运用小规模实验初步探讨了以中国人群为基础的遗传图绘制工作的必要性。18个无关汉族3代家系共131份血样采自甘肃省白银地区,常规PCR扩增9号染色体的10个STR基因座,采用非变性聚丙烯酰凝胶电泳分析。PCR产物经克隆测序确定核心序列重复次数,采用标准命名法命名各等位基因,用POPGENE软件包计算各基因座等位基因频率,并进行Hardy-Weiberg平衡检验,用Linkage软件包进行各基因座之间连锁关系分析。根据连锁分析结果绘制了中国人群由10个STR基因座构成的9号染色体遗传图。基于中国人群体的9号染色体10个STR连分析锁分析结果与GDB检索结果之间存在较显著的差异,这种差异同时表现在个别基因座之间和0号染色体遗传图总长度上。男、女遗传结果之间在较显著的差异,这种差异同时表现在个别基因之间和9号染色体总长度上。男、女遗传图总长度为129.21cM和178.4cM,均大高加索入。说明了在运用GDB数据之前,有必要根据实验群体进行基因座的初步评估,并且有必要对基于中国人群体的遗传图进行进一步的研究。  相似文献

新疆维吾尔族四个STR位点遗传多态性分析   总被引:2,自引:0,他引:2  
研究新疆维吾尔族人群D16S539、D13S317、D7S820和D5S818的STR基因位点的基因及基因型分布,获得4个基因座的群体遗传学数据。采用PCR扩增技术和基因扫描技术进行样本STR遗传结构分析,并与其他种族、人群的等位基因频率进行比较。结果表明4个基因位点在新疆维吾尔族人群中均具有遗传多态性。4个基因座的基因型分布均符合Hardy-Weinberg平衡定律(P>0.05),不同人群基因频率分布存在一定的差异,所得到的等位基因频率等数据可为遗传学研究、法医个体畜产品识别及亲子鉴定提供依据。  相似文献

为了分析陕西汉族人群中11号染色体上10个短串联重复序列(short tandem repeat, STR)基因座的多态性,采用荧光标记基因扫描方法,对11号染色体上的D11S1760、D11S4102、D11S4116、D11S4207、D11S4162、D11S914、D11S4127、D11S917、D11S4146和D11S915基因座的遗传多态性进行分析。结果显示:D11S1760、D11S4102、D11S4116、D11S4207、D11S4162、D11S914、D11S4127、D11S917、D11S4146和D11S915基因座分别检出17、11、15、11、4、6、7、12、11和13个等位基因,41、18、36、30、8、10、16、30、29和32个基因型,等位基因型符合Hardy-Weinberg平衡(P>0.05)。其杂合度分别为86.72%、67.95%、83.90%、85.96%、58.18%、57.63%、72.60%、72.73%、77.87%和86.40%,表明11号染色体上的这10个STR基因座在汉族人群中有较好的多态性,是理想的遗传标记物。  相似文献

为了调查X染色体上DXS6804、DXS9896和GATA144D04等3个STR基因座在中国汉族群体的遗传多态性及其法医学应用价值,来用PCR和聚丙烯酰胺凝胶电泳对X染色体3个STR基因座进行分型,并检验女性基因型频率分布是否符合Hardy Weinberg平衡,计算法医学常用各种概率。DXS6804、DXS9896和GATA144D04的非父排除率分别为0 5990、0 6220、0 4280,表明3个STR基因座在中国汉族群体均具有遗传多态性,χ2检验表明女性的基因型频率分布符合Hardy Weinberg平衡。X染色体上的基因座DXS6804、DXS9896和GATA144D04在中国汉族群体中具有较高的遗传多态性,可应用于法医学检验和群体遗传学分析。  相似文献

应用复合PCR及基因扫描技术, 对云南白族、傣族、彝族人群X染色体3个STR基因座DXS6804、DXS6799、DXS7132的遗传多态性进行研究。白族89个样本中共检出18个等位基因, 38个基因型, 等位基因频率分布在0.0200~0.6400之间, 基因型频率分布在0.0256~0.3333之间; 傣族100个样本中共检出17个等位基因, 24个基因型, 等位基因频率分布在0.0135~0.7500之间, 基因型频率分布在0.0385~0.5769之间; 彝族88个样本中共检出20个等位基因, 35个基因型, 等位基因频率分布在0.0125~0.5875之间, 基因型频率分布在0.0250~0.3500之间。群体遗传多态性指标及法医学应用指标统计结果显示, 3个基因座在云南3个少数民族人群中均具有高度多态性。聚类分析和系统进化关系分析发现, 彝族、白族、傣族与藏族之间的遗传关系较近。  相似文献

分析了中国汉族人群中12号染色体上7个短串联重复序列(short tandem repeat,STR)基因座的多态性。 采用荧光标记基因扫描对12号染色体上D12S1718、D12S1675、D12S358、D12S367、D12S1638、D12S1646和D12S1682基因座在80名陕西咸阳、榆林汉族人中的遗传多态性进行分析。结果在中国汉族人群中, D12S1718、D12S1675、D12S358、D12S367、D12S1638、D12S1646和D12S1682基因座分别检出7、10、8、8、6、9和11个等位基因,10、17、18、18、14、18和26个基因型,杂合度分别为44.28%、66.10%、78.89%、77.89%、73.69%、74.55%和82.39%。表明这7个STR基因座在中国人群中有较好的多态性,其基因型分布均符合Hard-Weinberg平衡(P>0.05)。  相似文献

运用复合PCR扩增,6%变性聚丙烯酰胺凝胶电泳结合银染技术对我国新疆102位无关的哈萨克族个体进行D16S539,D7S820,D13S317的STR位点的调查,为建立新疆哈萨克族群体数据库提供资料。经统计学检验,3个位点的基因型频率分布符合Hardy-Weinberg平衡定律,结果显示3个位点的期望杂合度为:0.9439、0.9356、0.9304,累积PIC=0.9905,DP=0.9998,PE=9572。紫外,比较新疆哈萨克族与其他4个人群的等位片段频率,发现除与北京汉族在D7S820位点上无统计学意义外(P>0.05),其他均可见显著性差异(P<0.05)。同时,在8个家系42人的调查中无一突变发现且均按孟德尔遗传规律传递。3个STR位点的联合分析在法医学应用及群体遗传学中显示了较高的价值。  相似文献

对河南省河南籍汉族群体的6个短串联重复序列(Short tandem repeats,STR)基因座等位基因频率进行研究,得到河南汉族群体F13A1,F13B,D8S1179,CSF1PO,D5S818,TPOX基因座的群体遗传学依据。EDTA抗凝血样采自河南122名无血缘关系的汉族个体,采用Chelex法抽提DNA,PCR扩增,非变性聚丙烯酰胺垂直凝胶电泳,银染显色分析,得到6个基因座的等位基因频率,各基因座的杂合度分别为:0.62,0.46,0.83,0.59,0.78,0.65;人体识别率分别为0.78,0.66,0.95,0.79,0.92,0.82。6个STR基因座具有较高的杂合度,等位基因分布符合Hardy-Weinberg平衡,是较理想的遗传标记,可用于法医学个本识别和亲权鉴定。  相似文献

为研究云南纳西族人群10个位于X染色体的短串联重复序列基因座及单倍型的遗传多态性, 采用PCR扩增, 变性聚丙烯酰胺凝胶电泳结合银染显色分型技术, 对98名云南纳西族无关男性个体X染色体的10个STR基因座进行基因分型。结果显示, 98名无关男性个体中, DXS7423、DXS7424、DXS6799、DXS7133、DXS6804、DXS8378、HPRTB、DXS7130、DXS7132 和DXS6789分别检出4、7、6、3、6、5、5、7、6和8个等位基因, 等位基因频率分布在0.0102(DXS7132、DXS6789)~0.7347(DXS7133)之间。由DXS8378与DXS7130基因座组成的单倍型共检出20种, 由DXS6789、DXS6799和DXS7424基因座组成的单倍型共检出56种, 单倍型多样性分别为0.8553和0.9649, 说明所选的10个X-STR位点有较高的多态性信息, 在基因组多样性研究、法医学个体识别、亲权鉴定中具有重要应用价值。  相似文献

利用多重PCR五色荧光(6FAM、VIC、NED、PET、LIZ)自动化检测技术检测西藏自治区藏族人群D8S1179、D21S11、D7S820、CSF1PO、D3S1358、TH01、D13S317、D16S539、D2S1338、D19S433、VWA、TPOX、D18S51、D5S818及FGA共15个STR基因座遗传多态性, 获得15个STR基因座的群体遗传学数据。结果显示:15个STR基因座的基因型分布符合Hardy-Weinberg平衡。15个STR基因座的个体鉴别力 (Discrimination power, DP)在0.7555~0.9602之间, 杂合度 (Heterozygosity, H)在0.5651~0.8530之间, 多态性信息含量 (Polymorphism information content, PIC)在0.5528~0.8456之间, 非父排除率(Probability of paternity exclusion, EPP)在0.3811~0.8549之间, 累积个体鉴别力为0.999999999, 累积非父排除率为0.999999998。15个短串联重复序列基因座适合作为西藏藏族人群的遗传标记, 用于人类学、疾病连锁分析、法医学亲子鉴定和个体识别等领域的研究。  相似文献

Population data was collected for the STR loci F13AO1, FES/FPS, HUMvWA, and HUMTHO1, in three major Philippine ethnolinguistic groups and used to estimate statistical parameters for identity testing in forensic work on Filipinos. The Cebuano, Ilocano, and Pampango populations in the Philippines were studied because they are among the biggest linguistic groups in the country, thus their genotypic profiles should substantially represent those of many Filipinos. The number of alleles varied from 4 to 9 at all loci, falling within the range observed for other local and world populations. Pairwise comparisons of the allele frequency distributions showed no statistical differences among the populations. The test for linkage equilibrium showed no evidence of non-random association of alleles across the physically unlinked loci in any of the three populations. The four loci combined gave an exclusion power of > or =0.9995 and a power of paternity exclusion of 0.8859-0.9389.  相似文献

The aim of this study was to investigate allelic frequency distribution and forensic genetic parameters of autosomal short tandem repeats (STR) loci of the population samples from 107 Tujia individuals from Chinese Hubei Province. Twenty-one autosomal STR genetic markers (D9S1122, D6S474, D6S1017, D5S2500, D4S2408, D3S4529, D2S441, D2S1776, D22S1045, D20S482, D1S1677, D1S1627, D1GATA113, D19S433, D18S853, D17S1301, D11S4463, D12ATA63, D10S1248, D10S1435 and D14S1434) were simultaneously amplified in a new multiplex polymerase chain reaction system. 155 alleles for all the STR loci from the Tujia population were observed and the corresponding allelic frequencies ranged from 0.005 to 0.589. Expected heterozygosity, polymorphic information content, power of discrimination and power of exclusion of the 21 STR loci in the Tujia population were from 0.579 to 0.824, from 0.525 to 0.802, from 0.773 to 0.945 and from 0.257 to 0.641, respectively. Our results indicate that the autosomal STRs multiplex system provides highly informative STR data and could be useful in forensic individual identification and parentage testing in this region.  相似文献

Genetic diversity and forensic parameters based on 15AmpFlSTR Identifiler short tandem repeat (STR) loci (D8S1179, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5S818 and FGA) were evaluated in a sample of 101 unrelated, autochthonous adults from Montenegro. After applying Bonferroni correction, the agreement with Hardy-Weinberg equilibrium (HWE) was confirmed for all loci with the exception of D5S818 (chi2 test) and D21S11 (exact test). The combined power of discrimination (PD) and the combined power of exclusion (PE) for the 15 studied loci were 0.9999999999999999844 and 0.99999382, respectively. According to measures of within-population genetic diversity, D2S1338, D18S51 and FGA may be considered as the most variable and most informative markers for forensic testing and population genetic analyses out of the 15 analysed loci in a population of Montenegro. D5S818 showed to be the least variable and together with TPOX, the least informative. Interpopulation comparisons were carried out and levels of genetic differentiation between population of Montenegro and five South-eastern European populations (Kosovo Albanians, Serbians from Vojvodina province, Macedonians, Bosnians and Croatians) were evaluated. The most differentiated population in relation to Montenegro is a population of Kosovo Albanians as suggested by both AMOVA and coefficients of genetic differentiation (F(ST) and R(ST)).  相似文献

Allele frequencies for 15 short tandem repeat (STR) loci (D8S1179, D21S11, D7S820, CSFIPO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5S818, and FGA) were obtained from 7,636 unrelated individuals of Chinese Han population living in Qinghai and Chongqing, China. Totally 206 alleles were observed, with the corresponding allele frequencies ranging from 0.0001-0.4982. Chi-square test showed that all of the STR loci agreed with the Hardy-Weinberg equilibrium. We also compared our data with previously published population data of other ethnics or areas. The results are valuable for human identification and paternity testing in Chinese Han population.  相似文献

Allele frequencies of 15 short tandem repeat (STR) loci, namely D5S818, D7S820, D13S317, D16S539, TH01, TPOX, Penta D, Penta E, D3S1358, D8S1179, D18S51, D21S11, CSF1PO, vWA, and FGA, were determined for 154 individuals from the Kadazan-Dusun tribe, an indigenous population of East Malaysia. All loci were amplified by polymerase chain reaction, using the Powerplex 16 system. Alleles were typed using a gene analyzer and the Genemapper ID software. Various statistical parameters were calculated and the combined power of discrimination for the 15 loci in the population was calculated as 0.999999999999999. These loci are thus, informative and can be used effectively in forensic and genetic studies of this indigenous population.  相似文献

Genetic data are reported for nine short tandem repeat (STR) loci (D3S1358, vWA, FGA, D8S1179, D21S11, D18S51, D5S818, D13S317, and D7S820) and six variable number of tandem repeat (VNTR) loci (D2S44, D10S28, D4S139, D1S7, D5S110, and D17S79) in samples of Utah African Americans, European Americans, and Hispanics. Little evidence of departures from Hardy-Weinberg equilibrium or gametic equilibrium was found in these populations. Because of their relatively higher mutation rates, the VNTR loci exhibited higher average heterozygosity and lower FST levels than did the STR loci. Genetic distance analysis showed congruence between the two types of systems, and a genetic distance analysis of the STR data showed that the three Utah populations are genetically similar to the same ethnic groups in other parts of the United States. In addition, this analysis showed that the African American population is the most genetically divergent, with greater similarity between the Hispanic and European American populations. This analysis demonstrates a high degree of consistency for population designations commonly used in forensic analysis.  相似文献

为了调查30个 InDel 位点在中国北京汉族人群中的群体遗传学数据, 并评估其法医学应用价值, 文章采集210名北京汉族无关健康个体外周血样, 提取样本DNA, 采用Investigator ® DIPplex 体系对HLD77等30个InDel 位点进行复合扩增, ABI3130 XL 遗传分析仪进行基因分型, 计算常用法医学参数, 分析群体遗传差异。经 Bonferroni 校正, 30个InDel 位点不存在连锁不平衡现象, 基因型分布符合Hardy-Weinberg 平衡; 各位点DP值为0.2690~0.6330, 累积个体识别力(TDP)为0.999999999985; 三联体累积非父排除率(CPEtrio)为0.98771049, 二联体累积非父排除率(CPEduo)为0.94579456。32例 STR 基因座发生突变的家系样本调查证实上述 InDel 位点未发现突变。结果表明, Investigator® DIPplex 试剂盒中包含的30个InDel 位点在北京汉族群体中具有较好的遗传多态性, 在 STR 存在突变及微量DNA检材等特殊检案中可作为有效的补充检测体系。  相似文献
