Summary Spores ofAspergillus sydowii, immobilized in 2.5% caleium alginate was used as inoculum in batch cultures for production of xylanase enzyme using xylan as the sole carbon source. Partially germinated mycelium from these entrapped spores produced significant amount of the enzyme in a short period of 24 hours and the same inoculum could be used repeatedly for at least 5 cycles with less than 10% loss of enzyme activity.  相似文献

Summary Alginate-entrappedChlorella demonstrate rapid uptake of phosphorus from synthetic growth medium in batch culture. Rates of phosphorus uptake demonstrated by immobilized algae were found to be much lower than those of non-immobilized cells. Uptake was dependent upon matrix stocking density, cell preculture conditions and cell viability, but not upon cell growth.  相似文献

Summary Tests made to utilize lignocellulosics as a substrate for the production of cellulases showed that the enzyme production from steam and explosion decompressed aspen wood (SED) by Tricoderma reesei RUT-C30 was low, and the enzyme system produced was deficient in exoglucanase and -glucosidase activities. Mixing this substrate with 10–20% pure cellulose lessened this deficiency and improved enzyme production. The enzyme system produced from the mixed substrate was rich in xylanase and had saccharifying ability equal to that produced in medium containing pure cellulose.  相似文献

A process for production of mold lactase was developed. Tests were carried out in pilot and industrial scale with an Aspergillus niger strain selected after screening a number of molds.A computer coupled autoanalyzer system was used for monitoring enzyme formation in the pilot fermentor. Lactase production was investigated using different pH- and temperature-profiles. A. niger lactase has an acid pH optimum, a high temperature optimum and good stability. It does not require any metal ions. It is suitable for immobilization for hydrolysis of lactose in acid whey.Three-fold enhancement of lactase production was obtained by mutagenizing A. niger using NTG as mutagenic agent.The lactases produced by the mutants have the same pH and temperature optima and stability but the growth properties of the mutants were different from those of the original strain.Sufficient specific activity of the enzyme preparation for immobilization was obtained by purifying the enzyme by selective adsorption on Na-Ca-silicate.  相似文献

Spinach thylakoid was immobilized by two different methods for the purpose of retention within a continuous-flow stirred tank reactor (CSTR). The glutaraldehyde crosslinked albumin polymer method completely inactivated the cyclic ATP photophosphorylation of thylakoid. In contrast, agarose-entrapped thylakoid retained about 17% of the activity of the cyclic photophosphorylation of non-immobilized thylakoid. This activity declined continuously during ATP production in the CSTR. Fifty percent of the initial activity was lost within about 5.5 h. Ascorbate was found to increase the stability of ATP photophosphorylation; about twice as much ATP was produced at the optimal ascorbate concentration of 5 mM. Under the optimal dilution rate of 2.36 h−1, about 60 μmol of ATP per mg chlorophyll were produced in 20 h by agarose-entrapped thylakoid in the CSTR. These results showed that, compared to non-immobilized thylakoid in batch operation, agarose-entrapped thylakoid produced only a low amount of ATP under continuous operation.  相似文献

The accumulation and volatilization of mercury by non-immobilized and immobilizedChlorella emersonii have been studied in batch culture systems. Reduction in the mercury concentration in the growth medium by non-immobilized cells was highly dependent on inoculum density, whilst reduction in mercury concentration by immobilized cells was rapid at all inoculum densities. Mercury accumulation by immobilized cell biomass was significantly greater than by non-immobilized cells with 106 and 105 cells bead–1 or ml–1. Volatilization of mercury by non-immobilized cell systems was greatest at higher inoculum densities, whereas more mercury was volatilized from immobilized cell systems at lower inoculum densities, and was greatest with unstocked alginate beads. Thus, in immobilized systems, mercury removal from solution is complex and involves mercury accumulation by the cells and volatilization by the matrix and cells. Further studies of mercury accumulation and volatilization by unstocked immobilization matrices revealed that agarose volatilized much less mercury than alginate or agar. The precise mechanism of mercury volatilization by alginate remains unclear, though it is thought to be a chemical effect.  相似文献

Summary Diacetyl production by (Citr*)Lactococcus lactis subsp.lactis 3022 was found to be an oxygen-dependent reaction. The diacetyl production by the cells immobilized in conventional Ca-alginate gel beads (Diameter: 3 mm) was lower than that of the cells immobilized in Ca-alginate gel fibers (Diameter: 0.2 mm), probably because oxygen transfer to the immobilized cells is better in gel fibers than in gel beads.  相似文献

Summary Various materials were tested and polycarbonate was chosen as the support material for attachment and film growth ofP. chrysogenum in rotating disc fermenters. Summary Enhanced rates of ethanol production byPachysolen tannophilus from D-xylose were obtained by performing the fermentation with recycled cells in suspension culture or immobilized in a Ca-alginate gel. Fermentation under these conditions did not require aeration. Summary A fungal strain isolated from soil and identified asAspergillus athecius, when grown on moistened wheat bran produced large amounts of extracellular invertase. Most of the invertase from the moldy bran was easily extracted by low ionic strength buffer (0.005 M, pH 5.7). The crude invertase immobilized on DEAE cellulose showed not only increased activity (45%) but also greater thermal and storage stability than the free enzyme. The free and the bound enzymes showed a temperature optimum of 50–55°C and a pH optimum of 5.7 and 4.8 respectively. The Km app. of the bound enzyme was lower than that of the free enzyme.  相似文献

Summary Bacillus subtilis CD4, when grown in nutrient broth or minimal medium in presence of xylan, produced extracellular xylanase that hydrolyzed xylan optimally at pH 5. The enzyme was induced by xylan, xylose and glucose. Addition of xylose or glucose in xylan containing medium did not affect enzyme production. The structural gene encoding xylanase was cloned and expressed in E. coli. The recombinant enzyme exhibited similar properties like that of native enzyme including resistance to repression by xylose and glucose.  相似文献

Summary Alpha-amylase production byBacillus licheniformis M27 in submerged fermentation was reduced from 480 to 30 units/ml when soluble starch concentration in medium was increased from 0.2 to 1.0%. In contrast, the enzyme production increased by 29 times even with 42 fold increase in the concentration of soluble starch and other starchy substrates in solid state fermentation system. The data establish regulation of the enzyme formation by enzyme end-product in submerged fermentation and ability of solid state fermentation to minimize it significantly. Summary Cryptococcus albidus var. albidus, strain IMAT-4735, grown on suitably treated oil-mill vegetation waters produced pectic enzyme. Growth and enzyme production were favoured by addition of calathide residues from sunflower cultivation, a low-cost pectic material, to the vegetation waters. Ultrafiltration of the centrifuged fermentation broth made it possible to almost completely recover the enzyme activity produced. The enzyme concentrate from ultrafiltration was directly reused in the me chanical extraction of olive oil.  相似文献

Summary Diacetyl production by (Citr+)Lactococcus lactis subsp.lactis 3022 cells immobilized in Ca-alginate fine fibers with single layer in the presence of catalase was three times higher than that in the absence of catalase. A co-immobilized culture system of the lactic acid bacterial cells (outer) and the homogenized bovine liver (inner layer) in Ca-alginate fibers with double gel layers was developed. Summary Aspergillus niger glucoamylase was adsorbed to -naphthyl cotton cloth by hydrophobic interaction. The adsorbed enzyme was cross-linked with glutaraldehyde. The immobilized glucoamylase exhibited greater pH dependence though the optimal pH did not change. The immobilized glucoamylase in a packed bed column completely hydrolysed 5% soluble starch at a specific velocity of about 4. Used naphthyl cloth could be regenerated by heating in 2 N NaOH at 100°C for 1 hour.  相似文献

Summary Candida cylindracea lipase adsorbed to silica gel produced a variety of flavor esters when hydrated and shaken in n-heptane containing substrates. Scale-up production of ethyl butyrate was examined in a packed column with recycling of n-hexane containing substrates. Increased substrate concentrations were stimulatory up to a point after which inhibition and enzyme destabilization in repeated runs occurred.  相似文献

Summary Saccharomyces anamensis having -galactosidase activity, has been immobilized in calcium alginate gel matrix that retained 78.6% enzyme activity to that of native cells. Optimum pH(7.0) was negligibly affected by immobilization. Km values for immobilized and native cells were 119 mM and 102 mM respectively. Protective agents like dithioerythritol, bovine serum albumin, enhance the enzyme activity when added prior to immobilization. Immobilized cells can be stored in refrigeration(4°C) for 42 days without a significant loss of enzyme activity.  相似文献

Summary The properties of glucose isomerase in native, heat-treated and immobilized cells of Streptomyces kanamyceticus after heat and mineral treatment have been compared. The optimum pH for glucose isomerase in native cells was shifted from 8.2 to 8.6 by heat treatment and immobilization. There is no change in the optimum temperature (90°C) for activity of the enzyme by the above treatment. Heat-treated cells and immobilized cells show greater pH and thermal stability of the enzyme. The Km values of the enzyme of native cells, heat-treated cells and immobilized heat-mineral-treated cells are 208 mM, 212 mM and 166 mM respectively; Mg++ and Co++ enhance the activity of isomerase in all cases.  相似文献
