荔枝与龙眼种质资源研究进展   总被引:3,自引:0,他引:3  
荔枝和龙眼是中国著名的南亚热带果树。本文分别就荔枝、龙眼种质资源在起源与分布、分类与鉴定等方面取得的研究进展进行了综述。荔枝和龙眼的分类研究大体经历了3个阶段:根据形态特征进行分类、应用同工酶分析和DNA分子标记进行分类。形态特征常常受到环境条件的影响,而同工酶分析可以检测到的多态性位点十分有限。应用分子标记研究荔枝、龙眼种质资源的遗传多样性克服了前述2种研究方法的不足。目前,荔枝和龙眼的品种选育仍然以实生选种和芽变选种为主。为了实现对荔枝、龙眼果实品质和农艺性状的改良,建议在分子水平上进一步深入地探讨荔枝、龙眼种质资源的遗传多样性;同时通过应用分子标记辅助选择技术提高荔枝和龙眼杂交育种的效率。  相似文献

Litchi (Litchi chinensis Sonn.) is a fruit crop with a clear niche for expansion in countries with subtropical climates. One of the main limitations for breeding purposes and optimum germplasm management in this species is the confusion in cultivar denomination among different producing countries and germplasm collections worldwide. Litchi cultivar identification is still mainly based on morphological characters, and homonymies and synonymies are very frequent. To address this gap, a molecular study was conducted to characterize litchi accessions from two of the main litchi-producing regions for the export market, Mauritius and Réunion, and to compare them to those obtained from litchi cultivars from different origins conserved in a germplasm collection in Spain. Eleven simple sequence repeat (SSR) loci were used to characterize molecular polymorphisms among 88 litchi accessions conserved in Mauritius, Réunion, and Spain. A total of 67 amplification fragments were detected with those 11 SSRs, with an average of 6.1 bands/SSR. Three primer pairs seemed to amplify more than one locus. The mean expected and observed heterozygosities over the eight single locus SSRs averaged 0.53 and 0.61, respectively. The total value for the probability of identity was 9.78?×?10?4. Molecular characterization revealed the existence of 42 different genetic profiles. Several synonymies and homonymies in litchi cultivar nomenclature both within and across geographical regions were found. This comparative study provides the basis for the standardization of litchi cultivar nomenclature over the studied regions and in other litchi-producing countries.  相似文献

Multiple shoot induction in Litchi chinensis Sonn. (litchi) has been achieved by two methods: (1) direct germination of litchi seeds in 6-benzylaminopurine (20 mg l–1)-supplemented MS liquid medium and supported on a filter-paper bridge and (2) in planta treatment with 6-benzylaminopurine (100 μg on alternate days) of the axillary bud regions of plants germinated and maintained under sterile conditions. While the former method resulted in as many as 27.5±8.6 shoot buds from the cotyledonary node within 4 weeks, the latter yielded on average approximately 8 shoot buds from each treated node in 8 weeks. The cytokinin treatment in planta consisted of placing sterile filter paper moistened with sterile distilled water over the node and adding different concentrations of 6-benzylaminopurine. Both methods of multiple shoot induction were found to be effective for the five genotypes of litchi that were tested. The shoots elongated and rooted directly in vermiculite after a pulse treatment with IBA (25 mg/ml) for 15 min. Fungus growth which is a serious problem in litchi tissue culture, was controlled using a fungicide, Bavistin, and by eliminating organic nutrients from the growth medium. Immature embryos of different sizes and ages from commercialvarieties of lychee (Litchi chinensis Sonn.) were cultured ina range of different media. Embryos as small as 3 mm could becultured using in vitro techniques and subsequently grown intoplants. MS solid medium with 2% sucrose supplemented with 150ml l–1 coconut water was most effective in stimulatingthe germination of immature lychee embryos. Embryos of lycheewere treated to induce adventitious buds from embryonic shootsas a means of achieving multiplication. The different varietiesexhibited differences in response, with Bengal embryonic shootsproducing 15 adventitious buds after pretreatment with 100 mgl–1 BAP for 3 h. Root formation was achieved in 65% ofadventitious shoots using MS medium supplemented with 0.5 mgl–1 NAA and activated charcoal. These plants were successfullydeflasked and grown on in the glasshouse. This technique providesof means of producing some multiple shoots from lychee embryosand has value for multiplication in a breeding program wherea method of micropropagation is unavailable. Content of total proanthocyanidins as well as total phenolics, flavonoids, antioxidant activities were evaluated for litchi (Litchi chinensis Sonn.) pulp of 32 cultivars. One cultivar, Hemaoli, showed the highest total proanthocyanidins and total phenolics, and DPPH or ABTS radical scavenging activities. ESI-MS and NMR analysis of the Hemaoli pulp crude extracts (HPCE) showed that procyandins composed of (epi)catechin unites with degree of polymerization (DP) of 2–6 were dominant proanthocyanidins in HPCE. After the HPCE was fractionated by a Sephadex LH-20 column, 32 procyanidins were identified by LC-ESI-Q-TOF-MS in litchi pulp for the first time. Quantification of individual procyanidin in HPCE indicated that epicatechin, procyanidin B2, procyanidin C1 and A-type procyanidin trimer were the main procyanidins. The radical scavenging activities of different fractions of HPCE as well as six procyanidins standards were evaluated by both DPPH and ABTS assays. HPCE fractions showed similar antioxidant activities with those of Vc and six individual procyanidins, the IC50 of which ranged from 1.88 ± 0.01 to 2.82 ± 0.10 μg/ml for DPPH assay, and from 1.52 ± 0.17 to 2.71 ± 0.15 μg/ml for ABTS assay. Such results indicate that litchi cultivars rich in proanthocyanidins are good resources of dietary antioxidants and have the potential to contribute to human health.  相似文献

荔枝、龙眼及龙荔的茎解剖结构比较   总被引:2,自引:0,他引:2  
为了观察荔枝、龙眼和龙荔茎解剖结构的异同,采用徒手切片法和组织离析法,对荔枝、龙眼和龙荔20份材料的茎横切面结构和导管分子的特征进行了比较研究。结果表明:(1)各试材的茎横切结构由外到内依次为:周皮、皮层、维管组织、髓和射线,但其结构特征总体上差异较大;龙眼种质的平均髓率(26.19%)显著高于龙荔(22.43%)和荔枝(6.96%),各试材间,褐毛荔枝的髓率最小(3.37%),‘三月红’居中(14.14%),野生龙眼最大(30.27%);荔枝的树皮率(41.93%)显著大于龙眼(33.21%)和龙荔(29.83%)。(2)各试材的导管密度未见明显规律,以‘三月红’的导管密度最小(93.17 个/mm2),龙荔次之,为101.78 个/mm2;荔枝的导管总面积/木质部面积的比值显著大于龙眼和龙荔;‘三月红’的射线宽度(22.13 μm)显著大于其它试材。(3)20份试材的导管分子绝大多数属于孔纹式或网纹式,极少数为环纹式或螺纹式或梯纹式;‘三月红’的导管分子内径(35.97 μm)显著大于其它试材,龙荔次之(33.90 μm);褐毛荔枝的导管分子(238.52 μm)显著长于其它试材。(4)导管分子短、孔径大、端壁倾斜度小(或水平)、无尾、孔纹、单穿孔板均是导管分子进化的特征;在荔枝试材中,‘三月红’的导管分子较为进化,而褐毛荔枝则较为原始。(5)对茎结构特征指标进行聚类分析,结果显示,20份试材首先聚为2大类,荔枝12份试材(除‘三月红’外)聚为第1大类;龙眼6份试材、龙荔及荔枝‘三月红’聚为第2大类;在这2大类的基础上又分为8个组,各组均有独自的显著特征。对荔枝、龙眼和龙荔3类植物茎结构特征的比较可知,总体上荔枝比龙眼原始,而龙荔居荔枝和龙眼之间。  相似文献

Immersion of litchi fruit in 49 degrees C water for 20 min followed by hydrocooling in ambient (24 +/- 4 degrees C) temperature water for 20 min was tested as a quarantine treatment against potential infestations of Mediterranean fruit fly, Ceratitis capitata (Wiedemann); and oriental fruit fly, Bactrocera dorsalis Hendel, eggs or larvae in Hawaiian litchi, Litchi chinensis Sonnerat. The 49 degrees C hot-water immersion of litchi provided probit 9 (99.9968% mortality with >95% confidence) quarantine security against eggs and first instars. There were no survivors from 15,000 each feeding and nonfeeding Mediterranean fruit fly or oriental fruit fly third instars immersed in a computer-controlled water bath that simulated the litchi seed-surface temperature profile during the 49 degrees C hot-water immersion treatment. Litchi served as the model for longan, Dimocarpus longan Lour., a closely related fruit that is smaller and also has commercial potential for Hawaii. Modified fruit infestation and holding techniques used to obtain adequate estimated treated populations from poor host fruit, such as litchi and longan, are described. Data from these experiments were used to obtain approval of a hot-water immersion quarantine treatment against fruit flies for litchi and longan exported from Hawaii to the U.S. mainland.  相似文献

在荔枝 (LitchichinensisSonn .)胚胎败育发生期 ,以系统溶剂法从正常或败育胚珠中初步提取酚类抑制物质 ,通过TLC分离与纯化 ,用GC MS联用仪进一步分离鉴定 ,并以标准品核对。试验首次从荔枝胚珠中分离鉴定出酚类抑制物质对羟基苯甲酸 (p_HBA)。生物活性测定表明 ,p_HBA是一种很强的生长抑制物质。在败育胚珠中其含量及IAA氧化酶活性均显著高于正常胚珠 ,IAA水平则明显低于正常胚珠 (P <0 .0 1)。因此认为 ,p_HBA参与了荔枝胚胎发育的调节 ,高含量的p_HBA是通过促进IAA侧链的氧化并影响促进和抑制生长的物质之间的平衡而导致荔枝胚胎的败育  相似文献

BackgroundLectins are highly important biomolecules to study several biological processes. A novel α-D-glucose/mannose specific lectin was isolated from the seeds of litchi fruits (Litchi chinensis) and its various biophysical and biochemical properties were studied.MethodsPurification was done by successive Sephadex G 100 and Con A-Sepharose 4B affinity chromatography. SDS-PAGE, Surface Plasmon Resonance (SPR), steady state absorbance, fluorescence, time-correlated single-photon counting, circular dichroism and antibiofilm activity by measuring total protein estimation and azocasein degradation assay have been performed.ResultsThe purified lectin is a homodimer of molecular mass ~ 54 kDa. The amount of lectin required for hemagglutination of normal human O erythrocytes was 6.72 µg/ml. Among the saccharides tested, Man-α-(1,6)-Man was found to be the most potent inhibitor (0.01 mM) determined by hemagglutination inhibition assay. Steady state and time resolved fluorescence measurements revealed that litchi lectin formed ground state complex with maltose (Ka=4.9 (±0.2)×104 M?1), which indicated static quenching (Stern-Volmer (SV) constant Ksv=4.6 (±0.2)×104 M?1). CD measurements demonstrated that litchi lectin showed no overall conformational change during the binding process with maltose. The lectin showed antibiofilm activity against Pseudomonus aeruginosa.ConclusionsA novel homodimeric lectin has been purified from the seeds of litchi fruits (Litchi chinensis) having specificity for α-d-glucose/mannose. The thermodynamics and conformational aspects of its interaction with maltose have been studied in detail. The antibiofilm activity of this lectin towards Pseudomonus aeruginosa has been explored.General significanceThe newly identified litchi lectin is highly specific for α-d-glucose/mannose with an important antibiofilm activity towards Pseudomonus aeruginosa.  相似文献

Litchi is an important fruit tree in tropical and subtropical areas of the world. However, there is widespread confusion regarding litchi cultivar nomenclature and detailed information of genetic relationships among litchi germplasm is unclear. In the present study, the potential of single nucleotide polymorphism (SNP) for the identification of 96 representative litchi accessions and their genetic relationships in China was evaluated using 155 SNPs that were evenly spaced across litchi genome. Ninety SNPs with minor allele frequencies above 0.05 and a good genotyping success rate were used for further analysis. A relatively high level of genetic variation was observed among litchi accessions, as quantified by the expected heterozygosity (He = 0.305). The SNP based multilocus matching identified two synonymous groups, ‘Heiye’ and ‘Wuye’, and ‘Chengtuo’ and ‘Baitangli 1’. A subset of 14 SNPs was sufficient to distinguish all the non-redundant litchi genotypes, and these SNPs were proven to be highly stable by repeated analyses of a selected group of cultivars. Unweighted pair-group method of arithmetic averages (UPGMA) cluster analysis divided the litchi accessions analyzed into four main groups, which corresponded to the traits of extremely early-maturing, early-maturing, middle-maturing, and late-maturing, indicating that the fruit maturation period should be considered as the primary criterion for litchi taxonomy. Two subpopulations were detected among litchi accessions by STRUCTURE analysis, and accessions with extremely early- and late-maturing traits showed membership coefficients above 0.99 for Cluster 1 and Cluster 2, respectively. Accessions with early- and middle-maturing traits were identified as admixture forms with varying levels of membership shared between the two clusters, indicating their hybrid origin during litchi domestication. The results of this study will benefit litchi germplasm conservation programs and facilitate maximum genetic gains in litchi breeding programs.  相似文献

Freshly harvested seeds of soybean (Glycine max L. Merr.) orseeds shelled, then dried, were non-viable. Seeds dried in intactpods, even when only 17% of normal size, matured into viableseeds and produced healthy plants. These seeds maintained activityof various enzymes but gained little soluble protein while air-dryingin intact pods. There was a qualitative change in seed proteinsassociated with maturation. Seeds matured in intact pods havea greater proportion of protein as slow-moving bands and havecompletely lost one fast-moving band compared with seeds shelledbefore drying. Seed maturation is a distinct phase of seed production,is independent of the parent plant, and can be imposed on theseed at many stages of development.  相似文献

To test the proposition that photoperiodic controls synchronizethe flowering of cowpeas, Vigna unguiculata (L.) Walp. [V. sinensis(L.) Savi], the day-length requirements for floral initiationand for flowering were investigated in several short-day accessions.No evidence was found of different critical photoperiods atdifferent stages of development, but exposure to only 2–4short days was required for floral initiation compared withabout 20 for development to open flowers. Pod setting was increasedafter exposure to even one short day more than the number requiredfor flower opening. Floral buds at higher nodes appeared to require fewer shortdays for development to flowering than buds at the lower nodes,and displayed faster rates of development. Inflorescence budsdid not resume development if they were exposed to 15 or morelong days following inflorescence initiation. Thus, any tendencytowards synchronous flowering in cowpeas is not due to the criticalday-length for flower development being shorter than that forflower initiation, but could be the result of cumulative photoperiodicinduction of plants and the more rapid development of later-formedflowers. 以‘马贵荔'ב焦核三月红'76株F_1代群体为试材,检测了酒石酸、苹果酸、蔗糖含量和单果重4个果实性状分离的情况。结果表明,4个性状表现为连续分布,具有数量性状的典型特征,与4个果实性状连锁的QTL位点23个,其中控制酒石酸的QTL为2个,控制苹果酸的QTL为4个,控制蔗糖的QTL为12个和控制单果重的QTL为5个。各QTL的LOD值在3.15~5.61之间,可解释13.85%~88.3%的表型变异。  相似文献

Polyphenol oxidase (PPO) plays a key role in the postharvest pericarp browning of litchi fruit, but its underlying mechanism remains unclear. In this study, we cloned the litchi PPO gene (LcPPO, JF926153), and described its expression patterns. The LcPPO cDNA sequence was 2120 bps in length with an open reading frame (ORF) of 1800 bps. The ORF encoded a polypeptide with 599 amino acid residues, sharing high similarities with other plant PPO. The DNA sequence of the ORF contained a 215-bp intron. After carrying out quantitative RT-PCR, we proved that the LcPPO expression was tissue-specific, exhibiting the highest level in the flower and leaf. In the pericarp of newly-harvested litchi fruits, the LcPPO expression level was relatively high compared with developing fruits. Regardless of the litchi cultivar and treatment conditions, the LcPPO expression level and the PPO activity in pericarp of postharvest fruits exhibited the similar variations. When the fruits were stored at room temperature without packaging, all the pericarp browning index, PPO activity and the LcPPO expression level of litchi pericarps were reaching the highest in Nandaowuhe (the most rapid browning cultivar), but the lowest in Ziniangxi (the slowest browning cultivar) within 2 d postharvest. Preserving the fruits of Feizixiao in 0.2-μm plastic bag at room temperature would decrease the rate of pericarp water loss, delay the pericarp browning, and also cause the reduction of the pericarp PPO activity and LcPPO expression level within 3 d postharvest. In addition, postharvest storage of Feizixiao fruit stored at 4°C delayed the pericarp browning while decreasing the pericarp PPO activity and LcPPO expression level within 2 d after harvest. Thus, we concluded that the up-regulation of LcPPO expression in pericarp at early stage of postharvest storage likely enhanced the PPO activity and further accelerated the postharvest pericarp browning of litchi fruit.  相似文献

Thermal analyses of freezing events in hydrated lettuce (LactucasativaL.) seeds show a correlation between low temperature exotherms(LTEs) (evidence of ice crystal formation) and seed death. Yet,weather patterns common to the Northern Great Plains of NorthAmerica regularly create conditions where non-dormant seedsof native plants hydrate with snow melt and are subsequentlyexposed to -30 °C or colder conditions. To determine ifsuch weather patterns decimate dispersed seeds, we measuredthe effects of freezing on fully hydrated winterfat (Eurotialanata(Pursh) Moq.) seeds harvested from the Northern Plainsat two USA and one Canadian location. Survival of hydrated seedsto -30 °C at a cooling rate of 2.5 °C h-1was similarto that of seeds not subjected to cooling, even though botha high temperature exotherm (HTE) and an LTE were observed.Although the LTE was not related to winterfat seed survival,freeze-stressed seeds had reduced germination rates and reducedseedling vigour, particularly for the collection with the lightestseeds. The temperature of LTEs was similar among seed collectionswith a mean of -17.6 °C, but was warmer when the seeds wereimbibed at 0 °C compared to 5, 10 or 20 °C. We founda significant correlation between the HTE and LTE temperatures.The difference and the correlation may be due to the highermoisture content of seeds imbibed at 0 °C. After pericarpremoval, only one exotherm in the range of the LTE was observed.This was also true for the naked embryo. 为探讨早春极端低温对龙眼成花的影响及芒果和荔枝花穗冷害发生时应对措施的效果,在2008年早春低温冷害时,通过抹除芒果和荔枝的冷害顶生花穗,研究该应对措施对促进腋芽再分化花芽并抽生花序的效果,并在低温冷害后,对不同龙眼品种的成花情况进行调查。结果表明:抹除荔枝冷害顶生花穗后能显著促进黑叶、钦州红荔、糯米糍、立夏红腋芽再生花序,平均单株花穗数分别为139、62.5、28和29穗,分别比对照的高119、22.5、25和26穗;而妃子笑、三月红、桂味和禾荔的处理树和对照树之间差异不明显。抹除芒果冷害顶生花穗后,台农1号、贵妃、桂热82号、红象牙和金穗芒的平均单株成花数分别为92、18、131、20.5和18穗,明显高于对照;而凯特芒、桂热120、吉尔、紫花芒和金穗芒处理树和对照树之间差异不显著。龙眼低温冷害后成花较好的有桂明、储良、石硖、小广眼、大乌圆和大广眼,平均单株花穗数分别为88、67、52.7、52、51和50穗;其次是桂香、乌龙岭、东壁、立冬本和早白露,平均单株花穗数分别为39、26、25、23.5和21.5穗。  相似文献

We report 12 microsatellites enriched in CT repeats obtained from a genomic library of the lychee (Litchi chinensis Sonn.) cultivar Mauritius. The polymorphisms revealed by these microsatellites were evaluated in a collection of 21 lychee cultivars. A total of 59 fragments were detected with these 12 SSRs, with an average of 4.9 bands/SSR. Three primer pairs seem to amplify more than a single locus. The mean expected and observed heterozygosities over the 9 single-locus SSRs averaged 0.571 (range: 0.137–0.864) and 0.558 (range: 0.169–0.779) respectively. The total value for the probability of identity was 7.53×10-5. In addition, the selected SSRs were used to amplify DNA from four longan cultivars. Eleven of the 12 SSRs produced amplification fragments in longan, and eight of these fragments were polymorphic. All except two of the products amplified from longan were the same size as those amplified from lychee, suggesting a close genetic proximity between the two species. The SSRs studied produced 22 different patterns, allowing the unambiguous identification of 16 lychee and the 4 longan cultivars studied. Discrimination was possible with just four selected microsatellites. Two groups with two and three undistinguishable cultivars were obtained, reflecting probable synonymies. Unweighted pair-group method of artimetic averages (UPGMA) cluster analysis divided the lychee cultivars studied into two main groups, one consisting of ancient cultivars and the other with more diverse recent cultivars. This is the first report of microsatellite development in the Sapindaceae, and the results demonstrate the usefulness of microsatellites for identification, similarity studies and germplasm conservation in lychee and related species.Communicated by H.F. Early and late varieties of the short day plant, Abelmoschusesculentus (L.) Moench. (= Hibiscus esculentus L.), were subjectedto various numbers of short days (SD; 10 h natural daylight)before transfer to 16 h long days. Short day treatments werestarted at the time of cotyledon release following seed germination,and their effects on the induction and development of flowersand fruits were recorded. While early okra required 6 SD cyclesfor the induction of flowering in all plants, late okra required14. However, both varieties needed more than 20 SD cycles forflower opening; flower buds produced by plants given fewer SDcycles abscinded with little or no development. In both varieties,the number of flower buds produced on each plant increased asthe number of short day cycles was increased from 5 to 20; flowerbuds were initiated at more axillary bud sites on the main stem.Three fruits were formed on each plant of both varieties subjectedto more than 30 SD cycles but, while the last formed fruit abscindedwithout development, the other two developed and became filledwith seeds. However, the greatest yield in fruit dry weightwas from plants kept under SD throughout and also in late okraplants given 55 SD. In general, the total number of seeds perplant increased as a larger number of SD cycles were imposed.  相似文献
