With tomato spotted wilt virus in petunia leaf strips, N-6 benzyladenine (BA) was as effective as kinetin in decreasing numbers of local lesions, a result which could not be attributed to an effect on the virus per se. Benzimidazole, adenine and ammonium nitrate were without effect. Benzyladenine was more effective than kinetin when supplied through the petioles of excised whole leaves. Local lesions and infectivity of TSWV in detached leaves of Nicotiana rustica were decreased by supplying BA before and after inoculation. Lesions and infectivity were also decreased in attached leaves when BA was applied 9 days before inoculation. BA supplied to attached leaves after inoculation increased infectivity. Supplying BA to the lower leaves of tomato plants before inoculating with TSWV decreased infectivity of unsprayed, systemically infected tip leaves taken as inoculum; BA supplied after inoculation increased infectivity. Local lesions caused by lucerne mosaic virus in excised leaves of Phaseolus vulgaris were decreased in number by supplying BA. The effects of pre- and post-inoculation sprays of BA are considered in relation to cell metabolism. Since pretreating leaves with kinins did not prevent infection, it is suggested that those which move freely through plants without adverse effects on normal growth may prove of value in increasing the tolerance of plants to virus infection.  相似文献

From the type strain of tobacco mosaic virus, defective strains were isolated that produced chlorotic or ringspot type symptoms in tobacco and were difficult to transmit without carborundum in the inoculum. Their concentration was less than 0–1 μg/ml of sap instead of the usual 2 mg/ml with the type strain. Phenol extracts of infected leaves were a little more infective than extracts in buffer, whereas phenol extracts of leaves infected with type strain were very much less infective than extracts in buffer. Electron microscopy of infective sap rarely showed any virus particles, but preparations concentrated by ultracentrifugation contained virus particles, many of which were broken or seemed inadequately assembled. Changing the ambient temperature at which infected plants were kept from 20 to 35°C did not increase the amount or improve the appearance of the virus. Some of the strains were inactivated during heating for 10 min between 70 and 80 °C. Undiluted sap lost its infectivity in 3 days at 20 °C, as did the type strain when diluted to 0–1 μg/ml in sap from healthy leaves. This is because substances that inhibit infection were produced by microbes in the sap. The ability of sap from healthy leaves to inhibit infection increased by more than twenty-five times when left 3 days at 20 °C. Infectivity of appropriate mixtures of type strain and aged sap was restored by diluting them in buffer. Sodium azide at 0·02% in sap prevented formation of the inhibitor. The infectivity of the defective strains increased when inoculated together with the type strain.  相似文献

Substances inhibiting local infection with virus X were isolated from leaves of potato cultivars Saphir, Saco, and Blaník by means of ultracentifugation and were concentrated with ammonium sulphate. These inhibitors were effective if rubbed into leaves of some host plants at least 2–4 days prior to infection without using any abrasives.  相似文献

Abstract Back transmission trials on young forest plants with isolates of purified viruses from the same tree species were performed using different inoculation techniques. Spruce seedlings and willow plants were successfully infected with tobacco necrosis virus (TNV) by the conventional method of mechanical inoculation of virus suspension mixed with celite as abrasive. Cherry leaf roll virus (CLRV) was transmitted to birches only after adding poly-L-orithine (PLO) to the inoculum. The same method was successful with brome mosaic virus (BMV) on beech seedlings. PLO also improved the rate of infection on TNV in willows. In only one case, was CLRV transmitted conventionally to a white ash seedling. The infection of white ash was increased when frozen powders, of infected ash leaves were directly rubbed onto leaves. BMV could not be transmitted to beech seedlings by carborundum pressure-inoculation. Stem slashing-inoculation of BMV and CLRV was successful with CLRV in one beech out of 60 seedlings.  相似文献

Big-vein diseased lettuce plants contained an agent that could consistently be transmitted mechanically to Chenopodium quinoa, in which it caused characteristic local lesions. Mechanical transmission was also possible to five other plant species including Nicotiana benthamiana, N. clevelandii and N. occidentalis, but not to lettuce. Symptoms in N. occidentalis were reminiscent of those of tobacco stunt disease. With zoospores of originally virus-free Olpidium brassicae, subcultured on the roots of N. occidentalis-P1, sap-inoculated either from lettuce or via C. quinoa, the agent could be transferred back to lettuce in which characteristic symptoms of big-vein were reproduced.
Infectivity in sap at room temperature was reduced by half after 2 h, and was practically lost after one day. Thermal inactivation was considerable at 45°C and complete at 50°C. Most infectivity was lost at dilution 1:5, and the dilution end-point was 1:10. The agent survived well in leaf material stored at -80°C, or in sap from leaves ground in buffer with DIECA and activated charcoal and freeze-dried. Mechanical transmission required low dilution (1:2) in the buffer with charcoal, and chilling of materials and utensils.
In lettuce, N. occidentalis-P1 and C. quinoa, with all isolates tested but one, infection was always associated with the presence of rod-shaped particles which in the literature have been associated with lettuce big-vein, and are similar to those described for tobacco stunt. Results obtained corroborate the assumption that these particles are the virions of lettuce big-vein virus. The virus also resembles tobacco stunt virus in mechanical transmissibility, instability in sap and symptoms on N. occidentalis.  相似文献

A virus causing a wilt of Datura stramonium was identified as a strain of henbane mosaic virus. It causes necrotic local lesions in Nicotiana rustica , and local lesions are demonstrable in tobacco by staining with iodine. Some of the factors affecting its transmission by Myzus persicae (Sulz.) were studied quantitatively using these lesions.
Infective aphids differed little in their ability to cause infection, and usually produced two or three lesions. The duration of the feeding puncture did not affect the number of infections and had little effect on the percentage of aphids becoming infective. Transmissible virus did not seem to be continually imbibed while aphids fed on infected plants, and there were indications that it was acquired immediately before aphids withdrew their stylets from the leaf. Aphids became infective when allowed to make feeding punctures into epidermis stripped from infected leaves.
M. persicae transmitted during feeding punctures as brief as 5–10 sec; the probability of single feeding punctures resulting in infection reached a maximum with those lasting from 20 to 30 sec, during which the stylets did not penetrate as far as the centre of the epidermal cell and little or no saliva appeared to be ejected. M. persicae did not transmit the virus when its stylets were artificially wetted with infective sap.
Periods of darkness before inoculation with datura wilt virus increased the susceptibility of Nicotiana rustica to infection by rubbing, but not to infection by aphids.  相似文献

The inhibition of infection by tobacco necrosis and tobacco mosaic viruses by tannic acid, and by extracts of raspberry and strawberry leaves, was associated with the precipitation of the viruses. Precipitation and inhibition were reversible, and infective virus was obtained from the precipitate formed between the viruses and tannins. Infectivity was fully restored by diluting mixtures of virus and tannin adequately and partially restored by adding alumina or nicotine sulphate.
Viruses and tannins are thought to form non-infective complexes, in which the virus and tannin components are held together by co-ordinate linkages or hydrogen bonds.
Macerating tobacco leaves infected with tobacco mosaic virus together with raspberry leaves greatly decreased the infectivity of the extracts; adding nicotine sulphate to the mixture of leaves before it was ground increased the infectivity, even though nicotine sulphate alone decreases the infectivity of tobacco mosaic virus. Even in the presence of nicotine sulphate, much of the virus was precipitated by substances from the raspberry leaves.
Extracts of roots of Fragaria vesca plants, infected with a tobacco necrosis virus, were more infective when made by macerating the roots with four times their weight of buffer at pH 8 than when made without buffer. Various methods are suggested for facilitating the transmission of viruses from plants that contain tannin.  相似文献

Among 11 epiphytic microorganisms one species each of Pseudomonas, Erwinia, and Aspergillus were antagonistic to Xanthomonas translucens subsp. oryzicola. Symptoms of bacterial leaf streak did not develop when the antagonists were sprayed on rice leaves 24 h before inoculation. Although the symptoms developed when the antagonists were applied 24 h after inoculation, the number of lesions and their length was significantly reduced over control. When the mixture of each antagonist and the pathogen was applied, no symptoms developed with Pseudomonas and Aspergillus sp. However, the symptoms could develop with Erwinia sp. although the number and length of the lesions was reduced over control.  相似文献

Potato plants (cv.‘Irish Cobbler’ with no major resistance genes to Phytophthora infestans), the lower or upper leaves of which were previously treated with hyphal wall components (HWC) of the fungus by rubbing with carborundum, acquired an induced resistance in other untreated leaves against cultivar-pathogenic races of P. infestans when challenged by spraying with a zoospore suspension. Such induced resistance was significantly shown to exist from at least 1 to 20 days after induction treatment with HWC. Thus, the treated plants were protected from severe late blight disease while non-induced control plants finally died of the disease. The induced resistance was due to a reduction of the number of successfully germinating zoospores, and subsequent penetration and then occurrence of hypersensitive-like cell response to the penetrating organisms. These results suggested that local leaf tissues of potato plants reacting to HWC may provide some systemic information that activates or enhances some resistance to P. infestans.  相似文献

Using carborundum as an abrasive and light rubbing with a culture of Agrobacterium tumefaciens, leaves of various species of bean and tobacco develop tumors on the leaf lamina. The induction of these tumors requires wounding, the presence of a virulent strain of the bacterium and is due to the bacterium, not substances released into the bacterial culture medium during growth. Observations of the histology and cytology of these tumors on the primary leaves of pinto bean show no significant differences from the more commonly studied stem tumors. The tumors on pinto beans first appear as chlorotic nests of dividing cells which gradually accumulate chlorophyll, eventually becoming dark green in color as opposed to the surrounding leaf tissue which is completely chlorotic at this stage. Tumor development is enhanced by a dark period following inoculation while growth of the leaf is essentially stopped. The tumors thus exhibit a pattern of growth and development independent of that of the normal leaf. The number of tumors obtained on pinto bean leaves was found to depend on the concentration of bacteria in the inoculum and on the age of the plants. A sharp peak in response was observed at about 7 days from planting. Best results were obtained by adding the bacterium at the time of wounding. The tumors were shown to differ from IAA-induced leaf proliferations with respect to their point of origin on the leaf, morphology, physiology and development.  相似文献

Tobacco mosaic virus (TMV) RNA was introduced directly into mesophyll cells of Nicotiana tabacum var. Samsun using electric-field pulses (electroinjection). The injected gene was successfully expressed in the recipient cells as judged by the assay for the virus coat protein using immunofluorescence and by the virus infectivity assay of the homogenate of the electroinjected cells for local lesions on tobacco leaves. As much as 50% of the cells that survived 24 days after electroinjection showed immunofluorescent specks.  相似文献

Large quantities of HIV are found trapped on the surface of follicular dendritic cells (FDCs), and virus persists on these cells until they ultimately die. We recently found that FDCs maintain HIV infectivity for long periods in vivo and in vitro. Because FDCs trap Ags (and virus) in the form of immune complexes and are rich in FcgammaRs, we reasoned that Ab and FcgammaRs may be required for FDC-mediated maintenance of HIV infectivity. To investigate this hypothesis, HIV immune complexes were formed in vitro and incubated for increasing times with or without FDCs, after which the remaining infectious virus was determined by HIV-p24 production in rescue cultures. FDCs maintained HIV infectivity in vitro in a dose-dependent manner but required the presence of specific Ab for this activity regardless of whether laboratory-adapted or primary X4 and R5 isolates were tested. In addition, Abs against either virally or host-encoded proteins on the virion permitted FDC-mediated maintenance of HIV infectivity. We found that the addition of FDCs to HIV immune complexes at the onset of culture gave optimal maintenance of infectivity. Moreover, blocking FDC-FcgammaRs or killing the FDCs dramatically reduced their ability to preserve virus infectivity. Finally, FDCs appeared to decrease the spontaneous release of HIV-1 gp120, suggesting that FDC-virus interactions stabilize the virus particle, thus contributing to the maintenance of infectivity. Therefore, optimal maintenance of HIV infectivity requires both Ab against particle-associated determinants and FDC-FcgammaRs.  相似文献

A virus that causes chlorotic streaks on ryegrass leaves was transmitted by the eriophyid mite Abacarus hystrix (Nalepa). Virus-free mites acquired the virus in 2 hr. feeding on infected ryegrass and the proportion that became infective increased with increased feeding time up to 12 hr.; vectors lost infectivity within 24 hr. of leaving the infected leaves. All instars of A. hystrix transmitted the virus.
The virus was transmitted by manual inoculation of sap to other species of Gramineae, including oats, rice, cocksfoot and meadow fescue, but none of these hosts seemed to contain as much virus as ryegrass; their saps did not precipitate specifically with antiserum prepared against the virus in ryegrass, whereas sap from infected ryegrass precipitated up to a dilution of 1/32. Infective sap of S22 Italian ryegrass contained flexuous rod-shaped particles; the dilution end-point of the virus was about 1 in 1000; the virus was inactivated when held for 10 min. at 60°C. and most of its infectivity was lost after 24 hr. at room temperature.  相似文献

Altered photosynthetic reactions in cucumber mosaic virus (CMV) inoculated leaves of virus resistant lines L113 and L57 and susceptible pepper (Capsicum annuum L.) plants cv. Albena grown in controlled environment and in the field were investigated. The CMV inoculated leaves of virus resistant lines developed different symptoms—necrotic local lesions on L113 and chlorotic spots on L57 while the same leaves of susceptible cv. Albena were symptomless. The changes in Photosystem II (PSII) and PSI electron transport were evaluated by chlorophyll fluorescence, and far-red (FR) light induced leaf absorbance A 810–860. CMV infection caused a decrease in maximal PSII quantum yield, F v/F m, in susceptible leaves. Increased non-photochemical fluorescence quenching in CMV-inoculated leaves of both resistant lines were observed. In CMV-inoculated leaves of all tested plants FR light induced P700 oxidation was decreased. In the present study, the viral-infected pepper plants grown in controlled environment to avoid the effects of abiotic factors were used as model system that allow us to investigate the differences in leaf senescence in CMV-inoculated leaves of susceptible and resistant pepper lines expressing different symptoms. Earlier leaf falls of inoculated leaves as a result of accelerated leaf senescence is important for building successful secondary virus resistance strategy following fast responses such as hypersensitive reaction.  相似文献

Abscisic acid (ABA) did not affect the infectivity of tobacco mosaic virus (TMV) in vitro. The same dilutions of ABA when applied on the leaves of Chenopodium amaranticolor Coste and Reyn. at different intervals before inoculation affected development of local lesions variably at different dilutions. The inhibition of local lesion formation was reduced at other intervals leading to stimulation at thirty minutes and six hours intervals. Post-inoculation treatments with 2 mg/l of ABA gave stimulation of local lesion formation, though other dilutions gave inhibition. Viral concentration was stimulated in the tomato seedlings root dipped in 0.2 mg/l of ABA for 6 hours and inoculated 24 hours after transplantation. Incorporation of different concentrations of ABA into tissue culture medium reduced the growth of the TMV infected tobacco callus tissue and stimulated the infectivity of the tissue grown over it assayed after three weeks.  相似文献

本研究探讨利用荧光定量PCR技术评价Sindbis病毒经亚甲蓝光化学处理后灭活效果的可行性。研究采用不同光照强度对Sindbis病毒进行亚甲蓝光化学灭活处理,并用SYBR Green I荧光定量PCR对Sindbis病毒的cDNA进行扩增,同时以细胞病变法做平行对照以测定病毒残余滴度。结果显示在亚甲蓝光化学处理过程中,随着光照强度的增强,病毒残余滴度由6.50 LgTCID50/mL逐渐降低至检测限以下,同时病毒核酸的拷贝数显著下降(P<0.05),并与病毒感染性的降低呈线性相关(R2>0.98)。以上结果表明,亚甲蓝光化学灭活法对Sindbis病毒核酸有破坏作用,病毒核酸损伤程度随光照强度的增强而增加,且与病毒感染性的降低存在相关性,提示荧光定量PCR技术评价亚甲蓝光化学法的病毒灭活效果具有可行性。  相似文献

Carbohydrate structures are often involved in the initial adhesion of pathogens to target cells. In the present study, a panel of anticarbohydrate monoclonal antibodies (MAbs) was tested for their ability to inhibit in vitro human immunodeficiency virus infectivity. MAbs against three different N- and O-linked carbohydrate epitopes (LeY, A1, and sialyl-Tn) were able to block infection by cell-free virus as well as inhibit syncytium formation. Inhibition of virus infectivity was independent of virus strain (HTLVIIIB or patient isolate SSI-002), the cell line used for virus propagation (H9 or MT4), and the cell type used as the infection target (MT4, PMC, or selected T4 lymphocytes). Inhibition was observed when viruses were preincubated with MAbs but not when cells were preincubated with MAbs before inoculation, and the MAbs were shown to precipitate 125I-labeled gp120. The MAbs therefore define carbohydrate structures expressed by the viral envelope glycoprotein gp120, indicating that glycans of the viral envelope are possible targets for immunotherapy or vaccine development or both.  相似文献

The infectivity of sap expressed from the lower epidermis stripped from leaves systemically infected with potato virus Y , henbane mosaic virus or tobacco mosaic virus was compared with that of sap from the underlying mesophyll. Results suggested that the concentration of virus in each of the two tissues was about the same.
Ultra-violet irradiation of leaves infected with potato virus Y or henbane mosaic virus greatly reduced the infectivity of sap expressed from subepidermal tissues.  相似文献
