Eggs of the common snapping turtle, Chelydra serpentina, were incubated at constant temperatures ranging from 20°C to 30°C, At hatching, the oviducts were absent or incomplete in males; the testes were differentiated. In females at hatching, the oviduct was intact hut in some cases the gonad retained bisexual characteristics. Three months after hatching, the ovary was differentiated and contained follicles. Eggs incubated at 20°C and at 30°C developed into females in 100% of the cases. At 26°C, 99% of the individuals were males; at 24°C, 100% were males. More males than females developed at incubation temperatures of 22°C and 28°C.  相似文献

In an experiment repeated for two separate years, incubation temperature was found to affect the body size and swimming performance of hatchling green turtles (Chelonia mydas). In the first year, hatchlings from eggs incubated at 26°C were larger in size than hatchlings from 28 and 30°C, whilst in the second year hatchlings from 25.5°C were similar in size to hatchings from 30°C. Clutch of origin influenced the size of hatchlings at all incubation temperatures even when differences in egg size were taken into account. In laboratory measurements of swimming performance, in seawater at 28°C, hatchlings from eggs incubated at 25.5 and 26°C had a lower stroke rate frequency and lower force output than hatchlings from 28 and 30°C. These differences appeared to be caused by the muscles of hatchlings from cooler temperatures fatiguing at a faster rate. Clutch of origin did not influence swimming performance. This finding that hatchling males incubated at lower temperature had reduced swimming ability may affect their survival whilst running the gauntlet of predators in shallow near-shore waters, prior to reaching the relative safety of the open sea.  相似文献

Incubation temperature and the amount of water taken up by eggs from the substrate during incubation affects hatchling size and morphology in many oviparous reptiles. The Brisbane river turtle Emydura signata lays hard-shelled eggs and hatchling mass was unaffected by the amount of water gained or lost during incubation. Constant temperature incubation of eggs at 24 °C, 26 °C, 28 °C and 31 °C had no effect on hatchling mass, yolk-free hatchling mass, residual yolk mass, carapace length, carapace width, plastron length or plastron width. However, hatchlings incubated at 26 °C and 28 °C had wider heads than hatchlings incubated at 24 °C and 31 °C. Incubation period varied inversely with incubation temperature, while the rate of increase in oxygen consumption during the first part of incubation and the peak rate of oxygen consumption varied directly with incubation temperature. The total amount of oxygen consumed during development and hatchling production cost was significantly greater at 24 °C than at 26 °C, 28 °C and 31 °C. We used eggs of Deinagkistrodon acutus to study the effects of incubation temperature on hatching success, embryonic expenditure of energy and hatchling phenotypes. One egg from each of the 15 fertile clutches was dissected for determination of egg composition, and a total of 164 eggs were incubated at five constant temperatures. Embryonic mortality increased dramatically at 30 °C, and none of eggs incubated at 32 °C hatched. Within the range from 24 to 30 °C, temperature affected incubation length and most hatchling traits examined. The mean incubation length at 24, 26, 28 and 30 °C was 36.4, 28.7, 21.8 and 15.7 days, respectively. Embryos developing at higher temperatures (28 and 30 °C) consumed more energy but produced less developed (and hence smaller) hatchlings, which characteristically had larger residual yolks but smaller carcasses. A principal component analysis resolved two components (with eigenvalues ⩾1) from ten size (initial egg mass)-free hatchling variables, accounting for 79.3% of variation in the original data. The first component (43.8% variance explained) had high positive loading for size-free values of dry mass, lipid mass, energy contents and ash mass of hatchlings, and the second component (35.5% variance explained) had high positive loading for size-free values of SVL, carcass dry mass and fatbody dry mass. Hatchlings from different incubation temperatures did not differ in scores on the first axis of the principal component analysis, whereas hatchlings from higher incubation temperatures (28 and 30 °C) had significantly lower scores on the second axis than did those from lower incubation temperatures (24 and 26 °C). As the second axis mainly represents traits relating to the developmental condition at hatching, the analysis therefore provided further evidence that eggs incubated at higher temperatures produced less developed hatchlings. Taken together, our data show that the optimal temperatures for embryonic development are relatively low in D. acutus largely due to its use of relatively cool habitats.  相似文献

Gonadal sex differentiation is temperature-dependent in Alligator mississippiensis; testis differentiation occurs in embryos incubated at 33°C and ovary differentiation occurs in embryos incubated at 30°C. Laminin and cytokeratin were examined immunohistochemically in the gonads of alligator embryos incubated at these temperatures. The aim of this study was to determine whether these structural proteins show the same sex-specific expression patterns reported for mammalian embryos, and to assess their usefulness as early markers of gonadal differentiation in species with temperature-dependent sex determination. Laminin delineated enlarged seminiferous cords in differentiating testes from developmental stage 23 to hatching. Laminin distribution was more diffuse and revealed smaller cords of cells in differentiating ovaries. Cytokeratin was also detected in developing gonads of both sexes. Cytokeratin became concentrated in the basal cytoplasm of differentiating Sertoli cells in developing testes. In developing ovaries, prefollicular cells of the ovarian cortex and cell cords in the medulla stained strongly for cytokeratin. Cytokeratin did not show the same basal distribution in female medullary cord cells as seen in the Sertoli cells of testes, however. These sex-specific patterns of laminin and cytokeratin distribution in embryonic alligator gonads may serve as early markers of sexual differentiation.  相似文献

In embryos of many reptiles, the sexual differentiation of gonads is temperature-dependent. In the turtle Emys orbicularis, all individuals become phenotypic males at 25 degrees C, whereas 100% phenotypic females are obtained at 30 degrees C. Steroid metabolism in embryonic gonads was studied at both temperatures, during and after the thermosensitive period for sexual differentiation. Pools of gonads were incubated for various times, with 3 beta-hydroxy-5-pregnen-20-one (pregnenolone), progesterone, dehydroepiandrosterone or 4-androstene-3,17- dione as substrates. The analysis of metabolites combined two successive chromatographies (HPLC and TLC) and autoradiography. Conversion of pregnenolone to progesterone and of dehydroepiandrosterone to 4-androstene-3,17-dione was more important in testes at 25 degrees C than in ovaries at 30 degrees C. In ovaries, a large amount of 5-pregnene- 3 beta,20 beta-diol was formed from pregnenolone, and 5-androstene-3 beta,17 beta-diol was produced from dehydroepiandrosterone. In both testes and ovaries, 5 alpha-pregnane and 5 alpha-androstane derivatives were the main metabolites obtained from progesterone and 4-androstene-3,17-dione, respectively. Progesterone was also converted to 20 beta-hydroxy-4-pregnen-3-one. Dehydroepiandrosterone and 4-androstene-3,17-dione were also metabolized into 11 beta-hydroxy-4-androstene-3,17-dione (only in testes), testosterone, 11 beta,17 beta-dihydroxy-4-androstene-3-one, 17 beta-hydroxy-4-androstene-3,11-dione (low amounts in testes, traces in ovaries), 17 alpha-hydroxy-4-androstene-3-one, estrone and estradiol-17 beta (traces).  相似文献

The evolutionary significance of sex-determining mechanisms, particularly temperature-dependent sex determination (TSD) in reptiles, has remained unresolved despite extensive theoretical work. To investigate the evolutionary significance of this unusual sex-determining mechanism, I incubated eggs of the common snapping turtle (Chelydra serpentina) at a male-producing temperature (26°C), a female-producing temperature (30°C), and an intermediate temperature that produced both sexes about equally (28°C). Laboratory experiments indicated that two performance variables, but no morphological measurements, were significantly influenced by incubation temperature (P ≤ 0.05): hatchlings from cooler incubation treatments swam faster than turtles from warmer incubation treatments, and hatchlings from 28°C exhibited a greater propensity to run than did individuals from 26°C and 30°C. In the field, hatchlings from the all-male and all-female producing temperatures had significantly higher first-year survivorship than did consexuals from the incubation temperature that produced both sexes (G = 6.622, P = 0.03). Significant directional selection was detected on propensity of hatchlings to run (β′ = –0.758, P = 0.05): turtles that tended to remain immobile had a higher probability of first-year survivorship than did individuals that moved readily. Thus, the effects of the gender × incubation temperature interaction on survivorship of hatchling turtles observed in the field experiment may have been mediated by temperature-dependent antipredator behavior. These results provide a possible functional explanation for the evolutionary significance of TSD in turtles that is consistent with predictions of theoretical models.  相似文献

Freshly-laid eggs of the Chinese three-keeled pond turtle (Chinemys reevesii) from captive cohorts in Hunan, Shanghai and Jiangxi were incubated at four constant temperatures of 24, 26, 28 and 30 °C to assess the effects of incubation temperature and cohort origin on incubation duration and hatchling phenotypes. Eggs from the three cohorts differed in size and shape. Egg mass and width were greatest in the Hunan cohort, smallest in the Jiangxi cohort, with the Shanghai cohort in between. Incubation duration decreased with increasing temperature and differed among the cohorts, with longer incubation duration for eggs from the Jiangxi cohort than those from the Hunan or Shanghai cohorts. Incubation temperatures significantly affected hatchling size and hatchlings from 30 °C were smaller than those from the lower temperatures in terms of carapace size and body mass. When incubated at a common temperature, hatchlings from the Hunan and Shanghai cohorts were larger than those from the Jiangxi cohort. The swimming capacity of hatchlings was affected by incubation temperature, but did not differ among the cohorts. The characteristics of eggs and hatchlings were similar among the Hunan and Shanghai cohorts, but they differed significantly from the Jiangxi cohort.  相似文献

Fluctuations in incubation temperature affect incubation duration but not morphology, locomotion and growth of hatchlings in the sand lizard Lacerta agilis (Lacertidae). —Acta Zoologica (Stockholm) 00 : 1–8. Studies looking for potential effects of temperature and temperature fluctuations on phenotypic traits of reptile hatchlings have shown species variation, but have not always allowed a distinction between effects of fluctuation per se and temperature extremes themselves. To examine whether incubation temperature fluctuation has a key role in influencing the phenotype of offspring, we incubated eggs of the sand lizard Lacerta agilis at one of the four temperature regimes (27, 27 ± 2, 27 ± 4 and 27 ± 6 °C). We found that: (1) hatchlings incubated under the four temperature regimes did not differ from each other in any of the morphological and physiological traits examined; (2) interactions that included temperature treatment did not affect any trait examined; (3) the mean incubation length was longer in the 27 ± 6 °C treatment than in the other three treatments; and (4) female hatchlings were shorter in head length and width but longer in snout‐vent length as well as abdomen length than males derived from the same‐sized egg. Our data show that both the type and the magnitude of temperature variation can affect incubation length. We found no evidence for phenotypic divergence in responses to temperature fluctuations during incubation and therefore suggest that temperature variation does not affect the phenotype of hatchlings in L. agilis.  相似文献

The pigmentation pattern of Alligator mississippiensis was examined. The number of white stripes on the dorsal side of embryos (stages 21-28) and hatchlings from eggs incubated at 30 degrees C (100% females) and 33 degrees C (100% males) was recorded. Total length, nape-rump length and tail length were recorded for each embryo and hatchling. The number of white stripes was affected by incubation temperature but not sex; hatchlings incubated at 33 degrees C had two more white stripes than those at 30 degrees C, despite being the same length. Five female hatchlings produced at 33 degrees C by manipulation of the temperature, had the same number of stripes as males that developed under the same incubation temperatures. The appearance of the pigmentation was accelerated in embryos incubated at 33 degrees C, occurring eight days earlier than at 30 degrees C. At the time just before the first signs of pigment deposition, embryos from 33 degrees C were longer than those at 30 degrees C. If the stripe formation is size dependent this explains why hatchlings at 33 degrees C have more stripes than hatchlings from 30 degrees C. The mechanism that produces the stripe patterns is unknown. We describe key elements a pattern formation mechanism must possess to produce such stripes and suggest a possible mechanism, based on cell movement driven by chemotaxis. We apply the mathematical model to dorsal patterning on A. mississippiensis. We show how length at pattern formation is the prime factor in determining stripe number and how the pattern can be formed in the observed anterior-posterior sequence. We present numerical simulations and show that the qualitative behaviour is consistent with the experimental results.  相似文献

In order to determine the temperature sensitive stages for sexual differentiation of the gonads in Emys orbicularis, eggs of this turtle were shifted at different stages of embryonic development from the male-producing temperature of 25°C to the female-producing temperature of 30°C and reciprocally. Based on the series of developmental stages described by Yntema (′68) for Chelydra serpentina, temperature begins to influence sexual differentiation of Emys orbicularis at stage 16, a stage in which the gonads are still histologically undifferentiated. Its action lasts over the first steps of histological differentiation of the gonads. The minimal exposure at 25°C required for male differentiation of all individuals extends from stage 16 to somewhat before stage 21. For 100% female differentiation, incubation at 30°C must be longer, from stage 16 to somewhat before stage 22. Shorter exposures at 25°C or 30°C during these periods result in different percentages of males, females, and intersexes. Our results show that there is a critical stage (stage 16) which is the same for both male and female differentiation of the gonads. The thermosensensitive periods are rather long, corresponding to 11–12 days at 25°C and 30°C.  相似文献

Cellular infiltrations forming lymphoid-like aggregates were previously observed in gonads of two turtle species exhibiting temperature-dependent sex determination (TSD): at hatching in Chelydra serpentina; at and after hatching in Emys orbicularis. We show here that such aggregates are also present in gonads of Testudo graeca by the end of embryonic development, suggesting that their occurrence is general in turtles. Since in C. serpentina, infiltrations were observed mainly in testes exhibiting remnants of the germinal epithelium, it was assumed that their occurrence was an expression of maleness leading to rejection of this epithelium. The generality of this hypothesis was tested in E. orbicularis by looking for lymphoid-like aggregates in three types of gonads (testes, ovotestes, and ovaries) and for the stages at which they occur. Gonads were from embryos, hatchlings, and young incubated at various temperatures. Ovotestes obtained by treatment with an aromatase inhibitor of eggs incubated at female-producing temperature were also examined. In these gonads, the differentiation of Sertoli cells in testicular cords/tubes was ascertained by expression of SOX9. Moreover, the cell composition of aggregates was determined on electron micrographs. Aggregates appear in ovaries and ovotestes by the end of embryonic development and are present in the majority of these gonads at hatching, and at least up to one year after hatching. They are composed mainly of lymphocytes and fibroblasts. Aggregates are not present in typical testes. Since they occur in most ovaries, they cannot be seen as an expression of maleness. Rather, lymphocytic infiltration and formation of lymphoid aggregates in turtle gonads can be seen as components of the immune system, and can be under the control of gonadal endogenous sex steroids.  相似文献

Temperature is one of the abiotic environmental factors most strongly affecting animal behaviour, physiology, and life history. In insects, lower temperatures generally slow down most physiological processes, reducing growth rate and prolonging the juvenile period. Here, we investigate temperature‐dependent ovariole and testis maturation in the anautogenous yellow dung fly, Scathophaga stercoraria L. (Diptera: Scathophagidae), and relate it to corresponding temperature effects on pre‐adult development time and the adult pre‐reproductive period. Flies were reared in the laboratory at three constant temperatures (18, 22, and 26 °C), and the size of the developing ovarioles and testes (reflecting sperm production) was measured over time (i.e., age). Ovariole size increased asymptotically over the first 12 days of adult life, while the testes continued to fill after day 10. In accordance with the temperature‐size rule, warmer temperatures resulted in smaller ovarioles (eggs) and smaller testes, independent of body size. Warmer temperatures also greatly reduced pre‐adult development time by more than half, from 12 to 25 °C, the larger males always taking 1–3 days longer than the females. Corresponding temperature effects on the adult pre‐reproductive period were small (<1 day between 15 and 25 °C), with males taking 5–6 days and females 10–13 days to first reproduction. Time lost by males during the pre‐adult stage, when ovaries and testes are produced, can thus be more than compensated‐for by time gained during the pre‐reproductive period, when eggs and sperm are produced, so males can nevertheless start reproducing sooner than females.  相似文献

The reproductive cycle of anglerfish was studied by examining fresh material obtained from commercial landings and research vessel surveys. The general morphology and histology of the gonads are described. This species has unusual gonads. The testes are tubular, and beanshaped in transverse section. The maturation process of the testis is similar to that of most other teleosts. The ovaries are confluent and form a single, flattened tube. During maturation, the width and, in particular, the length of the ovaries increases dramatically. A gelatinous matrix is produced during the maturation process, which contributes to the enormous size and weight of ripe ovaries. Ripe testes can be found throughout the year, whereas ripened ovaries were found only between November and May. Females outnumbered males in length classes greater than 70 cm and no males larger than 90 cm were found. The estimated mean length at maturity (150%) was 73 cm for females and 49 cm for males. Because only the large female anglerfish are capable of spawning, and because virtually all large anglerfish are female, trends in their abundance resulting from commercial exploitation should be monitored with care.  相似文献

LH/hCG receptors were disrupted by gene targeting in embryonic stem cells. The disruption resulted in infertility in both sexes. The gonads contained no receptor mRNA or receptor protein. Serum LH levels were greatly elevated, and FSH levels were moderately elevated in both sexes; estradiol and progesterone levels decreased but were not totally suppressed in females; testosterone levels were dramatically decreased and estradiol levels moderately elevated in males. The external and internal genitalia were grossly underdeveloped in both sexes. Abnormalities included ambiguous vaginal opening, abdominal testes, micropenis, dramatically decreased weights of the gonads and reproductive tract, arrested follicular growth beyond antral stage, disarray of seminiferous tubules, diminished number and hypotrophy of Leydig cells, and spermatogenic arrest beyond the round spermatid stage. LH/hCG receptor gene disruption had no effect on FSH receptor mRNA levels in ovaries and testes, progesterone receptor (PR) levels in ovaries and androgen receptor (AR) levels in testes. However, it caused a dramatic decrease in StAR and estrogen receptor-alpha (ERalpha) mRNA levels and an increase in ERbeta mRNA levels in both ovaries and testes. Estradiol and progesterone replacement therapy in females and testosterone replacement in males, to determine whether phenotype and biochemical changes were a consequence of decreased gonadal steroid levels or due to a loss of LH signaling, revealed complete restoration of some and partial restoration of others. Nevertheless, the animals remained infertile. It is anticipated that the LH receptor knockout animals will increase our current understanding of gonadal and nongonadal actions of LH and hCG.  相似文献

Summary A method for superficial pinealectomy of the adult white-footed mouse,Peromyscus leucopus, is presented. Histological examination of the brain of pinealectomized mice showed that the deep pineal gland was left intact. The survival rate of pinealectomized mice was 80%. Pinealectomized mice were exposed to a short day photoperiod (8L:16D) at 15°C for 7 weeks. After this time male mice maintained active gonads with a testicular index (TI, testis width×length/body weight) of 2.0±0.1. Testis weight was 202±35 mg, and the seminiferous tubules contained abundant spermatozoa (spermatogenic index [SI]=4.5±0.2). Sham operated animals had regressed testes. TI was 1.2±0.2, testis weight was 97±26 mg, and the SI was 2.7±0.7 (allP<0.05 relative to pinealectomized mice). Pinealectomized females were reproductively competent in that all of the mice had a perforate vagina, the reproductive tract weight (vagina, uterus, oviducts, and ovaries) was 111±15 mg, and the ovaries from each animal contained preovulatory follicles. Sham operated mice had an imperforate vagina, reproductive tract weights were 34±5 mg, and in only 1 out of 5 mice did the ovaries contain a preovulatory follicle (allP<0.05). The weight of the lipid-free interscapular brown fat was 28% less in pinealectomized mice relative to sham operated animals (P<0.01). These results support the role of the pineal gland as regulator of short day, cold induced reproductive regression and brown fat hypertrophy.  相似文献

Morphology of hermaphroditic gonads is studied in the individuals of walleye pollock Theragra chalcogramma collected in different regions of the seas of the North Pacific from 1987 to 2014. Based on the external morphological characteristics of sexual glands of 17 fishes, the most frequent types of the structure of bisexual gonads are described. Histological analysis shows the absence of differences in the composition and condition of oocytes in the left and right-paired ovaries, and the testes and ovaries of each individual are at the same maturity stage. In the ovigerous lamellae of ovaries in the areas adjacent to testes the zones of oocyte resorption are observed. These zones are located exclusively near the joining of the female and male parts of the gonads. The development of ovaries and testes of bisexual gonads is similar to that in the majority of normally maturing males and females collected during the same period. The occurrence of hermaphrodites is 0.0003–0.0016% from the total number of investigated exemplars.  相似文献

The present study was conducted to characterize the proximate composition and fatty/amino acid profile in muscle, liver and gonads obtained from farmed male and female Chinese sturgeon (Acipenser sinensis) with gonad development at stage II, and to discern the key nutrients for gonads by comparing the nutrients between tissues and between fish at different gonad stages. Chinese sturgeon were reared in an outdoor circular cement pool at temperatures ranging from 8°C to 26°C, and fed with 70% artificial diets and 30% frozen forage fish at a feeding rate of 0.2%–1.5% body weight according to the water temperature. Three male and three female A. sinensis were sampled for analyses. Results showed that the lipid content in gonads was significantly higher than in other tissues in both males and females (< .05). The protein content in male and female gonads was significantly lower than that in the muscle (< .05). Total content of mono‐unsaturated fatty acids (MUFA) in females was significantly higher than in males (< .05), while the total content of n‐6 PUFA in females was significantly lower than in males (< .05). Palmitic acid (C16:0) and oleic acid (C18:1n9) were the major saturated fatty acid (SFA) and MUFA, respectively, in both females and males. The contents of C18:2n6, C20:4n6 (ARA), C18:3n3 and C22:6n3 (DHA) in male gonads were significantly higher than in females (< .05). The contents of ARA, EPA and DHA in gonads were lower, possibly playing a pivotal role in gonad development. Comparing the nutrients of ovaries in broodstocks at stages III and IV and the eggs at stage IV, the present study suggests that the lipid and fatty acid profile in fish is vital to the gonad development and that proper improvement of lipid nutrition in the broodstock diet to provide sufficient energy and essential fatty acids would be beneficial for the gonad development of Acipenser sinensis.  相似文献
