Seventy-six day-8 bovine blastocysts were transferred by a paracervical non-surgical process to determine its feasibility as an alternative method of bovine embryo transfer. Forty-seven of the 76 transfers resulted in pregnancy (62%), as determined by rectal palpation at 45 to 90 days of gestation. This indicates that embryo transfer via the paracervical method is a viable alternative to surgical and non-surgical transfer in cattle.  相似文献

The objective of this study was to enhance procedures for producing piglets derived from in vitro-produced (IVP) pig embryos by non-surgical embryo transfer (ET). The effects of insertion length for the catheter, asynchrony between the age of donor IVP blastocysts and the recipient estrous cycle, and volume of transfer medium were investigated. The IVP blastocysts at 5 days after in vitro fertilization were placed into porcine zygote medium (PZM)-5 supplemented with 10% (v/v) fetal bovine serum (PZM+FBS) in a 0.25 mL plastic straw (21-40 blastocysts per straw) and then transferred into one uterine horn of recipients using the Takumi(?) catheter for deep intrauterine insertion. Successful production of piglets derived from IVP embryos was achieved following non-surgical ET when the catheter was inserted at more than 30 cm anterior to the spiral guide spirette. The efficiency of piglet production (percentage number of piglet(s) born based on the number of embryos transferred) was greater (P<0.05) in recipients whose estrous cycle was asynchronous to that of donors with a 1-day delay (8.3%) than in those with a 2-day (1.5%) or 3-day (0.9%) delay, while pregnancy and farrowing rates (10-40%) did not differ among treatments. When blastocysts were transferred into recipients with 1.0 or 2.5 mL PZM+FBS, there were no significant differences in farrowing rate (30-40%) or average litter size (4.5-6.7) between treatments. The results of the present study indicate that the insertion length of the deep intrauterine catheter and the degree of asynchrony between donor embryos and recipient estrous cycle influenced on pregnancy and birth outcome following non-surgical transfer of IVP blastocysts.  相似文献

The effects of dietary protein level on age and weight at onset of puberty were studied in the Mexican tropics using 12 Brown Swiss and 12 Zebu heifer calves in a completely randomized block design. The calves were weaned at 3 mo of age. They were confined in individual pens and fed either an adequate protein (12.8%) or a high protein (16.4%) diet ad libitum through the onset of puberty, which was defined as the first ovulation. To detect the first ovulation, heifers were palpated at 8 mo of age and then every 2 wk. At the detection of a developing follicle or any behavioral sign of estrus, they were palpated daily. Ovulation was confirmed by palpation of a corpus luteum. A blood sample for the determination of plasma progesterone concentration was taken with each palpation. There was a significant effect of breed on age at the onset of puberty, which was 9.4 and 12.3 mo for the Brown Swiss and Zebu heifers, respectively. Weight at puberty was 233 kg, independent of breed. Dietary protein had no effect on the onset of puberty. Brown Swiss heifers reach puberty earlier than Zebu heifers. Heifers in the tropics fed according to NRC recommendations reach puberty at ages comparable to those in temperate climate. This suggests that, if managed properly, calving age for Brown Swiss and Zebu heifers can be 24 and 27 mo, respectively. This is a considerable improvement from 36 to 48 mo of age now common in the area of study.  相似文献

The liveweights of 100 females sired by Holstein (H), Jersey (J) or Brown Swiss (BS) bulls out of Holstein-Friesian x Gir dams of 1/2 to 3/4 Holstein-Friesian fraction were compared. The animals were kept in a single herd under the same management. The data were analyzed separately for four age categories by least squares techniques. The models for cows and heifers older than 18 months included the fixed effects of breed of sire, Bos taurus fraction, season of weighing, days in milk class (for cows only), pregnancy status class, year and the random effect of animal nested within breed of sire x B. taurus fraction subclasses. The models for younger heifers included the same effects except for lactation and pregnancy status. Based on 2937 observations, the mean weights for 0- to 6-month-old calves for the groups with H, J or BS sires were 88 +/- 1, 77 +/- 1 and 75 +/- 1 kg, respectively, the mean weights for 1-year-old heifers were 168 +/- 2, 159 +/- 2 and 155 +/- 1 kg, for 2-year-old heifers, 327 +/- 4, 303 +/- 4 and 319 +/- 4 kg, for 3-year-old heifers, 380 +/- 5, 369 +/- 8 and 390 +/- 4 kg and for cows, 464 +/- 3, 413 +/- 2 and 478 +/- 2 kg. Cubic growth curves, which were different in the three sire breed groups, satisfactorily explained the cow weight changes with age (R(2)>or=0.98). Maximum weight was attained at 7.8, 9.5 and 9.2 years in cows with H, J or BS sires. Although breed of sire effects were not significant in heifers (P>0.05), those with H sires were the heaviest up to two years of age. The females with BS or J sires had similar weights up to one year of age, but thereafter the former reached similar weights as the females with H sires and declined less after attaining the maximum weight. The cows with J sires were the lightest at all ages. Since previous results showed similar protein and fat yields per day of calving interval in the three breed of sire groups, it is suggested that the lighter J crosses may be more economic than the other groups on account of their likely lower feed maintenance costs.  相似文献

Lifetime dairy production, reproduction and growth traits of 75 females sired by Holstein, Jersey or Brown Swiss bulls and Holstein-Friesian x Gir dams of 1/2 to 3/4 Holstein-Friesian fractions were compared. The animals were in a single herd under the same management. Milk, fat and protein yields, concentrates fed, reproduction, and weights were recorded throughout the lifetime of the cows. The data were analyzed by least squares techniques under models including the fixed effects of breed of sire, Bos taurus fraction and year of birth. Herd lifes for Holstein, Jersey and Brown Swiss crosses were 6.006 +/- 0.812, 8.129 +/- 0.863 and 7.247 +/- 0.777 years. Milk yields per day of herd life were 7.150 +/- 0.266, 6.757 +/- 0.282 and 6.249 +/- 0.254 kg. Weights of cull cows sold were 458 +/- 15, 415 +/- 15 and 457 +/- 13 kg. Based on these and on previously reported results of the same experiment, intakes of roughage and pasture were estimated from energy requirements. Lifetime expenditures on concentrates, roughages, pastures, milking, reproduction, and heifer rearing were calculated based on mean performance of each breed of sire, as well as on receipts from animals and milk sold (the latter with four sets of prices of protein, fat and carrier). The conclusion was that in systems of artificial female calf rearing and male calf wastage, the Jersey crosses appear to offer important economic benefits to farmers, which would be even greater if payment on milk protein and fat becomes effective.  相似文献

The international Brown Swiss cattle population pedigree was studied to measure genetic variations and to identify the most influential animals. Twenty-two countries provided pedigree information on 71 497 Brown Swiss bulls used for artificial insemination (AI). The total number of animals with the pedigree is 181 094. The mean inbreeding coefficient for the pedigree population was 0.77%. There was, in most cases, an increase in the mean inbreeding coefficient, with the highest value at 2.89% during the last 5-year period (2000 to 2004). The mean average relatedness for the pedigree population was 1.1%. The effective population size in 2004 was 204. There was notable variation between average generation intervals for the four parental pathways. The longest average generation interval, at 8.73 years, was observed in the sire–son pathway. The average generation interval for the whole population was 6.53 years. Most genetically influential individuals were sires. The highest contributing founder was a sire with a 3.22% contribution, and the highest contributing founder dam made a contribution of 1.75%. The effective number of founders and the effective number of ancestors were 141 and 88, respectively. The study showed that genetic variation within the pedigree population has been decreasing over recent years. Increasing the number of AI bulls with a low individual coefficient of inbreeding could help to maintain a good level of genetic variation in the Brown Swiss population.  相似文献

Background

Genomic selection estimates genetic merit based on dense SNP (single nucleotide polymorphism) genotypes and phenotypes. This requires that SNPs explain a large fraction of the genetic variance. The objectives of this work were: (1) to estimate the fraction of genetic variance explained by dense genome-wide markers using 54 K SNP chip genotyping, and (2) to evaluate the effect of alternative marker-based relationship matrices and corrections for the base population on the fraction of the genetic variance explained by markers.

Methods

Two alternative marker-based relationship matrices were estimated using 35 706 SNPs on 1086 dairy bulls. Both pedigree- and marker-based relationship matrices were fitted simultaneously or separately in an animal model to estimate the fraction of variance not explained by the markers, i.e. the fraction explained by the pedigree. The phenotypes considered in the analysis were the deregressed estimated breeding values (dEBV) for milk, fat and protein yield and for somatic cell score (SCS).

Results

When dEBV were not sufficiently accurate (50 or 70%), the estimated fraction of the genetic variance explained by the markers was around 65% for yield traits and 45% for SCS. Scaling marker genotypes with locus-specific frequencies of heterozygotes slightly increased the variance explained by markers, compared with scaling with the average frequency of heterozygotes across loci. The estimated fraction of the genetic variance explained by the markers using separately both relationships matrices followed the same trends but the results were underestimated. With less accurate dEBV estimates, the fraction of the genetic variance explained by markers was underestimated, which is probably an artifact due to the dEBV being estimated by a pedigree-based animal model.

Conclusions

When using only highly accurate dEBV, the proportion of the genetic variance explained by the Illumina 54 K SNP chip was approximately 80% for Brown Swiss cattle. These results depend on the SNP chip used and the family structure of the population, i.e. more dense SNPs and closer family relationships are expected to result in a higher fraction of the variance explained by the SNPs.  相似文献

In Exp. 1, embryo survival rates of 45 and 47% were recorded after artificial insemination and ipsilateral transfer respectively. In Exp. 2, pregnancy rates of 62 and 60% were recorded after artificial insemination and contralateral transfer to inseminated recipients respectively. In this experiment the contralateral transferred embryo survival rate was 44%. Transferred embryo survival was lower overall when donors and recipients were out of phase by 1 day than when exactly synchronous.  相似文献

The white belt pattern of Brown Swiss cattle is characterized by a lack of melanocytes in a stretch of skin around the midsection. This pattern is of variable width and sometimes the belt does not fully circle the body. To identify the gene responsible for this colour variation, we performed linkage mapping of the belted locus using six segregating half-sib families including 104 informative meioses for the belted character. The pedigree confirmed a monogenic autosomal dominant inheritance of the belted phenotype in Brown Swiss cattle. We performed a genome scan using 186 microsatellite markers in a subset of 88 animals of the six families. Linkage with the belt phenotype was detected at the telomeric region of BTA3. Fine-mapping and haplotype analysis using 19 additional markers in this region refined the critical region of the belted locus to a 922-kb interval on BTA3. As the corresponding human and mouse chromosome segments contain no obvious candidate gene for this coat colour trait, the mutation causing the belt pattern in the Brown Swiss cattle might help to identify an unknown gene influencing skin pigmentation.  相似文献

Adopting a mating system involving two different Robertsonian translocations with monobrachial homology, we studied the early development of mouse embryos trisomic or tetrasomic for chromosome 11. A developmental delay of 12-24 hours was evident in trisomic embryos at embryonic day (E)7.5, whereas tetrasomic embryos apparently had stopped growth by E6.5 without formation of extraembryonic structures. This extremely severe developmental abnormality found in tetrasomic embryos is similar to that reported in embryos having two active X chromosomes in extraembryonic cell lineages. Autosomal tetrasomy, but not autosomal trisomy, can lead to such early developmental errors. Thus, a reasonable inference would be that the X chromosome is twice as active as the autosome. Probably, the X chromosome became upregulated in response to the evolutionary necessity of minimizing haplo-insufficiency brought about by miniaturization of the Y chromosome.  相似文献

Surgical transfer of in vivo produced conventionally frozen-thawed embryos of farmed European polecat (Mustela putorius) was investigated as a part of an ex-situ preservation program which has the long-term aim of developing a genome resource bank for the endangered European mink (Mustela lutreola). Eighteen oestrous yearling European polecat donors were mated once daily on two consecutive days using 13 fertile males. The donors were surgically flushed for embryos 8-9 days after the first mating. The embryo recovery rate was 60% (116 embryos/193 corpora lutea). The embryos were cryopreserved with 1.5 M ethylene glycol in a programmable freezer using a conventional slow freezing protocol. The thawed embryos were surgically transferred either after dilution with 0.5 M sucrose or directly without removal of ethylene glycol. To induce ovulation, eight recipient females were mated once daily on two consecutive days with vasectomized males starting 7 or 8 days before embryo transfer. The recipients received 7-11 embryos each and three recipients delivered a total of nine pups after a gestation length of 44-46 days. The embryo survival rate was 10% (9 pups/93 frozen embryos). This report describes the first successful cryopreservation of embryos in the Mustelidae family resulting in viable offspring. The low embryo survival rate, however, indicates that the freezing-thawing protocol needs to be improved.  相似文献

The present theoretical analysis deals with biomechanics of the self-propulsion of a swimming sheet with heat transfer through non-isothermal fluid filling an inclined human cervical canal. Partial differential equations arising from the mathematical modeling of the proposed model are solved analytically. Flow variables like pressure gradient, propulsive velocity, fluid velocity, time mean flow rate, fluid temperature, and heat-transfer coefficients are analyzed for the pertinent parameters. Striking features of the pumping characteristics are explored. Propulsive velocity of the swimming sheet becomes faster for lower Froude number, higher Reynolds number, and for a vertical channel. Temperature and peak value of the heat-transfer coefficients below the swimming sheet showed an increase by the increment of Brinkmann number, inclination, pressure difference over wavelength, and Reynolds number whereas these quantities decrease with increasing Froude number. Aforesaid parameters have shown opposite effects on the peak value of the heat-transfer coefficients below and above the swimming sheet. Relevance of the current results to the spermatozoa transport with heat transfer through non-isothermal cervical mucus filling an inclined human cervical canal is also explored.  相似文献

Multiple ovulation-embryo transfer (MOET) protocols for farmed fallow deer (Dama dama ) were investigated in a series of 3 experiments. A total of 37 donors, of either European (D.d. dama ; n = 30) or Mesopotamian hybrid (D.d. mesopotamica x D.d. dama ; n =7) genotype, each received an intravaginal silastic device containing 0.3 g progesterone (CIDR((R))-type G device) for 14 d and injections of 0.5 units ovine FSH (8 x 0.06 unit injections from Days 10 to 14 of device insertion) and 100 IU PMSG (either with the first or last FSH injection). All donors received laparoscopic intrauterine inseminations of fresh semen (50 x 10(6) spermatozoa) from a Mesopotamian sire 36 h after withdrawal of CIDR((R)) devices. Embryos were recovered by laparotomy on Day 6 (Day 0 = estrus). Mean ovulation rates for the 3 experiments were 8.1, 9.8 and 7.0, with no effect of PMSG timing (P>0.10). However, embryo recovery rates, albeit low throughout the study (29.6%), were significantly improved with later PMSG administration (33.9 vs 20.1%; P<0.05). Hybrid and European donors performed in a similar manner. A range of embryo development stages was recovered throughout the study. In 2 experiments laparoscopic transfer of embryos to 48 recipient does treated previously with intravaginal CIDR((R)) devices for 14 d yielded a total pregnancy rate of 37.5%. In the experiment with fresh embryos, the use of clenbuterol to reduce uterine turgidity resulted in a higher proportion of does conceiving (3 4 ; 75%) compared with that of the untreated does (0 6 , 0%; P<0.05). In the second experiment, in which all the does routinely received clenbuterol, 10 19 (53%) and 5 19 (26%) does conceived following the transfer of fresh and cryopreserved embryos, respectively (P<0.05). While the overall efficiency of the MOET program was low (equivalent of 0.9 to 1.0 surrogate pregnancies per donor), improvements in the recovery rate of transferable embryos have considerable potential for genetic improvement of farm stock and captive propagation of endangered Mesopotamian fallow deer through maternal surrogacy programs.  相似文献

A pituitary extract was used to induce multiple ovulations in mares to determine whether day-7 embryos from multiple ovulators were viable as indicated by their ability to develop when transferred to recipients. There were more ovulations/donor for induced multiple-ovulating mares than for control single-ovulating mares (4.6 +/- 0.5 vs 1.0 +/- 0.0; n=14). The embryo collection rate per ovulation was similar for multiple ovulators (0.6 +/- 0.1 embryos/ovulation) and single ovulators (0.7 +/- 0.1). The embryo collection rate per donor, therefore, was higher (P<0.01) for the multiple ovulators (2.9 +/- 0.7 vs 0.7 +/- 0.1). The transfer success rate per embryo at day 21 was different (P<0.05) among recipients which received an embryo from control single-ovulating donors (7 8 ), multiple ovulators from which a single embryo was recovered (2 2 ), and multiple ovulators from which multiple embryos were recovered (9 19 ). The recipient pregnancy rate/donor at day 21 was 88% (7 8 ) for single-ovulating controls and 138% (11 8 ) for induced multiple ovulators. Results indicate that the survivability of day-7 embryos from multiple-ovulating donors was reduced. However, despite the reduced survival rate/embryo, the number of pregnant recipients/donor was increased by induction of multiple ovulations because of the increased number of embryos available for transfer.  相似文献

Apoptosis occurs during preimplantation development in both in vivo- and in vitro-produced embryos, and it may contribute to embryonic loss. The present study investigated the development of porcine nuclear transfer (NT) embryos reconstructed by using fetal fibroblasts as compared to embryos produced by in vitro fertilization (IVF). The onset and the frequency of apoptosis in NT and IVF embryos were examined via morphological and nuclear changes and TUNEL assay. The NT blastocysts had a similar number of nuclei as compared to IVF blastocysts and appeared to be morphologically similar. Relative to IVF embryos, the NT embryos had a lower cleavage rate (42.7% vs. 71.0%) and a lower developmental rate (11.1% vs. 28.6%) to the blastocyst stage. The earliest positive TUNEL signals were detected in the NT embryos on Day 5 of culture. The percentage of cells undergoing apoptosis in the NT embryos was higher than that of the IVF embryos and increased with time in vitro. Some of the abnormal morphological changes observed during early development related to apoptosis. Cytoplasmic fragmentation, developmental arrest, and nuclear condensation were typical characteristics of embryos undergoing apoptosis. Some mechanisms of the apoptotic pathway were triggered by changes in the NT embryos. The developmental rates of NT embryos might be improved by identifying specific apoptotic pathways and then intervening in these pathways to improve development.  相似文献
