The incidence matrix of a BIB design for v treatments has been used to construct a biased spring balance weighing design. Conditions under which an optimum biased spring balance weighing design exists are given. The paper deals with the problem of estimating the individual weights of objects under a biased spring balance weighing design with equal correlations of errors in the model. A lower bound for the variance of each of the estimated weights resulting from this biased spring balance weighing design is obtained and a necessary and sufficient condition for this lower bound to be attained is given. In recent years, a growing number of metabolic engineering strain design techniques have employed constraint-based modeling to determine metabolic and regulatory network changes which are needed to improve chemical production. These methods use systems-level analysis of metabolism to help guide experimental efforts by identifying deletions, additions, downregulations, and upregulations of metabolic genes that will increase biological production of a desired metabolic product. In this work, we propose a new strain design method with continuous modifications (CosMos) that provides strategies for deletions, downregulations, and upregulations of fluxes that will lead to the production of the desired products. The method is conceptually simple and easy to implement, and can provide additional strategies over current approaches. We found that the method was able to find strain design strategies that required fewer modifications and had larger predicted yields than strategies from previous methods in example and genome-scale networks. Using CosMos, we identified modification strategies for producing a variety of metabolic products, compared strategies derived from Escherichia coli and Saccharomyces cerevisiae metabolic models, and examined how imperfect implementation may affect experimental outcomes. This study gives a powerful and flexible technique for strain engineering and examines some of the unexpected outcomes that may arise when strategies are implemented experimentally.  相似文献

Mass balances of total nitrogen and total phosphorus were calculated for Lake Verevi (area 0.126 km2, maximum depth 11 m, mean depth 3.6 m), a sharply stratified small lake located in South Estonia within the borders of the town Elva. The lake has up to 10 small inflows but only three of them are nearly permanent. Accidental overflows from near-by oxidation ponds during high floods have been the major source of the nutrient load of the lake in the past. L. Verevi receives a significant part of its inflow from groundwater, which is difficult to measure. In dry years the outflow is temporary. During summer the lake is sharply thermally and chemically stratified. The spring turnover is often incomplete even in homothermal conditions, thus giving the lake some meromictic features. The influx of nitrogen exceeded the outflux at any supposed proportion (20%, 50%, 80%) of surface runoff. The lake retained 45–90% of the nitrogen influx by sedimentation and/or by denitrification. The largest nitrogen losses with loss rates more than 10 kg N d−1 occurred in May and June. The calculated phosphorus retention rate became strongly negative during mixing periods. From June to November, phosphorus release from the sediment exceeded sedimentation by 205 kg in 1991 and by 79 kg in 1993. Earlier stagnation and absence of a full spring turnover in the 2000 has slowed down the recovery of the lake because less phosphorus is flushed out. However, the stronger stratification and significantly smaller phosphorus content in the epilimnion limits biological activity and as a result improves the water quality of the surface layer.  相似文献

Applying heat balance sap flow gauges to plant stems <10 mm in diameter has been difficult because a miniature design is needed that can be attached to a range of stem geometries. This report presents a modified gauge design for use on small plant stems of irregular geometry and shows results from Glycine max with stem diameters of 3–4 mm. The gauge was evaluated on container-grown plants by comparing gauge measurements of flow to gravimetric estimates of transpiration. Experiments were conducted in the laboratory and greenhouse, using artificial and natural lighting, respectively. Laboratory comparisons of gauge versus gravimetric water loss measurements indicated that the instrument was accurate to within ±5% when soil water was not limiting. Similar results were obtained from greenhouse tests except when soil water availability was low and cumulative gauge estimates became 30–45% less than gravimetric measurements. Differences may have reflected reduced plant water uptake or errors in sap flow estimates associated with low flow rates. Gauge accuracy was not improved by including the rate change in heat storage (S) in the flow calculations because S was always less then 3% of the total heat balance. Relationships between system temperature and sap flow rate suggested a diagnostic test for determining optimum power input. A time constant of 15 s indicated potential application in many agronomic and physiological studies.  相似文献

Ion and acid–base balance were examined in the freshwater-adapted mummichog (Fundulus heteroclitus) using a series of treatments designed to perturb the coupling mechanisms. Unidirectional Cl uptake (JClin) was extremely low whereas JNain was substantial (three- to sixfold higher); comparable differences occurred in unidirectional efflux rates (JClout, JNaout). JClin was refractory to all treatments, suggesting that Cl/base exchange was unimportant or absent. Indeed, no base excretion or modulation of ion fluxes occurred for acid–base balance for up to 8 h after NaHCO3 loading (injections of 1000 or 3000 nequiv.·g−1). Acute environmental low pH (4.5) and amiloride (10−4 M) treatments caused concurrent inhibition of JNain and net H+ excretion (JH+net), indicating the presence of Na+/H+ exchange. JNain was elevated and JH+net restored during recovery from both treatments, but this exchange did not appear to be dynamically adjusted for acid–base homeostasis. High external ammonia exposure (1 mmol·l−1) initially blocked ammonia excretion (JAmmnet) but had no effect on JNain, whereas high pH (9.4) reduced both JAmmnet and JNain. Inhibition of JNain by the low pH and amiloride treatments had no effect on JAmmnet. These results indicate that ammonia excretion is entirely diffusive and independent of both Na+uptake and the protons that are transported via the Na+/H+ coupling. In addition, ureagenesis served as a compensatory mechanism during high external ammonia exposure, as a marked elevation in urea excretion partially replaced the inhibited JAmmnet. In all treatments, changes in the Na+–Cl net flux differential were consistent with changes in JH+net measured by traditional water titration techniques, indicating that the former can be used as an estimate of the acid–base status of the fish. Overall, the results demonstrate that the freshwater-adapted F. heteroclitus does not conform to the ion/acid–base relationships described in the standard model based on commonly studied species such as trout, goldfish, and catfish.  相似文献

摘要:加强农田土壤保持耕作管理,科学认识和调控农田耕作系统能流碳流,提高农业生态系统固碳减排能力,对于减缓农业对全球温室效应的贡献具有重要意义。本研究以北方半湿润偏旱区山西寿阳旱作春玉米土壤保持耕作试验研究为基础,利用田间定位观测数据、辅助能投入参数,土壤呼吸田间原位测定,以及农业生态系统能量/碳平衡分析及碳循环过程模拟方法,综合分析和比较不同耕作(CT传统、RT少耕和NT免耕)系统能量/碳平衡及能-碳关联影响。与CT比较,采用RT和NT措施下工业能耗CO2-C损失降低约4%—12%(相当11—35 kg CO2-C?hm-2?a-1)。在RT和NT系统下耗能系数可降低约6%—10%,能量生产效率可提高约7%—12%。2006—2007年由田间原位测定土壤呼吸CO2-C释放通量估算,在玉米休闲期(尤其是秋耕处理后),NT条件下土壤呼吸速率一般为最低(NT NT(2005380)>CT(1987375)。不同耕作下的玉米籽粒产量与生育期土壤呼吸通量趋势基本吻合,如2006-2007年玉米产量(kg?hm-2?a-1)平均为,RT(5614268)>NT(5533564)>CT(5487278)。玉米籽粒产量与生育期土壤呼吸通量之间呈密切相关(R2=0.88)。本研究结果得出,RT和NT对农田耕作系统的影响呈碳汇效应,且一般为NT >RT;而CT处理表现为碳源。RT和NT通过增加土壤碳投入是维持和提高土壤有机碳的有效途径。  相似文献

Growth performance, protein and energy utilization in triploid rainbow trout juveniles of two yearclasses, 0 + and 1 +, were compared to diploid control groups. Growth rate, food conversion and protein efficiency ratios of triploid rainbow trout did not differ (0 + juveniles) or were slightly lower (1 + juveniles) than the diploid control. Proximate body composition was also not affected by ploidy.
Apparent digestibility coefficients (ADC) ofprotein and ofenergy, oxygen uptake, and ammonia excretion rates were not different between the two groups. Nitrogen and energy balance studies showed that digestive and metabolic utilization of the diets did not differ in triploid and diploid rainbow trout juveniles.  相似文献

Cockroaches dehydrated for 8 days lost nearly 50% of their haemolymph volume and approx 25% of their tissue water. Haemolymph osmolality and sodium, potassium, and chloride concentrations in the haemolymph and tissue water were all regulated within narrow limits. It is confirmed that sodium and potassium ions are sequestered within the fat body during periods of dehydration. The increase in sodium and potassium ions in the fat body is shown to arise from ionic regulation of haemolymph and other tissues. During periods of rehydration, sodium and potassium concentrations decrease in the fat body and haemolymph volume and ionic concentrations return to near original levels. A small proportion of the surplus haemolymph chloride ions is shown to be associated with the cuticle during times of water deprivation.  相似文献
