50年代以来,国际上相继开展了一些关于化学信息在小家鼠繁殖中作用的研究,并在实验室中揭示了一些反应:如集群雌鼠气味使雌鼠性周期延长的Lee-Boot效应,雌鼠气味能解除集群雌鼠对性周期的抑制、并促使它们同期发情的Whitten效应,以及陌生雄鼠气味中断妊娠的Bruce效应等等。但是,在成鼠间起作用的几种化学信息,是否能被幼鼠感受,对幼鼠的发育和性成熟是否产生影响,还是一个未知的问题。 影响小鼠发育的因素很多,除环境因子外,近来还出现了有关社群因素中外激素对小鼠性成熟影响的报道,发现雄鼠或给予雄激素的雌鼠有促进幼雌鼠性成熟的作用,范志勤发现父本气味和陌生雄鼠气味对幼雌鼠性成熟均有促进作用。但是,目前关于种内雌鼠气味、集群雌鼠气味、幼鼠气味影响性成熟的研究尚不多见。有关各种气味对性成熟效应的比较的报道亦少。本文的目的在于通过实验方法,阐明种内的几种化学信息,诸如父本、陌生雄性、雌性、集群雌性、幼雌等几种气味,对幼雌小鼠性成熟的影响,并据此了解幼鼠识别这几种信息的能力。  相似文献

目的比较制备大鼠妊娠模型时在合笼前进行阴道涂片性周期筛选与不筛选两种方法对大鼠受孕率、受孕速度(合笼次数)的影响。方法 26只SD雌性大鼠随机分为阴道涂片性周期筛选合笼组(涂片组)、不筛选合笼组(自由组),筛选组雌鼠在合笼前进行阴道涂片性周期筛选,筛选处于动情期或动情前期的雌鼠进行合笼;自由组不进行性周期筛选,直接合笼;实验中大鼠分组、合笼均采用单因素完全随机方法。结果两种合笼方法对大鼠受孕率和受孕速度差异不显著(P〉0.05)。结论鉴于两种合笼方法在制备大鼠妊娠模型的受孕率及受孕速度方面无差异,同时为了减少人为干预对雌鼠的刺激和影响,建议在制备大鼠妊娠模型时考虑采用自由合笼法。  相似文献

目的研究海藻酸钠寡糖对小鼠免疫及抗氧化活性的影响。方法采用不同浓度的海藻酸钠寡糖分别对小鼠连续灌胃15d后,测量小鼠生长性能、小鼠胸腺和脾脏指数以及小鼠血浆中SOD和GSH的活性。结果海藻酸钠寡糖能促进小鼠的生长性能,提高小鼠的胸腺和脾脏指数。其中给小鼠灌注高浓度的海藻酸钠寡糖15d后,与对照组相比,小鼠的特定生长率和增重率分别提高了37.0%和48.5%(P0.01),小鼠胸腺和脾脏指数分别提高了18.8%和21.7%(P0.01)。海藻酸钠寡糖还能增加小鼠的抗氧化活性,与对照组相比,灌注高浓度海藻酸钠寡糖组小鼠的SOD和GSH-Px活性分别提高了92.6%和35.9%(P0.01)。结论海藻酸钠寡糖能促进小鼠的生长性能,增强小鼠的免疫和抗氧化功能。  相似文献

输卵管液是由输卵管粘膜上皮细胞分泌和输卵管血管渗出的混合液。人和浦乳动物的输卵管液除维持输卵管内渗透压和调节pH值外,还对生殖细胞的进一步成熟、受精、受精卵的卵裂和运输有极其重要的作用。输卵管液内含有水、无机盐、糖、蛋白质和各种氨基酸。有些内含物质还和动物的性周期有关。但有多种生理功能和生殖功能的维生素A、D、E在输卵管液中的含量未见报道。我们采用荧光分析法对不同性周期兔输卵管液中和血清中维生素A、D、E的含量进行了测定与比较。  相似文献

目的:研究枸杞粗多糖(Lycium Barbarum Polysaccharide,LBP)在小鼠体内的抗氧化作用。方法:用高(400mg/kg.d)、中(200mg/kg.d)、低(100mg/kg.d)剂量的枸杞粗多糖生理盐水溶液对D-半乳糖(100mg/kg.d)之衰老模型小鼠和正常小鼠灌胃。结果:枸杞粗多糖能较显著(P0.01)提高小鼠血清、肝脏及脑组织中SOD活性,降低MDA含量;极显著(P0.01)提高正常小鼠常压耐缺氧能力和游泳抗疲劳能力;此外小鼠的脾指数和胸腺指数均得到显著提高,表明枸杞粗多糖对提高小鼠的机体免疫水平具有重要的促进作用。结论:枸杞粗多糖对小鼠具有显著的抗氧化、抗衰老作用。  相似文献

为探讨天门冬多糖对免疫功能低下小鼠的免疫调节作用,本实验建立环磷酰胺诱导免疫抑制小鼠模型。通过称重计算脾脏指数和胸腺指数;MTT法检测T、B淋巴细胞增殖反应;双抗夹心ELISA方法检测小鼠血清中IL-2和IL-4水平,测定天门冬多糖对免疫抑制小鼠免疫功能的调节作用。结果表明天门冬多糖能够提高免疫抑制小鼠的脾脏指数和胸腺指数,在ConA或者LPS刺激下提高T、B淋巴细胞增殖率,提高血清中IL-2和IL-4水平。天门冬多糖对环磷酰胺诱导的免疫抑制小鼠有一定的保护作用。  相似文献

为研究千斤拔多糖对正常及免疫低下小鼠免疫功能的调节作用,该研究选用SPF级BALB/c小鼠,免疫抑制小鼠采用隔天皮下注射环磷酰胺(40 mg·kg~(-1))5次,通过测定脾脏、胸腺质量及计算脏器指数,采用碳粒廓清法计算单核巨噬细胞吞噬功能,在鸡红细胞免疫后测定小鼠血清溶血素抗体水平,同时观察高、低剂量千斤拔多糖(剂量分别为500、1 000 kg·d~(-1))对正常小鼠及免疫低下小鼠免疫调节的影响。结果表明:千斤拔多糖高、低剂量组对正常及免疫低下小鼠均有不同程度的增加脾脏指数、胸腺指数的作用;千斤拔多糖高、低剂量组对正常及免疫低下小鼠均能提高廓清指数,但没有统计学意义;千斤拔多糖高剂量组对正常及免疫低下小鼠的血清溶血素抗体水平都有显著性提高。这说明千斤拔多糖能有效提高正常及免疫低下小鼠免疫功能的作用。  相似文献

Ultrasound imaging in reproductive sciences offers new opportunities regarding optimization of the induction of the sexual cycle and ovulation, superovulation regimes, contraception programs, semen collection and testicular sperm extraction techniques, ovum pick up and ovarian transplantation procedures, as well as the application of artificial insemination, embryo collection and transfer. In non-domestic species, most of which lack basic data, ultrasonography is an ideal tool to study reproductive biology in both captive and wild populations. The use of this imaging modality led us to develop new, or modify established, reproductive technologies. Ultrasonography has been an integral part of over 200 assisted reproduction procedures in 17 mammalian species performed by our research team between 1992 and 1999. These procedures included the initial characterization of sexual cycles, hormonal cycle induction, semen collection by electroejaculation or manual stimulation, non-surgical artificial insemination (AI), non-surgical embryo transfer and temporary hormonal contraception. For these investigations, a variety of newly developed equipment was applied and species-specific hormonal treatments designed. We used several commercial and customized ultrasound systems with a variety of technical features. Some relevant improvements of these applications will be described and the role of ultrasonography elucidated to.  相似文献

The frequency of heterozygote carriers of (risk zone, alleles of FMR1 gene (40-47 CGG-repeats) was significantly higher in group of patients with ovarian dysfunction than in control group I. The tendency for higher frequency of those alleles was observed in patients with "poor response" to superovulation induction in IVF cycles. The average number of oocytes and follicles, which was obtained after stimulation of superovulation, was significantly decreased in FMR1 gene "risk zone" alleles carriers compared to patients with normal alleles of FMR1 gene. The average general dosage of exogenous gonadotrophin, necessary for superovulation induction was significantly higher in heterozygote carriers of FMR1 gene "risk zone" alleles than in patients with normal genotype. Thereby, the FMR1 gene "risk zone" alleles can be one of the hereditary susceptibility factors of impairment nature and stimulated ovulation.  相似文献

The objective of this study was to estimate the relative effects of genetic and phenotypic factors on the efficacy and efficiency of superovulation for Holstein-Friesian cows reared in Brazil. A database, established by the Associacao Brasileira de Criadores de Bovinos da Raca Holandesa, consisting of a total of 5387 superovulations of 2941 cows distributed over 473 herds and sired by 690 bulls was used for the analysis. The records were analyzed by MTDFREML (Multiple Trait Derivative-Free Restricted Maximum Likelihood), using a repeatability animal model. The fixed effects included in the model were contemporaneous group (veterinarian, herd, year and season of the superovulation); number of semen doses; cow age; and superovulation order. The estimated repeatability of the number of the transferable embryos was low (0.13), and the estimated heritability was 0.03. These results indicate that environmental factors play a critical role in the response of a cow to a superovulation treatment. There is little evidence that future responses to superovulation by individual females can be predicted by previous treatment(s) or that superovulation response is an heritable trait.  相似文献

Induction of ovulation with pregnant mare's serum (PMS) and the timing of human chorionic gonadotropin (hCG) injection on chromosomal normality were examined in preimplantation hamster embryos. Two separate experimental trials were done. The first compared superovulation (SO, PMS on Day 1 of the cycle followed by hCG on Day 4) to natural ovulation. Natural mating was used. In the second series of trials, precocious superovulation (PSO, PMS on Day 1 followed by hCG on Day 3) was used. Since there is poor sperm transport in PSO females, direct uterine artificial insemination (AI) was used to achieve fertilization. The control animals in the second series of trials were naturally ovulating females subjected to the artificial insemination procedure. Of 785 embryos analyzed in the SO group, 9 (1.1%) were aneuploid (5 hyperploidy and 4 hypoploidy) and 8 (1.0%) showed triploidy. In the PSO group, artificial insemination resulted in a normal development rate of 85.5% up to the 2-cell stage. A total of 2.6% karyotypically abnormal embryos, consisting of 5 (1.1%) aneuploid and 7 (1.5%) polyploid, were found among 460 embryos examined in PSO females. No significant difference in the incidence of chromosomal abnormalities was observed between the stages of development. The overall incidence of chromosomal imbalance in hormonally treated females was not significantly different from that in controls (2.2% in SO cycles vs. 1.2% in natural cycles, 2.6% in PSO with AI vs. 2.4% in natural cycles with AI). These results indicate that PMS-hCG treatment has no adverse effect on the chromosomal integrity of hamster preimplantation embryos.  相似文献

目的改造IVF-30和HTFplus培养液,观察在改造后培养液里卵的受精和早期胚胎的发育状况,寻找适合SD大鼠体外受精和早期胚胎培养的实验体系。方法采用两种方法(超数排卵及体内成熟)采集成熟卵母细胞用于体外受精。选择两种商品化的培养液IVF-30(Vitrolife),HTFplus(Lifeglobal)。分别在以上的培养基中增加10、20、30mmol/L的NaCl,将培养液的渗透压提升至325~355mOsM之间,观察在改造前与改造后的培养液内SD大鼠卵母细胞受精率和囊胚发育率等指标。结果经改造的培养液HTFplus和IVF-30,体外受精率分别从13%、15%提升至70%、67%。改造后的IVF-30和HTFplus的2细胞,大于4细胞及囊胚的发育率分别为85%,74%,22%;96%,75%,32%。HTFplus,IVF-30经过改造后更适合于SD大鼠的体外受精及早期胚胎培养。与超排相比较更多的在自然周期的体内成熟卵母细胞(16%,28%)发育到了囊胚。结论成功建立了SD大鼠的体外受精和体外培养系统。  相似文献

Six mature female rhesus monkeys were treated with HMG-HCG in control cycles at doses adjusted to induce ovulation while avoiding superovulation. Occurrence of ovulation was determined by observation of fresh ovulation points at laparotomy 48 to 120 hours following HCG. In subsequent cycles animals were treated with indomethacin (treatment days 4 through 10) together with the established dose of HMG-HCG. In 8 cycles indomethacin 5 mg/kg was given i.m. once daily; in 9 cycles 10 mg/kg i.m. was administered in 2 divided doses. Following this, PGF2alpha (3 mg t.i.d. s.c.) was administered for 3 days together with indomethacin 10 mg/kg and HMG-HCG, beginning on the day prior to HCG. Determinations of progesterone were performed by RIA on treatment days 4, 7, 10, and 11. Eleven of the 13 control cycles were ovulatory. With indomethacin 5 mg/kg/day, 5 of 8 cycles were ovulatory but ovulation was delayed in 2 instances. Of 9 cycles using indomethacin 10 mg/kg/day only 1 was ovulatory. When PGF2alpha was administered in susequent cycles along with indomethacin (10 mg/kg) and HMG-HCG, ovulation occurred in 13 of 19 cycles. These data suggest that local ovarian PGF2alpha may be essential in the mechanics of follicle rupture in gonadotropin-treated rhesus monkeys.  相似文献

Recombinant bovine somatotropin (rbST) has been shown to increase follicular growth in cattle and some studies have demonstrated an increase in superovulatory response for rbST-treated cows. Pregnancy rates have also been shown to increase when rbST was administered around the time of insemination or prior to embryo transfer. The application of rbST for the purpose of increasing superovulatory responses of donor cows and increasing pregnancy rates of recipient heifers was tested in a commercial embryo transfer program. In Experiment 1, embryo donor cows (n = 56) underwent three cycles of control superovulation (two before and one after weaning) and subsequently underwent up to four additional superovulations while being treated with either rbST (500 mg sustained-release rbST; Posilac, Monsanto, St. Louis, MO; n = 28) or excipient (control; n = 28) once every 14 days. In Experiment 2, lactating embryo donor cows (n = 37) underwent a control superovulation and then underwent a superovulation while lactating and being treated with either rbST (n = 16) or excipient (n = 21). In Experiment 3, embryo recipient heifers that were being implanted with either in vitro or in vivo produced embryos were treated with either rbST (n = 146) or excipient (n = 143) at the time of embryo transfer. Treatment of non-lactating (Experiment 1) or lactating (Experiment 2) donor cows with rbST during repeated superovulation did not affect the number of corpora lutea, the sum of transferable embryos, degenerate embryos, and unfertilized oocytes, or the number of transferable embryos. Treatment of recipient heifers with rbST (Experiment 3) did not affect pregnancy rates for either in vitro or in vivo produced embryos. We conclude that superovulatory response and pregnancy rates (respectively) are similar to control for rbST-treated cows undergoing repeated superovulations and rbST-treated recipient heifers treated at the time of embryo transfer.  相似文献

Models for the origin of the sex incorporate either obligate or facultative sexual cycles. The relevance of each assumption to the ancestral sexual population can be examined by surveying the sexual cycles of eukaryotes, and by determining the first lineage to diverge after sexuality evolved. Two protistan groups, the parabasalids and the oxymonads, have been suggested to be early-branching sexual lineages. A maximum-likelihood analysis of elongation factor-1α sequences shows that the parabasalids diverged prior to the oxymonads and thus represent the earliest sexual lineage of eukaryotes. Since both of these protist lineages and most other eukaryotes are facultatively sexual, it is likely that the common ancestor of all known eukaryotes was facultatively sexual as well. To understand the causes for poor response to superovulation in mature cows of high genetic potential, endocrine and follicular events during and after superovulation were compared in heifers (<2 yr old) yielding large numbers of embryos and cows (9 to 13 yr old) known to be poor embryo donors. Follicular development was monitored by daily ultrasonography. Blood samples were taken 2 to 3 times a day for the measurements of P4, E2, FSH and LH by RIA. Intensive blood collections at 15-min intervals for 6 h were also performed during preovulatory and luteal phases. The number of embryos produced in the heifers (15.2 +/- 2; mean +/- SEM) and the cows (0.6 +/- 0.4), was similar to the number of ovulatory follicles derived from ultrasonographic observations in the heifers (16.2 +/- 3.7), but not in the cows (7.8 +/- 2.8). Contrary to that observations in heifers, there was no increase in the number of 4- to 5-mm follicles in cows during superovulation. The number of larger follicles (>5 mm) increased during superovulation in both cattle groups, but it was significantly lower in cows than in heifers. During superovulation, the maximal E2 concentration was greater (P < 0.0001) in heifers than in cows. One cow showed delayed luteolysis during superovulation, while another had abnormally high FSH (>10 ng/ml) and LH (>3 ng/ml) concentrations following superovulation. All the cows had a postovulatory FSH rise which was not detected in the heifers. The results showed that attempts to improve superovulatory response in mature genetically valuable cows are hampered by a number of reproductive disorders that are not predictable from the study of the unstimulated cycle.  相似文献
