Anti-lipopolysaccharide factors (ALFs) are antimicrobial peptides with binding and neutralizing activities to lipopolysaccharide (LPS) in crustaceans. This study identified and characterized a novel ALF homolog (SpALF4) from the mud crab Scylla paramamosain. The complete cDNA of SpALF4 had 756 bp with a 381 bp open reading frame encoding a protein with 126 aa. The deduced protein contained a signal peptide and a LPS-binding domain. SpALF4 shared the highest identity with PtALF5 at amino acid level but exhibited low similarity with most of other crustacean ALFs. Furthermore, different from the previously identified three SpALF homologs and most of other ALFs, SpALF4 had a low isoelectric point (pI) for the mature peptide and the LPS-binding domain with the values of 6.93 and 6.74, respectively. These results indicate that SpALF4 may be a unique ALF homolog with special biological function in the mud crab. Similar to the spatial structure of ALFPm3, SpALF4 contains three α-helices packed against a four-strand β-sheet, and an amphipathic loop formed by a disulphide bond between two conserved cysteine residues in LPS-binding domain. SpALF4, mainly distributed in hemocytes, could be upregulated by Vibrio harveyi, Staphylococcus aureus, or white spot syndrome virus. Recombinant SpALF4 could inhibit the growth of Gram-negative bacteria (V. harveyi, Vibrio anguillarum, Vibrio alginolyticus, Aeromonas hydrophila, Pseudomonas putida), Gram-positive bacteria (S. aureus and Bacillus megaterium), and a fungus Candida albicans to varying degrees. Further study showed that it could also bind to all the aforementioned microorganisms except S. aureus. These results demonstrate that SpALF4 is a unique ALF homolog with potent antimicrobial activity against bacteria and fungi. This characteristic suggests SpALF4 plays an essential function in immune defense against pathogen invasion in mud crab.  相似文献

The circadian activity rhythm of the common marmoset, Callithrix j. jacchus was investigated by long-term recording of the locomotor activity of 15 individuals (5 males, 10 females) from 1.5 to 8 years old, both under constant illumination and under LD 12:12. The mean period of the spontaneous circadian rhythm was 23.2 ± 0.3 h. Neither sex-specific differences nor a systematic influence of light intensity on the spontaneous period were observed, but the period was dependent on the duration of the trial and on the age of the individual. Due to the short spontaneous period, in LD 12:12 there was a distinct advance of the activity phase with respect to the light time and a masking of the true onset of activity by the inhibitory direct effect of low light intensity during the dark time. After an 8 h delay shift of the LD 12:12, re-entrainment of the circadian activity rhythm required an average of 6.8 ± 0.7 days; the average re-entrainment time after an 8 h phase advance of the LD cycle was 8.6 ± 1.3 day. This directional effect is ascribed to characteristics of the phase-response curve. No ultradian components were observed, either in the LD-entrained or the free-running circadian activity rhythm.  相似文献

Activity period plays a central role in studies of primate origins and adaptations, yet fundamental questions remain concerning the evolutionary history of primate activity period. Lemurs are of particular interest because they display marked variation in activity period, with some species exhibiting completely nocturnal or diurnal lifestyles, and others distributing activity throughout the 24-h cycle (i.e., cathemerality). Some lines of evidence suggest that cathemerality in lemurs is a recent and transient evolutionary state (i.e., the evolutionary disequilibrium hypothesis), while other studies indicate that cathemerality is a stable evolutionary strategy with a more ancient history. Debate also surrounds activity period in early primate evolution, with some recent studies casting doubt on the traditional hypothesis that basal primates were nocturnal. Here, we used Bayesian phylogenetic methods to reconstruct activity period at key points in primate evolution. Counter to the evolutionary disequilibrium hypothesis, the most recent common ancestor of Eulemur was reconstructed as cathemeral at ~9-13 million years ago, indicating that cathemerality in lemurs is a stable evolutionary strategy. We found strong evidence favoring a nocturnal ancestor for all primates, strepsirrhines and lemurs, which adds to previous findings based on parsimony by providing quantitative support for these reconstructions. Reconstructions for the haplorrhine ancestor were more equivocal, but diurnality was favored for simian primates. We discuss the implications of our models for the evolutionary disequilibrium hypothesis, and we identify avenues for future research that would provide new insights into the evolution of cathemerality in lemurs.  相似文献

Many plants that bear extrafloral nectaries (EFNs) attract various ant species that can exclude herbivores. The aggressiveness of the attracted ants and their temporal activity patterns are important factors that can affect the efficiency of herbivore exclusion from the plant. However, the characteristics of this mutualistic relationship between EFN‐bearing plants and ants have not been sufficiently elucidated. We investigated the aggressiveness of six ant species against the common armyworm, Spodoptera litura Fabr., and temporal fluctuations in the abundance of four aggressive ant species on an EFN‐bearing plant, Mallotus japonicus (L.f.) Müll. Arg. Workers of Crematogaster teranishii Santschi, Pheidole noda Smith, Pristomyrmex punctatus Smith and Formica japonica Motschoulsky were observed to be highly aggressive. In contrast, workers of Camponotus vitiosus Smith showed low aggressiveness. Paratrechina flavipes Smith workers did not attack the herbivore. The activity patterns of the four aggressive ant species greatly differed. Crematogaster teranishii and Ph. noda workers were constantly active throughout the day and night. In contrast, F. japonica was diurnal. Pristomyrmex punctatus was principally nocturnal. Formica japonica workers foraged solitarily, whereas workers of the other three species foraged in a group or recruited nestmates. Our results suggest that the efficacy of the indirect defense in M. japonicus depends principally on the attracted ant species.  相似文献

【背景】谷氨酰胺转氨酶是一种能够催化酰基转移反应的酶,催化各种蛋白质分子之间或之内发生交联反应,在食品、化妆品、医药等领域具有重要的潜在价值。【目的】克隆来自拉达克轮丝菌(Streptoverticillium ladakanum) B1的谷氨酰胺转氨酶(TGase)基因并对其进行分子改造,使其在大肠杆菌中获得高效异源表达。【方法】分别克隆来自拉达克轮丝菌谷氨酰胺转氨酶的自身前导肽(pro)和除前导肽以外的成熟谷氨酰胺转氨酶(TGase)基因,以pET-22b为表达载体构建pro、TGase共表达和融合表达两种表达模式,在这两种表达模式的基础上进一步用定点突变的方法对成熟TGaseN端前4个氨基酸进行改造,检测不同表达模式以及突变对酶活的影响。【结果】当采用前导肽与TGase共表达时,可以直接得到活性形式的TGase,比酶活达到37.71 U/mg。在融合表达的基础上,将TGaseN端前3个氨基酸DSD突变为AAA,比酶活达到14.04U/mg,相较于原始表达模式提高了14.05%。【结论】前导肽与TGase共表达可以直接产生活性TGase,对于融合表达模式,合适位点的突变有利于提高TGase酶活。  相似文献

Macrovibrissae are specialized tactile sensory hairs present in most mammalian orders, used in maxillary mechanoreception or “face touch.” Some mammals have highly organized vibrissae and are able to “whisk” them. Movement of vibrissae is influenced by intrinsic vibrissa musculature, striated muscle bands that attach directly to the vibrissa capsule. It is unclear if primates have organized vibrissae or intrinsic vibrissa musculature and it is uncertain if they can move their vibrissae. The present study used histomorphological techniques to compare vibrissae among 19 primates and seven non‐primate mammalian taxa. Upper lips of these mammals were sectioned and processed for histochemical analysis. While controlling for phylogenetic effects the following hypotheses were tested: 1) mammals with well‐organized vibrissae possess intrinsic vibrissa musculature and 2) intrinsic vibrissa musculature is best developed in nocturnal, arboreal taxa. Our qualitative analyses show that only arboreal, nocturnal prosimians possess intrinsic musculature. Not all taxa that possessed organized vibrissae had intrinsic vibrissa musculature. Phylogenetic comparative analyses revealed a 70% probability that stem mammals, primates, and haplorhines possessed intrinsic vibrissa musculature and well‐organized vibrissae. These two traits most likely coevolved according to a discrete phylogenetic analysis. These results indicate that nocturnal, arboreal primates have the potential to more actively use their vibrissae in spatial recognition and navigation tasks than diurnal, more terrestrial species, but there is a clear phylogenetic signal involved in the evolution of primate vibrissae and “face touch.” Am J Phys Anthropol, 2013. © 2012 Wiley Periodicals, Inc.  相似文献

Recent fossil discoveries have demonstrated that Africa and Asia were epicentres for the origin and/or early diversification of the major living primate lineages, including both anthropoids (monkeys, apes and humans) and crown strepsirhine primates (lemurs, lorises and galagos). Competing hypotheses favouring either an African or Asian origin for anthropoids rank among the most hotly contested issues in paleoprimatology. The Afrocentric model for anthropoid origins rests heavily on the >45 Myr old fossil Algeripithecus minutus from Algeria, which is widely acknowledged to be one of the oldest known anthropoids. However, the phylogenetic position of Algeripithecus with respect to other primates has been tenuous because of the highly fragmentary fossils that have documented this primate until now. Recently recovered and more nearly complete fossils of Algeripithecus and contemporaneous relatives reveal that they are not anthropoids. New data support the idea that Algeripithecus and its sister genus Azibius are the earliest offshoots of an Afro–Arabian strepsirhine clade that embraces extant toothcombed primates and their fossil relatives. Azibius exhibits anatomical evidence for nocturnality. Algeripithecus has a long, thin and forwardly inclined lower canine alveolus, a feature that is entirely compatible with the long and procumbent lower canine included in the toothcomb of crown strepsirhines. These results strengthen an ancient African origin for crown strepsirhines and, in turn, strongly challenge the role of Africa as the ancestral homeland for anthropoids.  相似文献

Cathemeral animals distribute their activity more or less uniformly between the dark and light phases of the 24-hour day. This activity pattern is quite widely observed among major mammal taxa, although the number of species involved is relatively low. Among primates, cathemerality is seen principally, though not uniquely, in the strepsirhine (“lower primate”) family Lemuridae; it is ubiquitous in the genus Eulemur, among species of which it seems to be a stable activity pattern with a history going back at least 8 - 12 million years. Nocturnal primates have visual systems that accentuate sensitivity to light over visual acuity, showing large cornea size, high rod:cone ratios and large numbers of photoreceptors relative to ganglion cells. Diurnal forms do the reverse, while Eulemur species are intermediate in these respects, suggesting substantial accommodation of the visual system to the cathemeral lifestyle. Studies of one Eulemur species have revealed that its cathemerality is underpinned by a robust endogenous circadian timing mechanism with a free-running periodicity of over 25 hours. The lemurs appear to be entrained to the 24-hour day by the light/dark cycle, plausibly with dusk as the primary zeitgeber. The causes of cathemerality have been sought in such environmental influences as predator avoidance, energy conservation, dietary quality, and interspecies feeding competition. All of these factors seem to be relevant to the cathemeral lifestyle, but none appears to be a crucial determinant. Nothing is known of the underlying neural mechanisms that facilitate the lemurs' accommodation to the extremes to which 24-hour activity exposes them.  相似文献

The abundance, seasonal activity patterns and development rates of the tick Ixodes ricinus (L.) (Acari: Ixodidae), as well as microclimate features of the site of study, are described for a 9-year-long study (1994-2002) in north-central Spain. According to drag captures, larvae had a unimodal activity pattern, with a maximum observed around July-August, whereas nymphs displayed a bimodal pattern (May-June and August-September) with strong dominance of spring activity. An inversion of this pattern, with larger autumn peak, was observed in years with humid summers. Adults showed a small spring peak and a large autumn one. In the later years of the study, a small increase in the adult spring peak of activity was noticed, correlated with mild winters. Over the entire period of study, a clear increase in the total tick abundance was detected. Statistically significant differences between years were observed for some climate variables (saturation deficit, winter temperatures and number of days with temperatures above 6 degrees C), but a consistent and constant pattern of change was not observed in any climate variable. Temperature requirements for developing stages showed a sharp decrease in weeks 35-51 (eggs) and 38-50 (larvae and nymphs), a feature attributed to the presence of the morphogenetic diapause, beginning around September. Development rates obtained under quasi-natural conditions were almost twice those reported for other sites, suggesting an adaptation of this local, largely isolated I. ricinus population. According to drag captures and field-obtained development rates, interchange of nymphs between the two cohorts is common in this site, and seems to be influenced by the winter temperature and the date of larval engorgement.  相似文献

Multiple sclerosis (MS) is a demyelinating disease resulting in impairments in motor and mental performance and restrictions in activities. Self-report instruments are commonly used to measure activity patterns; alternatively, actigraphs can be placed on several parts of the body. The aims of this study were to evaluate the superiority and specificity of actigraph placement (wrist vs. ankle) in subjects with MS and healthy controls and explore the relationship between self-report and objective activity patterns. A total of 19 subjects with definite MS and 10 healthy volunteers wore actigraphs on the non-dominant wrist and ankle for three days while they kept a log to register performed activities every .5 h. Wrist and ankle actigraphs produced similar activity patterns during the most active hours (09:00-20:30 h) (ANOVA, time×location interaction: F=.901, df=23, p=.597) in individuals with MS and healthy controls (between subjects factor F=3.275, p=.083). Wrist placement of the actigraphs was better tolerated than ankle placement. Wrist actigraph data corresponded to a higher degree with self-reported activities of the upper limbs in the early afternoon, whereas ankle data seem to reflect better whole body movements in the later afternoon/early evening. Overall, actigraph data correlated moderately with self-reported activity (r=.57 for ankle and r=.59 for wrist). The regression model revealed that self-reported activities explained 44% of the variance in ankle and 50% of wrist data. Wrist and ankle actigraphs produce similar activity patterns in subjects with MS and in healthy controls; however, the placement of actigraphs on the wrist is better tolerated. Ankle actigraphs reflect general movement but underestimate upper body activity. Subjective registration of activity level partly matches with objective actigraph measurement. A combination of both objective and subjective activity registration is recommended to evaluate the physical activity pattern of subjects with MS.  相似文献

The distribution of tropical plant and animal diversity is still poorly documented, especially at spatial resolutions of practical use for conservation. In the present study, we evaluated the level to which geographical incomplete data availability of species occurrence affects the perception of biodiversity patterns (species richness and endemism) among pteridophytes in Bolivia. We used a data base of Bolivian pteridophytes (27,501 records), divided it into three time periods (1900–70, up to 1990 and up to 2006), and created grid-files at 15'-resolution for species richness and endemism. For each of these biodiversity properties we estimated the species richness (Chao 2) and the index of sampling completeness (C index) per grid, and then all these variables at both species richness and endemism were correlated. Patterns of richness were fairly consistent along all periods; the richest areas were placed along the humid-montane forest, even though they were strongly influenced by collecting intensity. Endemism had a lower degree of correlation with collecting intensity, but varied much more strongly through time than species richness. According to the C index, which gives the ratio between estimated (by Chao 2) and recorded values of species richness and endemism, both biodiversity properties tended to be undersampled in the richest grid cells. Inter-temporal correlations showed sharper differences of correlations for endemism than species richness. Consequently, already in 1970, botanists had a correct idea of the spatial distribution of pteridophyte richness in Bolivia (even though the magnitude was grossly underestimated). In contrast, patterns of endemism, which are of high conservation importance, may not even today be reliably known.  相似文献

在大鼠慢肌比目鱼肌(SOL)肌纤维的肌-腱接头(MTJ)上有较高的乙酰胆碱(ACh)敏感牲,而快肌伸趾长肌(EDL)的 MTJ却没有。SOL 肌纤维受 EDL神经交叉支配后,其 MTJ的 ACh敏感性消失,此点 Miledi等已有报道。本文首先验证了与此相对称的结果,即EDL 肌纤维受 SOL神经交叉支配后,其 MTJ获得与正常 SOL肌纤维 MTJ相似的 ACh敏感性,从而进一步肯定了MTJ 的ACh 敏感性的出现是由特殊神经支配决定的。本文的主要结果是:兼受 SOL神经和 EDL神经支配的EDL和SOL的纤维,其MTJ都没有AGh 敏感性。这一结果的兴趣,不但在于它显示当两种神经支配同时存在时,快肌神经的影响压倒慢肌神经,而且还在于此结果与以往用其他指标进行的双神经支配肌纤维实验的结果形成鲜明的对照:用 M-ATPase 组织化学染色和Z带宽度等变化为指标,在双神经支配肌纤维中,慢肌神经的影响总压倒快肌神经。我们也观察了长期电刺激对MTJ ACh敏感性的影响。SOL经“慢”型刺激2—3周后,其 MTJ的 ACh敏感性虽有降低,但不及“快”型刺激显著。综合各种有关的观察,本文对双神经支配肌纤维的 MTJ没有 ACh敏感性这一主要结果的解释进行了讨论。  相似文献

Spikes arriving at the synaptic connection produce short-term plastic changes of the synaptic efficacy. Model experiments have shown that paired-pulse facilitation attaining its maximum after a specific interval between a pair of arriving spikes might turn a “weak” plastic synapse attached to an integrate-and-fire neuron to a frequency-tuned device. Resulting computational capabilities create biologically plausible mechanisms of information processing relating to: (i) real-time identification of temporal patterns in a stream of random spiking activity (a recognition problem); and (ii) codetermination of the specific activity routing among neurons (an addressing problem) resulting in definite spatio-temporal patterns of the output activity (an input-output pattern problem).  相似文献

根据相机陷阱调查获得照片的时间,研究了2008年12月~2009年5月唐家河自然保护区内野生扭角羚Budorcastaxicolor的日活动模式。通过对24个1km×1km样方的调查,共完成686个相机工作日的取样量,获得有效的扭角羚捕捉次数709次(冬季322次,春季387次),并分别计算出扭角羚冬季和春季在各时间段的相对活动强度指数RAI。结果显示,扭角羚在冬季和春季的日活动模式没有显著差异(χ2检验,P>0.1),每天都有3个活跃时期(凌晨、早上和下午)和3个紧随的不活跃时期。其中,冬季日活动的最高峰出现在下午17:00~18:00(RAI=12.42),最低谷出现在日出前3:00~6:00(RAI=0.00);春季最高峰出现在早上6:00~7:00(RAI=20.16),最低谷出现在日出前2:00~5:00(RAI=0.26)。在春季,扭角羚在上午活跃期的活动强度明显高于下午活跃期,而在冬季则相反。春季与冬季相比,扭角羚上午和下午的两个活跃时间段都有提前,可能是由于不同季节气温和光照差异的影响。研究表明,相机陷阱调查技术是研究野生动物在野外活动模式和活动节律的有效工具,可为今后深入开展扭角羚行为生态学...  相似文献
