Abstract— The metabolism of single, first cortical rat brain slices was studied in response to incubation in media of various osmolalities. There was an inverse relationship between osmolality and the magnitude of the increase in tissue water content, and a direct relationship between osmolality and inulin space. Brain sodium varied directly with the media sodium, but hyperosmolal sucrose and glucose resulted in a drop in brain sodium. Brain potassium was constant in hyperosmolal sodium media, but it fell in hypo-osmolal media and hyperosmolal sucrose and glucose media.
Hypo-osmolality depressed the oxidation of [14C]glucose to 14CO2 whereas hyperosmolality obtained with sodium, sucrose, or glucose stimulated glucose oxidation. Lactate production was enhanced only by sodium hyperosmolality. Hypo-osmolality caused a 70% drop in phosphocreatine and a minor decrease in energy charge potential, analagous to the effects of hypoxia. Hyperosmolality had no effect on energy metabolism.
The total amino acid nitrogen released into the media was suppressed by hypo-osmolality but was increased by hyperosmolal incubation. Hyperosmolality also increased production of ammonia fourfold.
The time course of the change in tissue osmolality and ion content following incubation in hyperosmolal sodium media showed the tissue osmolality reached the media osmolality within 5 min, but it took 30 min for the tissue sodium to reach equilibrium with the media sodium. This indicates that unidentified or'idiogenic osmoles'are induced transiently by hyperosmolality associated with changes in amino acid and ammonia metabolism.  相似文献

油菜花蜜腺4枚,2故侧蜜腺和2枚中蜜腺。蜜腺由分泌表皮、产蜜组织和维管束组成。蜜腺原基是在花的各部分原基发生后,由花托局部区域的表面数层细胞经反分化形成。但它在短期内,完成了形态建成,与花的成熟同步。在蜜腺的发育过程中,产蜜组织细胞的液泡发生有规律的变化。同时,淀粉粒、蛋白质的动态也较为明显,以上变化与蜜汁的合成和分泌密切相关。其蜜腺的原蜜汁是由韧皮部提供,运转至产蜜组织内加工,最后由变态气孔和泌蜜通道泌出。  相似文献

Narcolepsy with cataplexy (NC) is a sleep disorder caused by the loss of the hypothalamic neurons producing hypocretin. The clinical hallmarks of the disease are excessive daytime sleepiness, cataplexy, other rapid eye movement (REM) sleep phenomena, and a fragmented wake-sleep cycle. Experimental data suggest that the hypocretin system is involved primarily in the circadian timing of sleep and wakefulness but also in the control of other biological functions such as thermoregulation. The object of this study was to determine the effects of the hypocretin deficit and of the wake-sleep cycle fragmentation on body core temperature (BcT) modulation in a sample of drug-free NC patients under controlled conditions. Ten adult NC patients with low cerebrospinal fluid (CSF) hypocretin levels (9 men; age: 38?±?12 yrs) were compared with 10 healthy control subjects (7 men; age: 44.9?±?12 yrs). BcT and sleep-wake cycle were continuously monitored for 44?h from 12:00?h. During the study, subjects were allowed to sleep ad libitum, living in a temperature- and humidity-controlled room, lying in bed except when eating, in a light-dark schedule (dark [D] period: 23:00–07:00?h). Sleep structure was analyzed over the 24-h period, the light (L) and the D periods. The wake-sleep cycle fragmentation was determined by calculating the frame-shift index (number of 30-s sleep stage shifts occurring every 15?min) throughout the 44-h study. The analysis of BcT circadian rhythmicity was performed according to the single cosinor method. The time-course changes in BcT and in frame-shift index were compared between narcoleptics and controls by testing the time?×?group (controls versus NC subjects) interaction effect. The state-dependent analysis of BcT during D was performed by fitting a mixed model where the factors were wake-sleep phases (wake, NREM stages 1 and 2, slow-wave sleep, and REM sleep) and group. The results showed that NC patients slept significantly more than controls during the 24?h due to a higher representation of any sleep stage (p?<?.001) during L, whereas the total amount of night sleep and its architecture were comparable in the two groups. Wake-sleep fragmentation was higher (p?<?.001) in NC subjects especially during L. Despite these differences, mesor (24-h mean), amplitude, and acrophase (peak time) of BcT circadian rhythm were comparable in narcoleptics and controls, and no between-group differences were detected in the time-course changes and in the state-dependent modulation at night of BcT. Synchronously dividing cells of Chlamydomonas reinhardtii Dang. (Chlorophyta) produce a single peak of cyclic adenosine monophosphate (cAMP), about sevenfold above the basal level, at the time of onset of the flagellar shortening that precedes mitosis. Cultures of a spontaneous palmelloid variant (which forms flagella-less cell clusters) of C. reinhardtii produce up to 15 times more cAMP per gram fresh weight of cells than do cultures of normal C. reinhardtii. Revertants from the palmelloid phenotype to the normal phenotype exhibit the low levels of cAMP characteristic of normal C. reinhardtii. Thus, elevation of cAMP level and decreased ability to form or maintain flagella are closely related phenomena. We propose that flagellar assembly/disassembly is regulated by endogenous cAMP in C. reinhardtii.  相似文献

低磷营养胁迫条件下,长穗偃麦草一中国春二体异附加系的RNA酶活性显著提高,RNase的IEF研究结果表明,对照条件下只有极微弱的活性带,而磷饥饿状态下则诱导出明显的特异活性带,但诱导的活性带无基因型间的差异,长穗偃麦草-中国春和灯芯偃麦草-中国春两套二体异附加系系分泌的酸性磷酸酶活性不同程度的显著提高。IEF结果表明,低磷胁迫诱导出低PH范围内的特异活性带,且活性带无基因型间的差异。  相似文献

Two outstanding questions in evolutionary biology are whether, and how often, the genetic basis of phenotypic evolution is predictable; and whether genetic change constrains evolutionary reversibility. We address these questions by studying the genetic basis of red flower color in Penstemon barbatus. The production of red flowers often involves the inactivation of one or both of two anthocyanin pathway genes, Flavonoid 3′,5′‐hydroxylase (F3′5′h) and Flavonoid 3′‐hydroxylase (F3′h). We used gene expression and enzyme function assays to determine that redundant inactivating mutations to F3′5′h underlie the evolution of red flowers in P. barbatus. Comparison of our results to previously characterized shifts from blue to red flowers suggests that the genetic change associated with the evolution of red flowers is predictable: when it involves elimination of F3′5′H activity, functional inactivation or deletion of this gene tends to occur; however, when it involves elimination of F3′H activity, tissue‐specific regulatory substitutions occur and the gene is not functionally inactivated. This pattern is consistent with emerging data from physiological experiments indicating that F3′h may have pleiotropic effects and is thus subject to purifying selection. The multiple, redundant inactivating mutations to F3′5′h suggest that reversal to blue‐purple flowers in this group would be unlikely.  相似文献

分布在甘肃河西走廊的4 种生态型芦苇(Phragm itescom m unisTrin.)的核酸代谢季节变化有差异。盐化草甸芦苇RNA 含量持续增加,DNA 含量相对稳定,其它3 种生态型芦苇的RNA 和DNA 含量以5月份为最高。过渡带芦苇的RNA 含量、沼泽芦苇及沙丘芦苇的DNA含量9 月份略有增高。盐化草甸芦苇与过渡带芦苇的DNase和RNase的活性7 月份最高,沼泽芦苇与沙丘芦苇的DNase和RNase活性5—9 月份呈增高趋势。盐化草甸芦苇的DNA 和RNA 合成活性不断升高,过渡带芦苇和沼泽芦苇的DNA 和RNA 合成活性及沙丘芦苇的RNA 合成活性5—9月份均降低,仅沙丘芦苇的DNA合成活性增强。RNA 聚丙烯酰胺梯度凝胶电泳分析结果表明,4 种生态型芦苇均含有25S、23S、18S、16S大分子量rRNA 和小分子量5.8S、5S、4.5SrRNA 及4StRNA。大分子量RNA 的含量高于小分子量RNA 含量。不同生态型及同一生态型芦苇的不同发育时期,相同的RNA 组分含量各不相同。且发育过程中23S、18S、16SrRNA 在不同月份发生不同程度的降解。由此,我们认为,核酸代谢的差异性是4 种生态型由生长转入衰  相似文献

The effects of silicon deficiency on the metabolism and composition of lipids in Cyclotella cryptica T13L Reimann, Lewin, and Guillard were examined. Silicon-deficient cells had higher levels of neutral lipids (primarily triacylglycerols) and higher proportions of saturated and monounsaturated fatty acids than silicon-replete cells. After 4 h of silicon deficiency, the percentage of newly assimilated NaH14CO3 partitioned into lipids increased from 27.6% to 54.1%, whereas the percentage partitioned into chrysolaminarin decreased from 21.6% to 10.6%. In addition, pulse-chase experiments with NaH14CO3 indicated that the amount of 14C in the total cellular lipid fraction increased by 32% after 12 h of silicon deficiency despite the absence of additional photoassimilable 14C. Therefore, the accumulation of lipids in response to silicon deficiency appears to be due to two distinct processes: (a) an increase in the proportion of newly assimilated carbon partioned into lipids, and (2) a slow conversion of previously assimilated carbon from non-lipid compounds into lipids  相似文献

镉污染后的水生维管束植物其叶绿素a/b值随水体镉浓度的增加而下降,叶绿素含量与镉浓度呈极显著的负相关。叶细胞膜透性与镉浓度呈极显著的正相关。不同抗性种叶片可溶性糖含量均随镉浓度增加而升高,但抗性种的增加率要小得多。镉对根系脱氢酶的活性产生抑制,且随镉浓度升高,活性相应下降。对过氧化物酶同工酶谱也产生明显的影响。这些在环境监测中可作为生理指标。凝胶层析表明,具有抗性的凤眼莲的可溶性成分中,存在着高分子镉复合物(>70,000)、分子量为13,300的镉复合物和低分子量镉螯合物及游离态镉。不抗种的紫背萍缺乏第二种镉复合物。  相似文献

本文用流式细胞术(FCM)和免疫细胞化学方法,检测了实验性小鼠脾的淋巴细胞增殖周期和5-溴脱氧尿苷阳性细胞(BrdU ̄+)在脾脏内的分布.以及脾小结中IgM ̄+细胞的反应。结果显示由利血平诱发的脾虚小鼠S期细胞数最少(占5%),G_1和G_2+M期细胞增加。经过健脾汤治疗后S期细胞增至10%,与对照组比较无显著性差异.但高于自然恢复组(P<0.001)。经免疫细胞化学显示:脾虚组小鼠的脾中5-溴脱氧尿苷阳性细胞(BrdU ̄+)的数量远低于其它组。脾小结缩小,帽状区IgM ̄+B细胞基本消失,经健脾汤治疗后,复健组的脾小结与自然恢复组比较,体积明显增大,生发中心的BrdU ̄+和帽状区IgM ̄+B淋巴细胞均相应增多。提示健脾汤有促进DNA和IgM合成及细胞增殖作用。  相似文献

本实验用大黄建立脾虚动物模型,以其肠道双歧杆菌水平为主要指标,配以电镜检查,观察回春生菌种及其四君子汤合剂对脾虚动物肠道菌群的调整作用。结果表明,单用回春生即可明显提高肠道双歧杆菌水平,但总体效果不如具有益气健脾作用的四君子汤。用回春生的目的在于直接补充肠道所需的双歧杆菌;四君子汤则主要使肠绒毛的形态恢复正常而改善微群落的生境。利用回春生中药合剂可作用于上述两个环节,产生协同作用,使疗效明显提高。  相似文献

为确定病原类型和造成虹鳟低死亡率的原因,研究对患病虹鳟进行了病理学观察、病毒的分离和鉴定以及动物感染实验。临床检查发现发病虹鳟体色变黑,肌肉和腹壁点状出血。病理学观察发现虹鳟造血器官脾和肾间组织严重坏死。通过反转录PCR法检测坏死组织和病变细胞中传染性造血器官坏死病毒、出血性败血病毒和传染性胰腺坏死病毒,并对得到的371 bp大小片段进行测序和构建进化树分析,发现感染病原为传染性造血器官坏死病毒。同时,给体重为1.5 kg健康虹鳟腹腔注射104 TCID50的组织滤液,累计死亡率达到35%。除此之外,将组织滤液接种到鲤鱼上皮瘤细胞系后出现了特征性病变。在实验过程中未发现细菌或寄生虫感染。结果证实引起虹鳟低死亡率的病原为传染性造血器官坏死病毒。  相似文献

目的探讨脾气虚胃溃疡证病结合模型结肠中5-羟色胺及其受体的变化,及加味四君子汤治疗脾虚证的机理。方法免疫组织化学染色和图像分析技术。结果脾气虚合并胃溃疡模型中结肠5-羟色胺及其受体阳性反应产物的含量均增加,经过加味四君子汤治疗后,上述指标的变化降低,接近对照组。结论大鼠脾气虚胃溃疡模型中结肠组织及其受体含量的增高可导致消化系统功能紊乱,可能是脾气虚胃溃疡的一种重要病因或病机之一,加味四君子汤能通过纠正这些变化而发挥治疗作用。  相似文献
