It is widely assumed that population size significantly affects the dynamics of plant populations. Smaller populations are threatened by genetic drift and inbreeding depression, both of which may result in a decrease of genetic variation and a resulting negative impact on plant fitness. In our study we analysed the patterns of random amplified polymorphic DNA (RAPD) variation among 10 Dictamnus albuspopulations of varying size. The aim was to examine local differentiation in relation to spatial isolation resulting from limited population size and geographical distancing between populations. Significant correlations were noted between population size and both percentage of polymorphic loci (P <0.01) and genetic diversity (P<0.01). The matrix correlation between genetic and geographical distances revealed that geographical differentiation was reflected in the RAPD profile (Mantel test: r2=0.34, P<0.001). We found the highest level of molecular variance of RAPD patterns among individuals within the populations (72.6%), whereas among-population variation accounted for only 21.6% of variation. These results were highly significant in that they indicated a restricted population differentiation, as would be expected from outcrossing species. An additional analysis of seed production showed that there was significant variation among populations in terms of mean seed number per flower and mean seed mass per population which could be attributed to differences in population size as well as levels of genetic variation.  相似文献

For improved fermentation of shoyu (soy sauce), a useful koji-making system has been developed using a mixed tane-koji of two shoyu koji moulds, namely Aspergillus oryzae K2 (length of conidiophores about 350 m) and the late-conidiation strain, A. oryzae HG (length of conidiophores about 2500 m). The mixed culture of strains K2 and HG had about twice the glutaminase activity of the single-strain cultures. In addition, the number of conidia in the mixed culture was about 10% of that in a culture of strain K2 alone.  相似文献

Solanum acaule Bitt. is a disomic tetraploid potato which has been assigned two endosperm balance numbers (EBN). It readily crosses with diploids but does not cross with other tetraploid species, although exceptions have been reported. The genetic basis of this behavior was studied in intra- and interspecific crosses involving plants of four introductions of this species and plants of one introduction of 2x S. commersonii Dun., one of 2x S. gourlayi Haw., and two of 4x S. gourlayi Haw., which have been assigned one EBN, two EBN, and four EBN respectively. Some of the pollinated pistils were used to analyze pollen-pistil compatibility reactions; the rest were left in the plants for seed production. At harvest, seeds were sorted according to size and plumpness, and the ploidy of the resulting plantlets determined from root tips. A model is proposed to explain the results of these crosses as well as the exceptions previously reported. It is based on the presence of two independent loci controlling the EBN, with two alleles in homozygosity: 1/2 and 0. This model, which is extended to cmm and grl, also explains the behavior of 3x (cmm x grl) hybrids in crosses with one-EBN, two-EBN, and four-EBN species reported in a previous work.  相似文献

Summary Previous experiments showed that the sympatric herbs Delphinium nelsonii and Ipomopsis aggregata compete for hummingbird pollination and that deleterious effects of the former species on seed set of the latter involve interspecific pollen transfer. However, seed set was not reduced when pollen of both species was applied simultaneously to I. aggregata stigmas. Hence a competitive effect may require arrival of foreign pollen before conspecific pollen. To explore this possibility we subjected I. aggregata flowers to a competition treatment in which they received D. nelsonii pollen 6 h before I. aggregata pollen, or to a control in which they received only the conspecific pollen. Foreign pollen precedence decreased mean seed set by almost 50%, which is consistent with effects observed in previous experiments. Reduced seed set can be explained by the fact that foreign pollen often caused stigma lobes to close together within 1.5–6 h, reducing subsequent receptivity. Stigma closure was also elicited by conspecific pollen, but not by mechanical stimulation, and was influenced by size of the pollen load and identity of the plant being pollinated.  相似文献

Gliadin and glutenin electrophoresis of F2 progeny from four crosses of durum wheat was used to analyse the linkage relationships between prolamin genes on chromosomes 1A and 1B. The results showed that these genes are located at the homoeoallelic lociGlu-1,Gli-3,Glu-3 andGli-1. The genetic distances between these loci were calculated more precisely than had been done previously for chromosome 1B, and the genetic distances betweenGli-A3,Glu-A3 andGli-A1 on chromosome 1A were also determined. Genes atGli-B3 were found to control some-gliadins and one B-LMW glutenin, indicating that it could be a complex locus.  相似文献

The frequency of outcrossing from a transgenic line of peas into three cultivars ('Carneval', 'Montana', 'Tipu') was studied in the field in 1997 and 1999. Two dominant traits, normal leaf form and a highly-expressed -glucuronidase (gusA) gene, were used as markers of pollen transfer. Because of heterogeneity in the commercial seed sources, leaf form alone was unreliable for assessing pollen migration into 'trap' plants. Of approximately 9000 offspring tested, only five plants scored positive for the presence of both markers. All five were located in 'trap' plots situated near the transgenic line. This represents a mean outcrossing rate of 0.07%.  相似文献

Summary The recently developed approach to deriving genetic markers via amplification of random DNA segments with single primers of arbitrary nucleotide sequence was tested for its utility in genetic linkage mapping studies with conifers. Reaction conditions were optimized to reproducibly yield clean and specific amplification products. Template DNA from several genotypes of Douglas-fir (Pseudotsuga menziesii) and white spruce (Picea glauca) were tested against eight ten-base oligonucleotide primers. Most of the tested primer/parent tree combinations yielded polymorphic PCR products (RAPD markers). Selected primers were then used in PCR reactions with template DNA isolated from offspring in Douglas-fir and black spruce diallel crosses among the same parental lines. The diallel study confirmed the appropriate inheritance of RAPD markers in the F1 generation. The value of these dominant RAPD markers for genetic linkage mapping in trees was established from both theoretical and applied perspectives.  相似文献

Restricted gene flow and localized selection should establish a correlation between physical proximity and genetic similarity in many plant populations. Given this situation, fitness may decline in crosses between nearby plants (inbreeding depression), and in crosses between more widely separated plants (“outbreeding depression”) mostly as a result of disruption of local adaptation. It follows that seed set and offspring fitness may be greatest in crosses over an intermediate “optimal outcrossing distance.” This prediction was supported for Ipomopsis aggregata, a long-lived herbaceous plant pollinated by hummingbirds. In six replicate pollination experiments, mean seed set per flower was higher with an outcrossing distance of 1–10 m than with selfing or outcrossing over 100 m. A similar pattern appeared in the performance of offspring from experimental crosses grown under natural conditions and censused for a seven-year period. Offspring from 10-m crosses had higher survival, greater chance of flowering, and earlier flowering than those from 1-m or 100-m crosses. As a result, 1-m and 100-m offspring achieved only 47% and 68%, respectively, of the lifetime fitness of 10-m offspring. Offspring fitness also declined with planting distance from the seed parent over a range of 1–30 m, so that adaptation to the maternal environment is a plausible mechanism for outbreeding depression. Censuses in a representative I. aggregata population indicated that the herbaceous vegetation changes over a range of 2–150 m, suggesting that there is spatial variation in selection regimes on a scale commensurate with the observed effects of outbreeding depression and planting distance. We discuss the possibility that differences in seed set might in part reflect maternal mate discrimination and emphasize the desirability of measuring offspring fitness under natural conditions in assessing outcrossing effects.  相似文献

Summary The effects of destruxin B, a host-specific toxin of Alternaria brassicae that causes black spot disease in oilseed brassicas, were studied on in vitro pollen germination and pollen-tube growth of Brassica campestris var brown sarson, B. juncea, B. napus cvs Westar and Cresor, B. nigra and Sinapis alba. Pollen grains of B. nigra, B. juncea and B. campestris were the most sensitive and those of S. alba the least sensitive to the toxin. Effects of the toxin were also studied on the leaves of these species, and the degree of sensitivity of leaves of different species was comparable to that of their pollen grains. The results on the responses of pollen grains as well as leaves to the toxin are in agreement with the degree of susceptibility/resistance of these species to A. brassicae reported in the literature, indicating that the genes imparting susceptibility/restistance are expressed in the pollen, a prerequisite for pollen selection. Results are also presented which show that the toxin fed to the cut end of isolated inflorescence axis is readily taken up by the developing pollen and results in the inhibition of germination of susceptible pollen. This technique offers a simple and effective method for application of selection pressure to eliminate pollen grains susceptible to the toxin from effecting fertilization.  相似文献

We have previously reported the isolation and characterization of a gene (Zm 13) from Zea mays which shows a pollen-specific pattern of expression. Stably transformed tobacco plants containing a reporter gene linked to portions of the Zm 13 5 flanking region show correct temporal and spatial expression of the gene. Here we present a more detailed analysis of the 5 regions responsible for expression in pollen by utilizing a transient expression system. Constructs containing the -glucuronidase (GUS) gene under the control of various sized fragments of the Zm13 5 flanking region were introduced into Tradescantia and Zea mays pollen via high-velocity microprojectile bombardment, and monitored both visually and with a fluorescence assay. The results suggest that sequences necessary for expression in pollen are present in a region from –100 to –54, while other sequences which amplify that expression reside between –260 and –100. The replacement of the normal terminator with a portion of the Zm13 3 region containing the putative polyadenylation signal and site also increased GUS expression. While the –260 to –100 region contains sequences similar to other protein-binding domains reported for plants, the –100 to –54 region appears to contain no significant homology to other known promoter fragments which direct pollen-specific expression. The microprojectile bombardment of Tradescantia pollen appears to be a good test system for assaying maize and possibly other monocot promoter constructs for pollen expression.  相似文献

The ability of two big brown bats (Eptesicus fuscus) to discriminate the distance to an electronically synthesized phantom target by echolocation was tested in the presence of interfering signals presented slightly before the target echo. Interfering signals were chosen to have differing degrees of similarity to the typical echolocation emission used by the bat in this task (which was the signal used to create the phantom target), and we predicted that the degree of disruption of ranging would be proportional to the similarity of the interference to the target echo. This prediction was not confirmed; rather, all interference signals not identical to the target echo increased the threshold to about twice that found with no interference. When the interference was identical to the target echo, the threshold increased to about 4 times that with no interference. When each bat was presented with phantom target echoes appropriate for the other bat, its range discrimination threshold increased about ten fold, and in this case the degree of interference of different signals was related to their similarity to the target echo, not to their similarity to the bat's normal signal. Restricted gene dispersion – resulting from both self-pollination and limited capability of pollen migration, as well as seed dispersion at short distances – has been considered the main reason for spatial genetic structuring in plant populations. This study evaluated the intrapopulation genetic structure and the mating system in four populations of Solanum lycocarpum, a woody bush occurring in Brazilian Cerrado vegetation. Two hundred and twenty-four individuals were genotyped through five nuclear SSR loci (30 alleles) and six cpSSR loci (82 haplotypes). The study evidenced that the species mates predominantly by outcrossings , that biparental inbreeding is not common, and that there are almost 10 trees participating as pollen donors per mother-tree. The populations were formed by many mother lineages, indicating efficient seed dispersion by the fauna and the occurrence of multiple foundation events. Spatial genetic structure was observed in three populations (average Sp=0.0184 ± 0.0030) and it resulted from both restricted seed dispersion and from vegetative reproduction. During the collection of seeds for ex situ conservation, seeds must be gathered from 150 to 200 mother trees, so that the effective size of 500 individuals is retained. The sampling must comprise the biggest possible number of populations in a wide area to enable the maintenance of the biggest possible haplotypic diversity.  相似文献

Summary Transmission of paternal chloroplasts was observed in Nicotiana, considered to inherit organelles in a strictly maternal way. Plants carrying streptomycin resistant plastids were used as pollen donors. Cell lines with paternal plastids in the offspring were selected as green (resistant) sectors on calli induced from the seedlings on streptomycin-containing media. The presence of paternal plastids in the regenerated plants was confirmed by restriction analysis. In the Nicotiana plumbaginifolia xN. plumbaginifolia Np(SR1)3 and the N. plumbaginifolia Np(gos)29 xN. tabacum SR1 crosses 2.5% and 0.07% of the offspring were found to contain paternal (tabacum) plastids, respectively. These plants, however, carried maternal mitochondria exclusively. This sexual cybridization method offers a simple way to transfer chloroplasts solely, a goal not accessible by protoplast fusion.  相似文献

Summary Ultrastructural analysis of the mature viable unhydrated pollen of maize,Zea mays from dehiscent anthers shows that the sperm cells are physically distant, each bounded by an envelope comprising their own plasma membrane and the inner plasma membrane of the vegetative cell. The chondriome is unusual in containing one or more filamentous complexes, up to 12m in length appressed to the side of the sperm nucleus. The extensions at each end of the elongate sperm cells contain longitudinally-oriented arrays of endoplasmic lamellae. We investigated the effect of petal-size variation in tall bellflower, Campanula americana, on pollen removal rates, seed set, and pollinator behavior. Pollen removal, seed set, and the behavior of two bee visitors (Bombus and Halictus) were assessed in flowers that had 0%, 50%, or 100% of their petal lobes removed. Pollen removal rates did not differ significantly among the three treatment groups, probably due to the presence of an ineffective (ugly) pollinator (Halictus), which quickly removed pollen from completely reduced flowers. Seed set was significantly positively correlated with petal size. Bumblebees discriminated against reduced flowers, thereby lowering pollen movement to reduced flowers. Halictid bees rarely contacted stigmatic surfaces and thus pollen delivery to reduced flowers was limited. The presence of an ugly pollinator probably obscured the effect of petal reduction on pollen receipt, thus masking the impact of attractive structures on male fitness. Given the positive relationship between seed set and petal size, we caution against ascribing expenditure on attractive structures exclusively to male function. In plants with many olules, expenditure on attractive structures may affect seed set.  相似文献

The mechanism of uptake of water-insoluble -sitosterol by a newly isolated strain of Arthrobacter simplex SS-7 was studied. The production of an extracellular sterol-pseudosolubilizing protein during growth of A. simplex on -sitosterol was demonstrated by isolating the factor from the cell-free supernatant and its subsequent purification by Sephadex G-150 column chromatography. The M r of the purified sterol-pseudosolubilizing protein determined by SDS–PAGE was 19kDa. The rate of sterol pseudosolubilization (5.2×10–3g l–1h–1) could not adequately account for the rate of sterol uptake (72×10–3g l–1h–1) and the specific growth rate (56×10–3 h–1). However in the unfavourable growth condition, when the cells were treated with sodium azide at the level of 30–60% of MIC, the sterol pseudosolubilization accounted for nearly 74% of the total growth containing 96% free cells. Cellular adherence to substrate particles was found to play an active role in the normal growth of the strain on -sitosterol. Unlike sodium acetate-grown cells, whose surface activity was negligible (60mNm–1), the sterol-grown cells had strong surface activity (40mNm–1). The high lipid content and long chain fatty acids in the cell-wall of -sitosterol-grown cells probably contribute to the high sterol adherence activity of the cells.  相似文献

Summary Numerous pollination treatments involving heavily irradiated (40–100 krad) pollen of diverse plant species failed to produce any clear cut egg transformants of the type reported by Pandey in Nicotiana. Genetic stocks of pea, rapeseed, and apple, bearing multiple Mendelian markers, were employed to detect any possible transformation events. For each plant species, an optimal level of irradiation was determined which would allow normal pollen tube growth leading to fertilization, but which would prevent the formation of normal hybrids due to the pulverized condition of the chromosomes contributed by the irradiated pollen. Pollination treatments included selfing, pollination with donor pollen mixed with self pollen, pollination with irradiated donor pollen mixed with self pollen, pollination with irradiated pollen followed by a delayed self pollination, and pollination with irradiated pollen by itself. None of these treatments produced clearly transformed seedlings. The total number of potential transformation events screened was in excess of 6,046 including 2,268 for pea, 3,309 for rapeseed, and 469 for apple. It is concluded that if egg transformation occurs outside of Nicotiana it is a rare event, and its frequent occurrence in Nicotiana must be, at best, an isolated phenomenon.Approved by the Director of the New York State Agricultural Experiment Station for publication as Journal Paper No. 3557  相似文献

Gold particles coated with -glucuronidase (GUS) mRNA with a 5 cap structure that had been synthesized in vitro were introduced, by use of a pneumatic particle gun, into pollen grains of lily (Lilium longiflorum), freesia (Freesia refracta) and tulip (Tulipa gesneriana). A fluorometric assay for the GUS activity indicated that in vitro synthesized GUS mRNA introduced into these pollen cells by particle bombardment was successfully expressed. GUS activity in extracts of the bombarded lily pollen became detectable fluorometrically within 30 min after bombardment, peaked at 6 h, then gradually decreased. This activity changed as a function of the developmental stage of the pollen cell of lily.  相似文献

Abies alba (Mill.) is native only to some parts of central Europe. The tree is indigenous to the Alps, the Black Forest, the Vosges, the Bayerischer Wald, and to some hilly and mountainous areas north of the Alps. A number of pollen diagrams from these regions demonstrate a clear Abies decline dating to the Roman period, and it appears that this Abies decline is a good time marker in central European pollen diagrams from the area mentioned. Firs were felled where they grew and their trunks were transported by the rivers Rhine and Danube to regions where Abies timber was needed for the construction of houses and bridges. During the excavation of sites situated far from indigenous fir stands, Abies needles and wood were found, indicating that the fir trunks were transported to these Roman towns by river. Fir timber is extremely valuable for the construction of large buildings, boats, and bridges. It is apparent that long distance transport on the central European rivers began in Roman times. Transport of timber was, of course, only a part of the new river commerce: building stone, wine, and crops were also transported for long distances.  相似文献

Pollen can function as a vehicle to disseminate introduced, genetically engineered genes throughout a plant population or into a related species. The measurement of the risk of inadvertent dispersal of transgenes must include the assessment of accidental dispersion of pollen. Factors to be considered include the rate of pollen spread, the maximal dispersion distance of pollen, and the spatial dynamics of pollen movement within seed production fields; none of which are known for alfalfa (Medicago sativa L.), an insect-pollinated crop species. Using a rare, naturally occurring molecular marker, alfalfa pollen movement was tracked from seed and hay production fields. Results indicated that leafcutter bees (Megachile spp.) used in commercial seed production show a directional, non-random bias when pollinating within fields, primarily resulting in the movement of pollen directly towards and away from the bee domicile. Within-field pollen movement was detected only over distances of 4 m or less. Dispersal of pollen from alfalfa hay and seed production fields occurs at distances up to 1000 m. By examining widely dispersed, individual escaped alfalfa plants and their progeny using RAPD markers, gene movement among escaped alfalfa plants has been confirmed for distances up to 230 m. The outcrossing frequency for large fields was nearly 10-times greater than that of research-sized plots. A minimum isolation distance of 1557 m may be required to prevent gene flow in alfalfa. Data suggest that complete containment of transgenes within alfalfa seed or hay production fields would be highly unlikely using current production practices.
