Water Pathways in Leaves of Hedera helix L. and Tradescantia virginiana L.   总被引:2,自引:0,他引:2  
Hydraulic conductances of leaf tissues of Hedera helix and Tradescantiavirginiana leaves were measured. It was found that water couldflow most easily through the veins, but that the cell wallsof at least the ventral epidermis were more efficient at resupplyingwater lost from the epidermal tissue than was the mesophyllat rehydrating itself. Vein and bundle-sheath extensions, whichare characteristic of mesomorphic leaves (e.g. T. virginiana),seem to be important in maintaining a close hydraulic connectionbetween the epidermis and the vascular tissue. In leaves notcontaining vein and bundle-sheath extensions, typically xeromorphicleaves (e.g. H. helix), there is not such a close connectionbetween the epidermis and vascular tissue. This was shown inexperiments involving the sudden application of a reduced pressurepotential to either the epidermis or the other tissues of leaves,and the measurement of transient stomatal opening.  相似文献

The composition of xylem sap and exudate from stem incisionsof Nicotiana glauca Grah. was compared in detail. Exudationfrom stem incisions occurred over a 5 min period in certainplants, enabling collection of 5–30 µl of sap. Therate of exudation showed an exponential decline. Exudate hada high dry matter content (170–196 mg ml–1) andhigh sugar (sucrose) levels. Xylem sap had a low pH (5.8) andexudate a pH of 7.9. Glutamine dominated the amino compoundsin xylem sap and exudate, and K+ was the major cation. Totalamino compounds in stem exudate reached 10.8 mg ml–1 whereasxylem sap contained much lower levels (0.28 mg ml–1).All mineral elements and amino compounds with the exceptionof calcium were more concentrated in stem exudate than in xylemsap. Sucrose was labelled heavily in stem exudate following pulsingof an adjacent leaf with 14CO2. A concentration gradient ofsugar (2.1 bar m–1) was recorded for stems. Levels ofsucrose, amino compounds and K+ ions in stem exudate showeda diurnal periodicity. Each commodity reached maximum concentrationat or near noon and minimum concentration about dawn. The evidencesuggests that exudate from stem incisions of N. glauca is arepresentative sample of solutes translocated in the phloem. Water uptake into the ventral epidermis and the other tissuesof excised T. virginiana leaves was measured separately andsimultaneously when the uptake rates were steady. It was foundthat uptake into the ventral epidermis was proportionately greater,compared with uptake into the remainder of a leaf, than wouldbe expected from the ratio of conductances of the two pathways.This result led to the conclusion that not only is the epidermisa pathway for water transport in the transpiration stream butthat it is an important site of evaporation for stomatal transpiration.  相似文献

Water uptake into the ventral epidermis and the other tissuesof T. virginiana leaves was recorded when the uptake rate wassuddenly increased. Results indicated that there was considerableevaporation, or stomatal transpiration, from the epidermal tissues,and that the water relationships of the epidermis are more affected,at least in the short term, by changes in factors which affectthe transpiration rate than are the water relationships of theother leaf tissues.  相似文献

Excised leaves of T. virginiana were supplied at their baseswith water containing a dilute solution of lead-EDTA chelate.After the leaf had been allowed to transpire for 3–5 hthe lead was precipitated in situ using hydrogen sulphide. Leafsections were examined microscopically and the path of the transpirationstream and the sites of evaporation inferred from the distributionof lead precipitate. From the distribution of precipitate itwas concluded that there is more evaporation from cells liningthe substomatal cavity, compared with cells in the bulk of themesophyll, than was previously thought to be the case.  相似文献

A combined system has been developed in which epidermal cell turgor, leaf water potential, and gas exchange were determined for transpiring leaves of Tradescantia virginiana L. Uniform and stable values of turgor were observed in epidermal cells (stomatal complex cells were not studied) under stable environmental conditions for both upper and lower epidermises. The changes in epidermal cell turgor that were associated with changes in leaf transpiration were larger than the changes in leaf water potential, indicating the presence of transpirationally induced within-leaf water potential gradients. Estimates of 3 to 5 millimoles per square meter per second per megapascal were obtained for the value of within-leaf hydraulic conductivity. Step changes in atmospheric humidity caused rapid changes in epidermal cell turgor with little or no initial change in stomatal conductance, indicating little direct relation between stomatal humidity response and epidermal water status. The significance of within-leaf water potential gradients to measurements of plant water potential and to current hypotheses regarding stomatal response to humidity is discussed.  相似文献

The postulated mechanical advantage of subsidiary cells overguard cells has been estimated using leaves of Tradescantiavirginiana. The turgor pressures of subsidiary cells were adjusted to bezero or maximal by plasmolytic treatments, and the resultingstomatal apertures were measured. The ‘mechanical advantage’was calculated from two mathematical models which define itas an ‘antagonism ratio’. In contradiction to Paulet’s (1965) data, we found that kinetin/IAA strongly affected organogenesis in callus tissue derived from the stem ofNicotiana glauca Grah. plants both in primary expiants and in subcultured calli. The effect of these substances was higher in the subcultured calli derived from mycoplasma-infected plants. Evidence of the absence of the infectious agent in de novo-formed plants in subcultured calli was given by grafting and by the electron micrograph.  相似文献

Callus tissue cultures were established from stems of tobacco plants (N. glauca Grah.) both healthy and mycoplasma (potato witches' broom disease) infected on a modified nutrient medium (with a lower content of mineral salts) according toMurashige andSkoog (1962) in the presence of 2,4-D (1 mg l?1) as a growth regulator. No differences were observed in the growth and development of both tissues. Organogenesis appeared on a nutrient medium (Petr? et al. 1972) supplemented with kinetin (0.64 mg or 2.56 mg l?1) and IAA (2 or 4 mg l?1). Callus derived from mycoplasma diseased plants started to form numerous buds after three months whereas organogenesis in callus from healthy controls appeared only after six months. We suppose that the reason of this difference is the fact that an expressively higher content of 2,4-D was found in the calli from healthy plants in comparison with the corresponding tissue from mycoplasma diseased ones. Reconstituted plants were isolated, rooted and transferred in the soil. The infectivity of these plants was assayed by grafting their stem tips on tomato plants which indicate very reliably and sensitively this mycoplasma disease. 31 reconstituted plants were obtained in the whole from calli isolated from mycoplasma infected plants and all of them were healthy. It was established that mycoplasma failed in the presence of 2,4-Din vitro. Stem pieces from diseased plants in which mycoplasma presence was proved, lose their infectivity after 4 weeks of cultivation on nutrient medium with this growth regulator. On the contrary 2,4-D which spreads and acts especially through phloem (Smith et al. 1947) does not kill mycoplasmain vivo even in doses evoking strong symptoms of 2,4-D effect on experimental plants.  相似文献

A technique is described for the establishment of infectious axenic callus cultures originated from stems of tobacco plants suffering from a mycoplasma disease, potato witches' broom. When stem pieces from a diseased tobacco plant (Nicotiana glauca Grah.) are culture on a modified nutrient medium according toMurashige andSkoog (1962) with added kinetin (0.64 mg or 2.56 mg 1 000 ml?1) and IAA (2 mg or 4 mg 1 000 ml?1), the pathogen persists and spreads in the newly formed callus tissue. The presence of the pathogen was proved by implantation of callus slices into stems of tomato plants which indicate the disease very reliably. Reconstituted tobacco plants too recovered the infectious agent of potato witches' broom; 27 plants from the 35 reconstituted plants were diseased. Similar results were obtained with Crimean yellows. Calli were cultured and subcultured from 1 to 18 monthsin vitro and some infectious cultures are maintained and their investigation goes on. We suppose that kinetin plays a very important role for infectivity preservation in callus cultures.  相似文献

Desiccation of 8- to 13-day-old seedlings, achieved by withholding nutrient solution from the vermiculite root medium, caused a reduction in nitrate reductase activity of the leaf tissue. Activity declined when leaf water potentials decreased below −2 bars and was 25% of the control at a leaf water potential of −13 bars. Experiments were conducted to determine whether the decrease in nitrate reductase activity was due to reduced levels of nitrate in the tissue, direct inactivation of the enzyme by low leaf water potentials, or to changes in rates of synthesis or decay of the enzyme.  相似文献

When the apertures of stomata in an epidermal strip changedthere was found to be a simultaneous change in the unbound watercontent of the tissue. Field beans were grown in three different irrigation treatments.The growth of each leaf was followed and estimates made of plantwater potential at dawn and of plant water potential, solutepotential, and pressure potential in the afternoon. The growthin area of the leaves against time was fitted with a logisticcurve from which the parameters of leaf growth were defined.The parameters were the area at which a leaf unfolds, the meangrowth rate, and the duration of growth, which combine to givethe final leaf size. Water stress reduced the final leaf sizeby reducing both the area at unfolding and the mean growth rate.The duration of growth was not consistently altered. Final leafsize was closely correlated with the plant water potential inthe afternoon and apparently more with pressure than solutepotential. 根据植物偏爱密码子优化设计、合成纳豆激酶基因sNK,利用重叠延伸PCR法在其中插入番茄果实特异性表达基因E8的第一内含子构成sNKi基因,通过农杆菌渗透法将这两种基因渗入到烟草NC89叶片中并实现瞬时表达。通过RT-qPCR法将两种基因在烟草叶片中转录水平的表达量进行比较,结果表明两种基因在烟草叶片中均表达,且sNKi基因的表达量显著高于sNK基因;通过纤维蛋白平板法在两种基因的瞬时表达样品中均能检测到纤溶酶活性,表明目的基因在烟草叶片中可正常翻译并表现出溶栓活性,且sNKi基因在翻译水平的表达量显著高于sNK基因。表明内含子对人工合成的纳豆激酶基因的瞬时表达具有显著的促进作用。  相似文献

在每盆施用0-8g纯氮、0—12gP2O5、0—12g K2O的肥料范围内,随着氮、磷、钾肥用量的增加,烤后烟叶中类胡萝卜素含量增加。每盆施钾量超过12gK2O时,再施钾肥对增加烤后烟叶中类胡萝卜素含量的效果不显著。随着土壤相对含水量的提高,类胡萝卜素总量表现为先增加后减少的趋势。70%左右的土壤相对含水量的烟叶中类胡萝卜素含量最高。  相似文献

Fluorescent in situ hybridization, C-banding/DAPI, and CMA(3)-fluorescence were performed to reveal the cytomolecular constitution of the standard (A) and supernumerary (B) chromosomes of an autotetraploid Tradescantia virginiana L. The analyses show that translocations and/or inversions have occurred during the evolution of the T. virginiana karyotype, generating a significant level of structural heterozygosity. Regarding the structural level, the present paper confirms the occurrence of small subterminal duplications and/or inversions in T. virginiana already suggested by previous authors. Interestingly, many of the distal chromosome segments in T. virginiana possess duplicated subterminal telomere clusters, heterochromatin, 5S and 45S rDNAs orderly intermixed and share this complex cytomolecular architecture with the common type of a B chromosome. Based on the obtained results, it is proposed that in T. virginiana the B chromosome may have arisen via excision from the distal region of an A chromosome. The nascent B could have retained much of the ancestral sequence arrangement, including duplicated telomeric cluster(s), heterochromatin and rDNA, but developed a new centromere/kinetochore to successfully propagate through the cell cycle.  相似文献

Leaf diffusion resistance was influenced by leaf age in well-wateredand water-stressed clonal tea plants. In well-watered plantsand in two of the three clones studied, young leaves showeda significantly lower diffusion resistance than old leaves.In water-stressed plants young leaves always exhibited a higherdiffusion resistance than old leaves. The highest diffusionresistance, irrespective of leaf age and water stress, occurredin clone DN which is known to be the most drought tolerant ofthe three clones studied, suggesting that drought tolerancein clone DN is caused, at least in part, by a stomatal regulationmechanism. Water release characteristic curves for the threeclones indicated differences in the water content-water potentialrelationship between young and old leaves as well as betweenclones. The drought tolerant clones had a higher relative watercontent for a given water potential compared with the drought-susceptibleclone. 1. Coproporphyrinogenase was extracted and purified from tobacco (Nicotiana tabacum L.). Enzyme activity was mainly located in mitochondria rather than in chloroplasts. The enzyme was purified by differential centrifugation, ammonium sulphate fractionation, calcium phosphate gel adsorption and dialysis. A 69-fold final purification was obtained. 2. An apparent K(m) value of 3.6x10(-5)m was found, the value being largely dependent on the amount of coproporphyrin III recovered after reduction with sodium amalgam to coproporphyrinogen III. Protoporphyrin formation was linear up to 3h and decreased with further incubation. The enzyme activity increased with the concentration of enzyme protein up to 30mug/ml of solution. 3. Enzyme activity was greatly enhanced by increasing Fe(2+) concentrations up to 0.5mm, beyond which inhibition occurred. Co(2+) and Mn(2+) were also found to activate at low concentrations (0.1mm) and inhibit at higher concentrations (5mm). Fe(3+) and Cu(2+), both at 0.1mm, and o-phenanthroline and EDTA, each at 1mm, were found to be inhibitory.  相似文献

烟草生长初期,超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量与pH增长呈正相关,而过氧化氢酶(CAT)和过氧化物酶(POD)活性则随着pH的增加而下降,pH值6.5和7.5时叶绿素含量和净光合速率(Pn)较高,旺长期达到高峰;生长后期,各处理的MDA含量和POD活性最高,SOD、CAT活性最低.各pH值处理的叶片中,叶绿素含量、Pn下降,比叶重达到高峰,但pH 8.5下的烟叶中叶绿素含量高,Pn大,比叶重较小.pH 6.5和7.5下的烟叶中蛋白质和可溶性糖总体含量高于其它pH的.烟碱含量在pH 5.5时最高,pH 8.5时最低.  相似文献

通过对烟草植株覆盖白、红、黄、蓝、紫色滤膜获得不同光质,研究了光质对烟叶生长及光合作用的影响。结果表明,与白膜处理相比,红膜与蓝膜处理下的烟草叶片较厚,比叶面积较小,叶绿素a/b比值、净光合速率、可变荧光强度(Fv)和最大荧光强度(Fm)的比值Fv/Fm(PSⅡ最大光化学量子效率)、PSⅡ实际光化学量子效率(ΦPSⅡ)、光饱和点和CO2饱和点均较高。黄膜处理下的叶片较白膜处理的更薄,净光合速率、Fv/Fm、ΦPSⅡ、光饱和点、CO2饱和点均较低。紫膜处理的叶片比叶面积比白膜处理的小,净光合速率和Fv/Fm比白膜的大。实验结果表明红光、蓝光和紫光促进了烟叶的生长,这种促进作用是与其高光合效率紧密相连的;而黄光对烟叶的生长有一定程度的抑制作用。  相似文献
