The root-rot fungus Heterobasidion annosum (Fr.) Bref. species complex consists of three intersterility groups (S, F, and P), separated by their host affinity. The phylogenetic relationship of the species complex was studied, with the focus on the S and F groups, by comparing DNA sequences of four nuclear gene fragments: calmodulin, glyceraldehyde 3-phosphate dehydrogenase, heat stress protein 80-1, and elongation factor 1-alpha, and one anonymous locus, from 29 fungal isolates originating from Europe, Asia, and North America. The phylogeny of each separate gene locus as well as the combined dataset consisted of three main clades: European F group isolates, Euroasian S group isolates, and North American S group isolates, suggesting them to be separated into phylogenetic species. The results also support the hypothesis of an early separation between the S and F groups, indicating that their distribution have followed their host tree species for a considerable time period.  相似文献

A genetic linkage map of the basidiomycete Heterobasidion annosum, casual agent of root rot in conifers, was constructed from a compatible mating between isolates from the North American S and P intersterility groups. In a population consisting of 102 progeny isolates, 358 AFLP markers were scored. The linkage analysis generated 19 large linkage groups, containing 6 or more markers, which covered 1468 cM. The physical size to genetic distance was approximately 11.1 kbp/cM. Segregation of three intersterility gene loci were analysed through mating of the progeny isolates with three tester strains carrying known intersterility genotypes. The loci for the two intersterility genes (S and P) were successfully located in the map. Segregation of the mating type locus was analysed by backcrossing the progeny isolates with their parental strains. The mating type locus could not be located in the map.  相似文献

Fungi in the basidiomycete species complex Heterobasidion annosum are significant root-rot pathogens of conifers throughout the northern hemisphere. We utilize a multilocus phylogenetic approach to examine hypotheses regarding the evolution and divergence of two Heterobasidion taxa associated with pines: the Eurasian H. annosum sensu stricto and the North American H. annosum P intersterility group (ISG). Using DNA sequence information from portions of two nuclear and two mitochondrial loci, we infer phylogenetic relationships via parsimony, Bayesian and median-joining network analysis. Analysis of isolates representative of the entire known geographic range of the two taxa results in monophyletic sister Eurasian and North American lineages, with North America further subdivided into eastern and western clades. Genetically anomalous isolates from the Italian presidential estate of Castelporziano are always part of a North American clade and group with eastern North America, upholding the hypothesis of recent, anthropogenically mediated dispersal. P ISG isolates from Mexico have phylogenetic affinity with both eastern and western North America. Results for an insertion in the mitochondrial rDNA suggest this molecule was obtained from the Heterobasidion S ISG, a taxon sympatric with the P ISG in western North America. These data are compatible with an eastern Eurasian origin of the species, followed by dispersal of two sister taxa into western Eurasia and into eastern North America over a Beringean land bridge, a pattern echoed in the phylogeography of other conifer-associated basidiomycetes.  相似文献

The genus Heterobasidion includes some of the most important pathogens of conifers in the world, and as such it is one of the most intensely studied genera of fungi. Because of the remarkable paucity of distinguishing morphological traits, the taxonomy of species within this genus has always been problematic. A partial resolution of the taxonomic issues regarding this genus was achieved by defining the most important and first described species within it, Heterobasidion annosum, as a species complex containing at least two partially intersterile biological species defined as intersterility groups (ISGs). With time, the number of ISGs has increased to include at least two distinct North American and three distinct Eurasian ISGs. Two additional, yet unnamed, taxonomic groups within Heterobasidion have been recently described in Japan. ISGs are distinguishable either by minor morphological differences, by partial intersterility, by ecological traits including host preference, and/or by their geographic range. Several studies employing a variety of molecular tools and analyses have confirmed the distinct genetic divergence among ISGs, identifying each of them as a monophyletic group. Using genetic markers, genotypes can always be unambiguously assigned to one ISG, and very few inter-ISG hybrids have been identified. In this paper, we summarize the available information, both genetic and ecological, that differentiates the two North American ISGs from each other and from other taxonomic units within the genus. We demonstrate that morphometric characteristics such as pore density and pore shape differentiate the two ISGs. Based on the cumulative genetic, ecological, and morphological evidence, we propose a disposition of ISGs of the North American H. annosum by replacing the P ISG with Heterobasidion irregulare, and the S ISG with Heterobasidion occidentale.  相似文献

Quantitative trait loci (QTL) for mycelial growth rate were identified and positioned on a genetic linkage map of Heterobasidion annosum sensu lato (s.l.), an important root rot pathogen on conifers. The mycelial growth rate among 84 progeny isolates were analysed in two different temperature regimes, 12 and 24 °C on malt extract agar and found to segregate as a continuous character. The assay identified three QTL for growth rate at low temperature positioned on the linkage groups 1, 17 and 19 with peak LOD values of 3.2, 2.9 and 4.8, respectively. At the QTL for high temperature growth, peak LOD values of 1.3, 2.8 and 2.2, were identified. The three QTL for the low temperature regime explained 20.9, 18.1 and 24.0 % of the variation in the mycelial growth rate, respectively. The broad-sense heritability was estimated to be 0.97 and 0.95 for growth rate at low and high temperatures, respectively. Two of the QTL for the mycelial growth rate were shown to be tightly linked to the intersterility genes S and P, which control mating within and between closely related species and intersterility groups of H. annosum s.lat. Isolates with a plus allele at the intersterility loci had a higher growth rate than isolates that harboured minus alleles.  相似文献

Polymerase chain reaction (PCR) was used to amplify ribosomal RNA (rRNA) genes of two isolates belonging to different intersterility groups of Heterobasidion annosum. Amplification of nuclear small rRNA or small mitochondrial rRNA yielded products of similar size in both intersterility groups. Digestion of PCR products with restriction enzymes did not reveal any differences between groups. Amplification of an internal transcribed spacer (ITS) region yielded an 0.3 kb product in both isolates. Digestion of this product with Hha I restriction enzyme revealed an intersterility specific banding pattern. These results indicate that PCR has a considerable potential for the diagnosis and characterization of fungal pathogens which are difficult to identify in conventional ways.  相似文献

在四川北部九寨沟和黄龙保护区的云杉、松树和铁杉上发现异担子菌 ,并从 6号标本中分离到 73个单孢菌株。在每个标本中随机选取 2个菌株分别与欧洲的原始多年异担子菌、小孔异担子菌和冷杉异担子菌的单孢菌株进行融合性交配。试验表明 ,这 6号标本都是小孔异担子菌。四川的菌株与欧洲的原始多年异担子菌交配不融合 ,而与欧洲的小孔异担子菌完全融合 ,并在交配后的菌落中形成锁状联合 ,且在交配的菌落中不产生拮抗线。虽然四川的菌株与欧洲的冷杉异担子菌有较高的融合性 ,但这些交配大部分为单项交配 ,即只在四川一侧的菌落中产生锁状联合 ,而且在交配的菌落中多数产生拮抗线。研究样品全部采自天然林 ,小孔异担子菌在四川经营林分中的致病性还有待进一步调查。  相似文献

Transmission of dsRNA viruses between homo- and heterokaryotic mycelia paired on agar plates and into conidia has been studied in Heterobasidion annosum. Horizontal transmission of dsRNA occurred between both homo- and heterokaryotic isolates, as well as between isolates belonging to different intersterility groups. The proportions of vertical transmission into conidia were 3% and 55%, respectively, for the two isolates included in the study. RT-PCR of dsRNA and PCR-RFLP of mitochondrial markers were used to confirm transmission of dsRNA between the cytoplasms of different mycelia. The identity of nuclei and nuclear migration during experiments were verified using PCR-RFLP of several nuclear markers.  相似文献

Four putative peroxidase-encoding gene fragments, named mnp1a, mnp1b, mnp2 and mnp3, were amplified with degenerative primers from the white-rot basidiomycete genus Heterobasidion. The fragments were cloned and sequenced. Similar fragments were produced and analyzed from the related genera Amylostereum, Bondarzewia and Echinodontium. Each amplified fragment contains three identically positioned introns. According to the predicted amino acid sequence, these fragments are most similar to two Mn peroxidase-encoding genes (MPGI and mnp2) and gene pgv of Trametes versicolor. Conserved residues thought to be essential for peroxidase function were identified. All four MnP gene loci of Heterobasidion were detected only in H. parviporum. Variation occurred in the predicted amino-acid sequences (131-132 amino acids) of all four fragments originating from the 47 Heterobasidion isolates tested. Amino acid variation in fragments of mnp2 and mnp3 separated European Heterobasidion parviporum ("S-type") and H. abietinum ("F-type"), known to have identical rDNA sequences. Asian and western North American isolates from fir, spruce and other hosts had the peroxidase amino acid sequences of European H. parviporum. American and European H. annosum ("P-type") isolates had different amino acid sequences and might be cryptic species.  相似文献

One-month-old Scots pine (Pinus sylvestris) and Norway spruce (Picea abies) seedlings were inoculated in vitro with Heterobasidion annosum strains, four each of the P-, S- and F-intersterility groups. Variation among strains and between the IS groups in virulence, expressed in mortality rate, was detected during twelve months after inoculation. Most of the strains were more virulent on spruce than on pine, and mortality of spruce seedlings was significantly higher. The P strains displayed similar virulence on both hosts, while S strains caused higher mortality of spruce seedlings and significantly lower mortality of pine seedlings. Strains of the F group were less virulent, but killed significantly (P < 0.05) more spruce than pine seedlings. In the interspecific analyses with two hosts, the isolates and IS groups accounted for most of the explained variation in the host mortality.  相似文献

Trichaptum abietinum and Trichaptum fuscoviolaceum (Hymenochaetales, Basidiomycota) are closely related saprotrophic fungi, widely distributed on coniferous wood in temperate regions worldwide. Three intersterility groups have previously been detected in T. abietinum, while no prezygotic barriers have been proven within T. fuscoviolaceum. The aim of this study was to reveal the phylogeography and genetic relationship between these two closely related species and to explore whether the previously observed intersterility groups in T. abietinum are reflected in the genetic data. We assembled worldwide fruit body collections of both species (N = 314) and generated DNA sequences from three nuclear (ITS2, LSU, IGS) and one mitochondrial rDNA region (mtLSU). The two species are genetically well separated in all analyses. In correspondence with observations from earlier mating studies, our results revealed that T. fuscoviolaceum is genetically more uniform than T. abietinum. Multiple genetic sub-groups exist in T. abietinum that may correspond to the previously observed intersterility groups. However, there is low consistency across the investigated loci in delimiting the different sub-groups, except for a consistent North American group. As for many other widespread fungi, a complex phylogeographic pattern is found in T. abietinum which may have been formed by geographic, as well as multiple genetic intersterility barriers.  相似文献

Little is known about the genetic relationship of pathogenic strains of Clostridium difficile from various parts of the world. We used Hind III restriction digestion of whole DNA to type isolates of C. difficile form hospitals in Argentina and Chile. The restriction pattern type of these South American isolates was classified according to the existing REA library of over 400 distinct REA types in over 90 groups, based on typing of more than 6000 clinical isolates. A total of 22 isolates of C. difficile was obtained and typed. The majority of isolates were matched to groups previously seen in North American and Europe. Three toxigenic groups, R, F, and Y, predominated in the isolates. Five isolates were found to be non-toxigenic and three belonged to group M, the most common non-toxigenic group found in North America. Several identical types were recovered from different hospitals, and types of the same group were found in both Buenos Aires and Santiago.  相似文献

Serpula himantioides is a widespread saprotrophic morphospecies mainly colonising coniferous wood in nature, but it appears frequently in buildings as well. From an earlier study, it is known that at least three divergent lineages occur within the S. himantioides species complex. In this study, a broader sample of S. himantioides isolates has been analysed by multi-locus sequencing, including new isolates from Asia, North and South America. Altogether five phylogenetical species (PS1-5) were detected, all recognised across independent gene phylogenies. A new southern South American phylogenetic species (PS1) was found, representing an early diverging lineage within the S. himantioides species complex. The two closely related PS2 and PS3 lineages included isolates from North America only, and PS4 was also dominated by North American isolates. Most of the investigated isolates (76%) clustered into PS5, a lineage that has been found on most continents, including North America. Overall, little phylogeographical structure was found in PS5, indicating frequent and recent long-distance dispersal events within this widespread lineage. Our analyses indicate that South and North America are the centres of divergence for the S.?himantioides species complex. Some of the lineages seem adapted to various substrates, but PS5 is able to decay a wide array of angiosperms and gymnosperms, which may have facilitated the spread of this lineage throughout the world.  相似文献

从亚洲、欧洲和北美收集18个蜜环菌狭义种菌株,PCR扩增其rDNA的IGS和ITS区域,用AluI、HaeIII、HinfI和TagI四种限制性内切酶进行酶切,同时用随机扩增微卫星(RAMS)多态性,对蜜环菌狭义种的进行了遗传多样性和分子系统学分析。结果蜜环菌狭义种的IGS和ITS-RFLP类型比以前报道的更多,亚洲、欧洲和北美菌株都具有比较明显的特征片段,通过ITS和IGS图谱可将三个大陆的菌株区分开,其中IGS-HinfI图谱能完全区分3个大陆的菌株。RFLP数据的系统学分析表明,该种存在明显的大陆遗传分化,亚、欧、北美的群体分属三个相互独立的进化系,北美群体IGS变异程度较小,ITS变异程度极高;而欧洲群体IGS的变异程度较大,多态性高,ITS变异程度非常小。RAMS系统分析表明,中国、日本和非洲的同宗配合蜜环菌属于一个系统发育系,该发育系同欧洲和北美的异宗配合种的两个发育系分属3个不同的独立进化分支。据此建议欧洲和北美的异宗配合蜜环菌应作为蜜环菌的两个亚种。  相似文献

To clarify the relationship between Bartonella grahamii strains and both the rodent host species and the geographic location of the rodent habitat, we have investigated 31 B. grahamii strains from ten rodent host species from Asia (Japan and China), North America (Canada and the USA), and Europe (Russia and the UK). On the basis of multi-locus sequencing analysis of 16S rRNA, ftsZ, gltA, groEL, ribC, and rpoB, the strains were classified into two large groups, an Asian group and an American/European group. In addition, the strains examined were clearly clustered according to the geographic locations where the rodents had been captured. In the phylogenetic analysis based on gltA, the Japanese strains were divided into two subgroups: one close to strains from China, and the other related to strains from Far Eastern Russia. Thus, these observations suggest that the B. grahamii strains distributed in Japanese rodents originated from two different geographic regions. In the American/European group, B. grahamii from the North American continent showed an ancestral lineage and strict host specificity; by contrast, European strains showed low host specificity. The phylogenetic analysis and host specificity of B. grahamii raise the possibility that B. grahamii strains originating in the North American continent were distributed to European countries by adapting to various rodent hosts. Bursaphelenchus xylophilus, pinewood nematode (PWN), is the most serious pest of pine forests in Japan, but in North America its role in pine wilt disease is still being studied. The PWN is known to infest many species of Pinus, with P. nigra, P. sylvestris, and P. thunbergii the most susceptible in the eastern United States. Because of its potential, several European countries (Finland, Norway, and Sweden) and Korea have established embargoes against the importation of coniferous wood from regions of the world known to be infested with the PWN. Although the PWN is not considered an economic pest in North American forests, the recent embargoes have established an impact on current forest management practices and an economic impact on North American export trade.  相似文献

A phylogenetic analysis was performed on 51 isolates of the commercially valuable basidiomycete, Grifola frondosa (maitake), using sequences from the Internal Transcribed Spacers and 5.8S region of the nuclear ribosomal DNA (rDNA) and a portion of the β-tubulin gene. The β-tubulin gene provided more than twice as many variable characters as the ITS/5.8S regions. The isolates analyzed comprised 21 from eastern North America, 27 from Asia, one from Europe, and two of unknown geographic origin, one of which was the major US commercial production strain in use. Grifola sordulenta was used as an outgroup. Combined and separate analysis of both genes showed a partition separating Asian versus eastern North American isolates. Bootstrap analysis showed strong support for these clades in the β-tubulin data alone and in the combined data. The major commercial isolate of unknown geographic origin is apparently of Asian descent based on its grouping within the Asian clade. The single European isolate analyzed was distinct from both the eastern North American and Asian clades. These results indicate strong support for a species partition separating eastern North American and Asian isolates of G. frondosa, despite previous studies indicating no morphological distinction between them.  相似文献

Evolution of viruses in the eastern equine encephalomyelitis (EEE) complex was studied by analyzing RNA sequences and oligonucleotide fingerprints from isolates representing the North and South American antigenic varieties. By using homologous sequences of Venezuelan equine encephalomyelitis virus as an outgroup, phylogenetic trees revealed three main EEE virus monophyletic groups. A North American variety group included all isolates from North America and the Caribbean. One South American variety group included isolates from the Amazon basin in Brazil and Peru, while the other included strains from Argentina, Guyana, Ecuador, Panama, Trinidad, and Venezuela. No evidence of heterologous recombination was obtained when three separate regions of the EEE virus genome were analyzed independently. Estimates of the overall rate of EEE virus evolution (nucleotide substitution) were 1.6 x 10(-4) substitution per nucleotide per year for the North American group and 4.3 x 10(-4) for the Argentina-Panama South American group. Evolutionary rate estimates for the North American group increased over 10-fold (from about 2 x 10(-5) to 4 x 10(-4)) concurrent with divergence of two monophyletic groups during the early 1970s. The North and South American antigenic varieties diverged roughly 1,000 years ago, while the two main South American groups diverged about 450 years ago. Analysis of multiple strains isolated from an upstate New York transmission focus during the same years suggested that, in certain locations, EEE virus may be relatively isolated for short time periods.  相似文献

Using taxon-specific competitive-priming polymerase chain reaction (TSCP-PCR) the intersterility groups (ISGs) S and P of the root and butt rot pathogen Heterobasidion annosum were found in an active pine mortality center in the forest region Tharandter Wald (Saxonia, Germany). All isolates from spruce were typed as S, and all isolates from pine were P, except for two isolates which were identified as hybrid 'SP’genets. These results indicate the possibility of gene flow in nature between the two ISGs in Europe.  相似文献
