Summary A second flowering gene, Sp, which influences sensitivity to photoperiod, is identified in the sweet pea, Lathyrus odoratus L. Genes Sp and Dn h act in a complementary manner to confer the summer-flowering phenotype and a near obligate long day requirement for flowering in the unvernalized state. Mutations sp and Dn i each diminish the response to photoperiod, and genotypes sp Dn h and Sp Dn i confer a spring-flowering phenotype. Response to photoperiod is further reduced in genotype sp Dn i, which flowers only marginally later than the day-neutral or winter-flowering phenotype characterized by genotypes Sp dn and sp dn (gene dn is epistatic to the gene pair Sp/sp). Like Dn i, gene sp reduces basal branching, while a branching gene, here resymbolized b, is shown to delay flowering in certain circumstances. Gene dn largely prevents basal branching in either b or B plants, but dn b plants do produce lateral shoots from the upper nodes, leading to a novel phenotype. The implications of the interactions between genes sp, Dn i, dn and b are discussed with respect to the control of flowering and branching.  相似文献

It has previously been concluded that regions tentatively designated trfA and trfB, located at 16–18.7 and 54–56 kb, respectively, on the genome of broad host range plasmid RK2 provide trans-acting functions involved in plasmid replication and maintenance in Escherichia coli (Thomas et al., 1980). A third region, the replication origin, oriRK2, located at 12 kb on the genome, is also required. A segment of DNA containing oriRK2 can be linked to a nonreplicating selective marker and can replicate as an autonomous plasmid so long as DNA of RK2 carrying the gene for one or more trans-acting replication functions is present in the same cell on an independent plasmid or integrated into the chromosome. It is demonstrated here that the trfA region alone can provide the trans-acting functions necessary for replication from oriRK2. Deletion of the trfB region in trans to an oriRK2 plasmid does not correlate with alteration in copy number or stability of the oriRK2 plasmid. Temperature-sensitive mutants defective in plasmid maintenance can apparently arise from mutations in both the trfA and trfB regions as indicated by complementation analysis of three different mutants. The trfA and trfB regions from two mutant plasmids have been cloned and used to allow a physically separate but functionally dependent oriRK2 plasmid to replicate at 30 °C. When the source of trfA and trfB is a trfB mutant the oriRK2 plasmid is temperature stable but is temperature sensitive when the source is a trfA mutant. This confirms that only trfA is essential for initiation at and elongation from oriRK2 which is probably the primary event in RK2 replication and suggests that the trfB region plays some other role in plasmid maintenance in plasmids carrying all three regions, oriRK2, trfA, and trfB.  相似文献

We describe genetic interactions between mutations in mgr, asp, and polo, genes required for the correct behaviour of the spindle poles in Drosophila. The phenotype of a polo 1 mgr double mutant is more similar to mgr than polo 1 , but the frequency of circular monopolar figures (CMFs) seen with either mutant alone is additive, suggesting that the two gene products are required for independent functions in the formation of bipolar spindles. The asp E3 mgr double mutant arrests much earlier in development than either mutant alone, indicative of a strong block to cell proliferation. We discuss whether the lack of microtubular structures in these cells reflects an extended mitotic arrest, or if it is a more direct consequence of the double mutant combination. A polo 1 asp E3 double mutant shows a dramatic synergistic increase in mitotic frequency. The loss of CMFs normally associated with the polo 1 phenotype suggests that the Asp microtubule-associated protein is required to maintain the structure of spindle poles. We speculate that Asp protein might be a substrate for the serine-threonine protein kinase encoded by polo. Received: 8 August 1998 / Accepted: 13 September 1998  相似文献

Summary Bf allele frequencies in a material of 172 unrelated Norwegians are given. Bf/HLA linkage relations in 49 informative matings with 178 children, and Bf/HLA association data of a material of 212 Bf-HLA haplotypes are presented. Of 171 informative meioses, there were no Bf-HLA-B recombinations, while 3 out of 158 Bf-HLA-A informative meioses showed recombination. There is significant association between the Bf F and the HLA-BW 35 allele.It is concluded that the Bf locus is situated on the HLA-B side of HLA-A within the HLA region, in very close proximity to HLA-B.  相似文献

Recently, two Bacillus thuringiensis strains were reported to synthesize parasporal inclusion bodies made not of the expected crystal (Cry) proteins but rather of the surface layer proteins (SLP) Sap (encoded by sap) and EA1 (encoded by eag), respectively. Whether the presence of the sap and eag genes is restricted to these two B. thuringiensis strains or ubiquitous in B. thuringiensis is unknown. We report here the distribution of the sap and eag genes in B. thuringiensis. Strains in the Bacillus cereus group were added for comparison purposes. We show that sap and eag are either present in tandem in 35% of the B. thuringiensis strains analysed and absent in 65% of the strains. When absent, a different tandem, the lytB/lytA cell wall hydrolase genes, is present. The distribution of the sap and eag S-layer and the lytB/lytA cell wall hydrolase genes is not species-specific in B. thuringiensis, B. cereus and Bacillus weihenstephanensis. Bacillus anthracis and Bacillus mycoides harbor sap and eag but not lytB/lytA. The sap, eag and lytB/lytA genes were absent in Bacillus pseudomycoides. Clearly, the distribution of the sap and eag S-layer and the lytB/lytA cell wall hydrolase genes in B. thuringiensis and in the Bacillus cereus group is mutually exclusive. We also showed that two genes involved in cell wall metabolism, csaA and csaB, are present not only upstream of the sap and eag S-layer genes, but also upstream of the lytB/lytA tandem in strains where sap and eag are absent. Bootstrapped neighbor-joining trees were inferred from the translated amino acid sequences of sap, eag and the tandem lytB/lytA, respectively.  相似文献

The size distributions of deletions, insertions, and indels (i.e., insertions or deletions) were studied, using 78 human processed pseudogenes and other published data sets. The following results were obtained: (1) Deletions occur more frequently than do insertions in sequence evolution; none of the pseudogenes studied shows significantly more insertions than deletions. (2) Empirically, the size distributions of deletions, insertions, and indels can be described well by a power law, i.e., f k = Ck b , where f k is the frequency of deletion, insertion, or indel with gap length k, b is the power parameter, and C is the normalization factor. (3) The estimates of b for deletions and insertions from the same data set are approximately equal to each other, indicating that the size distributions for deletions and insertions are approximately identical. (4) The variation in the estimates of b among various data sets is small, indicating that the effect of local structure exists but only plays a secondary role in the size distribution of deletions and insertions. (5) The linear gap penalty, which is most commonly used in sequence alignment, is not supported by our analysis; rather, the power law for the size distribution of indels suggests that an appropriate gap penalty is w k = a + b ln k, where a is the gap creation cost and blnk is the gap extension cost. (6) The higher frequency of deletion over insertion suggests that the gap creation cost of insertion (a i ) should be larger than that of deletion (a d ); that is, a i a d = In R, where R is the frequency ratio of deletions to insertions. Summary In Salmonella typhimurium, streptomycin resistance can occur by mutation at the strA or the strB mutants have altered ribosomes which are refractory to the drug in cell-free amino acid incorporation systems, and in 3H-dihydrostreptomycin binding studies. StrB mutants, unlike the strA mutants, are resistant to several aminoglycoside antibiotics and resistance is not due to a mutational change in the cell's protein synthetic machinery. Spectinomycin resistant mutants of S. typhimurium also fall into two classes, only one of which is ribosomal in mechanism. The spcA and spcB loci are closely linked to strA, aroC, and argG on the S. typhimurium linkage map.  相似文献

(L -Cys)n + N-base systems and (L -Cys)n + (L -Lys)n systems were studied by ir spectroscopy. It is shown that in the water-free systems, SH ?N ? S? ?H+N hydrogen bonds are formed. With the (L -Cys)n + N-base systems, both proton-limiting structures in the SH ?N ? S? ?H+N bonds have equal weight when the pKa of the protonated N-base is 2 pKa units larger than that of (L -Cys)n. The same is true with the water-free (L -Cys)n + (L -Lys)n system. Thus, with regard to the type of proton potentials present, these hydrogen bonds are proton-transfer hydrogen bonds showing very large proton polarizabilities. This is confirmed by the occurrence of continua in the ir spectra. Small amounts of water open these hydrogen bonds and increase the transfer of the proton to (L -Lys)n. In the (L -Lys)n + N-base systems, with increasing proton transfer the backbone of (L -Cys)n changes from antiparallel β-structure to coil. In (L -Cys)n + (L -Lys)n, the conformation is determined by the (L -Lys)n conformation and changes depending on the chain length of (L -Lys)n. Finally, the reactivity increase in the active center of fatty acid synthetase, which should be caused by the shift of a proton, is discussed on the basis of the great proton polarizability of the cysteine–lysine hydrogen bonds.  相似文献

The taxonomy of, and phylogenetic relationships among, African canaries typically assigned to the genus Serinus sensu lato (including the putative genera Alario, Pseudochloroptila, Serinops, Ochrospiza, Dendrospiza and Crithagra) were investigated, based on 823bp of the mitochondrial cytochrome b gene. Two clades emerged: (1) Palaearctic and Afrotropical taxa, including Serinus serinus, S. canaria, A. alario and the S. canicollis complex (S. c. canicollis, thompsonae and flavivertex), and (2) taxa endemic to the Afrotropics comprising Pseudochloroptila, Serinops, Ochrospiza, Poliospiza, Dendrospiza, Poliospiza and Crithagra spp. However, only clade one has jackknife and bootstrap support values above 50. The two clades were separated by Carduelis taxa, suggesting that Serinus sensu lato is not monophyletic. As it stands, the phylogeny does not support the recognition of Alario and Serinops and possibly Ochrospiza, Dendrospiza and Pseudochloroptila (Cape siskins) as distinct genera. Alario is sister to Serinus canicollis whereas the others are intermingled in a clade comprised primarily by Crithagra spp. The use of Serinus is confined to the Palaearctic canaries and allied species, and Crithagra for the strictly Afrotropical clade. Since the cytochrome b sequence divergence between the allopatric Cape (S. [c.] canicollis) and Yellow-crowned (S. [c.] flavivertex) canaries (2.8%) was similar to differences between other closely-related species and there are marked plumage differences between these taxa, we suggest that they be elevated to full species rank.  相似文献

 A population with birth rate function B(N) N and linear death rate for the adult stage is assumed to have a maturation delay T>0. Thus the growth equation N′(t)=B(N(tT)) N(tT) e d 1 TdN(t) governs the adult population, with the death rate in previous life stages d 1≧0. Standard assumptions are made on B(N) so that a unique equilibrium N e exists. When B(N) N is not monotone, the delay T can qualitatively change the dynamics. For some fixed values of the parameters with d 1>0, as T increases the equilibrium N e can switch from being stable to unstable (with numerically observed periodic solutions) and then back to stable. When disease that does not cause death is introduced into the population, a threshold parameter R 0 is identified. When R 0<1, the disease dies out; when R 0>1, the disease remains endemic, either tending to an equilibrium value or oscillating about this value. FLOWERING LOCUS M (FLM) is a MADS-domain gene that acts as an inhibitor of flowering in Arabidopsis. Here we describe the genetic interaction of FLM with genes in the photoperiod and autonomous flowering pathways. Although the sequence of FLM is most similar to that of FLC, FLM and FLC interact with different flowering pathways. It has been previously shown that flc lesions suppress the late-flowering phenotype of FRI-containing lines and autonomous-pathway mutants. However, flm lesions suppress the late-flowering phenotype of photoperiod-pathway mutants but not that of FRI-containing lines or autonomous-pathway mutants. Another MADS-domain flowering repressor with a mutant phenotype similar to FLM is SVP. The late-flowering phenotype of FLM over-expression is suppressed by the svp mutation, and an svp flm double mutant behaves like the single mutants. Thus FLM and SVP are in the same flowering pathway which interacts with the photoperiod pathway. Summary The ion and volume regulatory mechanisms ofTetrahymena pyriformis were studied in normal or hypotonic nutrient media and in isotonic inorganic media with different Na/K ratios, in conjunction with the effects of a general metabolic inhibitor (low temperature) and a specific inhibitor (iodoacetate). For K two mechanisms of active influx were found: The first is sensitive to IAc and seems to be the basic mechanism for the maintenance of the Ki/Ko gradient. The second is sensitive to cooling and related to the function of the contractile vacuole; it is also responsible for the high intracellular levels of K. The passive K efflux seems to be a basic factor for volume regulation, together with the contractile vacuole. It increases in hypotonic media and this seems to be related to structural changes of the membranes occurring in hypotonic media. For Na two mechanisms of active transport were also found: One for active Na efflux with highK m, which is associated with the contractile vacuole and another, for active Na influx with lowK m, which is inhibited by high levels of intracellular K.The electrochemical potentials of Na and K and the membrane potential (Cl Nernst potential) were also studied in isotonic inorganic media. The membrane is negatively polarized, except if Nao<5 mM when it becomes positive. Two calanoid copepod genera, Boeckella and Calamoecia have broadly similar geographic distributions in Australia but Boeckella is more common than Calamoecia. A field survey and literature search indicate that Boeckella is usually the first calanoid to colonize newly formed bodies of water, and Boeckella is more common in temporary pools and in high altitude environments. Boeckella usually produces larger clutches than Calamoecia, increasing its probability of establishment in lakes and ponds; calculations indicate that Boeckella's probability of establishment is far greater than that of Calamoecia. That Calamoecia is common on the North Island of New Zealand may be due to the presence of planktivorous fish; small species are more likely to survive in such environments. Calamoecia clutches may be able to move more often and farther than those of Boeckella but Calamoecia remains rare because its probability of establishment is low. Boeckella may be able to survive temporary pool environments not only because it can produce larger clutches, but also because many species in this genus are highly pigmented, and as such can survive brightly lighted environments. Patterns of abundance of North American diaptomids appear to be consistent with factors affecting those of Australian and New Zealand centropagids.  相似文献

A catalogue and a key to the non-lichenized, bulbilliferous basidiomycetes are given. The new genus Burgella is described for the lichenicolous B. flavoparmeliae, phylogenetically close to Sistotrema oblongisporum and Multiclavula. The genera Pneumatospora and Tricellulortus are placed in synonymy of Minimedusa, the new combination M. obcoronata is proposed, and the new facultative lichenicolous M. pubescens is described. The new facultative lichenicolous Burgoa angulosa is phylogenetically close to the generic type B. verzuoliana, whilst the new B. moriformis and B. splendens are provisionally included in the genus Burgoa. A Burgoa-like species in the Ceratobasidiaceae is left unnamed. Two new species of Marchandiomyces, M. buckii and M. nothofagicola, are described. As Marchandiomyces aurantiacus is phylogenetically more close to Erythricium than to Marchandiomyces, it is proposed to exclude it from that genus and to use the holomorphic generic name Marchandiobasidium for both anamorph and teleomorph of this species. The new genus Marchandiomphalina is introduced for the lichenized Omphalina foliacea, a taxon phylogenetically close to Marchandiobasidium. Abstract The consequences of ice and carbon dioxide anaesthetics on the survival and reproductive success of Drosophila simulans and Drosophila melanogaster were investigated after observations of high levels of mortality in D. simulans, possibly associated with brief chill coma. The association between brief chill coma and death is confirmed in female D. simulans but there is no mortality in male D. simulans or D. melanogaster of either sex. Mortality is unlikely to be associated with a strain specific cold intolerance because two geographically distinct populations of D. simulans were examined. In addition to the effect of ice anaesthesia, anaesthetizing newly‐eclosed male D. simulans with CO2 causes a reduction in fertility, which is evident 9–13 days after anaesthesia. This finding is important given that CO2 anaesthesia is a standard technique used in Drosophila and other insect cultures, and may have important consequences for studies of male fertility and sperm competition.  相似文献

The primary consideration in fermentor design is the supply of oxygen to the growing microorganisms. The oxygen-transfer characteristics of a two-stage splitcylinder airlift tower were compared to those of a similar single-stage airlift tower of equal liquid volume using a sodium sulfite–air system. At superficial gas velocities, from 720 to 1200 cm/min, no difference in KLa was apparent. The KLa was significantly larger in the two-stage tower for a gas velocity between 1200 and 2728 cm/min. At 2728 cm/min a KLa of 25.2 min?1 was achieved in the two-stage system, and at 2262 cm/min the two-stage tower had a 54% larger KLa than the single stage. A comparison of dispersion-volume based KLa showed a 27% larger value at a gas velocity of 2262 cm/min. The performance ratios for the two-stage tower were larger than those for the single-stage tower at oxygen-transfer rates greater than 180 mmol/liter hr. A comparison of the data with literature values is presented.  相似文献

The 10 Fagion associations bound to the western part of Czechoslovakia are conditioned both ecologically and chorologically in their species differentiation as well as geographical distribution. The division of the alliance into suballiances follows primarily the ecological phenomena. Within the Eu-Fagenion the associations form 3 groups conditioned by ecological factors.The Tilio platyphylli-Fagetum, Tilio cordatae-Fagetum, Melico-Fagetum and Carici pilosae-Fagetum represent associations of the submontane belt. The Tilio platyphylli-Fagetum is characterized by overlapping of Fagion and Carpinion species and by the absence of any Dentaria species due to chorological causes. The Tilio cordatae-Fagetum does not show any chorological phenomena in the species composition, however, it is limited to central, southern and western Bohemia only. The Melico-Fagetum is characterized by dominant Melica uniflora which is absent in the western and southern part of Bohemia. The Melico-Fagetum has even a more limited distribution occurring in northern and eastern Bohemia and in northern and central Moravia only. The Carici pilosae-Fagetum is characterized by Carex pilosa (dominant), Cephalanthera longifolia and Euphorbia amygdaloides; it is confined to the Carpathian province.The Dentario enneaphylli-Fagetum, Dentario glandulosae-Fagetum and Violo reichenbachianae-Fagetum represent a group of vicarious associations of the montane belt. The Dentario enneaphylli-Fagetum occurs mainly in the geographical province eská vysoina. Its eastern limit lies in the westernmost part of the Carpathian province where it forms a special subassociation and contacts the Dentario glandulosae-Fagetum. The latter association is characterized primarily by Dentaria glandulosa, a Carpathian endemic species. The Violo reichenbachianae-Fagetum is conditioned chorologically by the absence of any Dentaria species; it occurs in the mountains Kruné hory and Doupovské hory only.The Festuco-Fagetum is the single representant of the third group conditioned mainly by ecological factors.Nomenclature of species follows rothmaler et al.
