The fish gill is a multifunctional organ responsible for gas exchange and ionic regulation. It is hypothesized that both morphological and functional differentiation can be found in the gills of the aquatic air-breathing fish, Trichogaster leeri. To test this, we used the air-breathing fish, Trichogaster leeri, to investigate various morphological/functional parameters. First, we evaluated the importance of performing the aquatic surface respiration behavior in T. leeri. A reduced survival rate was observed when fish were kept in the restrained cages in hypoxic conditions. On the gross anatomy of gills, we found evidence of both morphological and functional modification in the first and the second gills and are responsible for ionic regulation. There were large-bore arterioarterial shunts in the fourth gill arch. It is specialized for the transport of oxygenated blood and is less responsive to environmental stress. In addition, the anterior and the posterior gills differed in the Na+, K+-ATPase activity upon ionic stresses. That is, only the Na+, K+-ATPase activity of the anterior two gills was up-regulated significantly in the deionized water. Lastly, we found that the number of mitochondria-rich cells in the first and the second gills increased following ionic stress and no difference was found in the third and the fourth gills following such an exposure. These results supported the hypothesis that there are morphological and functional differences between anterior and posterior gill arches within the air-breathing Trichogaster leeri. In contrast, no significant difference was found among gills in gross anatomy, filament density and Na+, K+-ATPase activity in the non-air-breather, Barbodes schwanenfeldi.  相似文献

Summary Autometallographic (AMG) silver enhancement of endogenous zinc was studied in seven organs of the rainbow trout Salmo gairdneri. Groups of trout were injected intraperitoneally with sodium selenite in doses ranging from 0.08 to 25 ppm, administered 1 h before being killed. The concentration of selenium obtained by each organ was determined by gamma-spectrometry, and compared with the autometallographic deposition of silver grains. The relative accumulation of selenium in the organs was: liver > spleen > kidney > intestine > gills > brain > muscle. In the fish labelled with 10 and 25 ppm Se, AMG-deposits were found (1) within lysosomes of liver cells, (2) within the granules and on the nuclear membrane of melanophores in the spleen, (3) on the microvilli and in the apical cytoplasm of renal proximal tubular cells, (4) within the granules and along the plasma membrane of intestinal eosinophilic granule cells, and in the apical portion of the intestinal epithelium, and (5) in the gills, within granule cells and on the surface of the ionocytes. In the trouts injected with 5 ppm Se, silver grains were still observed in the liver, the intestine, and the gills, whereas, no such grains were found in preparations from fish having received 1 ppm Se. The use of selenium for the histochemical demonstration of endogenous zinc versus exogenous metals is discussed. Also, consideration is given to the question of which part of the total tissue zinc that is histochemically reactive.  相似文献

Autoradiography at different levels of resolution was used to study the disposition of the polycyclic aromatic hydrocarbon benzo(a)pyrene (3H-BaP) in juvenile and sexually mature polar cod (Boreogadus saida). Exposure took place via the water or after intragastric administration at subzero temperatures. In water-exposed fish, high total tissue levels were found in the gills, olfactory organ, anterior kidney, liver, skin and intestinal wall. Only traces of radioactivity were present in the muscle, brain and gonads. No major differences in tissue levels or in general distribution pattern between males, females or juvenile fish were observed. The gills appeared to be the absorption site for exposure via water. After oral administration, tissue levels of 3H-BaP-derived radioactivity were negligible. Following both administration routes, levels of radioactivity were highest in the bile and intestinal contents while only traces were observed in the urine, indicating biliary excretion as the major excretory pathway in this aglomerular species. Tape-section autoradiography of fish exposed via water revealed tissue-bound residues of 3H-BaP in the olfactory organs, gills, kidney, liver, skin and intestinal mucosa. Light-microscopy autoradiography demonstrated that the bound residues in the olfactory organ, gills and anterior kidney were localized in epithelial cells, while those in liver and intestinal mucosa were evenly distributed. In conclusion, the present study shows that BaP is absorbed from the water via the gills at subzero temperatures, that tissue levels are considerably higher after water exposure than after dietary exposure, that biliary excretion is predominant and, finally, that site-specific tissue binding in the olfactory organs, gills and anterior kidney is confined to epithelial cells. A population of Gyrodactylus unicopula Glukhova, 1955, is described from the gill filaments of young plaice from a nursery ground on the north-west coast of Scotland. The seasonal incidence of this parasite was followed by sampling the 1965 year class of plaice at intervals of from one to three months from April 1965 to April 1967. The level of intensity is usually low and shows no progressive increase, although the fish are shown to be capable of supporting much higher populations of the parasite, which suggests the existence of some mechanism controlling the level of intensity in the natural population. The distribution of G. unicopula on the filamentous surface of the gills is shown in detail. χ2 analyses indicated no significant association or antagonism between this parasite and two ciliates commonly found on the gills of the same population of plaice. Gyrodactyliasis, the disease caused by parasites of the genus Gyrodactylus, is discussed with special reference to marine fish farming.  相似文献

Histological examination of gills of the rock crab (Cancer irroratus Say, 1917) taken from specimens collected in polluted and nonpolluted areas indicated that gill discoloration was not directly related to the degree of microbial fouling. Light microscope studies demonstrated that bacteria, peritrich and suctorian ciliates, and some naviculoid diatoms were the principal members of the fouling community. Examination of the bacteria by electron microscopy showed that several distinct types were present, and that some of these were attached to the gill by holdfast structures while others were enmeshed in a filamentous slime layer. An unusual small marine flagellate and an unidentified amoeba were also examined ultrastructurally.  相似文献

The liver and gills of five flounder species (Pleuronectes obscurus, P. punctatissimus, P. yokahamae, P. herzensteini, and Platichthys stellatus) from Sivuch'ya Bight were histologically studied. Specimens of all species were characterized by a wide spectrum of histomorphological changes. The proportions of most specific pathologies in P. obscurusfrom Sivuch'ya Bight were estimated. The pathologies comprised two major groups: those of the gills (edema and fusion of the respiratory lamellae; lifting and hyperplasia of the respiratory epithelium; vacuolization of the epithelium cells; and the presence of clavate lamellae, aneurysmal areas, and parasites) and those of the liver (vacuolization of the hepatocytes, aneurysms, necrosis of the parenchyma tissue, pathologies of the hepatocyte nuclei, pigment accumulation, and an increase in the number and size of melanomacrophage centers). The estimated proportions of pathological changes were compared with those observed in P. obscurusspecimens from the most contaminated area of Amurskii Bay. In both Sivuch'ya Bight and Amurskii Bay, the proportion of fish with various pathologies was high. The comparison of pathological changes in the gills and liver of P. obscurusand P. stellatusfrom Sivuch'ya Bight showed that, in terms of many parameters, the former species is more sensitive to pollution of the marine environment than the latter. Pathomorphological changes in the organs of the flounders studied are apparently caused by chronic pollutant action and suggest that the fish of both areas are in a damaged state.  相似文献

The current study was undertaken to enumerate Gram-positive bacteria in fresh sub-tropical marine fish and determine the effect of ambient storage (25°C) on the Gram-positive bacterial count. Total and Gram-positive bacteria were enumerated in the muscles, gills and gut of fresh and stored Pseudocaranx dentex, Pagrus auratus and Mugil cephalus on tryptone soya agar (TSA) and TSA with 0.25% phenylethyl alcohol (PEA), respectively. Initial studies indicated that PEA significantly reduced total aerobic bacterial count (TABC) whereas control Gram-positive bacteria were not affected by 0.25% PEA. TABC significantly increased in all fish body parts, whereas Gram-positive aerobic bacterial count (GABC) significantly increased only in the muscles and gills during ambient storage for 15 h. The TABC of the fish species increased from 4.00, 6.13 and 4.58 log cfu g−1, respectively in the muscles, gills, and gut to 6.31, 7.31 and 7.23 log cfu g−1 by the end of storage. GABC increased from 2.00, 3.52 and 2.20 log cfu g−1 to 4.70, 5.85 and 3.36 log cfu g−1. Within each species, TABC were significantly higher in the gills compared to that of muscles and gut; however, no significant differences were found in GABC between muscles and gills. This study demonstrated the potential importance of Gram-positive bacteria in sub-tropical marine fish and their spoilage.  相似文献

【目的】小瓜虫病是澳洲龙纹斑苗种阶段危害巨大的寄生虫病。探究小瓜虫在澳洲龙纹斑鳃器官上的分布及其影响可以丰富小瓜虫的致病性及病理学方面的研究内容,也可以为渔业生产中小瓜虫病的防治提供参考。【方法】采用光镜及扫描电镜技术确定病原,并观察小瓜虫在澳洲龙纹斑鳃丝、鳃小片及鳃盖上的分布情况及这些器官的变化情况。【结果】小瓜虫侵染澳洲龙纹斑的鳃器官后,分布在鳃丝、鳃小片及鳃盖的表面,上皮细胞之下及鳃小片之间,或是包裹在黏液细胞里,但侵染后期在鳃丝及鳃小片上的数量明显少于鳃盖。侵染后期,鳃丝、鳃小片及鳃盖出现一定程度的膨胀变形,黏液细胞分泌增多,鳃小片末端膨大变形甚至黏连融合,顶端充血呈球状或棒状。【结论】对于鳃部而言,侵染后期小瓜虫主要分布在澳洲龙纹斑的鳃盖上。小瓜虫主要通过侵染引发澳洲龙纹斑鳃部器官的变形膨大,使其丧失正常的功能,造成血液循环受阻、渗透压调节失衡,最终导致鱼体缺氧死亡。  相似文献

Three aquatic ecosystems in South Africa, the Hartbeespoort, Klipvoor and Bospoort Dams, are classified as hyper-eutrophic, because of high nutrient loads and chemical pollution. Water and two fish species, Clarias gariepinus and Cyprinus carpio, were collected from these dams to assess the impact of eutrophication and chemical pollutants on their health status. Water and muscle samples were analysed for organic and inorganic chemicals. Condition factor was determined and a necropsy performed to note any macroscopic abnormalities. A histology-based fish health assessment was done on the liver, kidney, gills and gonads. A number of fish from the three dams exhibited livers with fatty change and focal discoloration, skin lesions and parasites within the visceral cavity. The prevalence and severity of histopathology in the liver resulted in higher liver index values than the index values for kidneys and gills. Aluminium, silicon and chromium were detected in the water and muscle tissue. The DDT metabolite p,p’-DDE was present in both species, as well as in fish from the reference site, Marico-Bosveld Dam. Only C. gariepinus from Hartbeespoort Dam had p,p’-DDD levels higher than 5 µg g?1 per edible portion. Water from hyper-eutrophic dams adversely affects the health of freshwater fish.  相似文献

Summary

As is characteristic of freshwater Unionacea, the Australian species Hyridella depressa incubates its young within the demibranchs of its modified gills. The development of H. depressa was documented through light and scanning electron microscopic examination of the marsupial pouch in the inner demibranchs of brooding females. Transition from the gastrula to the glochidial stage was accompanied by a split in the larval integument. The glochidia of H. depressa have subtriangular shell valves each having a blunt tooth at the ventral margin. Each glochidium has a pair of hooks, one on each valve internal to the tooth. These hooks are used to attach to its fish host and are structured to interlock when the valves snap shut. One or two tufts of sensory hairs are located on the internal surface of the mantle dorsal to the hook, while a single tuft, encircled by a collar-like structure, is located centrally. The valves are pitted by pores and have concentric lines along the margins. Due to the similarity and phenotypic plasticity of adult Hyridella species, the morphology of their glochidia has potential for use as a taxonomic tool. In comparison to related species, the glochidia of H. depressa are medium-sized with a mean length of 243 μm and a mean height of 249 μm. The number of glochidia present in the gills was measured to determine the reproductive output of H. depressa. The embryos of an unknown mite, Unionicola sp., form cysts within the gill tissue of H. depressa and the adult mites were observed on the surface of the gills.  相似文献

Summary A fauna of small invertebrates in a perennial algal turf habitat showed marked seasonal fluctuations in abundance. These were characterised by a summer (December) peak in the abundance of gammarid amphipods and an autumn (April) peak in polychaete densities. Invertebrate abundance was lowest during the period February–March which coincided with an influx of very high densities of juveniles (0+ year class) of the sparid fish Chrysophrys auratus. Both juvenile C. auratus and also the mullid Upeneichthys porosus achieved their highest densities over coralline turf areas and fed on the associated invertebrates. Gammarid amphipods constituted the main food items of each species. All these observations suggested a key role of fish predators in determining the observed seasonal patterns of invertebrate abundance.This hypothesis was investigated by the use of replicated fish exclusion shields and cages which excluded both species from the algal turf and also permitted an assessment of Upeneichthys porosus feeding in the absence of Chrysophrys auratus. The experiment ran from November until June and covered the period of invertebrate and fish abundance changes. Little evidence of a fish predation effect on either the densities of or the timing of abundance peaks of the invertebrate fauna was detected. Sediment analysis of the cage and control experimental sites revealed no evidence of a cage effect on the microhabitat. It is suggested that seasonal changes in the abundance of invertebrates occurs independently of the high densities of predatory fish recorded in this habitat.  相似文献

1. Subtropical reservoirs of Australia are commonly subject to summer blooms of cyanobacteria. The potential for food web manipulation to control cyanobacterial blooms was investigated in Lake Maroon, south east Queensland using enclosures in which the density of the Australian gudgeon Hypseleotris spp. was manipulated. 2. Zooplankton biomass and community structure were strongly affected by fish density. A size dependent predation effect of Hypseleotris on zooplankton was observed at ambient fish densities, and the community shifted towards a dominance of copepod juveniles and nauplii. Substantial increases in the populations of Ceriodaphnia and calanoid copepods were observed at low fish densities and in the absence of fish. 3. At ambient fish densities total phytoplankton and the proportion of cyanobacteria were maintained at levels similar to those prevailing at day 0. Total phytoplankton and the proportion of cyanobacteria decreased substantially at low fish densities and in the absence of fish. Chlorophytes became dominant in the ‘no fish’ treatment and the grazing‐resistant species Oocystis and Dictyosphaerium were significantly higher than at ambient fish densities. 4. The experiment demonstrated a strong positive relationship between Hypseleotris density and cyanobacteria, and the results suggest that subtropical reservoirs may be suited to food web manipulation as a means of controlling summer cyanobacterial blooms.  相似文献

An infestation of young tilapia, Oreochromis mossambicus Peters, by the flagellate Cryptobia branchialiswas observed at the Salton Sea, California, in September, 1997. This is the first report of C. branchialis in a highly saline water-body (43 g l–1). Ultrastructure of C. branchialis as well as its effect on the gills of tilapia were studied using the scanning and transmission electron microscopy. No direct effect of C. branchialis on the epithelial cells of fish gills was observed. However, alterations of gill general structure, such as deposition of copious mucus on the gill surface, swelling of filaments, reduction of respiratory lamellae and their transformation into short club-shaped structures were found in infected fish. This suggests mortality of young tilapia may arise from decreased gill function in response to Cryptobia infestation.  相似文献

1. The effects of addition of juvenile perch (Perca fluviatilis) on the microcrustacean and rotifer communities associated with nymphaeid beds were studied, at three different plant densities [high (normal), medium (reduced by a half) and low (reduced to a third of normal)], in eighteen 2 m × 1 m enclosures in a shallow lake. 2. At the low and medium densities of lilies, Daphnia densities were high in the absence of perch but very low in the presence of perch. They increased, even in the presence of perch, to high densities (comparable with those in the absence of perch) at the highest plant density. Body sizes of Daphnia hyalina were consistent with high predation by perch at low and medium plant densities but reduced predation at high plant densities. Patterns of chlorophyll a concentration, in the presence of perch, inversely reflected those of D. hyalina density. 3. At naturally high densities but not at reduced densities, the plants appeared to act as refuges against predation for the Daphnia. Reductions in oxygen concentrations in the plant beds were not responsible for the refuge effect, nor could there be avoidance of the beds by the fish. The mechanism of the refuge effect must therefore lie in frustration of the process of capture of the Daphnia by the fish. 4. Numbers of other small Crustacea and rotifers were mostly unaffected by fish predation. Numbers of Asplanchna sp., Chydorus sphaericus and copepodites were higher in the presence of fish and, although there was no main effect of fish on numbers of Diaphanosoma brachyurum, there was a plant–fish interaction, with this species being less abundant in the presence of fish at low plant densities but more abundant in the presence of fish at medium and high plant densities. Main effects of plants were few, with only Asplanchna sp. and Keratella sp. decreasing in numbers with increasing plant density. Most taxa changed in numbers with time but interaction effects between time and plants, and fish and plants, were few.  相似文献

Several fish species are known to possess mechanisms that allow them to adapt to environments with different salinities. The aim of this study was to investigate the effects of salinity on the expression of aquaporins (aqp1a, aqp3a, aqp8a, and aqp9a) in the gills and intestines of Chinese black sleeper. After 30 days of acclimation, the expression of aqp1a, aqp3a, and aqp9a in the gills was significantly higher in fish transferred to 5 ppt than in those transferred to 40 ppt seawater, whereas aqp8 expression was lower. In contrast, aqp1a, aqp3a, and aqp8a expression in the intestines was higher in fish acclimated in 40 ppt than in those acclimated in 5 ppt. During abrupt salinity acclimation, the levels of aqp1a and aqp9a in the gills varied over time in fish acclimated in 5 ppt, but not in 40 ppt. The aqp3a levels in gills were higher in the 5 ppt group after 24 h than in the 40 ppt. The expression level of aqp8a in gills was higher in 40 ppt than in 5 ppt, except for that at 12 h. In the intestines, expression level of aqp1a and aqp8a were significantly upregulated from 12 to 48 h following acclimation in 40 ppt and aqp3a was higher in 40 ppt group than in 5 ppt, while aqp9a expression exhibited an opposite trend. These findings suggest that aqp1a, aqp3a, aqp8a and aqp9a may play a major osmoregulatory role in water transport in the gills and intestines during acclimation to different salinity environment.

A histology-based fish health assessment protocol was used in 2009–2010 to assess the health status of Clarias gariepinus from Pongolapoort Dam, South Africa. Nineteen fish were collected by angling. The histology of liver, kidney, gills and testes or ovaries was semi-quantitatively assessed and compared to that of fish from a reference site in the Okavango Panhandle, Botswana. Necropsy observations indicated that C. gariepinus from Pongolapoort Dam were in poorer condition than those from the Okavango Panhandle. Microscopic analysis showed that the mean liver index (I L), gill index (I G), testis index (I T) and ovary index (I O) were higher, although not statistically significant, in fish from the Okavango Panhandle. The difference in the results between the necropsy-based observations and semi-quantitative histological results was due to the external abnormalities detected in the necropsy analysis, including parasites and skin lesions, not expressing as changes to cell and tissue structure. According to the parameters measured, C. gariepinus from Pongolapoort Dam were in a healthy state.  相似文献

Invasive species represent a major threat with both direct and indirect effects on natural ecosystems, including effects on established and coevolved relationships. In a series of experiments, we examined how the interaction between two native species, a unionid mussel (Unio pictorum) and the European bitterling (Rhodeus amarus), a fish that parasitises unionids, was affected by the non-native zebra mussel (Dreissena polymorpha). The zebra mussel fouls hard substrates, including shells of living unionids, and its presence is often associated with a decrease in population density of native unionid mussels. Bitterling lay their eggs into live unionids and the embryos develop inside their gills. Using a range of zebra mussel densities, we demonstrated that zebra mussel fouling had a negative effect on the number of bitterling eggs inside the mussel host, with abundances of 5–10 zebra mussels (shell size 15–25 mm) per unionid critical for bitterling ability to utilise the host. In a further experiment, we found that bitterling did not discriminate between unfouled unionids and those fouled with five zebra mussels. Most ovipositions into fouled hosts, however, were unsuccessful as eggs failed to reach the unionid gills. We discuss implications of such unsuccessful ovipositions for bitterling recruitment and population dynamics.  相似文献

The existence of a layer of mucus covering the gill lamellae of healthy rainbow trout (Oncorhynchus mykiss) was investigated. Using cryo-scanning electron microscopy, a smooth, undulating, thin layer was observed which completely covered gill filaments and lamellae, thereby obscuring epithelial microridges. After processing cryopreserved gill arches in glutaraldehyde for conventional scanning electron microscopy, the layer was no longer present and epithelial microridges were clearly visible. The identity of this layer was investigated using cryopreserved gills which were treated in one of two ways. First, gills were incubated with a rabbit antiserum to gill mucus, with normal rabbit serum, or with phosphate-buffered saline. Following fixation in glutaraldehyde and processing, only the gill tissue incubated with the mucus-specific antiserum was still covered with the smooth layer. The layer was also retained on the gills of fish anesthetized in a solution containing mucusspecific antiserum and then processes in glutaraldehyde for conventional scanning electron microscopy. The tenacious nature of the mucous layer was demonstrated by its stability following exposure to formalin and a cationic detergent. Second, the presence of this layer was confirmed on gill tissue which was cryopreserved, followed by freeze-substitution and vapor fixation, and then examined by transmission electron microscopy.  相似文献

The concentrations of Fe, Mn, Ni, Pb and V in water, sediment and the gill, liver and muscle tissues of Synodontis resupinatus, Heterotis niloticus and Clarias gariepinus, all commercially important fish species of the lower Niger River, were investigated in 2015. Water, sediment and fish samples were collected for six months and heavy metals were determined using an Atomic Absorption Spectrometer. Fe ranked highest in water and sediment, with concentrations of 2.74 mg l?1 and 61.60 mg kg?1, respectively. Metals followed the magnitude of Fe > Mn > Ni > V > Pb in the water and Fe > Mn > V > Ni > Pb in the sediments. Metal concentrations were higher in the tissues of S. resupinatus compared with H. niloticus and C. gariepinus. Fe was also highest in the gills, liver and muscle of the three fish species. Its highest concentration of 132.97 mg kg?1 dry weight was recorded in the gills of S. resupinatus. Bioconcentration factors of metals ranged from 8.79 for Mn in H. niloticus muscle to 67.99 for Ni in S. resupinatus gills. The fish species studied pose no health risk for all metals studied, because the target hazard quotient was less than 1 and the estimated daily intakes of the metals were below the reference doses.  相似文献
