and their structure suggests that they initiate the growth of vegetative hyphae and spores after the stroma has passed through a period of rest.  相似文献

In this paper we report a case of onychomycosis caused by Chaetomium globosum. The patient had lesions of the fingernails of the left hand. The direct microscopical examination of the nails showed light-brown hyphae with thick-walled cells. The histopathological examination revealed thick aggregated hyphal element in the nail plate. Amongst the antimycotics tested oxiconazole with MIC values of 0.3 g/ml–1 was found to be most effective in vitro against Chaetomium globosum.  相似文献

We have described the isolation of Mycobacterium chelonei subspecies abscessus which on two occasions grew out within 4 days in routine cultures after inoculation with purulent material from injection abscesses in a patient with diabetes mellitus. Media isolation and culture characteristics of this organism in comparison to others in the Mycobacterium fortuitum complex are described. Mycobacteria in routine laboratory media can be mistaken for diphtheroids or other "contaminants" or can be overlooked entirely. The possibility of clinical infection of mycobacterial species should be kept in mind when confronted with "sterile" abscesses at injection sites.  相似文献

Cultures of Lavandula latifolia Medicus were established from axillary buds of mature field-grown plants. Explants were initially cultured on media with two different macronutrient combinations and benzyladenine or kinetin added either individually or with naphthaleneacetic acid. Subsequently, explants were subcultured in Murashige and Skoog medium supplemented with 20% coconut milk, 0.57 M indoleacetic acid and 8.88 M benzyladenine. Shoot proliferation from axillary buds was not affected by seasonal fluctuations in the stock plants but depended on the macronutrient composition and on the type and concentration of cytokinin tested. Best results were obtained in explants initially cultured in media with Murashige and Skoog constituents and supplemented with 5 M benzyladenine. In vitro-grown shoots were used to induce multiple shooting by transferring them to subculture medium. Shoots were rooted on Murashige and Skoog medium with macronutrients at half-strength. Plantlets were transferred to soil and grown to maturity.  相似文献

Summary An electron microscope study has been done on the structure of microconidia and stroma of Sclerotinia sclerotiorum. The microconidia showed the usual cell organelles already described for other species of this genus, e.g. a large nucleus and large lipid body, a few mitochondria and sparse endoplasmic reticulum. The stromatal ultrastructure presented hyphae with a rich content in lipid bodies and storage vacuoles as food reserve. Simple septa with plugged pores were occasionally seen. Although the existence of two kinds of hyphae, one with abundant food materials and the other type with a degenerated aspect, both hyphae showed a similar thickness in their cell walls approximately 0.2 , and the diameter of these hyphae was 1.5–3 .  相似文献

Summary Cell wall structure in ray and axial parenchyma cells in the wood ofCryptomeria was shown to be typically crossed polylamellate and dissimilar to the characteristically layered wall of fibers and tracheids. Ray cells differed from axial cells in terms of form and also in the relative inclination of crossed microfibrillar helices in the cell wall. This feature was reflected by positive birefringence in ray cells and negative birefringence in axial cells. Localized wall thickenings,viz. transverse bars in ray cells and longitudinal ribs in axial cells, also displayed crossed polylamellate structure. This observation contrasts with the exclusively longitudinal microfibrillar orientation previously reported for longitudinal ribs in elongated parenchyma cells of primary tissue. On the basis of similar microfibrillar orientations between outer and inner wall lamellae, the cell walls ofCryptomeria parenchyma were judged to be predominantly secondary.Lignin was heterogeneously distributed in lamellate fashion and a high concentration characterized the thin middle lamella. Both types of parenchyma suggested a higher lignin content than adjacent longitudinal tracheids.  相似文献

Summary The effects of methyl benzimidazole-2-yl carbamate (MBC) on microtubule and actin cytoskeleton were analyzed by indirect immunofluorescence and transmission electron microscopy in a wild-type strain and a benomyl-resistant mutant (benA 10) ofAspergillus nidulans. The treatment of the wild-type strain with sublethal doses of MBC not only caused depolymerization of cytoplasmic microtubules (MTs), but also changed the pattern of actin at the hyphal tips. In the MBC-treated hyphae, the actin fluorescence was concentrated at the very tip region of the hypha, whereas in the control hyphae, the actin fluorescence was weak at the very tip and strong below the tip. The dose of MBC used for the wild-type strain did not depolymerize the MTs or modify the actin organization at the apex in the mutant strain, which confirmed that the change in actin distribution in the wild-type strain was due to the disruption of MTs. In the mutant strain, a seven times higher concentration of MBC than in the wild-type strain was required to depolymerize MTs and to alter the actin organization at the apex. The ultrastructural study of the MBC-treated hyphae revealed that the area containing apical vesicles was larger and the number of microvesicles was higher than in control hyphae. These changes probably resulted from the disassembly of MTs and the reorientation of actin cytoskeleton in MBC-treated apexes and suggested that MTs would organize the actin at the apex, which in turn would restrict the vesicle fusion to a narrow area at the hyphal tip. Symbiosis between fungi and plant roots forming a mycorrhiza involves extensive interactions at the molecular level between both partners. The role of plant hormones in the regulation of mycorrhizal infection is not known to involve jasmonates. Their endogenous levels increase during pathogen attack; however, little has been done on their involvement in mycorrhizae. In our recent work, root growth patterns of 2-month-old spruce seedlings after inoculation withPisolithus tinctorius and/or jasmonic acid (JA) treatment were studied using a paper-sandwich technique. Changes in root length, the degree of branching, presence and length of root hairs, and infection parameters were followed using a stereomicroscope. The first mycorrhizal contact of hyphae with roots was significantly accelerated upon treatment with 0.5 M JA. Interactions between root hairs and fungal hyphae were seen by scanning electron microscopy. The multiplication of root hairs of non-mycorrhized seedlings treated with 5.0 M JA and changes of the root surface were observed by the same technique.  相似文献

The application of Ca2+ mobilizing secretagogues to rat parotid acini results in a significant decrease in cell volume (15-30%) due to isotonic salt loss. It is often assumed that the effects of such an isotonic volume decrease can be mimicked by anisotonic cell shrinkage. We demonstrate that the Na+-K+-2Cl- cotransporter in these cells is up-regulated by Ca2+ mobilizing secretagogues as well as by cell shrinkage in hypertonic media. However, we find that although the protein kinase inhibitors staurosporine (0.3 M) and K252a (0.6 M) significantly blunt the latter up-regulation, they are without effect on the former. These observations suggest that hypertonic and isotonic shrinkage do not result in the activation of the same intracellular signalling pathways, and indicate that anisotonic volume perturbations may not provide good experimental models of physiologic isotonic volume changes.  相似文献
